Abstract
L'invention concerne l'utilisation d'un extrait de Phaeodactylum tricornutum seul ou en association avec un extrait choisi parmi un extrait de guarana (Paullinia cupana), un extrait de ginseng (Panax ginseng), un extrait de Gingko biloba, un extrait de Nelumbo nucifera, un extrait de Centella asiatica, un extrait de Piper nigrum, un extrait de Bacopa monnieri ou l'une quelconque de leur combinaison et/ou un composé choisi parmi l'arginine, la créatine, la caféine, la théanine, la vitamine C, la vitamine E, la vitamine B, la théophylline, la paraxanthine, la théobromine, l'acide rosmarinique, l'acide ellagique, l'aldéhyde cinnamique ou l'une quelconque de leur combinaison, pour maintenir et/ou augmenter la vitesse de traitement des données et/ou la concentration chez des mammifères sains. L'association de l'extrait de P. tricornutum et d'un de ces extraits ou d'un ces composés est aussi décrite. L'invention est aussi relative à une composition comprenant l'association selon l'invention.
Description
Titre : Utilisation d'un extrait de microalgue seul ou en combinaison pour améliorer les capacités cognitives.
L'invention est relative à l'utilisation d'un extrait de la microalgue Phaeodacty/um tricornutum seul ou en association pour maintenir et/ou augmenter la vitesse de traitement des données et/ou la concentration chez des mammifères sains.
Les fonctions cognitives correspondent aux fonctions cérébrales responsables de l'acquisition, du traitement, de la mémoire et de l'utilisation de toutes les données provenant de l'environnement. Elles permettent de raisonner, s'adapter, se concentrer mais aussi communiquer. Certaines périodes de la vie, certaines habitudes ou certaines pratiques demandent chez les individus une attention, une mémoire et une vitesse d'acquisition accrues, ainsi qu'une vitesse accrue de tri et de sélection pertinentes des données en mémoire. En effet, les processus cognitifs peuvent être décomposés en phase d'apprentissage (stockage des données), phase de traitement des données au sein de la mémoire (vitesse et qualité de l'analyse) en fonction d'une situation nécessitant une décision, puis la réponse motrice permettant la mise en œuvre de la décision en la rendant réelle et effective. La phase de traitement des données est capitale : au-delà de la capacité et de la qualité de stockage de la mémoire d'un individu, c'est sa capacité à traiter les données mémorisées tant en qualité (précision) qu'en rapidité, qui lui permet de s'adapter de manière optimale à son environnement social, professionnel et privé. Un cas typique correspond aux étudiants, en conditions d'apprentissage et particulièrement en conditions d'examen, période durant laquelle leurs fonctions cognitives sont largement sollicitées. Un autre exemple est représenté par les joueurs de jeu vidéo, dont on sait que certaines de leurs fonctions cognitives en particulier telles que la vitesse de prise de décision, l'adaptation à un changement d'environnement, sont sollicitées durant la pratique. Ces populations d'individus ne présentent pas de trouble cognitif, ni même de trouble léger cognitif, définis comme des capacités cognitives altérées sans démence. Il ne s'agit pas d'une population âgée présentant un déclin cognitif. Il ne s'agit pas non plus d'individus ayant subi un quelconque stress prénatal pouvant expliquer un déficit cognitif acquis. Il ne s'agit à fortiori pas
d'individus souffrant d'une pathologie en lien avec un déficit cognitif tel que la maladie d'Alzheimer.
Des traitements médicamenteux à base de molécules telles que le donépézil, la rivastigmine, la galantamine, la mémantine, le donanemab, le TTP488 ou l'aducanumab sont déjà connus et ont montré leur effet sur le traitement ou la prévention d'un déficit ou d'un déclin cognitif. Des compléments alimentaires, notamment à base de caféine ou de vitamines, existent aussi sur le marché pour répondre à ce déficit. Toutefois, il existe un besoin dans le domaine nutraceutique d'ingrédients naturels alternatifs qui ne viennent ni prévenir ou traiter un déficit ni un déclin cognitif et qui ne consistent pas en un traitement pharmaceutique. On parle ici d'un besoin d'individus sains, de jeunes adultes ou adultes sans déficit, non âgés.
C'est précisément ce à quoi la présente invention se propose de répondre en proposant un extrait de Phaeodactylum tricornutum totalement naturel, actif sur des composantes spécifiques des fonctions cognitives. En outre, tel qu'il sera démontré dans la présente demande, la Demanderesse a de façon surprenante mis en évidence un effet synergique d'un extrait de P. tricornutum avec des extraits ou composés connus tels que les vitamines C, E, B6, B9, B12, le guarana Pauiiinia cupana), des extraits de Neiumbo nucifera, de Centeiia asiatica, de Piper nigrum, le ginseng Panax ginseng), le ginkgo biloba G. biioba), le bacopa Bacopa monnieri), la créatine, l'arginine, la caféine, la théanine, la théophylline, la paraparaxanthine, la théobromine, des composés phénoliques tels que l'acide rosmarinique, l'acide ellagique ou l'aldéhyde cinnamique.
Un des avantages de l'extrait de P. tricornutum selon l'invention est qu'il s'agit d'un extrait totalement naturel, ne présentant pas de toxicité pour les humains et qui ne génère pas d'allergie. Il peut être obtenu par culture de la microalgue en conditions contrôlées en photobioréacteur, et peut donc être produit facilement à l'échelle industrielle.
La microalgue P. tricornutum est une microalgue appartenant au genre Phaeodactylum et à la famille des Phaeodactylaceae. C'est une microalgue présente sous 3 morphotypes distincts (fusiforme, ovale, triradiée) que l'on
trouve dans de nombreuses régions océaniques du monde, dont le nord de l'Europe, l'Océanie, l'Atlantique.
La demande de brevet FR3092968A1 de la Demanderesse décrit des extraits de microalgue des espèces Phaeodactylum tricornutum et Tisochrysis iutea pour préparer un complément alimentaire pour prévenir ou lutter contre les troubles cognitifs liés au vieillissement ou les troubles cognitifs chez les enfants ou les jeunes adultes ayant subi un stress prénatal. Toutefois, il s'agit dans cette demande de prévenir un déclin de déficit d'apprentissage, d'améliorer notamment la mémoire de travail spatiale et la mémoire contextuelle à long terme. Les extraits décrits permettent de palier des altérations comportementales liées à des altérations biochimiques (augmentation du stress oxydatif et induction de processus neuro-inflammatoires mettant en œuvre la libération d'interleukines) que l'on observe en pratique chez des populations d'individus présentant des troubles cognitifs non pathologiques et en particulier chez de jeunes enfants ou adultes ayant subi un stress prénatal, ou chez des sujets âgés, ce qui n'est pas le cas de la population visée dans le cadre de la présente invention. Rien dans cette demande de brevet FR3092968A1 ne divulgue un effet d'un extrait de P. tricornutum pour maintenir et/ou augmenter la vitesse de traitement des données et/ou la concentration au sein de populations d'individus ne présentant aucun déficit cognitif. La demande ne divulgue à fortiori pas une utilisation de cet extrait en association avec un autre extrait ou composé pour en augmenter encore les effets. On notera qu'on distingue clairement au sens de l'invention un effet sur la mémoire d'un individu d'un effet sur la vitesse de traitement des données mémorisées. De même, un effet sur la concentration des individus est distinct de leur capacité à mémoriser les données.
Ainsi rien dans cette demande FR3092968A1 ne peut inciter l'homme du métier à utiliser un des extraits de microalgue décrits pour espérer maintenir et/ou augmenter la vitesse de traitement des données et/ou la concentration au sein d'une population par ailleurs distincte de celle décrite dans cette demande, encore moins en association avec un autre composé ou extrait pour en potentialiser les effets.
La demande WO2019193596A1 décrit par ailleurs une méthode pour réduire l'accumulation des lipides dans les cellules, à visée de prévention ou de traitement d'une dysfonction du foie ou des reins. Cette méthode emploie l'utilisation d'un extrait de la même microalgue P. tricornutum. L'utilisation de la présente invention en est toutefois parfaitement distincte. D'ailleurs, la méthode divulguée ici est une méthode de traitement thérapeutique, qui vise une population particulière d'individus. Ainsi rien dans ce document n'incite l'homme du métier à utiliser cet extrait de P. tricornutum pour mettre en œuvre l'invention décrite dans la présente demande.
Ainsi à la connaissance de la Demanderesse, aucun art antérieur ne divulgue l'utilisation décrite dans la présente demande. Aucun art antérieur ne décrit non plus l'association en tant que telle d'un extrait de P. tricornutum avec un extrait choisi parmi un extrait de guarana Pauiiinia cupana}, un extrait de ginseng (fanax ginseng), un extrait de Gingko biioba, un extrait de Neiumbo nucifera, un extrait de Centeiia asiatica, un extrait de Piper nigrum, un extrait de bacopa Bacopa monnieri} ou avec un composé choisi parmi la créatine, l'arginine, la caféine, la théanine, la vitamine C, la vitamine E, la vitamine B, la théophylline, la paraxanthine, la théobromine, l'acide rosmarinique, l'acide ellagique, l'aldéhyde cinnamique.
