Abstract
本発明は、D-アロースおよび/またはその誘導体および/またはその混合物を有効成分として含み、細胞周期のG0/1期に留まる期間を延長させる細胞増殖遅延剤、及びそれを含有する細胞の老化を遅延させる化粧品を提供する。また、本発明は、D-アロースおよび/またはその誘導体および/またはその混合物を有効成分として含む細胞増殖遅延剤を、それを必要とする対象に投与し、細胞周期のG0/1期に留まる期間を延長させることを含む、対象における細胞増殖を遅延させる方法を提供する。
Description
本発明は、D-アロースおよび/またはその誘導体および/またはその混合物を有効成分として含む、細胞増殖遅延剤、それを含む化粧品、及び細胞増殖を遅延させる方法に関する。
近年、細胞周期が停止(静止)するのではなく、全体的にゆっくりと周り、増殖するスピードが遅い集団が存在することがバクテリアや酵母で発見されている(非特許文献1及び2)。そのような細胞の現象は、生命体が進化の過程において、過酷な環境で生き残るために獲得された細胞機能として「細胞スローサイクリング」と呼ばれ、癌でも同様の細胞集団が存在することが確認されている(非特許文献3)。このような「細胞スローサイクリング」と呼ばれる細胞周期が停止せずにゆっくりと回る細胞の現象は、生命体が進化の過程で獲得した細胞内に潜在する能力であると考えられているものの、極限の状況のみで観察される現象であり、通常では生じ得ないと考えられている(非特許文献4)。
そのような状況の下、近年、希少糖の一つとして知られるD-アロースが、癌細胞株の増殖を抑制し得ることが報告されており、チオレドキシン相互作用タンパク質(TXNIP)を特異的に誘導ことにより、細胞周期タンパク質であるp27kip1タンパク質を安定化させることで細胞周期を「停止」させる作用があること(非特許文献5)や、細胞周期の「G2期」を遅延させる効果を有することが報告されている(特許文献1)。
また、別の希少糖の一つとして知られるアリトールには、抗脂肪蓄積抑制作用があることが知られている(特許文献2)。

本発明は、新たなメカニズムに基づく細胞増殖遅延剤、それを含む化粧品、及び細胞増殖を遅延させる方法を提供することを目的とする。
本発明者らは、鋭意研究を行なった結果、希少糖の一つであるD-アロースまたはその誘導体に、細胞周期のG0/1期に留まる期間を延長させる効果を有することを新たに見出し、本発明を完成させるに至った。
希少糖D-アロースが、TXNIPを介して細胞周期を停止させることや、細胞周期のG2期を遅延させることが報告されており(特許文献1及び非特許文献5)、D-アロースの誘導体であるアリトールが、抗肥満効果を有することが報告されているが(特許文献2)、いずれも、細胞周期のG0/1期に留まる期間を延長させ得ること、特に細胞周期を一時的に停止させることなく細胞周期のG0/1期に留まる期間を延長させ得ることについては記載されておらず、本願発明者らによって、初めて明らかにされたものである。
すなわち、本発明は、以下の発明を包含する。

本発明により、細胞周期を停止させることなく、細胞周期のG0/1期に留まる期間を特異的に延長させることが可能となり、これまで極限の状況のみで観察されると考えらえていた「細胞スローサイクリング」とよばれる現象を再現できるようになった。当該作用を有する本願発明により、様々な組織や臓器の加齢性変化を予防したり、癌の増殖を抑制したり、皮膚の抗老化剤(例えば化粧品)として利用可能とするなど、幅広く応用することが可能となる。
以下、本発明を実施するための形態について説明するが、本発明の技術的範囲は下記の形態のみに限定されない。なお、本明細書において引用されている先行技術文献は、本明細書においても援用され、その全体が本明細書において取り込まれる。
一実施態様において、本発明は、D-アロースおよび/またはその誘導体および/またはその混合物を有効成分として含み、細胞周期のG0/1期に留まる期間を延長させる、細胞増殖遅延剤を提供する。本発明は、医薬品として提供されるものであってもよく、食品として提供されるものであってもよい。食品としての本発明は、例えば、保健機能食品(例えば、特定保健用食品または栄養機能食品)、またはダイエタリーサプリメントであってもよい。

本発明は、希少糖であるD-アロースおよび/またはその誘導体および/またはその混合物が、これまで知られていなかった、細胞周期のG0/1期に留まる期間を延長させる効果を有することを新たに発見したことによって完成させたものである。
本明細書において、「細胞スローサイクリング」とは、細胞周期が停止(「細胞周期停止(cell cycle arrest)」)することなく、従来の細胞よりも細胞周期がゆっくりと進行する状態を示し、例えば、本願発明のようにD-アロースおよび/またはその誘導体および/またはその混合物を含まない培地で培養された細胞と比べて、D-アロースおよび/またはその誘導体および/またはその混合物を含む培地で培養された細胞は、細胞周期、特に細胞周期のG0/1期に留まる期間が特異的に延長され、その結果、細胞周期が延長する。
