Abstract
本発明は、一般式(I)(式中、n:mは0.25:0.75~0.95:0.05の範囲内であり、Rは水素原子、炭素数1~6のアルキル基、ウレタン結合を含む炭素数1~6のアルキル基、又はウレタン結合を含みかつ末端に置換されてもよいアリール基を含む炭素数1~6のアルキル基である)で示される繰り返し単位を有しかつ分子量5000以上を有する、2-メタクリロイルオキシエチルホスホリルコリンとメタクリル酸エステルとの共重合体を含む、固体分散体を製造するための可溶化剤を提供する。
Description
本発明は、固体分散体を製造するための可溶化剤及びそれを含有する固体分散体に関する。
難溶性薬物の溶解性改善技術として、固体分散体が知られている。固体分散体は、高分子キャリア中に薬物を非晶質状態で分散する手法であり、固体分散体には、溶出初期の速やかな薬物溶出とその溶解状態を保つ過飽和維持能が求められる。
現在、固体分散体の高分子キャリアとしては、ポリビニルピロリドン(PVP)用いることが一般的である(非特許文献1~3)。

本発明者らは、現在一般的に用いられている固体分散体の高分子キャリアであるPVP以外に固体分散体の製造に適した高分子キャリアを開発することを課題とした。
本発明者らは、上記課題に鑑み鋭意検討を重ねたところ、後述する特定の構造を有する共重合体が固体分散体の製造に適しており、かつ、PVPよりも固体分散体の製造に適した性質を有することを見出し、本発明を完成するに至った。
すなわち、本願は、要旨、以下の発明を提供するものである。
[1] 一般式(I)
(式中、n:mは0.25:0.75~0.95:0.05の範囲内であり、Rは水素原子、炭素数1~6のアルキル基、ウレタン結合を含む炭素数1~6のアルキル基、又はウレタン結合を含みかつ末端に置換されてもよいアリール基を含む炭素数1~6のアルキル基である)
で示される繰り返し単位を有しかつ分子量5000以上を有する、2-メタクリロイルオキシエチルホスホリルコリンとメタクリル酸エステルとの共重合体を含む、固体分散体を製造するための可溶化剤。
[2] Rが、炭素数1~6の直鎖アルキル基、ウレタン結合を含む炭素数1~6の直鎖アルキル基、又はウレタン結合を含みかつ末端に置換されていてもよいアリール基を含む炭素数1~6の直鎖アルキル基である、上記[1]記載の可溶化剤。
[3] Rが、ウレタン結合を含みかつ末端に置換されていてもよいアリール基を含む炭素数1~6の直鎖アルキル基である、上記[1]記載の可溶化剤。
[4] ウレタン結合を含みかつ末端に置換されていてもよいアリール基を含む炭素数1~6の直鎖アルキル基が、以下の式
-X-NHC(=O)-O-Y-Z
(式中、X及びYは、同一でも異なっていてもよく、それぞれが炭素数1~6の直鎖アルキレン基を示し、Zは置換されていてもよいアリール基を示す)
で表される、上記[3]記載の可溶化剤。
[5] X及びYの炭素数が、同一でも異なっていてもよく、それぞれが1~3である、上記[4]記載の可溶化剤。
[6] X及びYが、同一でも異なっていてもよく、それぞれがメチレン基又はエチレン基を示す、上記[4]記載の可溶化剤。
[7] Zが、置換されていてもよいフェニル基を示す、上記[4]~[6]のいずれか記載の可溶化剤。
[8] Zが、非置換のフェニル基又はメチル基に置換されたフェニル基である、上記[4]~[6]のいずれか記載の可溶化剤。
[9] 上記[1]~[8]のいずれか記載の可溶化剤と難溶性薬物とを含む、固体分散体。
[10] 難溶性薬物の水に対する溶解性が、10μg/mLである、上記[9]記載の固体分散体。
[11] 難溶性薬物が、インドメタシン、ニフェジピン、ダナゾール、イトラコナゾール、プロブコール、及びトログリタゾンからなる群より選択される、上記[9]記載の固体分散体。

本発明に係る共重合体は、固体分散体の製造に適しており、特に難溶性薬物を固体分散体中で可溶化するのに適している。
したがって、本発明の好ましい実施態様によれば、特定の構造を有する共重合体を含む可溶化剤と難溶性薬物とを含む固体分散体を提供することができる。

