Description
本開示は、糸状菌を用いて免疫グロブリンまたは多量体免疫グロブリンを製造する方法、上記製造方法に利用し得る糸状菌、および当該糸状菌を製造する方法に関する。
これまで、抗体などの有益なタンパク質の製造のために、トランスジェニック動物、哺乳類細胞培養、又は植物などが使用されている。しかしながら、トランスジェニック動物及び哺乳類細胞培養は、ウイルスやプリオンなどに汚染されるというリスクを有する。また、上記動物、動物細胞又は植物を用いた生産システムにおいて規模を拡大することは困難であり、遺伝子組換え植物では、組換えタンパク質の生産に約10ヶ月を要し、一方、哺乳類細胞培養では、約3ヶ月を要する。
糸状菌の一種である麹菌(アスペルギルス・オリゼ:Aspergillus oryzae)は日本酒、醤油、味噌といった日本における伝統的醸造製品の製造に古くから用いられてきた糸状菌であり、日本醸造学会により国菌として認定されている微生物である(非特許文献1:Kitamoto, 2002)。A.oryzaeは優れたタンパク質分泌能力を有しており、実際にA.oryzaeを用いて有用なタンパク質が生産されている(非特許文献2:Ito et al., 2007; 非特許文献3:Nakajima et al., 2006; 非特許文献4:Tsuchiya et al., 1992, 非特許文献5:Tsuchiya et al., 1994)。また、世界保健機関 (WHO) および国際食糧農業機関 (FAO) によりA.oryzaeは安全性の高い菌として認められており(非特許文献6:Barbesgaard et al., 1992)、食品・化成品・医療品向けの酵素剤および代謝化合物の生産にも利用されている。
糸状菌によって産生される異種タンパク質には、異種糸状菌由来の結晶セルロース緩和タンパク質(非特許文献7:Chen et al. (2010) Appl Environ Microbiol、特許文献1:特開2009-207368号公報)、植物由来の味覚修飾タンパク質(非特許文献3:Nakajima et al. 免疫グロブリンは重鎖定常領域の構造の相違によってIgG、IgA、IgM、IgD、IgEの5種類に分類され、それぞれ化学的特性が異なっている。特に、IgM、IgA免疫グロブリンは生体内で単量体の状態又は多量体の状態で機能することが知られている。当該多量体の状態のグロブリンは多量体免疫グロブリンと呼ばれる。

本開示は、糸状菌を用いて免疫グロブリンまたは多量体免疫グロブリンを製造する方法を提供する。別の態様において、本開示は上記製造方法に利用し得る糸状菌または当該糸状菌を製造する方法を提供する。
本開示の発明者らが鋭意研究を進めた結果、ゲノム上に免疫グロブリンをコードする核酸配列を有する糸状菌を培養することで、生理活性、例えば細菌増殖抑制活性等を保持した免疫グロブリンまたは多量体免疫グロブリンが産生されること、および当該免疫グロブリンまたは多量体免疫グロブリンが細胞培養培地中に分泌されること、並びに、産生された免疫グロブリンまたは多量体免疫グロブリンを分離してその生理活性を利用し得ることを予想外にも見出した。
当該知見に基づいて、本開示は一態様において、ゲノム中に免疫グロブリンをコードする核酸断片を含む糸状菌を用いて、多量体免疫グロブリンを製造する方法を提供する。
また、本開示は別の態様において、ゲノム中に哺乳類免疫グロブリンをコードする核酸断片を含有し、多量体哺乳類免疫グロブリンを製造することを特徴とする、糸状菌を提供する。
また、本開示は別の態様において、ゲノム中に免疫グロブリンをコードする糸状菌を製造する方法であって、前記免疫グロブリンをコードする核酸配列を、改変対象の糸状菌のゲノムに挿入することを含む、改変糸状菌を製造する方法を提供する。
また、本開示は別の態様において、ゲノム中に免疫グロブリンをコードする核酸断片を含有し、免疫グロブリンを製造することを特徴とする糸状菌を提供する。

本開示は、糸状菌を用いて免疫グロブリンまたは多量体免疫グロブリンを製造する方法を提供するという効果を有する。別の態様において、本開示は上記製造方法に利用し得る糸状菌または当該糸状菌を製造する方法を提供するという効果を有する。
本開示の発明者らは、既存の知見に基づいて糸状菌を用いて多量体免疫グロブリンを製造することを試みたが、単量体免疫グロブリンと異なり、多量体免疫グロブリンを製造すること、特に、生理活性を保持した多量体免疫グロブリンを細胞培地中に分泌させ、これを単離することで多量体免疫グロブリンを製造することは不可能であった。本開示の発明者らは、独自の試行錯誤を試みた結果、驚くべきことに、ゲノム上に免疫グロブリンをコードする核酸配列を有する糸状菌を培養することで、生理活性を保持した多量体免疫グロブリンが細胞培養培地中に分泌され、これを分離してその生理活性を利用し得ることを予想外にも見出した。典型的にはゲノム上に免疫グロブリンをコードする核酸配列を有する糸状菌は、ゲノム上に免疫グロブリンをコードする核酸配列を2コピー以上有している。

一態様において、糸状菌を培養する培地のpHは、4~11の間の任意のpHで調節することが可能である。好ましくは、培地のpHは4.0~11.0、4.5~10.0、5.0~9.0、5.5~8.5または6.0~8.0の範囲から選ばれる。一態様において、培地のpHは5.0以上、5.5以上、6.0以上、6.5以上、7.0以上、7.5以上、8.0以上、8.5以上、9.0以上および、9.5以上、並びに、11.0以下、10.5以下、10.0以下、9.5以下、9.0以下、8.5以下、8.0以下、7.5以下、7.0以下、6.5以下、6.0以下、5.5以下および5.0以下、またはこれら上限および下限の任意の組み合わせの範囲に設定することができる。
本開示の免疫グロブリンを製造する方法において、培養培地中の免疫グロブリンを他の培養培地成分から分離する工程では、菌体により(菌体内にまたは好ましくは菌体外に)生産された免疫グロブリンを定法に従って分離することができる。本開示の免疫グロブリンを製造する方法において使用される糸状菌は典型的には免疫グロブリンを菌体外に分泌生産するため、免疫グロブリンの単離および精製のために菌体を破壊するといった処理は必須ではない。好ましい態様において、本開示の免疫グロブリンを製造する方法は、菌体を破壊する処理を含まない。本開示の免疫グロブリンを製造する方法において使用される糸状菌は免疫グロブリンを菌体内に生産してもよい。免疫グロブリンを菌体内に含む糸状菌は、経口投与に適する。免疫グロブリンを菌体内に含む糸状菌は、経口投与する場合には、投与前に菌体を破壊してもよいが、破壊しなくてもよい。

