Abstract
本発明は、アスコルビン酸2-グルコシド塩基性アミノ酸塩及びアスコルビン酸2-グルコシドアルカリ金属塩を含有する食品組成物、化粧料組成物又は医薬組成物に関する。本発明の組成物は、安定で、皮膚に塗布したときの使用感が良好である。
Description
本発明は、アスコルビン酸2-グルコシド類を含有する安定な組成物に関する。
アスコルビン酸は、還元剤、コラーゲン合成促進剤、抗酸化剤として作用し、特に、アスコルビン酸の優れた抗酸化効果は皮膚内でのメラニン生成を抑制し、しみ、そばかすのような色素沈着を防ぐ成分として知られており、医薬、食品、化粧料等に広く配合されている。
しかし、アスコルビン酸は原末で吸湿状態やアルカリ条件下では不安定で、またアスコルビン酸の水溶液は強い酸性で刺激があるだけでなく、容易に酸化し、分解に関与する代表的なアミン類によってメイラード反応が起き褐変が生じ、褐変反応が進行する。これらによりアスコルビン酸を含有した化粧料、水分を含有する商品は褐変による異臭が発生しやすく、商品としての価値が失われ、商品への添加量が制限され効果に影響が余儀なくされる(非特許文献1)。
かかる観点から、アスコルビン酸の褐変を抑制するため、アスコルビン酸にフラボノイド配糖体を配合する技術が報告されている(特許文献1、2)。また、アスコルビン酸に代えてアスコルビン酸2-グルコシドを使用することも報告されている。
しかし、アスコルビン酸2-グルコシドの水溶液は、pHが低く、安定性が若干向上するものの褐変するという欠点は残っている。そこで、アスコルビン酸2-グルコシドに強塩基物質、レシチン、エーテル化合物、エステル化合物、クエン酸などを配合する技術も報告されている(特許文献3~6)。
しかし、アスコルビン酸は原末で吸湿状態やアルカリ条件下では不安定で、またアスコルビン酸の水溶液は強い酸性で刺激があるだけでなく、容易に酸化し、分解に関与する代表的なアミン類によってメイラード反応が起き褐変が生じ、褐変反応が進行する。これらによりアスコルビン酸を含有した化粧料、水分を含有する商品は褐変による異臭が発生しやすく、商品としての価値が失われ、商品への添加量が制限され効果に影響が余儀なくされる(非特許文献1)。
かかる観点から、アスコルビン酸の褐変を抑制するため、アスコルビン酸にフラボノイド配糖体を配合する技術が報告されている(特許文献1、2)。また、アスコルビン酸に代えてアスコルビン酸2-グルコシドを使用することも報告されている。
しかし、アスコルビン酸2-グルコシドの水溶液は、pHが低く、安定性が若干向上するものの褐変するという欠点は残っている。そこで、アスコルビン酸2-グルコシドに強塩基物質、レシチン、エーテル化合物、エステル化合物、クエン酸などを配合する技術も報告されている(特許文献3~6)。

しかしながら、いずれの手段でも、アスコルビン酸2-グルコシドの褐変反応は十分に抑制できず、これを含有する化粧料などの組成物の優れた作用効果を維持しつつその褐変を十分に抑制できるものではなかった。
そこで、本発明者は、アスコルビン酸2-グルコシドの塩基性アミノ酸塩を製造し、単離したところ、この物質が従来のアスコルビン酸やアスコルビン酸2-グルコシドに比べて、褐変せず安定であることを見出した。しかし、その安定性はさらに改良の余地があり、皮膚に塗布したときの使用感も改良の余地があった。
従って、本発明の課題は、アスコルビン酸2-グルコシド類を含有する安定で使用感の良好な組成物を提供することにある。
そこで、本発明者は、アスコルビン酸2-グルコシドの塩基性アミノ酸塩を製造し、単離したところ、この物質が従来のアスコルビン酸やアスコルビン酸2-グルコシドに比べて、褐変せず安定であることを見出した。しかし、その安定性はさらに改良の余地があり、皮膚に塗布したときの使用感も改良の余地があった。
従って、本発明の課題は、アスコルビン酸2-グルコシド類を含有する安定で使用感の良好な組成物を提供することにある。
