 供試菌としてStaphylococcus aureus NBRC12732及びStaphylococcus epidermidis ATCC12228を用いた。SCD寒天培地上で培養した供試菌を、SCD培地を1/10濃度で含有する生理食塩水中に懸濁し、1.0×10CFU/mL以上、9.0×10CFU/mL以下になるように菌液を調製した。菌液を各合成皮革表面に0.1mL添加し、4cm×4cmのストマッカーフィルムを被せ、菌液がフィルム全体に行き渡るように軽く押さえつけた。シャーレ内に入れて相対湿度90%以上、温度37±1℃で24±1時間培養した。これをフィルムごと不活化剤であるLP希釈液10mL中に投入し、よく揉んで試験菌を洗いだした。これをSCD寒天培地で混釈培養し、生菌数カウントを行い、下記式に基づき抗菌活性値を算出した。