Ainsi un premier objet concerne l'utilisation d'un extrait de Phaeodactyium tricornutum seul ou en association pour maintenir et/ou augmenter la vitesse de traitement des données et/ou la concentration chez des mammifères sains, en particulier des individus sains ne présentant pas de déclin et/ou de trouble cognitif. Un second objet concerne l'association d'un extrait de P. tricornutum avec un extrait choisi parmi un extrait de guarana Pauiiinia cupana , un extrait de ginseng (Panax ginseng), un extrait de Gingko biioba, un extrait de Neiumbo nucifera, un extrait de Centeiia asiatica, un extrait de Piper nigrum, un extrait de B. monnieri ou un composé choisi parmi la créatine, l'arginine, la caféine, la théanine, la vitamine C, la vitamine E, la vitamine B, la théophylline, la paraxanthine, la théobromine, l'acide rosmarinique, l'acide ellagique, l'aldéhyde cinnamique, avantageusement avec un extrait choisi parmi un extrait de guarana Pauiiinia cupana , un extrait de Neiumbo nucifera, un extrait de Centeiia
asiatica, un extrait de Piper nigrum, ou l'une quelconque de leur combinaison et/ou un composé choisi parmi la créatine, l'arginine, la caféine, la théophylline, la paraxanthine, la théobromine, l'acide rosmarinique, l'acide ellagique, l'aldéhyde cinnamique ou l'une quelconque de leur combinaison. Un troisième objet est relatif à une composition, avantageusement à usage de complément alimentaire, comprenant l'association selon l'invention.
Un premier objet de l'invention concerne ainsi l'utilisation d'un extrait de Phaeodactyium tricornutum seul ou en association avec un extrait choisi parmi un extrait de guarana P. cupana), un extrait de ginseng P. ginseng), un extrait de G. biioba, un extrait de N. nucifera, un extrait de C. asiatica, un extrait de P. nigrum, un extrait de B. monnierio l'une quelconque de leur combinaison et/ou un composé choisi parmi la créatine, l'arginine, la caféine, la théanine, la vitamine C, la vitamine E, la vitamine B, la théophylline, la paraxanthine, la théobromine, l'acide rosmarinique, l'acide ellagique, l'aldéhyde cinnamique ou l'une quelconque de leur combinaison, avantageusement avec un extrait choisi parmi un extrait de guarana Pauiiinia cupana), un extrait de Neiumbo nucifera, un extrait de Centeiia asiatica, un extrait de Piper nigrum, ou l'une quelconque de leur combinaison et/ou un composé choisi parmi la créatine, l'arginine, la caféine, la théophylline, la paraxanthine, la théobromine, l'acide rosmarinique, l'acide ellagique, l'aldéhyde cinnamique ou l'une quelconque de leur combinaison, pour maintenir et/ou augmenter la vitesse de traitement des données et/ou la concentration chez des mammifères sains, avantageusement chez des individus sains ne présentant pas de déclin et/ou de trouble cognitif. Avantageusement, il s'agit d'une utilisation non thérapeutique, en particulier nutraceutique.
On entend ici par « mammifère » un humain ou un animal, avantageusement un animal domestique, encore avantageusement le chien ou le chat. Il s'agit très avantageusement d'un humain. On entend par « individus sains » des individus qui ne présentent pas de déclin et/ou de trouble cognitif. Cette population exclut donc les individus âgés, en particulier les humains âgés, c'est-à-dire de plus de 60 ans, avantageusement de plus de 70ans, les individus souffrant de dépression, y inclus de dépression chronique, mais aussi les mammifères
présentant un déficit cognitif suite à un accident vasculaire cérébral, un cancer, un diabète, un traumatisme physique du cerveau, une maladie chronique des reins, ou encore la maladie d'Alzheimer, les individus présentant une quelconque modification cognitive comportementale liée à un stress oxydatif ou à un état neuro-inflammatoire, ainsi que les enfants ou jeunes adultes ayant subi un stress prénatal induisant des troubles non pathologiques, tels que l'hyperactivité, le déficit d'attention et de mémoire, le retard au niveau du langage et le comportement anxieux.
On entend par « utilisation non thérapeutique » une utilisation non pharmaceutique, qui n'est donc pas à visée de traitement thérapeutique au sens d'un spécialiste du domaine. Elle vise la population d'individus qualifiés de sains tel que définie ci-dessus.
On entend par « vitamine B » la vitamine B6, B9, B12 ou l'une quelconque de leur combinaison. On entend encore par « voie orale » l'ingestion de l'extrait de P. tricornutum seul ou en combinaison ou d'une composition le comprenant.
On entend par « augmenter la vitesse de traitement des données chez des mammifères sains », réduire le temps nécessaire à un mammifère sain, pour analyser les informations provenant d'un stimuli externe choisi parmi un stimulus visuel, un stimulus auditif, un stimulus olfactif ou un stimulus gustatif en vue d'une prise de décision. Dans un mode de réalisation avantageux, il s'agit d'une réduction du temps de réaction en réponse à un stimuli visuel, préférentiellement chez des individus sains.
Ainsi avantageusement, la réduction dudit temps de réaction est une diminution significative du temps de réponse d'au moins 9ms, avantageusement d'au moins 30ms, très avantageusement d'au moins 60ms mesurée au sein d'une population d'individus sains à laquelle on a administré une combinaison de guarana et de l'extrait de P. tricornutum selon l'invention, en comparaison du temps de réaction mesuré au sein de la même population recevant un ingrédient placebo.
La mesure est avantageusement effectuée dans le cadre d'une étude clinique par l'intermédiaire du test de Sternberg (Sternberg S., 1969) qui consiste à présenter aux participants des stimuli visuels, un par un, en les identifiant comme présents ou absents dans des séquences de 3, 6, 9, 12, 15 ou 18 secondes d'intervalle : le
nombre de stimuli visuel (données à traiter) est augmenté sur de courts intervalles de temps et le niveau de difficulté est ainsi croissant. Afin d'éviter la répétition, les participants reçoivent l'instruction de compter à rebours par trois et quatre jusqu'à un nombre aléatoire spécifique, jusqu'à ce qu'ils voient une lumière rouge apparaître sur l'écran de l'ordinateur. Ce test mesure spécifiquement la précision et le temps de réaction.
Dans un mode de réalisation avantageux, la réduction du temps de réaction est mesurée au sein d'une population composée de 51 hommes et 10 femmes sains, qualifiés de joueurs expérimentés, d'une moyenne d'âge de 17,7 à 25,7 ans, d'un poids de 60 à 86 kg, avec un indice de masse corporelle allant de 20,6 à 27,8 kg/m2. Avantageusement, les individus n'ont pas consommé de stimulants, suppléments ou compléments alimentaires comprenant de la théophylline, de la paraxanthine, de la théobromine, des vitamines, de la créatine, de la caféine, de la théanine, ou tout autre substance ou extrait contenant l'un quelconque des composés précédemment cités seul ou en association, ni consommé un extrait de guarana ou un extrait de ginseng ou un extrait de G. biloba, ni un extrait de P. tricornutum, ni d'aliments riches en arginine ou en nitrates, durant les 2 semaines avant le commencement de l'étude clinique. Encore avantageusement, la combinaison administrée est une combinaison de 500mg d'un extrait de guarana et d'une dose de 436mg/jour ou de 872mg/jour d'une composition comprenant l'extrait de P. tricornutum selon l'invention, du tocophérol, une huile MCT telle que décrite dans la suite de la présente demande, dans les conditions telles que décrites dans l'exemple 3. Très avantageusement, l'extrait de P. tricornutum compris dans la composition est l'extrait tel que préparé dans l'exemple 1 et l'extrait de guarana est l'extrait de graines de guarana tel que préparé dans l'exemple 2.
Dans un mode de réalisation alternatif, on entend par « augmenter la vitesse de traitement des données chez des individus sains » augmenter l'expression protéique d'une synaptotagmine choisie parmi la synaptotagmine 1 (SYT1), SYT2, SYT3, SYT4, SYT5, SYT6, SYT7, SYT8, SYT9, SYT10, SYT11, SYT12, SYT13, SYT14, SYT15, SYT16, SYT17, dans une culture de neurones en présence de l'extrait de P. tricornutum selon l'invention. Avantageusement, l'augmentation
de l'expression protéique est d'au moins 2%, avantageusement d'au moins 4% en présence de l'extrait selon l'invention en comparaison d'un contrôle sans extrait. Avantageusement il s'agit d'une augmentation de l'expression protéique de la synaptotagmine mesurée en présence de l'extrait de P. tricornutum tel que préparé selon l'exemple 1.
Dans un autre mode de réalisation alternatif, on entend par « augmenter la vitesse de traitement des données chez des individus sains » augmenter à l'échelle cellulaire neuronale la densité du réseau neuronal en augmentant la longueur et/ou la dendricité des prolongements neuronaux chez des individus sains. On entend par « dendricité » la formation de dendrites (prolongements du corps cellulaire des neurones). Il est bien connu en effet que l'ensemble des composantes cognitives, de façon particulière la vitesse de traitement des données des individus, est étroitement lié à la densité cellulaire neuronale.
