Die Erfindung betrifft ein Verfahren zur Herstellung eines neuen Diaminocyclohexandiols.
Die Erfindung bezieht sich somit auf ein Verfahren zur Herstellung des neuen 1D- (1, 3, 5/2) -1, 5-Di- amino-2,3-cyclohexandiols der Formel

Die Erfindung umfasst auch die Herstellung der Säureadditionssalze des Diaminocyclitols der Formel (1).
Diese Säureadditionssalze können entweder Einfach- (Mono)-salze sein, d. h., dass sie aus 1 Molekül des Diaminocyclitols und 1 Molekül einer organischen oder anorganischen Säure gebildet werden, oder sie können Doppel- (Di)-salze sein, d. h., dass sie aus 1 Molekül Diaminocyclitol und 2 Molekülen einer organischen oder anorganischen Säure gebildet werden.

Die übrigen Verbindungen der allgemeinen Formel (vit) können nach dem gleichen Verfahren hergestellt werden, wie es in der obgenannten belgischen Patentschrift bzw. deutschen Offenlegungsschrift zur Herstellung der Verbindungen der obigen allgemeinen Formel (VI) beschrieben wird.
Das Diaminocyclitol der Formel (I) und seine Salze stellen sehr wertvolle Zwischenprodukte für die Herstellung von Substanzen mit antibakteriellen Eigenschaften nach bekannten Verfahren dar.
Sie sind deshalb so besonders wertvoll, weil die chemische Struktur des Diaminocyclitols eine sterische Anordnung von Amino- und Hydroxygruppen aufweist, die analog, aber trotzdem verschieden von den natürlichen Diaminocyelitolen ist, die im Molekülaufbau von Antibiotika der Hybrimycinreihe enthalten sind.
Die Erfindung wird durch das folgende Beispiel, auf welches sie jedoch nicht beschränkt ist, näher erläutert : Beispiel : Herstellung von 1D- (1, 3, 5/2) -1, 5-Diamino-2, 3-cyclohexandiol-Dihydrochlorid :
Zu einer Lösung von 0, 49 g ID- (l, 3, 5/2) -1, 5-iazido-2, 3-cyclohexandiol, das wie oben beschrieben hergestellt wurde, in 25 cm3 Äthanol werden 2 cm3 Raney-Nickel (d. i. ein Katalysator aus feinteiligem Nickel, der erhalten wird, indem das Aluminium aus einer Nickel/Aluminium-Legierung mit Alkali herausgelöst wird) zugesetzt.
Das Reaktionsmedium wird 5 h hydriert. Der Katalysator wird dann durch Filtrieren auf einem im Handel erhältlichen Kieselgur-Produkt abgetrennt und mehrmals mit einer 50 Teile Methanol/50 Teile WasserMischung gewaschen. Die hydrierte Lösung und die Waschflüssigkeit wird vereinigt, und das Ganze wird zur Trockne eingedampft. Auf diese Weise erhält man 0, 33 g 1D- (1, 3, 5/2) -1, 5-Diamino-2, 3-cyclohexandiol als leicht violett gefärbten Schaum. Dieser Schaum bleibt homogen in einer Dünnschichtchromatographieprüfung auf Cellulose unter Verwendung einer Mischung von 2 Teilen Pyridin + 1 Teil Ammoniaklösung + 2 Teilen Äthanol + 1 Teil Wasser als Lösungsmittel.

Die so erhaltenen 0, 33 g Diamin werden in 5 ems abs. Methanol und 3,5 om3 einer Methanol/10% Salz- säure-Lösung aufgenommen.
Die Lösung wird 12 h bei 00C stehen gelassen, und die entstandenen Kristalle werden auszentrifugiert.
