本発明は、FGFR阻害剤を用いた軟骨形成異常症(軟骨無形成症、軟骨低形成症、タナトフォリック骨異形成症)の治療に関する。

本発明は、FGFR阻害剤を用いた軟骨形成異常症(軟骨無形成症、軟骨低形成症、タナトフォリック骨異形成症)の治療に関する。
線維芽細胞増殖因子(FGF;fibroblast growth factor)は幅広い組織で発現が見られ細胞の増殖分化を司る増殖因子の一つである。FGFの生理学的活性は、特異的な細胞表面受容体である線維芽細胞増殖因子受容体(FGFR;fibroblast growth factor receptor)によって媒介される。FGFRは、受容体型のタンパク質チロシンキナーゼファミリーに属し、細胞外リガンド結合ドメイン、1回膜貫通ドメイン及び細胞内チロシンキナーゼドメインから構成されており、これまでに4種類のFGFRが同定されている(FGFR1、FGFR2、FGFR3及びFGFR4)。FGFRは、FGFの結合によって二量体を形成し、リン酸化により活性化される。受容体の活性化は、下流の特定のシグナル伝達分子の動員及び活性化を誘導し、生理機能を発現する。
FGF/FGFRシグナル伝達の異常は、ヒトの軟骨細胞分化異常に関する疾患との関連性について報告がある。ヒトの軟骨細胞分化異常における疾患におけるFGF/FGFRシグナルの異常な活性化は、FGFRの遺伝子変異に起因するとされている(非特許文献1、2)。
軟骨形成異常症においては、FGFR3のG380R、N540K、K650E等の点突然変異が報告されており、このような遺伝子変異が軟骨形成異常症の原因となっている可能性が示唆されている(非特許文献3、4、5)。また、FGFR阻害剤であるNVP-BGJ398がキナーゼドメイン外の活性化変異であるFGFR3Y367Cにおいて発症する軟骨無形成症マウスモデルにおいて、治療効果を持つことが報告されている(非特許文献6)。また、NVP-BGJ398がFGFR3G380R変異に対して50nMでキナーゼ阻害効果を示すことが報告されている。一方で、FGFR3G380Rにおいて発症する軟骨無形成症マウスモデルにおける治療効果は報告されていない。
FGFR阻害効果を有する二置換ベンゼンアルキニル化合物が報告されており(特許文献1)、これらの化合物が特定のFGFR2変異を持つ癌に有効であること(特許文献2)、投与スケジュールとして間歇投与が有用であり得ること(特許文献3)も報告されている。

本発明は、軟骨形成異常症を治療するための新規医薬及び当該医薬組成物を用いた治療方法を提供することを課題とする。
本発明者らは、前記課題を解決すべく鋭意検討を重ねた結果、1-[(3S)-3-[4-アミノ-3-[2-(3,5-ジメトキシフェニル)エチニル]-1H-ピラゾロ[3,4-d]ピリミジン-1-イル]-1-ピロリジニル]-2-プロペン-1-オン(Futibatinib)又はその薬学的に許容される塩が、変異を有するFGFR3のリン酸化を阻害し、軟骨形成異常症に対して優れた骨延長効果を有することを見出した。
すなわち本発明は、次の[1]~[11]を包含する。
[1]1-[(3S)-3-[4-アミノ-3-[2-(3,5-ジメトキシフェニル)エチニル]-1H-ピラゾロ[3,4-d]ピリミジン-1-イル]-1-ピロリジニル]-2-プロペン-1-オン又はその薬学的に許容される塩を含有する、軟骨形成異常症を治療するための医薬組成物。
[2]軟骨形成異常症が、軟骨無形成症、軟骨低形成症、又はタナトフォリック骨異形成症である、[1]記載の医薬組成物。
[3]軟骨形成異常症が、軟骨無形成症である、[1]又は[2]記載の医薬組成物。
[4]軟骨形成異常症が、FGFR3変異を有する軟骨形成異常症である、[1]~[3]のいずれか1つに記載の医薬組成物。
[5]FGFR3変異が、FGFR3の248番目のアルギニン、380番目のグリシン、FGFR3の540番目のアスパラギン、又は650番目のリジンの変異である、[4]記載の医薬組成物。
[6]FGFR3変異を有する軟骨形成異常症が、FGFR3変異を有する軟骨無形成症である、[4]記載の医薬組成物。
[7]軟骨無形成症が、FGFR3の380番目のグリシンがアルギニンに変異したFGFR3変異を有する軟骨無形成症である、[6]記載の医薬組成物。