Ainsi avantageusement, l'extrait de P. tricornutum selon l'invention augmente d'au moins 2,5%, préférentiellement d'au moins 5% la longueur des prolongements neuronaux in vitro, dans une culture de neurones, en comparaison de la longueur desdits prolongements mesurée sans extrait. Avantageusement, l'augmentation des prolongement neuronaux est mesurée dans une culture de neurones en présence d'une quantité d'extrait selon l'invention en poids par rapport au poids total du milieu de culture et de l'extrait comprise entre 0,0005% et 1,5%, avantageusement entre 0,001% et 0,5%. Avantageusement, l'extrait de P. tricornutum est celui tel que préparé à l'exemple 1. Très avantageusement, la longueur des neurones est mesurée après marquage des neurones avec un anticorps anti-p-tubuline puis révélé à l'aide d'un anticorps secondaire couplé à un fluorochrome à l'aide de prises photographiques par un microscope automatique, en particulier dans les conditions telles que décrites dans l'exemple 5a).
Alternativement, l'augmentation de la vitesse de traitement des données chez des individus sains est évaluée en mesurant la dendricité neuronale dans le même modèle de culture de neurones in vitro que celui décrit ci-dessus, en présence d'une quantité d'extrait selon l'invention en poids par rapport au poids total du milieu de culture et de l'extrait comprise entre 0,0005% et 1,5%,
avantageusement entre 0,001% et 0,5%. Avantageusement, l'extrait de P. tricornutum est celui tel que préparé à l'exemple 1. Très avantageusement, la dendricité neuronale est évaluée en mesurant la formation des bifurcations des neurones, par microscopie tel que détaillé ci-dessus.
Dans encore un autre mode de réalisation alternatif de l'invention, « augmenter la vitesse de traitement des données chez des individus sains » signifie augmenter in vitro la vitesse de mobilisation du calcium au niveau des synapses durant la transmission synaptique (transmission de l'influx nerveux entre deux neurones). Les canaux calciques sont en effet essentiels à la transmission de l'influx nerveux ; leur ouverture sous l'effet de la dépolarisation des boutons synaptiques suite à un potentiel d'action permet un afflux d'ions calcium dans les boutons synaptiques déclenchant la fusion de vésicules synaptiques, lesquelles libèrent les neurotransmetteurs tels que l'acétylcholine. Ainsi une mesure in vitro du taux de calcium mobilisé dans une culture de neurones en présence de l'extrait selon l'invention permet de manière plausible de refléter une augmentation de la vitesse de traitement des données. Dans un mode de réalisation avantageux, la mesure du taux de calcium mobilisé est effectuée à l'aide d'une sonde fluorescente se liant au calcium sous l'effet de sa libération suite à l'activation des récepteurs neuronaux, en particulier dans les conditions telles que décrites dans l'exemple 5b). Avantageusement, l'augmentation de la vitesse de traitement des données par l'extrait selon l'invention est mesurée dans une culture de neurones en présence d'une quantité comprise entre 0,0005% et 1,5%, avantageusement entre 0,001% et 0,5%. Avantageusement, l'extrait selon l'invention est l'extrait de P. tricornutum est celui tel que préparé à l'exemple 1.
On entend d'autre part par « maintenir la vitesse de traitement des données chez des mammifères sains » maintenir le temps de réaction nécessaire à un mammifère sain, avantageusement un individu sain, en réponse à un nombre croissant de stimuli (données à traiter). Dans un mode de réalisation préférentiel, il s'agit du maintien du temps de réaction mesuré en présence de la combinaison de guarana et de l'extrait de P. tricornutum selon l'invention, en comparaison du
temps de réaction mesuré au sein de la population ayant reçu l'ingrédient placebo. On entend avantageusement par « maintenir la vitesse de traitement des données chez des individus sains », un maintien du temps de réaction mesuré en présence de la combinaison de guarana et de l'extrait de P. tricornutum lorsque le nombre de stimuli visuel (données à traiter d) sur un temps donné est augmenté de 4 à 6. Avantageusement encore, il s'agit de la mesure du temps de réaction mesuré dans le cadre de l'étude clinique décrite précédemment. Encore avantageusement, la combinaison administrée est une combinaison de 500mg d'un extrait de guarana et d'une dose de 436mg/jour ou de 872 mg/jour d'une composition comprenant l'extrait de P. tricornutum selon l'invention, du tocophérol, une huile MOT telle que décrite dans la suite de la présente demande, dans les conditions telles que décrites dans l'exemple 3. Très avantageusement, l'extrait de P. tricornutum compris dans la composition est l'extrait tel que préparé dans l'exemple 1 et l'extrait de guarana est l'extrait de graines de guarana tel que préparé dans l'exemple 2.
On entend par ailleurs par « maintenir et/ou augmenter la concentration de mammifères sains » maintenir et/ou augmenter la précision d'analyse des informations provenant d'un stimuli externe choisi parmi un stimulus visuel, un stimulus auditif, un stimulus olfactif ou un stimulus gustatif en vue d'une prise de décision chez un mammifère sain. Dans un mode de réalisation avantageux, il s'agit d'une augmentation de la précision en réponse à un stimulus visuel, avantageusement chez des individus sains. Ainsi, l'amélioration de la concentration est une augmentation significative du pourcentage de précision d'une population d'individus sains d'au moins 0,02%, avantageusement d'au moins 0,05% après 30 jours de supplémentation mesuré au sein d'une population d'individus sains à laquelle on a administré la combinaison de l'extrait de guarana et de l'extrait de P. tricornutum selon l'invention, en comparaison d'une population recevant un ingrédient placebo. La mesure est effectuée dans le cadre de l'étude clinique décrite précédemment mais par l'intermédiaire du test Go-NoGo (Donders, 1969 ; Wessel, 2017) qui évalue précisément la capacité à maintenir une attention soutenue (concentration) ainsi que le contrôle des réponses (prise de décision) via la mesure de la précision de la réponse à des
stimuli visuels en appuyant sur une touche représentant "Go" ou en inhibant une réponse en n'appuyant pas sur la touche représentant "No-Go". Cette évaluation a été réalisée avec la même population d'étude décrite précédemment et dans des conditions strictement similaires. Encore avantageusement, la combinaison administrée est une combinaison de 500mg d'un extrait de guarana et d'une dose de 436mg/jour ou de 872 mg/jour d'une composition comprenant l'extrait de P. tricornutum selon l'invention, du tocophérol, une huile MCT telle que décrite dans la suite de la présente demande, dans les conditions telles que décrites dans l'exemple 4. Très avantageusement, l'extrait de P. tricornutum compris dans la composition est l'extrait tel que préparé dans l'exemple 1 et l'extrait de guarana est l'extrait de graines de guarana tel que préparé dans l'exemple 2.
Dans un mode de réalisation de l'invention, l'extrait de P. tricornutum est utilisé seul.
Avantageusement, l'extrait est obtenu par toute méthode d'extraction connue de l'homme du métier. De façon avantageuse, l'extrait est obtenu à partir de la biomasse d'une culture de la microalgue. La biomasse peut être au préalable centrifugée puis filtrée pour éliminer l'eau. Une étape d'extraction solide-liquide peut être mise en œuvre par la suite. Préférentiellement, l'extrait est obtenu par extraction dans un solvant ou mélange de solvant, et avantageusement dans l'eau, l'acétone, l'hexane, l'acétate d'éthyle, le méthyltétrahydrofurane, le 2- méthyloxolane, l'heptane, un alcool choisi parmi l'éthanol, le méthanol, l'isopropanol, une huile naturelle ou ramifiée, un glycol, un polyol, un mélange eau/alcool, eau/glycol en proportion de 100/1 à 1/100 (p/p) ou tout autre solvant permettant d'extraire tout ou partie des composés de nature hydrophobe et amphiphile. Très préférentiellement, l'extrait de P. tricornutum est obtenu par extraction dans un mélange eau/éthanol en proportion de 40/60 (p/p) à 1/100 (p/p), y inclus de 30/70 (p/p) et de 20/80 (p/p). Le solvant ou le mélange de solvants est séparé de la biomasse résiduelle après extraction par des procédés de type centrifugation, filtration et peut par la suite être concentré, ou le solvant éliminé, par des techniques telles que l'évaporation sous vide ou tout autre technique permettant l'évaporation sélective du solvant considéré.
Alternativement, l'extrait de P. tricornutum selon l'invention est obtenu par extraction en conditions subcritiques ou supercritiques. Avantageusement dans ce cas, l'extrait est obtenu par extraction au CO2 supercritique.
L'extraction peut être réalisée à une température allant de 4°C à 300°C, y inclus la température ambiante, c'est-à-dire une température de 20°C. Dans un mode préférentiel de réalisation de l'invention, l'extraction est réalisée à température ambiante. Dans tous les cas, l'extrait obtenu est filtré, éventuellement séché. Avantageusement dans ce cas, l'étape de séchage est effectuée par lyophilisation, séchage sous vide, séchage en tambour, ou atomisation, par lit fluidisé en couplant par toute technique permettant une encapsulation ou une microencapsulation via une matrice de support et/ou la formation d'une émulsion. L'extrait selon l'invention se présente alors sous forme de poudre. Il pourra être incorporé dans une composition seul ou en association comme décrite ci-après. Alternativement, l'extrait de P. tricornutum est filtré mais non séché et se présente sous forme liquide.
