Abstract
본 발명은 세포에서 분리된 미토콘드리아를 동결 및/또는 동결건조하는 방법, 및 동결 및/또는 동결건조된 미토콘드리아의 용도에 관한 것이다. 본 발명에 따른 동결된 미토콘드리아는 동결되지 않은 미토콘드리아와 유사한 항염증 활성 및 에너지 대사 관련 효소의 활성을 나타내었다. 또한, 동결건조된 미토콘드리아도 수복 후 동결건조 전의 미토콘드리아와 유사한 수준의 항염증 활성, 에너지 대사 관련 효소들의 활성 및 장기 보존 효능을 나타내었다. 따라서, 본 발명에 따른 동결 및/또는 동결건조된 미토콘드리아는 미토콘드리아 기능저하 또는 기능장애로 인해 유발되는 다양한 질환 치료에 분리된 미토콘드리아를 폭넓게 적용할 수 있게 함으로써 미토콘드리아를 포함하는 치료제의 상업적 이용 가능성을 크게 향상시킬 것으로 기대된다.
Description
본 발명은 세포에서 분리된 미토콘드리아를 동결 및/또는 동결건조하는 방법, 및 동결 및/또는 동결건조된 미토콘드리아의 용도에 관한 것이다. 보다 상세하게는, 미토콘드리아를 동결 및/또는 동결건조하는 방법, 및 상기 방법에 의해 동결 및/또는 동결건조된 미토콘드리아를 유효성분으로 포함하는 약학 조성물 및 장기 보존액 조성물에 관한 것이다.
동결건조는 수분을 함유하는 시료를 동결하고 감압 하에 두어 시료 중의 수분을 승화시켜 제거하는 건조법으로서, 식물이나 동물의 세포, 조직, 또는 식품 등의 물을 포함하는 물질을 장기간 보존하기 위해 사용되고 있다.
그러나 물을 포함하는 물질을 동결하면 동결 중에 물 분자끼리 얼음 결정(빙정)을 형성하여, 물을 포함하는 물질 중에서 용질이나 혼입물의 매질이 불균일하게 확산되는 동결 농축이 발생하여 조직의 안정성을 높이기 위하여 동결 보호제를 사용한다. 그러나 진핵세포의 경우 동결 보호제를 사용하여도 동결건조 시 세포막 등의 세포 구조의 손상 없이 세포를 안정하게 보존하기 어렵고, 이에 따라 세포 내 유효 성분의 기능을 유지할 수 없게 되는 문제점이 있다.
미토콘드리아는 에너지 공급원으로서 ATP 합성, 활성산소 생산, 세포사멸 등 다양한 생리과정에서 핵심적인 역할을 한다. 따라서, 미토콘드리아가 손상되면 다양한 질병이 유발될 수 있는데, 대부분의 미토콘드리아 장애는 미토콘드리아 DNA에서 발생하는 유전성 또는 후천성 돌연변이에 기인한다. 예를 들어, 미토콘드리아 막전위 이상으로 인한 팽윤, 활성산소종 또는 자유라디칼 등에 의한 산화적 스트레스, 그리고 미토콘드리아의 에너지 생성을 위한 산화적 인산화 기능의 결함 등에 의해 미토콘드리아의 기능이 변형될 수 있다. 이러한 미토콘드리아의 기능 이상은 미토콘드리아 유전병, 류마티스 관절염과 같은 염증성 질환, 허혈성 질환, 감염성 질환, 심장질환, 근질환, 파킨슨병이나 알츠하이머병 등과 같은 퇴행성 질환 및 각종 암의 발생과 암전이 등이 보고된 바 있다.
이러한 미토콘드리아 기능장애와 관련된 질환을 치료하기 위하여, 세포나 조직으로부터 미토콘드리아를 분리하여 이를 치료의 목적으로 생체에 다시 주입하는 시도가 최근 증가하고 있다(대한민국 등록특허 제10-2126199호, 대한민국 등록특허 제10-2019277호).
이와 같이, 미토콘드리아를 이용한 특정 질환의 치료에 대한 가능성 제시 등 다양한 연구가 이루어지고 있음에도 불구하고, 미토콘드리아를 안정적으로 유지 및 보관할 수 있는 방법, 및 안정적인 제형과 관련한 응용기술의 개발은 미진한 상황이다. 따라서, 미토콘드리아와 이에 포함된 생체활성 인자들을 안정적으로 유지·보관할 수 있는 새로운 방법 개발이 필요하다.
이에 본 발명자들은 미토콘드리아 관련 질환 치료의 일환으로서 이식 치료를 위한 미토콘드리아를 효율적으로 보존하기 위한 방법을 연구하였다. 그 결과, 본 발명의 방법으로 동결건조된 미토콘드리아가 상온 및 저온에서 보관 가능하면서, 동결 및/또는 동결건조된 미토콘드리아는 동결 전의 미토콘드리아의 활성을 유지하는 것을 확인함으로써 본 발명을 완성하였다.
상기 과제를 해결하기 위하여, 본 발명의 일 측면은 동결 또는 동결건조된 미토콘드리아 및 이를 포함하는 약학 조성물 및 장기 보존액 조성물 키트를 제공한다.
본 발명의 다른 측면은 분리된 미토콘드리아를 안정적으로 동결 또는 동결건조 시키는 방법 및 동결 보호제; 및 상기 동결 보호제 및 미토콘드리아를 포함하는 동결 조성물을 제공한다.
본 발명의 또 다른 측면은 분리된 동결 또는 동결건조된 미토콘드리아의 미토콘드리아 관련 질환의 예방 또는 치료용 용도를 제공한다.
본 발명의 또 다른 측면은 분리된 동결 또는 동결건조된 미토콘드리아를 투여하는 단계를 포함하는 미토콘드리아 관련 질환의 예방 또는 치료 방법을 제공한다.
본 발명에 따른 동결 및/또는 동결건조된 미토콘드리아는 동결되지 않은 미토콘드리아와 유사한 것을 확인하였다. 또한, 동결 및/또는 동결건조된 미토콘드리아는 동결건조되지 않은 미토콘드리아와 유사한 수준의 항염증 활성 및 에너지 대사 관련 효소들의 활성을 나타내었다. 또한, 동결건조된 미토콘드리아는 동결건조되지 않은 미토콘드리아와 유사한 수준의 장기 보존 효능을 나타내었다. 따라서, 본 발명에 따른 동결 및/또는 동결건조된 미토콘드리아는 미토콘드리아 기능저하 또는 기능장애로 인해 유발되는 다양한 질환 치료에 분리된 미토콘드리아를 폭넓게 적용할 수 있게 함으로써 미토콘드리아를 포함하는 치료제의 상업적 이용 가능성을 크게 향상시킬 것으로 기대된다.

동결 및/또는 동결건조된 미토콘드리아
본 발명의 일 측면은 동결 및/또는 동결건조된 미토콘드리아를 제공한다.
본 명세서에 사용된 용어 "미토콘드리아(mitochondria)"는 세포 내 에너지 공급원인 아데노신 트라이포스페이트(ATP)의 합성 및 조절에 관여하는 진핵세포의 세포 소기관으로, 생체 내 다양한 대사 경로, 예를 들어, 세포 신호처리, 세포 분화, 세포 사멸 뿐만 아니라, 세포 주기 및 세포 성장의 제어와 연관이 있다. 따라서, 유전적, 환경적 또는 원인 불명의 원인으로 인한 미토콘드리아 기능저하 또는 기능장애는 미토콘드리아 관련 유전병, 류마티스 관절염과 같은 염증성 질환, 허혈성 질환, 감염성 질환, 심장 질환, 근 질환, 파킨슨병이나 알츠하이머병 등과 같은 퇴행성질환 및 각종 암의 발생과 암 전이 등과 같은 다양한 질환의 발병에 관련 있다고 보고된 바 있다.
상기 미토콘드리아는 포유동물로부터 수득된 것일 수 있으며, 인간으로부터 수득된 것일 수 있다. 구체적으로, 상기 미토콘드리아는 세포 또는 조직으로부터 분리된 것일 수 있으며, 혈액세포 또는 혈소판으로부터 분리된 것일 수 있다. 또한, 상기 미토콘드리아는 미토콘드리아의 생물학적 활성이 정상인 세포로부터 수득된 정상적인 미토콘드리아일 수 있다. 이때, 상기 미토콘드리아는 조직 또는 세포를 농축하여 파쇄 후 분리하여 사용하거나 동결 보관한 후 해동한 조직 또는 세포 시료로부터 파쇄 후 분리된 것일 수 있다. 또한, 상기 미토콘드리아는 세포 및 조직을 체외에서 배양하여 분리하거나 동결 보관한 후 해동한 시료로부터 분리된 것일 수 있다. 예를 들어, 상기 미토콘드리아는 체세포, 생식세포 또는 줄기세포로부터 수득된 것일 수 있다.
구체적으로, 상기 체세포는 근육세포, 간세포, 신경세포, 섬유아세포, 상피세포, 지방세포, 골세포, 백혈구, 림프구, 혈소판 또는 점막세포일 수 있다.
또한, 상기 줄기세포는 여러 종류의 조직 세포로 분화할 수 있는 능력을 가진 미분화 세포로서, 이에 제한되지는 않으나 중간엽줄기세포, 성체줄기세포, 역분화줄기세포, 배아줄기세포, 골수줄기세포, 신경줄기세포, 윤부줄기세포 및 조직 유래 줄기세포로 구성된 군으로부터 선택되는 어느 하나일 수 있다. 이때, 상기 중간엽줄기세포는 탯줄, 제대혈, 골수, 지방, 근육, 신경, 피부, 활액, 정소, 양막 및 태반으로 구성된 군으로부터 선택되는 어느 하나로부터 수득된 것일 수 있다.
또한, 상기 미토콘드리아는 자가(autologous), 동종(allogenic) 또는 이종(xenogenic)으로부터 수득된 것일 수 있다. 구체적으로, 자가 미토콘드리아는 동일 개체의 조직 또는 세포로부터 수득된 미토콘드리아를 의미한다. 또한, 동종 미토콘드리아는 개체와 같은 종에 속하면서 대립유전자에 대해서는 다른 유전자형을 가지는 개체로부터 수득된 미토콘드리아를 의미한다. 또한, 이종 미토콘드리아는 개체와 다른 종에 속하는 개체로부터 수득된 미토콘드리아를 의미한다. 또한, 이종 미토콘드리아는 개체와 다른 종에 속하는 개체로부터 수득된 미토콘드리아를 의미한다.