本明細書において、「細胞周期」とは、真核生物において、有糸***、細胞質***および間期を含む細胞***を構成する事象のサイクルをいう。歴史的には、増殖中の体細胞では細胞周期を***期(M期)と間期に分けていたが、DNA合成が間期の一部で行われていることが見出されたことに伴い、この時期(S期)をはさんで、G1期、S期およびG2期に、M期を加えて、細胞周期全体を4期に分けるようになり、通常はこのサイクルを細胞周期という。従って、1つの局面では、細胞周期は、S期に倍加した遺伝情報をM期に等分配して細胞を複製していくサイクルを意味する。本明細書において、1細胞周期の各期の所要時間は、公知の方法によって算出することが可能であり、例えば、生体細胞の直接観察、***指数、オートラジオグラフ法の導入、セルソーターによる各細胞内DNA含量の測定などによって算出され得る。増殖を停止した多くの細胞種ではG1期の途中から細胞周期を離脱し、G0期へ移行する。
本明細書において「G0期」とは、細胞が細胞周期の進行をG1期で止めている状態にある期間をいう。細胞周期の進行の停止は、G1期、G2期、M期などで起こるが、一般的にG0期はG1期で細胞周期を離脱した場合をさし、本明細書においても一般的な定義に従う。細胞が分化あるいは老化して増殖停止したときがG0期の代表例として知られる。G0期とG1期との出入は、G1期中におけるチェックポイントの前で起こると考えられている。
細胞周期の進行を制御する主な因子群は、従来、CDK、サイクリン、CDK阻害因子(CKI)が知られている。CDKがタンパク質リン酸化酵素の触媒サブユニットである。細胞周期進行の駆動装置であるともいえる。CDKの調節因子として、サイクリンは活性化因子であり、CKIは抑制因子ということができる。CDKとサイクリンとは、それぞれタンパク質ファミリーを構成し、それらのサブタイプの組合せに応じて細胞周期の進行を促す時期が異なる。哺乳類体細胞における代表例としては、サイクリンB-Cdc2(CDK1)複合体(Cdc2キナーゼ)はM期、サイクリンD-CDK4複合体およびサイクリンE-CDK2複合体はG1期、サイクリンA-CDK2複合体はS期の進行をもたらすとされている。CKIは、G1期とS期の進行の抑制あるいはG0期への離脱をもたらすとされている。
本明細書において「細胞周期のG0/1期に留まる期間」とは、G1期が開始する時(すなわち、M期が終了する時)からS期が開始する時(すなわち、DNAの合成を開始する時)までの期間、及び/又は、G0期にある細胞が細胞周期に入り(すなわち、細胞周期がG0期からG1期に戻る)、S期が開始する時までの期間をいう。本願発明者らは、D-アロースは、細胞周期を停止させることなく、細胞周期のG0/1期に留まる期間を特異的に延長させ得ることを見出した。細胞周期のG0/1期に留まる期間が特異的に延長するために、その後のS期への移行が遅延し、さらにその後のG2/M期への移行が遅れることが見出された。
一実施態様において、本願発明は、細胞における解糖系の活性を抑制する効果を発揮する。本明細書において「解糖系」とは、生体(細胞)内に存在する生化学反応経路であり、グルコースをピルビン酸などの有機酸に分解(異化)し、グルコースに含まれる高い結合エネルギーを生物が使いやすい形に変換していくための代謝過程をいい、一般に用いられている用語と同一のものを指す。嫌気状態でも起こりうる代謝系の代表的なものである一方で、得られる還元力やピルビン酸が電子伝達系やクエン酸回路に受け渡されることで好気呼吸の一部としても機能する。
解糖系の活性は、細胞内活性を測定すること評価することができる。限定されるわけではないが、解糖系の活性は、例えば、細胞の酸素消費速度(OCR値)、細胞外酸性化速度(ECAR値)、又はOCR及びECAR値を測定することによって評価することができる(Jing Zhang, and Qing Zhang, Using Seahorse Machine to Measure OCR and ECAR in Cancer CellsMethods Mol Biol. 2019;1928:353-363.を参考のこと)。本明細書において、「解糖系の活性を抑制する」とは、例えば、細胞外酸性化速度(ECAR値)を、コントロール(例えば、D-アロース及び/又はアリトールの非存在下で培養した細胞)と比較して有意に低下させることであってもよく、酸素消費速度(OCR値)を、コントロール(例えば、D-アロース及び/又はアリトールの非存在下で培養した細胞)と比較して有意に高い状態に維持するものであってもよい。
細胞内活性は、例えば、Agilent Technologies(旧Seahorse Bioscience)社製の細胞外フラックスアナライザーXFe24(24well用)を用いることによって細胞の主要なエネルギー代謝経路である解糖系、及びミトコンドリアによる好気呼吸の状態を、細胞に対して無侵襲・高感度に経時的計測を行うことができる。しかしながら、上記装置は細胞内活性を測定し得る装置の例示であり、細胞内活性の測定には任意の装置、方法を用いて評価することができるため、当該装置の利用に限定されない。
一実施態様において、本発明は、細胞周期のG0/1期に留まる期間を延長させる効果を発揮することができることから、癌細胞の増殖を遅延させ得る。本発明が適用し得る癌としては、例えば、白血病(例えば、急性骨髄性白血病、慢性骨髄性白血病、急性リンパ性白血病、慢性リンパ性白血病)、悪性リンパ腫(ホジキンリンパ腫、非ホジキンリンパ腫(例えば、成人T細胞白血病、濾胞性リンパ腫、びまん性大細胞型B細胞性リンパ腫))、多発性骨髄腫、骨髄異形成症候群、頭頸部癌、消化器癌(例えば、食道癌、食道腺癌、胃癌、大腸癌、結腸癌、直腸癌)、肝臓癌(例えば、肝細胞癌)、胆嚢・胆管癌、胆道癌、膵臓癌、甲状腺癌、肺癌(例えば、非小細胞肺癌(例えば、扁平上皮非小細胞肺癌、非扁平上皮非小細胞肺癌)、小細胞肺癌)、乳癌、卵巣癌(例えば、漿液性卵巣癌)、子宮頚癌、子宮体癌、子宮内膜癌、膣癌、外陰部癌、腎癌(例えば、腎細胞癌)、尿路上皮癌(例えば、膀胱癌、上部尿路癌)、前立腺癌、精巣腫瘍(例えば、胚細胞腫瘍)、骨・軟部肉腫、皮膚癌(例えば、ブドウ膜悪性黒色腫、悪性黒色腫、メルケル細胞癌)、神経膠腫、脳腫瘍(例えば、膠芽腫)、胸膜中皮腫および原発不明癌)が挙げられる。本発明を適用可能な癌細胞は、例えば、膵臓癌細胞、又は脳腫瘍細胞である。