そこで本発明者は、アスコルビン酸2-グルコシド塩基性アミノ酸塩に種々の成分を配合し、その安定性と皮膚に塗布したときの使用感について検討してきたところ、アスコルビン酸2-グルコシド塩基性アミノ酸塩とアスコルビン酸2-グルコシドアルカリ金属塩を併用すれば、安定性が向上し、かつ皮膚に塗布したときの使用感も向上することを見出し、本発明を完成した。
本発明は、次の発明[1]~[6]を提供するものである。
[1]アスコルビン酸2-グルコシド塩基性アミノ酸塩及びアスコルビン酸2-グルコシドアルカリ金属塩を含有する食品組成物、化粧料組成物又は医薬組成物。
[2]アスコルビン酸2-グルコシド塩基性アミノ酸塩が、アスコルビン酸2-グルコシドリジン塩、アスコルビン酸2-グルコシドアルギニン塩、アスコルビン酸2-グルコシドヒスチジン塩、及びアスコルビン酸2-グルコシドトリプトファン塩から選ばれる1種又は2種以上である[1]記載の組成物。
[3]アスコルビン酸2-グルコシドアルカリ金属塩が、アスコルビン酸2-グルコシドナトリウム塩及びアスコルビン酸2-グルコシドカリウム塩から選ばれる1種又は2種である[1]又は[2]記載の組成物。
[4]組成物のpHが5.0以上7.5以下である[1]~[3]のいずれかに記載の組成物。
[5]アスコルビン酸2-グルコシド塩基性アミノ酸塩が、アスコルビン酸2-グルコシドアルギニン塩である[1]~[4]のいずれかに記載の組成物。
[6]アスコルビン酸2-グルコシドアルカリ金属塩が、アスコルビン酸2-グルコシドナトリウム塩である[1]~[5]のいずれかに記載の組成物。
[1]アスコルビン酸2-グルコシド塩基性アミノ酸塩及びアスコルビン酸2-グルコシドアルカリ金属塩を含有する食品組成物、化粧料組成物又は医薬組成物。

アスコルビン酸2-グルコシド塩基性アミノ酸塩及びアスコルビン酸2-グルコシドアルカリ金属塩を含有する水溶液は、着色が顕著に抑制され、そのpHが弱酸性から中性であるから、アスコルビン酸類の多様な機能であるメラニン生成抑制による美白効果及びコラーゲン生成活性化によるしわ改善効果を有し、皮膚刺激性のない安全な食品、医薬又は化粧料の配合成分として有用である。
本発明の一態様は、アスコルビン酸2-グルコシド塩基性アミノ酸塩及びアスコルビン酸2-グルコシドアルカリ金属塩を含有する組成物である。この組成物は、特に、食品組成物、化粧料組成物又は医薬組成物として有用である。
本発明におけるアスコルビン酸とは、IUPAC名を、(R)-3,4-ジヒドロキシ-5-((S)-1,2-ジヒドロキシエチル)フラン-2(5H)-オンという化合物であり、ビタミンC、L-アスコルビン酸とも呼ばれる物質である。アスコルビン酸は、前述のごとく、還元剤、コラーゲン合成促進剤、抗酸化剤として作用し、特に、アスコルビン酸の優れた抗酸化効果は皮膚内でのメラニン生成を抑制し、しみ、そばかすのような色素沈着を防ぐ成分として知られており、医薬、食品、化粧料に広く使用されている。
アスコルビン酸2-グルコシドとは、アスコルビン酸の2位の炭素原子にグルコースが結合した物質であり、生体内で代謝されてアスコルビン酸になるので、アスコルビン酸のプロビタミンとされている。本発明で用いられるアスコルビン酸2-グルコシド塩基性アミノ酸塩は、アスコルビン酸2-グルコシドの塩基性アミノ酸塩である。
ここで、アスコルビン酸2-グルコシド塩基性アミノ酸としては、アスコルビン酸2-グルコシドリジン塩、アスコルビン酸2-グルコシドアルギニン塩、アスコルビン酸2-グルコシドヒスチジン塩、及びアスコルビン酸2-グルコシドトリプトファン塩から選ばれる1種又は2種以上が挙げられる。このうち、アスコルビン酸2-グルコシドリジン塩、アスコルビン酸2-グルコシドアルギニン塩がより好ましく、アスコルビン酸2-グルコシドアルギニン塩がさらに好ましい。
