WERKWIJZE VOOR DE BEREIDING VAN OPTISCH AKTIEVE ALCOHOLEN
EN ESTERS, EN ALCOHOLEN EN ESTERS TOEGEPAST EN BEREID
IN DERGELIJKE WERKWIJZEN
De uitvinding betreft een werkwijze voor de bereiding van een optisch aktieve N-gesubstitueerde-3pyrrolidinol, waarbij een mengsel van de enantiomeren van een overeenkomstige N-gesubstitueerde-3-acyloxypyrrolidine verbinding wordt onderworpen aan een enzymatische enantioselektieve hydrolyse met behulp van een enzym met esterhydrolase aktiviteit.
Uit JP-A-1141600 is de enantioselektieve enzymatische hydrolyse van N-benzyl-3-acyloxypyrrolidine bekend.
De in JP-A-1141600 beschreven werkwijze heeft echter het nadeel dat de enzymatische hydrolyse wordt uitgevoerd bij lage substraatconcentraties. Bovendien kan de verkregen optisch aktieve N-benzyl-3-pyrrolidinol slechts via hydrogenolyse worden omgezet in optisch aktief 3-pyrrolidinol, hetgeen in de praktijk speciale voorzieningen vereist.
Optisch aktieve 3-pyrrolidinol, met name (R)-3pyrrolidinol, is een bekend tussenprodukt voor diverse farmaceutica. Het is derhalve van belang de optisch aktieve 3-pyrrolidinol met hoge enantiomere overmaat te verkrijgen. In de praktijk is het vaak nuttig, zo niet noodzakelijk, om bij de bereiding van optisch aktieve 3pyrrolidinol gebruik te maken van 3-pyrrolidinol waarvan het N-atoom beschermd is.
De enantiomere overmaat, die een maat is voor de enantiomere zuiverheid en wordt aangeduid als"e. e." (enantiomeric excess), is een veelal gebruikte grootheid.
Kort gezegd is de enantiomere overmaat gelijk aan het verschil van de hoeveelheden enantiomeren gedeeld door de som van de hoeveelheden enantiomeren, welk quotiënt door

vermenigvuldiging met 100 als percentage kan worden uitgedrukt.
De uitvinding beoogt een werkwijze te verschaffen welke het mogelijk maakt via enzymatische hydrolyse ook bij hoge substraatconcentratie optisch aktieve N-gesubstitueerde-3-pyrrolidinol met hoge e. e. te bereiden, die gemakkelijk kan worden omgezet in optisch aktieve 3-pyrrolidinol met hoge e. e.
Dit wordt volgens de uitvinding bereikt wanneer de N-gesubstitueerde-3-pyrrolidinol een verbinding is met de algemene formule (l) :

groepen in R of Rl, genoemd in formule (1) en/of (2), onafhankelijk van elkaar, 1-10 C-atomen de lineaire of vertakte alkyl, aryl, alkaryl of aralkylgroepen in R en kunnen, onafhankelijk van elkaar, tevens gesubstitueerd zijn met een of meerdere halo-atomen.
Verrassenderwijze is namelijk gevonden dat, wanneer als N-substituent een acylgroep of een acyloxygroep, bij voorkeur een acylgroep, wordt toegepast, zowel een zeer selectieve enzymatische hydrolyse kan worden uitgevoerd in een commercieel aantrekkelijk proces, als een, gemakkelijk tot de alcohol 3-pyrrolidinol hydrolyseerbaar produkt, wordt verkregen.
De reaktieomstandigheden voor enantioselectieve hydrolyse zijn niet erg kritisch. Men voert de hydrolyse meestal uit in waterig milieu, maar ook mengsels van water met organische oplosmiddelen kunnen worden gebruikt. Het is echter ook goed mogelijk gebleken de hydrolyse uit te voeren, waarbij het substraat niet is opgelost in een oplosmiddel. Water mengbare en niet water mengbare oplosmiddelen zoals DMF, DMSO, ethanol, methanol, tolueen, hexaan, isoöctaan, MTBE, dichloormethaan, e. a. kunnen in het reaktiemengsel aanwezig zijn. De concentratie van het substraat in het reaktiemengsel is bij voorkeur zo hoog mogelijk. In de praktijk blijken substraatconcentraties in het reaktiemengsel van meer dan 25% in het bijzonder meer dan 50% mogelijk.
Tevens is gebleken dat ook de overeenkomstige enantioselektieve (trans) verestering bijzonder goed verloopt. De uitvinding heeft dan ook tevens betrekking op een werkwijze voor de bereiding van een optisch aktieve Ngesubstitueerde-3-acyloxy-pyrrolidine verbinding, waarbij een mengsel van de enantiomeren van de overeenkomstige Ngesubstitueerde-3-pyrrolidinol wordt onderworpen aan een enzymatische, enantioselektieve (trans) verestering in aanwezigheid van een geschikt enzymatisch veresterings-