또한, 상기 미토콘드리아는 세포에서 분리된 미토콘드리아일 수 있다. 또한, 상기 미토콘드리아는 손상되지 않고, 미토콘드리아 활성을 가지고 있는 것일 수 있다.
본 발명의 동결된 미토콘드리아는 냉장에서 해동될 수 있다. 구체적으로 약 0℃ 내지 약 10℃에서 해동될 수 있다. 또한, 상기 해동된 미토콘드리아는 냉장에서 보관될 수 있다. 구체적으로 약 0℃ 내지 약 10℃에서 보관될 수 있다.
본 발명의 동결건조된 미토콘드리아는 상온 또는 냉장에서 보관할 수 있다. 구체적으로, 상기 미토콘드리아는 약 0℃ 내지 약 37℃에서 보관할 수 있다. 상기 미토콘드리아는 약 0℃ 내지 약 8℃, 약 9℃ 내지 약 15℃, 약 16℃ 내지 약 22℃, 약 23℃ 내지 약 29℃ 또는 약 30℃ 내지 약 37℃에서 보관할 수 있다.
또한, 상기 동결건조된 미토콘드리아는 수복하여 사용 시, 미토콘드리아 활성을 나타낼 수 있다.
본 명세서에서 사용된 용어, "수복"은 동결건조된 미토콘드리아를 원상태로 회복시키는 것을 의미하며, 수용액을 상기 동결건조된 미토콘드리아에 첨가함으로써 수행될 수 있다. 이때, 주사용수, 생리적 식염수, 인산염 완충용액, 정제수 또는 탈이온수일 수 있다. 바람직하게는 탈이온수일 수 있다.
이때, 상기 수복과정은 상온 또는 냉장에서 수행될 수 있다. 구체적으로, 약 0℃ 내지 37℃에서 수행될 수 있다. 상기 미토콘드리아는 약 0℃ 내지 약 8℃, 약 9℃ 내지 약 15℃, 약 16℃ 내지 약 22℃, 약 23℃ 내지 약 29 또는 약 30℃ 내지 약 37℃에서 수행될 수 있다. 또한, 상기 수복과정은 약 1분 내지 약 1시간, 약 1분 내지 약 50분, 약 1분 내지 약 40분, 약 1분 내지 약 30분, 약 1분 내지 약 20분, 약 1분 내지 약 10분 또는 약 1분 내지 약 5분 동안 수행될 수 있다. 상기 수복과정은 동결건조된 미토콘드리아가 수복되어 건조되어 있지 않은 상태로 되는 한, 특별히 제한되지 않는다.
본 발명의 일 구체예에서 상기 동결건조된 미토콘드리아를 수복하여 사용 시, 미토콘드리아에 존재하는 에너지 합성 관련 효소들의 활성이 동결건조되지 않은 미토콘드리아와 유사한 것을 확인할 수 있었다(도 5).
또한, 동결 및/또는 동결건조된 미토콘드리아는 항염증 및 분리된 장기의 보호 효능을 나타낼 수 있다. 본 발명의 일 구체예에서 혈소판 또는 탯줄 유래 중간엽 줄기세포 유래의 동결 및/또는 동결건조된 미토콘드리아를 대식세포에 처리 후 염증반응을 유도 시, 염증반응 유도가 억제된 것을 확인할 수 있었다(도 1, 도 2, 도 6 및 도 7). 또한, 상기 동결건조된 미토콘드리아를 포함하는 장기 보관 용액에 분리된 심장을 보관 시, 심장 보호 효능을 나타냈다(도 8).
동결 및 동결건조 방법
본 발명의 또 다른 측면은 분리된 미토콘드리아를 안정적으로 동결 및/또는 동결건조 시키는 방법을 제공한다.
상기 동결 방법은 미토콘드리아를 분리하는 단계; 및 분리된 미토콘드리아를 글라이신을 포함하는 미토콘드리아의 동결 보호제를 처리하여 냉동시키는 단계를 포함할 수 있다. 이때, 상기 동결 보호제는 트레할로오스, 트리스 및 이들의 조합으로 이루어진 군에서 선택되는 어느 하나를 추가로 포함할 수 있다.
또한, 상기 동결건조 방법은 미토콘드리아를 분리하는 단계; 분리된 미토콘드리아를 글라이신을 포함하는 미토콘드리아의 동결 보호제로 처리하여 냉동시키는 단계; 및 냉동된 미토콘드리아를 동결건조 시키는 단계를 포함할 수 있다. 여기서 미토콘드리아는 상술한 바와 동일하다. 이때, 상기 동결 보호제는 트레할로오스, 트리스 및 이들의 조합으로 이루어진 군에서 선택되는 어느 하나를 추가로 포함할 수 있다.
상기 미토콘드리아를 특정 세포로부터 분리하는 경우에는, 예를 들어, 특정 버퍼 용액을 사용하거나 전위차 및 자기장을 이용하는 등 공지된 다양한 방법을 비롯하여 이를 변형한 다양한 방법을 통해 미토콘드리아를 분리할 수 있다.
상기 미토콘드리아 분리는 미토콘드리아 활성 유지 측면에서, 조직 또는 세포를 파쇄하고 원심분리하여 수득할 수 있다. 일 구체예로, 세포를 배양하고, 이러한 세포를 포함하는 조성물을 제1차 원심분리하여 펠렛을 생성하는 단계, 상기 펠렛을 버퍼 용액에 재현탁시키고, 균질화하는 단계, 상기 균질화된 용액을 제2차 원심분리하여 상층액을 제조하는 단계 및 상기 상층액을 제3차 원심분리하여 미토콘드리아를 정제하는 단계로 수행될 수 있다. 이때, 제2차 원심분리가 수행되는 시간은 제1차 및 제3차 원심분리가 수행되는 시간보다 짧도록 조절되는 것이 세포 활성 유지 면에서 바람직하며, 제1차 원심분리에서 제3차 원심분리로 갈수록 속도를 높일 수 있다.
구체적으로, 상기 제1차 내지 제3차 원심분리는 약 0℃ 내지 약 10℃의 온도, 바람직하게는 약 3℃ 내지 약 5℃의 온도에서 수행될 수 있다. 또한, 상기 원심분리가 수행되는 시간은 약 1분 내지 50분 동안 수행될 수 있으며, 원심분리 횟수 및 샘플의 함량 등에 따라 적절히 조정될 수 있다.
아울러, 상기 제1차 원심분리는 약 100xg 내지 약 1,000xg, 약 200xg 내지 약 700xg 또는 약 300xg 내지 약 450xg의 속도로 수행될 수 있다. 또한, 상기 제2차 원심분리는 약 1xg 내지 약 2,000xg, 약 25xg 내지 약 1,800xg 또는 약 500xg 내지 약 1,600xg의 속도로 수행될 수 있다. 또한, 상기 제3차 원심분리는 약 100xg 내지 약 20,000xg, 약 500xg 내지 약 18,000xg 또는 약 800xg 내지 약 15,000xg의 속도로 수행될 수 있다.
아울러, 상기 수득된 미토콘드리아의 안정화를 위하여 약학적으로 허용 가능한 당이 사용될 수 있다. 구체적으로, 수크로오스(sucrose), 만니톨(mannitol), 트레할로오스(trehalose) 등의 당일 수 있으며, 이에 제한되는 것은 아니다. 또한, pH 완충제로서 이에 제한되지는 않으나, 약학적으로 허용가능한 트리스(tris), HEPES(hydroxyethyl piperazine ethane sulfonic acid), 인산(phosphate) 등이 사용될 수 있다.
한편, 상기 미토콘드리아 수득 후 이온(ion) 유출에 의한 손상을 제거하고, 산화 스트레스를 억제하기 위한 킬레이터, 항산화제 등의 첨가제를 사용할 수 있으며, 이는 당업계에서 널리 알려진 시약을 제한없이 포함하여 사용할 수 있다. 예컨대, EDTA, EGTA, 시트레이트(citrate), 글라이신(glycine), 타우린(taurine), ATP 등일 수 있으며, 이에 제한되는 것은 아니다.
또한, 상기 미토콘드리아 분리는 미토콘드리아 활성 유지 측면에서, 냉동된 세포 또는 조직을 해동하여 파쇄하고 원심분리하여 수득할 수 있다. 상기 미토콘드리아 수득 방법은 세포 또는 조직을 냉동하는 단계, 상기 세포 또는 조직을 해동하는 단계 및 해동된 세포 및 조직을 파쇄하는 단계로 수행될 수 있다.
구체적으로, 본 발명에서 상기 분리된 미토콘드리아는 글라이신을 포함하는 미토콘드리아의 동결 보호제와 혼합되어 냉동될 수 있다. 이때, 상기 동결 보호제는 트레할로오스, 트리스 및 이들의 조합으로 이루어진 군에서 선택되는 어느 하나를 추가로 포함할 수 있다. 본 발명에서 상기 동결 보호제는 수용액 형태로 사용할 수 있으며, 상기 수용액은 주사용수, 생리적 식염수, 인산염 완충용액, 정제수 또는 탈이온수일 수 있다.
상기 동결 보호제는 미토콘드리아를 동결시킬 시, 미토콘드리아의 안정성을 유지 및/또는 강화할 수 있으며, 미토콘드리아의 활성 또한 안정적으로 유지시킬 수 있다.
본 발명의 동결 보호제는 글라이신(glycine)을 포함할 수 있다. 상기 글라이신은 이에 제한되지는 않으나, 상기 보호제 내에 약 15 mM 이상의 농도로 포함될 수 있다. 구체적으로는 약 15 mM 내지 약 150 mM의 농도, 약 17 mM 내지 약 130 mM의 농도, 약 20 mM 내지 약 120 mM의 농도, 약 22 mM 내지 약 110 mM의 농도 또는 약 25 mM 내지 약 100 mM의 농도로 포함될 수 있다. 또한, 상기 글라이신(glycine)은 이에 제한되지는 않으나 히스티딘, 이소류신, 류신, 리신아세테이트, 메티오닌, 페닐알라닌, 트레오닌, 트립토판, 발린, 알라닌, 아르기닌, 아스파라긴산, 시스테인, 글루타민산, 프롤린, 세린 및 티로신으로 이루어진 군으로부터 하나 이상 선택되는 아미노산과 함께 사용 가능하다.
