Description
本発明は、PGAM-Chk1結合阻害剤を含む肺疾患、肝疾患又は腎疾患の治療剤に関する。
高齢化が進む現代社会においては、高齢者全般の健康増進への取り組みと共に、多彩な加齢性疾患、特に生命活動に重要な肺、肝臓、腎臓等における加齢性疾患への斬新なアプローチによる医薬品開発が喫緊の課題となりつつある。
例えば、肺線維症は、肺胞壁におけるびまん性線維増殖を特徴とし、乾性咳嗽や労作時呼吸困難を主症状とする疾患であり、狭義には間質性肺炎の終末病態である特発性肺線維症を指すが、広義には肺の線維化と間質性肺炎との併存状態を意味するとされている。そして、あらゆる間質性肺炎が肺線維症の原因となり得る。
間質性肺炎は、肺の間質を中心に炎症を来す疾患の総称であり、感染、膠原病、放射線、薬剤、粉塵など特定の原因によるものと、原因が不明の特発性間質性肺炎とを含む。特発性間質性肺炎には、特発性肺線維症、非特異性間質性肺炎、特発性器質化肺炎、呼吸細気管支炎を伴う間質性肺疾患、剥離性間質性肺炎、急性間質性肺炎、及びリンパ性間質性肺炎の疾患が知られており、特発性肺線維症が最も発生頻度が高く、単に肺線維症とも呼ばれる。
特発性肺線維症では、肺間質にびまん性に線維性結合組織が過剰形成をおこし、肺の機能が障害され、診断確定後の平均生存期間が2.5~5年間と報告されている(非特許文献1)。特発性肺線維症の患者の3~15%でテロメア異常があることが判明したことから特発性肺線維症は代表的な加齢性疾患の一つとも考えられている(非特許文献2)。原因が特定できる間質性肺炎は、その原因の除去や、ステロイド剤などの抗炎症剤の投与などにより治癒する場合が多い。一方、肺線維症や線維化を伴う間質性肺炎の治療には、一般的にステロイド剤や免疫抑制剤が用いられているが、予後を改善するような効果的な治療法は無いのが現状であり、新たな治療薬の開発が強く望まれている。
一方、解糖系代謝とは、細胞が取り込んだグルコースを分解することにより、ATPという高エネルギー分子を合成する代謝過程の総称である。解糖系代謝は、ほとんどの通常細胞においてエネルギー代謝の重要な役割を担っており、個体が生きていく上で不可欠である。そのため、解糖系代謝の異常はヒトの疾患に密接に関連する。解糖系代謝が低下すると、神経変性疾患、心機能低下、糖尿病、筋肉萎縮、貧血等の原因となる一方、解糖系代謝が亢進する病態もいくつか知られている。解糖系代謝が亢進する病態としては、例えば癌、虚血(血流低下)、局所炎症等が挙げられる。
解糖系代謝経路は10個の解糖系酵素の順次反応よりなる。解糖系酵素ホスホグリセリン酸ムターゼPGAMはその8番目の酵素として、3-Phosphoglycerate(3PG)から2-Phosphoglycerate(2PG)への変換反応を触媒する。本発明者らは、細胞内でPGAMがセリンスレオニンタンパク質キナーゼであるChk1と結合すること、そしてこの物理的な結合が解糖系代謝において重要な役割を果たすこと、また、この物理的結合を制御することで解糖系を制御することができることを見出した(特許文献1)。しかし、解糖系代謝の制御と線維症等の疾患との関連については何ら知られていない。

上述のような状況下、本発明は、肺疾患、肝疾患又は腎疾患、特に肺線維症、腎硬化症に対して優れた治療効果を奏する新規治療薬を提供することを課題とする。
本発明者らは、上記課題を解決するために鋭意研究した結果、解糖系代謝酵素であるPGAMと、セリンスレオニンタンパク質キナーゼであるChk1との物理的結合の阻害剤が、肺疾患、肝疾患及び腎疾患に対する治療効果を奏することを見出した。中でも、肺疾患のうちの肺線維症に対する治療効果に優れていた。即ち、本発明は、以下に示すとおりである。
[1]PGAMとChk1との結合阻害剤を有効成分として含有する、肺疾患、肝疾患又は腎疾患の治療剤。

本発明の治療剤は、PGAMとChk1との結合阻害剤を有効成分として含有することで、肺疾患、肝疾患又は腎疾患、特に肺疾患、なかでも肺線維症に対して優れた治療効果を奏する。本発明の治療剤は、これまで有効な治療剤が存在しなかった特発性肺線維症(IPF)等に対しても優れた治療効果が期待できる。
以下、本発明について詳細に説明する。
<肺疾患、肝疾患又は腎疾患治療剤>
本発明の治療剤は、PGAMとChk1との結合阻害剤を有効成分として含有することを特徴とする。

