他方の限外ろ過法では、主には低分子量の溶質が限外ろ過膜のフィルターを通り抜けてしまうことがあるので、十分な収率が得られないことがある。つまり、限外ろ過膜の分子分画分子量以下の物質は、その一部又はすべてが膜を透過してしまうため、原料液中の成分組成を保ったまま濃縮することができない。

本実施形態の原料液濃縮システムは、
溶媒及び溶質を含有する原料液と、誘導溶質を含有する誘導溶液とを、正浸透膜を介して接触させ、原料液中の溶媒を誘導溶液中に移動させるとともに誘導溶液中の誘導溶質を原料液中に移動させることにより、濃縮原料液及び希釈誘導溶液を得るための、第一のユニットと、
濃縮原料液を凍結乾燥処理して更に濃縮された生成物を得るための、第二のユニットと
を有する。
本実施形態の原料液濃縮システムの好ましい例は、上記の第一のユニットと第二のユニットとを有し、第一のユニットにおける誘導溶液中の誘導溶質を原料液中へ移動させる誘導溶質の逆拡散速度が、200g/(m2×hr)以下のシステムである。
また、本実施形態の原料液濃縮システムの他の好ましい例は、上記の第一のユニットと第二のユニットとを有し、上記の誘導溶質の逆拡散速度が、0.001g/(m2×hr)以上、50g/(m2×hr)以下のシステムである。
上記の医薬品原料とは、医薬品を製造するための原料化合物を意味する。上記の医薬品製造プロセスにおける反応生成物とは、医薬品を製造するために進められる化学反応で得た生成物を意味する。
(1)原料液と、誘導溶液とを、正浸透膜を介して接触させ、原料液中の溶媒を誘導溶液中に移動させること。これによって、原料液の濃縮を行うとともに、誘導溶液が希釈され、濃縮原料液及び希釈誘導溶液を得る。
(2)誘導溶液中の誘導溶質を原料液中へ移動させること。
第二のユニットでは、第一のユニットで得られた濃縮原料液を凍結乾燥法によって更に濃縮し、水分量が好ましくは5質量%以下にまで減少された生成物(濃縮物)を得る。
本明細書では、第一のユニットで行われる工程を「第一の工程」と称し、本実施形態の第二のユニットで行われる工程を「第二の工程」と称する。すなわち、本明細書において、「第一の工程」及び「第二の工程」は、それぞれ「第一のユニット」及び「第二のユニット」に置き換えることができ、また逆に、「第一のユニット」及び「第二のユニット」は、それぞれ「第一の工程」及び「第二の工程」に置き換えることができる。
凍結乾燥法の前段階を行うユニットとしては、例えば、予備凍結ユニット、殺菌ユニット、脱塩ユニット、pH調整ユニット等が挙げられる。
第一のユニットによる第一の工程と第二のユニットによる第二の工程とは、時間間隔を置かずに連続して行ってもよいし、所定の時間間隔を開けて行ってもよい。例えば、第一のユニットによって得られた濃縮原料液を一時的に貯蔵し、所定の時間が経過した後に、第二のユニットに供して、第二の工程を行ってもよい。しかしながら、第一のユニットと第二のユニットとを連結させ、第一の工程と第二の工程との間に時間間隔を置かずに、連続して濃縮を行うことが、時間的な効率の点でより好ましい。
図1の原料液濃縮システムの第一のユニットは、正浸透膜oを有し、正浸透処理を行う正浸透膜ユニットである。この正浸透膜ユニットの内部空間は、正浸透膜oによって、原料液側空間R及び誘導溶液側空間Dの2つに分割されている。正浸透膜ユニットの原料液側空間Rに、濃縮対象物である原料液aを導入する。他方、正浸透膜ユニットの誘導溶液側空間Dには、誘導溶液dを導入する。
原料液aは、溶質及び溶媒bを含有する。誘導溶液dは、誘導溶質(例えば、無機塩)を含有し、更に溶媒bを含有することが好ましい。誘導溶液dの浸透圧は、原料液aよりも高くなるように設定されている。
同時に、(2)誘導溶質の分圧差を駆動力として、誘導溶液d中の誘導溶質sは、正浸透膜oを通過して、原料液a中に移動する。これを誘導溶質sの逆拡散rとする。
これにより、濃縮された原料液である濃縮原料液cと、希釈された誘導溶液である希釈誘導溶液eとが得られる。図1における第一のユニットでは、原料液aと誘導溶液dとを向流させているが、並流でもよい。