ここで、「一般式(I)で示される繰り返し単位を有し」とは、2-メタクリロイルオキシエチルホスホリルコリンとメタクリル酸エステルとからなり、任意の重合反応によりn:mのモル比率で形成されるランダム共重合体であることを示す。
ここで、「固体分散体」とは、薬物を固体状態の不活性な担体中に微粒子または分子状態で分散させたものであり、難溶性薬物の可溶化のために用いるものである。
ここで、「可溶化剤」とは、薬物の溶解度を増加させるために使用または配合される添加物をいう。薬物が胃や腸で吸収されるためには、胃液や腸液に溶解している必要があるため、溶解性向上のために可溶化剤が使用される。
ここで、「炭素数1~6のアルキル基」には、例えば、メチル基、エチル基、プロピル基、ブチル基、ペンチル基、ヘキシル基が挙げられる。

好ましい実施態様では、Rは、炭素数1~6の直鎖アルキル基、ウレタン結合を含む炭素数1~6の直鎖アルキル基、又はウレタン結合を含みかつ末端に置換されていてもよいアリール基を含む炭素数1~6の直鎖アルキル基である。
別の好ましい実施態様では、Rは、ウレタン結合を含みかつ末端に置換されていてもよいアリール基を含む炭素数1~6の直鎖アルキル基である。
より好ましい実施態様では、Rのウレタン結合を含みかつ末端に置換されていてもよいアリール基を含む炭素数1~6の直鎖アルキル基は、以下の式

特に好ましい実施態様では、Zは、置換されていてもよいフェニル基を示し、好ましくは、Zは、非置換のフェニル基又はメチル基に置換されたフェニル基である。
一実施態様では、Rのウレタン結合を含みかつ末端に置換されていてもよいアリール基を含む炭素数1~6の直鎖アルキル基は、以下の式で表される。
上記共重合体は、例えば、Polymer Journal, Vol 22, No. 5, pp355-360 (1990)に記載の方法に準じた方法で、製造できる。
具体的には、2-メタクリロイルオキシエチルホスホリルコリン及び疎水性モノマーを、好ましくは0.25:0.75~0.90:0.10のモル比で用い、好ましくはテトラヒドロフラン及びエタノールの混合溶媒中、開始剤、好ましくはα、α’-アゾビスイソブチロニトリルの存在下で、好ましくは60~65℃で4~20時間反応させることにより製造できるが、この製造方法に限定されるわけではない。
具体的には、2-メタクリロイルオキシエチルホスホリルコリン及び疎水性モノマーを、好ましくは0.25:0.75~0.90:0.10のモル比で用い、好ましくはテトラヒドロフラン及びエタノールの混合溶媒中、開始剤、好ましくはα、α’-アゾビスイソブチロニトリルの存在下で、好ましくは60~65℃で4~20時間反応させることにより製造できるが、この製造方法に限定されるわけではない。
本発明に係る上記可溶化剤は、難溶性薬物を固体分散体に可溶化するのに適していることから、一実施態様によれば、上記可溶化剤と難溶性薬物とを含む固体分散体が提供される。
ここで、「難溶性薬物」の「難溶性」の程度は、例えば、薬物の水に対する溶解性により決定することができる。
難溶性を薬物の水に対する溶解性で決定する場合、難溶性薬物の水溶性は、例えば、約10μg/mL以下、約9μg/mL以下、約8μg/mL以下、約7μg/mL以下、約6μg/mL以下、約5μg/mL以下、約4μg/mL以下、約3μg/mL以下、約2μg/mL以下、約1μg/mL以下が挙げられ、好ましくは約8μg/mL以下、より好ましくは約1μg/mL以下である。
ここで、上記溶解性は、過剰量の薬物を37℃の水20mLの入った容器内に投入、攪拌し、120分後に紫外吸光光度計により測定した濃度と定義される。