本開示の方法で製造される、多量体免疫グロブリンは多量体化を促進するための異種アミノ酸配列を含んでもよい。このような異種アミノ酸配列として、例えばコイルドコイルドメインを形成するアミノ酸配列を利用することができる。一態様において、当該異種アミノ酸配列は、軟骨オリゴマー基質タンパク質(COMP)、マンノース結合タンパク質A、コイルドコイルセリンリッチタンパク質1、ポリペプチド放出因子2、SNAP-25、SNARE、Lacリプレッサー、またはアポリポタンパク質Eのうちのいずれかに由来するものを利用することができる。好ましい態様において、多量体化を促進するための異種アミノ酸配列として、例えば特表2016―512309に記載される軟骨オリゴマー基質タンパク質(COMP)のコイルドコイル集合ドメインすることができる。当該異種アミノ酸配列は、免疫グロブリンタンパク質のN末端またはC末端、好ましくはC末端に融合させることができる。当該融合には多量体免疫グロブリンの所望の生理機能を喪失しない限り様々なリンカー配列を使用することができるが、使用しなくてもよい。一態様において、アミノ酸配列SSADDAKKDAAKKDDAKKDDAKKDASからなるリンカーあるいは、アミノ酸配列SSADDAAADAAAADDAAADDAAADASからなるリンカーを使用することができる。一態様において、当該多量体化を促進するための異種配列リンカーまたはリンカーは、免疫グロブリンの短縮されたN末端またはC末端に融合することができる。
(免疫グロブリンまたは多量体免疫グロブリンを製造することを特徴とする改変糸状菌) 本開示は別の態様において、ゲノム中に哺乳類免疫グロブリンをコードする核酸断片を含有し、免疫グロブリンまたは多量体免疫グロブリンを製造することを特徴とする改変糸状菌を提供する。当該改変糸状菌は、上記本開示の(免疫グロブリンを製造する方法)または(多量体免疫グロブリンを製造する方法)に使用することができ、菌体内または菌体外に免疫グロブリンまたは多量体免疫グロブリンを産生または分泌する。
本開示の多量体免疫グロブリンを製造することを特徴とする改変糸状菌は、ゲノム上に免疫グロブリンをコードする核酸断片を典型的には2コピー以上有している。一態様において、本開示の改変糸状菌は、ゲノム上に免疫グロブリンをコードする核酸配列を2コピー以上、3コピー以上、4コピー以上、5コピー以上、6コピー以上、7コピー以上、8コピー以上、9コピー以上、10コピー以上有している。ゲノム上に含まれる、免疫グロブリンをコードする核酸配列のコピー数が多いほど、より効率的に多量体免疫グロブリンを産生または分泌する改変糸状菌を取得することが容易である。二量体免疫グロブリンを産生する糸状菌は、例えば、ゲノム上に免疫グロブリンをコードする核酸配列を2コピー以上、3コピー以上、4コピー以上、5コピー以上、6コピー以上、7コピー以上、8コピー以上、9コピー以上、10コピー以上有している。三量体免疫グロブリンを産生する糸状菌は、例えば、ゲノム上に免疫グロブリンをコードする核酸配列を3コピー以上、4コピー以上、5コピー以上、6コピー以上、7コピー以上、8コピー以上、9コピー以上、10コピー以上有している。五量体免疫グロブリンを産生する糸状菌は、例えば、ゲノム上に免疫グロブリンをコードする核酸配列を5コピー以上、6コピー以上、7コピー以上、8コピー以上、9コピー以上、10コピー以上有している。免疫グロブリンをコードする核酸断片は、ゲノム上の単一領域又は複数の領域内に挿入又は置換される。典型的には、免疫グロブリンをコードする核酸断片はゲノム上の単一領域に挿入または置換される。ゲノム上の単一領域とは、当該糸状菌ゲノム上の一つの連続した領域を意味する。一態様において、ゲノム上の単一領域は、例えば1遺伝子座程度の大きさを有する。例えば、ゲノム上の単一領域は、3~12kbp程度の大きさを有するが、特に限定されない。
本開示の免疫グロブリンまたは多量体免疫グロブリンを製造することを特徴とする改変糸状菌は、ゲノム改変処理を行った糸状菌を培養し、菌体内または培養培地中に免疫グロブリンまたは多量体免疫グロブリンを産生または分泌する糸状菌を同定することで取得することができる。
ゲノム改変処理を行った糸状菌が分泌する多量体免疫グロブリンが多量体形態であることは、本開示の技術分野で一般的に使用される各種方法を用いて確認できる。例えば、培養培地から分離された免疫グロブリンを、非還元状態のドデシルナトリウム硫酸ポリアクリルアミドゲル電気泳動(SDS-PAGE)にて解析することにより、当該多量体免疫グロブリンが多量体であることを確認できる。
本開示のゲノム改変糸状菌を製造するために使用するゲノム改変技術(ゲノム編集技術)には、本開示の技術分野に一般的に使用される各種方法を利用することができる。例えば、CRISPR-Casシステム、メガヌクレアーゼ(MN)、ジンクフィンガーヌクレアーゼ(ZFN)、および転写活性化様エフェクターヌクレアーゼ(TALEN)などを利用することができる。一態様において、改変後の糸状菌内にゲノム改変用のプラスミドが残存しない改変技術を用いることが好ましい。例えば、特開2018-191551に開示される、多段階による多重変異株作成用のゲノム編集プラスミドを単一段階の変異操作または多段階の変異操作に利用することで、改変後の糸状菌内にゲノム改変用のプラスミドが残存しないゲノム改変糸状菌を得ることができる。
本開示に係るゲノム改変糸状菌は、従来公知の糸状菌のゲノムを改変することで作製できる。改変対象とする糸状菌としては特に限定されない。使用できる糸状菌の非限定的な例として、Aspergillus aculeatus、Aspergillus luchuensis、Aspergillus caesiellus、Aspergillus candidus、Aspergillus carneus、Aspergillus clavatus、Aspergillus deflectus、Aspergillus fischerianus、Aspergillus flavus、Aspergillus fumigatus、Aspergillus glaucus、Aspergillus nidulans、Aspergillus niger、Aspergillus ochraceus、Aspergillus oryzae、Aspergillus parasiticus、Aspergillus penicilloides、Aspergillus restrictus、Aspergillus sojae、Aspergillus sydowi、Aspergillus tamari、Aspergillus terreus、Aspergillus ustus及びAspergillus versicolor等のAspergillus属糸状菌、Trichoderma aggressivum、Trichoderma asperellum、Trichoderma atroviride、Trichoderma aureoviride、Trichoderma austrokoningii、Trichoderma brevicompactum、Trichoderma candidum、Trichoderma caribbaeum var. aequatoriale、Trichoderma caribbaeum var. また、改変対象とする糸状菌は、野生株でも良く、各種の変異が導入された変異株でも良い。例えば、WO2007/099776号公報に記載される、液胞内酸性プロテアーゼ遺伝子を破壊した変異株、特開2012-179011号公報に開示されるオートファジー関連遺伝子の機能を低減させた変異株などを用いてもよい。
別の態様において、本開示の改変糸状菌として、免疫グロブリンをコードする核酸配列をゲノムに導入する改変に加えて別の改変を加えた糸状菌を用いることができる。例えば、WO2007/099776号公報に記載される、液胞内酸性プロテアーゼ遺伝子の破壊、特開2012-179011号公報に開示されるオートファジー関連遺伝子の機能低減といった改変を加えることができる。
本開示の改変糸状菌のゲノムに含まれる核酸断片がコードする免疫グロブリンまたは多量体免疫グロブリンは、様々な生物の免疫グロブリンであってよい。例えば、ヒト、ヒト以外の霊長類、マウス、ラット、モルモット、ウサギ、ハムスター、イヌ、ネコ、イタチ、ウシ、ブタ、ウマ、シカ、イノシシ、ヒツジ、ヤギ、ラクダ等の免疫グロブリンであってもよい。免疫グロブリンまたは多量体免疫グロブリンは、好ましくはIgAまたはIgMである。
(免疫グロブリンまたは多量体免疫グロブリンを産生する改変糸状菌を製造する方法)
本開示は一態様において、ゲノム中に免疫グロブリンをコードする糸状菌を製造する方法であって、前記免疫グロブリンをコードする核酸配列を、改変対象の糸状菌のゲノムに挿入することを含む、免疫グロブリンまたは多量体免疫グロブリンを産生する改変糸状菌を製造する方法を提供する。
本開示は一態様において、ゲノム中に免疫グロブリンをコードする糸状菌を製造する方法であって、前記免疫グロブリンをコードする核酸配列を、改変対象の糸状菌のゲノムに挿入することを含む、免疫グロブリンまたは多量体免疫グロブリンを産生する改変糸状菌を製造する方法を提供する。
本開示の改変糸状菌を製造する方法は、典型的には
改変対象の糸状菌を準備する工程;
単一段階のゲノム改変操作によって、免疫グロブリンをコードする核酸断片を、改変対象の糸状菌のゲノムに挿入する工程;および
前記核酸断片の挿入されたゲノム改変糸状菌が、培養培地中に免疫グロブリンまたは多量体免疫グロブリンを分泌することを確認する工程を含む。
改変対象の糸状菌を準備する工程;
単一段階のゲノム改変操作によって、免疫グロブリンをコードする核酸断片を、改変対象の糸状菌のゲノムに挿入する工程;および
前記核酸断片の挿入されたゲノム改変糸状菌が、培養培地中に免疫グロブリンまたは多量体免疫グロブリンを分泌することを確認する工程を含む。
本開示の改変糸状菌を製造する方法において、単一段階のゲノム改変操作とは、免疫グロブリンをコードする核酸断片を糸状菌に形質導入する操作を2回以上含まないことを意味する。典型的には、単一段階のゲノム改変操作は、免疫グロブリンをコードする核酸断片を糸状菌に形質導入する工程、および、形質導入された糸状菌をネガティブマーカーまたはポジティブマーカを利用して同定する工程を含む。
本開示の改変糸状菌を製造する方法は、単一段階のゲノム改変操作によって、免疫グロブリンをコードする核酸断片を改変対象の糸状菌のゲノムに挿入する工程を含んでいる限り、さらに追加のゲノム改変操作を伴ってもよい。例えば、免疫グロブリンをコードする核酸断片を2コピー以上含む改変糸状菌を製造する場合、単一段階のゲノム改変操作によって、免疫グロブリンをコードする核酸断片の2コピー以上を改変対象の糸状菌のゲノムに挿入する工程に加えて、単一段階のゲノム改変操作によって、免疫グロブリンをコードする核酸断片を1コピー形質導入する工程を伴ってもよい。一態様において、免疫グロブリンの産生に有益な他の任意のゲノム改変操作を伴ってもよい。
一態様において、本開示の改変糸状菌を製造する方法において、ゲノム改変以外の方法で糸状菌の遺伝子構成を修飾することができる。例えば、プラスミドによって異種遺伝子を形質導入してもよい。
本開示の改変糸状菌を製造する方法において、追加のゲノム改変または他の修飾を含む場合、当該ゲノム改変または修飾工程を複数組み合わせて含んでもよく、当該工程は、免疫グロブリンをコードする核酸断片を、改変対象の糸状菌のゲノムに挿入する工程の前、もしくは後あるいはその両方であってもよい。