Dans un autre mode de réalisation, l'extrait de P. tricornutum est utilisé en association avec un extrait choisi parmi un extrait de guarana P. cupana), un extrait de ginseng P. ginseng), un extrait de G. biioba, un extrait de N. nucifera, un extrait de C asiatica, un extrait de P. nigrum, un extrait de B. monnieri ou l'une quelconque de leur combinaison et/ou un composé choisi parmi la créatine, l'arginine, la caféine, la théanine, la vitamine C, la vitamine E, la vitamine B, la théophylline, la paraxanthine, la théobromine, l'acide rosmarinique, l'acide ellagique, l'aldéhyde cinnamique ou l'une quelconque de leur combinaison.
En particulier l'extrait de P. tricornutum est utilisé en association avec un extrait choisi parmi un extrait de guarana Pauiiinia cupana), un extrait de Neiumbo nucifera, un extrait de Centeiia asiatica, un extrait de Piper nigrum, ou l'une quelconque de leur combinaison et/ou un composé choisi parmi la créatine, l'arginine, la caféine, la théophylline, la paraxanthine, la théobromine, l'acide rosmarinique, l'acide ellagique, l'aldéhyde cinnamique ou l'une quelconque de leur combinaison,
Ainsi un extrait de guarana P. cupana), de ginseng P. ginseng), de G. biioba, de N. nucifera, de C. asiatica, de P. nigrum peut être obtenu par extraction de tout
ou partie de la plante d'intérêt choisie parmi les bourgeons, les feuilles, la tige, les fleurs, les graines, le rhizome. L'extraction de guarana est avantageusement une extraction des graines, l'extraction de G. bi/oba est avantageusement une extraction des bourgeons ou des feuilles, avantageusement des feuilles. L'extraction de P. ginseng est avantageusement une extraction du rhizome, l'extraction de N. nucifera est avantageusement une extraction des fleurs et l'extraction de P. nigrum avantageusement une extraction des graines.
L'extraction peut être mise en œuvre par toute méthode connue de l'homme du métier choisie parmi la macération, la décoction à chaud, par broyage dont le broyage aux ultrasons, à l'aide d'un mixeur, ou encore l'extrait peut être obtenu par extraction dans l'eau, notamment en conditions subcritiques ou supercritiques (dioxyde de carbone). L'extraction peut être conduite à partir de matière sèche ou fraîche, avantageusement sèche, en quantité de 0,1 % à 20 % en poids, avantageusement de 1 % à 20 %, très avantageusement de 5 % à 15 %, et encore plus avantageusement de 15 % en poids par rapport au poids total de la matière et du solvant d'extraction. Au sens de l'invention, on entend par « matière sèche » une matière végétale déshydratée comprenant moins de 15%, avantageusement moins de 10%, encore avantageusement moins de 5% et très avantageusement moins de 1% d'eau. L'extraction peut être réalisée à une température allant de 4°C à 300°C, y inclus la température ambiante, c'est-à-dire une température de 20°C. Dans un mode préférentiel de réalisation de l'invention, l'extraction sera réalisée à température ambiante.
Dans un mode de réalisation de l'invention, l'extraction est réalisée dans l'eau en conditions subcritiques, à une température allant de 100°C à 300°C, avantageusement de 120°C à 250°C, encore avantageusement à 120°C. L'extraction peut être réalisée à une température donnée ou à des températures successives croissantes. Dans un mode de réalisation avantageux de l'invention, l'extraction est réalisée à une température de 120°C. Dans un mode alternatif, elle sera conduite selon un gradient de trois températures croissantes comprises entre 100°C et 200°C, tel que 120°C, 140°C puis 160°C ou 110°C, 130°C puis 150°C, ou encore 120°C, 145°C puis 170°C.
On entend par extraction en « conditions subcritiques » une extraction en présence d'eau, dans des conditions de température supérieures à 100°C et de pression inférieure à 221 bars, l'eau restant à l'état liquide mais possédant une viscosité et une tension de surface inférieures à celle de l'eau à température ambiante, augmentant sa constante diélectrique. Ainsi, la pression d'extraction est comprise entre 150 bars et 250 bars, préférentiellement entre 200 et 221 bars, avantageusement dans un autoclave d'extraction sous pression.
Dans un mode de réalisation alternatif, l'extrait de guarana P. cupana), de ginseng P. ginseng) , de G. biioba, de N. nucifera, de C asiatica ou de P. nigrum peut être obtenu par extraction dans un solvant ou mélange de solvant, et avantageusement dans l'eau, l'acétone, l'hexane, l'acétate d'éthyle, le méthyltétrahydrofurane, le 2-méthyloxolane, l'heptane, un alcool choisi parmi l'éthanol, le méthanol, l'isopropanol, une huile naturelle ou ramifiée, un glycol, un polyol, un mélange eau/alcool, eau/glycol en proportion de 100/1 à 1/100 (p/p) ou tout autre solvant permettant d'extraire tout ou partie des composés de nature hydrophobe et amphiphile tels que les liquides ioniques comme par exemple des cations imidazolium ou pyrrolidinium combinés avec les anions chlorure, acétate et tosylate. L'extrait obtenu est ensuite filtré puis éventuellement séché par lyophilisation, séchage sous vide, séchage en tambour, ou atomisation. L'extrait séché se présente ainsi sous forme de poudre. Alternativement il peut se présenter sous forme liquide.
Dans un mode de réalisation avantageux de l'invention, l'extrait de P. tricornutum est utilisé en association avec un extrait de guarana Pauiiinia cupana), un extrait de Neiumbo nucifera, un extrait de Centeiia asiatica, un extrait de Piper nigrum, ou l'une quelconque de leur combinaison. Dans un mode de réalisation avantageux de l'invention, l'extrait de P. tricornutum est utilisé en association avec un extrait de guarana, avantageusement un extrait des graines de guarana.
Dans encore un autre mode de réalisation, l'extrait de P. tricornutum est utilisé en association avec un composé choisi parmi la créatine, l'arginine, la caféine, la théanine, la vitamine C, la vitamine E, la vitamine B, la théophylline, la paraxanthine, la théobromine, , l'acide rosmarinique, l'acide ellagique, l'aldéhyde cinnamique ou l'une quelconque de leur combinaison, avantageusement avec un
composé choisi parmi la créatine, l'arginine, la caféine, la théophylline, la paraxanthine, la théobromine, l'acide rosmarinique, l'acide ellagique, l'aldéhyde cinnamique ou l'une quelconque de leur combinaison.
La caféine (C8Hi0N4O2 ; masse molaire : 194,19 g/mol, n° CAS 58-08-2), la théophylline (C7H8N4O2 ; masse molaire : 180,164 g/mol ; n° CAS : 58-55-9), la paraxanthine (C5H4N4O2, masse molaire : , n° CAS 69-89-6) et la théanine (C7HI4N2O, masse molaire : 174,2 g/mol, n° CAS : 3081-61-6) peuvent être obtenues par extraction de toute plante en contenant, notamment choisies parmi Coflea arabica, P. cupana, Camellia sinensis, Theobroma cacao. La théobromine (C7H8N4O2, masse molaire : 180,164 g/mol, n° CAS : 83-67-0) peut être obtenue par extraction de T. cacao. La créatine (C4H7N3O, masse molaire : 113,1 g/mol, n° CAS : 60-27-5) et l'arginine (C6Hi4N4O2, masse molaire : 174,2 g/mol, n° CAS : 7200-25-1 ou 157-06-2 pour la forme D ou R-) peuvent être obtenues dans le commerce. L'acide rosmarinique (C18H16O8, masse molaire : 360,31 g/mol, n° CAS : 20283-92-5) peut être extrait de la plante Saivia rosmarinus. L'acide ellagique (CI4H6O8, masse molaire : 302,19 g/mol, n° CAS : 476-66-4) peut être extrait à partir de fruits rouges tels que cerise, framboise, mûres. L'aldéhyde cinnamique (C9H8O, masse molaire : 132,16 g/mol, n° CAS : 104-55-2) est le composant principal de l'essence de cannelle.
Ces composés sont extraits par un solvant organique non polaire ou mélange de solvant organique non polaire choisis parmi l'hexane, le cyclohexane, l'heptane, le trichlorométhane, le dichlorométhane, l'acétone en proportion de 100/1 à 1/100 (p/p), le 2-méthyloxolane ou les liquides ioniques comme par exemple des cations imidazolium ou pyrrolidinium combinés avec les anions chlorure, acétate et tosylate. Ils peuvent être purifiés par Chromatographie Liquide Haute Performance (HPCL) préparative. Alternativement, ils peuvent être obtenus dans le commerce. La vitamine C (Acide L-ascorbique, C6H8O6, masse molaire 176,12 g/mol, n° CAS 50-81-7) peut être obtenue dans le commerce.
L'utilisation de l'extrait de P. tricornutum selon l'invention, seul ou en association, est avantageusement une utilisation par voie orale. L'extrait seul ou en association peut être compris dans une composition, avantageusement à usage de complément alimentaire.