また、一実施態様において、本願発明は、細胞周期のG0/1期に留まる期間を延長させる効果を発揮することができることから、細胞の老化を遅延させるために用いられてもよく、例えば、アンチエージングに用いられる化粧品や、移植用組織又は臓器の保護剤の他、将来的には、生体老化を抑制する食品や医薬品への応用も可能である。
本発明で用いられ得るD-アロースは、自然界に大量に存在するD-グルコース(ブドウ糖)に比べて圧倒的に存在量が少ない。糖の基本単位である単糖(炭素数が6つの単糖(ヘキソース)は全部で34種類あり、アルドースが16種類、ケトースが8種類、糖アルコールが10種類ある)のうち、自然界に大量に存在するD-グルコース(ブドウ糖)に代表される「天然型単糖」に対して、自然界に微量にしか存在しない単糖(アルドース、ケトース)およびその誘導体(糖アルコール)を「希少糖」と定義付けられている。現在、大量生産が可能な希少糖は、D-プシコースとD-アロースである。D-アロースは、アルドースに分類されるアロースのD体であり、六炭糖である。
「D-アロース」を得る方法としては、L-ラムノース・イソメラーゼを用いてD-プシコースから合成する方法や、D-プシコース含有溶液にD-キシロース・イソメラーゼを作用させて得る方法などが開示されているが、本発明におけるD-アロースは、それらの方法に限定されず、化学的な処理方法により異性化されたものなど、何れの方法によって得られたものでもよい。D-アロースの原料となるD-プシコースは、フラクトースを酵素(エピメラーゼ)処理して得られる製法が一般的であるが、それに限定されず、当該酵素を生産する微生物を利用した製法により得られたものでも良いし、天然物から抽出されたものや、天然物の中に含まれるものをそのまま用いても良く、化学的な処理方法により異性化されたものでも良い。また、酵素を利用してD-プシコースを精製する方法は公知である。
D-アロースは、D-アロース含有シロップの形態で用いることもできる。D-アロース含有シロップは、一般的なシロップ(液糖)に適宜混合することでも得られるが、市販品「レアシュガースウィート」(発売元:(株)レアスウィート、販売者:松谷化学工業(株))として、一般店頭で販売されている「食品」であり、容易に入手することができる。
D-アロース含有シロップを得る方法は、例えば、単糖(D-グルコースやD-フラクトース)にアルカリを作用させ、ロブリー・ドブリュイン-ファン エッケンシュタイン転位反応やレトロアルドール反応とそれに続くアルドール反応を起こさせ(以上の反応をアルカリ異性化反応と呼ぶ)、生じた各種単糖(希少糖含む)を含むシロップを広く「希少糖含有シロップ」と呼ぶことができ、D-グルコースおよび/もしくはD-フラクトースを原料として、D-グルコースおよび/もしくはD-フラクトース含量が55~99質量%になるまでアルカリ異性化したシロップが例示される。上記の「レアシュガースウィート」は、異性化糖を原料とし、国際公開第2010/113785号に開示される手法により得られる希少糖を含有するシロップであり、希少糖として主にD-プシコースおよびD-アロースが含まれるように製造されたものである。当該手法により得られる希少糖含有シロップに含まれる希少糖は、全糖に対する割合でD-プシコース0.5~17質量%、D-アロース0.2~10質量%である。高橋らの文献(応用糖質科学、第5巻、第1号、44-49(2015))によると、D-プシコース5.4g/100g、D-ソルボース5.3g/100g、D-タガトース2.0g/100g、D-アロース1.4g/100g、D-マンノース4.3g/100gが含まれるシロップであることが報告されている。
前記希少糖含有シロップの製造に使用される原料としては、でん粉、砂糖、異性化糖、フラクトース、グルコースなどが挙げられる。異性化糖とは、特定組成比のD-グルコースとD-フラクトースを主組成分とする混合糖として広く捉えられ、一般的には、でん粉をアミラーゼ等の酵素または酸により加水分解して得られた、主にブドウ糖からなる糖液を、グルコースイソメラーゼまたはアルカリにより異性化したブドウ糖および果糖を主成分とする液状の糖のことを指す。JAS規格においては、果糖含有率(糖のうちの果糖の割合)が50%未満のものを「ブドウ糖果糖液糖」、50%以上90%未満のものを「果糖ブドウ糖液糖」、90%以上のものを「高果糖液糖」、およびブドウ糖果糖液糖にブドウ糖果糖液糖を超えない量の砂糖を加えたものを「砂糖混合果糖ブドウ糖液糖」とよぶが、本発明の希少糖含有シロップの原料としては、何れの異性化糖を用いても構わない。
例えば、D-フラクトースを原料とした希少糖含有シロップは、D-プシコース5.2%、D-アロース1.8%、グルコース15.0%、D-フラクトース69.3%を含んでいる。また、異性化糖を原料とした希少糖含有シロップは、D-プシコース3.7%、D-アロース1.5%、グルコース45.9%、D-フラクトース37.7%を含み、D-グルコースを原料とすると、D-プシコース5.7%、D-アロース2.7%、グルコース47.4%、D-フラクトース32.1%を含んでいるが、原料および処理方法の違いにより含有糖組成は変化する。これらのシロップからD-アロースを分離精製して使用しても良いが、シロップのままでの使用も考えられる。