本発明においてPGAMとは、ホスホグリセリン酸ムターゼという、解糖系代謝経路における10個の解糖系酵素の順次反応のうち、その8番目の反応を担う酵素であり、3-Phosphoglycerate(3PG)から2-Phosphoglycerate(2PG)への変換反応を触媒する酵素のことをいう。PGAMは、細胞の老化と癌化の接点を担う。PGAMにはPGAM1とPGAM2の二つのアイソフォームが存在する。PGAM1とPGAM2は組織発現パターンに違いが見られるものの、アミノ酸配列上高い相同性を有しており、機能や酵素活性は同等であると考えられる。ヒトPGAM1、PGAM2のアミノ酸配列はそれぞれ配列番号1、配列番号2として、cDNA配列はそれぞれ配列番号3、配列番号4として配列表に示した。
本発明においてChk1とは、セリンスレオニンタンパク質キナーゼであり、細胞周期のG2期でDNA損傷チェックポイントの活性化において役割を担うチェックポイントキナーゼである。Chk1は、本発明者らによって解糖系代謝においても重要な役割を担っていることが見出された。ヒトChk1のアミノ酸配列は配列番号5として、cDNA配列は配列番号6として配列表に示した。
本発明における解糖系代謝とは、細胞が取り込んだグルコースを分解することにより、ATPを合成する代謝過程の総称である。解糖系代謝には、グルコースをその中間産物であるピルビン酸に分解し、乳酸を合成する経路と、ミトコンドリアでの酸化的リン酸化へと進む経路の、二通りの経路が存在する。前者は、酸素呼吸を必要としないので嫌気的解糖系代謝と呼ばれ、後者はミトコンドリア酸素呼吸によりATPを合成するので好気的解糖系代謝と呼ばれる。この解糖系代謝は、ほとんどの通常細胞においてエネルギー代謝の重要な役割を担っており、個体が生きていく上で不可欠である。
本発明におけるPGAM及びChk1には、上記の公知のPGAM及びChk1と機能的に同等なタンパク質も含まれる。このようなタンパク質には、例えば、PGAM及びChk1の変異体、アレル、バリアント、ホモローグ、PGAM及びChk1の部分ペプチド、他のタンパク質との融合タンパク質、タグによって標識されたものなどが挙げられるが、これらに限定されることはない。
本発明におけるPGAM及びChk1の変異体としては、上述のアミノ酸配列において1もしくは複数のアミノ酸が置換、欠失、挿入、及び/又は付加したアミノ酸配列からなる天然由来のタンパク質であって、既述のアミノ酸配列からなるタンパク質と機能的に同等なタンパク質を挙げることができる。また、上述の塩基配列からなるDNAとストリンジェントな条件下でハイブリダイズする天然由来のDNAよりコードされるタンパク質であって、既述のアミノ酸配列からなるタンパク質と機能的に同等なタンパク質も、PGAM及びChk1のそれぞれの変異体として挙げることができる。
本発明において、変異するアミノ酸数は特に制限されないが、通常、30アミノ酸以内であり、好ましくは15アミノ酸以内であり、さらに好ましくは5アミノ酸以内(例えば、3アミノ酸以内)であると考えられる。変異するアミノ酸残基においては、アミノ酸側鎖の性質が保存されている別のアミノ酸に変異されることが望ましい。例えば、アミノ酸側鎖の性質としては、疎水性アミノ酸(A,I,L,M,F,P,W,Y,V)、親水性アミノ酸(R,D,N,C,E,Q,G,H,K,S,T)、脂肪族側鎖を有するアミノ酸(G,A,V,L,I,P)、水酸基含有側鎖を有するアミノ酸(S,T,Y)、硫黄原子含有側鎖を有するアミノ酸(C,M)、カルボン酸及びアミド含有側鎖を有するアミノ酸(D,N,E,Q)、塩基含有側鎖を有するアミノ酸(R,K,H)、芳香族含有側鎖を有するアミノ酸(H,F,Y,W)を挙げることができる(括弧内はいずれもアミノ酸の一文字標記を表す)。あるアミノ酸配列に対する1又は複数個のアミノ酸残基の欠失、付加及び/又は他のアミノ酸配列による置換により修飾されたアミノ酸配列を有するポリペプチドがその生物学的活性を維持することは既に知られている。
本発明において機能的に同等とは、対象となるタンパク質が、PGAM又はChk1と同等の生物学的機能や生化学的機能を有することを指す。PGAMの生物学的機能や生化学的機能としては、解糖系酵素としての機能であり、具体的には、3-Phosphoglycerate(3PG)から2-Phosphoglycerate(2PG)への変換反応を触媒する機能を挙げることができる。また、Chk1の生物学的機能や生化学的機能としては、セリンスレオニンタンパク質キナーゼとしての機能を挙げることができる。
目的のタンパク質と機能的に同等なタンパク質をコードするDNAを調製するために、当業者に良く知られた方法としては、ハイブリダイゼーション技術やポリメラーゼ連鎖反応(PCR)技術を利用する方法が挙げられる。即ち、当業者にとっては、PGAM又はChk1の塩基配列もしくはその一部をプローブとして、またPGAM又はChk1に特異的にハイブリダイズするオリゴヌクレオチドをプライマーとして、PGAM又はChk1と高い相同性を有するDNAを単離することは通常行いうることである。このようにハイブリダイズ技術やPCR技術により単離しうるPGAM又はChk1と同等の機能を有するタンパク質をコードするDNAもまた本発明のDNAに含まれる。
本発明におけるPGAM及びChk1の状態としては、特に制限はなく、例えば精製された状態、細胞内に発現した状態、細胞抽出液内に発現した状態等であってもよい。
本発明におけるPGAM及びChk1の由来となる生物種としては、特に限定されるものではないが、例えば、ヒト、サル、イヌ、ネコ、マウス、ラット、モルモット、ブタ、ウシ、酵母、昆虫等が挙げられ、中でも治療剤の対象となり得るヒト、イヌ、ネコが好ましく、ヒトがより好ましい。
本発明の治療剤が有効成分として含有するPGAMとChk1との結合阻害剤は、PGAMとChk1との結合を阻害する化合物であればよく、天然化合物、有機化合物、無機化合物、核酸、タンパク質(抗体を含む)、ペプチド等の単一物質;化合物ライブラリー、核酸ライブラリー、ペプチドライブラリー、遺伝子ライブラリーの発現産物;細胞抽出物、細胞培養上清、発酵微生物産生物、海洋生物抽出物、植物抽出物、原核細胞抽出物、真核単細胞抽出物又は動物細胞抽出物の抽出物等のいずれであってもよい。
本発明において特定の物質がPGAMとChk1との結合を阻害するか否かを判定する方法は特に限定されず、PGAM及びChk1の状態に応じて行うことができる。例えば、PGAM及びChk1が精製された状態であれば、精製標品に被験物質を添加することにより行うことができる。また、細胞内に発現した状態又は細胞抽出液内に発現した状態であれば、それぞれ、細胞の培養液若しくは該細胞抽出液に被験物質を添加することにより、又は実験動物に直接投与することにより行うことができる。被験物質がタンパク質の場合には、例えば、該タンパク質をコードするDNAを含むベクターをPGAM及びChk1が発現している細胞へ導入する、又は該ベクターをPGAM及びChk1が発現している細胞抽出液に添加することで行うことも可能である。また、例えば、酵母又は動物細胞等を用いた2ハイブリッド法を利用して判定することも可能である。
本発明において特定の物質がPGAMとChk1との結合を阻害するか否かを判定する際には、PGAM及びChk1を含む試料に被験物質を加えることにより、PGAM及び/又はChk1と被験物質とを接触させてもよい。また、PGAM又はChk1のどちらか一方を含む試料に被験物質を加えた後、含んでいなかったもう一方を加えてそれぞれと被験物質とを接触させてもよい。
本発明において特定の物質がPGAMとChk1との結合を阻害するか否かを判定する際には、上記接触の後、PGAMとChk1との結合量を測定する。なお、PGAM及びChk1を含む試料に被験物質を加えなかった場合のPGAMとChk1との結合量を測定してコントロールとする。具体的には、被験物質を加えた場合(接触させた場合)と、被験物質を加えなかった場合(接触させなかった場合)のPGAMとChk1との結合量を測定し、それらの値を比較する。被験物質を加えた場合に、被験物質を加えなかった場合と比較して、PGAMとChk1との結合量が減少していれば、この被験物質はPGAMとChk1との結合を阻害する効果がある(PGAMとChk1との結合阻害活性がある)と判断できる。
PGAMとChk1との結合量を測定する方法としては、例えば、具体的に以下のような方法が挙げられる。PGAM2-FLAG及びChk1-myc-His発現ベクターを共発現させた細胞の蛋白質抽出液から、プロテインGアガロースにコンジュゲートされたFLAGタグ抗体により、PGAM2-FLAG蛋白を免疫沈降させる。この時PGAM2-FLAG蛋白に結合して共沈降してくるChk1-myc-His蛋白をウェスタンブロットにより検出し、定量する。あるいは、同様の蛋白質抽出液からNi-NTAアガローズビーズにより、Chk1-myc-His蛋白を沈降させる。この時Chk1-myc-His蛋白に結合して共沈降してくるPGAM2-FLAG蛋白をウェスタンブロットにより検出し、定量する。
PGAMとChk1との結合量を測定する方法としては、上記以外にも例えば、互いに結合し得るタグで標識されたPGAM及びChk1を用いる方法も好ましい方法として挙げられ、具体的には、Nanobitのシステムを利用した以下のような方法が挙げられる。NanobitではPGAMとChk1にそれぞれルシフェラーゼをベースに作成された発光蛋白質の大サブユニットと小サブユニットをそれぞれタグとして取り付ける。これらのタグは互いに結合し得る構造となっている。それぞれのサブユニットはそれ自体発光しないが、PGAMとChk1の結合により大サブユニットと小サブユニットが会合し完全な発光蛋白となることで発光シグナルを発する。このときの発光シグナルの強度を測定することで高感度かつ高い定量性をもって結合量を測定することができる。
本発明の治療剤が有効成分として含有するPGAMとChk1との結合阻害剤は、さらにセノリシス活性を有する化合物であることが好ましい。肺疾患、肝疾患、腎疾患のうち、加齢性疾患、特に肺線維症には老化細胞が関与すること、老化細胞を除去することで肺線維症の症状が改善するという報告もあることから、PGAMとChk1との結合阻害活性に加えてセノリシス活性を有する化合物は肺線維症等の加齢性疾患の治療効果に優れると考えられる。
ここで、セノリシス(Senolysis;老化細胞除去)とは、慢性炎症の原因である老化細胞を除去すれば、個体老化が改善できるという発想に基づき提案されている考え方である。本発明においてセノリシス活性とは、老化細胞を選択的に除去できる能力の程度をいう。
化合物についての上記セノリシス活性の確認は以下の方法により行うことができる。被験物質の、若い細胞に対する傷害活性と、老化細胞に対する傷害活性を測定し、若い細胞に対する傷害活性を有さず、老化細胞に対する傷害活性を有する被験物質はセノリシス活性を有すると確認できる。また、被験物質の、若い細胞に対する傷害活性と、老化細胞に対する傷害活性を測定し、老化細胞に対する傷害活性が、若い細胞に対する傷害活性と比較して顕著に高い被験物質である場合、被験物質は優れたセノリシス活性を有すると確認できる。
ここで、本発明において、若い細胞として、マウス培養細胞では、マウス初代線維芽細胞(MEFs;mouse embryonic fibroblasts、あるいはREFs;rat embryonic fibroblasts)、マウス初代皮膚ケラチノサイト等が挙げられる。また、本発明において、若い細胞として、ヒト培養細胞では、ヒト胎児肺線維芽細胞(IMR90やWI-38、TIG、MRC細胞)、ヒト皮膚初代細胞(NHDF細胞、BJ細胞、NHEK細胞)、正常ヒト乳腺上皮細胞(NHMEC細胞)、正常ヒト血管内皮細胞(HUVEC細胞、HAEC細胞、HPAEC細胞、HCAEC細胞)、正常ヒト前立腺上皮細胞(HPEC)等が挙げられる。なお、細胞種により、その増殖スピードに違いがあり、老化速度も異なるものであるが、おおむねマウス細胞は、最初の樹立から継続培養1か月ほどで老化細胞となり、ヒト細胞は3か月ほどで老化細胞となることが知られている。よって、マウス細胞では、最初の樹立から継続培養の2-3週間以内を若い細胞といい、ヒト細胞では、最初の樹立から継続培養の1か月以内を若い細胞ということができる。一方、本発明において、老化細胞としては、上記の継続培養期間を超えて培養したもの、マウスなら3~4週以上、ヒトなら1~2か月以上培養したものが挙げられる。さらに、マウスやヒトの若い細胞でも、細胞老化を誘導するストレス(酸化ストレスや、発癌ストレス、DNA傷害ストレスなど)により、短期間で老化細胞へと移行させることもできる。
本発明の治療剤は、PGAMとChk1との結合阻害活性及びセノリシス活性を有する化合物を有効成分として含有することが好ましく、このような化合物としては、RSK1キナーゼ阻害剤、Fakキナーゼ阻害剤、Fakキナーゼが関与するシグナル経路の阻害剤、CDK阻害剤、カルシウム拮抗剤、強心配糖体、DNA障害剤、抗菌剤、オーロラキナーゼ阻害剤、フラボノイド、PI3Kキナーゼ阻害剤、HDAC阻害剤、加齢黄斑変性治療薬、p38MAPK阻害剤、mTOR阻害剤、チロシンキナーゼ阻害剤、Bcl-2阻害剤、HIF-2α阻害剤、スタチン、セロトニン受容体拮抗薬、その他キナーゼ阻害剤、ナトリン3b等が好ましい化合物として挙げられる。
(RSK1キナーゼ阻害剤)
RSK1はRSK(リボソームS6キナーゼ)ファミリーメンバーであり、増殖因子制御型セリンスレオニンキナーゼである。RSK1は2つの異なるキナーゼ触媒ドメインをもち、***促進因子活性化キナーゼ(MAPK)シグナル伝達経路メンバーを含むさまざまな基質をリン酸化する。本発明の治療剤が含有するRSK1キナーゼの阻害剤としては、上記RSK1キナーゼ活性を阻害する物質であれば特に限定されないが、例えば、RSK1をコードするDNAの転写産物と相補的なアンチセンス核酸や、siRNA、BI-D1870(CAS番号:501437-28-1)等の化合物が好ましい例として挙げられる。
RSK1はRSK(リボソームS6キナーゼ)ファミリーメンバーであり、増殖因子制御型セリンスレオニンキナーゼである。RSK1は2つの異なるキナーゼ触媒ドメインをもち、***促進因子活性化キナーゼ(MAPK)シグナル伝達経路メンバーを含むさまざまな基質をリン酸化する。本発明の治療剤が含有するRSK1キナーゼの阻害剤としては、上記RSK1キナーゼ活性を阻害する物質であれば特に限定されないが、例えば、RSK1をコードするDNAの転写産物と相補的なアンチセンス核酸や、siRNA、BI-D1870(CAS番号:501437-28-1)等の化合物が好ましい例として挙げられる。
本発明で用いられるアンチセンス核酸は、転写、スプライシング又は翻訳の過程を阻害して、RSK1遺伝子の発現を抑制するアンチセンス核酸であることが好ましい。アンチセンス配列の態様としては、RSK1のmRNAの5’端近傍の非翻訳領域に相補的なアンチセンス配列を設計すれば、遺伝子の翻訳阻害に効果的であるものと考えられる。しかし、コード領域もしくは3’側の非翻訳領域に相補的な配列も使用し得る。このように遺伝子の翻訳領域だけでなく非翻訳領域に相補的な配列も使用しうる。このように、遺伝子の翻訳領域だけでなく、非翻訳領域の配列のアンチセンス配列を含む核酸も、本発明で利用されるアンチセンス核酸に含まれる。アンチセンス核酸は標的とする遺伝子の転写産物に対して好ましくは90%以上、最も好ましくは95%以上の相補性を有する。アンチセンス配列を用いて、効果的に標的遺伝子の発現を阻害するアンチセンスRNAの長さは、特に制限されない。
本発明で用いられるRSK1のsiRNAは、細胞で毒性を示さない範囲の短鎖からなる二重鎖RNAを意味し、例えば15~49塩基対と、好適には15~35塩基対と、さらに好適には21~30塩基対とすることが出来る。
RSK1のsiRNAは、RSK1遺伝子と完全に同一である必要はないが、少なくとも70%以上、好ましくは80%以上、さらに好ましくは90%以上、最も好ましくは95%以上の配列の相同性を有する。siRNAにおけるRNA同士が対合した二重鎖RNAの部分は、完全に対合しているものに限らず、ミスマッチ(対応する塩基が相補的でない)、バルジ(一方の鎖に対応する塩基がない)などにより不対合部分が含まれていてもよい。本発明においてはdsRNAにおけるRNA同士が対合する二重鎖RNA領域中に、バルジ及びミスマッチの両方が含まれていてもよい。
本発明において、RSK1のsiRNAとしては、例えば配列番号7又は配列番号8の配列を含むものが挙げられる。なお、本発明におけるsiRNAには、ヌクレアーゼ分解耐性向上等を目的として、3’末端にdTdT等のオーバーハング2塩基を付加してもよい。
(Fakキナーゼ阻害剤)
Fak(接着斑キナーゼ)は非受容体型タンパク質チロシンキナーゼであり、細胞外基質との細胞接着を媒介するインテグリン濃縮接着点からのシグナル伝達に関与する。FAKにより促進されたシグナルは、付着依存性細胞の生存に関与し、増殖因子受容体やインテグリン刺激に応答した効率的な細胞遊走に非常に重要であることが知られている。本発明の治療剤が含有するFakキナーゼ阻害剤としては、上記Fakキナーゼ活性を阻害する物質であれば特に限定されないが、例えばPF-00562271(CAS No.939791-41-0)、PF-431396(CAS No.717906-29-1)やこれらの塩又は誘導体であってこれらと同等の活性を有する化合物等が好ましい例として挙げられる。
Fak(接着斑キナーゼ)は非受容体型タンパク質チロシンキナーゼであり、細胞外基質との細胞接着を媒介するインテグリン濃縮接着点からのシグナル伝達に関与する。FAKにより促進されたシグナルは、付着依存性細胞の生存に関与し、増殖因子受容体やインテグリン刺激に応答した効率的な細胞遊走に非常に重要であることが知られている。本発明の治療剤が含有するFakキナーゼ阻害剤としては、上記Fakキナーゼ活性を阻害する物質であれば特に限定されないが、例えばPF-00562271(CAS No.939791-41-0)、PF-431396(CAS No.717906-29-1)やこれらの塩又は誘導体であってこれらと同等の活性を有する化合物等が好ましい例として挙げられる。
(Fakキナーゼが関与するシグナル経路の阻害剤)
本発明の治療剤が含有する、Fakキナーゼが関与するシグナル経路の阻害剤としては、c-Met阻害剤、Mdr-1阻害剤等が挙げられる。本発明の治療剤が含有するc-Met阻害剤としては、PHA-665752(CAS No.477575-56-7)や、これの塩又は誘導体であって同等の活性を有する化合物等が好ましい例として挙げられる。また、本発明の治療剤が含有するMdr-1阻害剤としては、CP-100356(CAS No.142715-48-8)やこれの塩又は誘導体であってこれと同等の活性を有する化合物等が好ましい例として挙げられる。

