Description
本発明は、エラスチン様ブロックペプチド配列を含むポリペプチド等に関する。
人工血管は、代表的には心臓のバイパス手術に使用されるが、その他にも、動脈瘤・末梢瘤、慢性動脈閉塞症、大動脈-下肢動脈の再建、急性動脈閉塞、血管外傷、血行再建合併症、吻合部動脈瘤、人工血管劣化や代用血管の再狭窄・急性閉塞の治療など、多くの手術において用いられる。人工血管の種類としては、布製人工血管、PTFE性人工血管、生体材料由来の人工血管、合成高分子材料製人工血管、人工素材-生体素材ハイブリッド人工血管、結合組織管等があり、各種人工血管材料の開発が進められている。
エラスチンは生体組織に弾性・伸縮性を付与する重要な機能性タンパク質であるが、その高い疎水性に由来するハンドリングの難しさから、材料利用が大幅に遅れている。本発明者は、水中で自己集合性ナノファイバーを形成する二重疎水性エラスチン類似ポリペプチド(GPG)を開発している。得られるファイバーは物理ゲルを形成することができ、生体材料への応用が期待できる。また、非特許文献1では、GPGに細胞接着配列が付加されてなるエラスチン類似ポリペプチドが報告されている。

血管は内壁が内皮細胞で覆われていることから、人工血管材料についても、内皮細胞接着性を有する材料が好ましい。ただ、細胞に対する接着性を有する場合、同時に血小板に対しても接着性を有するので、このような性質の人工血管材料は血栓を生じさせ易いと考えられる。本発明者は、GPGに細胞接着配列が付加されてなるエラスチン類似ポリペプチドの血小板に対する接着性を評価したところ、内皮細胞に対する接着性と同様に、血小板に対する接着性も比較的高いものであった。
そこで、本発明は、内皮細胞に対する接着性を備えながらも、血小板に対する接着性が低く抑えられているポリペプチドを提供することを課題とする。
本発明者は上記課題に鑑みて鋭意研究を進めた結果、(配列番号1:X1GGX2G)nからなるG配列ブロックと(配列番号2:VPGX3G)mからなるP配列ブロックを含むエラスチン様ブロックペプチド配列、及び末端に配置された内皮細胞接着配列を含む、ポリペプチド、であれば、上記課題を解決できることを見出した。本発明者はこの知見に基づいてさらに研究を進めた結果、本発明を完成させた。即ち、本発明は、下記の態様を包含する。
項1. (配列番号1:X1GGX2G)nからなるG配列ブロック(式中、X1は同一又は異なってV又はLを示し、X2は同一又は異なってV又はLを示し、nは4以上の整数を示す。)と(配列番号2:VPGX3G)mからなるP配列ブロック(式中、X3は同一又は異なって任意のアミノ酸を示し、mは5以上の整数を示す。)を含むエラスチン様ブロックペプチド配列、及び末端に配置された内皮細胞接着配列を含む、ポリペプチド。
項2. 前記エラスチン様ブロックペプチド配列の配列構造が、N末端側から、G配列ブロック-P配列ブロック-G配列ブロック、G配列ブロック-P配列ブロック-P配列ブロック-G配列ブロック、P配列ブロック-G配列ブロック、又はG配列ブロック-P配列ブロックである、項1に記載のポリペプチド。
項3. 前記ブロック間にリンカー配列が介在する、項1又は2に記載のポリペプチド。