一実施態様において、本発明に用いられ得るD-アロースは、D-アロースおよび/またはその誘導体および/またはその混合物であってもよい。一般に、ある出発化合物から分子の構造を化学反応により変換した化合物を出発化合物の誘導体と呼称する。本明細書において、「D-アロースの誘導体」は、出発化合物としてのD-アロースの分子構造を化学反応により変換して得られる化合物;及び、D-アロースと類似の化合物(例えばD-グルコース)を出発化合物として、その出発化合物の分子構造を化学反応により変換して得られる化合物であって、D-アロースの分子構造を化学反応により変換して得られる化合物と同一の構造を有する化合物(「D-アロースの構造類似体」という。)を含む意味で用いられる。一実施態様において、本発明に用いられ得る「D-アロースの誘導体」は、D-アロースの還元物である、アリトールを含むものであってもよい。
本明細書において、「アリトール(D-allo-ヘキシトール;又は(2S,3S,4R,5R)-ヘキサン-1,2,3,4,5,6-ヘキサオール、ともいう。)」は、ポリオール(糖アルコール)に分類される希少糖である。アリトールは、D-プシコース又はD-アロースを還元することで得られる炭素数6の糖アルコールであり、自然界にはほとんど存在しないが、ズイナ(ズイナ属、随菜属、学名:Itea spp.)(地域によってズイナのことを「リョウブ」とも呼ばれている。)の植物体に含有することが知られている(特許文献2)。従って、本発明に適用され得るアリトールは、ズイナ属の植物体から抽出されたものを用いてもよく、D-プシコースから周期律表の第8族の元素から選ばれる金属を含有する触媒の存在下、水素添加反応により製造されたアリトールを使用してもよく、これらに限定されない。また、本発明に適用され得るアリトールは、アリトールおよび/またはその誘導体および/またはその混合物であってもよい。
本明細書において、D-アロースの誘導体は、D-アロースのカルボニル基がアルコール基となった糖アルコール(すなわち、アリトール)、D-アロースのヒドロキシメチル基がカルボキシ基に変換されたウロン酸、D-アロースの1位のホルミル基がカルボキシル基になったアルドン酸、D-アロースの1位のホルミル基と、主鎖の末端のヒドロキシメチル基がともにカルボキシル基に変わったアルダン酸、あるいはD-アロースのアルコール基がアミノ基で置換されたアミノ糖であってもよい。
他の実施態様において、D-アロースの誘導体は、D-アロースの任意のヒドロキシ基(例えば、2位、3位、4位、5位及び/又は6位のヒドロキシ基)が水素原子、ハロゲン原子、アミノ基、カルボキシル基、ニトロ基、シアノ基、低級アルキル基、低級アルコキシ基、低級アルカノイル基、低級アルカノイルオキシ基、低級アルコキシカルボニル基、モノ又はジ低級アルキル置換アミノ基、アラルキル基、アリール基又はヘテロアリール基で置換されたD-アロース誘導体であってもよい。
一実施態様において、「D-アロースの誘導体」は、出発化合物としてのアリトールの分子構造を化学反応により変換して得られる化合物;及び、アリトールと類似の化合物(例えばD-マンニトール)を出発化合物として、その出発化合物の分子構造を化学反応により変換して得られる化合物であって、アリトールの分子構造を化学反応により変換して得られる化合物と同一の構造を有する化合物(「アリトールの構造類似体」という。)を含むものであってもよい。一実施態様において、D-アロースの誘導体は、例えば、アリトールの任意のヒドロキシ基(例えば、1位、2位、3位、4位、5位及び/又は6位のヒドロキシ基)が水素原子、ハロゲン原子、アミノ基、カルボキシル基、ニトロ基、シアノ基、低級アルキル基、低級アルコキシ基、低級アルカノイル基、低級アルカノイルオキシ基、低級アルコキシカルボニル基、モノ又はジ低級アルキル置換アミノ基、アラルキル基、アリール基又はヘテロアリール基で置換されたものであってもよい。
ハロゲン原子は、フッ素、塩素、臭素、ヨウ素の各原子を示す。低級アルキル基及び低級アルコキシ基、低級アルコキシカルボニル基、モノ又はジ低級アルキル置換アミノ基におけるアルキル部分は、直鎖、分岐もしくは環状のC1~C6アルキル基を示し、具体的例としては、メチル、エチル、プロピル、イソプロピル、ブチル、イソブチル、sec-ブチル、tert-ブチル、ペンチル、イソペンチル、ネオペンチル、ヘキシル、イソヘキシル、シクロプロピル、シクロブチル、2-メチルシクロプロピル、シクロプロピルメチル、シクロペンチル、シクロヘキシル等を挙げることが出来る。
低級アルカノイル基及び低級アルカノイルオキシ基の低級アルカノイル部分は、直鎖、分岐もしくは環状のC1~C7アルカノイル基を示し、具体的例としては、ホルミル、アセチル、プロピオニル、ブチリル、イソブチリル、バレリル、イソバレリル、ピバロイル、ヘキサノイル、シクロプロピルカルボニル、シクロブチルカルボニル、2-メチルシクロプロピルカルボニル、シクロヘキシルカルボニル等を挙げることができる。
アラルキル基は、C7~C20のアラルキル基を示し、具体的例としては、ベンジル、フェネチル、α-メチルベンジル、ベンズヒドリル、トリチル、ナフチルメチル等を挙げることが出来る。
アリール基は、C6~C14のアリール基を示し、具体的例としては、フェニル、ナフチル等を挙げることが出来る。