(CDK阻害剤)
サイクリン依存性キナーゼ(cyclin-dependent kinase:CDK)は、細胞周期を司る核内キナーゼ群でサイクリンと結合してはじめて活性をもつタンパク質である。細胞周期の異なった時点で活性化される複数のCDKが存在する。活性は、CDK自身のチロシン、スレオニン残基のリン酸化と脱リン酸化で制御されている。本発明の治療剤が含有するCDK阻害剤は、このようなCDKの活性を阻害、抑制する物質であり、例えば、SNS-032(CDK2/7/9阻害剤;CAS No.345627-80-7)、AZD5438(CDK1/2/9阻害剤;CAS No.602306-29-6)、Flavopiridol(Alvocidib)(CDK1/2/4/6/9阻害剤;CAS No.146426-40-6)、PHA-793887(CDK7/9阻害剤;CAS No.718630-59-2)、AT7519(CDK1/2/4/6/9阻害剤;CAS No.844442-38-2)、PHA-767491(CDK7/9阻害剤; CAS No.942425-68-5)やこれらの塩又は誘導体であってこれらと同等の活性を有する化合物等が好ましい例として挙げられる。
サイクリン依存性キナーゼ(cyclin-dependent kinase:CDK)は、細胞周期を司る核内キナーゼ群でサイクリンと結合してはじめて活性をもつタンパク質である。細胞周期の異なった時点で活性化される複数のCDKが存在する。活性は、CDK自身のチロシン、スレオニン残基のリン酸化と脱リン酸化で制御されている。本発明の治療剤が含有するCDK阻害剤は、このようなCDKの活性を阻害、抑制する物質であり、例えば、SNS-032(CDK2/7/9阻害剤;CAS No.345627-80-7)、AZD5438(CDK1/2/9阻害剤;CAS No.602306-29-6)、Flavopiridol(Alvocidib)(CDK1/2/4/6/9阻害剤;CAS No.146426-40-6)、PHA-793887(CDK7/9阻害剤;CAS No.718630-59-2)、AT7519(CDK1/2/4/6/9阻害剤;CAS No.844442-38-2)、PHA-767491(CDK7/9阻害剤; CAS No.942425-68-5)やこれらの塩又は誘導体であってこれらと同等の活性を有する化合物等が好ましい例として挙げられる。
(カルシウム拮抗剤)
カルシウム拮抗剤は、細胞膜上のカルシウムチャネルに結合し、細胞内へのカルシウムイオン流入を阻害する薬剤である。本発明の治療剤が含有するカルシウム拮抗剤としては、例えば、Manidipine(CAS No.89226-50-6)、Lomerizine 2HCl(CAS No.101477-54-7)やこれらの塩又は誘導体であってこれらと同等の活性を有する化合物等が好ましい例として挙げられる。