第一のユニットにおける正浸透処理は、全量ろ過方式によってもクロスフローろ過方式によってもよいが、クロスフローろ過方式によることが、ろ過流速及び膜汚染抑制の観点から好ましい。
凍結乾燥は、典型的には、チャンバー内における以下の3つの段階を含む:
(A)凍結段階;
(B)一次乾燥段階;及び
(C)二次乾燥段階
段階(A)は、第一のユニットによって得られた濃縮原料液c中の溶媒を凍結する段階である。段階(B)においては、チャンバー内の圧力を低下させ(例えば、13.3Pa(0.1トール)以下まで)、熱を加えて、凍結溶媒を昇華させる。段階(C)において、チャンバー内の温度を上昇させて、結晶水等の結合溶媒も含めて、残留溶媒含有量が所望のレベルに下がるまで、取り除く。このようにして、濃縮原料液cが更に濃縮された、生成物fを得る。ここで、脱溶媒を促進する方法として、チャンバー圧力の低下に代えて、乾燥された気体を流入させる方法を用いてもよい。
図2の原料液濃縮システムにおける第一のユニット及び第二のユニットは、それぞれ、図1の原料液濃縮システムにおける第一のユニット及び第二のユニットと同じであってよい。しかしながら、図2の原料液濃縮システムでは、第一のユニットで得られた濃縮原料液を、第一のユニットにおける原料液として再使用する循環機構を有している。この実施態様では、例えば、原料液aを第一のユニットに1回通して得られた濃縮原料液の浸透圧が、所定の値に到達しないときに、当該濃縮原料液の少なくとも一部を第一のユニットに複数回通すことにより、所定の浸透圧の濃縮原料液cを得られる利点を有する。
この場合、原料液aを第一のユニットに通す回数(すなわち、第一のユニットで得られた濃縮原料液を、第一のユニットにおける原料液として再使用する回数)は任意である。
図3の原料液濃縮システムにおける第一のユニット及び第二のユニットは、それぞれ、図1の原料液濃縮システムにおける第一のユニット及び第二のユニットと同じであってよい。しかしながら、図3の原料液濃縮システムでは、誘導溶液再生ユニット(第一の誘導溶液再生ユニット)を更に有している。図3では、本実施形態の第一の誘導溶液再生工程を行うためのユニットが「第一の誘導溶液再生ユニット」として示されている。本明細書において、「第一の誘導溶液再生工程」は、「第一の誘導溶液再生ユニット」に置き換えることができ、また逆に、「第一の誘導溶液再生ユニット」は、「第一の誘導溶液再生工程」に置き換えることができる。この第一の誘導溶液再生ユニットは、第一のユニットで得られた希釈誘導溶液eから溶媒bを除去して濃縮し、再生誘導溶液gを得るとともに、得られた再生誘導溶液gを、再び誘導溶液dとして循環させる機能を有していてよい。希釈誘導溶液eからの溶媒bの除去は、公知の濃縮手段、例えば蒸発手段等によって行われてよい。
なお、再生誘導溶液gに溶媒bの一部が含まれてもよい。例えば、溶媒bが水を含む他成分系であり、他成分が共沸成分である場合は、溶媒bの除去は困難となる。そのため、再生誘導溶液gに溶媒bの一部が含まれることになるが、システム上問題にはならない。
図4の原料液濃縮システムにおける第一のユニット及び第二のユニットは、それぞれ、図1の原料液濃縮システムにおける第一のユニット及び第二のユニットと同じであってよい。しかしながら、図4の原料液濃縮システムでは、図3の原料液濃縮システムとは別の態様の誘導溶液再生ユニット(第二の誘導溶液再生ユニット)を有している。図4では、本実施形態の第二の誘導溶液再生工程を行うためのユニットが「第二の誘導溶液再生ユニット」として示されている。本明細書において、「第二の誘導溶液再生工程」は、「第二の誘導溶液再生ユニット」に置き換えることができ、また逆に、「第二の誘導溶液再生ユニット」は、「第二の誘導溶液再生工程」に置き換えることができる。図4の原料液濃縮システムにおける第二の誘導溶液再生ユニットでは、先ず、誘導溶液dから溶媒bを除去して濃縮誘導溶液hを得る。そして、ここで得られた濃縮誘導溶液hと、第一の工程で得られた希釈誘導溶液eとを混合して混合物(再生誘導溶液g)を得て、この再生誘導溶液gを誘導溶液dとして使用する機能を有していてよい。この第二の誘導溶液再生ユニットにおいて、誘導溶液dからの溶媒bの除去は、公知の濃縮手段、例えば蒸発手段等によって行われてよい。濃縮誘導溶液hと希釈誘導溶液eとの混合は、例えば、図4に示したようなバッファタンク内で行われてよい。