ヘテロアリ-ル基は、C3~C8のヘテロアリ-ル基であって、同一または異なってN、O、S各原子の1~4個を含む単環、多環または縮合環であり、具体的例として、2-ピリジル、3-ピリジル、4-ピリジル、2-キノニル、3-キノニル、4-キノニル、5-キノニル、6-キノニル、7-キノニル、8-キノニル、2-インドリル、3-インドリル、4-インドリル、5-インドリル、6-インドリル、7-インドリル、2-フリル、3-フリル、2-チエニル、3-チエニル、2-ピロリジル、3-ピロリジル、2-イミダゾリル、4-イミダゾリル、5-イミダゾリル、2-オキサゾリル、4-オキサゾリル、5-オキサゾリル、2-チアゾリル、4-チアゾリル、5-チアゾリル等を挙げることが出来る。
一実施態様において、本発明において用いられ得るD-アロースの誘導体は、例えば2-デオキシ-D-アロース、5-デオキシ-D-アロース、6-デオキシ-D-アロース、3-デオキシ-D-アロース(3-デオキシ-D-グルコース)等のD-アロースの誘導体等であってもよい。
本発明において用いられ得る「D-アロースおよび/またはその誘導体および/またはその混合物」は、薬理学的に許容され得るその塩または/および水和物も含まれる意味で解釈される。
本明細書において、「食品」は、食品全般を意味するが、いわゆる健康食品を含む一般食品の他、特定保健用食品や栄養機能食品等の保健機能食品をも含むものであり、さらにダイエタリーサプリメント(サプリメント、栄養補助食品)、飼料、食品添加物等も本発明の食品に包含される。本発明のサイトカインの産生を抑制するための組成物は、D-アロースという食品を有効成分とするものであり、甘味料、調味料、食品添加物、食品素材、飲食品、健康飲食品、医薬品・医薬部外品又は飼料の形態で用いられてもよく、いずれの形態で用いても、サイトカインの産生を抑制することを可能とする。
一実施態様において、本発明の組成物は、任意の投与経路で投与することができる。投与経路としては、局所投与(皮膚上、吸入、注腸、点眼、点耳、経鼻、膣内等)、経腸投与(経口、経管、経注等)、非経口投与(経静脈、経動脈、経皮、筋肉注等)が挙げられる。
一実施態様において、本発明の細胞増殖遅延剤として適用し得るD-アロースおよび/またはその誘導体および/またはその混合物の用量は、本発明の効果が発揮される用量であれば、適宜調整することができるが、例えば、1mg/kg体重/日~1000mg/kg体重/日で投与されるものであってもよく、年令、症状により適宜増減することも可能である。例えば、経腸管投与で摂取可能な1mg/kg体重/日~1000mg/kg体重/日、例えば、10mg/kg体重/日~800mg/kg体重/日、50mg/kg体重/日~500mg/kg体重/日の用量で投与されてもよい。
希少糖D-アロースおよび/またはその誘導体および/またはその混合物を食品の一成分として使用した場合、日常の食生活のなかで、希少糖D-アロースおよび/またはその誘導体および/またはその混合物の有効量を安全に摂取することができる。その根拠として希少糖D-アロースは、アルドースであり、その面からヒトに投与できる安全性の高い化合物である。
本明細書において、「医薬品」とは、医薬品及び医薬部外品を含む意味で用いられる。
本発明の細胞増殖遅延剤は、組成物として提供されてもよく、D-アロースおよび/またはその誘導体および/またはその混合物は、一般的賦形剤、安定剤、保存剤、結合剤、崩壊剤等の適当な添加剤を配合し、錠剤、散剤、顆粒剤、カプセル剤や、溶液剤、シロップ剤、エリキシル剤、または油性ないし水性の懸濁等の適宜な剤型を選んで製剤化されて提供されてもよい。
固形製剤としては、D-アロースおよび/またはその誘導体および/またはその混合物は、さらに、製剤学上許容されている添加物を含み、例えば充填剤類や増量剤類、結合剤類、崩壊剤類、溶解促進剤類、湿潤剤類、または潤滑剤類等を必要に応じて選択して混合し、製剤化することができる。また、固形製剤としては、D-アロースおよび/またはその誘導体および/またはその混合物に賦形剤、崩壊剤、結合剤、および滑沢剤等を加えて混合し、圧縮整形することにより製造することができる。賦形剤には、乳糖、デンプン、あるいはマンニトール等が一般に用いられる。崩壊剤としては、炭酸カルシウムやカルボキシメチルセルロースカルシウム等が一般に用いられる。結合剤には、アラビアゴム、カルボキシメチルセルロース、あるいはポリビニルピロリドンが用いられる。滑沢剤としては、タルクやステアリン酸マグネシウム等が公知である。
錠剤は、マスキングや、腸溶性製剤とするために、公知のコーティングを施すことができる。コーティング剤には、エチルセルロースやポリオキシエチレングリコール等を用いることができる。
一実施態様において、本発明の組成物は、上述の医薬品(医薬組成物)の他に、食品(例えば、医療用食品、特定保健用食品、健康補助食品、健康食品、栄養機能食品、サプリメント、ダイエタリーサプリメント、またはハーブティなど)としても提供可能である。D-アロースは、1mg/kg体重/日~1000mg/kg体重/日の用量(例えば、50kgの成人の場合、50mg~50g/日)で投与されてもよい。
本発明の剤を化粧品として用いる場合は、化粧水、乳液、ファンデーション、口紅、リップクリーム、クレンジングクリーム、マッサージクリーム、パック、ハンドクリーム、ハンドパウダー、ボディシャンプー、ボディローション、ボディクリーム、浴用化粧品等の形態として用いてもよい。
上記成分に加えて、さらに必要により、本発明の効果を損なわない範囲内で、通常化粧品、医薬品、医薬部外品等の皮膚外用剤に用いられる成分、例えば酸化防止剤、油分、紫外線防御剤、界面活性剤、増粘剤、アルコール類、粉末成分、色材、水性成分、水、各種皮膚栄養剤等を必要に応じて適宜配合することができる。