상기 동결 보호제는 당류를 추가로 포함할 수 있다. 이때, 당류는 수크로오스(sucrose), 트레할로오스(trehalose), 만니톨(mannitol), 소르비톨, 글루코오스, 프락토오스, 만노오스, 말토오스, 락토오스, 이소말토오스, 덱스트란 및 덱스트린으로 이루어진 군으로부터 선택되는 하나 이상인 것일 수 있다. 바람직하게, 상기 당류는 트레할로오스일 수 있다. 구체적으로 상기 동결 보호제 내 트레할로오스는 약 40 mM 내지 약 400 mM의 농도, 약 50 mM 내지 약 350 mM의 농도, 약 60 mM 내지 약 300 mM의 농도, 약 70 mM 내지 약 250 mM의 농도 또는 약 80 mM 내지 약 200 mM의 농도로 포함될 수 있다.
상기 동결 보호제는 버퍼를 추가로 포함할 수 있다. 상기 주사용 액상 조성물 내 포함되는 상기 버퍼는 이에 제한되지는 않으나 트리스(tris) 버퍼, HEPES 버퍼, MOPS(3-(N-morpholino)propanesulfonic acid) 버퍼 및 아세테이트 또는 포스페이트 함유 버퍼로 이루어진 군으로부터 선택되는 것일 수 있으나, 이에 제한되지 않는다. 이때, 상기 버퍼의 pH는 이에 제한되지는 않으나, 약 7.0 내지 약 7.8 범위, 약 7.2 내지 약 7.6 범위 또는 약 7.3 내지 약 7.5 범위일 수 있다. 바람직하게, 본 발명에서 상기 버퍼는 트리스(tris) 버퍼일 수 있다. 구체적으로 상기 동결 보호제는 약 5 mM 내지 약 50 mM의 농도, 약 8 mM 내지 약 40 mM의 농도, 약 10 mM 내지 약 35 mM의 농도, 약 13 mM 내지 약 30 mM의 농도 또는 약 15 mM 내지 약 25 mM의 농도로 포함될 수 있다.
보다 구체적으로, 본 발명의 동결 보호제는 글라이신을 포함할 수 있으며, 트레할로오스, 트리스 및 이들의 조합으로 이루어진 군에서 선택되는 어느 하나를 추가로 포함할 수 있다. 일 구체예로 상기 동결 보호제는 글라이신 및 트레할로오스를 포함할 수 있다. 상기 동결 보호제는 글라이신 및 트리스를 포함할 수 있다. 일 구체예로 상기 동결 보호제는 글라이신, 트레할로오스 및 트리스를 포함할 수 있다. 본 발명의 일 실시예에서 글라이신, 트리할로오스 및 트리신을 포함하는 혼합 보존용액(TTG)는 트레할로오스 및 트리스를 포함하는 혼합 보존용액(TT)와 비교하여 동결 및 해동 단계에서 발생하는 미토콘드리아의 손상을 감소시켰다(도 9 내지 도 12).
또한, 상기 동결 보호제는 킬레이트제를 더 포함할 수 있다.
상기 킬레이트제는 이에 제한되지는 않으나, 주사가능한 등급의 EGTA(ethylene glycol tetraacetic acid), EDTA(ethylene diamine tetraacetic acid) 및 BAPTA(1,2-bis(o-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid)로 이루어진 군으로부터 하나 이상 선택되는 것일 수 있다. 상기 킬레이트제는 상기 주사용 액상 조성물에 포함되는 미토콘드리아 수득 후 이온(ion) 유출에 의한 손상을 저해할 수 있다.
또한, 상기 동결 보호제는 첨가제로, 미토콘드리아의 기능 유지 및 활성에 유효한 항산화제, ATP, 마그네슘(magnesium) 등을 포함할 수 있다.
상기 냉동은 이에 제한되지는 않으나, -1℃ 이하일 수 있다. 구체적으로, 약 -5℃ 내지 약 -200℃, 약 -15℃ 내지 약 -180℃, 약 -25℃ 내지 약 -160℃, 약 -40℃ 내지 약 -140℃, 약 -55℃ 내지 약 -120℃, 약 -60℃ 내지 약 -100℃ 또는 약 -70℃ 내지 약 -90℃의 온도 범위로 수행될 수 있다.
상기 냉동은 액체 질소(LN2) 또는 냉동 장치를 이용하여 수행될 수 있다. 구체적으로, 냉동(refrigeration) 또는 동결(freezing) 과정에서 LN2를 사용하여 급속 냉동하거나, 초저온냉동고(deep freezer), 냉동고 등 냉동 장치를 이용하여 수행될 수 있다. 상기 냉동은 미토콘드리아를 냉동 또는 동결할 수 있는 방법인 한, 특별히 제한되지 않는다. 상기 미토콘드리아를 냉동 장치를 이용하여 냉동시키는 경우, 냉동 컨테이너(freezing container)를 사용할 수 있다. 상기 냉동 컨테이너는 1분 당 약 1℃씩 온도를 낮추어 세포를 냉동시킬 수 있다. 바람직하게는, 초저온냉동고를 이용하는 경우 상기 냉동 컨테이너를 사용할 수 있다. 또한, 상기 미토콘드리아를 냉동 장치를 이용하여 냉동시키는 경우, 냉동 컨테이너(freezing container)를 사용하지 않을 수 있다.
상기 냉동은 약 24시간 이상, 약 48시간 이상 또는 약 96시간 이상 동안 수행될 수 있으나, 냉동된 미토콘드리아가 정상적인 활성을 갖는 한, 특별히 제한되지 않는다.
상기 동결건조는 이에 제한되지 않으나, 미토콘드리아를 냉동시키는 단계 및 상기 미토콘드리아를 건조하는 단계로 나누어 수행할 수 있다. 이때, 동결건조는 진공 상태에서 수행할 수 있다. 상기 미토콘드리아의 동결건조는 진공상태에서 미토콘드리아를 냉동한 뒤, 점차 온도를 상승시키면서 수행할 수 있다.
미토콘드리아의 동결건조 시, 상기 냉동 단계는 약 -40℃ 이하에서 수행할 수 있다. 구체적으로, 약 -80℃ 내지 약 -40℃, 약 -70℃ 내지 약 -40℃, 약 -60℃ 내지 약 -40℃ 또는 약 -50℃ 내지 약 -40℃에서 수행할 수 있다. 이때, 상기 냉동은 약 5분 내지 약 120분, 약 10분 내지 약 100분, 약 15분 내지 약 80분, 약 20분 내지 약 60분 또는 약 25분 내지 약 40분 동안 수행할 수 있다. 본 발명의 일 실시예에서 상기 동결은 약 -40℃에서 30분 동안 수행하였다.
미토콘드리아 동결건조 시, 상기 건조 단계는 3단계로 나누어 수행할 수 있다. 제1 단계 건조는 약 -30℃ 내지 약 0℃에서 수행할 수 있다. 보다 구체적으로 약 -40℃ 내지 약 0℃, 약 -30℃ 내지 약 -5℃ 또는 약 -20℃ 내지 약 -10℃에서 수행할 수 있다. 이때, 제1 단계 건조는 약 1시간 내지 약 50시간 동안 수행할 수 있다. 구체적으로 약 1시간 내지 약 50시간, 약 5시간 내지 약 50시간, 약 10시간 내지 약 50시간, 약 20시간 내지 약 50시간 또는 약 30시간 내지 약 50시간 동안 수행할 수 있다. 본 발명의 일 실시예에서 상기 제1 단계 건조는 약 -20℃에서 25시간 동안 수행하였고, 미토콘드리아 상태에 따라 약 -20℃에서 8.3시간 동안 추가 수행하였다.
제2 단계 건조는 약 0℃ 내지 약 10℃ 이하에서 수행할 수 있다. 보다 구체적으로, 약 0℃ 내지 약 10℃, 약 2℃ 내지 약 9℃, 약 4℃ 내지 약 8℃ 또는 약 5℃ 내지 약 7℃에서 수행할 수 있다. 이때, 제2 단계 건조는 약 1시간 내지 약 24시간 동안 수행할 수 있다. 구체적으로, 상기 2단계 건조는 약 1시간 내지 약 16시간, 약 2시간 내지 약 16시간, 약 4시간 내지 약 16시간, 약 6시간 내지 약 16시간 또는 약 8시간 내지 약 16시간 동안 수행할 수 있다. 본 발명의 일 실시예에서 상기 제2 단계 건조는 약 6℃에서 8.3시간 동안 수행하였다.
제3 단계 건조는 약 11℃ 내지 약 30℃ 이하에서 수행할 수 있다. 보다 구체적으로, 약 11℃ 내지 약 30℃, 약 15℃ 내지 약 30℃, 약 20℃ 내지 약 30℃ 또는 약 25℃ 내지 약 30℃에서 수행할 수 있다. 이때, 제3 단계 건조는 약 1시간 내지 약 18시간 동안 수행할 수 있다. 구체적으로, 상기 제3 단계 건조는 약 1시간 내지 약 18시간, 약 3시간 내지 약 18시간 또는 약 6시간 내지 약 18시간 또는 약 9시간 내지 약 18시간 동안 수행할 수 있다. 본 발명의 일 실시예에서 상기 제3 단계 건조는 약 25℃에서 9시간 동안 수행하였다.
상기 동결 보호제와 혼합된 미토콘드리아의 농도는 ㎖당 약 0.1 mg 내지 약 10 mg, 약 0.2 mg 내지 약 9 mg, 약 0.3 mg 내지 약 8 mg, 약 0.4 mg 내지 약 7 mg, 약 0.5 mg 내지 약 6 mg, 약 0.6 mg 내지 약 5 mg, 약 0.7 mg 내지 약 4 mg, 약 0.8 mg 내지 약 3 mg 또는 약 0.9 mg 내지 약 2 mg일 수 있다.