(強心配糖体)
強心配糖体とは、心房細動、心房粗動等の上室性頻脈や浮腫を伴う鬱血性心不全あるいは不整脈に用いられるステロイド配糖体の総称であるが、本発明の治療剤が含有する強心配糖体としては、例えば、Proscillaridin A (CAS No.466-06-8)、Digoxin(CAS No.20830-75-5)やこれらの塩又は誘導体であってこれらと同等の活性を有する化合物等が好ましい例として挙げられる。
強心配糖体とは、心房細動、心房粗動等の上室性頻脈や浮腫を伴う鬱血性心不全あるいは不整脈に用いられるステロイド配糖体の総称であるが、本発明の治療剤が含有する強心配糖体としては、例えば、Proscillaridin A (CAS No.466-06-8)、Digoxin(CAS No.20830-75-5)やこれらの塩又は誘導体であってこれらと同等の活性を有する化合物等が好ましい例として挙げられる。
(DNA傷害型抗癌剤)
DNA傷害型抗癌剤とは、癌細胞のDNAに傷害を起こすことで癌細胞を選択的に細胞死に導くタイプの抗癌剤の創傷であるが、本発明の治療剤が含有するDNA傷害型抗癌剤としては、例えば、Doxorubicin(Adriamycin)HCl(CAS No.25316-40-9)、Daunorubicin HCl(CAS No.23541-50-6)、Epirubicin HCl(CAS No.56390-09-1)、Mitoxantrone 2HCl(CAS No.70476-82-3)、Hydroxy Camptothecine(CAS No.64439-81-2)やこれらの塩又は誘導体であってこれらと同等の活性を有する化合物等が好ましい例として挙げられる。
DNA傷害型抗癌剤とは、癌細胞のDNAに傷害を起こすことで癌細胞を選択的に細胞死に導くタイプの抗癌剤の創傷であるが、本発明の治療剤が含有するDNA傷害型抗癌剤としては、例えば、Doxorubicin(Adriamycin)HCl(CAS No.25316-40-9)、Daunorubicin HCl(CAS No.23541-50-6)、Epirubicin HCl(CAS No.56390-09-1)、Mitoxantrone 2HCl(CAS No.70476-82-3)、Hydroxy Camptothecine(CAS No.64439-81-2)やこれらの塩又は誘導体であってこれらと同等の活性を有する化合物等が好ましい例として挙げられる。
(抗菌剤)
本発明の治療剤が含有する抗菌剤としては、例えば、Aminoacridine(CAS No.90-45-9)、Triclosan(CAS No.3380-34-5)、Ethacridine lactate(TAZ活性化;CAS No.1837-57-6)やこれらの塩又は誘導体であってこれらと同等の活性を有する化合物等が好ましい例として挙げられる。
本発明の治療剤が含有する抗菌剤としては、例えば、Aminoacridine(CAS No.90-45-9)、Triclosan(CAS No.3380-34-5)、Ethacridine lactate(TAZ活性化;CAS No.1837-57-6)やこれらの塩又は誘導体であってこれらと同等の活性を有する化合物等が好ましい例として挙げられる。
(オーロラキナーゼ阻害剤)
オーロラキナーゼは、ヒトではAURKA遺伝子にコードされる酵素であり、セリン/スレオニンキナーゼファミリーのメンバーである。正常な細胞増殖には、有糸***と減数***の過程においてオーロラキナーゼが適切に機能することが必要不可欠である。オーロラキナーゼは1つまたは複数箇所のリン酸化によって活性化され、その活性は細胞周期のG2期からM期への移行時にピークに達することが知られている。本発明の治療剤が含有するオーロラキナーゼ阻害剤としては、例えば、AMG-900(CAS No.945595-80-2)、JNJ-7706621(CDK1,2,Aurora A,B阻害;CAS No.443797-96-4)やこれらの塩又は誘導体であってこれらと同等の活性を有する化合物等が好ましい例として挙げられる。