本発明によれば、内皮細胞に対する接着性を備えながらも、血小板に対する接着性が低く抑えられているポリペプチドを提供することができる。また、本発明によれば、平滑筋細胞の合成型への分化を抑制しつつもその培養が可能なポリペプチドを提供することができる。
本明細書中において、「含有」及び「含む」なる表現については、「含有」、「含む」、「実質的にからなる」及び「のみからなる」という概念を含む。
本明細書中において、アミノ酸配列中のアミノ酸残基を、単にアミノ酸または具体的なアミノ酸名称(バリン、ロイシン)等を示すこともある。また、アミノ酸配列中のアミノ酸残基をアミノ酸の一文字表記で示すこともある。
1.ポリペプチド
本発明は、その一態様において、(配列番号1:X1GGX2G)nからなるG配列ブロックと(配列番号2:VPGX3G)mからなるP配列ブロックを含むエラスチン様ブロックペプチド配列、及び末端に配置された内皮細胞接着配列を含む、ポリペプチド(本明細書において、「本発明のポリペプチド」と示すこともある。)に関する。以下に、これについて説明する。
本発明は、その一態様において、(配列番号1:X1GGX2G)nからなるG配列ブロックと(配列番号2:VPGX3G)mからなるP配列ブロックを含むエラスチン様ブロックペプチド配列、及び末端に配置された内皮細胞接着配列を含む、ポリペプチド(本明細書において、「本発明のポリペプチド」と示すこともある。)に関する。以下に、これについて説明する。
G配列ブロックは、配列番号1に示されるアミノ酸配列(X1GGX2G)のリピート配列(X1GGX2G)nからなるブロックである限り、特に制限されない。G配列ブロックは、通常、β-シート構造をとり得るブロックである。G配列ブロックにより、ポリペプチドに自己集合による繊維形成能を付与することができる。
X1は同一又は異なってV又はLを示し、X2は同一又は異なってV又はLを示す。X1及びX2は好ましくはVである。
nは4以上の整数を示す。nを4以上とすることにより、本発明のポリペプチドは、繊維状自己集合体を形成することができる。nは、好ましくは4~20、より好ましくは4~12、さらに好ましくは4~8、よりさらに好ましくは4~6である。
P配列ブロックは、配列番号2に示されるアミノ酸配列(VPGX3G)のリピート配列(VPGX3G)mからなるブロックである限り、特に制限されない。P配列ブロックは、通常、β-ターン構造をとり得るブロックである。P配列ブロックにより、ポリペプチドが下限臨界共溶温度(LCST)を持つようになり、LCST以上で自己集合して水溶液から相分離するようになる。
本発明のポリペプチドのLCST(溶媒:水、濃度:0.03 wt%の場合)は、例えば10~30℃、好ましくは15~25℃である。
X3は同一又は異なって任意のアミノ酸を示す。X3で示されるアミノ酸としては、例えば、リジン、アルギニン、ヒスチジン等の塩基性側鎖を有するアミノ酸; アスパラギン酸、グルタミン酸といった酸性側鎖を有するアミノ酸; グリシン、アスパラギン、グルタミン、セリン、スレオニン、チロシン、システインといった非帯電性極性側鎖を有するアミノ酸; アラニン、バリン、ロイシン、イソロイシン、プロリン、フェニルアラニン、メチオニン、トリプトファンといった非極性側鎖を有するアミノ酸; スレオニン、バリン、イソロイシンといったβ-分枝側鎖を有するアミノ酸; チロシン、フェニルアラニン、トリプトファン、ヒスチジンといった芳香族側鎖を有するアミノ酸等が挙げられる。X3で示されるアミノ酸は、プロリン以外のアミノ酸であることが好ましい。
X3で示されるアミノ酸により、LCSTを調整することができる。X3で示されるアミノ酸が親水性アミノ酸である場合、LCSTが高くなりすぎるので、好ましくない。LCSTを適切な(生理的に許容可能な)温度(例えば30~40℃)に調整し易いという観点から、P配列ブロック中のX3で示されるアミノ酸の一部はバリン、ロイシン等の非芳香族性疎水性アミノ酸(好ましくはバリン)であり、他のアミノ酸としては、フェニルアラニン、トリプトファン等の芳香族性疎水性アミノ酸(好ましくはフェニルアラニン)であることが好ましい。この場合、P配列ブロック中のX3の総数に対するX3で示される非芳香族性疎水性アミノ酸の数の割合は、例えば60~95%、好ましくは70~80%、より好ましくは75~85%であり、P配列ブロック中のX3の総数に対するX3で示される芳香族性疎水性アミノ酸の数の割合は、例えば5~40%、好ましくは20~30%、より好ましくは15~25%である。さらに、この場合、P配列ブロック中、X3で示される芳香族性疎水性アミノ酸はできるだけ分散して出現することが好ましく、例えば1~7つ、好ましくは1~5つ、より好ましくは2~4つ、さらに好ましくは3つのリピート単位(VPGX3G)の繰返しに対して1つの割合で、X3が芳香族性疎水性アミノ酸であるリピート単位が出現することが好ましい。