一実施態様では、難溶性薬物は、インドメタシン、ニフェジピン、ダナゾール、イトラコナゾール、プロブコール、及びトログリタゾンからなる群より選択され、好ましくは、インドメタシン、ニフェジピン、ダナゾール、イトラコナゾール、プロブコール、又はトログリタゾンである。
固体分散体を製造する際に、使用する本発明に係る可溶化剤の配合量は、固体分散体とする薬物(好ましくは、難溶性薬物)の配合量や難溶性の度合いにより異なることから、一概に設定することは困難であるが、薬物:可溶化剤の質量比で、例えば0.10:0.90~0.90:0.10程度、好ましくは0.10:0.90~0.70:0.30程度、より好ましくは0.10:0.90~0.50:0.50程度配合することが好ましい。
本発明に係る固体分散体の製造方法は、特段限定されるわけではなく、本技術分野で一般的に用いられている方法を使用できる。
一般に、難水溶性薬物および可溶化剤としての高分子を有機溶媒に溶解させ、その後溶媒を除去することにより調製することができる。溶媒を除去する方法としては、加温や噴霧乾燥法(スプレードライ)等が挙げられる。また、乾式の混合粉砕や溶融法等により、溶媒を用いずに固体分散体を調製することもできる。
一般に、難水溶性薬物および可溶化剤としての高分子を有機溶媒に溶解させ、その後溶媒を除去することにより調製することができる。溶媒を除去する方法としては、加温や噴霧乾燥法(スプレードライ)等が挙げられる。また、乾式の混合粉砕や溶融法等により、溶媒を用いずに固体分散体を調製することもできる。
以下、実施例を用いて、本発明を更に詳細に説明するが、本発明はこれら実施例に記載される態様に限定されるものではない。
合成例1
親水性単量体として2-メタクリロイルオキシエチルホスホリルコリン(MPC)を用い、疎水性単量体としてブチルメタクリレート(BMA)を用いて、それぞれの濃度が0.3mol/L及び0.7mol/Lとなるように、これらの単量体をメタノールに溶解し混合した。単量体溶解液に、触媒及び配位子として、臭化銅(I)及び2,2’-ビピリジルをそれぞれ0.01mol/L及び0.02mol/Lとなるように添加した。この溶液に、窒素を20分間吹き込み、重合開始剤としてエチルα-ブロモイソブチレートを0.01mol/Lとなるように添加した。反応系を密封後、引き続いて、25℃で24時間攪拌し、重合反応を実施した。この溶液をジエチルエーテルに滴下し、再沈殿法により、沈殿物を回収した。回収物を水に溶解し、透析膜(スペクトラム・メディカル・インダストリーズ社製、商品名Spectra/por 7,MWCO 3500)に挿入し、水を用いて透析操作を行い、1日2回の溶媒交換を5日間続けることで、重合体水溶液を得た。更に、得られた水溶液を凍結乾燥することで、重合体粉末を得た。
親水性単量体として2-メタクリロイルオキシエチルホスホリルコリン(MPC)を用い、疎水性単量体としてブチルメタクリレート(BMA)を用いて、それぞれの濃度が0.3mol/L及び0.7mol/Lとなるように、これらの単量体をメタノールに溶解し混合した。単量体溶解液に、触媒及び配位子として、臭化銅(I)及び2,2’-ビピリジルをそれぞれ0.01mol/L及び0.02mol/Lとなるように添加した。この溶液に、窒素を20分間吹き込み、重合開始剤としてエチルα-ブロモイソブチレートを0.01mol/Lとなるように添加した。反応系を密封後、引き続いて、25℃で24時間攪拌し、重合反応を実施した。この溶液をジエチルエーテルに滴下し、再沈殿法により、沈殿物を回収した。回収物を水に溶解し、透析膜(スペクトラム・メディカル・インダストリーズ社製、商品名Spectra/por 7,MWCO 3500)に挿入し、水を用いて透析操作を行い、1日2回の溶媒交換を5日間続けることで、重合体水溶液を得た。更に、得られた水溶液を凍結乾燥することで、重合体粉末を得た。
<分子量測定(GPC)>
得られた重合体を、10mmol/Lの塩化リチウムを含むメタノール:水=7:3(v:v)に対して、1mg/mLとなるように溶解し、試料溶液とした。GPCの分析条件は、溶離液:10mmol/Lの塩化リチウムを含むメタノール:水=7:3(v:v)、流速:0.5mL/分、カラム温度:40℃、カラム:Shodex社製OHpak LB-806M、検出器:示差屈折率計、標準物質:ポリエチレンオキサイドとした。
得られた重合体を、10mmol/Lの塩化リチウムを含むメタノール:水=7:3(v:v)に対して、1mg/mLとなるように溶解し、試料溶液とした。GPCの分析条件は、溶離液:10mmol/Lの塩化リチウムを含むメタノール:水=7:3(v:v)、流速:0.5mL/分、カラム温度:40℃、カラム:Shodex社製OHpak LB-806M、検出器:示差屈折率計、標準物質:ポリエチレンオキサイドとした。
<単量体比測定(1H-NMR)>
得られた重合体10mgを重水素化エタノール0.65mLに溶解した。得られた重水素化エタノール溶液を、日本電子社製α-500を用いて1H-NMR分析を行い、親水性単量体と疎水性単量体のモル比を求めた。
得られた重合体poly(MPC-co-BMA)を以下PMBと称する。重合体の分析結果を表1に示す。
得られた重合体10mgを重水素化エタノール0.65mLに溶解した。得られた重水素化エタノール溶液を、日本電子社製α-500を用いて1H-NMR分析を行い、親水性単量体と疎水性単量体のモル比を求めた。
得られた重合体poly(MPC-co-BMA)を以下PMBと称する。重合体の分析結果を表1に示す。
合成例2~6
表1に示す疎水性単量体及び組成比を用いて、合成例1と同様にして、重合体を合成し、分析した。表1中、
MEBUは2-メタクリロイルオキシエチルブチルウレタン、
MEBZUは2-メタクリロイルオキシエチルベンジルウレタン、
MEPUは2-メタクリロイルオキシエチルフェニルウレタン、
MEPEUは2-メタクリロイルオキシエチルフェネチルウレタン、
MEMBZUは2-メタクリロイルオキシエチル4-メチルベンジルウレタン
を示す。
表1に示す疎水性単量体及び組成比を用いて、合成例1と同様にして、重合体を合成し、分析した。表1中、
MEBUは2-メタクリロイルオキシエチルブチルウレタン、
MEBZUは2-メタクリロイルオキシエチルベンジルウレタン、
MEPUは2-メタクリロイルオキシエチルフェニルウレタン、
MEPEUは2-メタクリロイルオキシエチルフェネチルウレタン、
MEMBZUは2-メタクリロイルオキシエチル4-メチルベンジルウレタン
を示す。
実施例1-1
<固体分散体の調製>
合成例で得られた重合体0.25g及びインドメタシン0.25gを、10mLのエタノール:アセトン=1:1(v:v)に溶解した。ロータリーエバポレーターを用いて、溶解液から溶媒を蒸発させ、固体分散体の粉末を得た。エバポレーターの条件は、温度:40℃、圧力:20mmHgとした。
<固体分散体の調製>
合成例で得られた重合体0.25g及びインドメタシン0.25gを、10mLのエタノール:アセトン=1:1(v:v)に溶解した。ロータリーエバポレーターを用いて、溶解液から溶媒を蒸発させ、固体分散体の粉末を得た。エバポレーターの条件は、温度:40℃、圧力:20mmHgとした。