アスコルビン酸2-グルコシドの塩基性アミノ酸塩におけるアスコルビン酸2-グルコシドと塩基性アミノ酸との構成モル比率は、1:1.5~1.5:1が好ましく、1:1.2~1.2:1がより好ましい。
ここで、アスコルビン酸2-グルコシド塩基性アミノ酸としては、アスコルビン酸2-グルコシドリジン塩、アスコルビン酸2-グルコシドアルギニン塩、アスコルビン酸2-グルコシドヒスチジン塩、及びアスコルビン酸2-グルコシドトリプトファン塩から選ばれる1種又は2種以上が挙げられる。このうち、アスコルビン酸2-グルコシドリジン塩、アスコルビン酸2-グルコシドアルギニン塩がより好ましく、アスコルビン酸2-グルコシドアルギニン塩がさらに好ましい。
アスコルビン酸2-グルコシドの塩基性アミノ酸塩におけるアスコルビン酸2-グルコシドと塩基性アミノ酸との構成モル比率は、1:1.5~1.5:1が好ましく、1:1.2~1.2:1がより好ましい。
アスコルビン酸2-グルコシド塩基性アミノ酸塩は、アスコルビン酸2-グルコシドに塩基性アミノ酸を反応させることにより製造することができる。具体的には、超音波処理などで溶存酸素濃度を低下させたイオン交換水中、40~60℃で、アスコルビン酸2-グルコシド及び塩基性アミノ酸を添加し、pHを5.22~5.55に調整することにより、塩基性アミノ酸の添加量を調整して反応させる。反応終了後は、凍結乾燥、噴霧乾燥などにより、アスコルビン酸2-グルコシド塩基性アミノ酸塩を粉末として単離することができる。
得られたアスコルビン酸2-グルコシド塩基性アミノ酸塩粉末は、長期間安定であり、褐変もしないので、化粧料や医薬組成物に高濃度配合するための原料として有用である。溶液では他の基剤と混合するために、どうしても濃度上限に限界があった。液体状態で原料を保存するためには、防腐剤などを添加する、フィルター通液により無菌化し、無菌容器に充填する、あるいは低温にて保管するなどの必要があるが、粉末化することで、他の成分を添加せずに、室温で安定的にしかもコンパクトに保存することが可能となる。
得られたアスコルビン酸2-グルコシド塩基性アミノ酸塩粉末は、長期間安定であり、褐変もしないので、化粧料や医薬組成物に高濃度配合するための原料として有用である。溶液では他の基剤と混合するために、どうしても濃度上限に限界があった。液体状態で原料を保存するためには、防腐剤などを添加する、フィルター通液により無菌化し、無菌容器に充填する、あるいは低温にて保管するなどの必要があるが、粉末化することで、他の成分を添加せずに、室温で安定的にしかもコンパクトに保存することが可能となる。
本発明組成物中のアスコルビン酸2-グルコシド塩基性アミノ酸塩の含有量は、安定性、使用感及びアスコルビン酸2-グルコシド塩基性アミノ酸塩のアスコルビン酸としての機能発揮性の観点から、1~40質量%が好ましく、2~35質量%がより好ましく、5~30質量%がさらに好ましい。
アスコルビン酸2-グルコシドアルカリ金属塩としては、アスコルビン酸2-グルコシドナトリウム塩、アスコルビン酸2-グルコシドカリウム塩、アスコルビン酸2-グルコシドリチウム塩が挙げられる。このうち、アスコルビン酸2-グルコシドナトリウム塩、アスコルビン酸2-グルコシドカリウム塩が好ましく、アスコルビン酸2-グルコシドナトリウム塩がさらに好ましい。
アスコルビン酸2-グルコシドアルカリ金属塩におけるアスコルビン酸2-グルコシドとアルカリ金属との構成モル比率は、1:1.5~1.5:1が好ましく、1:1.2~1.2:1がより好ましい。
アスコルビン酸2-グルコシドアルカリ金属塩におけるアスコルビン酸2-グルコシドとアルカリ金属との構成モル比率は、1:1.5~1.5:1が好ましく、1:1.2~1.2:1がより好ましい。