エラスチン様ブロックペプチド配列は、G配列ブロック及びP配列ブロックを含むものである限り、特に制限されない。エラスチン様ブロックペプチド配列中のG配列ブロックの数は、特に制限されないが、例えば1~5、好ましくは2~5、より好ましくは2~4、さらに好ましくは2~3、特に好ましくは2である。エラスチン様ブロックペプチド配列中のP配列ブロックの数は、特に制限されないが、例えば1~5、好ましくは1~4、より好ましくは1~3、さらに好ましくは1~2である。
ブロック間(G配列ブロックとP配列ブロックとの間、G配列ブロックとG配列ブロックとの間、P配列ブロックとP配列ブロックとの間)は、直接連結されていてもよいし、リンカー配列が介在していてもよい。好ましくは、ブロック間には、リンカー配列が介在する。
リンカー配列としては、エラスチン様ブロックペプチド配列の自己集合性による繊維形成能を著しく低減させない限りにおいて特に制限されず、通常は、大きな制限なく任意のアミノ酸またはアミノ酸配列を採用することができる。リンカー配列のアミノ酸長は、例えば1~20、好ましくは1~15、より好ましくは1~10、さらに好ましくは1~8、よりさらに好ましくは2~8である。リンカー配列の具体例としては、配列番号10:LWLGSGで示されるアミノ酸配列、KLで示されるアミノ酸配列等が挙げられ、さらにこれらのアミノ酸配列に対して1または複数個(例えば1~5個、好ましくは1~3個、より好ましくは1~2個、さらに好ましくは1個)のアミノ酸が変異(例えば置換、欠失、挿入、付加、好ましくは置換、より好ましくは保存的置換)されてなるアミノ酸配列を採用することができる。
本明細書中において、「保存的置換」とは、アミノ酸が類似の側鎖を有するアミノ酸に置換されることを意味する。例えば、リジン、アルギニン、ヒスチジンといった塩基性側鎖を有するアミノ酸同士で置換されることが、保存的な置換にあたる。その他、アスパラギン酸、グルタミン酸といった酸性側鎖を有するアミノ酸;グリシン、アスパラギン、グルタミン、セリン、スレオニン、チロシン、システインといった非帯電性極性側鎖を有するアミノ酸;アラニン、バリン、ロイシン、イソロイシン、プロリン、フェニルアラニン、メチオニン、トリプトファンといった非極性側鎖を有するアミノ酸;スレオニン、バリン、イソロイシンといったβ-分枝側鎖を有するアミノ酸;チロシン、フェニルアラニン、トリプトファン、ヒスチジンといった芳香族側鎖を有するアミノ酸同士での置換も同様に、保存的な置換にあたる。
エラスチン様ブロックペプチド配列の配列構造は、自己集合による繊維形成能の観点から、N末端側から、G配列ブロック-P配列ブロック-G配列ブロック、G配列ブロック-P配列ブロック-P配列ブロック-G配列ブロック、P配列ブロック-G配列ブロック、又はG配列ブロック-P配列ブロックであることが好ましい。該配列構造は、特に、G配列ブロック-P配列ブロック-G配列ブロック、又はG配列ブロック-P配列ブロック-P配列ブロック-G配列ブロックであることが好ましい。これらの構造中、「-」は(ブロック間の)直接連結又は他の配列(例えばリンカー配列等)を示す。
内皮細胞接着配列は、内皮細胞接着性を有するアミノ酸配列である限り、特に制限されない。本発明においては、内皮細胞に対する高い接着性、及び血小板に対する低い接着性の観点から、該配列はREDV(配列番号3)を含むアミノ酸配列であることが特に好ましい。中でも、同様の観点から、該配列はGREDV(配列番号4)であることがとりわけ好ましい。
内皮細胞接着配列は、本発明のポリペプチドの末端(N末端及び/又はC末端)に配置され、好ましくはC末端に配置される。内皮細胞接着配列は、N末端及びC末端のどちらか一方にのみ配置されていてもよいし、両方に配置されていてもよい。本発明の好ましい一態様においては、内皮細胞接着配列は、一方の末端にのみ配置される。
本発明のポリペプチドが内皮細胞接着性を発揮できる限り、内皮細胞接着配列の末端側(本発明のポリペプチドの末端側、例えば内皮細胞接着配列が本発明のポリペプチドのC末端に配置される場合は、内皮細胞接着配列のC末端側)にさらに他のアミノ酸配列(例えば1~5、1~3、1~2、1アミノ酸長)が付加されていてもよい。本発明の特に好ましい態様においては、内皮細胞接着配列の末端側には他のアミノ酸配列は存在しない。
エラスチン様ブロックペプチド配列と内皮細胞接着配列とは、直接連結されていてもよいし、両者の間に他のアミノ酸配列(例えば3~30、7~25、10~20アミノ酸長の配列)が介在していてもよい。この他のアミノ酸配列としては、内皮細胞接着性の観点から、親水性アミノ酸リッチ配列が好ましい。換言すれば、内皮細胞接着配列の末端側とは反対側に親水性アミノ酸リッチ配列が配置されていることが好ましい。親水性アミノ酸リッチ配列は、親水性アミノ酸の割合が高い配列限り特に制限されず、例えば50%以上、60%以上、70%以上、80%以上、90%以上の割合で親水性アミノ酸を含む、例えば3~15、5~10アミノ酸長のアミノ酸配列が挙げられる。親水性アミノ酸リッチ配列としては、本発明のポリペプチドの精製を容易にできるという観点から、ヒスチジン連続配列(Hisタグ配列)が好ましい。他のアミノ酸配列内には、架橋反応に使用できる官能基を有するアミノ酸(例えばリシン、システイン等)が含まれることが好ましい。
本発明の特に好ましい態様においては、本発明のポリペプチドは、配列番号9で示されるアミノ酸配列からなるポリペプチド(GPG-REDV)、又は配列番号9に対して1又は複数個(例えば1~10個、好ましくは1~5個、より好ましくは1~2個、さらに好ましくは1個)のアミノ酸が変異(例えば置換、欠失、挿入、付加、好ましくは置換、より好ましくは保存的置換)されてなるアミノ酸配列からなるポリペプチドである。
本発明のポリペプチドは、内皮細胞に対する高い接着性、及び血小板に対する低い接着性が著しく損なわれない限りにおいて、化学修飾されたものであってもよい。
本発明のポリペプチドは、C末端がカルボキシル基(-COOH)、カルボキシレート(-COO-)、アミド(-CONH2)またはエステル(-COOR)の何れであってもよい。
ここでエステルにおけるRとしては、例えば、メチル、エチル、n-プロピル、イソプロピル、n-ブチルなどのC1-6アルキル基;例えば、シクロペンチル、シクロヘキシルなどのC3-8シクロアルキル基;例えば、フェニル、α-ナフチルなどのC6-12アリール基;例えば、ベンジル、フェネチルなどのフェニル-C1-2アルキル基;α-ナフチルメチルなどのα-ナフチル-C1-2アルキル基などのC7-14アラルキル基;ピバロイルオキシメチル基などが用いられる。
本発明のポリペプチドは、C末端以外のカルボキシル基(またはカルボキシレート)が、アミド化またはエステル化されていてもよい。この場合のエステルとしては、例えば上記したC末端のエステルなどが用いられる。
さらに、本発明のポリペプチドには、N末端のアミノ酸残基のアミノ基が保護基(例えば、ホルミル基、アセチル基などのC1-6アルカノイルなどのC1-6アシル基など)で保護されているもの、生体内で切断されて生成し得るN末端のグルタミン残基がピログルタミン酸化したもの、分子内のアミノ酸の側鎖上の置換基(例えば-OH、-SH、アミノ基、イミダゾール基、インドール基、グアニジノ基など)が適当な保護基(例えば、ホルミル基、アセチル基などのC1-6アルカノイル基などのC1-6アシル基など)で保護されているもの、あるいは糖鎖が結合したいわゆる糖タンパク質などの複合タンパク質なども包含される。
本発明のポリペプチドは、酸または塩基との薬学的に許容される塩の形態であってもよい。塩は、薬学的に許容される塩である限り特に限定されず、酸性塩、塩基性塩のいずれも採用することができる。例えば酸性塩の例としては、塩酸塩、臭化水素酸塩、硫酸塩、硝酸塩、リン酸塩等の無機酸塩; 酢酸塩、プロピオン酸塩、酒石酸塩、フマル酸塩、マレイン酸塩、リンゴ酸塩、クエン酸塩、メタンスルホン酸塩、パラトルエンスルホン酸塩等の有機酸塩; アスパラギン酸塩、グルタミン酸塩等のアミノ酸塩等が挙げられる。また、塩基性塩の例として、ナトリウム塩、カリウム塩等のアルカリ金属塩; カルシウム塩、マグネシウム塩等のアルカリ土類金属塩等が挙げられる。
本発明のポリペプチドは、溶媒和物の形態であってもよい。溶媒は、薬学的に許容されるものであれば特に限定されず、例えば水、エタノール、グリセロール、酢酸等が挙げられる。
本発明のポリペプチドは、公知の遺伝子工学的手法に従って容易に製造することができる。例えば、PCR、制限酵素切断、DNA連結技術、in vitro転写・翻訳技術、リコンビナントタンパク質作製技術等を利用して製造することができる。
2.ポリヌクレオチド、細胞
本発明は、その一態様において、本発明のポリペプチドのコード配列を含む、ポリヌクレオチド(本明細書において、「本発明のポリヌクレオチド」と示すこともある。)に関する。以下、これについて説明する。