또한, 상기 냉동된 미토콘드리아는 동결건조될 수 있다.
동결 보호제 및 동결 조성물
본 발명의 또 다른 측면은 글라이신을 포함하는 미토콘드리아 동결 보호제, 및 상기 동결 보호제 및 미토콘드리아을 포함하는 동결 조성물을 제공한다. 이때, 상기 동결 보호제는 트레할로오스, 트리스 및 이들의 조합으로 이루어진 군에서 선택되는 어느 하나를 추가로 포함할 수 있다.
상기 미토콘드리아 및 동결 보호제는 상술한 바와 동일하다.
약학 조성물
본 발명의 또 다른 측면은 상기 동결 및/또는 동결건조된 미토콘드리아를 유효성분으로 포함하는 질환의 예방 또는 치료용 약학 조성물을 제공한다.
이때, 약학 조성물의 용도는 미토콘드리아 관련 질환의 예방 또는 치료를 위한 것일 수 있다.
본 명세서에서 사용된 용어 "미토콘드리아 관련 질환"은 유전적, 환경적 또는 원인 불명의 원인으로 인한 미토콘드리아 기능저하 또는 기능장애가 관련된 질환을 총체적으로 의미한다. 예컨대, 상기 질환에는 레버씨 선천성 시신경병증(Leber's Hereditary Optic Neuropathy; LHON), 레이 증후군(Leigh syndrome), 불균일 적색근육 섬유를 갖는 근간대성 간질(Myoclonic Epilepsy Associated with Ragged-Red Fibers; MERRF), 미토콘드리아 근병증, 뇌병증, 고젖산혈증, 뇌졸중(Mitochondrial Myopathy, Encephalopathy, Lactacidosis, Stroke; MELAS) 등의 유전 질환과 파킨슨, 알츠하이머와 같은 퇴행성질환, 당뇨병, 비만과 같은 대사성 질환, 패혈증, 류마티스 관절염, 골 관절염, 전신성 염증 반응 증후군(SIRS)과 같은 염증성 질환, 난청, 암, 허혈성 질환, 신경 질환, 심장 질환, 근 질환, 퇴행성 질환, 섬유화 질환, 눈 질환, 탈모, 신경계 자가면역 질환 등이 포함될 수 있으나, 이에 제한되지 않는다.
상기 암은 이에 제한되지 않으나, 유방암, 폐암, 췌장암, 신경교모종, 위암, 간암, 대장암, 전립선암, 난소암, 자궁경부암, 갑상선암, 후두암, 급성 골수성 백혈병, 뇌종양, 신경모세포종, 망막 모세포종, 두경부암, 침샘암 및 림프종으로 구성된 군에서 선택되는 어느 하나의 암일 수 있다.
또한, 상기 허혈성 질환은 이에 제한되지는 않으나, 중증하지허혈증, 허혈성 뇌졸중, 허혈성 심장 질환, 허혈성 대장염 등일 수 있다. 이때, 상기 허혈성 질환은 미토콘드리아 이상에 의해 발병하는 질환일 수 있으며, 미토콘드리아 기능 저하로 인해 발생하는 허혈성 세포 장애를 모두 포함한다.
상기 신경 질환은 간질, 약물-내성 간질, 양극성 장애 또는 양극성 장애의 조울증, 두통, 편두통, 불안 장애, 주의력 결핍 장애, 중독성 질환, 인격장애, 자폐, 외상성 뇌 손상, 프리히드라이히 운동실조증 또는 시신경 위축 등일 수 있으나, 이에 제한되지 않는다.
또한, 상기 감염성 질환은 이에 제한되지는 않으나, B형 간염, C형 간염, 인간 파필로마 바이러스(human papilloma virus: HPV) 감염, 사이토메갈로바이러스(cytomegalovirus) 감염, 바이러스성 호흡기 질환, 인플루엔자 바이러스 감염 등일 수 있다.
또한, 상기 근 질환은 이에 제한되지는 않으나, MELAS 증후군, MERRF 증후군, Kearns-Sayre 증후군, 근병증, 뇌근육병증, 근무력증, 중증 근무력증, 근위축성 측삭 경화증, 근육퇴행위축, 근위축증, 근긴장저하, 근력약화, 근경직증 등일 수 있다. 이때, 상기 근 질환은 미토콘드리아 기능 저하로 인해 발생하는 근육 세포 장애를 모두 포함할 수 있다.
퇴행성질환은 알츠하이머병(Alzheimer's disease), 파킨슨병(Parkinson's disease), 루게릭병(amyotrophic lateral sclerosis), 헌팅턴병(Huntington's Disease), 다발성 경화증(Multiple sclerosis), 면역계이상 뇌기능 부전, 진행성 신경퇴행질환, 대사성 뇌질환, 니만-픽병(Niemann-Pick disease), 뇌 허혈 및 뇌출혈로 인한 치매(dementia)일 수 있으나, 이에 제한되지 않는다.
상기 눈 질환은 녹내장, 당뇨병성 망막병증과 에이지 관련 황반 변성 등일 수 있으나, 이에 제한되지 않는다.
상기 신경계 자가면역 질환은 다발성경화증(multiple sclerosis), 시신경척수염(neuromyelitis optica), 급성 파종성뇌척수염(acute disseminated encephalomyelitis), 상행성 척수염(ascending myelitis), 중심성 척수염(central myelitis), 하행성 척수염(descending myelitis) 횡단성 척수염(transverse myelitis), 중증근무력증(myasthenia gravis), 길랑바레증후군(guillain-barre syndrome), 자가면역성 포도막염(autoimmuneuveitis), 자가면역성뇌증(autoimmune encephalopathy) 및 만성 염증성 탈수초성 다발신경병증(chronic inflammatory demyelinating polyneuropathy) 등일 수 있으나, 이에 제한되지 않는다.
본 발명의 동결 및/또는 동결건조된 미토콘드리아를 유효성분으로 포함하는 약학 조성물에 있어서, 상기 미토콘드리아는 이에 제한되지는 않으나 약 0.1 내지 약 500 ㎍/㎖, 약 0.2 ㎍/㎖ 내지 약 450 ㎍/㎖ 또는 약 0.5 ㎍/㎖ 내지 약 400 ㎍/㎖의 농도로 포함될 수 있다. 상기 범위로 미토콘드리아를 포함함으로써, 투여 시 미토콘드리아 용량 조절이 용이하고, 환자의 미토콘드리아 관련 질환, 구체적으로 염증성 질환의 증상 개선 정도가 보다 향상될 수 있다. 이때, 미토콘드리아의 용량은 상기 동결된 미토콘드리아를 해동하거나 동결건조된 미토콘드리아를 수복시킨 후, 막 단백질을 정량함으로써, 미토콘드리아를 정량할 수 있다. 구체적으로, 상기 분리된 미토콘드리아를 브래드포드 분석법(bradford protein assay)을 통해 정량할 수 있다[James D. McCully가 저술한 논문(J Vis Exp. 2014;(91): 51682)].
특히, 본 발명에 따른 상기 약학 조성물은 투여할 개체의 체중을 기준으로 1회 약 0.01 ㎎/㎏ 내지 약 5 ㎎/㎏, 약 0.1 ㎎/㎏ 내지 약 4 ㎎/㎏ 또는 약 0.25 ㎎/㎏ 내지 약 2.5 ㎎/㎏ 양의 미토콘드리아를 투여할 수 있으나, 이에 제한되는 것은 아니다. 즉, 상기 약학 조성물이 미토콘드리아 관련 질환이 유발된 개체의 체중을 기준으로 상기 범위의 함량으로 미토콘드리아가 투여되는 것이 세포 활성 측면에서 가장 바람직하다.
또한, 상기 약학 조성물은 1회 내지 10회, 3회 내지 8회 또는 5회 내지 6회 투여할 수 있으며, 바람직하게는 5회 투여할 수 있다. 이때, 투여 간격은 1일 내지 7일 또는 2일 내지 5일 간격으로 할 수 있으며, 바람직하게는 3일 간격으로 투여할 수 있다.
본 명세서에서 사용하는 용어, "예방"은 상기 약학적 조성물의 투여에 의해 암 발생을 억제하거나 그의 발병을 지연시키는 모든 행위를 말한다. 또한, "치료"는 상기 약학적 조성물의 투여에 의해 암 증세가 호전되거나 이롭게 변경하는 모든 행위를 말한다.
본 명세서에서 사용된 용어, "효능(efficacy)"은 1년, 5년 또는 10년과 같이 일정 기간에 걸쳐 생존 또는 질병이 없는 상태에서 생존(disease-free survival)과 같은 하나 이상의 파라미터에 의해 결정될 수 있다. 뿐만 아니라, 상기 파라미터는 개체에서 적어도 하나의 종양의 크기가 억제되는 것을 포함할 수 있다.
상기 약학 조성물의 바람직한 투여량은 환자의 상태 및 체중, 질병의 정도, 약물형태, 투여경로 및 기간에 따라 다르지만, 당업자에 의해 적절하게 선택될 수 있다. 본 발명의 암 예방 또는 치료용 약학 조성물에서 그 유효성분은 항암 활성을 나타낼 수 있는 한, 용도, 제형, 배합 목적 등에 따라 임의의 양(유효량)으로 포함될 수 있다. 여기서 "유효량"이란 항암 효과를 유도할 수 있는 유효성분의 양을 말한다. 이러한 유효량은 당업자의 통상의 능력 범위 내에서 실험적으로 결정될 수 있다.
본 발명의 상기 약학 조성물은 약학적으로 허용 가능한 담체를 더 포함할 수 있다. 상기 약학적으로 허용 가능한 담체는 환자에게 전달하기에 적절한 비-독성 물질이면 어떠한 담체라도 가능하다. 증류수, 알코올, 지방, 왁스 및 비활성 고체가 담체로 포함될 수 있다. 약물학적으로 허용되는 애쥬번트(완충제, 분산제) 또한 약학 조성물에 포함될 수 있다.
구체적으로, 상기 약학 조성물은 유효성분 이외에 약제학적으로 허용되는 담체를 포함하여 당업계에 공지된 통상의 방법으로 투여 경로에 따라 비경구용 제형으로 제조될 수 있다. 여기서 "약제학적으로 허용되는" 의미는 유효성분의 활성을 억제하지 않으면서 적용(처방) 대상이 적응 가능한 이상의 독성을 지니지 않는다는 의미이다.