本発明の剤は、外皮に適用される化粧料、医薬部外品等として適用可能であり、その剤型も皮膚に適用できるものであれば限定されず、溶液系、可溶化系、乳化系、粉末分散系、水-油二層系、水-油-粉末三層系、軟膏、化粧水、ゲル、エアゾール等、任意の剤型が適用される。
本発明を適用可能な対象は、ヒトを含む動物(ヒト、ウシ、ブタ、イヌ、ネコ等の哺乳類、ニワトリ等の鳥類等)であってもよく、特に限定されない。
以下に、本発明を実施例に基づいて更に詳しく説明するが、これらは本発明を何ら限定するものではない。
<実施例1:培養ヒト脳腫瘍細胞を使用した解析>
培養ヒト脳腫瘍細胞株であるU251細胞又はU87細胞に対し、糖の投与なし(コントロール)あるいは各濃度(3、5、10、30又は50mM)のD-アロース又はD-グルコースを添加し、48時間培養後に、WST-1アッセイを行い、細胞増殖に与える影響を比較した。
その結果、希少糖であるD-アロースは培養ヒト脳腫瘍細胞株であるU251細胞とU87細胞において、図1に示すような細胞増殖抑制作用を用量依存的に示した。
D-アロースによる脳腫瘍細胞の増殖抑制効果のメカニズムを解析するため、アポトーシスについて、アネキシンV染色後の細胞をフローサイトメトリーによって評価した(図2)。
その結果、脳腫瘍細胞増殖抑制効果を生じるD-アロースの用量では、アポトーシスを生じないことが明らかとなった。すなわち、D-アロースによる脳腫瘍細胞増殖抑制効果は、アポトーシスを介さないということが示された(図2)。
そこで、D-アロースが細胞周期を停止さることにより脳腫瘍細胞の増殖抑制を生じている可能性を考え、フローサイトメトリー(FACS解析)を使用して、細胞周期の変化を同定した(参照:Yamaguchi F, Takata M, Kamitori K, et al. Rare sugar D-allose induces specific up-regulation of TXNIP and subsequent G1 cell cycle arrest in hepatocellular carcinoma cells by stabilization of p27kip1. Int J Oncol. 2008;32(2):377-385.)。具体的には、細胞周期は、Propidium Iodide (PI)/ RNase Staining Solution (Cell Signaling Technology)で評価した。細胞は、DMEM、50mM-Dグルコースまたは50mM-Dアロースを用いて24時間培養した。その後、培養した細胞をトリプシンで回収し、PBSで2回洗浄した後、-20℃で一晩かけて70%エタノールで固定した。PBSで洗浄した後、500μlのPI試薬で30分間、室温、暗室で細胞を染色し、PBSで洗浄した後、フローサイトメトリーを使用し、617nmでの発光をモニターした。データはKaluza解析ソフトウェア(Ver.1.2, Beckman Coulter)で解析し、細胞周期への影響は、細胞周期の各相における細胞分布の割合の変化によって評価した。
その結果、図3に示すように、脳腫瘍細胞の増殖抑制効果を生じるD-アロース 50mMの投与は、各脳腫瘍細胞周期の停止を誘導しなかった。すなわち、D-アロースによる脳腫瘍細胞の増殖抑制効果は、細胞周期の停止を介さないということが示された。
次に、最近、細胞増殖モニタリング試薬として使用されているフローサイトメトリー用の蛍光プローブであるCytoTell(商標)を脳腫瘍細胞に24時間取り込ませ、その後、糖の投与なし(コントロール)、D-グルコース 50mM、D-アロース 50mMを投与した状態で96時間放置し、各細胞での蛍光プローブ強度の減衰を評価した(図4-1)。
その結果、脳腫瘍細胞増殖抑制効果を生じるD-アロース 50mMの投与は、著しく各脳腫瘍細胞での蛍光プローブ強度の減衰を抑制することが明らかとなった(図4-2)。
以上の結果より、D-アロースはアポトーシスによる細胞死や細胞周期の停止を生じることなく、細胞の増殖スピードを遅延させる効果が示された。すなわち、細胞周期を停止させずに全体をゆっくりと減速させる「細胞スローサイクリング」を生じている可能性が強く考えられたため、さらなる解析を培養ヒト膵臓癌細胞株であるPK-1細胞を使用して実施した。
<実施例2:培養ヒト膵癌細胞を使用した解析>
培養ヒト膵癌細胞株であるPK-1細胞に対し、25もしくは50mMのD-アロース又はD-グルコースを添加し、48時間培養後に、WST-1アッセイを行い、細胞増殖に与える影響を比較した。
培養脳腫瘍細胞において得られた結果と同様、D-アロースは培養膵癌細胞株であるPK-1細胞において、図5に示すような細胞増殖抑制作用を示した。
アネキシンV染色を行い、フローサイトメトリーを使用してアポトーシスについて評価したが、膵癌PK-1細胞の増殖抑制効果を生じた用量である50mMのD-アロースでは、アポトーシスは生じていないことが確認された(図6)。
そこで、D-アロースによる培養ヒト膵癌PK-1細胞の増殖抑制効果が細胞周期の停止を介している可能性を考え、フローサイトメトリー(FACS解析)を使用して細胞周期の変化を同定した。具体的には、PK-1細胞を一晩血清飢餓状態にした後、70%エタノールを用いて-20℃で一晩固定した。細胞DNAを染色するために、遠心分離したペレット状の細胞を450μLのヨウ化プロピジウム(PI)/RNase溶液(Cell Signaling Technology, Danvers, MA, USA)に再懸濁し、室温で30分間、暗所で培養した。その後、サンプルあたり10,000個の細胞をフローサイトメーター(Cytomics FC500 flow cytometry system, Beckman Coulter, Indianapolis, IN, USA)で分析した。