本発明のポリペプチドの発現カセットは、細胞内で本発明のポリペプチドを発現可能なDNAである限り特に制限されない。本発明のポリペプチドの発現カセットの典型例としては、プロモーター、及びそのプロモーターの制御下に配置された本発明のポリペプチドのコード配列を含むDNAが挙げられる。
対象細胞の由来生物種としては、特に制限されず、例えば腸内細菌科細菌等の細菌、酵母等の真菌、動物、植物が挙げられる。動物としては、特にヒト、サル、マウス、ラット、イヌ、ネコ、ウサギ、ブタ、ウマ、ウシ、ヒツジ、ヤギ、シカ等の種々の哺乳類動物以外にも、非哺乳類脊椎動物、無脊椎動物等も挙げられる。本発明の好ましい一態様において、対象細胞の由来生物種としては、哺乳類、大腸菌、酵母、枯草菌、ゼブラフィッシュ、メダカ、昆虫等が挙げられる。また、細胞の種類としても、特に制限されず、各種組織由来又は各種性質の細胞、例えば血液細胞、造血幹細胞・前駆細胞、配偶子(***、卵子)、線維芽細胞、上皮細胞、血管内皮細胞、神経細胞、肝細胞、ケラチン生成細胞、筋細胞、表皮細胞、内分泌細胞、ES細胞、iPS細胞、組織幹細胞、がん細胞等が挙げられる。
本発明のポリペプチドの発現カセットに含まれるプロモーターとしては、特に制限されず、対象細胞に応じて適宜選択することができる。プロモーターとしては、例えばpol II系プロモーターを各種使用することができる。pol II系プロモーターとしては、特に制限されないが、例えばCMVプロモーター、EF1プロモーター、SV40プロモーター、MSCVプロモーター、hTERTプロモーター、βアクチンプロモーター、CAGプロモーター等が挙げられる。その他にも、プロモーターとして、例えばtrcやtac等のトリプトファンプロモーター、lacプロモーター、T7プロモーター、T5プロモーター、T3プロモーター、SP6プロモーター、アラビノース誘導プロモーター、コールドショックプロモーター、テトラサイクリン誘導性プロモーター等が挙げられる。
本発明のポリペプチドの発現カセットは、必要に応じて、他のエレメント(例えば、マルチクローニングサイト(MCS)、薬剤耐性遺伝子、複製起点、エンハンサー配列、リプレッサー配列、インスレーター配列、レポータータンパク質(例えば、蛍光タンパク質等)コード配列、薬剤耐性遺伝子コード配列などが挙げられる。)を含んでいてもよい。MCSは複数(例えば2~50、好ましくは2~20、より好ましくは2~10)個の制限酵素サイトを含むものであれば特に制限されない。本発明のポリペプチドが機能性ドメインを含まない場合であれば、該MCSは、任意の機能性ドメインのコード配列を挿入に使用することができる。
薬剤耐性遺伝子としては、例えばクロラムフェニコール耐性遺伝子、テトラサイクリン耐性遺伝子、ネオマイシン耐性遺伝子、エリスロマイシン耐性遺伝子、スペクチノマイシン耐性遺伝子、カナマイシン耐性遺伝子、ハイグロマイシン耐性遺伝子、ピューロマイシン耐性遺伝子等が挙げられる。
レポータータンパク質としては、特定の基質と反応して発光(発色)する発光(発色)タンパク質、或いは励起光によって蛍光を発する蛍光タンパク質である限り特に限定されない。発光(発色)タンパク質としては、例えばルシフェラーゼ、βガラクトシダーゼ、クロラムフェニコールアセチルトランスフェラーゼ、βグルクロニダーゼ等が挙げられ、蛍光タンパク質としては、例えばGFP、Azami-Green、ZsGreen、GFP2、HyPer、Sirius、BFP、CFP、Turquoise、Cyan、TFP1、YFP、Venus、ZsYellow、Banana、KusabiraOrange、RFP、DsRed、AsRed、Strawberry、Jred、KillerRed、Cherry、HcRed、mPlum等が挙げられる。
本発明のポリペプチドの発現カセットは、これのみで、或いは他の配列と共に発現ベクターを構成していてもよい。ベクターの種類は、特に制限されず、例えば動物細胞発現プラスミド等のプラスミドベクター; レトロウイルス、レンチウイルス、アデノウイルス、アデノ随伴ウイルス、ヘルペスウイルス、センダイウイルス等のウイルスベクター等が挙げられる。また、その他にも、ベクターとしては、例えば、大腸菌においてはpBR322誘導体に代表されるColE1系プラスミド、p15Aオリジンを持つpACYC系プラスミド、pSC系プラスミド、Bac系等のF因子由来ミニFプラスミドが挙げられる。
本発明のポリヌクレオチドは、公知の遺伝子工学的手法に従って容易に作製することができる。例えば、PCR、制限酵素切断、DNA連結技術等を利用して作製することができる。