[11]タナトフォリック骨異形成症が、FGFR3の248番目のアルギニンがシステインに変異したFGFR3変異、又は650番目のリジンがグルタミン酸に変異したFGFR3変異を有するタナトフォリック骨異形成症である、[10]記載の医薬組成物。
本発明は、以下の態様にも関する。
・1-[(3S)-3-[4-アミノ-3-[2-(3,5-ジメトキシフェニル)エチニル]-1H-ピラゾロ[3,4-d]ピリミジン-1-イル]-1-ピロリジニル]-2-プロペン-1-オン又はその薬学的に許容される塩を含む、軟骨形成異常症の治療剤。
・軟骨形成異常症の治療における使用のための、1-[(3S)-3-[4-アミノ-3-[2-(3,5-ジメトキシフェニル)エチニル]-1H-ピラゾロ[3,4-d]ピリミジン-1-イル]-1-ピロリジニル]-2-プロペン-1-オン又はその薬学的に許容される塩。

本発明によれば、軟骨形成異常症に対し、優れた骨延長効果を奏する治療を行うことが可能である。
本発明は、1-[(3S)-3-[4-アミノ-3-[2-(3,5-ジメトキシフェニル)エチニル]-1H-ピラゾロ[3,4-d]ピリミジン-1-イル]-1-ピロリジニル]-2-プロペン-1-オン又はその薬学的に許容される塩を含む軟骨形成異常症の治療剤、1-[(3S)-3-[4-アミノ-3-[2-(3,5-ジメトキシフェニル)エチニル]-1H-ピラゾロ[3,4-d]ピリミジン-1-イル]-1-ピロリジニル]-2-プロペン-1-オン又はその薬学的に許容される塩を有効成分とする、軟骨形成異常症を治療するための医薬組成物、及び当該医薬組成物を用いた治療方法に関する。
1-[(3S)-3-[4-アミノ-3-[2-(3,5-ジメトキシフェニル)エチニル]-1H-ピラゾロ[3,4-d]ピリミジン-1-イル]-1-ピロリジニル]-2-プロペン-1-オン(本明細書において「化合物1」と示すことがある)は、下記の構造を有する二置換ベンゼンアルキニル化合物であり、特に限定するものではないが、例えば国際公開WO2013/108809号に記載の製造方法に基づき合成することができる。
本発明において、化合物1はそのまま、又は薬学的に許容される塩の形態で使用することができる。化合物1の薬学的に許容される塩としては、特に限定するものではないが、例えば塩酸、硫酸等の無機酸、酢酸、クエン酸、酒石酸、マレイン酸等の有機酸との付加塩、カリウム、ナトリウム等のアルカリ金属との塩、カルシウム、マグネシウム等のアルカリ土類金属との塩、アンモニウム塩、エチルアミン塩、アルギニン塩等の有機塩基との塩等が挙げられる。
本発明において「FGFR3」とは、ヒト又は非ヒト哺乳動物のFGFR3を含み、好ましくはヒトFGFR3である。ヒトFGFR3のNCBI Gene IDは2261である。また、FGFR3タンパク質は、そのスプライシングバリアントであるアイソフォームを含み、ヒト由来のものであれば、例えば、NCBI Reference Sequence:NP_000133で示されるアミノ酸配列(配列番号1)からなるポリペプチドが挙げられる。
本発明において「FGFR3変異」は、軟骨形成異常症の原因となるアミノ酸変異を有するFGFR3タンパク質若しくは当該アミノ酸をコードするFGFR3遺伝子であれば特に限定されないが、好ましくは配列番号1で示されるアミノ酸配列において、248番目のアルギニン、380番目のグリシン、540番目のアスパラギン及び650番目のリジンからなる群から選択される少なくとも1つのアミノ酸が変異したアミノ酸配列からなるFGFR3タンパク質若しくは野生型のアミノ酸配列が配列番号1と異なるFGFR3タンパク質の場合において、配列番号1における上記位置に相当する位置の少なくとも1つのアミノ酸が変異したアミノ酸配列からなるFGFR3タンパク質又は当該アミノ酸配列をコードするFGFR3遺伝子であり;より好ましくは配列番号1で示されるアミノ酸配列において、248番目のアルギニン、380番目のグリシン及び650番目のリジンからなる群から選択される少なくとも1つのアミノ酸が変異したアミノ酸配列からなるFGFR3タンパク質若しくは野生型のアミノ酸配列が配列番号1と異なるFGFR3タンパク質の場合において、配