その結果、膵癌PK-1細胞増殖を顕著に抑制するD-アロース 50mM投与により、膵癌PK-1細胞のG0/G1期の割合が若干上昇する傾向が認められた(図7)。
しかしながら、この程度のD-アロースによる変化が、非常に強い増殖抑制効果をもたらしたかについては疑問が残った。そこで、D-アロースが細胞周期の変化を全体的に遅延させるのではないかと考え、図8に示す検証を実施した。
まず、培養ヒト膵癌PK-1細胞にノコダゾール50nMを12時間作用させ、細胞周期をいったんストップさせた。その後、ノコダゾールを培養液から取り除くことにより、再び細胞周期が動き始めるため、リセットした状態から細胞周期を経時的に観察することが可能となる。このような実験条件下において、D-アロースが細胞周期のスピードに対してどのような効果を示すのかについて、詳細に検討を実施した。
その結果、D-アロースを投与した培養ヒト膵癌PK-1細胞では、G0/1期からS期への移行が有意に遅延することが明らかとなった。このような遅延作用により、結果的にG2/M期への移行も遅延することも示された。
以上、D-アロースは細胞周期を停止させず、G0/1期からS期への移行を有意に遅らせる作用を介して、細胞周期のスピードを全体的に遅延させ、増殖する抑制する効果につながるのではないかと考えられた。
しかし、このようなD-アロースの効果がなぜ生じるのかについては明らかとなっていないため、Agilent Technologies(旧Seahorse Bioscience)社製の細胞外フラックスアナライザーXFe24(24well用)を使用して、培養ヒト膵癌PK-1細胞の酸素消費速度(OCR値)及び細胞外酸性化速度(ECAR値)を測定し、解糖系に対する影響を検討した(図10)。
その結果、培養ヒト膵癌PK-1細胞にD-アロースを50mMの濃度で16時間投与すると、その細胞の解糖系が著しく低下していることが明らかとなった。しかも、当該細胞の最大限の解糖系の能力、すなわち予備能を示す「解糖系能力」も低下させていることが示された。
<実施例3:培養ヒト膵癌細胞を使用した解析>
D-アロースの誘導体にも同様に細胞周期のG0/1期に留まる期間を延長させる効果を有するか確かめるために、D-アロースの誘導体であるアリトールを用いて、実施例2と同様の実験を行った。コントロールとして糖アルコールの一つであるD-マンニトールを用いた。

培養ヒト膵癌細胞株であるPK-1細胞に対し、50mMのアリトール又はD-マンニトールを添加し、48時間培養後に、WST-1アッセイを行い、細胞増殖に与える影響を比較した。また、
アリトールは、D-アロースと同様、培養膵癌細胞株であるPK-1細胞において、胞増殖抑制作用を示した(図11)。
アネキシンV染色を行い、フローサイトメトリーを使用してアポトーシスについて評価した。その結果、膵癌PK-1細胞の増殖抑制効果を生じた用量である50mMのアリトールでは、アポトーシスは生じていないことが確認された(図12)。
そこで、アリトールによる培養ヒト膵癌PK-1細胞の増殖抑制効果が細胞周期の停止を介している可能性を考え、実施例2と同様の方法によりフローサイトメトリー(FACS解析)を使用して細胞周期の変化を同定した。
その結果、D-アロースの効果よりは弱いが、D-アロースを投与した時と同様にアリトールにおいても、培養ヒト膵癌PK-1細胞では、G0/1期からS期への移行が有意に遅延することが明らかとなった。このような遅延作用により、結果的にG2/M期への移行も遅延することも示された。
アリトールも、D-アロースと同様に細胞周期を停止させず、G0/1期からS期への移行を有意に遅らせる作用を介して細胞周期のスピードを全体的に遅延させ、細胞増殖する抑制する効果を発揮する可能性が示唆された。
実施例2と同様に、Agilent Technologies(旧Seahorse Bioscience)社製の細胞外フラックスアナライザーXFe24(24well用)を使用して、培養ヒト膵癌PK-1細胞の酸素消費速度(OCR値)及び細胞外酸性化速度(ECAR値)を測定し、解糖系に対する影響も検討した。
その結果、培養ヒト膵癌PK-1細胞にアリトールを50mMの濃度で18時間投与すると、その細胞の解糖系が低下していることが明らかとなった。しかも、当該細胞の最大限の解糖系の能力、すなわち予備能を示す「解糖系能力」も有意に低下させていることが示された。
以上、これまでの実験データにより、D-アロース及びアリトールは解糖系を抑制することによってG0/1期からS期への移行を有意に遅らせ、これが「細胞スローサイクリング」につながり、癌細胞の抑制作用を生じているのではないかと考えられた。
<実施例4:ヒト膵癌細胞株を使用したメタボローム解析>
培養ヒト膵癌細胞株であるPK-1細胞及びPANC-1(5.0×106個)に対し、糖の添加なし(コントロール)あるいは50 mMのD-グルコース又はD-アロースを添加し、12時間、37℃、5%CO2雰囲気下で培養した。その後、各細胞について、ヒューマン・メタボローム・テクノロジーズ株式会社(山形県鶴岡市、日本)が提供するプロトコールに従ってサンプルを調製し、同社が提供する解析サービス(サービス名:C-SCOPE(https://humanmetabolome.com/jpn/service/analysis/cscope/))により、中心エネルギー代謝(解糖系、TCA回路、アミノ酸代謝、及び核酸代謝)に関連する116物質(表1参照)について測定し、その測定結果に基づいて、主成分解析(PCA)(図15)、ヒートマップ解析(図16~図19)及びパスウェイマップ解析(図20)を行った。