なお、濃縮誘導溶液hに溶媒bの一部が含まれてもよい。例えば、溶媒bが水を含む他成分系であり、他成分が共沸成分である場合は、溶媒bの除去は困難となる。そのため、濃縮誘導溶液hに溶媒bの一部が含まれることになるが、システム上問題にはならない。
図5の原料液濃縮システムにおける第一のユニット、及び第二のユニットは、それぞれ、図1の原料液濃縮システムにおける第一のユニット及び第二のユニットと同じであってよい。図5の原料液濃縮システムは、図2に示した循環機構と、図4に示した第二の誘導溶液再生ユニットとを、重畳的に有する。
図5の原料液濃縮システムにおいて、第二の誘導溶液再生ユニットに代えて、図3に示した態様のもの(第一の誘導溶液再生ユニット)を採用してもよい。或いは、第一の誘導溶液再生ユニットと第二の誘導溶液再生ユニットとを重畳的に有していてもよい。
なお、濃縮誘導溶液hに溶媒bの一部が含まれてもよい。例えば、溶媒bが水を含む他成分系であり、他成分が共沸成分である場合は、溶媒bの除去は困難となる。そのため、濃縮誘導溶液hに溶媒bの一部が含まれることになるが、システム上問題にはならない。
引き続き、本実施形態の原料液濃縮システムを構成する各要素について、以下に詳説する。
原料液aとは、溶質及び溶媒bを含有する流体であり、本実施形態の原料液濃縮システムによって濃縮されることが予定されている。この原料液aは、流体である限りにおいて、乳化物であってもよい。
本実施形態に適用される原料液aの溶質としては、糖、アミノ酸、オリゴペプチド、酵素、及び核酸から成る群から選択される有用物質が挙げられる。原料液aの溶質として好ましくは、数平均分子量が100~6,000である化合物である。分子量が100未満の溶質は、第一のユニット中の正浸透膜を透過する場合がある。分子量が6,000を超える溶質は、正浸透膜表面に付着する場合がある。溶質が、分子量500以下の低分子量体である場合、その数平均分子量は、構造式から計算される分子量であってよい。溶質が、分子量500を超える高分子量体である場合、その数平均分子量は、ゲルパーミエーションクロマトグラフィーによって測定された、ポリエチレンオキシド換算の数平均分子量であってよい。
非天然アミノ酸の一例として、例えば、標識化合物と結合したアミノ酸である「標識化アミノ酸」が挙げられる。標識化アミノ酸としては、例えば、側鎖にベンゼン環等の芳香環を含むアミノ酸骨格を有するアミノ酸に標識化合物を結合させたアミノ酸等が挙げられる。また、特定の機能が付与された非天然アミノ酸の例として、例えば、光応答性アミノ酸、光スイッチアミノ酸、蛍光プローブアミノ酸、蛍光標識アミノ酸等が挙げられる。
誘導溶液dは、誘導溶質を含有し、更に溶媒bを含有することが好ましい。そして、誘導溶液dは、原料液aよりも高い浸透圧を持ち、かつ、正浸透膜oを著しく変性させない流体である。
本実施形態で使用可能な誘導溶質としては、例えば、無機塩、糖、アルコール、重合体等を挙げることができる。したがって本実施形態における誘導溶液は、無機塩、糖、アルコール、重合体等から選択される1種又は2種以上を含む溶液であってよい。なかでも、本実施形態における誘導溶液は、高い浸透圧を有するとの観点から、溶質として、無機塩及びアルコールから成る群から選択される1種又は2種以上を含むことが好ましい。
無機塩としては、例えば、塩化ナトリウム、塩化カリウム、塩化マグネシウム、塩化カルシウム、硫酸ナトリウム、硫酸マグネシウム、チオ硫酸ナトリウム、亜硫酸ナトリウム、塩化アンモニウム、硫酸アンモニウム、炭酸アンモニウム等を;