L'invention concerne encore l'association d'un extrait de Phaeodactylum tricornutum et d'un extrait choisi parmi un extrait de guarana P. cupana), un extrait de ginseng P. ginseng), un extrait de G. biioba, un extrait de N. nucifera, un extrait de C asiatica, un extrait de P. nigrum, un extrait de B. monnieriou un composé choisi parmi la créatine, l'arginine, la caféine, la théanine, la vitamine C, la vitamine E, la vitamine B, la théophylline, la paraxanthine, la théobromine, l'acide rosmarinique, l'acide ellagique, l'aldéhyde cinnamique et l'une quelconque de leurs combinaison. Dans un mode de réalisation, l'association est caractérisée en ce que le ratio en poids entre l'extrait de P. tricornutum et l'extrait choisi parmi un extrait de guarana, un extrait de ginseng, un extrait de G. biioba, un extrait de N. nucifera, un extrait de C. asiatica, un extrait de B. monnieri ou du composé est respectivement de 1/6 à 6/1, avantageusement de 2/1 à 4/1, encore avantageusement de 3/1 à 7/2. Cette association est synergique. Très avantageusement selon l'invention, il s'agit d'une association d'un extrait de P. tricornutum et d'un extrait de guarana P. cupana). Préférentiellement, l'extrait de guarana est un extrait de graines P. cupana). Ainsi une association particulièrement avantageuse comprend en poids par rapport au poids total de l'association :
- Un extrait de P. tricornutum présent en quantité comprise entre 50mg et 600mg, avantageusement de 220mg à 440mg,
- Un extrait de guarana P. cupana) présent en quantité comprise entre 300mg et 700mg, avantageusement de 500mg.
Dans un mode de réalisation avantageux, l'extrait de P. tricornutum est préparé par extraction solide-liquide tel que déjà exposé dans l'invention. Préférentiellement, l'extrait de P. tricornutum est obtenu par extraction dans un
mélange eau/éthanol en proportion de 40/60 (p/p) à 1/100 (p/p), y inclus de 30/70 (p/p) et de 20/80 (p/p), encore préférentiellement dans l'éthanol. Encore avantageusement, l'extrait de guarana P. cupan ) est obtenu par extraction hydroalcoolique à chaud puis concentration avec évaporation sous vide. Les 2 extraits sont filtrés puis séchés par lyophilisation, séchage sous vide, séchage en tambour, ou atomisation. Chacun des 2 extraits se présente sous forme de poudre et peuvent être combinés dans les ratios décrits pour obtenir l'association.
L'association selon l'invention est particulièrement utile pour maintenir et/ou augmenter la vitesse de traitement des données et/ou la concentration chez des mammifères sains, avantageusement des individus sains ne présentant pas de déclin et/ou de trouble cognitif. En effet, la Demanderesse a mis en évidence de manière inattendue un effet synergique de l'association de l'extrait de P. tricornutum et d'un extrait de guarana P. cupanà) dans un ratio en poids de respectivement 1/6 à 6/1, avantageusement dans un ratio en poids de respectivement de 2/1 à 4/1, très avantageusement dans le ratio en poids de 3/1 à 7/2.
Ainsi dans un mode de réalisation, « augmenter la vitesse de traitement des données chez des mammifères sains », signifie ici réduire significativement le temps nécessaire d'au moins 5ms, avantageusement d'au moins 10ms, très avantageusement d'au moins 20 ms pour analyser les informations provenant d'un stimuli externe choisi parmi un stimulus visuel, un stimulus auditif, un stimulus olfactif ou un stimulus gustatif en vue d'une prise de décision, avantageusement un stimuli visuel, chez un mammifère sain, avantageusement un individu sain.
Avantageusement, cette réduction du temps de réaction est mesurée au sein d'une population d'individus sains à laquelle a été administrée l'association selon l'invention en comparaison du temps de réaction mesuré au sein d'une population recevant un ingrédient placebo. La mesure est avantageusement effectuée dans le cadre d'une étude clinique par l'intermédiaire du test de Sternberg (Sternberg S., 1969) tel que décrit précédemment. Quatre populations d'individus sains sont constituées, la première recevant une administration de
placébo, la seconde recevant une administration de l'association d'un extrait de P. tricornutum et d'un extrait de guarana, la 3eme une administration de l'extrait de P. tricornutum seul et la 4eme une administration d'un extrait de guarana seul. Avantageusement encore, l'association est l'association de l'extrait de P. tricornutum et d'un extrait de guarana, avantageusement un extrait de graines, dans le ratio en poids de respectivement de 1/6 à 6/1, avantageusement de 2/1 à 4/1, très avantageusement dans le ratio en poids de 3/1 à 7/2.
Très avantageusement, il s'agit de l'extrait de P. tricornutum tel que préparé selon l'exemple 1 et l'extrait de guarana est celui tel que préparé dans l'exemple 2.
Dans un mode de réalisation alternatif, l'association d'un extrait de P. tricornutum et d'un extrait de guarana P. cupana) augmente l'expression protéique d'une synaptotagmine choisie parmi la synaptotagmine 1 (SYT1), SYT2, SYT3, SYT4, SYT5, SYT6, SYT7, SYT8, SYT9, SYT10, SYT11, SYT12, SYT13, SYT14, SYT15, SYT16, SYT17, dans une culture de neurones.
Avantageusement, l'augmentation de l'expression protéique est d'au moins 3%, avantageusement d'au moins 5% l'expression protéique de la synaptotagmine dans la culture neuronale en comparaison d'un contrôle sans ladite association, tandis que l'extrait de P. tricornutum seul n'augmente que de 2% ladite expression protéique. De même, l'extrait de guarana seul n'augmente que de 1% l'expression protéique de la synaptotagmine en comparaison du contrôle sans extrait de guarana. Dans un mode de réalisation particulièrement avantageux, il s'agit d'une augmentation de l'expression protéique de de la synaptotagmine mesurée en présence de l'extrait de P. tricornutum et d'un extrait de guarana P. cupana), avantageusement un extrait de graines, dans le ratio en poids respectivement de 2/1 à 4/1, très avantageusement dans le ratio en poids de 3/1 à 7/2. Encore avantageusement, il s'agit de l'extrait de P. tricornutum tel que préparé selon l'exemple 1 et l'extrait de guarana est celui tel que préparé dans l'exemple 2.
Dans un autre mode de réalisation alternatif, l'association synergique d'un extrait de P. tricornutum et d'un extrait de guarana P. cupana) augmente la vitesse de traitement des données chez des mammifères sains, avantageusement des
individus sains, en augmentant in vitro la longueur et/ou la dendricité des prolongements neuronaux. Ainsi avantageusement, la longueur des prolongements neuronaux est augmentée dans une culture in vitro de neurones d'au moins 2,5%, préférentiellement d'au moins 5% en présence de l'association d'un extrait de P. tricornutum et d'un extrait de guarana P. cupanâ), en comparaison de l'augmentation des prolongements neuronaux mesurée en présence de l'extrait de P. tricornutum seulement ou de l'extrait de guarana seulement. Dans un mode de réalisation avantageux, l'association synergique est telle que le ratio en poids de l'extrait de P. tricornutum et de l'extrait de guarana P. cupanâ), est de respectivement 1/6 à 6/1, avantageusement de 2/1 à 4/1, très avantageusement de 3/1 à 7/2.
Très avantageusement, il s'agit de l'extrait de P. tricornutum tel que préparé selon l'exemple 1 et l'extrait de guarana est celui tel que préparé dans l'exemple 2.
Dans encore un autre mode de réalisation alternatif, l'association d'un extrait de P. tricornutum et d'un extrait de guarana P. cupanâ) augmente la vitesse de traitement des données chez des mammifères sains, avantageusement des individus sains, en augmentant in vitro la vitesse de mobilisation du calcium au niveau des synapses durant la transmission synaptique. Une mesure in vitro du taux de calcium mobilisé dans une culture de neurones en présence de l'association ou de chacun des extraits la caractérisant permet de manière plausible de refléter une augmentation de la vitesse de traitement des données. Dans un mode de réalisation avantageux, la mesure du taux de calcium mobilisé est effectuée à l'aide d'une sonde fluorescente se liant au calcium sous l'effet de sa libération suite à l'activation des récepteurs neuronaux. Avantageusement, l'augmentation de la vitesse de traitement des données est mesurée dans une culture in vitro de neurones en présence de l'association de l'extrait de P. tricornutum et de l'extrait de guarana telle que leur ratio en poids est de respectivement 1/6 à 6/1, avantageusement de 2/1 à 4/1, très avantageusement de 3/1 à 7/2. Très avantageusement, il s'agit de l'extrait de P. tricornutum tel que préparé selon l'exemple 1 et l'extrait de guarana est celui tel que préparé dans l'exemple 2.
Un autre objet de l'invention est encore relatif à une composition comprenant l'association selon l'invention. Dans un mode de réalisation, la composition est à usage de complément alimentaire.