本開示の改変糸状菌を製造する方法において準備される糸状菌は特に限定されない。例えば、上記(免疫グロブリンまたは多量体免疫グロブリンを製造することを特徴とする改変糸状菌)においてゲノム改変対象として言及した各種糸状菌を用いることができる。
本開示の改変糸状菌を製造する方法において使用するゲノム改変操作は特に限定されない。例えば、上記(免疫グロブリンまたは多量体免疫グロブリンを製造することを特徴とする改変糸状菌)において言及した各種ゲノム改変技術を用いることができる。
本開示の改変糸状菌を製造する方法に含まれるゲノム改変操作に用いられるベクターまたは核酸構築物として、本開示の技術分野で一般的に使用される各種ベクターまたは核酸構築物を使用することができる。例えば、特開2012-179011に開示される選択可能マーカー遺伝子、クローニング部位及び制御領域(プロモーター及びターミネーター)を有するベクターを用いることができる。一態様において、ゲノム改変操作に CRISPR-Casシステムを用いる場合、特開2018-191551に開示される、プラスミドの自律複製を可能とするA. nidulans由来のDNA断片AMA1および高発現により生育を著しく悪化させるAoace2遺伝子を特定の条件で高発現できるよう設計したDNA断片を含むベクターを好適に使用することができる。
本開示の改変糸状菌を製造する方法に含まれる、前記核酸断片の挿入されたゲノム改変糸状菌が、培養培地中に多量体免疫グロブリンを分泌することを確認する工程は、本開示の技術分野で一般的に使用される各種方法を用いて確認できる。例えば、培養培地から分離された免疫グロブリンを、非還元状態のドデシルナトリウム硫酸ポリアクリルアミドゲル電気泳動(SDS-PAGE)にて解析することにより、当該免疫グロブリンが多量体であることを確認できる。
(組成物)
本発明の改変糸状菌で製造される免疫グロブリンまたは多量体免疫グロブリンは、精製したグロブリンを含む組成物として提供される他、免疫グロブリンまたは多量体免疫グロブリンを発現する改変糸状菌自体を含む組成物、免疫グロブリンまたは多量体免疫グロブリンを発現する改変糸状菌の培養液の一部または全体を含む組成物、または、前記改変糸状菌と培養液の一部もしくは全体を含む組成物として提供することができる。本発明の組成物は、好ましくは医薬組成物、食品組成物、飼料組成物などの形態で提供されるが、これらに限定されない。一態様において、本発明の組成物は医薬組成物、食品組成物および飼料組成物は、それぞれ最終的な製品を製造するために用いられる材料組成物を含む。
本発明の改変糸状菌で製造される免疫グロブリンまたは多量体免疫グロブリンは、精製したグロブリンを含む組成物として提供される他、免疫グロブリンまたは多量体免疫グロブリンを発現する改変糸状菌自体を含む組成物、免疫グロブリンまたは多量体免疫グロブリンを発現する改変糸状菌の培養液の一部または全体を含む組成物、または、前記改変糸状菌と培養液の一部もしくは全体を含む組成物として提供することができる。本発明の組成物は、好ましくは医薬組成物、食品組成物、飼料組成物などの形態で提供されるが、これらに限定されない。一態様において、本発明の組成物は医薬組成物、食品組成物および飼料組成物は、それぞれ最終的な製品を製造するために用いられる材料組成物を含む。
(医薬組成物)
本発明に係る医薬組成物は、改変糸状菌で製造される免疫グロブリンまたは多量体免疫グロブリンの効果を利用して、疾患または障害の予防または治療に用いられる。
本発明に係る医薬組成物は、改変糸状菌で製造される免疫グロブリンまたは多量体免疫グロブリンの効果を利用して、疾患または障害の予防または治療に用いられる。
一態様において、本発明に係る医薬組成物は、C.difficile細菌と関連する疾患の治療のために使用される。
C.difficile細菌と関連する疾患とは、特に限定はされないが、例えば、炎症性腸疾患、潰瘍性大腸炎、クローン病、アレルギー、喘息、肥満、自己免疫疾患、新生児壊死性腸炎等が挙げられ、中でも炎症性腸疾患が好ましい。
このような本発明に係る医薬組成物は、本発明に係る改変糸状菌で製造される免疫グロブリンまたは多量体免疫グロブリンの有効量を含んでいればよく、例えば、医薬組成物100重量%中の本発明に係る抗体の含有割合が0.001~99.99重量%の範囲になるように、対象とする疾患の種類、剤型、投与方法、投与対象、投与対象者の症状の度合い、並びに投与によって発揮される効果の程度等を勘案して、適宜設定することができる。
本明細書にて使用する用語「有効量」とは、本発明に係る改変糸状菌で製造される多量体免疫グロブリンが生体において所望の生理効果を奏する量を意味する。
本発明に係る医薬組成物には、本発明に係る本発明に係る改変糸状菌で製造される免疫グロブリンまたは多量体免疫グロブリンと共に、薬学的に許容可能な担体或いは添加物を配合しても良い。薬学的に許容可能な担体或いは添加物とは、任意の担体、希釈剤、賦形剤、懸濁剤、潤滑剤、アジュバント、媒体、送達システム、乳化剤、錠剤分解物質、吸収剤、保存剤、界面活性剤、着色剤、香料、または甘味料を意味し、公知のものを採用すればよい。
この様な投与対象となる個体は、特に限定はされないが、例えばヒト、マウス、ラット、モルモット、ウサギ、ハムスター、イヌ、ネコ、イタチ、ウシ、ブタ等の哺乳類、ニワトリなどの鳥類等が挙げられる。
当該医薬組成物の投与量並びに投与方法は、投与対象となる個体が罹患する腸内疾患の種類、性別、種、年齢、全身状態、疾患の重篤度、所望する効果の程度等に区々ではある。投与量としては、通常は、0.001~100mg/kg/日の範囲で適宜設定すればよい。
また、投与方法については、特に限定はされないが、消化管に直接投与することが好ましく、このような投与方法として、例えば経口投与、経鼻投与、経粘膜投与、経腸投与等が挙げられる。
なお、経腸投与とは、肛門を介した投与には限られず、例えば胃瘻等の様に個体外からチューブ等を消化管に挿入して、それを経由した投与も含まれる。消化管を挿入する位置は腸に限らず、食道、胃、小腸(十二指腸、空腸、回腸等を含む。)、大腸(盲腸、結腸、直腸等を含む。)等が挙げられる。
なお、このような本発明に係る医薬組成物の投与は、上記の量を1日に1度に投与してもよく、数回に分けて投与してもよい。また、上記疾患に対する治療効果を有する範囲において、投与間隔は、毎日、隔日、毎週、隔週、2~3週毎、毎月、隔月または2~3ヶ月毎でもよい。
(食品組成物および飼料組成物)
本発明に係る食品組成物または飼料組成物は、本発明に係る改変糸状菌で製造される免疫グロブリンまたは多量体免疫グロブリンを含む。本発明に係る食品組成物は、一般の食品の他、条件付き特定保健用食品を含む特定保健用食品、栄養補助食品、機能性食品、病者用食品等、食品として摂取されるあらゆる形態を包含する。この様な食品組成物または飼料組成物を使用することによって、本発明に係る改変糸状菌で製造される免疫グロブリンまたは多量体免疫グロブリンの備える生理活性を利用することができる。食品組成物は、整腸用、腸内環境改善用、腸内環境最適化用、腸内腐敗防止用等と表示された食品組成物として提供することができる。
本発明に係る食品組成物または飼料組成物は、本発明に係る改変糸状菌で製造される免疫グロブリンまたは多量体免疫グロブリンを含む。本発明に係る食品組成物は、一般の食品の他、条件付き特定保健用食品を含む特定保健用食品、栄養補助食品、機能性食品、病者用食品等、食品として摂取されるあらゆる形態を包含する。この様な食品組成物または飼料組成物を使用することによって、本発明に係る改変糸状菌で製造される免疫グロブリンまたは多量体免疫グロブリンの備える生理活性を利用することができる。食品組成物は、整腸用、腸内環境改善用、腸内環境最適化用、腸内腐敗防止用等と表示された食品組成物として提供することができる。
この様な食品組成物または飼料組成物における改変糸状菌で製造される免疫グロブリンまたは多量体免疫グロブリンの配合割合は特に限定はされず、食品組成物または飼料組成物の形態、用途等に応じて適宜調整すればよい。通常は組成物の総量に対して、0.001~99重量%程度とすればよい。
本発明に係る食品組成物または飼料組成物の摂取量は特に限定はされず、目的とする効果、その目的とする度合い、並びにその他の諸条件などに応じて設定することができる。例えば、本発明に係る免疫グロブリンまたは多量体免疫グロブリンの量に換算して、通常は0.001~100mg/kg/日程度とすればよく、これを1日1回~数回に分けて摂取してもよい。
上述の食品組成物の具体的な形態は特に限定はされないが、例えば、清涼飲料、炭酸飲料、栄養飲料、果実飲料、乳酸飲料、乳飲料等の飲料;アイスクリーム、アイスシャーベット、かき氷等の冷菓;飴、キャンディー、ガム、チョコレート、錠菓、スナック菓子、ビスケット、ゼリー、ジャム、クリーム、焼き菓子等の菓子類;そば、うどん、はるさめ、中華麺、即席麺等の麺類;かまぼこ、ハム、ソーセージ等の水産・畜産加工食品;加工乳、発酵乳等の乳製品;サラダ油、てんぷら油、マーガリン、マヨネーズ、ショートニング、ホイップクリーム、ドレッシング等の油脂及び油脂加工食品;ソース、たれ等の調味料;スープ、シチュー、サラダ、惣菜、ふりかけ、漬物、パン、シリアル等を例示できる。また、特定保健用食品、栄養補助食品、機能性食品等の場合であれば、粉末、顆粒、カプセル、トローチ、タブレット、シロップ等の形態が挙げられる。
本発明に係る食品組成物または飼料組成物は、精製したグロブリンを含む組成物として提供される他、免疫グロブリンまたは多量体免疫グロブリンを発現する改変糸状菌自体を含む組成物、免疫グロブリンまたは多量体免疫グロブリンを発現する改変糸状菌の培養液の一部または全体を含む組成物、または、前記改変糸状菌と培養液の一部もしくは全体を含む組成物として提供することができる。一態様において、本発明の食品組成物または飼料組成物は、糸状菌の培養過程、好ましくは発酵過程を経て製造される組成物として提供される。このような場合、培養物あるいは醸造発酵物をそのまま食品組成物としてもよく、醸造発酵物の一部を精製したものを食品組成物としてもよい。例えば、培養物あるいは醸造発酵物から糸状菌を含む固形成分をろ過して液体組成物として提供することができる。このような組成物は、糸状菌自体、糸状菌の一部、糸状菌の抽出物、糸状菌の培地となる成分、あるいはこれらの混合物を含むことができる。このような食品組成物の非限定的な例は、みりん、酒、甘酒、味噌、醤油、塩麹、米麹、醤油麹、麦麹、豆麹、麹納豆、酒粕魚麹:魚醤のもと、ラギ(インドネシア)、ケチャップ(インドネシアの味噌、トマトは一切なし)、醤:ひしお酒醸、醪糟、餅麹、撒麹、チョングッチャン、鰹節およびその他の雑節等を含む。
本発明の飼料組成物は、各種動物に対する整腸用、腸内環境改善用、腸内環境最適化用、腸内腐敗防止用等と表示された飼料組成物として提供することができる。
上述の飼料組成物の具体的な形態は特に限定はされないが、例えば上述した本発明に係る飼料組成物が発揮する効果が損なわれない限りにおいて、通常の飼料に混合、又は必要に応じて通常の飼料に配合可能な成分と共に混合して飼料組成物とすればよく、飼料組成物そのものを飼料としてもよい。例えば、各種糸状菌発酵食品の製造に用いた搾りかす配合して飼料とすることができる。
以下、実施例により本発明をさらに詳細に説明するが、これらは例示に過ぎず本開示を限定するものではない。
方法と材料
1.DNA操作方法
DNA操作には大腸菌DH5αを使用した。PCRには、PrimeSTARHS DNAポリメラーゼ(TaKaRa)を使用した。In-Fusion HDクローニングキット(TaKaRa)をプラスミド作製に使用した。プラスミドはアルカリSDS法により抽出した。塩基配列は、ファスマック株式会社での受託により解析を行った。
1.DNA操作方法

