Abstract

Važan intermedijer za izradu cefalosporinskih antibiotika, 7-aminodesacetoksi cefalosporanske kiseline (7-ADCK), izradjuje se novim biopostupkom u kome se transformisani soj Penicillium chrysogenum-a gaji u prisustvu adipatne sirovine da se proizvede adipoil-6-APK (6-amino penicilanske kiseline); i ekspresija in situ gena ekspandaze, na pr. iz Streptomyces clavuligerus-a, pomoću koga je P. chrysogenum bio transformisan, preobraća adipoil-6-APK proširenjem prstena u adipoil-7-ADCK. Krajnji proizvod, 7-ADCK, se potom izradjuje ocepljivanjem adipoil bočnog lanca koristeći adipoil acilazu. Sledstveno tome, celokupna sinteza se izvodi upotrebljavajući biopostupak, i delotvorna je i ekonomična.An important intermediate for the manufacture of cephalosporin antibiotics, 7-aminodesacetoxy cephalosporic acid (7-ADCK), is made by a new bio-process in which a transformed Penicillium chrysogenum strain is grown in the presence of an adipate feed to produce adipoyl-6-APIC (acid-penic acid). ); and in situ expression of the expansion gene, e.g. from Streptomyces clavuligerus, by which P. chrysogenum was transformed, converts adipoyl-6-APK by extending the ring into adipoyl-7-ADCK. The final product, 7-ADCK, is then made by cleavage of the adipoyl side chain using adipoyl acylase. Consequently, the entire synthesis is performed using a bioprocess, and is effective and economical.
