Abstract
patente de invenção: "clonagem, expressão e uso de fosfolipases ácidas". a presente invenção refere-se a uma nova sequência de dna, que codifica um polipeptídeo com atividade de fosfolipase essencialmente sem atividade de lipase, caracterizada pelo fato de que a sequência de dna é selecionada de a) sequência de dna que compreendem uma sequência de nucleotídeos de acordo com seq id no: 1, b) sequência de dna, que compreendem a sequência codificante de acordo com seq id no: 1, c) sequência de dna, que codificam a sequência de proteína de acordo com seq id no: 2, d) sequência de dna, que são codificadas pelo plasmídeo ppl3949-topo2.5 com a carta de restrição de acordo com a figura 7 e depositada sob o número de depósito dsm 22741, e) sequência de dna, que hibridizam em condições rigorosas com uma das sequências de dna de acordo com a),b) ou d), f) sequências de dna, que devido à degeneração do código genético são análogas às sequências de dna de acordo com a), b),c),d) ou e) e g) filamentos complementares às sequências de acordo com a) até f), em que a sequência de dna é preferencialmente derivada de aspergillus e de modo ainda mais preferido, de aspergillus fumigatus, bem com um polipeptídeo com atividade de fosfolipase essencialmente sem atividade de lipase selecionado de a) um polipeptídeo, que é codificado pela parte codificante de uma sequência de dna de acordo com uma das reivindicaçãoes 1 a 2, b) um polipeptídeo com a sequência de acordo com a seq id no: 2 ou de uma sequência derivada dessa, que pode ser obtido através de substituição, adição, deleção de um ou mais aminoácidos, c) um polipeptídeo com uma sequência, que apresenta uma identidade de pelo menos 83% para os aminoácidos 1 a 299 de seq id no: 2, d) um polipeptídeo, que é codificado por uma sequência de ácido nucleico, que hibridiza em condições rigorosas com (i) os nucleotídeos 55 a 1106 da seq id no: 1, (ii) a sequência de cdna contida nos nucleotídeos 55 a 1106 da seq id no: 1, (iii) uma sequência parcial de (i) ou (ii) de pelo menos 100 nucleotídeos ou (iv) um filamento complementar de (i), (ii) ou (iii), e) uma variante do polipeptídeo com seq id no: 2 compreendendo uma substituição, deleção e/ou inserção de um ou mais aminoácidos, f) variantes alélicas para as sequências de aminoácido a) até e).