2.wA遺伝子への遺伝子導入のためのゲノム編集プラスミドの作製
wA遺伝子の上流を標的として二本鎖DNA切断を行うsgRNAを発現するゲノム編集プラスミドpRGE-gwAup(図2)は、Katayama et al.[Appl. Environ. Microbiol.,85:e01896-18, 2019]に記載のとおり作製した。sgRNAコード配列とU6ターミネーターを含むDNA断片を、SmaIで切断したppAsATC9a2と接続し、pRGE-gRT6を作製した。U6プロモーターは、SmaIで切断したpRGE-gRT6と接続して、pRGE-gwAupを作製した。
wA遺伝子の上流を標的として二本鎖DNA切断を行うsgRNAを発現するゲノム編集プラスミドpRGE-gwAup(図2)は、Katayama et al.[Appl. Environ. Microbiol.,85:e01896-18, 2019]に記載のとおり作製した。sgRNAコード配列とU6ターミネーターを含むDNA断片を、SmaIで切断したppAsATC9a2と接続し、pRGE-gRT6を作製した。U6プロモーターは、SmaIで切断したpRGE-gRT6と接続して、pRGE-gwAupを作製した。
3.プラスミドの作製方法
2量体IgAの生産株取得の形質転換のために、ドナーDNAプラスミドpwAup-DN H+L+J(co)(図1)を以下のとおり作製した。
最初に、プラスミドpwAup-DN H+LlinkerSmaI(co)を構築した。amyBプロモーター、AmyBのシグナル配列とW27 IgAのH鎖のコード領域からなるDNA断片を増幅した。amyBターミネーター、amyBプロモーター、AmyBのシグナル配列とW27 IgAのL鎖のコード領域、およびamyBターミネーターからなるDNA断片を増幅し、同時にリンカー配列とSmaI切断部位もamyBターミネーターの3’末端に導入した。これらの2つのDNA断片を、In-Fusion HDクローニングキットを使用してSmaIで切断したプラスミドpwAup-DNと接続し、pwAup-DN H+LlinkerSmaI(co)を作製した。
2量体IgAの生産株取得の形質転換のために、ドナーDNAプラスミドpwAup-DN H+L+J(co)(図1)を以下のとおり作製した。
最初に、プラスミドpwAup-DN H+LlinkerSmaI(co)を構築した。amyBプロモーター、AmyBのシグナル配列とW27 IgAのH鎖のコード領域からなるDNA断片を増幅した。amyBターミネーター、amyBプロモーター、AmyBのシグナル配列とW27 IgAのL鎖のコード領域、およびamyBターミネーターからなるDNA断片を増幅し、同時にリンカー配列とSmaI切断部位もamyBターミネーターの3’末端に導入した。これらの2つのDNA断片を、In-Fusion HDクローニングキットを使用してSmaIで切断したプラスミドpwAup-DNと接続し、pwAup-DN H+LlinkerSmaI(co)を作製した。
続いて、プラスミドpwAup-DN H+L+J(co)を構築した。amyBプロモーター、AmyBのシグナル配列とW27 IgAのJ鎖のコード領域、およびamyBターミネーターからなるDNA断片を増幅した。このDNAを、In-Fusion HDクローニングキットを使用してSmaIで切断したプラスミドpwAup-DN H+LlinkerSmaI(co)と接続し、pwAup-DN H+L+J(co)を作製した。
5量体IgA発現DNAの生産株取得の形質転換のために、ドナーDNAプラスミドpwAup-DN HAAComp+L(co)(図10)を、以下のとおり作製した。
AmyBのシグナル配列とW27 IgAのH鎖およびC末側のリンカーのコード領域を増幅した。リンカー配列の一部、COMPドメイン、amyBターミネーター、amyBプロモーター、AmyBのシグナル配列およびW27 IgAのL鎖のコード領域からなるDNA断片を増幅した。これらのDNA断片の増幅の際に、H鎖とCOMPドメインをつなぐリンカーにもともと含まれていたリジン残基をアラニンに置換した。これら2つのDNA断片を、In-Fusion HDクローニングキットを使用してSmaIで切断したプラスミドpwAupADNと接続し、pwAup-DN HAAComp+L(
co)を作製した。

