Abstract
Se revelan métodos para realizar una modificación planificada en un gen de fertilidad masculina en el genoma de una planta. Los métodos comprenden poner en contacto una célula de la planta con un agente de inducción de rotura bicatenaria diseñado capaz de inducir una rotura bicatenaria en una secuencia blanco en el gen de fertilidad masculina e identificar una célula que comprende una alteración en la secuencia blanco. Además se revelan plantas, células de plantas, partes de plantas, y semillas que comprenden un gen de fertilidad masculina con una alteración en el mismo. Asimismo se revelan moléculas de ácidos nucleicos que comprenden genes de la fertilidad masculina con por lo menos una modificación planificada. moléculas de ácidos nucleicos optimizadas que codifican endonucleasas que son agentes de inducción de rotura bicatenaria diseñados y cassettes de expresión, células huéspedes, y plantas que comprenden una o más de las moléculas de ácidos nucleicos.