アスコルビン酸2-グルコシドアルカリ金属塩は、特開2002-88095号公報、特許第6895436号公報などに記載の方法によって製造することができる。例えば、アスコルビン酸2-グルコシドに水酸化ナトリウム、炭酸ナトリウム、炭酸水素ナトリウムなどのアルカリ金属塩を反応させればよい。具体的には、音波処理など溶存酸素濃度を低下させたイオン交換水中、40~60℃で、アスコルビン酸2-グルコシド及びアルカリ金属塩を添加し、pHを6.0-6.5に調整することにより、アルカリ金属塩の添加量を調整して反応させる。反応終了後は、凍結乾燥、噴霧乾燥などにより、アスコルビン酸2-グルコシドアルカリ金属塩を粉末として単離することができる。
アスコルビン酸2-グルコシドアルカリ金属塩は、アスコルビン酸と同様の生理活性を有することが知られているが、アスコルビン酸2-グルコシド塩基性アミノ酸塩と併用することにより、安定性が向上すること及び使用感が向上することは、知られていない。

本発明組成物中のアスコルビン酸2-グルコシドアルカリ金属塩の含有量は、安定性、使用感及びアスコルビン酸2-グルコシドアルカリ金属塩のアスコルビン酸としての機能発揮性の観点から、1~50質量%が好ましく、2~45質量%がより好ましく、5~40質量%がさらに好ましい。
本発明組成物中のアスコルビン酸2-グルコシド塩基性アミノ酸塩(a)とアスコルビン酸2-グルコシドアルカリ金属塩(b)の含有モル比(a/b)は、組成物の着色防止効果、アスコルビン酸としての機能発揮効果、及びべたつき感などの使用感の観点から、0.2~5.0が好ましく、0.3~3.0がより好ましく、0.5~2.0がさらに好ましい。
本発明の組成物のpHは、着色防止効果及び皮膚に適用した際の刺激性低減の観点から、5.0~7.5が好ましく、5.0~6.5がより好ましく、5.0~6.0がさらに好ましい。
アスコルビン酸2-グルコシド塩基性アミノ酸塩及びアスコルビン酸2-グルコシドアルカリ金属塩を含有する組成物としては、食品組成物、医薬組成物、化粧料組成物が挙げられる。これらの組成物の形態としては、液状組成物、乳化組成物、粉体状組成物、固形組成物が挙げられる。
食品組成物としては、機能性食品、特定保健用食品などが挙げられる。また、医薬組成物としては、錠剤、顆粒剤、シロップ剤などの経口投与用製剤、クリーム剤、軟膏剤、液剤などの経皮投与用製剤、点眼薬、注射剤などが挙げられる。
化粧料組成物としては、液状化粧料、乳化化粧料、粉体状化粧料、固形化粧料などとすることができる。これらの化粧料には、アスコルビン酸2-グルコシド塩基性アミノ酸塩及びアスコルビン酸2-グルコシドアルカリ金属塩以外に、アスコルビン酸又はその塩、水、油剤、界面活性剤、保湿成分、アルコール類、ポリオール類、紫外線吸収剤、紫外線防御剤、各種粉体、各種美容成分、酸化防止剤、抗菌剤、香料などが含まれているのが好ましい。
また、本発明の化粧料は、アスコルビン酸2-グルコシド塩基性アミノ酸塩及びアスコルビン酸2-グルコシドアルカリ金属塩の機能を十分に得る観点から、皮膚用化粧料であるのがより好ましい。またその形態は、水性液状化粧料、水中油型乳化化粧料、油中水型乳化化粧料であるのが好ましい。
食品組成物としては、機能性食品、特定保健用食品などが挙げられる。また、医薬組成物としては、錠剤、顆粒剤、シロップ剤などの経口投与用製剤、クリーム剤、軟膏剤、液剤などの経皮投与用製剤、点眼薬、注射剤などが挙げられる。
化粧料組成物としては、液状化粧料、乳化化粧料、粉体状化粧料、固形化粧料などとすることができる。これらの化粧料には、アスコルビン酸2-グルコシド塩基性アミノ酸塩及びアスコルビン酸2-グルコシドアルカリ金属塩以外に、アスコルビン酸又はその塩、水、油剤、界面活性剤、保湿成分、アルコール類、ポリオール類、紫外線吸収剤、紫外線防御剤、各種粉体、各種美容成分、酸化防止剤、抗菌剤、香料などが含まれているのが好ましい。
また、本発明の化粧料は、アスコルビン酸2-グルコシド塩基性アミノ酸塩及びアスコルビン酸2-グルコシドアルカリ金属塩の機能を十分に得る観点から、皮膚用化粧料であるのがより好ましい。またその形態は、水性液状化粧料、水中油型乳化化粧料、油中水型乳化化粧料であるのが好ましい。