본 발명의 약학 조성물이 비경구용 제형으로 제조될 경우, 적합한 담체와 함께 당업계에 공지된 방법에 따라 주사제, 경피 투여제 및 비강 흡입제의 형태로 제제화될 수 있다. 주사제로 제제화할 경우 적합한 담체로서는 멸균수, 에탄올, 글리세롤이나 프로필렌글리콜 등의 폴리올 또는 이들의 혼합물을 사용할 수 있으며, 바람직하게는 링거 용액, 트리에탄올 아민이 함유된 PBS(phosphate buffered saline)나 주사용 멸균수, 5% 덱스트로스 같은 등장 용액 등을 사용할 수 있다.
본 발명의 약학 조성물은 주사용 제제일 수 있다. 따라서, 본 발명에 따른 약학 조성물은 주사제 처방의 유통에 따른 제품 안정성을 확보하기 위하여, 주사제로 사용 가능한 산수용액 또는 인산염 등의 완충용액을 사용하여 pH를 조절함으로써, 물리적으로나 화학적으로 매우 안정한 주사제로 제조될 수 있다.
구체적으로, 본 발명의 약학 조성물은 주사용수를 포함할 수 있다.
상기 주사용수는 고형주사제의 용해나 수용성 주사제를 희석하기 위하여 만들어진 증류수로서, 글루코스 주사, 자일리톨 주사, D-만니톨 주사, 프룩토스 주사, 생리식염수, 덱스트란 40 주사, 덱스트란 70 주사, 아미노산 주사, 링거액, 락트산-링거액 또는 약 pH 3.5 내지 약 7.5 범위의 인산염 완충용액 또는 인산이수소나트륨-구연산 완충용액 등일 수 있다.
본 발명의 약학 조성물은 안정화제 또는 용해보조제를 추가로 포함할 수 있다. 예를 들어, 안정화제는 피로설파이트(pyrosulfite) 또는 에틸렌 디아민테트라아세트산(ethylene diaminetetraacetic acid)일 수 있고, 용해보조제는 염산, 아세트산, 인산수산화칼륨, 탄산수소칼륨, 탄산칼륨 또는 트리스(tris)일 수 있다. 일 구체예로, 상기 약학 조성물은 TTG(trehalose-tris-glycine) 용액과 같은 혼합 보존용액을 포함할 수 있으며, 약학적으로 허용되는 약품제재에 통상적으로 이용될 수 있다.
구체적으로, 본 발명의 약학 조성물은 동결 및/또는 동결건조된 미토콘드리아 이외에 주사용 액상 조성물을 포함할 수 있다. 이때, 동결 및/또는 동결건조된 미토콘드리아는 상술한 바와 동일하다. 본 발명의 약학 조성물이 주사용 액상 조성물을 포함함으로써 미토콘드리아 기능 관련 질환의 예방 또는 치료를 위한 조성물로, 주사를 통해 미토콘드리아 투여 시 미토콘드리아의 응집, 혈소판의 감소 및 응집 등에 의해 야기될 수 있는 혈전 생성을 억제할 수 있고, 미토콘드리아의 안정성을 유지 및/또는 강화할 수 있으며, 미토콘드리아의 활성 또한 안정적으로 유지시킬 수 있다.
여기서, 상기 액상 조성물은 글라이신을 포함할 수 있다. 또한, 상기 액상 조성물은 당류 및/또는 버퍼(buffer) 추가로 포함할 수 있다.
상기 글라이신(glycine)은 이에 제한되지는 않으나, 주사용 액상 조성물 중 약 15 mM 이상의 농도로 존재할 수 있고, 구체적으로는 약 15 mM 내지 약 150 mM의 농도, 약 17 mM 내지 약 130 mM의 농도, 약 20 mM 내지 약 120 mM의 농도, 약 22 mM 내지 약 110 mM의 농도 또는 약 25 mM 내지 약 100 mM의 농도로 존재할 수 있다. 또한, 상기 글라이신은 이에 제한되지는 않으나 히스티딘, 이소류신, 류신, 리신아세테이트, 메티오닌, 페닐알라닌, 트레오닌, 트립토판, 발린, 알라닌, 아르기닌, 아스파라긴산, 시스테인, 글루타민산, 프롤린, 세린 및 티로신으로 이루어진 군으로부터 하나 이상 선택되는 아미노산과 함께 사용 가능하다.
또한, 상기 주사용 액상 조성물 내 포함되는 상기 당류는 이에 제한되지는 않으나 수크로오스, 트레할로오스, 만니톨, 소르비톨, 글루코오스, 프락토오스, 만노오스, 말토오스, 락토오스, 이소말토오스, 덱스트란 및 덱스트린으로 이루어진 군으로부터 선택되는 하나 이상인 것일 수 있다. 특히, 상기 당류는 트레할로오스, 만니톨 또는 수크로오스일 수 있다. 바람직하게, 상기 당류는 트레할로오스일 수 있다.
상기 주사용 액상 조성물 내 포함되는 상기 버퍼는 이에 제한되지는 않으나 트리스(tris) 버퍼, HEPES 버퍼, MOPS 버퍼 및 아세테이트 또는 포스페이트 함유 버퍼로 이루어진 군으로부터 선택되는 것일 수 있다. 바람직하게, 상기 버퍼는 주사가 가능한 트리스(tris) 버퍼일 수 있다.
바람직하게, 상기 액상 조성물은 글라이신, 트레할로오스 및 트리스를 포함할 수 있다.
이때, 상기 버퍼의 pH는 이에 제한되지는 않으나, 약 7.0 내지 약 7.8 범위, 약 7.2 내지 약 7.6 범위 또는 약 7.3 내지 약 7.5 범위일 수 있다.
또한, 상기 버퍼는 이에 제한되지는 않으나, 주사용 액상 조성물 중 약 5 mM 내지 약 50 mM의 농도, 약 8 mM 내지 약 40 mM의 농도, 약 10 mM 내지 약 35 mM의 농도, 약 13 mM 내지 약 30 mM의 농도 또는 약 15 mM 내지 약 25 mM의 농도로 존재할 수 있다.
상기 주사용 액상 조성물은 약 200 내지 약 400 mOsm, 약 230 내지 약 380 mOsm, 약 250 내지 약 350 mOsm, 약 260 내지 약 320 mOsm, 약 270 내지 약 330 mOsm 또는 약 280 내지 약 300 mOsm 범위의 오스몰농도(osmolarity)를 갖는 것일 수 있다. 이때, 상기 범위의 오스몰농도는 2℃ 내지 8℃ 또는 그 이상의 온도에서의 장시간 보관을 촉진하는 한편, 조성물을 비경구용 투여, 예컨대, 혈관내, 근육내 또는 피하 주사를 통해 개체에 부작용 유발없이 투여가 적합하도록 한 것이다.
본 명세서에서 사용된 용어 "오스몰농도(osmolarity)"는 용매의 킬로그램 당 용액의 삼투압에 기여하는 용질의 몰을 지칭하는 것으로, 오스몰농도는 삼투압계(osmometer)를 사용하여 샘플의 빙점강하(freezing point depression)를 측정함으로써 결정된다.
또한, 상기 주사용 액상 조성물은 킬레이트제를 더 포함할 수 있다.
상기 킬레이트제는 이에 제한되지는 않으나, 주사가능한 등급의 EGTA, EDTA 및 BAPTA로 이루어진 군으로부터 하나 이상 선택되는 것일 수 있다. 상기 킬레이트제는 상기 주사용 액상 조성물에 포함되는 미토콘드리아 수득 후 이온 유출에 의한 손상을 제거할 수 있다.
또한, 상기 약학 조성물은 첨가제로, 미토콘드리아의 기능 유지 및 활성에 유효한 항산화제, ATP, 마그네슘 등을 포함할 수 있다.
본 발명의 약학 조성물은 바이알, 카트리지, 주사기 및 자동주사기(autoinjector)로 이루어진 군으로부터 선택된 용기에 보관될 수 있다. 또한, 상기 약학 조성물이 보관된 용기는 치료가 필요한 개체에게 투여될 때까지 실온, 약 2℃ 내지 약 8℃ 냉장온도 또는 약 25℃ 내지 약 40℃로 저장될 수 있다.
상기 개체는 이에 제한되지는 않으나, 사람, 개, 소, 말, 돼지, 양, 염소, 고양이, 마우스, 토끼, 래트(rat)와 같은 포유동물일 수 있으며, 바람직하게는 인간일 수 있다.
한편, 본 발명의 약학적 조성물은 약학적으로 유효한 양으로 투여한다. 상기 용어 "치료학적으로 유효한 양" 또는 "약학적으로 유효한 양"이란 대상 질환을 예방 또는 치료하는데 유효한 화합물 또는 조성물의 양으로서, 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분하며 부작용을 일으키지 않을 정도의 양을 의미한다. 상기 유효량의 수준은 환자의 건강상태, 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 방법, 투여 시간, 투여 경로 및 배출 비율, 치료 기간, 배합 또는 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 일 구체예에서 치료학적으로 유효한 양은 질환을 치료하는데 효과적인 약물의 양을 의미한다.
본 명세서에서 사용하는 용어 "투여"란, 적절한 방법으로 개체에게 소정의 물질을 도입하는 것을 의미하며, 상기 조성물의 투여 경로는 목적 조직에 도달할 수 있는 한 어떠한 일반적인 경로를 통하여 투여될 수 있다. 복강내 투여, 정맥내 투여, 근육내 투여, 피하 투여, 피내 투여, 국소 투여, 비내 투여, 직장내 투여될 수 있으나, 이에 제한되지 않는다.