列番号1における上記位置に相当する位置の少なくとも1つのアミノ酸が変異したアミノ酸配列からなるFGFR3タンパク質又は当該アミノ酸配列をコードするFGFR3遺伝子であり;より好ましくは配列番号1で示されるアミノ酸配列において、248番目のアルギニン及び380番目のグリシンからなる群から選択される少なくとも1つのアミノ酸が変異したアミノ酸配列からなるFGFR3タンパク質若しくは野生型のアミノ酸配列が配列番号1と異なるFGFR3タンパク質の場合において、配列番号1における上記位置に相当する位置の少なくとも1つのアミノ酸が変異したアミノ酸配列からなるFGFR3タンパク質又は当該アミノ酸配列をコードするFGFR3遺伝子であり;より好ましくは配列番号1で示されるアミノ酸配列において、380番目のグリシンが変異したアミノ酸配列からなるFGFR3タンパク質若しくは野生型のアミノ酸配列が配列番号1と異なるFGFR3の場合において、配列番号1における380番目に相当する位置のグリシンが変異したアミノ酸配列からなるFGFR3タンパク質又は当該アミノ酸配列をコードするFGFR3遺伝子である。本明細書において、あるFGFR3タンパク質について、単に配列番号1におけるX番目のアミノ酸と示した場合、そうでないことが明記されない限り、配列番号1で示されるアミノ酸配列におけるX番目のアミノ酸及び野生型のアミノ酸配列が配列番号1と異なるFGFR3タンパク質の場合において配列番号1におけるX番目に相当する位置のアミノ酸を示す。なお、野生型のアミノ酸配列が配列番号1と異なるFGFR3タンパク質のあるアミノ酸が、配列番号1で示されるアミノ酸のどの位置に相当するアミノ酸であるかどうかは、例えば、BLASTのMultiple Alignmentにより確認することができる。
380番目のグリシン又は配列番号1における380番目に相当する位置のグリシンが変異したFGFR3としては、アルギニンに変異したものが好ましい。本明細書おいて、380番目のグリシン又は配列番号1における380番目に相当する位置のグリシンが変異したFGFR3をG380Rと示すことがある。後述するR248C、N540K及びK650Eも同様である。248番目のアルギニン又は配列番号1における248番目に相当する位置のアルギニンが変異したFGFR3としては、システインに変異したR248Cが好ましい。540番目のアスパラギン又は配列番号1における540番目に相当する位置のアスパラギンが変異したFGFR3としては、リジンに変異したN540Kが好ましい。650番目のリジン又は配列番号1における650番目に相当する位置のリジンが変異したFGFR3としては、グルタミン酸に変異したK650Eが好ましい。
FGFR3変異として、好ましくはR248C、G380R、N540K及びK650Eからなる群から選択される少なくとも1つのアミノ酸変異を有するアミノ酸配列からなるFGFR3タンパク質若しくは当該アミノ酸配列をコードするFGFR3遺伝子であり、より好ましくはR248C、G380R及びK650Eからなる群から選択される少なくとも1つのアミノ酸変異を有するアミノ酸配列からなるFGFR3タンパク質若しくは当該アミノ酸配列をコードするFGFR3遺伝子であり、より好ましくはR248C及びG380Rからなる群から選択される少なくとも1つのアミノ酸変異を有するアミノ酸配列からなるFGFR3タンパク質若しくは当該アミノ酸配列をコードするFGFR3遺伝子であり、より好ましくはG380R変異を有するFGFR3タンパク質若しくは当該アミノ酸配列をコードするFGFR3遺伝子である。
また、前述のように、あるFGFR3アイソフォームにおける変異において、アミノ酸の欠失、挿入等によって、変異の位置が配列番号1で示されるアミノ酸の位置とは異なる場合であっても、配列番号1で示されるアミノ酸の位置に相当する位置の変異と同様であると解される。そのため、例えば、配列番号1で表されるFGFR3における380番目のグリシンは、NCBI Reference Sequence:NP_001156685で示されるアミノ酸配列(配列番号2)からなるFGFR3においては、382番目のグリシンに相当する。そのため、本発明において、そうでないことが明示されない限り、例えば、「G380R」は、配列番号1で表されるFGFR3の380番目のグリシンがアルギニンに変異していることだけでなく、NCBI Reference Sequence:NP_001156685で示されるアミノ酸配列からなるFGFR3における382番目のグリシンがアルギニンに変異したものも包含する。なお、あるFGFR3アイソフォームのあるアミノ酸が、配列番号1で示されるアミノ酸のどの位置に相当するアミノ酸であるかどうかは、例えば、BLASTのMultiple Alignmentにより確認することができる。