Ainsi, la composition comprend l'association d'un extrait de P. tricornutum et d'un extrait choisi parmi un extrait de guarana P. cupana), un extrait de ginseng P. ginseng), un extrait de G. biioba un extrait de N. nucifera, un extrait de C asiatica, un extrait de P. nigrum, un extrait de B. monnieri ou l'une quelconque de leur combinaison et/ou un composé choisi parmi la créatine, l'arginine, la caféine, la théanine, la vitamine C, la vitamine E, la vitamine B, la théophylline, la paraxanthine, la théobromine, l'acide rosmarinique, l'acide ellagique, l'aldéhyde cinnamique ou l'une quelconque de leur combinaison, en particulier telle que le ratio en poids entre l'extrait de P. tricornutum et l'extrait ou le composé choisi est de 1/6 à 6/1, avantageusement de 2/1 à 4/1 très avantageusement de 3/1 à 7/2. Très avantageusement, la composition comprend l'association d'un extrait de P. tricornutum et d'un extrait choisi parmi un extrait de guarana Pauiiinia cupana), un extrait de Neiumbo nucifera, un extrait de Centeiia asiatica, un extrait de Piper nigrum ou l'une quelconque de leur combinaison, ou un composé choisi parmi la créatine, l'arginine, la caféine, la théophylline, la paraxanthine, la théobromine, l'acide rosmarinique, l'acide ellagique, l'aldéhyde cinnamique ou l'une quelconque de leur combinaison. Encore plus avantageusement, la composition comprend l'association d'un extrait de P. tricornutum et d'un extrait de guarana P. cupana), avantageusement un extrait de graines, en particulier dans un ratio en poids de respectivement 1/6 à 6/1, avantageusement de 2/1 à 4/1, très avantageusement de 3/1 à 7/2.
La composition peut comprendre en outre une huile végétale et/ou de la vitamine E. Par « huile végétale », on entend toute huile extraite d'une plante ou d'une algue, incluse une microalgue, notamment choisie parmi l'huile d'olive, l'huile de colza, l'huile de lin, l'huile de tournesol, une huile à triglycérides à chaîne moyenne (MCT). Par triglycérides à chaines moyennes (MCT, Medium Chain Triglycerides), on entend des esters de glycérol et d'acides gras saturés, dont la chaîne hydrocarbonée a de 6 à 12 atomes de carbone. Une huile MCT peut être ainsi choisie parmi l'huile de coco, avantageusement l'huile de noix de
coco, l'huile de palmiste et l'huile de palme mais peuvent être obtenus à partir d'autres graisses ou huiles. Avantageusement au sens de l'invention, l'huile végétale est une huile MCT, avantageusement encore l'huile de coco, très avantageusement l'huile de noix de coco. Dans un mode de réalisation de l'invention particulièrement avantageux, l'huile végétale est présente en quantité de 20% à 80%, avantageusement encore de 30% à 60% en poids par rapport au poids total de la composition. On entend par « vitamine E » un tocophérol choisi parmi l'o-tocophérol, le y-tocophérol, le p-tocophérol ou le 5-tocophérol, ou un tocotriénol choisi parmi l'o-tocotriénol, le p-tocotriénol, le y-tocotriénol ou le 5-tocotriénol. Avantageusement, il s'agit de l'o-tocophérol. Dans un mode de réalisation avantageux, la vitamine E est présente dans la composition en quantité en poids par rapport au poids total de la composition de 0,15% à 1,25%, avantageusement de 0,25% à 1%, très avantageusement de 0,5%. Ainsi une composition avantageuse de l'invention comprend au moins, en poids par rapport au poids total de la composition :
- Un extrait de P. tricornutum en quantité de 10% à 25%, avantageusement de 11% à 16%,
- Une huile MCT présente en quantité de 30% à 60%,
- De l'o-tocophérol présent en quantité de 0,25% à 1%, avantageusement de 0,5%,
- De la caféine ou de la théine en quantité de 5% à 25%, avantageusement de 12%.
Une autre composition particulièrement avantageuse comprend au moins, en poids par rapport au poids total de la composition :
- Un extrait de P. tricornutum en quantité de 10% à 25%, avantageusement de 11% à 16%,
- Une huile MCT présente en quantité de 30% à 60%,
- De l'o-tocophérol présent en quantité de 0,25% à 1%, avantageusement de 0,5%,
- De l'arginine en quantité de 5% à 25%.
Une autre composition avantageuse comprend au moins encore, en poids par rapport au poids total de la composition :
-Un extrait de P. tricornutum en quantité de 10% à 25%, avantageusement de 11% à 16%,
-Une huile MCT présente en quantité de 30% à 60%,
-De l'a-tocophérol présent en quantité de 0,5%,
-Un extrait de guarana P. cupanà) en quantité de 35% à 55%, avantageusement un extrait de graines de guarana P. cupanà).
En outre, la composition de l'invention comprend au moins un additif choisi parmi les agents conservateurs, les colorants, les arômes, les agents de délitement, les agents lubrifiants, les agents d'enrobage ou d'encapsulation. La composition se présente sous la forme de gélules, de capsules, de comprimés, de pastilles, de liquide ou de poudre libre. Elle est avantageusement conditionnée en doses ayant un poids unitaire compris entre llmg et 2g, avantageusement entre 20mg et 2g. Lorsqu'elle se présente sous forme liquide, la composition peut être intégrée à une boisson énergisante, un jus de fruit, un sirop, de l'eau, ou bien être formulée sous forme de gélules de type « soft gels ».
Elle est à usage de complément alimentaire, particulièrement à destination des sportifs, des joueurs vidéo, des étudiants. Dans ce cas, la composition est administrée par voie orale à une dose comprise entre 1 g et 2 g durant une période d'au moins 1 jour, avantageusement d'au moins 15 jours, très avantageusement de 1 mois. Alternativement, la composition est utilisée en tant que complément alimentaire chez les animaux, avantageusement les animaux domestiques, encore avantageusement le chien et le chat, très avantageusement le chien. Dans ce cas, la composition est administrée par voie orale seule ou mélangée à un aliment, à une dose journalière comprise entre llmg et 25mg par kg de masse corporelle.
La composition selon l'invention est utilisée pour maintenir et/ou augmenter la vitesse de traitement des données et/ou la concentration chez des mammifères sains, avantageusement des individus sains.
Des exemples faisant référence à la description sont présentés ci-après. Ces exemples sont illustratifs et ne sauraient limiter la portée de l'invention. Les exemples font partie intégrante de la présente invention et toute caractéristique
nouvelle par rapport à un état de la technique antérieure à partir de la description prise dans son ensemble fait partie intégrante de l'invention.
Sauf mention contraire, les pourcentages sont exprimés en poids/poids, la température est donnée en degré Celsius et le temps est donné en millisecondes (ms).

Exemples
Exemple 1 : mode de préparation d'un extrait de Phaeodactylum tricornutum
L'extrait de microalgue de P. tricornutum est obtenu à partir de la biomasse d'une culture de la microalgue d'une souche dont l'origine est la France. La
biomasse est au préalable centrifugée puis filtrée pour éliminer l'eau. Une étape d'extraction solide-liquide est ensuite mise en œuvre.
L'extrait est ainsi obtenu par extraction dans un mélange eau/éthanol en proportion de 40/60 (p/p) à 1/100 (p/p), à une température de 20°C, c'est-à-dire à température ambiante, durant une période de 2 heures. Il est ensuite filtré puis séché par lyophilisation. Il se présente sous forme de poudre.
Exemple 2 : mode de préparation d'un extrait de guarana Paullina cupana) et association d'un extrait de Phaeodactyium tricornutum et de l'extrait de guarana (P, cupana
2. a) : préparation d'un extrait de guarana :
L'extrait de guarana est obtenu par extraction des graines de la plante Paullinia cupana dont l'origine est la France, dans un mélange eau/éthanol en proportion (20,80 ; v/v).
L'extraction est conduite à une température de 80°C durant une période de 2 heures.
L'extrait est ensuite filtré puis séché par lyophilisation. Il se présente sous forme de poudre.
2. b) Extrait de guarana commercial
L'extrait hydroalcoolique (eau/éthanol) de graines de la plante Paullinia cupana (guarana) peut être acheté auprès de la société Natac ou la société Nexira. Il se présente sous forme de poudre.
2.c) Préparation de l'association extrait de Phaeodactyium tricornutum et extrait de quarana {P. cupana)
L'extrait de guarana obtenu selon l'exemple 2. a) ou 2. b) peut être mélangé à la poudre obtenue dans l'exemple 1 pour obtenir l'association selon l'invention dans les proportions en poids respectives d'extrait de P. tricornutum et de guarana de 2/9 à 3/7 (exemple 2cl) ou de 3/1 à 7/2 (exemple 2c2).
Exemple 3 : Effet d'un extrait de Phaeodactylum tricornutum sur le maintien et/ou l'augmentation de la vitesse de traitement des données et/ou la concentration chez des individus sains.
Protocole : Une étude clinique en double aveugle avec placebo a été menée sur une population composée de 51 hommes et 10 femmes recrutés en tant que joueurs de jeux-vidéos expérimentés (21.7±4 ans d'âge moyenne, 73.0±13 kg de poids moyen, 24.2±3.6 kg/m2 de répartition massique moyenne). La population d'intérêt a été divisée au hasard en 3 groupes et s'est vu administrer les suppléments comme suit :
-Groupe contrôle : 2 gélules placebo de 436 mg/jour (436 mg d'huile de tournesol en gélule ressemblant à une gélule comprenant l'extrait de P. tricornutum) et 1 gélule de 500 mg/jour contenant de la microcellulose (ressemblant à une gélule de guarana).
-Traitement 1 - dit de faible dose : combinaison d'une gélule comprenant l'extrait de P. tricornutum tel que préparé selon l'exemple 1 (dose : 436mg), de l'o- tocophérol et une huile MCT, d'une gélule placebo de 436 mg/jour et d'une gélule d'extrait de guarana de 500 mg/jour.