糖としては、例えば、ショ糖,果糖,ブドウ糖等の一般的な糖類、及びオリゴ糖,希少糖等の特殊な糖類を;
アルコールとしては、例えば、メタノール、エタノール、1-プロパノール、2-プロパノール等のモノアルコール;エチレングルコール、プロピレングリコール等のグリコール等を、挙げることができる。アルコールとして好ましくは、エタノール及び2-プロパノールから成る群から選択される1種又は2種である。
重合体としては、例えば、ポリエチレンオキシド,プロピレンオキシド等の重合体、及びこれらの共重合体等を挙げることができる。
二つの液体間の浸透圧差を判断するには、例えば、以下のいずれかの方法によることができる。
(1)二つの液体を混合後、二相分離する場合:二相分離後に、体積が増えた方の液体の浸透圧が高いと判断する、又は、
(2)二つの液体を混合後、二相分離しない場合:正浸透膜oを介して二つの液体を接触させ、一定時間の経過後に体積が大きくなった液体の浸透圧が高いと判断する。このときの一定時間とは、その浸透圧差に依存するが、一般的には数分から数時間の範囲である。
本実施形態における溶媒bは、例えば、
水と、
アセトニトリル、メタノール、及び2-プロパノールから成る群から選択される1種又は2種以上と
を含有していてよく、これらを主成分とすることが好ましい。ここでいう主成分とは、溶媒b中に、50質量%超え、60質量%以上、80質量%以上、95質量%以上又は100質量%の割合で含まれていることを意味する。
本実施形態における誘導溶液dの溶媒は、水であってよい。
第一のユニットによって濃縮されて得られる濃縮原料液cは、原料液a中の成分が維持され、かつ、溶媒bの少なくとも一部が選択的に分離されている。本実施形態の原料液濃縮システムでは、第一のユニットにおいて、原料液aから分離される溶媒bの量又は割合を、任意に制御することができる。
このように、第一のユニットにおいて、原料液aの浸透圧が十分に高くなるまで正浸透処理を行うことにより、第二のユニットにおける凍結乾燥工程を効率化でき、凍結乾燥における時間的及びエネルギー的な負荷を低減することができる。
本実施形態の原料液濃縮システムの第一の工程では、正浸透処理が行われる。
本実施形態における正浸透処理は、例えば、正浸透膜oによって内部空間が原料液側空間R及び誘導溶液側空間Dの2つに分割された正浸透膜ユニットを用いて実施されてよい。
正浸透膜ユニットの正浸透膜oとは、原料液a中の溶媒bは透過させるが、溶質は透過させない、又は透過させ難い機能を有し、かつ、誘導溶液d中の誘導溶質sを濃縮原料液c中に逆拡散rさせる機能を有する。
本実施形態の原料液濃縮システムの第一のユニットで用いられる正浸透膜oは、逆浸透膜としての機能を有する膜であってもよい。しかしながら、圧力によって溶媒を除去する逆浸透処理と、原料液と誘導溶液との浸透圧の差を利用する正浸透処理とは、溶媒除去に活用される駆動力の違いに起因して、適切な膜構造が異なる。
本実施形態の原料液濃縮システムのように、一連のユニットに正浸透処理が組み込まれているシステムにおいては、正浸透膜としての機能がより高い膜を使用することが好ましい。プロセス中に限外ろ過膜(UF膜)のようなフィルターを配置して、80%以上の溶質を分離し、その透過液中の溶質を回収する目的で、正浸透膜を活用してもよい。
正浸透膜oは、支持層(支持膜)上に分離活性層を有する複合型の膜が好ましい。上記支持膜は、平膜であっても中空糸膜であってもよい。支持層と分離活性層は同一の素材であってもよいし、異なる素材であってもよい。
平膜を支持膜とする場合、支持膜の片面又は両面に分離活性層を有するものであってよい。
中空糸膜を支持膜とする場合、中空糸膜の外表面若しくは内表面、又はこれらの双方の面上に分離活性層を有するものであってよい。