4.A.oryzaeの形質転換によるIgA生産株の取得
2量体IgAの生産株取得のための形質転換には、ドナーDNAプラスミドpwAup-DN H+L+J(co)(図1)およびゲノム編集プラスミドpRGE-gwAup(図2)を用いた。5量体IgAの生産株取得のための形質転換には、ドナーDNAプラスミドpwAup-DN HAAComp+L(co)(図10およびゲノム編集プラスミドpRGE-gwAup(図2)を用いた。形質転換の宿主として、プロテアーゼ遺伝子10重破壊株NSlD1-ΔP10 [Yoon et al. Appl Microbiol Biotechnol. 89:747-759, 2011]を使用した。A.oryzaeの形質転換は、既報[Maruyama et al., Methods Mol Biol 765:447-456,2011]の方法に従い行った。

5.W27 IgAの生産と硫安沈殿
IgA抗体を生産するために、5×DPY培地(10% Dextrin, 5% Hipolypeptone, 2.5% Yeast extract, 0.5% K2HPO4,0.05% MgSO4・7H2O,pH 8.0)、またはDPY培地(2% Dextrin,1% Hipolypeptone,0.5% Yeast extract,0.5% K2HPO4、0.05% MgSO4・7H2O,pH 8.0)を使用した。IgA生産株の分生子を100mLあたり1×107個となるように液体培地に植菌して、30℃で8日間150rpmで振とう培養を行った。Miracloth(Merck Millipore)でろ過することにより培養上清を回収した。形質転換体の培養液へのIgA生産量は、ELISAアッセイまたはウエスタン解析により解析した。

6.生産したIgAの硫安沈殿による濃縮
生産したIgAの硫安沈殿による濃縮は以下のとおり行った。Miracloth(Merck Millipore)を用いたろ過により得た培養上清について、5×DPY培地による培養では10000×g,4℃,10分間、DPY培地を使用した場合は3,500 rpm(2,150×g),4℃,10分間で遠心して、その上清を回収した。1/10容量の10×PBSバッファーを加えてpHを調整し、5×DPY 培地による培養ではさらにPBSバッファーで5倍に希釈した。1 Lあたり390g(NH4)2SO4(FUJIFILM Wako Chemicals)を添加して60%飽和で、混合物を穏やかに攪拌しながら4℃で一晩インキュベートした。その後、10,000×g,4℃,20分間遠心して沈殿を得て、少量の60%飽和(NH4)2SO4溶液を加えて10,000×g,4℃,10分間遠心により洗浄した。沈殿をPBSバッファーで溶解し、14,000MWCO透析膜(和光純薬工業)を使用してPBSバッファーで24時間透析した。途中でPBSバッファーを2回交換した。透析後、5×DPY培地による培養では15,000rpm(20,620×g),4℃,10分間、DPY培地を使用した場合は3,500rpm(2,150×g),4℃,10分間遠心し、その上清を3.0μm、0.45μm、0.2μmのフィルターで順次ろ過した。
生産したIgAの硫安沈殿による濃縮は以下のとおり行った。Miracloth(Merck Millipore)を用いたろ過により得た培養上清について、5×DPY培地による培養では10000×g,4℃,10分間、DPY培地を使用した場合は3,500 rpm(2,150×g),4℃,10分間で遠心して、その上清を回収した。1/10容量の10×PBSバッファーを加えてpHを調整し、5×DPY 培地による培養ではさらにPBSバッファーで5倍に希釈した。1 Lあたり390g(NH4)2SO4(FUJIFILM Wako Chemicals)を添加して60%飽和で、混合物を穏やかに攪拌しながら4℃で一晩インキュベートした。その後、10,000×g,4℃,20分間遠心して沈殿を得て、少量の60%飽和(NH4)2SO4溶液を加えて10,000×g,4℃,10分間遠心により洗浄した。沈殿をPBSバッファーで溶解し、14,000MWCO透析膜(和光純薬工業)を使用してPBSバッファーで24時間透析した。途中でPBSバッファーを2回交換した。透析後、5×DPY培地による培養では15,000rpm(20,620×g),4℃,10分間、DPY培地を使用した場合は3,500rpm(2,150×g),4℃,10分間遠心し、その上清を3.0μm、0.45μm、0.2μmのフィルターで順次ろ過した。
7.W27 IgAのウエスタン解析
生産したIgAが含まれるサンプルのウエスタン解析は、以下の抗体を使用して行った。
H鎖の検出には、抗H鎖抗体Goat Anti-Mouse IgA-UNLB(Cat.No. 1040-01,SouthernBiotech)、および二次抗体Rabbit anti-goat IgG(H+L)/(H&L)antibody,HRP conjugate(Cat.No.SA00001-4,Proteintech Group,Inc.)を使用した。
L鎖の検出には、抗L鎖抗体Goat Anti-Mouse Kappa-UNLB(Cat.No.1050-01,SouthernBiotech)、および二次抗体Rabbit anti-goat IgG(H+L)/(H&L)antibody,HRPconjugate(Cat.No.SA00001-4,Proteintech Group,Inc.)を使用した。
J鎖の検出には、抗J鎖抗体Ig J chain(M-159)(Cat.No.sc-66929,Santa Curz Biotechnology)、および二次抗体Anti-rabbit IgG, HRP-linked antibody(Cat.No.7074,Cell Signaling Technology)を使用した。

8.モノクローナルIgA抗体の定量と非還元SDS-PAGE
精製した抗体の定量は、ELISA法で行った。抗体の定量方法の詳細は以下の通りである。まず、Anti-goat-mouse IgA(Southern Biotech:1040-01)を2μg/mlになるように0.05M Na2CO3で希釈して、C96MaxiSorp Nunc-Immuno Plate(Thermo Fisher)(以下ELISAプレートとする)の各wellに50μl/wellずつ加え、一晩4℃で固相化させた。Vaccu-Pette/96multiwell pipetter(Sigma-Aldrich)(以下プレートウォッシャー)を用いて、PBSによりプレートを3回洗浄後、1% bovine serum albumin (BSA)(Wako)を含むPBS(PBS-BSA)を150μl/wellとなるように加えて、4℃で一晩静置してブロッキングを行った。抗体液の系列を96穴プレートで作成した。抗体液の希釈系列は、等倍(x1、精製抗体のx1/100とx1/1000をx1とする)、3倍(x1/3)、10倍(x1/10)、30倍(x1/30)、100倍(x1/100)、300倍(x1/300)、1000倍(x1/1000)、3000倍(x1/3000)で、PBS-BSAを用いて段階希釈液を作成した。プレートからPBS-BSAを完全に取り除いた後、段階希釈液を50μl/wellとなるように加えて室温で1時間静置して反応させた。
次に0.05%Tween 20(Chem Cruz)を含むPBS(PBS-T)によりプレートウォッシャーで3回プレートを洗浄した後、2次抗体Alkaline Phosphatase(ALP)-conjugatedanti-goat mouseIgA (Southern Biotech: 1040-04)をPBS-BSAに、終濃度0.5 μg/mlとなるように希釈し、50μl/wellで加え、室温で1時間静置して反応させた。PBS-Tによりプレートウォッシャーで3回プレートを洗浄後、5mlの炭酸バッファー(6.5mM Na2CO3、18.5mM NaHCO3)に1M MgCl2 10μlとSubstrate for Alkaline Phosphatase(Sigma-Aldrich)1錠を加えた基質溶液を50μl/wellで加えた。室温で遮光して静置後、十分に発色していることを目視で確認して、TriStar2 LB942(BERTHOLD TECHNOLOGIES)を使用してoptical density(O.D.)405nmの吸光度を測定した。