본 발명의 약학 조성물의 바람직한 투여량은 투여할 개체의 체중을 기준으로 1회 약 0.01 ㎎/㎏ 내지 약 5 ㎎/㎏, 약 0.1 ㎎/㎏ 내지 약 4 ㎎/㎏ 또는 약 0.25 ㎎/㎏ 내지 약 2.5 ㎎/㎏ 양의 미토콘드리아를 투여할 수 있으나, 이에 제한되는 것은 아니다. 즉, 상기 약학 조성물이 질환을 앓고 있는 개체의 체중을 기준으로 상기 범위의 함량으로 동결 및/또는 동결건조된 미토콘드리아가 투여되는 것이 세포 활성 측면에서 가장 바람직하다. 또한, 상기 약학 조성물은 1 내지 10회, 3 내지 8회 또는 5 내지 6회 투여할 수 있으며, 바람직하게는 5회 투여할 수 있다. 이때, 투여 간격은 1 내지 7일 또는 2 내지 5일 간격으로 할 수 있으며, 바람직하게는 3일 간격으로 투여할 수 있다. 이러한 투여량은 어떠한 측면으로든 본 발명의 범위를 제한하는 것으로 해석되어서는 아니 된다.
이때, 상기 약학 조성물은 이에 제한되지는 않으나, 약 5 ㎖ 이하, 약 3 ㎖ 이하 또는 약 2 ㎖ 이하의 부피로 18G 내지 32G 바늘을 사용하여 비경구 투여될 수 있다.
상기 용어 "개체"란 본 발명의 조성물이 적용(처방)될 수 있는 대상을 의미하며, 질환을 앓고 있는 개체일 수 있다. 또한 상기 개체는 인간을 포함한 쥐, 생쥐, 가축 등의 포유동물일 수 있으나, 바람직하게는 인간일 수 있다. 본 발명의 조성물은 상기 동결 및/또는 동결건조된 미토콘드리아 이외에, 안전성이 검증되고 각 질환에 대한 예방 또는 치료효과를 갖는 것으로 공지된 임의의 화합물이나 천연 추출물을 추가로 포함할 수 있다. 이때, 상기 약학 조성물 및 상기 활성을 갖는 화합물 또는 천연 추출물은 동시 또는 순차적으로 투여할 수 있다.
본 발명의 또 다른 측면은 질환 예방 또는 치료용 약학 조성물을 제조하기 위한 상기 동결 및/또는 동결건조된 미토콘드리아의 용도를 제공한다. 또한, 본 발명은 상기 약학 조성물을 개체에 투여하는 단계를 포함하는 질환의 예방 또는 치료 방법을 제공할 수 있다. 여기서 개체는 포유동물일 수 있으며, 바람직하게는 인간일 수 있다. 동결된 미토콘드리아, 동결건조된 미토콘드리아, 예방 및 치료는 상술한 바와 동일하다.
상기 질환은 미토콘드리아 관련 질환으로, 구체적 질환은 상술한 바와 동일하다. 이때, 투여는 주사제를 통해 환부나 피하주사, 정맥 주사를 통해 투여될 수 있다. 이를 통해, 본 발명에 따른 약학 조성물은 질환이 발생한 환부에 직접적으로 정상적인 활성을 미토콘드리아를 공급할 수 있어, 미토콘드리아 기능이 저하된 세포의 활성을 증가시키거나 미토콘드리아 기능 이상 세포 재생에 유용하며, 미토콘리아 관련 질환의 예방 또는 치료에 이용될 수 있다.
장기 보존액 조성물 키트
본 발명의 또 다른 측면은 동결 및/또는 동결건조된 미토콘드리아 및 장기 보존액을 포함하는 분리된 세포, 조직 또는 장기(organ)의 보존을 위한 장기 보존액 조성물 키트를 제공한다. 상기 동결 및 동결건조된 미토콘드리아는 상술한 바와 동일하다.
상기 장기 보존액은 히스티딘, 트립토판 및 케토글루타레이트로 이루어진 군으로부터 선택된 하나 이상을 포함할 수 있다. 상기 케토글루타레이트는 알파-케토글루타레이트(alpha-ketoglutrate)일 수 있다.
상기 키트의 동결 및/또는 동결건조된 미토콘드리아는 사용하기 전에 히스티딘, 트립토판 및 케토글루타레이트로 이루어진 군으로부터 선택된 하나 이상을 포함하는 장기 보존액과 혼합하여 사용할 수 있다.

상기 장기 보존액 조성물 키트는 관류(perfusion) 또는 재관류(re-perfusion)을 위한 것일 수 있다. 상기 관류(perfusion)은 분리된 세포, 조직 또는 장기에 관류액을 주입하거나 계속 흘려보내 원형질을 인공액으로 치환하는 것일 수 있다. 상기 재관류(re-perfusion)는 분리된 세포, 조직 또는 장기에 혈액 공급이 일시적으로 중단되었다가 다시 혈액의 흐름을 복구하는 것일 수 있다.
상기 장기 보존액 조성물 키트는 비경구 투여용일 수 있다. 상기 조성물은 액제일 수 있다. 또한, 상기 조성물은 하나 이상의 첨가제(additives)를 포함할 수 있다. 상기 첨가제는 부형제(excipients), 희석제(diluents), 부착방지제(anti-adherents), 보존제(preservatives), 담체(vehicles), 계면활성제 또는 이들의 조합을 포함할 수 있다. 또한, 상기 조성물은 염류, 예를 들어 MgCl2, CaCl2, NaCl, KCl 또는 이들의 조합을 더 포함할 수 있다.
상기 장기 보존액 조성물은 분리된 세포, 조직 또는 장기의 장기보존 효율을 높일 수 있는 첨가제를 포함할 수 있다. 상기 첨가제는 물, 당류(예, 수크로스, 라피노스(raffinose), 전분), pH 완충물질(예, 인산나트륨, 인산칼륨), ATP, 완충제, 알부민 또는 항산화물질(예, 비타민 E)일 수 있다.
본 발명의 또 다른 측면은 장기 보존액 조성물 키트를 제조하기 위한 상기 동결 및/또는 동결건조된 미토콘드리아의 용도를 제공한다. 또한, 본 발명의 또 다른 측면은 분리된 세포, 조직 또는 장기를 상기 동결 및/또는 동결건조된 미토콘드리아가 포함된 장기 보존액 조성물의 존재 하에서 보관하는 단계를 포함하는, 분리된 세포, 조직 또는 장기를 보관하는 방법을 제공한다.
상기 미토콘드리아, 동결 및/또는 동결건조된 미토콘드리아, 세포, 조직, 장기, 장기 보존액 조성물 및 보관은 상술한 바와 동일하다.
상기 보관은 냉장 상태에서 수행되는 것일 수 있다. 상기 냉장 상태는 약 -4℃ 내지 약 25℃, 약 -4℃ 내지 약 20℃, 약 -4℃ 내지 약 15℃, 약 -4℃ 내지 약 10℃, 약 -2℃ 내지 약 10℃, 약 -2℃ 내지 약 8℃, 약 0℃ 내지 약 15℃, 약 0℃ 내지 약 10℃, 약 0℃ 내지 약 8℃, 약 0℃ 내지 약 6℃, 약 0℃ 내지 약 5℃, 약 0℃ 내지 약 4℃, 약 0℃ 내지 약 3℃ 또는 약 2℃ 내지 약 5℃일 수 있다.
상기 보관은 약 0시간 내지 약 2일, 약 0시간 내지 약 42시간, 약 0시간 내지 약 36시간, 약 0시간 내지 약 30시간, 약 0시간 내지 약 24시간, 약 0시간 내지 약 18시간, 약 0시간 내지 약 12시간, 약 0시간 내지 약 9시간, 약 0시간 내지 약 6시간 또는 약 0시간 내지 약 3시간 동안 수행될 수 있다.
이때, 동결된 미토콘드리아를 사용할 경우, 상기 방법은 미토콘드리아를 분리하는 단계; 및 분리한 미토콘드리아를 동결 보호제와 혼합하여 냉동하는 단계를 더 포함할 수 있다. 또한, 동결건조된 미토콘드리아를 사용할 경우, 상기 방법은 미토콘드리아를 분리하는 단계; 및 분리한 미토콘드리아를 동결 보호제와 혼합하여 냉동하는 단계; 및 냉동된 미토콘드리아를 동결건조하는 방법을 더 포함할 수 있다. 상기 미토콘드리아를 분리하는 방법, 분리된 미토콘드리아를 냉동하는 방법 및 냉동된 미토콘드리아를 동결건조하는 방법 및 동결 보호제는 상술한 바와 동일하다.
이하, 본 발명을 실시예를 통하여 보다 상세하게 설명한다. 그러나, 이들 실시예는 하나 이상의 구체예를 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
준비예 1. 미토콘드리아 준비
농축된 혈소판, 탯줄 유래 중간엽 줄기세포 또는 HEK293 세포를 물리적으로 파쇄한 후, 분별원심분리법을 이용하여 침전된 미토콘드리아를 수득하였다.
실시예 1. 미토콘드리아 동결 및 동결건조
수득한 미토콘드리아를 1 mg/㎖의 농도로 TTG 혼합 보존용액(0.195 M 트레할로오스, 20 mM 트리스, 65 mM 글라이신)으로 재현탁 하였다. 이후, LN2를 이용하여 급속 냉동하거나, -80℃에서 냉동하였다. 미토콘드리아를 -80℃에서 냉동할 시, 냉동 컨테이너(freezing container, FC)를 사용 또는 사용하지 않는 조건으로 동결하였다. 동결 건조의 경우, 동결 건조기를 이용하여 진공상태에서 순차적으로 온도를 -40℃, -20℃, 6℃ 및 25℃로 변경하여 수행하였다. 구체적으로 -40℃에서 30분 동안 동결과정을 수행한 뒤, -20℃에서 25시간 동안 반응시켰다. 미토콘드리아의 상태를 확인하면서 -20℃에서 추가적으로 8.3시간 반응시켰다. 그 다음으로 각각 6℃에서 8.3시간 및 25℃에서 9시간 동안 반응을 수행하여 최종적으로 동결건조된 미토콘드리아를 수득하였다.
실시예 2. 동결된 미토콘드리아의 항염증 효능 확인
1% FBS(fetal bovine serum)가 포함된 RPMI 1640 배지로 4×105개의 THP-1 인간 단핵구 세포를 배양하였다. 배양 16시간 내지 24시간 뒤, 24시간 내지 96시간 동안 동결한 혈소판 유래 미토콘드리아(40 ㎍)를 THP-1 세포에 처리하였다.