middel met behulp van een enzym met esterhydrolase aktiviteit, waarbij een N-gesubstitueerde-3-pyrrolidinol met formule (1), waarin R als in conclusie 1 gedefinieerd is, wordt toegepast, en dat de N-gesubstitueerde-3acyloxy-pyrrolidine verbinding een ester volgens formule (2)
is waarin R en R1 als in conclusie 1 gedefinieerd zijn.
De enantioselektieve enzymatische (trans) verestering van N-benzyl-3-pyrrolidinol is beschreven in JP-A-4131093. De hier beschreven werkwijze heeft naast de hierboven met betrekking tot JP-A-1141600 beschreven nadelen van lage substraatconcentratie tijdens de enzymatische reaktie en minder gemakkelijke omzetting in 3-pyrrolidinol, tevens als nadeel dat produkt met lagere e. e. wordt verkregen.
Daarnaast is uit Tetrahedron : Asymmetry Vol. 3, No. 8, p. p. 1049-1054,1992 een enzymatische hydrolyse of verestering bekend van een ander substraat, nl. 3-

(hydroxymethyl) danwel een overeenkomstige ester, m. lipase. Ook hier wordt de enzymatische piperidine,(trans) verestering of hydrolyse uitgevoerd bij lage substraat-concentraties, waardoor een commercieel minder aantrekkelijk proces wordt verkregen. Bovendien blijkt uit het genoemde artikel dat de scheiding van de reaktieprodukten via extractie - welk als een veel gemakkelijker vorm van opwerking wordt gezien dan de voorgestelde selektieve kristallisatie-praktisch niet mogelijk is.
De enzymatische (trans) verestering kan zowel in puur substraat als in een geschikt organisch oplosmiddel met een zeer laag watergehalte plaatsvinden. Als oplosmiddelen kunnen bijvoorbeeld gebruikt worden ketonen, alkanen, cyclische ethers, dialkylethers, chloorhoudende alkanen en alkylaromaten, in het bijzonder methyl-ethylketon, methyl-isobutyl-keton, 2-pentanon, isoöctaan, dioxaan, di-n-butylether, tolueen, hexaan, cyclohexaan, dichloor-methaan, met als voorkeur dichloormethaan, 2pentanon en methyl-ethyl-keton. De concentratie van het

substraat in het reaktiemengsel is bij voorkeur zo hoog mogelijk. In de praktijk blijken substraatconcentraties, berekend ten opzichte van de totale hoeveelheid oplosmiddel en substraat, in het reaktiemengsel hoger dan 25% in het bijzonder hoger dan 50% mogelijk.
Bij voorkeur wordt een substraatconcentratie groter dan 90% toegepast.
Daarnaast bevat het reactiemengsel tevens een geschikt enzymatisch veresteringsmiddel waarin het substraat in principe ook opgelost kan zijn.
Verrassenderwijze is gebleken dat in de werkwijzen volgens de uitvinding hoge substraatconcentraties kunnen worden toegepast.
Tevens heeft aanvraagster gevonden dat de scheiding van reaktieprodukten eenvoudig via extractie kan worden uitgevoerd. Gebleken is namelijk dat het mogelijk is om door geschikte keuze van het extractiemiddel eerst de ester te isoleren. Vervolgens kan desgewenst, door een ander extractiemiddel te kiezen, de alcohol worden geïsoleerd. Verrassenderwijze is namelijk gebleken, dat zelfs wanneer zowel de ester als de alcohol ieder voor zieh, in het extractiemiddel oplosbaar zijn, bij toepassing van een geschikt gekozen extractiemiddel op het verkregen reaktiemengsel met daarin water, alleen de ester geëxtraheerd werd en de alcohol in het resterende reaktiemengsel achter bleef.
Geschikte extractiemiddelen om de ester te extraheren bleken onder meer apolaire oplosmiddelen zoals bijvoorbeeld alkanen met 5 of meer Catomen, en dialkylethers met 4 of meer C-atomen ; voorbeelden van bijzonder geschikte extractiemiddelen om de ester te isoleren zijn hexaan en MTBE (methyl-tertiairbutyl-ether). Geschikte extractiemiddelen om de alcohol te isoleren zijn bijvoorbeeld chloorhoudende alkanen met 3 of minder C-atomen en alkylaromaten met 7-10 C-atomen ; voorbeelden van bijzonder geschikte extractiemiddelen om de alcohol te isoleren zijn dichloormethaan en tolueen.
De enzymen die tijdens de hydrolyse of (trans)- verestering kunnen worden toegepast zijn enzymen met