本発明において「軟骨形成異常症」は、例えば、軟骨無形成症、軟骨低形成症、タナトフォリック骨異形成症等の成長軟骨の機能低下による疾患を意味し、好ましくは軟骨無形成症、軟骨低形成症、又はタナトフォリック骨異形成症であり、より好ましくは軟骨無形成症である。
また、本発明において、「FGFR3変異を有する軟骨形成異常症」としては、配列番号1における380番目のグリシン又は配列番号1における380番目に相当する位置のグリシン、540番目のアスパラギン又は配列番号1における540番目に相当する位置のアスパラギン、又は248番目のアルギニン若しくは配列番号1における248番目に相当する位置のアルギニン又は650番目のリジン若しくは配列番号1における650番目に相当する位置のリジンの変異による成長軟骨の機能低下による疾患等が挙げられ;

本発明において、FGFR3変異は、当業者に周知の方法で検出することが可能である。例えば、FGFR3遺伝子の変異の検出方法としては、サザンブロッティング法、PCR法、DNAマイクロアレイ法、シークエンス解析法等の通常慣用の方法が挙げられる。また、FGFR3タンパク質の変異の検出方法としては、FGFR3変異に特異的に結合する抗体を用いた手法(ELISA法、ウエスタンブロッティング法、免疫染色法等)、マススペクトル分析等の通常慣用の方法が挙げられる。FGFR3変異に特異的に結合する抗体は、市販品を使用すること、又は通常慣用の方法で作製することが可能である。
本発明において用語「試料」は、生体試料(例えば、細胞、組織、臓器、体液(血液、リンパ液等)、消化液、尿)のみならず、これらの生体試料から得られる核酸抽出物(ゲノムDNA抽出物、mRNA抽出物、mRNA抽出物から調製されたcDNA調製物、cRNA調製物等)及びタンパク質抽出物も包含する。また、前記試料は、ホルマリン固定処理、アルコール固定処理、凍結処理又はパラフィン包埋処理が施してあるものでもよい。前記生体試料としては、生体から採取したものを使用することができる。また、生体試料の採取方法は、生体試料の種類に応じて適宜選択することができる。
本発明において、化合物1又はその薬学的に許容される塩そのものを軟骨形成異常症の治療剤として用いても、化合物1又はその薬学的に許容される塩を薬学的担体と組み合わせた医薬組成物として用いてもよい。従って、一実施形態において、本発明は、化合物1又はその薬学的に許容される塩を含有する医薬組成物を提供する。
化合物1又はその薬学的に許容される塩を有効成分として製剤中に含有せしめる場合、任意選択で薬学的担体と配合し、予防又は治療目的に応じて各種の投与形態を採用可能である。投与形態として、例えば、経口剤、注射剤、坐剤、軟膏剤、貼付剤等が挙げられるが、経口剤が好ましい。経口剤としては、錠剤、カプセル剤、顆粒剤、粉剤、シロップ剤等の形態とすることができ、特に限定するものではない。これらの投与形態は、各々当業者に公知慣用の製造方法により製造できる。製剤又は医薬組成物には、投与形態によって、また任意選択で適切な賦形剤、希釈剤、増量剤、崩壊剤等の担体を添加することができる。
上記の各投与単位形態中に配合されるべき化合物1又はその薬学的に許容される塩の量は、これを適用すべき患者の症状により、或いはその剤形等により一定ではないが、一般に投与単位形態あたり、経口剤では0.05~1000mg、注射剤では0.01~500mg、坐剤では1~1000mgとするのが望ましい。
また、化合物1又はその薬学的に許容される塩の1日あたりの投与量は、患者の症状、体重、年齢、性別等によって異なり一概には決定できないが、通常成人(体重60kg)1日あたり化合物1又はその薬学的に許容される塩として約1~1000mgであり、好ましくは1日あたり約10~500mgであり、より好ましくは1日あたり約10~300mgである。