-Traitement 2 - dit de dose élevée : combinaison de 2 gélules comprenant chacune l'extrait de P. tricornutum tel que préparé selon l'exemple 1 (dose : 436mg), de l'o-tocophérol et une huile MCT, et d'une gélule d'extrait de guarana (500 mg/jour).
Les suppléments ont été préparés pour maintenir l'administration en double aveugle pendant toute la durée de l'étude. Les critères d'inclusions étaient les suivants :
1.) Hommes et femmes en bonne santé ;
2.) Entre 18 et 40 ans ;
3.) Antécédents autodéclarés de jeu vidéo pendant 5 heures ou plus par semaine pendant les 6 mois précédant le dépistage ;
4.) Indice de Masse Corporelle (IMC) compris entre 18 et 34,9 Kg/m2 ;
5.) Les sujets ont accepté de fournir leur propre jeu vidéo d'action ou de stratégie orienté opérateur, auquel ils ont joué au moins 21 fois au cours des 3 derniers mois précédent le début de l'étude clinique ;
6.) Pas d'ingestion récente (<2 semaines) de complément alimentaire affectant la fonction cognitive ;
7.) Être capable de donner un consentement éclairé écrit et de consommer le produit expérimental quotidiennement pendant la durée de l'étude ;
8.) Vivre librement (vivre dans une maison privée, seul ou en famille, et être capable de maintenir sa santé et son hygiène sans assistance).
9.) Est disposé à maintenir une durée de sommeil constante le soir précédant les visites d'étude.
10.) Sont d'accord pour continuer leur utilisation habituelle du jeu entre les visites d'étude.
Les critères de non inclusions étaient les suivants (Les participants n'étaient pas autorisés à participer à l'étude dans les cas) :
1.) Consommation de caféine et d'alcool durant les 12 heures précédant chaque visite de l'étude ;
2.) Consommation de compléments alimentaires susceptibles d'affecter la cognition et/ou ayant un effet stimulant au moins 7 jours avant la visite 2 ;
3.) Femmes enceintes, femmes allaitant ou souhait de grossesse ;
4.) Présence d'un trouble psychotique ou dépressif majeur non traité ou tout antécédent de déficit cognitif ;
5.) Hypertension, diabète, maladie de la thyroïde, maladie cardiaque non contrôlée, cancer.
6.) Maladie neurologique importante ;
7.) Changements majeurs prévus dans le mode de vie (régime alimentaire, niveau d'exercice, déplacements) pendant la durée de l'étude ;
8.) Antécédents d'abus d'alcool ou de drogues au cours des 12 derniers mois ;
9.) Allergie connue à l'un des ingrédients du produit complémentaire ;
10.) Individus non disposés à fournir leur propre système de jeu et/ou leur propre jeu.
Au cours d'une séance de familiarisation (Visite 1), les participants ont été informés de l'étude et ont signés un consentement éclairé. Les participants ont répondu à des questionnaires sur leurs antécédents médicaux, subi un examen physique général comprenant la détermination de leur taille, de leur poids, de leur fréquence cardiaque au repos et de leur tension artérielle, et ont été informés des méthodes générales de l'étude. Les personnes admissibles à
participer ont été programmées pour les évaluations de base et s'exercer aux évaluations de l'étude. Il a été demandé aux participants de noter leur consommation d'aliments et de liquides pendant les 4 jours précédant le test, de s'abstenir de consommer des quantités atypiques de caféine et d'autres stimulants non consommés normalement dans leur régime alimentaire pendant 48 heures, et de rester à jeun pendant 12 heures avant le test. Au cours de cette visite, la classification du jeu vidéo (orienté action ou stratégie) a été confirmée. Chaque sujet devait apporter le jeu vidéo choisi et approuvé au préalable, ainsi qu'une plate-forme de jeu compatible et ses accessoires. Les sujets devaient avoir joué au jeu choisi au moins 21 fois au cours des 3 mois précédant la sélection. Les sujets ont accepté d'apporter et de jouer au même jeu vidéo à la visite 1 et à la visite 2 et de jouer au jeu choisi de manière régulière, sans excès entre les visites de l'étude, afin de minimiser les changements de biais de courbe d'apprentissage. Le dernier score du jeu choisi avant le dosage a été enregistré. Il était fourni sous forme de photo ou de capture d'écran, mais pas sous forme de réponse verbale.
Expérience 1 (Visite 2) : Les participants se sont présentés au laboratoire et ont rendu un journal alimentaire de 4 jours, ont été pesés, ont rempli des questionnaires sur la sensibilité aux stimulants et les effets secondaires, et ont effectué les tests de go no-go, de Sternberg. Ils ont ensuite été répartis au hasard pour ingérer les suppléments tel que décrits. Quinze minutes après l'ingestion, les participants ont répétés les tests (Post-15-SUPP). La durée entre l'ingestion du traitement et le début du jeu vidéo a été enregistrée. Les participants ont joué ensuite à leur jeu vidéo pendant 1 heure. Immédiatement après, les participants ont effectué à nouveau la batterie de tests ainsi qu'un test sur les stimulants et les effets secondaires. Les participants sont ensuite sortis du laboratoire et ont reçu la quantité appropriée du traitement assigné avec des instructions sur la façon de le prendre (Post-Gaming).
Expérience 2 (Visite 3) : Après 4 semaines de supplémentation quotidienne, les participants se sont présentés au laboratoire et ont rendu les journaux alimentaires de 4 jours, ont été pesés et ont effectué la même batterie de tests que précédemment (Expérience 1) (Pre-SUPP). Ensuite, ils ont ingéré le
traitement attribué à l'étude et/ou le placebo juste après les tests de pré-jeu et ils ont attendu la même durée que celle enregistrée lors de l'expérience 1. Les participants ont joué ensuite à leur jeu vidéo (même jeu vidéo que lors de l'expérience 1) pendant 1 heure. Immédiatement après, les participants ont refait la batterie de tests et ont complété des questionnaires sur la sensibilité aux stimulants et les effets secondaires ainsi qu'un test sur les stimulants (Post- Gaming). Cette étude a été menée conformément aux directives et aux principes de la Food and Drug Administration (FDA), de la Conférence internationale sur l'harmonisation (ICH) et des bonnes pratiques cliniques (BPC).
Les sujets ont reçu pour instruction de prendre leur supplément dédié une fois par jour pendant 1 mois (±2 jours). La première et la dernière dose ont été administrées sur le site clinique lors de la visite 2 (expérience 1) et de la visite 3 (expérience 2), respectivement.
Les données ont été analysées à l'aide de modèles linéaires généraux (GLM) univariés, multivariés et à mesures répétées, en tenant compte des caractéristiques de base respectives comme covariables si nécessaire. De plus, les changements par rapport à la ligne de base ont été évalués par des moyennes avec des intervalles de confiance à 95% et analysés par une analyse de variance à sens unique. À la fin des tests, une analyse de puissance a également été effectuée.
Résultats : Les résultats du test de Sternberg sont présentés dans les 2 tableaux ci-dessous (Les graphes correspondants sont présentés à la Figure 1 correspondante) et correspondent aux résultats du test de la lere expérience (1 supplémentation unique reçue).

Conclusion : Les résultats ont montré un effet de la combinaison de guarana et de l'extrait de P. tricornutum en comparaison de l'ingrédient placebo, avec un effet prépondérant de l'extrait de P. tricornutum au sein de cette combinaison, sur l'augmentation de la vitesse de traitement des données chez des individus sains, cet effet étant caractérisé à la fois par :
- un temps de réaction plus court des individus ayant reçu une dose faible ou haute de la composition en réponse au stimulus visuel par rapport aux individus du groupe placebo.
- une diminution plus importante du temps de réaction due à la composition comprenant l'extrait de P. tricornutum à haute dose (872mg) qu'à dose faible (436mg), traduisant un bénéfice majoré de l'extrait de P. tricornutum.
- un maintien de ce temps de réaction constant lorsque le nombre de stimuli visuels d (données à analyser) a augmenté. Il est à noter que l'évolution des temps de réactions associées à la supplémentation en fonction du nombre de stimuli visuels (n), sont opposées aux résultats communément obtenus lors du test de Sternberg. En effet, lorsque le nombre de stimuli entrants (le nombre de données à évaluer avant de prendre une décision) augmente, le temps de réaction augmente mécaniquement car le traitement des données est sériel et exhaustif (Sternberg, 1966), et, chaque individu a un temps de traitement natif par unité de donnée constant. Or, la supplémentation de la combinaison de l'extrait de P. tricornutum à faible dose (436mg) et de l'extrait de guarana induit un temps de réaction constant à partir d'un nombre de stimuli de 4 jusqu'à 6. La
supplémentation de la combinaison de l'extrait de P. tricornutum à haute dose (872mg) et de l'extrait de guarana, induit quant à elle un temps de réaction décroissant, dès 2 stimuli et jusqu'à 6.
Exemple 4 : Effet d'un extrait de Phaeodacty/um tricornutum sur le maintien et/ou l'augmentation de la concentration chez des individus sains.
Protocole : le protocole est celui présenté à l'exemple 3.
