Verfahren zur Herstellung von neuen Piperidinderivaten
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen Piperidinderivaten mit wertvollen pharmakologischen Eigenschaften.
Es wurde überraschenderweise gefunden, dass Piperidinderivate der allgemeinen Formel I :

in welcher Ri einen Alkylrest mit höchstens 12 Kohlenstoffatomen, einen Alkenylrest mit 3-5 Kohlenstoffatomen, den Cyc1opropylmethylrest oder einen Phenyl-alkylrest mit 7-9 Kohlenstoffatomen, R Wasserstoff oder den Methylrest, Rg einen Alkylrest mit höchstens 4 Kohlenstoffatomen, den Phenylrest, einen Phenylalkylrest mit

Trimethylen- bis Hexamethylenrest und R4 Wasserstoff oder den Methylrest bedeutet, und ihre Salze mit anorganischen und organischen Säuren wertvolle pharmakologische Eigenschaften,
insbesondere eine ausgezeichnete analgetische Wirksamkeit bei oraler wie parenteraler Applikation und starke antitussive Wirksamkeit besitzen. Im Gegensatz zu andern Analgetica besitzen sie keine para-sympathicolytischen Eigenschaften, sondern wirken eher para- sympathicomimetisch. Sie sind zugleich relativ wenig toxisch und eignen sich deshalb z. B. zur
Linderung und Behebung von Schmerzen verschiedener Genese und auch des Hustenreizes.
In den Verbindungen der allgemeinen Formel I und den zugehörigen, weiter unten genannten Ausgangsstoffen ist Ri z. B. durch Alkylreste, wie den Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n-Butyl-, Isobutyl-, sek. Butyl-, n-Amyl-, Isoamyl-, n-Hexyl-, n-Octyl-, n-Decyl-oder n-Dodecylrest ; durch Alkenylreste wie den Allyl-, Crotyl-, Methallyl-oder y. y-Dimethylallylrest ; durch den Cyclopropylmethylrest oder durch Phenylalkylreste, wie den Benzyl-, ss-Phenyl-äthyl-oder y-Phenyl-propyl-rest verkörpert. Die Verbindungen der allgemeinen Formel I lassen sich, wie weiter gefunden wurde, in überraschend einfacher Weise herstellen, indem man eine Verbindung der allgemeinen Formel II :

wobei R2, R3 und R4 die oben angegebene Bedeutung haben, mit einem reaktionsfähigen Ester einer Verbindung der allgemeinen Formel III :
R1-OH, (III) in welcher R die oben angegebene Bedeutung hat, umsetzt. Die Umsetzung erfolgt beispielsweise bei Raumtemperatur oder mässig erhöhter Temperatur in einem geeigneten organischen Lösungsmittel, wie z. B. Äthanol, Aceton, Äthylacetat oder Dimethylformamid. Gegebenenfalls wird die Umsetzung durch Zusatz säurebindender Mittel, wie z. B. Kaliumcarbonat, und/oder von Kaliumjodid beschleunigt. Als reaktionsfähige Ester eignen sich insbesondere Halogenwasserstoffsäureester, wie Bromide, Chloride und Jodide, weiter Arylsulfonsäureester, z.
B. p-Toluolsulfonsäureester sowie leicht zugängliche Schwefelsäureester, wie Dimethylsulfat und Diäthylsulfat.
Mit anorganischen und organischen Säuren wie Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Salpetersäure, Phosphorsäure, Methansulfonsäure, Äthandisulfonsäure, ss-Hydroxy-äthansulfonsäure, Essigsäure, Propionsäure, Maleinsäure, Fumarsäure, Milchsäure, Äpfelsäure, Weinsäure, Citronensäure, Benzoesäure, Salicylsäure, Phenylessigsäure und Mandelsäure bilden die Verbindungen der allgemeinen Formel I Salze, die teilweise, gut wasserlöslich sind.
Die nachfolgenden Beispiele erläutern die Durchführung der erfindungsgemässen Verfahren, stellen jedoch keineswegs die einzigen Ausführungsformen derselben vor. Die Temperaturen sind in Celsiusgraden angegeben.

:Quecksilber-lI-sulfat 15 Stunden bei Raumtemperatur stehen gelassen. Dann wird das Reaktionsgemisch mit Natronlauge alkalisch gestellt und mit Chloroform ausgezogen. Die Chloroformlösung wird getrocknet und eingedampft. Der Rückstand von 4, 1 g wird in 20 ml Methanol gelöst, mit 10 g an Benzylbromid versetzt und 48 Stunden bei 200 stehen gelassen. Dann wird das Gemisch unter Vakuum eingedampft, Wasser zugefügt und die sauer reagierende Lösung mit Äther geschüttelt.
Die wässerige Phase wird alkalisch gestellt, und mit Chloroform ausgezogen, die Chloroformlösung getrocknet und eingedampft und der Rückstand im Hochvakuum destilliert. Das l- (l'-Benzyl-4'-hydroxy-4'-piperidyl]-2-propanon siedet im Kugelrohr unter 0, 01 Torr bei 115-125 o. Das Hydrochlorid schmilzt bei 166-168 o. b) In analoger Weise wird unter Verwendung von 8 g Methyljodid an Stelle des Benzylbromids das 1- (1' -Methvl-4'-hydroxy-4'-piperidyl) -2-propanon erhalten, Smp. des Citrats 103-105 .
Beispiel 2 : g g l- (4'-Hydroxy-4'-piperidyl)-2-propanon werden in 5 ml Äthanol gelöst mit 3 ml Allylbromid versetzt und 24 Stunden stehen gelassen. Hierauf wird das Reaktionsgemisch eingedampft, der Rückstand in 2 n Salzsäure gelöst, die mit Äther geschüttelt, die wässerige Phase alkalisch gestellt und mit Chloroform ausgegossen. Die Chloroformlösung wird getrocknet, eingedampft und der Rückstand im Hochvakuum destilliert. Das l- (l'-Allyl-4'-hydroxy-4'-piperidyl)-2-propanon geht unter 0, 01 Torr bei 105-125 Luftbadtemperatur über, Smp. des Citrats 75-780.
Beispiel 3 : 2 g 1- (4'-Hydroxy-4'-piperidyl-2-propanon werden mit 4 g n-Hexylbromid, l g Kaliumjodid und l g Kaliumcarbonat in 25 ml Aceton 4 Stunden unter Rückfluss gekocht. Dann wird das Reaktionsgemisch eingedampft, mit wenig Wasser versetzt und die alkalische Lösung mit Chloroform extrahiert.
Die Chloroformlösung wird getrocknet, und im Vakuum eingedampft und der Rückstand im Hochvakuum