なお、化合物1又はその薬学的に許容される塩の1日あたりの投与量を連日投与する場合、その投与量は、例えば、1日あたり化合物1又はその薬学的に許容される塩として約1~200mgであり、好ましくは1日あたり2~100mgであり、より好ましくは1日あたり4~50mgであり、さらに好ましくは1日あたり10~40mgである。
なお、化合物1又はその薬学的に許容される塩の1日あたりの投与量を間歇投与する場合、その投与量は、例えば、1日あたり化合物1又はその薬学的に許容される塩として約2~1000mgであり、好ましくは1日あたり10~500mgであり、より好ましくは1日あたり20~200mgであり、さらに好ましくは1日あたり50~160mgである。
また、化合物1又はその薬学的に許容される塩の投与スケジュールは、連日投与、又は間歇投与が挙げられる。
本明細書において、「連日投与」とは、例えば、21日間連続して投与するスケジュールを1サイクルとした投与スケジュールが挙げられ、1サイクルが終了する毎に休薬期間を設けてもよい。
本明細書において「間歇投与」とは、1週間に2回以上かつ投与間隔(ある投与日と次の投与日との間隔の日数)を1日以上空ける条件を満たす限り、特に限定されない。
例えば、1週間で1サイクルの投与スケジュールであって、化合物1又はその薬学的に許容される塩が、1サイクル当り1~3日おき(ある投与日と次の投与日との間隔が1~3日)に2回以上投与され、当該サイクルが1回又は2回以上繰り返して実施される、投与スケジュール;
14日間で1サイクルの投与スケジュールであって、化合物1又はその薬学的に許容される塩が、投与日と次の投与日との間隔が1~3日空けて1サイクル当りに4~7回投与され、当該サイクルが1回又は2回以上繰り返して実施される、投与スケジュール;
14日間で1サイクルの投与スケジュールであって、1サイクルに含まれる14日間のうち、化合物1又はその薬学的に許容される塩が、第1日目、第4日目、第8日目及び第11日目に投与される、投与スケジュール;
14日間で1サイクルの投与スケジュールであって、1サイクルに含まれる14日間のうち、化合物1又はその薬学的に許容される塩が、第1日目、第3日目、第5日目、第7日目、第9日目、第11日目及び第13日目に投与される、投与スケジュール;
14日間で1サイクルの投与スケジュールであって、1サイクルに含まれる14日間のうち、化合物1又はその薬学的に許容される塩が、第1日目、第3日目、第5日目、第8日目、第10日目及び第12日目に投与される、投与スケジュール等が挙げられる。本発明において、「投与日と次の投与日との間隔をX日空ける」とは、第n日目に投与をした場合、次の投与日を第n+(1+X)日目とすることを意味する。例えば、投与日と次の投与日との間隔を1日空けるとは、第1日目に投与をした場合、次の投与日を第3日目とすることを意味する。
14日間で1サイクルの投与スケジュールであって、化合物1又はその薬学的に許容される塩が、投与日と次の投与日との間隔が1~3日空けて1サイクル当りに4~7回投与され、当該サイクルが1回又は2回以上繰り返して実施される、投与スケジュール;

本発明は、また、化合物1又はその薬学的に許容される塩の有効量を、軟骨形成異常症の患者に投与する工程を含む、軟骨形成異常症の治療方法を提供する。化合物1又はその薬学的に許容される塩、その投与方法等については前述の通りである。患者としては、ヒト、非ヒトの哺乳動物等が挙げられる。非ヒトの哺乳動物としては、例えば、サル、イヌ、ネコ、ウサギ、マウス、ラット、モルモット等が挙げられる。また、前述したように、軟骨形成異常症としては、FGFR3変異軟骨形成異常症等が挙げられる。
従って、一実施形態において、本発明は、また、以下の(1)及び(2)の工程を含む、FGFR3変異軟骨形成異常症の治療方法を提供する:
(1)患者由来の試料中から、FGFR3タンパク質又はFGFR3遺伝子の変異を検出する工程、
(2)上記(1)の工程において、FGFR3タンパク質又はFGFR3遺伝子の変異が検出された患者に、化合物1又はその薬学的に許容される塩の有効量を投与する工程。