3.材料、用途

に関する。以下に、これらについて説明する。

本発明の繊維状自己集合体は、本発明のポリペプチドが自己集合して形成される繊維であり、この限りにおいて特に制限されない。
本発明の繊維状自己集合体の直径は、特に制限されないが、ナノサイズであることが好ましい。具体的には、例えば5~300nm、好ましくは10~150nm、より好ましくは30~100nmである。
本発明の繊維状自己集合体は、例えば、本発明のポリペプチドをLCST未満の温度(例えば0~15℃、2~10℃)で溶媒中に溶解させた後、得られた溶液をLCST以上(例えば30~45℃、30~40℃)に加温して一定期間静置することにより、得ることができる。溶媒としては水が使用されるが、水と有機溶媒の混合溶媒も使用することができる。溶液中の本発明のポリペプチドの濃度は、特に制限されないが、例えば1~50μM、好ましくは5~40μM、より好ましくは10~30μMである。静置時間は、特に制限されないが、例えば1~15日間、好ましくは2~10日間、より好ましくは5~10日間である。
本発明の成型体は、例えば、次のようにして得ることができる。本発明の繊維状自己集合体を比較的高い濃度(例えば、0.2~0.4%)で含む溶液を所定の温度(例えば30~40℃)で一定期間静置することにより、溶液がゲル化する。このゲルを、そのまま本発明の成型体として使用することもできるし、適宜乾燥させて得られた物を本発明の成型体として使用することもできる。成型体は、シート状、管状等の任意の形状をとることができる。
本発明の複合材料に使用される基材としては、特に制限されず、軟質材料、硬質材料等を使用することができる。基材としては、例えば、PET、PTFEなどの各種高分子材料を使用することができる。
医療機器、生体材料としては、例えば人工血管、人工軟骨、癒着防止シート材料、人工心臓弁、眼内レンズ等が挙げられる。実験器具としては、例えば細胞培養基材等が挙げられる。これらの中でも、本発明のポリペプチドの特性の観点から、人工血管が特に好ましい。
以下に、実施例に基づいて本発明を詳細に説明するが、本発明はこれらの実施例によって限定されるものではない。
試験例1.ポリペプチドの合成
下記3つのポリペプチド(GPG1、GPG3、及びGPG-REDV):
(GPG1)N末端側から(配列番号5:VGGVG)5からなるG配列ブロック、(配列番号6:VPGXG)25からなるP配列ブロック(式中、Xは同一又は異なってV又はFを示す。)、(配列番号5:VGGVG)5からなるG配列ブロックの順に、各ブロックがリンカー配列を介在して連結してなるエラスチン様ブロックペプチド配列を含むポリペプチド(配列番号7)、
(GPG3)GPG1のC末端側に細胞接着配列が配置されてなるポリペプチド(配列番号8)、及び
(GPG-REDV)GPG1のC末端側に内皮細胞接着配列が配置されてなるポリペプチド(配列番号9)、
を合成した。GPG1及びGPG3は既報のアミノ酸配列からなるポリペプチドであり、GPG-REDVは今般新たに設計したポリペプチドである。
下記3つのポリペプチド(GPG1、GPG3、及びGPG-REDV):