培養ヒト膵癌細胞株であるPK-1細胞及びPANC-1(5.0×106個)に対し、糖の添加なし(コントロール)あるいは50 mMのD-グルコース又はD-アロースを添加し、12時間、37℃、5%CO2雰囲気下で培養した。その後、各細胞について、ヒューマン・メタボローム・テクノロジーズ株式会社(山形県鶴岡市、日本)が提供するプロトコールに従ってサンプルを調製し、同社が提供する解析サービス(サービス名:C-SCOPE(https://humanmetabolome.com/jpn/service/analysis/cscope/))により、中心エネルギー代謝(解糖系、TCA回路、アミノ酸代謝、及び核酸代謝)に関連する116物質(表1参照)について測定し、その測定結果に基づいて、主成分解析(PCA)(図15)、ヒートマップ解析(図16~図19)及びパスウェイマップ解析(図20)を行った。
主成分解析(図15)及びヒートマップ解析(図16~図19)の結果より、膵癌細胞は、D-アロースを添加することにより、細胞内の代謝プロファイルが大きく変化することが明らかとなった。特にD-アロースを膵癌細胞に添加することにより、解糖系におけるグルコース6リン酸(Glucose 6-phosphate)及びフルクトース6リン酸(Fructose 6-phospate)を増加させる一方で、フルクトース1,6-二リン酸(Fructose 1,6-diphosphate)を減少させた。また、D-アロースの添加により、ポリオールパスウェイにおけるフルクトース1リン酸(Fructose 1-phospate)が増加していた。これらの結果より、D-アロースは、解糖系やポリオールパスウェイにおいて、これらの物質の代謝を媒介する経路に作用している可能性が示唆された(図20)。
<実施例5:D-アロースを添加したヒト膵癌細胞株PK-1細胞の細胞周期の可視化>
ヒト膵癌細胞株PK-1に、Addgene(マサチューセッツ州、米国)より購入したtFucci(CA)5(Plasmid #153521)(参考:Ando R., et al., Two new coral fluorescent proteins of distinct colors for sharp visualization of cell-cycle progression. bioRxiv, 2020, https://www.biorxiv.org/content/10.1101/2020.03.30.015156v2)をトランスフェクトし、細胞周期を可視化した。tFucci(CA)5をトランスフェクトした細胞は、蛍光顕微鏡を用いて観察した場合、G1期では赤、S期では緑、G2/M期では黄~オレンジを呈する(図21)。当該細胞を用いて、D-アロース(50 mM)の添加の有無によるヒト膵癌細胞株PK-1細胞の細胞周期への影響を観察した。
ヒト膵癌細胞株PK-1に、Addgene(マサチューセッツ州、米国)より購入したtFucci(CA)5(Plasmid #153521)(参考:Ando R., et al., Two new coral fluorescent proteins of distinct colors for sharp visualization of cell-cycle progression. bioRxiv, 2020, https://www.biorxiv.org/content/10.1101/2020.03.30.015156v2)をトランスフェクトし、細胞周期を可視化した。tFucci(CA)5をトランスフェクトした細胞は、蛍光顕微鏡を用いて観察した場合、G1期では赤、S期では緑、G2/M期では黄~オレンジを呈する(図21)。当該細胞を用いて、D-アロース(50 mM)の添加の有無によるヒト膵癌細胞株PK-1細胞の細胞周期への影響を観察した。
その結果、D-アロースを添加することにより、ヒト膵癌細胞株PK-1細胞のG1期からS期への移行を中心とした細胞周期が遅延することが示された(図22)。
Claims (9)
- D-アロースおよび/またはその誘導体および/またはその混合物を有効成分として含み、細胞周期のG0/1期に留まる期間を延長させる、細胞増殖遅延剤。
- D-アロースの誘導体が、アリトールである、請求項1に記載の細胞増殖遅延剤。
- 細胞周期を停止しない、請求項1または2に記載の細胞増殖遅延剤。
- 細胞における解糖系の活性を抑制する、請求項1~3のいずれか1項に記載の細胞増殖遅延剤。
- 癌細胞の増殖を遅延させる、請求項1~4のいずれか1項に記載の細胞増殖遅延剤。
- 膵臓癌細胞、又は脳腫瘍細胞の増殖を遅延させる、請求項1~5のいずれか1項に記載の細胞増殖遅延剤。
- 細胞の老化を遅延させる、請求項1~4のいずれか1項に記載の細胞増殖遅延剤。
- 請求項1~4のいずれか1項に記載の細胞増殖遅延剤を含む、細胞の老化を遅延させる化粧品。
- D-アロースおよび/またはその誘導体および/またはその混合物を有効成分として含む細胞増殖遅延剤を、それを必要とする対象に投与し、細胞周期のG0/1期に留まる期間を延長させること
を含む、対象における細胞増殖を遅延させる方法。
