La présente invention est relative ä des préparations pharmaceutiques en suppositoires bioadhésives, qui contiennent un ou plusieurs polymères hydrophiles et des quantités relativement grandes d'eau, c'est-à-dire excédant 30 % en poids de Ja préparation pour obtenir une adherence et une rétention améliorées de médicament soluble dans l'eau ou insoluble dans l'eau en un site desire.

La formulation de suppositoire bioadhésive de l'invention est formée d'un ingrédient ou médicament thérapeutiquement actif soluble dans l'eau ou insoluble dans l'eau, d'un hydrocolloide ou polymère hydrophile soluble dans t'eau ou insoluble dans J'eau qui s'hydrate, devient adhésif et augmente le temps de rétention du médicament au site de traitement, d'eau en une quantité d'au moins environ 30 % en poids de la formulation, et d'une composition de base pour suppositoire de faible point de fusion qui fond à la température corporelle, forme une émulsion avec l'eau, et favorise la dispersion de l'hydrocolloide et du médicament autour des zones désirées.

On chauffe le Witepsol 558 jusqu'à fusion (environ
35 C) et on le conserve à une temperature d'environ 45 C jusqu'à ce que les autres composants soient prêts à être ajoutés.
On réduit la température de la masse à 40 C par refroidissement dans de l'air avec agitation et on agite le liquide résultant. On ajoute de l'eau et on agite le melange dans un mélangeur Sitverson pour former une émulsion. On ajoute ensuite la nystatine suivie de I'hydrocolloide (carboxyméthylcelJutose de sodium). On poursuit l'agitation pendant 10 minutes jusqu'à l'obtention d'une suspension uniforme. Tout au long du processus ci-dessus, la température des divers me ! anges est maintenue au dessus d'environ 35-37 C.
On poursuit alors l'agitation et on verse la suspension liquide résultante dans des moules appropries pour former des suppositoires d't g.
Exemple 2
On prépare des suppositoires à Ja nystatine Cl g chacun) répondant à la composition suivante en suivant la procédé de l'exemple 1, à l'exception que l'on ajoute un agent de neutralisation à l'eau de manière à obtenir un pH de 6 avec l'émulsion et qu'on utilise le Witepsol S55 comme base de suppositoire.