(1)患者由来の試料中から、FGFR3タンパク質又はFGFR3遺伝子の変異を検出する工程、
(2)上記(1)の工程において、FGFR3タンパク質又はFGFR3遺伝子の変異が検出された患者に、化合物1又はその薬学的に許容される塩の有効量を投与する工程。
一実施形態において、本発明は、また、以下の方法も提供する:
FGFR3タンパク質又はFGFR3遺伝子の変異を有する患者に、化合物1又はその薬学的に許容される塩の有効量を投与する工程を含む、
FGFR3変異軟骨形成異常症の治療方法であって、
該患者由来の試料中から、FGFR3タンパク質又はFGFR3遺伝子の変異が検出される、
方法。
FGFR3タンパク質又はFGFR3遺伝子の変異を有する患者に、化合物1又はその薬学的に許容される塩の有効量を投与する工程を含む、
FGFR3変異軟骨形成異常症の治療方法であって、
該患者由来の試料中から、FGFR3タンパク質又はFGFR3遺伝子の変異が検出される、
方法。
上記の治療方法において、FGFR3タンパク質又はFGFR3遺伝子の変異が検出された患者は、化合物1又はその薬学的に許容される塩の有効量を投与する化学療法が十分な治療効果を示すと予測される。ここで、「治療効果」は、骨延長効果等により評価することができる。また、FGFR3の機能阻害の活性(例えば、FGFR3リン酸化を指標とした阻害活性)の程度により治療効果を推定することができる。
以下、実施例を挙げて本発明を更に具体的に説明するが、本発明はこれらによって何ら限定されるものではない。本発明は実施例により十分に説明されているが、当業者により種々の変更及び修飾が可能であろうことは理解される。したがって、そのような変更及び修飾が本発明の範囲を逸脱するものでない限り、それらは本発明に包含される。
実施例1:インビトロにおける化合物1によるFGFR3変異体に対する阻害活性の評価
1-1 FGFR3変異体発現ベクターの構築
FGFR3ベクターはORIGENE社より購入したFGFR3(NM_000142)Human Tagged ORF Clone(FGFR3野生型(WT)発現ベクター)を用い、各変異体(G380R、N540K及びK650E)の発現用ベクターは、前記ベクターをテンプレートとして、構築した。

1-2 FGFR3リン酸化を指標としたFGFR3阻害活性測定
ヒト胎児腎細胞HEK293Tを、10%ウシ胎仔血清を含むDMEM培地にて培養し、細胞を常法により回収後、10%ウシ胎仔血清を含むDMEM培地に懸濁し、Lipofectamine3000 Reagent(ThermoFisherSCIENTIFIC)を用いたリポトランスフェクション法を用いて、上記で示したFGFR3野生型又は変異体発現ベクターをそれぞれ細胞に導入し、96プレートに1ウェルあたり1.5×104個/100μLで播種した。
ヒト胎児腎細胞HEK293Tを、10%ウシ胎仔血清を含むDMEM培地にて培養し、細胞を常法により回収後、10%ウシ胎仔血清を含むDMEM培地に懸濁し、Lipofectamine3000 Reagent(ThermoFisherSCIENTIFIC)を用いたリポトランスフェクション法を用いて、上記で示したFGFR3野生型又は変異体発現ベクターをそれぞれ細胞に導入し、96プレートに1ウェルあたり1.5×104個/100μLで播種した。
薬液として、Vehicle(DMSO)群、各希釈系列(3000nMを最大終濃度とし、1000、300、100、30、10、3、1、0.3nMまでの9濃度の希釈系列)の化合物1を準備した。播種した細胞を37℃、5% CO2で24時間インキュベートしたのち、薬液を含む培地を11μL添加し、さらに1時間インキュベートした。
FGFR3の自己リン酸化能に対する機能阻害は、Human Phospho-FGF R3 DuoSet IC ELISA(R&D SYSTEMS)を用いて測定及び評価した。具体的には、キットに付属の細胞溶解液にProtease inhibitor(Roche)及びPhosphatase inhibitor(Roche)を添加し、それを用いて細胞を溶解し、キットのプロトコルに従い実験を実施し、各ウェルに対してプレートリーダー(SpectraMAX384,Molecular Devices)で比色定量を行った。薬剤添加ウェルの相対的なFGFR3のリン酸化率は、次式に従い、コントロール群を100%とした場合の比率として算出した。なお、実験は2連(1処理群につき2ウェル)で行い、2ウェルの各データの平均値を解析に用いた。