Résultat : les résultats sont présentés dans le tableau 3 (Figure 2) (Test du Go- NoGo)

Conclusion : la supplémentation de la combinaison de l'extrait de guarana et de la composition comprenant l'extrait de P. tricornutum a amélioré le niveau de précision des réponses apportées après 30 jours de supplémentation, quelle que soit la dose (faible ou haute) de la composition comprenant l'extrait de P. tricornutum, tandis que le niveau de précision diminue dans le groupe placebo (Figure 2). Les bénéfices sur le niveau de concentration sont majorés avec la composition comprenant l'extrait de P. tricornutum à dose élevée (Figure 2 droite).
Exemple 5 : Effet in vitro d'une association synergique d'un extrait de P. tricornutum et d'un extrait de guarana ( . eu pana) sur le maintien et/ou l'augmentation de la vitesse de traitement des données chez des individus sains.
Exemple 5a) Augmentation de la longueur et de la dendricité des prolongements neuronaux.
Des neurones sensitifs ont été obtenus à partir de cellules hiPS (human induced Pluripotent Stem cells) elles-mêmes obtenues à partir de fibroblastes humains d'un donneur sain. Les cellules ont été ensemencées en plaque de 96 puits et maintenues durant une période de 6 jours en culture dans un milieu induisant leur différenciation à une température de 37°C (5% de CO2). Le milieu de culture a été changé tous les 2 jours. Après une période de 8 jours de culture, le milieu de culture a été changé par un milieu de maturation (milieu avec facteurs de croissance dont Nerve Growth Factor). Les cellules ont été maintenues en culture à une température de 37°C sous 5% de CO2.
Après 1 jour d'induction, les facteurs de croissance du milieu de culture ont été éliminés et remplacés par un contrôle comprenant du facteur de croissance NGF seulement ; ou l'extrait de P. tricornutum selon l'exemple 1 ; ou l'extrait de guarana selon les exemples 2a) ou 2b) ; ou encore l'association selon l'exemple 2c) dans les ratios respectifs en poids P. tricornutum/ guarana de 2/9 à 3/7 (exemple 2cl) ou de 3/1 à 7/2 (exemple 2c2).
Les milieux ont été cultivés durant une période de 4 jours puis les cellules ont été récupérées et marquées avec un anticorps anti-p-tubuline ensuite révélé à l'aide d'un anticorps secondaire couplé à un fluorochrome (marquage de la structure des neurones i.e. Corps cellulaire et neurites). La longueur des prolongements neuronaux et le nombre de bifurcations au niveau de ces prolongements ont été mesurés grâce à l'établissement d'un algorithme détectant automatiquement les neurites et leurs bifurcations sur 20 images acquises au x20 pour chaque puits de culture. Une moyenne de 6 puits a été établie pour chaque condition, comparée au contrôle puis une analyse statistique a été conduite (One Way Anova avec correction de Dunnett).
Exemple 5b) Augmentation in vitro de la vitesse de mobilisation du calcium au niveau svnaptigue.
Des cellules de neurones sensitifs telles que décrites à l'exemple 5a) ont été ensemencées en plaque de 96 puits et maintenues durant une période de 6 jours en culture dans un milieu induisant leur différenciation à une température de 37°C (5% de CO2). Le milieu de culture a été changé tous les 2 jours. Après une
période de 9 jours de culture, le milieu de culture a été changé par un milieu de maturation (milieu avec facteurs de croissance dont Nerve Growth Factor) et l'extrait de P. tricornutum selon l'exemple 1 ; ou l'extrait de guarana selon l'exemple 2a) ou 2b) ; ou l'association des deux extraits selon l'exemple 2c) aux ratios respectifs en poids de P. tricornutum/ guarana de 2/9 à 3/7 (exemple 2cl) ou de 3/1 à 7/2 (exemple 2c2) ; ou un contrôle (Facteur de croissance NGF) ont été ajoutés au milieu de culture. Les cellules ont été maintenues en culture à une température de 37°C sous 5% de CO2 durant une période de 19 jours, période après laquelle les récepteurs neuronaux deviennent fonctionnels. Les récepteurs neuronaux TRPM8 (Transient receptor potential cation channel subfamily M (melastatin) member ont été spécifiquement inhibés puis les milieux incubés avec une sonde fluorescente (Fluo-4-AM ; ThermoFisher) afin d'activer spécifiquement ces récepteurs. Une augmentation de la fluorescence traduit une activation des récepteurs, donc une entrée massive de calcium au niveau des neurones, reflétant une augmentation de la vitesse de traitement des données chez les individus sains. Les variations de fluorescence ont été enregistrées sur une période de 2 minutes puis la moyenne pour chaque condition a été calculée (n=6). Une analyse statistique a été conduite (One way Anova).

Claims
1. Utilisation d'un extrait de Phaeodactylum tricornutum seul ou en association avec un extrait choisi parmi un extrait de guarana Pauiiinia cupana , un extrait de Nelumbo nucifera, un extrait de Centella asiatica, un extrait de Piper nigrum, ou l'une quelconque de leur combinaison et/ou un composé choisi parmi la créatine, l'arginine, la caféine, la théophylline, la paraxanthine, la théobromine, l'acide rosmarinique, l'acide ellagique, l'aldéhyde cinnamique ou l'une quelconque de leur combinaison, pour maintenir et/ou augmenter la vitesse de traitement des données et/ou la concentration chez des individus sains ne présentant pas de déclin et/ou de trouble cognitif.
2. Utilisation selon la revendication 1 caractérisée en ce que l'extrait de Phaeodactylum tricornutum est obtenu par extraction dans un solvant ou mélange de solvant, avantageusement dans l'eau, l'acétone, l'hexane, l'acétate d'éthyle, le méthyltétrahydrofurane, le 2-méthyloxolane, l'heptane, un alcool choisi parmi l'éthanol, le méthanol, l'isopropanol, une huile naturelle ou ramifiée, un glycol, un polyol, un mélange eau/alcool, eau/glycol en proportion de 100/1 à 1/100 (p/p), avantageusement dans un mélange eau/éthanol en proportion de 40/60 (p/p) à 1/100 (p/p).
3. Utilisation selon la revendication 1 ou 2 par voie orale.
4. Utilisation selon l'une quelconque des revendications 1 à 3 dans une composition, avantageusement à usage de complément alimentaire.
5. Association d'un extrait de Phaeodactylum tricornutum et d'un extrait choisi parmi un extrait de guarana Pauiiinia cupana'), un extrait de Nelumbo nucifera, un extrait de Centella asiatica, un extrait de Piper nigrum, ou un composé choisi parmi la créatine, l'arginine, la caféine, la théophylline, la paraxanthine, la théobromine, l'acide rosmarinique, l'acide ellagique, l'aldéhyde cinnamique et l'une quelconque de leurs combinaison.
6. Association selon la revendication 5 caractérisée en ce que le ratio en poids entre l'extrait de Phaeodactylum tricornutum et l'extrait ou le composé est de 1/6 à 6/1.
7. Association selon l'une quelconque des revendications 5 à 6 caractérisée en ce que l'extrait est un extrait de guarana Pauiiinia cupanâ), avantageusement un extrait de graines.
8. Association selon la revendication 7 caractérisée en ce qu'elle contient :
Un extrait de P. tricornutum présent en quantité comprise entre 50mg et 600mg, avantageusement de 220mg à 440mg,
Un extrait de guarana P. cupanâ) présent en quantité comprise entre 300mg et 700mg, avantageusement de 500mg.
9. Composition, avantageusement à usage de complément alimentaire, comprenant l'association selon l'une quelconque des revendications 5 à 8, une huile végétale et de la vitamine E, caractérisée en ce que l'huile végétale est présente dans la composition en concentration de 30% à 60% en poids et la vitamine E est présente en concentration de 0,25 à 1% en poids par rapport au poids total de la composition.
10. Composition selon la revendication 9 caractérisée en ce qu'elle comprend :
-Un extrait de P. tricornutum en quantité de 10% à 25%, avantageusement de 11% à 16%
-Une huile MCT présente en quantité de 30% à 60%,
-De l'o-tocophérol présent en quantité de 0,5%,
-Un extrait de guarana P. cupanâ) en quantité de 35% à 55%, avantageusement un extrait de graines.
11. Composition selon la revendication 9 ou 10 caractérisée en ce qu'elle comprend en outre au moins un additif choisi parmi les agents conservateurs, les colorants, les arômes, les agents de délitement, les agents lubrifiants, les agents d'enrobage ou d'encapsulation.
12. Composition selon l'une quelconque des revendications 9 à 11 caractérisée en ce qu'elle se présente sous la forme de gélules, de capsules, de comprimés, de pastilles, de liquide ou de poudre libre.
13. Composition selon l'une quelconque des revendications 9 à 12 caractérisée en ce qu'elle est conditionnée en doses ayant un poids unitaire compris entre 20 mg et 2 g.
14. Composition selon l'une quelconque des revendications 9 à 13 caractérisée en ce qu'elle est administrée à une dose comprise entre lg et 2g durant une période d'au moins 1 jour, avantageusement d'au moins 15 jours, très avantageusement de 1 mois.
15. Utilisation de l'association selon l'une quelconque des revendications 5 à 8 ou de la composition selon l'une quelconque des revendications 9 à 14 pour maintenir et/ou augmenter la vitesse de traitement des données et/ou la concentration chez des individus sains ne présentant pas de déclin et/ou de trouble cognitif.