を合成した。GPG1及びGPG3は既報のアミノ酸配列からなるポリペプチドであり、GPG-REDVは今般新たに設計したポリペプチドである。
具体的には、これらのポリペプチドをコードするプラスミドDNAを用いてE.coli BLR(DE3)株を形質転換し、ポリペプチドを発現させた。His-tagを用いた金属イオンアフィニティークロマトグラフィーで精製して、ポリペプチドを得た。SDS-PAGE及びMALDI-TOF-MSにより、目的のポリペプチドが得られていることを確認した。
試験例2.ファイバー形成の確認
GPG-REDVを20μMの濃度で冷水に溶解後、37℃で7日間静置し、自己集合性ナノファイバーを得た。ナノファイバーは透明分散液として得られた。ナノファイバーの形成は、原子間力顕微鏡観察により確認した。

以上より、GPG-REDVは、既報のGPG1及びGPG3と同様に、自己集合性ナノファイバーを形成することが分かった。
試験例3.コーティング効率の評価1
ポリペプチド(GPG1、GPG3、及びGPG-REDV)をファイバー形成させた場合と、ファイバー未形成(単分子状)の場合とで、基板へのコーティング率を測定した。具体的には以下のようにして行った。

各ポリペプチドを20μMの濃度で冷水に溶解後に2つに分け、一方は37℃で7日間静置し(ファイバー形成サンプル)、他方は4℃で7日間静置した(ファイバー未形成サンプル)。両サンプルに超純水を添加して、ポリペプチド濃度を0.1mg/mLに調整した。得られたサンプルを、ホウケイ酸ガラス製組織培養用プレート上に滴下し(100μL/cm2)、37℃で4時間静置した。プレート上の余分な液を除去して、37℃で12時間乾燥させた。プレート上にUVを12時間照射して滅菌した後、HEPES緩衝生理食塩水でプレート上を洗浄し、コーティングプレートを得た。洗浄液に含まれるタンパク質量をMicroBCAアッセイで求めることで、基板に残存したタンパク量を算出した。
結果を表1に示す。
試験例4.コーティング効率の評価2
GPG1、GPG3、及びGPG-REDVとコラーゲンとで、コーティング率を比較した。GPG1、GPG3、及びGPG-REDVについては、ファイバー形成サンプルを使用し、サンプルを組織培養用プレート上に滴下した後の静置時間を2時間とする以外は、試験例3と同様にして行った。また、コラーゲンについても、ファイバー形成用静置処理(37℃7日間)を行わない以外は、GPG1、GPG3、及びGPG-REDVと同様にして行った。
GPG1、GPG3、及びGPG-REDVとコラーゲンとで、コーティング率を比較した。GPG1、GPG3、及びGPG-REDVについては、ファイバー形成サンプルを使用し、サンプルを組織培養用プレート上に滴下した後の静置時間を2時間とする以外は、試験例3と同様にして行った。また、コラーゲンについても、ファイバー形成用静置処理(37℃7日間)を行わない以外は、GPG1、GPG3、及びGPG-REDVと同様にして行った。
結果を表2に示す。
試験例5.血管内皮細胞の接着性の評価
試験例4で得られた各コーティングプレート(1wellの面積:0.3cm2)に、HUVEC細胞(ヒト臍帯静脈内皮細胞)を1.0×104cells/wellになるように播種し、37℃で1時間又は24時間培養した。培養後、細胞を4%パラホルムアルデヒドで固定(4℃、30分間)し、続いて0.5%TritonX-100で透過処理(4℃、25分間)し、続いて1%BSAでブロッキング処理(25℃、1時間)した。一次抗体(抗ビンキュリン抗体及び抗アクチン抗体)溶液で処理(4℃、12時間)し、続いて二次抗体処理(抗ビンキュリン抗体に対する蛍光標識抗体溶液で25℃で1時間処理後に、抗アクチン抗体に対する蛍光標識抗体溶液で4℃で30分間処理)し、続いて核染色処理(25℃、5分間)を行った。細胞を、蛍光顕微鏡で観察し、核の数を計測して、得られた値を元に、プレートの単位面積当たりの細胞数を算出した。さらに、観察像から、プレート面積に対する細胞の占有面積の割合を算出した。
試験例4で得られた各コーティングプレート(1wellの面積:0.3cm2)に、HUVEC細胞(ヒト臍帯静脈内皮細胞)を1.0×104cells/wellになるように播種し、37℃で1時間又は24時間培養した。培養後、細胞を4%パラホルムアルデヒドで固定(4℃、30分間)し、続いて0.5%TritonX-100で透過処理(4℃、25分間)し、続いて1%BSAでブロッキング処理(25℃、1時間)した。一次抗体(抗ビンキュリン抗体及び抗アクチン抗体)溶液で処理(4℃、12時間)し、続いて二次抗体処理(抗ビンキュリン抗体に対する蛍光標識抗体溶液で25℃で1時間処理後に、抗アクチン抗体に対する蛍光標識抗体溶液で4℃で30分間処理)し、続いて核染色処理(25℃、5分間)を行った。細胞を、蛍光顕微鏡で観察し、核の数を計測して、得られた値を元に、プレートの単位面積当たりの細胞数を算出した。さらに、観察像から、プレート面積に対する細胞の占有面積の割合を算出した。
結果を図1及び2に示す。GPG-REDVコーティングプレートに対する血管内皮細胞の接着性は、他のコーティングプレートやコーティング無しのプレート(組織培養用プレート)に対する接着性と同程度であった。
試験例6.血管内皮細胞の増殖性の評価
試験例4で得られた各コーティングプレート(1wellの面積:0.3cm2)に、HUVEC細胞(ヒト臍帯静脈内皮細胞)を1.0×103cells/wellになるように播種し、37℃で1、3、5、又は7日間培養した。培養後、細胞を4%パラホルムアルデヒドで固定(4℃、30分間)し、続いて0.5%TritonX-100で透過処理(4℃、25分間)し、続いて1%BSAでブロッキング処理(25℃、1時間)した。一次抗体(抗フォン・ヴィレブランド因子抗体及び抗アクチン抗体)溶液で処理(4℃、12時間)し、続いて二次抗体処理(抗フォン・ヴィレブランド因子抗体に対する蛍光標識抗体溶液で25℃で1時間処理後に、抗アクチン抗体に対する蛍光標識抗体溶液で4℃で30分間処理)し、続いて核染色処理(25℃、5分間)を行った。細胞を、蛍光顕微鏡で観察し、核の数を計測して、得られた値を元に、プレートの単位面積当たりの細胞数を算出した。