IC50値(50%阻害濃度)は、コントロール群に対して50%阻害を達成する濃度として算出した。
FGFR3野生型(WT)、又は変異体(G380R、N540K又はK650E)を発現させた293T細胞株において、化合物1は下記のような阻害活性を示した(表1)。
実施例2:軟骨無形成症モデルマウスを用いた化合物1の骨延長効果
FGFR3ACHマウス(Naski,M.C.et al.Development 1998,125(24):4977-88。以下、単にACHマウスと示す)を用いた。FVB系統のACHマウスに人工授精したF1 hybridマウスを大量作出し、ゲノムDNAを抽出しPCR法で遺伝子型決定を行った。化合物1は0.5%HPMCに溶解して各濃度の溶液を作成し、投与日の個々体重に応じて、化合物1の投与用量が0.1mg/kg、1mg/kg、又は3mg/kgとなるように10mL/kgで投与した。化合物1投与群と対照群(vehicle=0.5%HPMC投与)に群分け(WT及びvehicle投与群:n=5、化合物1投与群:n=6)して日齢21から42まで1日1回腹腔内注射にて週5日投薬(5投2休)を行った。日齢43に安楽死処置を施し、体重測定ならびにX線撮影による骨長測定(大腿骨、脛骨、尺骨)を行った。X線撮影はfaxitron X-ray DX-50(アクロバイオ株式会社)を用いて行い、計測はImageJで行った。Dunnett's multiple comparisons testを用いて、有意差検定を行い、図1~図5中の*はp<0.05、**はp<0.01、***はp<0.001を示す。
図1~図5に示すように測定時(日齢43)において、vehicle投与群と比較して化合物1の投与群は濃度依存的な骨延長効果が見られた。
次に投薬後のマウスから大腿骨を単離し、軟骨成長板をサフラニンO染色して、その長さを計測した。その結果を図4に示した。ACHマウスの成長板平均幅は148.3であって、ACHマウスに化合物1を投薬した群では、206.4であり、vehicle投与群に比べて伸長があることが確認出来た。
次に投薬後のマウスから大腿骨を単離し、軟骨成長板をサフラニンO染色して、その長さを計測した。その結果を図4に示した。ACHマウスの成長板平均幅は148.3であって、ACHマウスに化合物1を投薬した群では、206.4であり、vehicle投与群に比べて伸長があることが確認出来た。
実施例3:軟骨無形成症モデルマウスを用いた化合物1の骨延長効果
実施例2と同様の方法により、化合物1の投与用量(雄マウス:1mg/kg、3mg/kg、又は10mg/kg、雌マウス:1mg/kg、3mg/kg、又は6mg/kg)を変更し、雄及び雌の両方のマウス(n=10)を用いて骨延長効果の評価を行った。Dunnett's multiple comparisons testを用いて、有意差検定を行い、図6~図11中の*はp<0.05、**はp<0.01、***はp<0.001、****はp<0.0001を示す。
図6~図11に示すように、測定時(日齢43)において、vehicle投与群と比較して化合物1の投与群は濃度依存的な骨延長効果が見られた。
実施例4:疾患iPS細胞モデルでの薬効評価
タナトフォリック骨異形成症I型(TD1,R248C)と軟骨無形性症(ACH,G380A)患者の皮膚線維芽細胞と健常者の皮膚線維芽細胞からiPS細胞株を樹立し、軟骨細胞へと分化誘導した。健常iPS細胞からは軟骨組織が形成されたのに対し、TD1-iPS細胞とACH-iPS細胞から誘導した組織では軟骨成分が減少していた(Yamashita et al.,Nature,2014,513(7519):507-11)。