次に実施例を挙げて本発明をさらに詳細に説明するが、本発明はこれら実施例に限定されるものではない。
〔製造例1〕
イオン交換水を0.5kW-1.0kW出力の超音波で脱気を行った。0.8kW設定時で20-40分間脱気するが、脱気の確認は溶存酸素濃度計で測定し溶存酸素濃度値0.5ppm以下に成ったことを確認しイオン交換水を得た。この水50mlを45-60℃に設定するが、より好ましくは50℃±0.5に設定した。アスコルビン酸2-グルコシド5.0gを採り攪拌溶解後、アルギニン14.464mmolを加えた。この時pHは5.22-5.55とするのが好ましく、より好ましくは5.35である。この時アルギニン添加モル濃度をpHで微調整する。この溶液を凍結乾燥し、アスコルビン酸2-グルコシドアルギニン塩の粉末(7.55g、90.03%)を得た。得られたアスコルビン酸2-グルコシドアルギニン塩の粉末のFT/IRスペクトルを図1に示す。スペクトル結果は1720cm-1(非イオンCOOH,逆対称伸縮振動)と特有バンド1400cm-1が観測され、アルギニンのイオン結合導入が確認された。

〔製造例2〕
溶解に使用されるイオン交換水処理法は製造例1と同様に行った。このイオン交換水100mlを50℃±0.5に設定し、アスコルビン酸2-グルコシド5.0gを添加攪拌溶解後、リジン17.97mmolを加えた。pHは5.1-5.6を呈するがモル濃度を微調整してpH5.3とした。この溶液を凍結乾燥し、アスコルビン酸2-グルコシドリジン塩の粉末(7.62g、98.42%)が得られた。
溶解に使用されるイオン交換水処理法は製造例1と同様に行った。このイオン交換水100mlを50℃±0.5に設定し、アスコルビン酸2-グルコシド5.0gを添加攪拌溶解後、リジン17.97mmolを加えた。pHは5.1-5.6を呈するがモル濃度を微調整してpH5.3とした。この溶液を凍結乾燥し、アスコルビン酸2-グルコシドリジン塩の粉末(7.62g、98.42%)が得られた。
〔実施例1〕
(アスコルビン酸2-グルコシドアルギニン塩(ASA2-G・Arg)とアスコルビン酸2-グルコシドナトリウム塩(ASA2-G・Na)の混合)
ASA2-Gの溶解度は、879g/L(25℃)すなわち、46.8重量%とされている。ASA2-G・ArgとASA2-G・Naは、水に対する溶解度が向上しており、70重量%以上の高濃度で水に溶解することができた。また、ASA2-G・ArgとASA2-G・Naを同時に水に溶かすことができ、両者あわせてのアスコルビン酸2-グルコシド誘導体の濃度を高くすることも可能であった。
ASA2-G・ArgとASA2-G・Naを混合した美容液(これらの成分の合計濃度は40重量%)を調製し、6名の被験者の顔に塗布してもらい、使用感(べたつき)のアンケート結果を得た。良いから悪いまでの5段階のスコア(スコアが高いほど良い)を平均した。混合の重量比率は、ASA2-G・Arg:ASA2-G・Na=100:0、80:20、60:40、40:60、20:80、0:100の4種類(試験品2-5)とし、ASA2-G・Argが単独で溶解している試験品1と比較した。
さらに、これら試験品1-5を透明なガラス製容器に充填し、40℃に2週間保管して、着色の変化を肉眼観察した。
得られた結果を、表1に示す。
(アスコルビン酸2-グルコシドアルギニン塩(ASA2-G・Arg)とアスコルビン酸2-グルコシドナトリウム塩(ASA2-G・Na)の混合)

ASA2-G・ArgとASA2-G・Naを混合した美容液(これらの成分の合計濃度は40重量%)を調製し、6名の被験者の顔に塗布してもらい、使用感(べたつき)のアンケート結果を得た。良いから悪いまでの5段階のスコア(スコアが高いほど良い)を平均した。混合の重量比率は、ASA2-G・Arg:ASA2-G・Na=100:0、80:20、60:40、40:60、20:80、0:100の4種類(試験品2-5)とし、ASA2-G・Argが単独で溶解している試験品1と比較した。