(HIF-2α阻害剤)
HIF-2αは、転写因子HIF-1αの機能性ホモローグであり、進化上保存されている。HIF-ファミリータンパクは、細胞や組織が低酸素状況に暴露されたときに活性化される転写因子である。低酸素状況では、ミトコンドリア酸素呼吸ができないために、細胞はエネルギー産生を解糖系代謝に依存せざるを得ない。低酸素環境で、HIF蛋白の活性化により、解糖系代謝mRNAが亢進し、解糖系代謝亢進が可能となる(Iyer et al.,Genes and Development, 1998,Vol12,p149-62)。通常の酸素環境では、転写因子HIF蛋白は、ユビキチン化修飾を受けやすく、蛋白分解され不安定な蛋白である一方、低酸素状況では、HIFのユビキチン化は阻害され、蛋白が安定化し、活性を亢進させる。本発明の治療剤が含有するHIF-2α阻害剤としては、HIF-2αの機能を阻害する物質であれば特に限定されないが、例えば、HIF-2αをコードするDNAの転写産物と相補的なアンチセンス核酸や、siRNA(例えば、配列番号9、配列番号10)等が好ましい例として挙げられる。
HIF-2αは、転写因子HIF-1αの機能性ホモローグであり、進化上保存されている。HIF-ファミリータンパクは、細胞や組織が低酸素状況に暴露されたときに活性化される転写因子である。低酸素状況では、ミトコンドリア酸素呼吸ができないために、細胞はエネルギー産生を解糖系代謝に依存せざるを得ない。低酸素環境で、HIF蛋白の活性化により、解糖系代謝mRNAが亢進し、解糖系代謝亢進が可能となる(Iyer et al.,Genes and Development, 1998,Vol12,p149-62)。通常の酸素環境では、転写因子HIF蛋白は、ユビキチン化修飾を受けやすく、蛋白分解され不安定な蛋白である一方、低酸素状況では、HIFのユビキチン化は阻害され、蛋白が安定化し、活性を亢進させる。本発明の治療剤が含有するHIF-2α阻害剤としては、HIF-2αの機能を阻害する物質であれば特に限定されないが、例えば、HIF-2αをコードするDNAの転写産物と相補的なアンチセンス核酸や、siRNA(例えば、配列番号9、配列番号10)等が好ましい例として挙げられる。
(ナトリン3b)
ナトリン3b(Nutlin-3b;CAS No.675576-97-3)は、ナトリン-3a(Nutlin-3a)の光学異性体である。本発明の治療剤は、ナトリン3b(Nutlin3b;CAS No.675576-97-3)を有効成分として含有する物であることが好ましい。

上記以外にも、フラボノイド(Chrysin;CAS No.480-40-0)、PI3Kキナーゼ阻害剤(PF-05212384;CAS No.1197160-78-3、XL147;CAS No.934526-89-3)、HDAC阻害剤(Trichostatin A(TSA);CAS No.58880-19-6)、加齢黄斑変性治療薬(Verteporfin;CAS No.129497-78-5)、p38MAPK阻害剤(SB203580;CAS No.152121-47-6、Doramapimod(BIRB 796);CAS No.285983-48-4)、mTOR阻害剤(AZD8055;CAS No.1009298-09-2、KU-0063794;CAS No.938440-64-3)、チロシンキナーゼ阻害剤(Neratinib(HER2、EGFR阻害);CAS No.698387-09-6、Vargatef;CAS No.656247-17-5、NVP-BHG712(C-raf、C-src阻害);CAS No.940310-85-0)、Bcl-2阻害剤(TW-37;CAS No.877877-35-5)、スタチン(Mevastatin;CAS No.73573-88-3)、セロトニン受容体拮抗薬(RS-127445;CAS No.199864-87-4)、その他キナーゼ阻害剤(BMS-345541;CAS No.445430-58-0、CX-4945;CAS No.1009820-21-6)や、これらの塩又は誘導体は、本発明の治療剤の有効成分として採用することができる。
本発明の治療剤は、上記有効成分に加えて、他の任意成分として、製剤上、薬理学的又は生理学的に許容される一種又は二種以上の担体、賦形剤、結合剤、崩壊剤、滑沢剤、分散剤、界面活性化剤、可塑剤、懸濁剤、乳化剤、希釈剤、緩衝剤、抗酸化剤、細菌抑制剤等を含有していてもよい。また、その他低分子量のポリペプチド、血清アルブミン、ゼラチンや免疫グロブリン等の蛋白質、アミノ酸、多糖及び単糖等の糖類や炭水化物、糖アルコールを含んでいてもよい。さらに必要に応じて他の有効成分を含有していてもよい。
本発明の治療剤は、医薬品、医薬部外品、食品等の分野において従来公知の任意の方法により製造することができる。その製造過程において、上記有効成分、その他の任意成分を公知のいかなる方法で添加・混合してもよい。
注射用の水溶液とする場合には、例えば生理食塩水、ブドウ糖やその他の補助薬を含む等張液、例えば、D-ソルビトール、D-マンノース、D-マンニトール、塩化ナトリウムが挙げられ、適当な溶解補助剤、例えばアルコール(エタノール等)、ポリアルコール(プロピレングリコール、PEG等)、非イオン性界面活性剤(ポリソルベート80、HCO-50)等と併用してもよい。必要に応じてさらに希釈剤、溶解補助剤、pH調整剤、無痛化剤、含硫還元剤、酸化防止剤等を含有してもよい。
また、マイクロカプセル(ヒドロキシメチルセルロース、ゼラチン、ポリ[メチルメタクリル酸]等のマイクロカプセル)に封入したり、コロイドドラッグデリバリーシステム(リポソーム、アルブミンミクロスフェア、マイクロエマルジョン、ナノ粒子及びナノカプセル等)としたりすることもできる。さらに、薬剤を徐放性の薬剤とする方法も公知であり、本発明に適用し得る。
本発明の治療剤をヒトや他の動物の医薬として使用する場合には、これらの物質自体を直接患者に投与する以外に、公知の製剤学的方法により製剤化して投与を行うことも可能である。製剤化する場合には、上記に記載の製剤上許容される成分を添加してもよい。
本発明におけるすべての薬剤は、医薬品の形態で投与することが可能であり、経口的又は非経口的に全身あるいは局所的に投与することができる。例えば、点滴などの静脈内注射、筋肉内注射、腹腔内注射、皮下注射、坐薬、注腸、経口性腸溶剤などを選択することができ、患者の年齢、症状により適宜投与方法を選択することができる。有効投与量は、一回につき体重1kgあたり0.001mgから100mgの範囲で選ばれる。あるいは、患者あたり0.1~1000mg、好ましくは0.1~50mgの投与量を選ぶことができる。具体的な例としては、体重1kgあたり1ヶ月(4週間)に0.1mgから40mg、好ましくは1mgから20mgを1回から数回に分けて、例えば2回/週、1回/週、1回/2週、1回/4週などの投与スケジュールで点滴などの静脈内注射、皮下注射、経口投与等の方法で、投与する。投与スケジュールは、投与後の状態の観察及び血液検査値の動向を観察しながら2回/週あるいは1回/週から1回/2週、1回/3週、1回/4週のように投与間隔を延ばしていくなど調整することも可能である。
本発明の治療剤は、肺、肝臓、腎臓等において優れた機能改善効果を奏し、肺疾患、肝疾患、腎疾患の治療剤として好適に用いられ、肺疾患、腎疾患の治療剤としてより好適に用いられ、特に、肺線維症治療剤、腎硬化症治療剤としてさらに好適に用いられる。肺線維症としては、例えば、間質性肺炎、特発性肺線維症、肺移植後の慢性あるいは急性拒絶による肺線維症、コロナ後遺症後の肺線維症、非特異性間質性肺炎、特発性器質化肺炎、剥離性間質性肺炎、呼吸細気管支炎関連間質性肺炎、急性間質性肺炎、リンパ球性間質性肺炎、サルコイドーシス、慢性好酸球性肺炎、急性好酸球性肺炎、リンパ脈管筋腫症、肺胞蛋白症、ヘルマンスキー・パドラック症候群、肺ランゲルハンス細胞組織球症、鉄肺症、アミロイドーシス、肺胞微石症、過敏性肺炎、じん肺、感染性肺疾患、薬剤性肺炎、放射線肺炎等の、肺における線維症を伴う疾患が挙げられる。また、本発明の治療剤は、高齢者における肺疾患、肝疾患、腎疾患の治療剤として特に好適に用いられる。さらに、後述するとおり、PGAM‐Chk1結合阻害変異ノックインマウスは、野生型マウスと比較して寿命が長くなることから、本発明の治療剤は、寿命を延長する効果、老化を抑制する効果等を有し、寿命延長剤、抗老化剤、老化抑制剤、老化遅延剤としても有効であるといえる。
<PGAM‐Chk1結合阻害変異ノックインマウス>
本発明のPGAM‐Chk1結合阻害変異ノックインマウスは、Pgam1遺伝子の変異により、PGAMタンパクがCHK1タンパクに結合できなくなっているマウスである。このマウスにおいては、PGAMタンパクの、CHK1タンパクへの結合部位を構成するアミノ酸が別のアミノ酸に置換され、PGAMタンパクがCHK1タンパクに結合できなくなっている。上記のアミノ酸置換は、この置換によりPGAMタンパクがCHK1タンパクに結合できなくなるものであれば特に限定されないが、例えば、PGAMタンパクの68番目のトリプトファンがアラニンに置換されるものが好ましい例として挙げられる。具体的には、本発明のPGAM‐Chk1結合阻害変異ノックインマウスは、配列番号11で表されるアミノ酸配列からなるマウス型変異PGAMタンパクをコードする変異Pgam1遺伝子がノックインされているマウスである。