この分化誘導系において、分化誘導開始後3日目より1nMの化合物1を培地交換の度(培地交換は2-7日に1回)に添加し、分化誘導10週目に解析を行った。陽性コントロールとしてRosuvastatinを用いた。評価項目は組織切片染色(Safranin染色)、COL2A1及びAggrecan(ACAN)のmRNA発現を指標とした。Unpaired testを用いて、有意差検定を行い、図13中の*はp<0.05を示す。
図12に示すように、化合物1の添加によって、軟骨細胞外マトリックスの構成成分であるグリコサミノグリカンを染色するサフラニン染色陽性の染色像が認められた。
また、図13に示すように、化合物1を添加した細胞において、COL2A1及びACANのmRNAの発現は添加していない細胞と比べ発現が増加した。
また、図13に示すように、化合物1を添加した細胞において、COL2A1及びACANのmRNAの発現は添加していない細胞と比べ発現が増加した。

- 軟骨形成異常症が、軟骨無形成症、軟骨低形成症、又はタナトフォリック骨異形成症である、請求項1記載の医薬組成物。
- 軟骨形成異常症が、軟骨無形成症である、請求項1又は2記載の医薬組成物。
- 軟骨形成異常症が、FGFR3変異を有する軟骨形成異常症である、請求項1~3のいずれか1項に記載の医薬組成物。
- FGFR3変異が、FGFR3の248番目のアルギニン、380番目のグリシン、FGFR3の540番目のアスパラギン、又は650番目のリジンの変異である、請求項4記載の医薬組成物。
- FGFR3変異を有する軟骨形成異常症が、FGFR3変異を有する軟骨無形成症である、請求項4記載の医薬組成物。
- 軟骨無形成症が、FGFR3の380番目のグリシンがアルギニンに変異したFGFR3変異を有する軟骨無形成症である、請求項6記載の医薬組成物。
- FGFR3変異を有する軟骨形成異常症が、FGFR3変異を有する軟骨低形成症である、請求項4記載の医薬組成物。
- 軟骨低形成症が、FGFR3の540番目のアスパラギンがリジンに変異したFGFR3変異を有する軟骨低形成症である、請求項8記載の医薬組成物。
- FGFR3変異を有する軟骨形成異常症が、FGFR3変異を有するタナトフォリック骨異形成症である、請求項4記載の医薬組成物。
- タナトフォリック骨異形成症が、FGFR3の248番目のアルギニンがシステインに変異したFGFR3変異、又は650番目のリジンがグルタミン酸に変異したFGFR3変異を有するタナトフォリック骨異形成症である、請求項10記載の医薬組成物。
- 1-[(3S)-3-[4-アミノ-3-[2-(3,5-ジメトキシフェニル)エチニル]-1H-ピラゾロ[3,4-d]ピリミジン-1-イル]-1-ピロリジニル]-2-プロペン-1-オン又はその薬学的に許容される塩を含む、軟骨形成異常症の治療剤。
- 軟骨形成異常症の治療における使用のための、1-[(3S)-3-[4-アミノ-3-[2-(3,5-ジメトキシフェニル)エチニル]-1H-ピラゾロ[3,4-d]ピリミジン-1-イル]-1-ピロリジニル]-2-プロペン-1-オン又はその薬学的に許容される塩。
- 軟骨形成異常症の治療のための、1-[(3S)-3-[4-アミノ-3-[2-(3,5-ジメトキシフェニル)エチニル]-1H-ピラゾロ[3,4-d]ピリミジン-1-イル]-1-ピロリジニル]-2-プロペン-1-オン又はその薬学的に許容される塩の使用。
- 軟骨形成異常症を治療するための医薬を製造するための、1-[(3S)-3-[4-アミノ-3-[2-(3,5-ジメトキシフェニル)エチニル]-1H-ピラゾロ[3,4-d]ピリミジン-1-イル]-1-ピロリジニル]-2-プロペン-1-オン又はその薬学的に許容される塩の使用。
- 1-[(3S)-3-[4-アミノ-3-[2-(3,5-ジメトキシフェニル)エチニル]-1H-ピラゾロ[3,4-d]ピリミジン-1-イル]-1-ピロリジニル]-2-プロペン-1-オン又はその薬学的に許容される塩の有効量を、軟骨形成異常症の患者に投与する工程を含む、軟骨形成異常症の治療方法。
- 活性成分としての1-[(3S)-3-[4-アミノ-3-[2-(3,5-ジメトキシフェニル)エチニル]-1H-ピラゾロ[3,4-d]ピリミジン-1-イル]-1-ピロリジニル]-2-プロペン-1-オン又はその薬学的に許容される塩を、被験者における軟骨形成異常症を治療するためのその使用のための指示書と共に含む、コマーシャルパッケージ。