처리 1시간 후, 2 ㎍/㎖ 농도의 LPS(lipopolysaccharide)를 처리하여 염증 세포 모델을 구축하였다. LPS를 처리하고 4시간 뒤, 상기 THP-1 세포의 배지를 채취하여 20,000xg에서 10분간 원심분리 하여 상층액 내 염증성 사이토카인인 TNF-α의 농도를 ELISA로 측정하였다(도 1). 또한, 동일 방법으로 줄기세포 유래 미토콘드리아 및 혈소판 유래 미토콘드리아(3명의 혈액으로부터 각각 분리하였음)의 항염증 효능도 확인하였다. 이때, 미토콘드리아는 40 ㎍의 농도로 처리하였다.
그 결과, 도 1 및 도 2에 나타난 바와 같이, 부형제(컨트롤)만 처리한 경우에 비해 동결한 혈소판 유래 미토콘드리아 또는 줄기세포 유래 미토콘드리아의 항염증 효과를 확인할 수 있었다.
실시예 3. 동결건조된 미토콘드리아의 수복
동결건조된 미토콘드리아는 상온 및 냉장에서 보관하고 사용하기 직전, 초순수 증류수를 첨가하여 동결건조전의 부피로 수복하여 사용하였다.
동결건조 전의 미토콘드리아의 단백질 농도와 수복 후 미토콘드리아의 단백질 농도는 BCA(bicinchoninic acid) 방법으로 측정하였다(도 3).
실시예 4. 동결건조된 미토콘드리아의 세포 내 이동 확인
적색 형광 단백질이 미토콘드리아에 표지된 세포(HEK293-MTdsRED)로부터 미토콘드리아를 수득하여 실시예 3의 방법으로 동결건조 후 수복하였다. 상기 수복된 미토콘드리아를 배양 중인 HEK293-MTS-GFP 세포(녹색형광 표지 단백질이 미토콘드리아에 표지됨)에 10 ㎍의 농도로 1일간 처리한 뒤, 공초점 현미경으로 세포를 관찰하였다.
그 결과, 도 4에 나타난 바와 같이, 동결건조 후 수복된 미토콘드리아는 이식한 세포 내부로 이동하였다.
실시예 5. 동결 및 동결건조된 미토콘드리아의 에너지 합성 관련 효소 활성 확인
동결 또는 동결건조에 따른 미토콘드리아 활성 변화를 확인하기 위하여, 동결건조 또는 동결건조하지 않은 혈소판 유래 미토콘드리아를 BCA 방법을 이용하여 단백질 농도를 측정하고, 동일 단백질량 당 에너지 합성 관련 효소의 활성을 비교하였다.
구체적으로, 미토콘드리아 내막에 존재하는 산화적 인산화 복합체 complex Ⅰ및 Ⅲ의 활성은 산화된 cytochrome c(Sigma aldrich, cat no. C2037)가 환원됨에 따라 변화하는 흡광도의 세기를 550 nm 파장에서 20분 동안 1분 간격으로 측정하여 평가하였다. 또한, complex Ⅳ의 활성은 cytochrome c를 sodium hydrosulfite(Sigma aldrich, cat no. 157953)를 이용하여 환원시킨 후, 산화됨에 따라 변화하는 흡광도의 세기를 550 nm 파장에서 20분 동안 1분 간격으로 측정하여 평가하였다. ATP 합성능은 ADP를 첨가한 후 CellTiter-Glo luminescence kit(Promega)를 이용하여 발광도를 20분간 1분 간격으로 측정하였다. 미토콘드리아의 시트르산 합성효소 활성능은 Acetyl-CoA(Sigma aldrich, cat no. A2181) 및 Oxaloacetic acid(Sigma aldrich, cat no. O4126)가 미토콘드리아 내 시트르산 합성효소를 통해 조효소 A(CoA-SH)로 전환되고, 상기 조효소 A가 DTNB(5,5'-Dithiobis(2-nitrobenzoic acid), Sigma aldrich, cat no. D218200)와 반응하여 450 nm 파장에서 발색이 되는 원리를 이용하여 20분 동안 1분 간격으로 측정하였다(도 5).
그 결과, 도 5에 나타난 바와 같이, 동결 또는 동결건조 이후에도 상기 미토콘드리아에 존재하는 효소들의 활성이 유지되는 것을 확인할 수 있었다.
실시예 6. 동결건조된 미토콘드리아의 항염증 효능 확인
1% FBS(fetal bovine serum)가 포함된 RPMI 1640 배지로 4×105개의 THP-1 인간 단핵구 세포를 배양하였다. 배양 16시간 내지 24시간 뒤, 동결건조되지 않은 미토콘드리아 또는 혈소판 또는 탯줄 유래 중간엽 줄기세포에서 분리하여 상기 실시예 3의 방법으로 준비한 동결건조된 미토콘드리아(40 ㎍)를 THP-1 세포에 처리하였다.
처리 1시간 후, 2 ㎍/㎖ 농도의 LPS를 처리하여 세포 내 염증 반응을 유도하였다. LPS를 처리하고 4시간 뒤, 상기 THP-1 세포의 배지를 채취하여 20,000xg에서 10분간 원심분리하여 상층액 내 염증성 사이토카인인 TNF-α의 농도를 ELISA로 측정하였다. 또한, 동일 방법으로 동결건조 또는 동결건조하지 않은 미토콘드리아의 농도별 항염증 효능도 확인하였다.
그 결과, 도 6 및 도 7에 나타난 바와 같이, 부형제(컨트롤)만 처리한 경우에 비해 동결건조하지 않은 미토콘드리아 또는 동결건조 미토콘드리아 처리군에서 미토콘드리아 농도 의존적으로 항염증 효과를 확인할 수 있었다.
실시예 7. 동결건조된 미토콘드리아의 심장 기능 보호 효과 측정
실시예 7.1. 심장 적출 및 안정화
심장의 기능을 확인하기 위하여, 먼저 래트에서 심장을 적출하여 안정화시켰다.
구체적으로, Sprague-Dawley 래트(SD, 12주령, 수컷)를 케타민으로 마취한 뒤 희생시켰다. 래트의 심장을 적출하고, 적출된 심장은 대동맥-카테터를 통해 랑겐도르프(langendorff)에 설치하였다. 그 뒤, 적출한 심장을 크렙스-헨셀라이트(Krebs-Henseleit) 용액으로 약 20분간 관류시켜 세척하였다. 심장의 대동맥을 통해 냉장 보관된 히스티딘-트립토판-케토글루타레이트(histidine-tryptophan-ketoglutarate: HTK) 용액을 주사하고 약 10분간 관류하여 심장을 안정화시켰다. 이때, 상기 HTK 용액은 198 mM 히스티딘, 2 mM 트립토판, 1 mM 케토글루타레이트, 30 mM 만니톨, 4 mM MgCl2, 15 μM CaCl2, 15 mM NaCl 및 9 mM KCl을 포함하였다.
실시예 7.2. 적출된 심장에서 미토콘드리아-함유 용액에 의한 관류액의 유지
동결건조된 미토콘드리아 또는 미토콘드리아를 HTK 용액에 첨가하여 5 ㎍/㎖의 동결건조 미토콘드리아-HTK 및 미토콘드리아-HTK 용액을 준비하였다.
실시예 7.1에서 준비된 안정화된 심장에 5 ㎍/㎖의 미토콘드리아-HTK 용액(MT), 5 ㎍/㎖의 동결건조된 미토콘드리아-HTK 용액(lyophilized MT) 또는 미토콘드리아 불포함-HTK 용액(컨트롤)을 약 2분 동안 주입하였다. 그 후, 심장을 각각 150 ㎍/30 ㎖의 미토콘드리아-HTK 용액(MT), 동결건조 미토콘드리아-HTK 용액(lyophilized MT) 또는 미토콘드리아-불포함 HTK 용액에 담그고, 약 4℃에서 보관하였다. 9시간 경과 후, 보관된 심장을 대동맥-카테터를 통해 랑겐도르프에 설치하고, 크렙스-헨셀라이트 용액의 흘려주었다. 심장을 통해 흘러나오는 관류액의 양을 측정하였다(도 8).
그 결과, 도 8에 나타난 바와 같이, HTK 용액(컨트롤)에서만 보관한 경우에 비해 미토콘드리아-HTK 용액에 보관한 경우 심장의 관류액이 약 1.97배, 동결건조 미토콘드리아-HTK 용액에서 약 1.48배 증가하였다.
실시예 8. 동결 보존 시 혼합 보존용액에 따른 미토콘드리아의 손상 감소 효과 확인
실시예 8.1. 혼합 보존용액에 따른 동결 및 해동 작용에 대한 미토콘드리아 보호 효과 비교
탯줄 유래 중간엽 줄기세포에서 분리된 미토콘드리아를 각각 SHE(250 mM sucrose, 20 mM HEPES, 2 mM EGTA) 혼합 보존용액, TTG(20 mM tris, 195 mM trehalose, 66.6 mM glycine) 혼합 보존용액 또는 TT(20 mM tris, 195 mM trehalose) 혼합 보존용액에 혼합하여 동결한 뒤, 해동하여 12,000g에서 10분간 원심분리 하였다. 원심분리 후 상층액을 수득하여 미토콘드리아 손상 유발시 유출되는 사이토크롬 C(cytochrome C)의 양을 웨스턴 블롯을 이용하여 확인하였다. 대조군은 초음파로 파쇄된 미토콘드리아도 상기와 동일한 조건으로 원심분리하고 상층액을 수득하여 사용하였다.
그 결과, 도 9에 나타낸 바와 같이, 미토콘드리아를 TTG 혼합 보존용액에 혼합하여 동결한 미토콘드리아를 해동시킨 상층액에 비하여 SHE 또는 TT 혼합 보존용액에 혼합하여 동결한 미토콘드리아의 상층액에서 사이토크롬 C의 유출이 증가한 것을 확인할 수 있었다. 상기 결과를 통해, TTG 혼합 보존용액이 동결 및 해동 과정에서의 미토콘드리아 손상으로부터 보다 안정적으로 미토콘드리아를 보호할 수 있음을 확인할 수 있었다.