本実施形態において好ましい支持膜は、微細孔性中空糸支持膜である。この微細孔中空糸支持膜は、その内表面に、孔径が好ましくは0.001μm以上2.0μm以下、より好ましくは0.005μm以上1.0μm以下の微細孔を有する。他方、微細孔性中空糸支持膜の内表面から膜の深さ方向に外表面までの構造については、透過する流体の透過抵抗を小さくするために、強度を保ち得る限りでできるだけ疎な構造であることが好ましい。この部分の疎な構造は、例えば網状、指状ボイド等、又はそれらの混合構造のいずれかであることが好ましい。
更に好ましくは、ポリスルホン、ポリエーテルスルホン、ポリフッ化ビニリデン、ポリアクリロニトリル、及びポリアミドから選ばれる少なくとも1種を主成分とすることであり、特に好ましくはポリアミドの層である。
分離活性層におけるポリアミドは、多官能性酸ハライド及び多官能性芳香族アミンの界面重合により形成されることができる。
正浸透膜ユニットとしては、複数の正浸透膜の糸束が好ましくは適当なハウジング内に収納されて構成される、正浸透膜モジュールの形態にあるものを使用することが好ましい。
中空糸膜が、微細孔性支持膜と、微細孔性支持膜の内表面に設けられ、高分子重合体薄膜の分離活性層とを備え、
中空糸糸束を有する中空糸膜の膜面積の合計が0.01m2以上であり、
そして分離活性層の厚み方向の断面を撮影した走査型電子顕微鏡画像における分離活性層部分の質量を測定する方法により算出された、中空糸糸束の半径方向及び長さ方向における分離活性層の厚みの変動係数が0~60%であることが好ましい。
より好ましくは、中空糸糸束を有する中空糸膜の膜面積の合計が1m2以上である。
本実施形態の複合中空糸膜モジュールにおける分離活性層の表面が、このような微細凹形状となる機構につき、本発明者等は以下のように推察している。ただし本発明は、以下の理論に拘束されるものではない。

ここで、Fは溶媒bについての透過流束(L/(m2×hr))であり、Lは透過した溶媒bの容量(L)であり、Mは正浸透膜oの表面積(m2)であり、Hは時間(hr)である。
溶媒bが水である場合の透過流束は、一般に「透水量」と呼ばれ、例えば、処理液として純水を用い、誘導溶液として3.5質量%食塩水を用いて測定することができる。
この逆拡散速度が200g/(m2×hr)以下であれば、原料液中に移動する誘導溶質量が比較的少ないと評価できる。この場合には、第二の工程である凍結乾燥の処理時間が長くなることを回避できる。誘導溶質の逆拡散が過度に多いと、凍結乾燥の処理時間が長くなる。その理由は定かではないが、本発明者等は、次のように推察している。
濃縮原料液中の水は、凍結乾燥処理中に、いったん氷の結晶となったうえ、昇華して除去される。ここで、濃縮原料液中に誘導溶質が存在すると、当該誘導溶質の周囲の水は、非晶質のまま不凍水として固定化される。この固定化された不凍水の除去には長時間を要するため、誘導溶質の逆拡散が過度に多いときには、凍結乾燥の処理時間が長くなると推察される。
一方、この誘導溶質sの逆拡散速度が0.001g/(m2×hr)以上であれば、原料液中の溶質が、正浸透膜oの膜表面にファウリングし難くなり、正浸透膜oが目詰まりすることなく、高濃度の濃縮原料液cを得ることができる。そして、このことにより、原料液a中の溶質の損失を抑制しつつ、溶質を回収することができ、かつ、第二のユニットにおける凍結乾燥処理の時間を短縮することが可能となる。
以上のことから、効率的な濃縮を行う観点から、誘導溶質sの逆拡散速度は、0.001g/(m2×hr)以上50g/(m2×hr)以下が好ましい。

ここで、RFは誘導溶液dについての透過流束(g/(m2×hr))であり、gは透過した誘導溶質の量(g)であり、Mは正浸透膜oの表面積(m2)であり、Hは時間(hr)である。
原料液側空間Rへ移動した誘導溶質の量(g)は、Thermo Fishier Scientific社製のICP-MS、形式「iCAP Q」を用いて測定した。数式(3)より運転により移動した溶質の透過流束を計算した。
正浸透膜ユニットの原料液側空間Rには濃縮対象物である原料液aが導入され、誘導溶液側空間Dには誘導溶液dが導入される。これらの流れの方向は、向流でも並流でもよい。