得られた固体分散体の粉末を、含まれる薬物量が2.5mgとなるように秤量した。秤量した固体分散体末を水20mLの入った容器内に投入し、溶出プロファイルを測定した。μDissの測定条件は、試験液:水、試験液温度:37℃、検出器:紫外吸光光度計とした。結果を図面に記載する。
得られた固体分散体の粉末を、含まれる薬物量が2.5mgとなるように秤量した。秤量した固体分散体末を水20mLの入った容器内に投入し、溶出プロファイルを測定した。μDissの測定条件は、試験液:水、試験液温度:37℃、検出器:紫外吸光光度計とした。結果を図面に記載する。

ポリビニルピロリドン(PVP)0.4g及びインドメタシン0.1gを、10mLのエタノール:アセトン=1:1(v:v)に溶解した。ロータリーエバポレーターを用いて、溶解液から溶媒を蒸発させ、固体分散体の粉末を得た。エバポレーターの条件は、温度:40℃、圧力:20mmHgとした。

ポリビニルピロリドン(PVP)0.4g及びインドメタシン0.1gを、10mLのエタノール:アセトン=1:1(v:v)に溶解した。ロータリーエバポレーターを用いて、溶解液から溶媒を蒸発させ、固体分散体の粉末を得た。エバポレーターの条件は、温度:40℃、圧力:20mmHgとした。
<溶出プロファイル測定(μDiss)>

図1~図6の結果から、実施例の高分子共重合体は、汎用される高分子であるPVPと比較して、難溶性薬物の溶出性を顕著に改善することがわかる。
