Claimed is a planktonic eurybiontic strain of unicellular green microalgae Chlorella sorokiniana AGT, capable of photosynthesis at temperatures of from 6°С, deposited in the Russian National Collection of Industrial Microorganisms under number VKPM AI-29. The strain Chlorella sorokiniana AGT VKPM AI-29 can be used for improving the ecological condition of bodies of water of any type or purpose, helping to accelerate the processes of biotic self-purification in bodies of water by increasing the concentration of dissolved oxygen and reducing the level of biogenic elements in the water.

Планктонный эврибионтный штамм Chlorella sorokiniana AGT ВКПМ AI-29 был получен в результате селекции исходного штамма микроводоросли Chlorella sorokiniana. Селекцию проводили на питательных средах при заданных условиях посредством осуществления селекционных циклов, повторяющихся в условиях изменения режимов культивирования и отбора новых колоний после серии циклов для продолжения селекции. В процессе получения нового штамма было проделано более 37 циклов культивирования для получения штамма с заданными параметрами, более 40 манипуляций с составом питательной среды и её концентрации, более 12 различных лабораторных опытов с изменением температур и взаимодействием нового штамма с эндемическими видами цианобактерий и водорослей, распространенных в пресноводных водоёмах. Поскольку целью селекции было получение эврибионтного штамма планктонной хлореллы, сохраняющего активность при широких колебаниях температуры, устойчивого к воздействиям широкого диапазона природных условий гидробиоценоза, таким как изменения pH, повышенная концентрация растворенного в воде СОг, повышенное содержание фосфатов, аммония и других шоковых воздействий, в процессе циклов селекции эти условия менялись искусственным образом в широких пределах. Перед каждым этапом селекционного цикла отобранные штаммы тщательно анализировались на устойчивость к вышеперечисленным воздействиям, и по результатам анализа подбирались определенные условия культивирования в следующем цикле, с целью получения в конечном итоге штамма, соответствующего заданным требованиям. Необходимо отметить, что условия, в которых проводилось культивирование хлореллы в процессе селекции, существенно отличались от природных. Так, режим освещения на некоторых этапах состоял из фазы день - 20 часов/ ночь - 4 часа при температуре от 12 до 28°С и pH 7,0; на других этапах температурные режимы планомерно понижались с 21 до 6°С. Был подобран оптимальный состав питательной среды для культивации штамма, который позволил развиваться планктонному штамму при низких температурах от 6°С. Полученный штамм делится дважды в сутки синхронно. Before each stage of the breeding cycle, the selected strains were carefully analyzed for resistance to the above effects, and based on the results of the analysis, certain cultivation conditions were selected in the next cycle, in order to ultimately obtain a strain that meets the specified requirements. It should be noted that the conditions under which the cultivation of chlorella was carried out during the selection process differed significantly from natural ones. Thus, the lighting regime at some stages consisted of a phase day - 20 hours / night - 4 hours at a temperature of 12 to 28°C and pH 7.0; at other stages, the temperature regimes systematically decreased from 21 to 6°C. The optimal composition of the nutrient medium for the cultivation of the strain was selected, which allowed the development of the planktonic strain at low temperatures from 6°C. The resulting strain is divided twice a day synchronously.
Проведённые лабораторные исследования подтвердили наличие эврибионтных свойств штамма по отношению к температурным колебаниям, составу питательной среды, его высокую резистентность к прочей альгофлоре, включая бактерии, наличие антибактериального растительного эффекта, доминирование штамма при определенной плотности клеток, способность значительно повышать концентрацию растворенного кислорода в воде. В результате проведенной работы созданный новый штамм был адаптирован для культивирования при температурах от 6°С до 30°С, демонстрируя повышенную резистентность к температурным границам, а также колебаниям уровня pH при культивировании от 6,5 до 11,5.
Культурально-морфологические особенности полученного штамма Chlorella sorokiniana AGT приведены ниже. Из приведенных данных следует, что предлагаемый штамм обладает более высокой скоростью прироста биомассы при температуре 10°С по сравнению с другими известными штаммами, при этом нормативная оптическая плотность суспензии штамма Chlorella sorokiniana AGT в пределах 1 ,4 D (440) достигалась за пять дней, что более чем в 4 раза быстрее штамма-аналога (Chlorella vulgaris BIN) при данной температуре. Таким образом, срок культивирования штамма Chlorella sorokiniana AGT до состояния нормативной оптической плотности 1 ,4 D (440) значительно меньше по сравнению с другими штаммами. Так, по сравнению даже с ближайшим аналогом (штаммом Chlorella vulgaris BIN) состояние нормативной оптической плотности достигается более чем в 4 раза быстрее при температуре 10°С и в 3 раза быстрее при температурах 28-30°С. Для изучения способности нового штамма восстанавливать равновесие в водных системах посредством снижения уровня загрязняющих веществ, а также повышения уровня концентрации растворенного кислорода в воде, были проведены следующие эксперименты. Было взято по 10 л воды из загрязненного водоема (эвтрофированный водоём с ярко выраженными абиотическими и биотическими признаками, включая устойчивое «цветение» воды, наличие неприятного запаха и т.д.) для каждого тестируемого штамма, после чего в пробы добавлены суспензии штаммов с одинаковой нормативной оптической плотностью 1 ,4-1 , 8 D (440). Суспензии штаммов представляли собой культуральную среду одинаковой оптической плотности, т.е. с примерно одинаковым количеством живых клеток хлореллы в равном объёме. Кроме того, в пробы был введен раствор углекислого газа на основе содержащего клетчатку материала - голозёрного овса. Через 48 часов был замерен остаточный осадок органических веществ, содержащийся в 10-литровых емкостях анализируемой среды. После этого была рассчитана относительная величина эффективности поглощения биогенных элементов для каждого анализируемого штамма, отражающая степень эффективности изъятия биогенов. Для ее расчета, значение осадка, полученное для штамма Chlorella vulgaris ИФР С-1 11 , было принято за единицу. After 48 hours, the residual sediment of organic substances contained in 10-liter containers of the analyzed medium was measured. After that, the relative value of the nutrient uptake efficiency for each analyzed strain was calculated, reflecting the degree of nutrient uptake efficiency. For its calculation, the sediment value obtained for the Chlorella vulgaris strain IGF C-1 11 was taken as one.

