Verfahren zur Herstellung von Guajacolverbindungen.
Im Stammpatent Nr. 150477 ist ein Verfahren zur Darstellung von Guajacolverbindungen geschützt, das darin besteht, dass man Phosphorsäuremono-bzw.-bis- [2-methoxyphenyl]-ester mit

'ausgeführt werden, dass man anorganische, vorzugsweise in wasserhaltigen Flüssigkeiten lösliche Salze, wie Alkalisalze des Phosphorsäure-mono-bzw. -bis- [2-methoxyphenyl] -esters, mit Salzen organischer Basen, z. B. halogenwasserstoffsauren Salzen, zur Umsetzung bringt. Ferner kann man auch Säurederivate, vorzugsweise Halogenide des Phosphorsäure-mono-bzw.-bis-ss-methoxyphenyl]-esters, mit organischen Basen in Gegenwart von Wasser zur Umsetzung bringen.
Es hat sich gezeigt, dass man bei Anwendung von aliphatischen Aminen, z. B. n-Amylamin, Isoamylamin, als basische Komponente salzartige Guajacolverbindungen mit ebenfalls sehr wertvollen therapeutischen Eigenschaften erhält. Es entstehen so neue Verbindungen von sehr guter Löslichkeit und leichter Resorbierbarkeit durch den Organismus, in dem das Guajacol seine Wirksamkeit sehr gut entfaltet, ohne die nachteiligen Wirkungen des freien Guajacols auszuüben.
Unter den nach vorliegendem Verfahren erhältlichen Verbindungen haben sich jene vorzüglich bewährt, die man erhält, wenn man Oxyalkylamine, z. B. Mono-, Di-oder Triäthylolamin bzw. mehrwertige aliphatische Amine, wie z. B. Äthylendiamin, Tetramethylendiamin, zur Umsetzung verwendet.
Durch Einwirkung eines Überschusses bzw. einer mehrfach molekularen Menge eines aliphatisehen Amins können auch in manchen Fällen Molekülverbindungen der erfindungsgemäss herstellbaren Salze mit den freien Basen erhalten werden.
Beispiel 1-620-4 g (2 Mol) Phosphorsäure-bis-[2-methoxyphenyl]-ester und 61-pa--- (1 Mol + 2-3% Überschuss) Äthylendiamin werden vermischt und dann unter mässiger Erwärir auf dem Wasserbade eingedampft. Der honigartige Rückstand kristallisiert nach längerer Küh ; mit Eis ; es scheidet sich eine gelbliche Kristallmasse ab, die zwischen Filtrierpapier abgep und aus wenig Alkohol umkristallisiert wird. Man erhält so ein weisses, kristallines, leicht war lösliches Produkt vom F = 143 (unkorr. ). Die Methoxylbestimmung ergab : 17'9% ; beree für C20H28O12N2P2 : 18-2%.
Beispiel 2 : 310'2 g (1 Mol) Phosphorsäure-bis-[2-methoxyphenyl]-ester und 149'2 g (1 Triäthylolamin werden vermischt und in einer Schale auf dem Wasserbade eingedampft. Das h' entstehende Reaktionsprodukt wird auf der Nutsche abgesaugt und hierauf zweimal aus Alk umkristallisiert. Die erhaltene Verbindung ist eine farblose, gut kristallisierende, in Wasser l ( lösliche Masse von angenehmem Geschmack, die vom Organismus äusserst leicht resorbiert v F = 1000 (unkorr.).
Beispiel 3 : 330 g Phosphorsäure-bis-[2-methoxyphenyl]-esterchlorid werden mit einer Lösung von 300 g Triäthylolamin in 500 cm3 Wasser vermischt. Alsbald tritt Umsetzung ein, die durch etwa einstündiges Erhitzen des Reaktionsgemisches auf dem Wasserbade vervollständigt wird. Nach dem Abkühlen werden etwa 2500 cm3 Äthylalkohol zugefügt. Hiebei scheidet sich als Nebenprodukt Tri- äthylolamin-hydrochlorid aus, welches abfiltriert wird. Das Filtrat hinterlässt nach Abdampfen der alkoholischen Flüssigkeit einen Rückstand, der aus Alkohol umkristallisiert das entstandene Triäthylol- aminsalz liefert ; F = 99-1000 (unkorr. ).

1. Ausführungsform des Verfahrens gemäss Stammpatent Nr. 150477, dadurch gekennzeichnet, dass man als organische Basen aliphatische Amine verwendet.

Among the compounds obtainable by the present process, those that are obtained when using oxyalkylamines, e.g. B. mono-, di- or triethylolamine or polyvalent aliphatic amines, such as. B. ethylenediamine, tetramethylenediamine, used for the implementation.
By the action of an excess or a multiple molecular amount of an aliphatic amine, it is also possible in some cases to obtain molecular compounds of the salts which can be prepared according to the invention with the free bases.
Example 1-620-4 g (2 mol) of phosphoric acid bis [2-methoxyphenyl] ester and 61-pa --- (1 mol + 2-3% excess) ethylenediamine are mixed and then, with moderate heating, on the water bath evaporated. The honey-like residue crystallizes after a long period of cooling; with ice ; a yellowish crystal mass separates out, which is popped between filter paper and recrystallized from a little alcohol. A white, crystalline, easily soluble product with an F = 143 (uncorrected) is obtained. The methoxyl determination gave: 17'9%; beree for C20H28O12N2P2: 18-2%.
Example 2: 310'2 g (1 mol) of phosphoric acid bis [2-methoxyphenyl] ester and 149'2 g (1 triethylolamine are mixed and evaporated in a bowl on the water bath. The reaction product formed is on the suction filter sucked off and then recrystallized twice from alk. The compound obtained is a colorless, well crystallizing, water-soluble mass with a pleasant taste, which is extremely easily absorbed by the organism v F = 1000 (uncorr.).
Example 3 330 g of bis [2-methoxyphenyl] phosphoric acid ester chloride are mixed with a solution of 300 g of triethylolamine in 500 cm3 of water. Immediately a reaction occurs, which is completed by heating the reaction mixture on the water bath for about one hour. After cooling, about 2500 cm3 of ethyl alcohol are added. Triethylolamine hydrochloride separates out as a by-product, which is filtered off. After evaporation of the alcoholic liquid, the filtrate leaves a residue which, when recrystallized from alcohol, yields the triethylolamine salt formed; F = 99-1000 (uncorrected).