PGAMタンパクの、CHK1タンパクへの結合部位を構成するアミノ酸が別のアミノ酸に置換され、PGAMタンパクがCHK1タンパクに結合できなくなっているタンパクとしては、配列番号11で表されるアミノ酸配列において、1もしくは複数のアミノ酸が置換、欠失、挿入、及び/又は付加したアミノ酸配列からなるタンパク質であって、配列番号11で表されるアミノ酸配列からなるタンパク質と機能的に同等なタンパク質を挙げることができる。これらのタンパク質のアミノ酸配列は、上記の68番目のトリプトファンがアラニンに置換されており、かつ、PGAM解糖系酵素ホスホグリセリン酸ムターゼを有するものである必要がある。
本発明のPGAM‐Chk1結合阻害変異ノックインマウスには、配列番号11で表されるアミノ酸配列において、1もしくは複数のアミノ酸が置換、欠失、挿入、及び/又は付加したアミノ酸配列からなるタンパク質であって、配列番号11で表されるアミノ酸配列からなるタンパク質と機能的に同等なタンパク質であるマウス型変異PGAMタンパクをコードする遺伝子(変異Pgam1遺伝子)がノックインされているマウスも含まれる。
本発明のPGAM‐Chk1結合阻害変異ノックインマウスは、若年における健康状態には異常が見られず繁殖性も正常であるが、老年期になると野生型マウスとの違いが現れる。例えば、加齢と共に発現が上がることが知られている各臓器(肺、肝臓、腎臓等)における老化マーカー、炎症マーカー発現や、慢性炎症の症状等も抑えられ、老化の進行が抑えられたマウスといえる。また、野生型マウスと比較して寿命が長く、長寿の傾向がみられるマウスである。
本発明のPGAM‐Chk1結合阻害変異ノックインマウスは様々な加齢性疾患あるいはPGAM-Chk1結合が亢進する病態に対するPGAM-Chk1結合阻害の治療効果を検証するための実験系として好適に用いられる。
以下の実施例にて本発明を具体的に説明するが、本発明は実施例によって限定されるものではない。
1.C57BL/6J PGAM‐Chk1結合阻害変異ノックインマウスの作製
マウス受精卵のPgam1遺伝子の68番目のトリプトファンをCRISPA-Cas9システムをもちいてアラニンに置換した。2細胞期まで培養した受精卵を仮親(ICR8週齢)に移植し出産させた。出産後に耳介よりDNA抽出を行い、遺伝子型を決定した。

2.PGAM‐Chk1結合阻害変異ノックインマウスの特徴
上述の方法で作製したPGAM‐Chk1結合阻害変異ノックインマウスは、若年における健康状態には異常は見られず、繁殖性も正常であった。通常の飼育環境下では老年期にのみ野生型マウスと違いが発現した。

C57BL/6J、90週齢の野生型(+/+)と上記ノックインマウス(KI/KI)について、肺組織における炎症・老化マーカー発現を比較した。老化マーカーとしては、p16lnk4のmRNA、炎症マーカーとしては、IL1α及びIL6のmRNAの相対発現量を測定した。図1(老化マーカー)、図2(炎症マーカー)に示す通り、ノックインマウスでは、肺における老化マーカー、炎症マーカーの発現が低下していた。また、肺のHE染色像を図3に示す。野生型(control)老齢マウスでは胸膜側から免疫細胞の浸潤がみられ慢性炎症が起こっているのに対して、結合変異ノックインマウス(KI群)ではそのような慢性炎症の所見は見られなかった。
C57BL/6J、90週齢の野生型(+/+)と上記ノックインマウス(KI/KI)について、肝臓における炎症・老化マーカー発現を比較した。老化マーカーとしては、p16lnk4のmRNA、炎症マーカーとしては、IL6のmRNAの相対発現量を測定した。図4に示す通り、ノックインマウスでは、肝臓における老化マーカー、炎症マーカーの発現が低下していた。
90週齢のC57BL/6J野生型(+/+)と上記ノックインマウス(KI/KI)について、腎臓における炎症・老化マーカー発現、腎機能マーカー発現を比較した。老化マーカーとしては、p16lnk4のmRNA、炎症マーカーとしては、IL6のmRNAの相対発現量を測定した。図5に示す通り、ノックインマウスでは、腎臓における老化マーカー、炎症マーカーの発現が低下していた。またノックインマウスでは、腎機能マーカーの改善が見られた(図6)。
3.ブレオマイシン(BLM)による肺線維化モデル
20週齢のPGAM‐Chk1結合阻害変異ノックインマウス(KI/KI)及び野生型マウス(WT)に対して、2.5mg/kgのブレオマイシンを気管内投与した。マウスは毎日生存確認を行い、ブレオマイシン投与から14日後にマウス体重を測定し(図7)、その後サクリファイスした。肺組織をサンプリングし重量を測定した(図8)。その後、肺組織の一部を十分量のRNAlater(Invitrogen社製)に浸し、4℃で一晩保存した。翌日、RNAlaterより肺組織を取り出し、TRIzole(Invitrogen社製)を用いてTotalRNAを回収した。その後、ReverTra Ace qPCR RT kit(TOYOBO社製)によりmRNAを逆転写しcDNAを調製した。調製したcDNAに対し線維化関連遺伝子(Col1a2、Col5a2、Fibronectin、Toropelastin)に特異的なプライマーを用いてリアルタイムPCRを実施した。結果を図9に示す。また、一部の肺組織は4%パラホルムアルデヒドで48時間固定処理された。固定後の肺組織はパラフィンに埋包された後、5μmの薄切り切片にされ、スライドガラスにセットされた。この肺組織の薄切切片に対して、マッソントリクロム染色を行い、線維化を評価した(図10)。