esterhydrolase activiteit. Bij de enzymatische (trans) verestering kunnen in principe dezelfde enzymen (microorganismen) gebruikt worden als bij de hydrolyse.
Bekende groepen zijn lipasen en esterasen. Geschikte voorbeelden zijn enzymen afkomstig van het genus, Candida, Mucor, Rhizopus, Pseudomonas, Penicillum, Chromobacterium, Asperqillus en Humicola.
Dergelijke enzymen zijn via algemeen bekende kweekmethoden verkrijgbaar. Vele worden op technische schaal geproduceerd en zijn in de handel verkrijgbaar. Het enzympreparaat zoals dat bij de onderhavige uitvinding wordt gebruikt, wordt niet beperkt door zuiverheid e. d. en kan zowel een ruwe enzymoplossing als een gezuiverd enzym zijn, maar het kan ook bestaan uit (gepermeabiliseerde en/of geimmobiliseerde) cellen, die de gewenste aktiviteit bezitten, of uit een homogenaat van cellen met een dergelijke aktiviteit. Het enzym kan ook in geimmobiliseerde vorm of in chemische gemodificeerde vorm worden gebruikt. Wanneer in het gebruikte enzympreparaat ook enige ongewenste tegengestelde enzymaktiviteit aanwezig is verdient het aanbeveling deze ongewenste aktiviteit te verwijderen of te onderdrukken teneinde een maximale enantioselektiviteit te verkrijgen.
De uitvinding wordt op geen enkele wijze beperkt door de vorm waarin het enzym voor de onderhavige uitvinding wordt gebruikt.
Binnen het kader van de uitvinding kan uiteraard ook een enzym worden gebruikt dat afkomstig is van een mutant of van een genetisch gemodificeerd microörganisme.
Bijzonder geschikt is gebleken Chromobacterium viscosum en lipase, afkomstig van Candida antarctica. Dit lipase kan bijvoorbeeld worden geproduceerd via recombinant-DNA-technologie. Het gen coderend voor het betreffende lipase wordt daarbij heteroloog tot expressie gebracht in een gastheer micro-organisme, bijvoorbeeld Asperqillus oryzae. Het lipase van Candida antarctica is in de handel verkrijgbaar, bijvoorbeeld onder de merknaam SP435 en SP535 van NOVO.

Gebleken is dat het lipase van Candida antarctica nagenoeg volledig selektief is, waardoor het mogelijk is om zowel de beide enantiomeren van de ester als de alcohol met hoge e. e. te verkrijgen.
De werkwijze volgens de uitvinding is niet beperkt tot een specifiek type reaktor en kan bijvoorbeeld uitgevoerd worden in een continu-, een batch-, een fedbatch of een membraanreaktor.
In de werkwijze volgens de uitvinding zal meestal worden uitgegaan van een (nagenoeg) racemisch mengsel van N-gesubstitueerde-3-pyrrolidinol, wanneer een enantioselektieve enzymatische (trans) verestering wordt uitgevoerd, of van N-gesubstitueerd-3-acyloxypyrrolidine, wanneer een enantioselektieve enzymatische hydrolyse wordt uitgevoerd. Met een nagenoeg racemisch mengsel wordt in het kader van de uitvinding bedoeld een mengsel van enantiomeren, waarbij de e. e. lager is dan 10%.
De uitgangsverbindingen kunnen op bekende wijze worden bereid via N-acylering van racemisch 3pyrrolidinol, gevolgd door verestering van het zo verkregen produkt.
De enzymatische hydrolyse, danwel (trans) verestering, zal, hoewel dit geen kritische parameter is, meestal worden uitgevoerd bij een

temperatuur van 0-50 C, bij voorkeur bij een temperatuur van 25-45 C. De druk, waarbij de reaktie wordt uitgevoerd, kan binnen wijde grenzen variëren. In de praktijk zal de reaktie meestal bij atmosferische druk worden uitgevoerd.
De pH, waarbij de reaktie wordt uitgevoerd, is evenmin kritisch en ligt meestal tussen 3 en 10, bij voorkeur tussen 6 en 9.
Geschikte oplosmiddelen die kunnen worden toegepast in de enzymatische hydrolyse of (trans)- verestering zijn bijvoorbeeld chloroform, dichloormethaan, benzeen, xyleen, trichloormethaan, isopropylether, 3-pentanon, methyl tertiairbutylether, methyl-isobutylketon, cyclohexanon, isooctaan, ethylacetaat, e. a.