次に0.05%Tween 20(Chem Cruz)を含むPBS(PBS-T)によりプレートウォッシャーで3回プレートを洗浄した後、2次抗体Alkaline Phosphatase(ALP)-conjugatedanti-goat mouseIgA (Southern Biotech: 1040-04)をPBS-BSAに、終濃度0.5 μg/mlとなるように希釈し、50μl/wellで加え、室温で1時間静置して反応させた。PBS-Tによりプレートウォッシャーで3回プレートを洗浄後、5mlの炭酸バッファー(6.5mM Na2CO3、18.5mM NaHCO3)に1M MgCl2 10μlとSubstrate for Alkaline Phosphatase(Sigma-Aldrich)1錠を加えた基質溶液を50μl/wellで加えた。室温で遮光して静置後、十分に発色していることを目視で確認して、TriStar2 LB942(BERTHOLD TECHNOLOGIES)を使用してoptical density(O.D.)405nmの吸光度を測定した。
上記とは別に2量体抗体が産生されているかどうかを非還元SDS-PAGEによって確認した。サンプル調製は、抗体(5μg)に3×Native buffer(0.2M Tris-HCl pH6.8、30% Glycerol、0.015% Bromophenol blue)を加えた。WIDE RANGE Gel Preparation Buffer(4×)forPAGE(Nacalai Tesque)を用いた6%のSDS-PAGE用ゲルを用いて泳動後、Coomassie brilliant blue(CBB)染色を行った。還元SDS-PAGEは2-ME添加のbufferをサンプルに加えたのちに95℃で10分間変性後に非還元と同じ6%のSDS-PAGE用ゲルを用いて泳動後、Coomassie brilliant blue(CBB)染色を行った。
9.大腸菌に対する精製IgA抗体の結合力の解析
大腸菌に対する精製IgA抗体の結合力を比較するために、大腸菌に対するELISAを行った。大腸菌(DH5αもしくはBW38092及びその変異体、いずれもK12株)について37℃好気培養(LB培地)を一晩行い、遠心分離により集めた。PBSで菌体を洗浄後0.05M Na2CO3バッファーに細菌を懸濁し、ELISAプレートにコーティングした(4℃一晩)。上記の精製IgA抗体定量ELISAと同様に1%BSA添加PBSによるブロッキング後に、96穴プレート中の各抗体希釈系列を作製して加え、室温で約1時間反応させた。その後、プレートを0.05%Tween 20添加PBSで洗浄し、2次抗体Alkaline Phosphatase(ALP)-conjugated anti-goat mouse IgA(Southern Biotech: 1040-04)をPBS-BSAに、終濃度0.5μg/mlとなるように希釈し、50μl/wellで加え、室温で1時間静置して反応させた。PBS-Tによりプレートウォッシャーで3回プレートを洗浄後、Alkali Phosphatase tablet(Sigma)を使用して上記と同様に発色反応を行った。十分な反応のために、発色後のELISAプレートを4度で一晩反応させ、TriStar2 LB942(BERTHOLD TECHNOLOGIES)を使用し、OD405 nmを測定した。

10.大腸菌およびC. difficile菌増殖抑制試験(嫌気)
大腸菌をLB培地(一晩37℃)で嫌気培養した。以後のすべての操作は嫌気チャンバー内で行った。培養液を遠心分離し、細菌を集菌後、LB培地で洗浄を行った。細菌液を培養液で10倍に希釈後に、SYTO24(Invitrogen)とCell Viability Kit(Becton Dickinson)に含まれるCounting beadsを加え、フローサイトメトリー(測定は好気下)によりSYTO24陽性の生細菌数をCounting beadsとの比較から算出した。LB培地で細菌が300cells/5μlになるように希釈した。エッペンチューブに細菌希釈液5μl加え、さらに嫌気化したPBS、または嫌気化した精製した抗体を25μl加え、さらにLB培地を30μl加え37℃嫌気で静置培養した(抗体最終濃度は0.42mg/ml)。その後、20時間に細菌液を希釈してLB plateに播種し、一晩37℃で嫌気培養した。次の日にコロニー数を計測して生細菌数を測定した。
上記大腸菌に対する試験と同様に、C. difficile 10,000細胞を、2量体IgA発現化改変麹株(KojiHLJ#1)から得られた多量体W27 IgA抗体(417mg/ml)とともに37度で20時間嫌気培養した後、大腸菌用培地プレート(LB plate)に播種し、コロニー数を計測した。

実施例1:2量体IgAの生産株の作製
A.oryzaeにおいて、W27 IgA抗体を2量体で生産することを試みた。IgAの2量体は、H鎖、L鎖、およびJ鎖によって構成される。それぞれをコードする遺伝子について、A.oryzaeでもっとも発現量が高い遺伝子の1つtef1のコドン使用頻度をもとにして、コドン改変を行った。これらに対して、A.oryzaeでもっとも分泌量が多いα-アミラーゼAmyBのシグナル配列をコードする配列を連結して、翻訳後に小胞体に移行して分泌されるように設計した。プロモーターとターミネーターは、amyB遺伝子のものを使用した(図1)。
A.oryzaeにおいて、W27 IgA抗体を2量体で生産することを試みた。IgAの2量体は、H鎖、L鎖、およびJ鎖によって構成される。それぞれをコードする遺伝子について、A.oryzaeでもっとも発現量が高い遺伝子の1つtef1のコドン使用頻度をもとにして、コドン改変を行った。これらに対して、A.oryzaeでもっとも分泌量が多いα-アミラーゼAmyBのシグナル配列をコードする配列を連結して、翻訳後に小胞体に移行して分泌されるように設計した。プロモーターとターミネーターは、amyB遺伝子のものを使用した(図1)。
遺伝子導入には、A.oryzaeにおけるゲノム編集技術CRISPR/Cas9システムによる遺伝子改変技術[Katayama et al., Appl. Environ. Microbiol., 85:e01896-18, 2019]を利用した。IgA抗体として、国際公開2014/142084号に記載されるW27抗体および特願2021-110421情報に基づいて、W27抗体のCHO細胞産生リコンビナントIgAであって、軽鎖にプロテインL結合のための変異を導入したRS_H000_L001を用いた。RS_H000_L001の重鎖可変領域および軽鎖可変領域のアミノ酸配列を以下に示す。以下、実施例1~4において、RS_H000_L001を「W27 IgA」と記載する。
なお、上記配列はマウス由来のシグナル配列を含んでいるが、本実施例において糸状菌のゲノム改変に使用したW27 IgAは、以下に示すように、α-アミラーゼAmyBのシグナル配列を含んでいる。
W27 IgA抗体のH鎖、L鎖、およびJ鎖を発現するカセットをタンデムに接続した。このような発現カセットを、色素合成遺伝子wAの上流領域とORF内部領域の内側に位置することで、wA遺伝子座に相同組換えで挿入されるように設計して、ドナーDNAプラスミドpwAup-DN H+L+J(co)を作製した(図1)。
wA遺伝子上流を標的として二本鎖DNA切断を行うsgRNAを発現するために、ゲノム編集プラスミドpRGE-gwAup [Katayama et al., Appl. Environ. Microbiol., 85:e01896-18, 2019]を使用した(図2)。
A.oryzaeの形質転換の宿主として、プロテアーゼ遺伝子10重破壊株であるNSlD1-ΔP10 [Yoon et al., Appl. Microbiol. Biotechnol., 89:747-759, 2011]を使用した。この株を使用することで、ヒト由来リゾチームおよびウシ由来キモシン[Yoon et al., Appl. Microbiol. Biotechnol., 89:747-759, 2011]、ヒト由来IgG抗体[Huynh et al., Fungal Biol. Biotechnol.,7:7,2020]について生産量が増加することが示されている。
なお、上記配列はマウス由来のシグナル配列を含んでいるが、本実施例において糸状菌のゲノム改変に使用したW27 IgAは、以下に示すように、α-アミラーゼAmyBのシグナル配列を含んでいる。