Как видно из таблицы 2.1., планктонный эврибионтный штамм Chlorella sorokiniana AGT значительно эффективнее снижает концентрацию основных биогенных элементов по сравнению со штаммами сравнения, в том числе в 3,8 раза эффективнее штамма-аналога (Chlorella vulgaris BIN), что способствует активному подавлению новым штаммом Chlorella sorokiniana AGT роста цианобактерии Oscillatoria princeps, тогда как другие известные штаммы не оказывали никакого влияния на ее рост. Для проведения эксперимента было взято 20 литров воды из естественного водоёма, с начальной концентрацией растворенного кислорода 5,5 мг/л и значением водородного показателя pH 8,0 и разделено на два образца (опытный и контрольный). В опытный образец было добавлено 100 мл суспензии планктонного эврибионтного штамма Chlorella sorokiniana AGT. Через 24 и 48 часов были проведены замеры концентраций растворенного кислорода и значений pH. В результате эксперимента концентрация растворенного кислорода в опытном образце через 24 часа после добавления суспензии нового штамма составила 9,7 мг/л, pH - 8,4, через 48 часов - 10,3 мг/л, pH - 8,0. В контрольном образце не было зафиксировано наблюдаемых изменений. Оба образца содержались на свету в лабораторных условиях при температуре около 24°С. For the experiment, 20 liters of water were taken from a natural reservoir, with an initial concentration of dissolved oxygen of 5.5 mg/l and a pH value of 8.0, and divided into two samples (experimental and control). 100 ml of a suspension of planktonic eurybiont strain Chlorella sorokiniana AGT was added to the test sample. Dissolved oxygen concentrations and pH values were measured after 24 and 48 hours. As a result of the experiment, the concentration of dissolved oxygen in the test sample 24 hours after adding the suspension of the new strain was 9.7 mg/l, pH - 8.4, after 48 hours - 10.3 mg/l, pH - 8.0. No observed changes were recorded in the control sample. Both samples were kept in the light under laboratory conditions at a temperature of about 24°C.

Основной причиной эвтрофикации водоемов является чрезмерное использование удобрений в сельском хозяйстве: избыток биогенных элементов, в особенности нитрата и фосфора, вымывается, стекает по поверхности или вымывается грунтовыми водами и таким образом попадает в озера и водоемы. Биогены могут вызывать обильный рост фитопланктона, который препятствует прохождению солнечного света в толщу воды, вследствие чего нарушаются процессы фотосинтеза бентосных растений и тем самым уменьшается концентрация кислорода. Более того, происходит накопление большой массы отмершего фитопланктона - детрита; бактерии, потребляя детрит, уменьшают содержание кислорода, таким образом фитопланктоном вытесняются бентосные растения, наступает гибель рыбы. Предлагаемый штамм, вселяемый в водоемы в составе суспензии, способен эффективно поглощать биогенные элементы, например, избыточной азот и фосфор, за счет чего полностью устраняется интенсивное размножение синезеленых водорослей, которые являются начальным звеном развития эвтрофикации. Кроме того, штамм Chlorella sorokiniana AGT является природным антиоксидантом, замедляя процессы гниения, которые начинаются из-за недостатка кислорода. Процесс гниения сопровождается выделением углекислого газа, поэтому торможение процессов гниения новым штаммом способствует предотвращению образования углекислого газа, а выделяемый СОг утилизируется штаммом в процессе фотосинтеза. As shown by the experiments, the new strain of Chlorella sorokiniana AGT is highly productive and capable of active growth already at 10°C. A feature of this strain is its high adaptive ability to reproduce in a wide temperature range, which distinguishes it from other known strains. In addition, the new strain has the ability to restore disturbed balance in water bodies, which is inevitably lost in the process of eutrophication. The main cause of eutrophication of water bodies is the excessive use of fertilizers in agriculture: excess nutrients, especially nitrate and phosphorus, are washed out, run off the surface or are washed away by groundwater and thus enter lakes and water bodies. Nutrients can cause abundant growth of phytoplankton, which prevents the passage of sunlight into the water column, as a result of which the processes of photosynthesis of benthic plants are disrupted and thereby the concentration of oxygen decreases. Moreover, there is an accumulation of a large mass of dead phytoplankton - detritus; bacteria, consuming detritus, reduce the oxygen content, thus benthic plants are displaced by phytoplankton, and fish die. The proposed strain, introduced into water bodies as part of a suspension, is able to effectively absorb biogenic elements, such as excess nitrogen and phosphorus, thereby completely eliminating the intensive reproduction of blue-green algae, which are the initial link in the development of eutrophication. In addition, the Chlorella sorokiniana AGT strain is a natural antioxidant, slowing down the decay processes that begin due to lack of oxygen. The process of decay is accompanied by the release of carbon dioxide, so the inhibition of the processes of decay by a new strain helps to prevent the formation of carbon dioxide, and the released CO2 is utilized by the strain in the process of photosynthesis.
