Abstract

Pronalazak obezbedjuje postupke za identifikovanje bioloski aktivnih biomolekula. U jednom vidu pronalaska, jedan bioloski aktivan biomolekul kao sto je RNK ili neki peptid identifikovan je ugradnjom nasumice modifikovanih nukleotidnih sekvenci u jednu resetku koju sacinjava jedan modulator delovanja enzima, transformisanjem u sustini identicnih celija domacina napred dobijenim strukturama i pretrazivanjem tansformisanih celija da bi se identifikovale one kod kojih je promenjeno jedno unapred izabrano fenotipsko svojstvo. Potom se nasumice modifikovana DNK izdvaja iz fenotipski promenjenih celija i odredjuje se peptid i/ili RNK kodiran nasumice modifikovanom sekvencom. Sa svoje strane, partneri za medjusobno delovanje koji su moguci ciljni lekovi identifikovani su i izdvojeni koriscenjem peptida i/ili RNK kao dela afinitetnih reagensa. Jedna preporucljiva resetka je izvedena od krompirovog inhibitora I familije proteinskih inhibitora a kao primer prikazan je jecmeni inhibitor 2A (Cl-2A) himotripsina. Drugi vid pronalaska odnosi se na identifikaciju novih enzimskih inhibitora koristeci u sustini isti pristup, ali uz specificno pretrazivanje promena delovanja ciljnih enzima. Takodje su prikazani postupci izvodjenja relevantnih vektora transformacija i izrazavanja kao i postupci za identifikovanje vodecih jedinjenja i ciljnih lekova za primenu u razvoju lekova. Konacno, u opseg pronalaska takodje spada i postupak za pripremanje jednog lekovitog proizvoda.