前述の2量体IgA発現用のドナーDNAプラスミドpwAup-DN H+L+J(co)、およびゲノム編集プラスミドpRGE-gwAupを用いて、NSlD1-ΔP10株を形質転換した。この際、既報[Katayama et al., Appl. Environ. Microbiol., 85:e01896-18, 2019]にもとづき、ゲノム編集プラスミドに対するドナーDNAプラスミドの使用量を増やすことで、発現カセットが多コピー導入された形質転換体が取得されやすいようにした。取得した形質転換株について、2量体IgAの発現カセットのコピー数と生産量を解析して、コピー数が多く生産量が高かった株を「H+L+J(co) #1」と命名し、以降の生産実験に使用した。
2量体IgA生産株「H+L+J(co) #1」を液体培地に植菌し、8日間培養ののち培養上清を回収して、硫安沈殿により約10倍に濃縮した。ウエスタン解析を行った結果、H鎖、L鎖およびJ鎖は培養上清および濃縮サンプルで検出され、それぞれ培養液への生産が確認された(図3)。
プラスミドpwAup-DN H+L+J(co)に含まれるW27 IgA H(重鎖)、W27 IgA L(軽鎖)およびW27 IgA J(J鎖)の塩基配列およびアミノ酸配列を、図4~9に示す。
実施例2:5量体IgAの生産株の作製
W27 IgA抗体を5量体で生産するために、COMPドメインを使用した。COMP (cartilage oligomeric matrix protein)ドメインは、軟骨オリゴマーマトリックスタンパク質由来のコイルドコイルドメインであり、5量体形成の誘導に使用されている。
遺伝子導入には、A. oryzaeにおけるゲノム編集技術CRISPR/Cas9システムによる遺伝子改変技術[Katayama et al., Appl. Environ. Microbiol., 85:e01896-18, 2019]を利用した。W27 IgA抗体のH鎖のC末側にCOMPドメインを融合して、L鎖とともに発現するためのカセットをタンデムに接続した。この際、H鎖とCOMPドメインをつなぐリンカーにもともと含まれていたリジン残基をアラニンに置換した。プロモーターとターミネーターは、amyB遺伝子のものを使用したこのような発現カセットを、色素合成遺伝子wAの上流領域とORF内部領域の内側に位置することで、wA遺伝子座に相同組換えで挿入されるように設計して、ドナーDNAプラスミドpwAup-DN HAAComp+L(co)を作製した(図10)。wA遺伝子上流を標的として二本鎖DNA切断を行うsgRNAを発現するために、ゲノム編集プラスミドpRGE-gwAup [Katayama et al., Appl. Environ. Microbiol., 85:e01896-18, 2019]を使用した。
W27 IgA抗体を5量体で生産するために、COMPドメインを使用した。COMP (cartilage oligomeric matrix protein)ドメインは、軟骨オリゴマーマトリックスタンパク質由来のコイルドコイルドメインであり、5量体形成の誘導に使用されている。

Environ. Microbiol., 85:e01896-18, 2019]を使用した。
作製したドナーDNAプラスミドpwAup-DN HAAComp+L(co)とゲノム編集プラスミドpRGE-gwAupを用いて、NSlD1-ΔP10株を形質転換した。この際、既報[Katayama et al., Appl. Environ. Microbiol., 85:e01896-18, 2019]にもとづき、ゲノム編集プラスミドに対するドナーDNAプラスミドの使用量を増やすことで、発現カセットが多コピー導入された形質転換体が取得されやすいようにした。取得した形質転換株について、5量体IgAの発現カセットのコピー数と生産量を解析して、コピー数が多く生産量が高かった株を「HAAComp+L(co) #1」と命名し、以降の生産実験に使用した。
5量体IgA生産株「HAAComp+L(co) #1」を液体培地に植菌し、8日間培養ののち培養上清を回収して、硫安沈殿により約10倍に濃縮した。ウエスタン解析を行った結果、H鎖およびL鎖は培養上清および濃縮サンプルで検出され、それぞれ培養液への生産が確認された(図11)。
プラスミドpwAup-DN HAAComp+L(co)に含まれるW27 IgA H(重鎖)およびW27 IgA L(軽鎖)の塩基配列およびアミノ酸配列を、図12~15に示す。
実施例3:IgA融合タンパク質の構成の影響
IgAのH鎖のC末端にCOMP配列を追加した融合タンパク質について、IgAのH鎖C末端領域の構成およびリンカーの構成が多量体形成に与える影響を検討した。
天然のL鎖および以下の構成を有する融合タンパク質をH鎖とするIgAをコードする核酸構築物を用いてゲノム改変CHO細胞を作製し、分泌されたIgAの多量体形成状態について検討した。
IgAのH鎖のC末端にCOMP配列を追加した融合タンパク質について、IgAのH鎖C末端領域の構成およびリンカーの構成が多量体形成に与える影響を検討した。
天然のL鎖および以下の構成を有する融合タンパク質をH鎖とするIgAをコードする核酸構築物を用いてゲノム改変CHO細胞を作製し、分泌されたIgAの多量体形成状態について検討した。
図16に示すように、IgAのH鎖C末端領域の構成およびリンカーの構成はIgAの多量体形成に影響し得ることが確認された。特に、リンカーを用いずにCOMP配列を結合したH鎖を用いた場合、特に、IgAのテールピース領域を一部削除してCOMP配列を結合したH鎖を用いた場合に、より多量体として産生されるIgAの割合が多くなることが観察された。
実施例4:ゲノム改変糸状菌で製造された多量体免疫グロブリンの結合特性
麹菌及びCHO細胞から分泌された多量体化されたW27 IgAについて、大腸菌に対する結合力を確認した。なお、国際公開2014/142084号に記載するように、哺乳類細胞で発現したW27 IgA抗体は大腸菌に結合することが知られている。麹菌及びCHO細胞から分泌された多量体化されたW27 IgAを精製して、これらの多量体IgAによる大腸菌結合能を試験した。使用した多量体IgAを還元状態(左)および非還元状態でSDS-PAGEにかけ、CBB染色した結果を図17、18に示す。非還元状態のSDS-PAGEの結果に示されるように、2量体IgA発現化改変麹株(Koji HLJ#1)(図17)および5量体IgA発現化改変麹株(Koji HAA COMP+L#1)(図18)のいずれからも、多量体IgAの発現が確認された。なお、麹菌で産生したIgA抗体は非還元状態のSDS-PAGE上でバンドが広がる傾向がみられた。
大腸菌に対する結合力を測定した結果、図19~20に示すように、麹菌から分泌された多量体IgA抗体は、CHO細胞から分泌された多量体IgA抗体と同様に、SHMTタンパク質を発現する大腸菌に対して結合することが確認された。
より具体的には、2量体IgA発現化改変麹株「H+L+J(co) #1」から取得した多量体IgA(KojiHLJ#1)について上記大腸菌BW38029株との結合を確認した試験結果を図19に示す。
同様に、5量体IgA発現化改変麹株「HAAComp+L(co) #1」から取得した多量体抗体(Koji HAACOMP#1)について大腸菌BW38029株との結合を確認した試験結果を図20に示す。比較のために、IgA重鎖のC末端テールピースを一部欠損した状態でCOMP配列を追加した5量体IgA発現化改変CHO細胞から取得した多量体IgA(CHO W27notailCOMP)、2量体IgA発現化改変CHO細胞で発現した多量体IgA(CHO W27dimer)、2量体IgA発現化改変麹株「H+L+J(co) #1」から発現した多量体IgA(2回の別試行により得たKoji HLJ#1-No.1およびKoji HLJ#1-No.2を用いた)、並びに陰性対照を用いた(図20)。
図20に示すように、5量体IgA発現化改変麹株「HAAComp+L(co) #1」から取得した多量体抗体は、2量体IgA発現化改変麹株「H+L+J(co) #1」から取得した多量体抗体に比して、より強い結合性を有することが確認された。
麹菌及びCHO細胞から分泌された多量体化されたW27 IgAについて、大腸菌に対する結合力を確認した。なお、国際公開2014/142084号に記載するように、哺乳類細胞で発現したW27 IgA抗体は大腸菌に結合することが知られている。麹菌及びCHO細胞から分泌された多量体化されたW27 IgAを精製して、これらの多量体IgAによる大腸菌結合能を試験した。使用した多量体IgAを還元状態(左)および非還元状態でSDS-PAGEにかけ、CBB染色した結果を図17、18に示す。非還元状態のSDS-PAGEの結果に示されるように、2量体IgA発現化改変麹株(Koji HLJ#1)(図17)および5量体IgA発現化改変麹株(Koji HAA COMP+L#1)(図18)のいずれからも、多量体IgAの発現が確認された。なお、麹菌で産生したIgA抗体は非還元状態のSDS-PAGE上でバンドが広がる傾向がみられた。
大腸菌に対する結合力を測定した結果、図19~20に示すように、麹菌から分泌された多量体IgA抗体は、CHO細胞から分泌された多量体IgA抗体と同様に、SHMTタンパク質を発現する大腸菌に対して結合することが確認された。
より具体的には、2量体IgA発現化改変麹株「H+L+J(co) #1」から取得した多量体IgA(KojiHLJ#1)について上記大腸菌BW38029株との結合を確認した試験結果を図19に示す。
同様に、5量体IgA発現化改変麹株「HAAComp+L(co) #1」から取得した多量体抗体(Koji HAACOMP#1)について大腸菌BW38029株との結合を確認した試験結果を図20に示す。比較のために、IgA重鎖のC末端テールピースを一部欠損した状態でCOMP配列を追加した5量体IgA発現化改変CHO細胞から取得した多量体IgA(CHO W27notailCOMP)、2量体IgA発現化改変CHO細胞で発現した多量体IgA(CHO W27dimer)、2量体IgA発現化改変麹株「H+L+J(co) #1」から発現した多量体IgA(2回の別試行により得たKoji HLJ#1-No.1およびKoji HLJ#1-No.2を用いた)、並びに陰性対照を用いた(図20)。
図20に示すように、5量体IgA発現化改変麹株「HAAComp+L(co) #1」から取得した多量体抗体は、2量体IgA発現化改変麹株「H+L+J(co) #1」から取得した多量体抗体に比して、より強い結合性を有することが確認された。
実施例5:ゲノム改変糸状菌で製造された多量体免疫グロブリンの生理活性
(5-1)ゲノム改変糸状菌で製造された多量体IgAについて、SHMTを発現する大腸菌に対する増殖抑制効果を検証した。
具体的には、大腸菌(BW38029株にSHMT VenusHis融合遺伝子をノックインしたもの)300細胞を、2量体IgA発現化改変麹株(KojiHLJ#1)から得られた多量体抗体(417mg/ml)とともに37度で20時間嫌気培養した後、大腸菌用培地プレート(LB plate)に播種し、コロニー数を計測した(n=3)。結果を図21に示す。比較として、抗体を含まない陰性対照、2量体IgA発現化改変CHO細胞で発現した多量体IgA、およびCHO細胞で発現した W27 IgG抗体を用いた。
2量体IgA発現化改変麹株(KojiHLJ#1)から取得した多量体W27IgAは、2量体IgA発現化改変CHO細胞で発現した多量体W27 IgAと同様に、SHMTを発現する大腸菌に対して増殖抑制効果を示した。一方、CHO細胞で発現したW27 IgG抗体は、このような増殖抑制効果をほとんど示さなかった。
上記データが示すように、多量体IgA発現化改変麹株から分泌された多量体IgA抗体は、CHO細胞から分泌された多量体IgA抗体と同様に、大腸菌に対して増殖抑制効果を示すことが確認された。
(5-1)ゲノム改変糸状菌で製造された多量体IgAについて、SHMTを発現する大腸菌に対する増殖抑制効果を検証した。