正浸透膜ユニットの原料液側空間Rに導入される原料液aの流量は任意であるが、典型的な例として、正浸透膜ユニット中の正浸透膜oの表面積1m2当たり、1分間当たり、50mL/(m2・分)以上20,000mL/(m2・分)以下の範囲を例示することができ、100mL/(m2・分)以上15,000mL/(m2・分)以下とすることが好ましい。
第一のユニットにおいて、原料液側空間R(中空糸状の正浸透膜の内部)に原料液aを流通させるとき、原料液側空間Rに、10kPa以上200kPa以下の圧力をかけてもよい。この加圧により、正浸透膜oの、溶媒bについての透過流束を高くすることができ、好ましい。
第一の工程において、正浸透膜ユニットの原料液側空間Rに導入される原料液aの温度は、好ましくは3℃以上60℃以下であり、より好ましくは5℃以上50℃以下である。理由は定かではないが、原料液aの温度が3℃以上では透過流速が遅くなることを回避し易くなりでき、60℃以下では原料液a中の成分の一部が変性することを回避し易くなる。
正浸透膜ユニットの誘導溶液側空間Dに導入される誘導溶液dの温度は、好ましくは5℃以上60℃以下であり、より好ましくは10℃以上50℃以下である。理由は定かではないが、誘導溶液dの温度が5℃以上又は60℃以下のときは、正浸透膜oを介して誘導溶液dから原料液aへ誘導溶質が移動する量が多くなることを回避し易くなる。
本実施形態の原料液濃縮システムにおける第二のユニットでは、第一のユニットで得られた濃縮原料液cを凍結乾燥させ、更に濃縮された生成物fを得る。
本実施形態の原料液濃縮システムにおける凍結乾燥には、公知であるいずれの技術を使用してもよく、その典型的なプロセスに関しては前述したとおりである。
本実施形態の原料液濃縮システムにおいて任意に採用される誘導溶液再生ユニットは、以下のいずれかであってよい。
(1)希釈誘導溶液eから溶媒bを除去して、希釈誘導溶液eの濃縮物である再生誘導溶液gを得て、得られた再生誘導溶液gを誘導溶液dとして使用するためのユニット(第一の誘導溶液再生ユニット、例えば図3の場合)、又は
(2)誘導溶液dから溶媒bを除去して、誘導溶液dの濃縮物である濃縮誘導溶液hを得て、得られた濃縮誘導溶液hと希釈誘導溶液eとを混合して混合物(再生誘導溶液g)を得て、得られた再生誘導溶液gを誘導溶液dとして使用するためのユニット(第二の誘導溶液再生ユニット、例えば図4の場合)。
蒸留プロセスとは、希釈誘導溶液e又は誘導溶液dを所定の温度に調整した後、蒸留塔に送入し、塔頂部から溶媒bを得るとともに、塔底部からは、溶媒bが除去されて濃縮された希釈誘導溶液である再生誘導溶液g、又は溶媒bが除去されて濃縮された誘導溶液である濃縮誘導溶液hを得るプロセスである。
膜蒸留プロセスとは、撥水性多孔質膜によって液相部と気相部とに分割された分離室を有する膜ユニットを用いる方法である。この場合、膜蒸留用の膜ユニットの液相部に希釈誘導溶液e又は誘導溶液dを導入し、気相部を減圧とすることにより、希釈誘導溶液e又は誘導溶液dに含有される溶媒bが、液相部から撥水性多孔質膜を通過して減圧の気相部に移動する。これによって希釈誘導溶液e又は誘導溶液dから溶媒bを除去して、再生誘導溶液g又は濃縮誘導溶液hを得ることができる。
以下、膜蒸留プロセスに使用される要素について説明する。
膜蒸留プロセスに用いる撥水性多孔質膜の形状としては、例えば、中空糸膜状、平膜状、スパイラル膜状等が挙げられる。
平膜状の撥水性多孔質膜は、例えば、単一の層から構成されるものであってもよいし、支持層と、該支持層上の分離活性層とを有するものであってもよい。中空糸状の撥水性多孔質膜は、例えば、単一の層から構成される中空糸であってもよいし、中空糸状の支持層と、該支持層の外表面若しくは内表面、又はこれらの双方の面上の分離活性層とを有するものであってもよい。
撥水性多孔質膜における支持層及び分離活性層の素材は、それぞれ、第一のユニットにおける正浸透膜oについて上記に例示した素材から選択される任意のものから構成されていてよい。
この透過流束は、第一のユニットにおける正浸透膜oの、溶媒bについての透過流束と同様に定義される。