Bij de enantioselektieve enzymatische (trans)- verestering kunnen in principe alle bekende, voor dergelijke processen geschikte veresteringsmiddelen worden toegepast. Voorbeelden van dergelijke veresteringsmiddelen zijn zuren in een geactiveerde vorm zoals bijvoorbeeld anhydriden en esters.
Door de keuze van de uitgangsverbindingen is het mogelijk gebleken een commerciëel aantrekkelijke route voor de bereiding van optisch aktieve 3-pyrrolidinol te ontwerpen. De uitvinding heeft derhalve ook betrekking op de N-acyl-3-pyrrolidinol verbindingen en de N-acyl-3acyloxy-pyrrolidine verbindingen, toegepast in de werkwijze volgens de uitvinding, alsmede op de (R)-en

(S)-enantiomeren daarvan.

In het bijzonder bevatten de lineaire, cyclische of vertakte alkyl-, alkenyl-, aryl-, alkaryl- of aralkylgroepen in R of Rl, genoemd in formule (1) en/of (2), onafhankelijk van elkaar, 1-10 C-atomen ; de lineaire, cyclische of vertakte alkyl, aryl, alkaryl of aralkylgroepen in R en Rl kunnen, onafhankelijk van elkaar, tevens gesubstitueerd zijn met één of meerdere halo-atomen.
Een voorkeur gaat uit naar verbindingen waarin R en Rl identiek zijn, aangezien deze verbindingen eenvoudig met een acyleringsmiddel kunnen worden bereid. Daarnaast geldt enerzijds globaal dat wanneer R en/of Rl meer Catomen bevatten een betere procesvoering wordt bereikt, terwijl anderzijds het werken met verbindingen waarbij R en/of Rl minder C-atomen bevatten, commerciëel aantrekkelijker is. Bij voorkeur geldt voor de verbindingen toegepast en/of verkregen in de werkwijze volgens de uitvindingen, dat R en/of R1 2-5 koolstofatomen bevatten. De beste resultaten werden bereikt, wanneer R en Rl beide ethyl of propyl voorstellen.
De uitvinding zal nu verder worden toegelicht aan de hand van de volgende voorbeelden zonder evenwel daartoe te worden beperkt.
Voorbeeld I Enzymatische hydrolyse
4, 95 gram racemisch N-butyryl-3-pyrrolidinylbutyraat werd toegevoegd aan 20 ml Kpi (kaliumfosfaat) buffer van pH 8. Na toevoeging van 1000 mg Candida antarctica lipase (SP 435 ; NOVO) werd de pH constant gehouden op 8 middels een automatische titratie met 1 N kalium-hydroxide oplossing. De temperatuur werd op 300C

gehandhaafd. De reaktie werd gestopt door filtratie van het reaktiemengsel waarbij de biokatalysator als residu achterbleef. De biokatalysator werd gewassen met 2 * 10 ml water. Na het instellen van de pH op 3 werd uit de verzamelde waterlagen d. m. v. een continue extractie met hexaan de (S)-N-butyryl-3-pyrrolidinyl-butyraat geïsoleerd.
Uit de overgebleven waterlaag werd vervolgens m. b. v. een continue extractie met tolueen de (R)-Nbutyryl-3-pyrrolidinol geïsoleerd. De geïsoleerde (R)-N-