上記データが示すように、多量体IgA発現化改変麹株から分泌された多量体IgA抗体は、CHO細胞から分泌された多量体IgA抗体と同様に、大腸菌に対して増殖抑制効果を示すことが確認された。
(5-2)ゲノム改変糸状菌で製造された多量体IgAについて、C. difficileに対する増殖抑制試験を行った。
具体的には、上記大腸菌に対する試験と同様に、C. difficile 10,000細胞を、2量体IgA発現化改変麹株(KojiHLJ#1)から得られた多量体W27 IgA抗体(417mg/ml)とともに37度で20時間嫌気培養した後、大腸菌用培地プレート(LB plate)に播種し、コロニー数を計測した(n=3)。結果を図22に示す。比較として、抗体を含まない陰性対照、2量体IgA発現化改変CHO細胞で発現した多量体W27 IgA、およびCHO細胞で発現したW27 IgG抗体を用いた。 麹菌から得られた2量体IgA抗体(KojiHLJ#1)は、2量体IgA発現化改変CHO細胞で発現した多量体W27 IgAと同様に、C. difficileに対して増殖抑制効果を示した。一方、CHO細胞で発現したW27IgG抗体は、このような増殖抑制効果をほとんど示さなかった。
上記データが示すように、麹菌から分泌されたIgA抗体は、CHO細胞から分泌されたIgA抗体と同様に、C. difficileに対して増殖抑制効果を示すことが確認された。
具体的には、上記大腸菌に対する試験と同様に、C. difficile 10,000細胞を、2量体IgA発現化改変麹株(KojiHLJ#1)から得られた多量体W27 IgA抗体(417mg/ml)とともに37度で20時間嫌気培養した後、大腸菌用培地プレート(LB plate)に播種し、コロニー数を計測した(n=3)。結果を図22に示す。比較として、抗体を含まない陰性対照、2量体IgA発現化改変CHO細胞で発現した多量体W27 IgA、およびCHO細胞で発現したW27 IgG抗体を用いた。 麹菌から得られた2量体IgA抗体(KojiHLJ#1)は、2量体IgA発現化改変CHO細胞で発現した多量体W27 IgAと同様に、C. difficileに対して増殖抑制効果を示した。一方、CHO細胞で発現したW27IgG抗体は、このような増殖抑制効果をほとんど示さなかった。
上記データが示すように、麹菌から分泌されたIgA抗体は、CHO細胞から分泌されたIgA抗体と同様に、C. difficileに対して増殖抑制効果を示すことが確認された。
以上の結果から、多量体IgA発現化改変麹株から分泌された多量体IgA抗体は、CHO細胞から分泌された多量体IgA抗体と同様の、優れた生理活性を備えていることが確認された。
Claims (13)
- ゲノム中に免疫グロブリンをコードする核酸断片を含む糸状菌を準備する工程;
前記糸状菌を培養する工程;および
培養培地中の多量体免疫グロブリンを他の培養培地成分から分離する工程を含む、多量体免疫グロブリンを製造する方法。 - 前記糸状菌がゲノム中に免疫グロブリンをコードする核酸断片を2コピー以上含有することを特徴とする、請求項1に記載の方法。
- 前記2コピー以上の免疫グロブリンをコードする核酸断片が、糸状菌ゲノムの単一領域に挿入されている、請求項1または2に記載の方法。
- 前記免疫グロブリンが、細菌増殖抑制作用を有するIgAまたはIgMである、請求項1に記載の方法。
- 前記免疫グロブリンにおいて、重鎖がC末端テールピースの一部または全部領域を欠く請求項1に記載の方法。
- 請求項1または2に記載の方法で用いられる、ゲノム中に免疫グロブリンをコードする核酸断片を含む糸状菌。
- 請求項1または2に記載の方法で製造された多量体免疫グロブリンまたは多量体免疫グロブリンを含む糸状菌。
- 請求項1または2に記載の方法で製造された多量体免疫グロブリンまたは多量体免疫グロブリンを含む糸状菌を含む組成物。
- 改変対象の糸状菌を準備する工程;
単一段階のゲノム改変操作によって、免疫グロブリンをコードする核酸断片の2コピー以上を、前記改変対象の糸状菌に挿入する工程;および
前記核酸断片の挿入されたゲノム改変糸状菌が、培養培地中に多量体免疫グロブリンを分泌することを確認する工程を含む、
ゲノム中に免疫グロブリンをコードする核酸断片を含む糸状菌糸状菌を製造する方法。 - ゲノム中に免疫グロブリンをコードする核酸断片を2コピー以上含む遺伝子改変糸状菌であって、多量体免疫グロブリンを産生することができる、糸状菌。
- 前記免疫グロブリンが、IgAまたはIgMである、請求項10に記載の糸状菌。
- 請求項10または11に記載の糸状菌を含む、組成物。
- 請求項10または11に記載の糸状菌の培養上清を含む、組成物。