得られた結果を、表1に示す。
試験品1では、塗布後にベタベタするとの印象を持った被験者がいた。試験品2-5では使用感はほぼ同じで、ベタベタすると印象を持った被験者はいなかった。ASA2-G・NaをASA2-G・Argに対して1/4以上配合すると使用感が大幅に向上することがわかった。さらに、これら試験品2-5は、40℃に2週間保管した場合の褐変の程度は同程度で、差はなかった。
〔実施例2〕
アスコルビン酸2-グルコシドアルギニン塩(ASA2-G・Arg)を5質量%及びアスコルビン酸2-グルコシドナトリウム塩(ASA2-G・Na)を5質量%配合して美容液を製造する場合の配合例及び製法を以下に示す。
(1)配合(単位は質量%)
1. 水 67.98%
2. プロパンジオール 17.0%
3. ASA2-G・Arg 5.0%
4. ASA2-G・Na 5.0%
5. グリセリン 3.9%
6. クエン酸Na 0.5%
7. 1,3-BG 0.30%
8. スフィンゴ糖脂質 0.15%
9. キサンタンガム 0.12%
10. ヒアルロン酸Na 0.02%
11. クエン酸 0.02%
12. アセチルヒドロキシプロリン 0.01%
13. 加水分解コラーゲン 0.001%
14. 加水分解エラスチン 0.00001%
(2)製法
油相成分を混合し、約80℃に加熱する。次に残りの原料(水溶成分)を混合して、約80℃に加熱する。80℃に保ちつつ、ホモミキサーで撹拌しながら水溶成分に油相成分を徐々に加えて乳化する。最後に温度を室温に冷やしてから、ASA2-G・Arg及びASA2-GNaを添加し、よく混合し、均一としてから製品とする。
アスコルビン酸2-グルコシドアルギニン塩(ASA2-G・Arg)を5質量%及びアスコルビン酸2-グルコシドナトリウム塩(ASA2-G・Na)を5質量%配合して美容液を製造する場合の配合例及び製法を以下に示す。
(1)配合(単位は質量%)
1. 水 67.98%
2. プロパンジオール 17.0%
3. ASA2-G・Arg 5.0%
4. ASA2-G・Na 5.0%
5. グリセリン 3.9%
6. クエン酸Na 0.5%

(2)製法
油相成分を混合し、約80℃に加熱する。次に残りの原料(水溶成分)を混合して、約80℃に加熱する。80℃に保ちつつ、ホモミキサーで撹拌しながら水溶成分に油相成分を徐々に加えて乳化する。最後に温度を室温に冷やしてから、ASA2-G・Arg及びASA2-GNaを添加し、よく混合し、均一としてから製品とする。
Claims (6)
- アスコルビン酸2-グルコシド塩基性アミノ酸塩及びアスコルビン酸2-グルコシドアルカリ金属塩を含有する食品組成物、化粧料組成物又は医薬組成物。
- アスコルビン酸2-グルコシド塩基性アミノ酸塩が、アスコルビン酸2-グルコシドリジン塩、アスコルビン酸2-グルコシドアルギニン塩、アスコルビン酸2-グルコシドヒスチジン塩、及びアスコルビン酸2-グルコシドトリプトファン塩から選ばれる1種又は2種以上である請求項1記載の組成物。
- アスコルビン酸2-グルコシドアルカリ金属塩が、アスコルビン酸2-グルコシドナトリウム塩及びアスコルビン酸2-グルコシドカリウム塩から選ばれる1種又は2種である請求項1又は2記載の組成物。
- 組成物のpHが5.0以上7.5以下である請求項1~3のいずれか1項に記載の組成物。
- アスコルビン酸2-グルコシド塩基性アミノ酸塩が、アスコルビン酸2-グルコシドアルギニン塩である請求項1~4のいずれか1項に記載の組成物。
- アスコルビン酸2-グルコシドアルカリ金属塩が、アスコルビン酸2-グルコシドナトリウム塩である請求項1~5のいずれか1項に記載の組成物。