結果を図3に示す。GPG-REDVコーティングプレートにおける血管内皮細胞の増殖性は、コーティング無しのプレート(組織培養用プレートそのもの)における増殖性よりも高かった。
試験例7.血小板の接着性の評価
全血を200gで5分間遠心して上清を回収した。上清を1500gで10分間遠心して、沈殿(血小板)を細胞培養培地に懸濁して、血小板懸濁液を得た。試験例4で得られた各コーティングプレート(1wellの面積:0.9cm2)に、血小板懸濁液を3.5×106cells/wellになるように播種し、37℃で1時間静置した。血小板をPBSで洗浄後、血小板を1%グルタルアルデヒドで固定(37℃、2時間)した。血小板をPBSで洗浄し、続いて50%PBS(超純水希釈)で洗浄し、続いて超純水で洗浄した。血小板を、電界放出型走査電子顕微鏡(FE-SEM)で観察し、プレートの単位面積当たりの各形態(Type1:偽足0本、Type2:偽足1-2本、Type3:偽足3本以上)の血小板数を算出した。
全血を200gで5分間遠心して上清を回収した。上清を1500gで10分間遠心して、沈殿(血小板)を細胞培養培地に懸濁して、血小板懸濁液を得た。試験例4で得られた各コーティングプレート(1wellの面積:0.9cm2)に、血小板懸濁液を3.5×106cells/wellになるように播種し、37℃で1時間静置した。血小板をPBSで洗浄後、血小板を1%グルタルアルデヒドで固定(37℃、2時間)した。血小板をPBSで洗浄し、続いて50%PBS(超純水希釈)で洗浄し、続いて超純水で洗浄した。血小板を、電界放出型走査電子顕微鏡(FE-SEM)で観察し、プレートの単位面積当たりの各形態(Type1:偽足0本、Type2:偽足1-2本、Type3:偽足3本以上)の血小板数を算出した。
結果を図4に示す。GPG-REDVコーティングプレートに対する血小板の接着性は、細胞接着配列を有するGPG3又はコラーゲンでコートされたプレートに対する接着性、さらにはコーティング無しのプレート(組織培養用プレートそのもの)に対する接着性よりも大幅に低く、細胞接着配列を有しないGPG1コーティングプレートに対する接着性と同程度であった。
以上より、GPG-REDVは、内皮細胞高接着性と血小板低接着性の両方を併せ持ち、人工血管材料として適していることが分かった。
試験例8.血管平滑筋細胞の接着性及び増殖性の評価
平滑筋細胞には、収縮型と合成型が存在する。収縮型は、正常な血管に存在し、紡錘形である、収縮性を有する、アルファ平滑筋アクチン(αSMA)の発現する等の特徴を有する。一方、合成型は、病理状態の血管に存在し、丸い形状を有する、細胞外マトリックスの合成が活発である、高い増殖性と遊走能を有する等の特徴を有する。人工血管材料は、平滑筋細胞が問題なく培養でき、且つ平滑筋細胞の合成型への分化を誘導しないことが求められる。平滑筋細胞が活性化(合成型へ分化)してしまうと、血管が肥厚する(開存性が保たれない)原因となる。そこで、血管平滑筋細胞の接着性、増殖性等を評価した。具体的には、以下のようにして行った。