실시예 8.2. 혼합 보존용액에 따른 동결 후 해동된 미토콘드리아의 활성 비교
동결 및 해동 과정에서 발생하는 스트레스에 대한 TTG 혼합 보존용액의 미토콘드리아 보호 효과를 확인하기 위하여, 탯줄 유래 중간엽 줄기세포에서 분리된 미토콘드리아를 각각 SHE 혼합 보존용액 단독, TTG 혼합 보존용액 단독, TT 혼합 보존용액 단독, DMSO를 포함하는 TTG 혼합 보존용액(TTG+DMSO) 또는 DMSO를 포함하는 TT 혼합 보존액(TT+DMSO)과 혼합하여 동결한 뒤, 해동하여 미토콘드리아의 활성을 비교하였다. 이때, DMSO는 통상적으로 사용되는 동결 보존액으로서 10%의 농도로 사용하였다. 미토콘드리아 활성은 미토콘드리아 특이적 효소인 시트르산 합성효소(citrate synthase) 및 ATP 합성효소(ATP synthase)의 활성을 실시예 5와 동일한 방법으로 확인하여TEk.
그 결과, 도 10에 나타낸 바와 같이, TTG 혼합 보존용액에 혼합되어 동결된 미토콘드리아가 SHE 혼합 보존용액에 혼합되어 동결된 미토콘드리아에 비하여 높은 시트르산 합성효소 및 ATP 합성효소의 활성을 나타내었다.
실시예 8.3. 혼합 보존용액에 따른 동결 후 해동된 미토콘드리아의 항염증 효능 비교
탯줄 유래 중간엽 줄기세포에서 분리된 미토콘드리아를 각각 SHE 혼합 보존용액, TTG 혼합 보존용액 단독, TT 혼합 보존용액 단독, DMSO를 포함하는 TTG 혼합 보존용액(TTG+DMSO) 또는 DMSO를 포함하는 TT 혼합 보존액(TT+DMSO)과 혼합하여 동결한 뒤, 해동하여 미토콘드리아의 항염증 효능을 확인하였다. 이때, 미토콘드리아의 항염증 효능은 실시예 2와 동일한 방법으로 확인하였다.
그 결과, 도 11에 나타낸 바와 같이, TTG 혼합 보존용액에 혼합되어 동결된 미토콘드리아가 SHE 혼합 보존용액, TT 혼합 보존용액, TT+DMSO 혼합 보존용액에 혼합되어 동결된 미토콘드리아에 비하여 높은 항염증 효능을 나타내었다. 상기 결과는 TTG 혼합 보존용액과 혼합하여 동결된 미토콘드리아의 활성이 다른 보존 용액과 혼합되어 동결된 미토콘드리아의 활성보다 우수함을 나타낸 결과로서, TTG 혼합 보존용액이 동결 및 해동 과정에서의 발생하는 미토콘드리아의 손상으로부터 보다 안정적으로 미토콘드리아를 보호할 수 있음을 시사하는 결과이다.
실시예 8.4. 혼합 보존용액에 따른 동결 후 해동된 미토콘드리아의 혈소판 응집 억제 효과 비교
탯줄 유래 중간엽 줄기세포에서 분리된 미토콘드리아를 SHE 혼합 보존용액 또는 TTG 혼합 보존용액에 혼합하여 동결한 뒤, 해동하여 미토콘드리아의 혈소판 응집에 대한 억제효과를 비교하였다.
구체적으로, 동결 후 해동된 미토콘드리아를 0.1 μM의 MitoTracker Orange로 10분간 염색하고 미토콘드리아 분리용 용액으로 2회 세척하였다. 상기 미토콘드리아(5 ㎍)를 1 μM의 Mitotracker Green으로 염색된 혈소판(1×107)과 혼합하여 하루 동안 진탕배양 하였다. 상기 진탕배양된 용액(5 ㎕)을 슬라이드 글라스에 도포한 뒤, 공초점 레이저 주사 현미경을 사용하여 관찰하였다.
그 결과, 도 12에 나타낸 바와 같이, TTG 혼합 보존용액에 혼합되어 동결된 미토콘드리아의 경우에는 혈소판의 응집을 유발하지 않았다. 반면, SHE 혼합 보존용액에 혼합되어 동결된 미토콘드리아의 경우, 혈소판 응집이 유발되었다.
실시예 9. TTG 혼합 보존용액과 혼합하여 동결 및 해동된 미토콘드리아 동의 손상 확인
탯줄 유래 중간엽 줄기세포에서 분리된 미토콘드리아를 TTG 혼합 보존용액에 혼합하여 동결한 뒤, 한달 뒤 해동하여 미토콘드리아의 손상을 확인하였다.
구체적으로, 미토콘드리아의 총 단백량, ATP 합성율, Mitotracker 염색 비율, 외막내 특이 단백질인 Tom20의 발현 비율을 측정한 뒤, 동일 미토콘드리아를 TTG 혼합 보존용액과 혼합하여 동결하였다. 동결 후 한달 뒤, 상기 미토콘드리아를 해동하여 상기 항목을 다시 측정하였다. 이때, 미토콘드리아의 총 단백량은 BCA assay kit을 이용하여 측정하였고, ATP 합성율은 실시예 5의 방법으로 측정하였다. Mitotacker 염색 비율 및 Tom20 발현율은 0.5 μM의 Mitotracker green 또는 Mitotracker Orange로 염색하거나, 형광단백질이 부착된 Tom20 항체(Santacruz, sc-17764-FITC)로 염색한 뒤에 유세포 분석기를 이용하여 분석하였다.
그 결과, 도 13과 같이 TTG 혼합 보존용액에서 동결 보관 시, 미토콘드리아의 총 단백량, ATP 합성율, Mitotracker 염색 비율 및 Tom20의 발현율에 있어서 미토콘드리아 동결 전 및 후에 큰 변화가 없음을 확인할 수 있었다.
Claims (22)
- 동결 및/또는 동결건조된 미토콘드리아.
- 제1항에 있어서,상기 동결건조된 미토콘드리아는 37℃ 이하에서 보관하는 것인, 동결 및/또는 동결건조된 미토콘드리아.
- 분리된 미토콘드리아를 안정적으로 동결시키는 방법.
- 제3항에 있어서,상기 미토콘드리아를 동결시키는 방법은미토콘드리아를 분리하는 단계; 및분리된 미토콘드리아를 글라이신을 포함하는 미토콘드리아의 동결 보호제를 처리하여 냉동시키는 단계를 포함하는, 미토콘드리아를 동결시키는 방법.
- 제4항에 있어서,상기 동결 보호제는 트레할로오스, 트리스 및 이들의 조합으로 이루어진 군에서 선택되는 어느 하나를 추가로 포함하는 것인, 미토콘드리아를 동결시키는 방법.
- 제4항에 있어서,상기 냉동은 -1℃ 이하의 온도에서 수행되는 것인, 미토콘드리아를 동결시키는 방법.
- 제4항에 있어서,상기 미토콘드리아는 혈소판, 근육세포, 간세포, 섬유아세포, 상피세포, 신경세포, 지방세포, 골세포, 백혈구, 림프구, 줄기세포 또는 점막세포로부터 수득되는 것인, 미토콘드리아를 동결시키는 방법.
- 분리된 미토콘드리아를 안정적으로 동결건조 시키는 방법.
- 제8항에 있어서,상기 미토콘드리아를 동결건조 시키는 방법은미토콘드리아를 분리하는 단계;분리된 미토콘드리아를 글라이신을 포함하는 미토콘드리아의 동결 보호제를 처리하여 냉동시키는 단계; 및냉동된 미토콘드리아를 동결건조 시키는 단계를 포함하는, 미토콘드리아를 동결건조 시키는 방법.
- 제9항에 있어서,상기 동결 보호제는 트레할로오스, 트리스 및 이들의 조합으로 이루어진 군에서 선택되는 어느 하나를 추가로 포함하는 것인, 미토콘드리아를 동결건조 시키는 방법.
- 제9항에 있어서,상기 냉동은 -1℃ 이하의 온도에서 수행되는 것인, 미토콘드리아를 동결건조 시키는 방법.
- 제9항에 있어서,상기 미토콘드리아는 혈소판, 근육세포, 간세포, 섬유아세포, 상피세포, 신경세포, 지방세포, 골세포, 백혈구, 림프구, 줄기세포 또는 점막세포로부터 수득되는 것인, 미토콘드리아를 동결건조 시키는 방법.
- 글라이신을 포함하는 미토콘드리아 동결 보호제.
- 제13항에 있어서,상기 미토콘드리아 동결 보호제는 트레할로오스, 트리스 및 이들의 조합으로 이루어진 군에서 선택되는 어느 하나를 추가로 포함하는 것인, 미토콘드리아 동결 보호제.
- 글라이신을 포함하는 미토콘드리아 동결 보호제; 및 미토콘드리아를 포함하는 미토콘드리아 동결 조성물.
- 제15항에 있어서,상기 동결 보호제는 트레할로오스, 트리스 및 이들의 조합으로 이루어진 군에서 선택되는 어느 하나를 추가로 포함하는 것인, 미토콘드리아 동결 조성물.
- 동결 및/또는 동결건조된 미토콘드리아를 유효성분으로 포함하는 질환의 예방 또는 치료용 약학 조성물.
- 제17항에 있어서,상기 질환은 미토콘드리아 관련 질환들로 만성 염증성 질환, 급성 염증성 질환, 허혈성 질환, 신경 질환, 심장 질환, 근 질환, 퇴행성 질환, 대사 질환, 섬유화 질환, 관절 질환, 눈 질환, 탈모, 암, 난청, 면역 관련 질환의 예방 또는 치료용 약학 조성물.
- 동결 및/또는 동결건조된 미토콘드리아 및 장기 보존액을 포함하는 분리된 세포, 조직 또는 장기의 보존을 위한 장기 보존액 조성물 키트.
- 제19항에 있어서,상기 동결 및/또는 동결건조된 미토콘드리아는 사용하기 전에 장기 보존액과 혼합하여 사용하는 것인, 장기 보존액 조성물 키트.
- 동결 및/또는 동결건조된 미토콘드리아의 미토콘드리아 관련 질환의 예방 또는 치료용 용도.
- 동결 및/또는 동결건조된 미토콘드리아를 투여하는 단계를 포함하는 미토콘드리아 관련 질환의 예방 또는 치료 방법.