図7に示すとおり、野生型マウス(WT)は、ブレオマイシンの気管内投与によって体重が減少したが、PGAM‐Chk1結合阻害変異ノックインマウス(KI/KI)では、体重の減少は起こらなかった。一方、肺重量については、野生型マウス(WT)は、ブレオマイシンの気管内投与によって増加し、肺線維化が起こっていることが示唆された。一方、PGAM‐Chk1結合阻害変異ノックインマウス(KI/KI)では、変化がなく、ブレオマイシンの投与により引き起こされる肺線維化から回復していることが示唆された(図8)。また、図9に示すとおり、野生型マウス(WT)は、ブレオマイシンの気管内投与によって肺における線維化関連遺伝子発現が顕著に増加したのに対して、PGAM‐Chk1結合阻害変異ノックインマウス(KI/KI)では、その増加の度合いが小さかった(図9)。肺組織切片のマッソントリクロム染色の結果からも、PGAM‐Chk1結合阻害変異ノックインマウス(KI/KI)では野生型マウス(WT)と比較して肺線維化が回復していることがわかる。
4.PGAM-Chk1結合阻害物質の探索
Cell-base NanoBiTシステムを用いたPGAM-Chk1結合阻害HTP(ハイスループット)スクリーニングを実施した。具体的には以下の方法により行った。NanoBiTではPGAMとChk1にそれぞれルシフェラーゼをベースに作成された発光蛋白質の大サブユニットと小サブユニットをそれぞれタグとして取り付ける。これらのタグは互いに結合し得る構造となっている。それぞれのサブユニットはそれ自体発光しないが、PGAMとChk1の結合により大サブユニットと小サブユニットが会合し完全な発光蛋白となることで発光シグナルを発する。このときの発光シグナルの強度を測定することで高感度かつ高い定量性をもって結合量を測定することができる。このときナトリンのようなPGAMとChk1の結合を阻害する物質の存在下では発光シグナルが低下する。よってこの発光シグナルの減少を指標として、薬剤ライブラリーの中からPGAMとChk1との結合を阻害する物質の探索を行った。

このPGAM-Chk1結合可視化Cell-base NanoBiTシステムを用いて、京都大学医学研究支援センターのサポートを受け、安全性評価が確定している既存薬ライブラリー(2300余)を対象にPGAM-Chk1結合阻害HTPスクリーニング(1次スクリーニング)を行い、約200強の結合阻害候補薬を獲得した(図11、図12)。
これらの候補薬について、若いIMR90細胞と老化IMR90細胞の薬剤感受性を比較し、老化IMR90細胞に対する細胞傷害活性が高く、若いIMR90細胞に対する細胞傷害活性が低い物質をセノリシス活性の高い物質として次のステップに進めることとした(2次スクリーニング)。老化IMR90細胞は発癌性Ras変異を誘導することで作成した。96ウェルプレートに若いIMR90細胞と老化IMR90細胞をそれぞれ播種し、2日後に上記で得られた候補薬を5μM及び10μMの2段階の濃度で処理した。72時間後に生き残った細胞をホルマリンで固定し、クリスタルバイオレットで染色した。その後、各ウェルのクリスタルバイオレットを2% Triton X100で抽出し定量化した(図13-1、図13-2)。若いIMR90細胞と老化IMR90細胞でそれぞれ溶媒(DMSO)処理細胞を陰性対照とし、陰性対照に対する生存率を、以下の式により算出した。さらに若いIMR90細胞と老化IMR90細胞の薬剤感受性を比較し、若い細胞生存率/老化細胞生存率の値が1.14以上となる化合物を選定した。

以上のとおり、現時点で、ナトリン3b及びRSKキナーゼ阻害剤BI-D1870及び、2次スクリーニングで獲得された薬剤を含めると、図14にまとめたとおり、40個以上の治療剤の候補化合物が得られた。2次スクリーニングで獲得されたこれらの化合物は、機能分類により、およそ7グループに分類できた。なお、図11に示すとおり、さらに3次スクリーニングとして肺線維症モデル動物を用いた試験を行うことで、肺線維症に対する治療効果を奏する薬剤を選択することができる。
5.BLM肺線維化モデルマウスへのナトリン3bの投与効果の検討-1
(1)生存率
10-12週齢のC57BL6マウスに対して、2.5mg/kgのブレオマイシンを気管内投与した。ブレオマシン投与8日後から20mg/kgのナトリン3bあるいは溶媒のみを週に4回、2週間にわたり腹腔内投与した。マウスは毎日生存確認を行い、ブレオマイシン投与から21日後までに死亡したマウスをカウントした。本試験においては、ナトリン3b非投与群(溶媒のみ)が6匹、ナトリン3b投与群が7匹とした。結果を図15に示した。図15に示すとおり、ナトリン3bの投与により、マウスの肺線維化モデルマウスの生存率が向上した。図中、3Bはナトリン3bを示す。以下の図においても同様である。
(1)生存率

(2)線維化関連mRNA発現
10-12週齢のICRマウスに対して、2.5mg/kgのブレオマイシンを気管内投与した。ブレオマシン投与8日後から、14、20、及び24mg/kgのナトリン3bあるいは溶媒のみを週に4回、2週間にわたり腹腔内投与した。ブレオマイシン投与から21日後にナトリン3b投与群あるいは溶媒投与群のマウスをサクリファイスし、肺組織をサンプリングした。試験においては、ナトリン3b非投与群(溶媒のみ)が3匹、ナトリン3b投与群が3匹とした。以下の試験についても同様である。採取した肺組織は十分量のRNAlaterに浸し、4℃で一晩保存した。翌日、RNAlaterより肺組織を取り出し、TRIzoleを用いてTotalRNAを回収した。その後、ReverTra Ace qPCR RT kitによりmRNAを逆転写しcDNAを調製した。調製したcDNAに対し線維化関連遺伝子(Col1a2、Col5a2、Fibronectin、Toropelastin)に特異的なプライマーを用いてリアルタイムPCRを実施した。結果を図16-1~4に示す。