試験例4で得られた各コーティングプレート(1wellの面積:0.3cm2)に、HUASMC細胞(ヒト臍帯動脈平滑筋細胞)を1.0×104cells/wellになるように播種し、37℃で24時間培養した。培養後、細胞を4%パラホルムアルデヒドで固定(4℃、30分間)し、続いて0.5%TritonX-100で透過処理(4℃、25分間)し、続いて1%BSAでブロッキング処理(25℃、1時間)した。一次抗体(抗ビンキュリン抗体及び抗アクチン抗体)溶液で処理(4℃、12時間)し、続いて二次抗体処理(抗ビンキュリン抗体に対する蛍光標識抗体溶液で25℃で1時間処理後に、抗アクチン抗体に対する蛍光標識抗体溶液で4℃で30分間処理)し、続いて核染色処理(25℃、5分間)を行った。細胞を、蛍光顕微鏡で観察し、核の数を計測して、得られた値を元に、プレートの単位面積当たりの細胞数を算出した。さらに、観察像から、プレート面積に対する細胞の占有面積の割合を算出した。

<試験例8-2.増殖性評価>

<試験例8-3.遺伝子発現量の測定>
試験例4で得られた各コーティングプレート(1wellの面積:0.3cm2)に、HUASMC細胞(ヒト臍帯動脈平滑筋細胞)を2.0×103cells/wellになるように播種し、37℃で7日間培養した。CellAmpTM Direct Lysis and RT set(タカラバイオ社製)を用い、推奨プロトコルに従ってセルライセート作製と逆転写反応を行った。続いて、インターカレーター法を用いたリアルタイムポリメラーゼ連鎖反応(rt-PCR)によりαSMA (ACTA2) の遺伝子発現量を測定した。ハウスキーピング遺伝子としてグリセルアルデヒドー3-リン酸脱水素酵素(GAPDH)を用いた。Primerの配列を下表に示す。TB GreenTM Premix Ex TaqTMII(タカラバイオ社製)12.5 μL、forward primer (10 μmol/L)およびreverse primer(10 μmol/L) 各1μL、上記で調製済の逆転写反応液 2.5 μL、滅菌精製水 8 μL を氷上で混合してPCR反応液を調製した。この反応液から Thermal Cycler Dice Real Time System III(タカラバイオ社製)を用いて標準プロトコルに従いPCR反応を行った。増幅曲線と閾値線の交点(Ct値)をCrossing Point法で求め、ΔΔCt法でACTA2発現量を算出した。使用したプライマー(配列番号11~14)を表3に示す。

<結果>
試験例8-1の結果を図5に示す。平滑筋細胞は、GPG-REDVに、細胞培養用ガラス基板よりも有意に多く接着した。細胞占有率は、GPG-REDVとガラス基板で同程度であった。

試験例8-2の結果を図6に示す。平滑筋細胞は、GPG-REDV上で増殖性を有しつつも、その増殖性はガラス基板よりも抑制されていた。
試験例8-3の結果を図7に示す。GPG-REDVの場合、平滑筋細胞が収縮型を保っていることを示すαSMAの遺伝子発現量が多い。これは、平滑筋細胞の過増殖を抑制できることを示している。
以上より、GPG-REDVは人工血管材料として適していることが分かった。
Claims (13)
- (配列番号1:X1GGX2G)nからなるG配列ブロック(式中、X1は同一又は異なってV又はLを示し、X2は同一又は異なってV又はLを示し、nは4以上の整数を示す。)と(配列番号2:VPGX3G)mからなるP配列ブロック(式中、X3は同一又は異なって任意のアミノ酸を示し、mは5以上の整数を示す。)を含むエラスチン様ブロックペプチド配列、及び末端に配置された内皮細胞接着配列を含む、ポリペプチド。
- 前記エラスチン様ブロックペプチド配列の配列構造が、N末端側から、G配列ブロック-P配列ブロック-G配列ブロック、G配列ブロック-P配列ブロック-P配列ブロック-G配列ブロック、P配列ブロック-G配列ブロック、又はG配列ブロック-P配列ブロックである、請求項1に記載のポリペプチド。
- 前記ブロック間にリンカー配列が介在する、請求項1又は2に記載のポリペプチド。
- 前記内皮細胞接着配列がREDV(配列番号3)を含む、請求項1~3のいずれかに記載のポリペプチド。
- 前記内皮細胞接着配列がGREDV(配列番号4)である、請求項1~4のいずれかに記載のポリペプチド。
- 前記内皮細胞接着配列の末端側とは反対側に親水性アミノ酸リッチ配列が配置されている、請求項1~5のいずれかに記載のポリペプチド。
- 請求項1~6のいずれかに記載のポリペプチドのコード配列を含む、ポリヌクレオチド。
- 請求項1~6のいずれかに記載のポリペプチドの発現ベクターである、請求項7に記載のポリヌクレオチド。
- 請求項7又は8に記載のポリヌクレオチドを含む、細胞。
- 請求項1~6のいずれかに記載のポリペプチドの繊維状自己集合体。
- 請求項10に記載の繊維状自己集合体を含む、成型体。
- 請求項10に記載の繊維状自己集合体及び請求項11に記載の成型体からなる群より選択される少なくとも1種と、基材とを含む、複合材料。
- 請求項10に記載の繊維状自己集合体、請求項11に記載の成型体、及び請求項12に記載の複合材料からなる群より選択される少なくとも1種を含む、人工血管。