butyryl-3-pyrrolidinol in tolueen werd m. 6 N zoutzuur gedurende 17 uur bij 1000C gehydrolyseerd tot (R)-3pyrrolidinol als zoutzure zout, waarna m. b. v. tolueen het aanwezige water azeotropisch werd verwijderd. De (R)-3pyrrolidinol. HCl kristalliseerde vervolgens uit en kon na filtratie als hygroscopische kristallen verkregen worden met een e. e. > 98%.
Voorbeeld II Enzymatische hydrolyse
23, 3 gram racemisch N-butyryl-3-pyrrolidinylbutyraat werd toegevoegd aan 2, 5 ml Kpi buffer van pH 8.
Na toevoeging van 1980 mg Candida antarctica lipase (SP 435 ; NOVO) werd de pH constant gehouden op 8 middels een automatische titratie. De temperatuur werd op 400C gehandhaafd. De reactie werd gestopt door filtratie van het reaktiemengsel waarbij de biokatalysator als residu achterbleef. De bio-katalysator werd gewassen met 2 * 10 ml water. De e. e. van het gevormde (R)-N-butyryl-3pyrrolidinol bedroeg 91%.
Voorbeeld III Enzymatische hydrolyse
24, 1 gram racemisch N-butyryl-3-pyrrolidinylbutyraat werd toegevoegd aan 0, 5 ml Kpi buffer van pH 8.
Na toevoeging van 1900 mg Candida antarctia lipase (SP 435 ; NOVO) werd de pH constant gehouden op 8 middels een automatische titratie. De reaktie werd gestopt door

filtratie van het reaktiemengsel waarbij de biokatalysator als residu achterbleef. De biokatalysator werd gewassen met 2 * 10 ml water. De temperatuur werd op 400c gehandhaafd. De e. e. van het gevormde (R)-N-butyryl-3pyrrolidinol bedroeg 91%.
Voorbeeld IV Enzymatische hydrolyse
0, 2 gram racemisch N-butyryl-3-pyrrolidinylbutyraat werd toegevoegd aan 2 ml Kpi-buffer van pH 8. Na toevoeging van 30 mg enzym werd de pH constant gehouden op 8 middels een automatische titratie. De temperatuur werd op 400C gehandhaafd. De reaktie werd gestopt door verlaging van de pH naar pH 3. Vervolgens werd de e. e. bepaald van het gevormde (R)-N-butyryl-3-pyrrolidinol met behulp van HPLC. De resultaten, zonder optimalisatie naar

Voorbeeld V Enzymatische (trans) veresterina
20 gram racemisch N-acetyl-3-pyrrolidinol werd toegevoegd aan 500 ml dichloormethaan en 25 ml butaanzuuranhydride. Na toevoeging van 2 g Candida antarctica lipase (SP 435 ; NOVO) werd de reaktie na 2 uur, d. m. v. filtratie van het enzym, gestopt.

gehandhaafd. De e. e van het gevormde (R)-N-acetyl- 3-pyrrolidinyl-butyraat bedroeg 95%. Het reaktieprodukt, (R)-N-acetyl-3-pyrrolidinyl-butyraat werd na verwijderen van de dichloormethaan, d. m. v. een filmverdamper, verwarmd met 6 N zoutzuur op 1000c gedurende 17 uur. Na de hydrolyse werd het (R)-3-pyrrolidinol als zoutzure zout verkregen.
Voorbeeld VI Enzymatische (trans) veresterincr
Aan 0, 5 gram racemisch N-acetyl-3-pyrrolidinol werd 0, 5 ml butaanzuuranhydride en 1 microliter water toegevoegd. Na toevoeging van 50 mg Candida antarctica (SP 435 ; NOVO) lipase werd de reaktie na 60 minuten door filtratie van het enzym gestopt. De temperatuur werd op 300C gehandhaafd. De e. e. van het gevormde (R)-N-acetyl-3pyrrolidinyl-butyraat bedroeg 87%. Na hydrolyse met behulp van 6 N zoutzuur werd het (R)-3-pyrrolidinol als zoutzure zout verkregen.

IN SUCH METHODS
The invention relates to a method for the preparation of an optically active N-substituted-3-pyrrolidinol, wherein a mixture of the enantiomers of a corresponding N-substituted-3-acyloxypyrrolidine compound is subjected to an enzymatic enantioselective hydrolysis by means of an enzyme with ester hydrolase activity .
JP-A-1141600 discloses the enantioselective enzymatic hydrolysis of N-benzyl-3-acyloxypyrrolidine.
However, the method described in JP-A-1141600 has the drawback that the enzymatic hydrolysis is performed at low substrate concentrations. Moreover, the resulting optically active N-benzyl-3-pyrrolidinol can only be converted into optically active 3-pyrrolidinol via hydrogenolysis, which in practice requires special provisions.
Optically active 3-pyrrolidinol, in particular (R) -3pyrrolidinol, is a known intermediate for various pharmaceuticals. It is therefore important to obtain the optically active 3-pyrrolidinol with a high enantiomeric excess.