図16-1~4に示すとおり、ブレオマシン投与で上昇した線維化関連遺伝子の発現が、ナトリン3b投与により低下した。
(3)病理比較
10-12週齢のICRマウスに対して、2.5mg/kgのブレオマイシンを気管内投与した。ブレオマシン投与8日後から、14、20、及び24mg/kgのナトリン3bあるいは溶媒のみを週に4回、2週間にわたり腹腔内投与した。ブレオマイシン投与から21日後にナトリン3b投与群あるいは溶媒投与群のマウスをサクリファイスし、肺組織をサンプリングした。肺組織は4%パラホルムアルデヒドで48時間固定処理された。固定後の肺組織はパラフィンに埋包された後、5μmの薄切り切片にされ、スライドガラスにセットされた。この肺組織の薄切切片に対して、マッソントリクロム染色を行い、線維化を評価した。具体的には、マッソントリクロム染色では、核を鉄ヘマトキシリン染色で黒褐色、細胞質を核フクシンで赤色、膠原繊維をアニリンブルーで青色に染色する。そこで、線維化の評価にはImageJ(画像解析ソフト)を用いて染色像を三原色に分離し、青色に染色された面積を線維化領域として定量化した。得られた各群のシグナルをコントロール群(PBS+PEG)に対する相対値として表した。マッソントリクロム染色の結果を図17-1に、線維化領域の定量化結果を図17-2に示す。
10-12週齢のICRマウスに対して、2.5mg/kgのブレオマイシンを気管内投与した。ブレオマシン投与8日後から、14、20、及び24mg/kgのナトリン3bあるいは溶媒のみを週に4回、2週間にわたり腹腔内投与した。ブレオマイシン投与から21日後にナトリン3b投与群あるいは溶媒投与群のマウスをサクリファイスし、肺組織をサンプリングした。肺組織は4%パラホルムアルデヒドで48時間固定処理された。固定後の肺組織はパラフィンに埋包された後、5μmの薄切り切片にされ、スライドガラスにセットされた。この肺組織の薄切切片に対して、マッソントリクロム染色を行い、線維化を評価した。具体的には、マッソントリクロム染色では、核を鉄ヘマトキシリン染色で黒褐色、細胞質を核フクシンで赤色、膠原繊維をアニリンブルーで青色に染色する。そこで、線維化の評価にはImageJ(画像解析ソフト)を用いて染色像を三原色に分離し、青色に染色された面積を線維化領域として定量化した。得られた各群のシグナルをコントロール群(PBS+PEG)に対する相対値として表した。マッソントリクロム染色の結果を図17-1に、線維化領域の定量化結果を図17-2に示す。
図17-1~2に示すとおり、ナトリン3b投与群の肺では、コントロール群と比較して、ブレオマシンによって誘導された線維化面積が小さく、線維症の治療効果が確認された。
6.BLM肺線維化モデルマウスへのナトリン3bの投与効果の検討-2
ブレオマイシン誘導肺線維症におけるFoxM1標的因子へのナトリン3b投与の影響を検討した。具体的には、10-12週齢のICRマウスに対して、2.5mg/kgのブレオマイシンを気管内投与した。ブレオマシン投与8日後から、20mg/kgのナトリン3bあるいは溶媒のみを週に4回、2週間にわたり腹腔内投与した。ブレオマイシン投与から21日後にナトリン3b投与群及び溶媒投与群のマウスをサクリファイスし、肺組織をサンプリングした。試験においては、ナトリン3b非投与群(溶媒のみ)が3匹、ナトリン3b投与群が3匹とした。採取した肺組織は十分量のRNAlaterに浸し、4℃で一晩保存した。翌日、RNAlaterより肺組織を取り出し、TRIzoleを用いてTotalRNAを回収した。その後、ReverTra Ace qPCR RT kitによりmRNAを逆転写しcDNAを調製した。調製したcDNAに対しFoxM1、及びその下流標的遺伝子(Plk1、Ccnb2、Aspm、Cep55)に特異的なプライマーを用いてリアルタイムPCRを実施した。結果を図18-1~18-2に示す。
ブレオマイシン誘導肺線維症におけるFoxM1標的因子へのナトリン3b投与の影響を検討した。具体的には、10-12週齢のICRマウスに対して、2.5mg/kgのブレオマイシンを気管内投与した。ブレオマシン投与8日後から、20mg/kgのナトリン3bあるいは溶媒のみを週に4回、2週間にわたり腹腔内投与した。ブレオマイシン投与から21日後にナトリン3b投与群及び溶媒投与群のマウスをサクリファイスし、肺組織をサンプリングした。試験においては、ナトリン3b非投与群(溶媒のみ)が3匹、ナトリン3b投与群が3匹とした。採取した肺組織は十分量のRNAlaterに浸し、4℃で一晩保存した。翌日、RNAlaterより肺組織を取り出し、TRIzoleを用いてTotalRNAを回収した。その後、ReverTra Ace qPCR RT kitによりmRNAを逆転写しcDNAを調製した。調製したcDNAに対しFoxM1、及びその下流標的遺伝子(Plk1、Ccnb2、Aspm、Cep55)に特異的なプライマーを用いてリアルタイムPCRを実施した。結果を図18-1~18-2に示す。
図18に示すとおり、BLM肺線維症組織ではFoxM1及びその下流標的遺伝子の発現上昇が観察された。BLM誘導肺線維症に対してナトリン3bを投与するとFoxM1標的遺伝子の発現抑制が観察された。
7.老齢マウス肺へのナトリン3bの投与効果の検討
C57BL/6J、20か月齢老齢マウスに対して、週に1回、3か月間ナトリン3bを投与した。3か月後にマウスをサクリファイスし、肺組織のHE染色およびmRNA発現解析をおこなった。比較対象として、若齢マウス(C57BL/6J 10週齢)、及びナトリン3bの非投与の老齢マウス(Vehicle)の肺組織を用いた。結果を図19-1~19-2に示す。
C57BL/6J、20か月齢老齢マウスに対して、週に1回、3か月間ナトリン3bを投与した。3か月後にマウスをサクリファイスし、肺組織のHE染色およびmRNA発現解析をおこなった。比較対象として、若齢マウス(C57BL/6J 10週齢)、及びナトリン3bの非投与の老齢マウス(Vehicle)の肺組織を用いた。結果を図19-1~19-2に示す。
図19-1~19-2に示すとおり、老齢肺組織(Vehicle群)では、老化マーカーであるp21陽性細胞が増加した。一方、老齢肺組織でのp21細胞はナトリン3b投与により有意に抑制された。
8.老齢マウス腎臓へのナトリン3bの投与効果の検討
C57BL/6J、20か月齢老齢マウスに対して、週に1回、3か月間ナトリン3bを投与した。3か月後にマウスをサクリファイスし、腎臓組織のHE染色およびmRNA発現解析をおこなった。比較対象として、若齢マウス(C57BL/6J 10週齢)、及びナトリン3bの非投与の老齢マウス(Vehicle)の腎臓組織を用いた。結果を図20-1~20-3に示す。
C57BL/6J、20か月齢老齢マウスに対して、週に1回、3か月間ナトリン3bを投与した。3か月後にマウスをサクリファイスし、腎臓組織のHE染色およびmRNA発現解析をおこなった。比較対象として、若齢マウス(C57BL/6J 10週齢)、及びナトリン3bの非投与の老齢マウス(Vehicle)の腎臓組織を用いた。結果を図20-1~20-3に示す。
腎臓組織のHE染色から糸球体ヒアリン化を4段階のスコアで評価、および間質ヒアリン化陽性率を評価したところ、図20-1~20-2に示すとおり、ナトリン3b投与群では糸球体及び間質領域でヒアリン化の有意な改善が見られた。また、老齢腎臓組織で誘導される老化マーカー及びSASPマーカー上昇は、ナトリン3b投与により抑制された(図20-3)。
9.PGAM‐Chk1結合阻害変異ノックインマウス(W68A)の寿命延長効果の検討
PGAM‐Chk1結合阻害変異ノックインマウス(W68A)の雄、及び野生型マウスを長期間SPF環境で飼育した。マウスの生存曲線をカプラン・マイヤー方でプロットした結果を図21に示す。

図21に示すとおり、PGAM‐Chk1結合阻害変異ノックインマウス(W68A)は野生型マウスと比較し、長寿の傾向が見られた。
本発明の治療剤は、PGAMとChk1との結合阻害剤を有効成分として含有することで、肺疾患、肝疾患、腎疾患、特に肺疾患、中でも肺線維症に対して優れた治療効果を奏する。これまで有効な治療剤が存在しなかった特発性肺線維症(IPF)等に対しても優れた治療効果が期待できる。

- PGAMとChk1との結合阻害剤を有効成分として含有する、肺疾患、肝疾患又は腎疾患の治療剤。
- PGAMとChk1との結合阻害剤を有効成分として含有する、肺疾患又は腎疾患の治療剤。
- 肺疾患が、肺線維症である、請求項2に記載の治療剤。
- 高齢者における肺疾患、肝疾患又は腎疾患の治療剤である、請求項1に記載の肺疾患、肝疾患又は腎疾患の治療剤。
- 上記結合阻害剤が、RSK1キナーゼ阻害剤、Fakキナーゼ阻害剤、Fakキナーゼが関与するシグナル経路の阻害剤、CDK阻害剤、カルシウム拮抗剤、強心配糖体、DNA障害剤、抗菌剤、オーロラキナーゼ阻害剤、フラボノイド、PI3Kキナーゼ阻害剤、HDAC阻害剤、加齢黄斑変性治療薬、p38MAPK阻害剤、mTOR阻害剤、チロシンキナーゼ阻害剤、Bcl-2阻害剤、HIF-2α阻害剤、スタチン、セロトニン受容体拮抗薬、その他キナーゼ阻害剤、及びナトリン3bからなる群より選択される少なくとも1種である、請求項1又は2に記載の治療剤。