希釈誘導溶液e又は誘導溶液dは、液相部に導入される前に、20℃以上90℃以下の範囲に温度調整されていることが好ましい。この温度が20℃以上であれば、膜蒸留による溶媒bの分離の効率が損なわれる場合を回避し易くなり、90℃以下であれば、希釈誘導溶液e又は誘導溶液dに含まれる誘導溶質が、撥水性多孔質膜を通過して溶媒bへ移動する量が増大する場合を回避し易くなる。
希釈誘導溶液e又は誘導溶液dを加熱するための熱源として、例えば熱交換器を用いることができ、又は産業プロセス等の排熱を用いることができる。熱源として排熱を利用すると、溶媒bの分離のために新たに消費されるエネルギー量を削減することができるため、好ましい。
膜蒸留プロセスに用いる膜蒸留用の膜ユニットの気相部は、所定の圧力まで減圧されていることが好ましい。気相部の圧力は、装置のスケール、誘導溶液dの濃度、所望の溶媒bの生成速度等に応じて適宜に設定されてよいが、例えば、0.1kPa以上80kPa以下とすることが好ましく、1kPa以上50kPa以下とすることがより好ましい。
膜蒸留用の膜ユニットの気相部を減圧するための減圧装置としては、例えば、ダイアフラム真空ポンプ、ドライポンプ、油回転真空ポンプ、エジェクタ、アスピレーター等が挙げられる。
第一の誘導溶液再生ユニットによると、希釈誘導溶液eから溶媒bが分離されて、濃縮された希釈誘導溶液である再生誘導溶液gとなり、膜蒸留用の膜ユニットから排出される。得られた再生誘導溶液gは、必要に応じて希釈誘導溶液eと混合されて所定の濃度に調整されたうえで、誘導溶液dとして再利用することができる。再生誘導溶液gの再利用の際、冷却装置を用いて再生誘導溶液gの温度を調整してもよい。
第二の誘導溶液再生ユニットによると、誘導溶液dから溶媒bが分離されて、濃縮された誘導溶液である濃縮誘導溶液hとなり、膜蒸留用の膜ユニットから排出される。得られた濃縮誘導溶液hは、希釈誘導溶液eと混合されて所定の濃度に調整されて再生誘導溶液gとなる。再生誘導溶液gをそのまま誘導溶液dとして、又は再生誘導溶液gを誘導溶液に混合した混合物を誘導溶液dとして再利用することができる。濃縮誘導溶液hの再利用の際、冷却装置を用いて濃縮誘導溶液hの温度を調整してもよい。
上記における冷却装置としては、例えば、チラー、熱交換器等を用いることができる。
これらの誘導溶液再生ユニットによって誘導溶液dから分離された溶媒bは、必要に応じて再利用してよい。
上記のような本実施形態の原料液濃縮システムによると、第一のユニットにおいて、原料液aに含有される成分(溶質)の組成が実質的に維持されたまま、高濃度に濃縮された濃縮原料液cを、高い効率で得ることができる。更に、第二のユニットにおいて、濃縮原料液cの凍結乾燥が行われ、更に濃縮された生成物fが得られる。
ここで、第一のユニットで得られる濃縮原料液fにおいて、原料液aに含有される成分の組成が維持される程度が高いほど、第二のユニットで得られる生成物fにおける、原料液の含有成分(溶質)の回収率が高くなる。
得られた生成物f中の溶質の分析は、原料液a及び濃縮原料液cに含まれる成分の種類に応じて、適宜に選択されてよい。例えば、重量法、ICP-MS(誘導結合高周波プラズマ質量分析)、核磁気共鳴分光(NMR)法、ガスクロマトグラフィー質量分析(GC/MS)法、比色法、蛍光法、高速液体クロマトグラフ(HPLC)等の各種の公知の分析方法を用いることができる。
一方、生成物fにおける溶質の回収率を過度に高くしようとすると、かえって過分のコストを要することとなる。この観点から、本実施形態の原料液濃縮システムでは、生成物fにおける溶質の回収率は、99.9%以下に留めてもよい。
溶媒及び溶質を含有する原料液と、誘導溶質を含有する誘導溶液とを、正浸透膜を介して接触させ、前記原料液中の前記溶媒を前記誘導溶液中に移動させるとともに前記誘導溶液中の前記誘導溶質を前記原料液中に移動させることにより、濃縮原料液及び希釈誘導溶液を得る、第一の工程と、
前記濃縮原料液を凍結乾燥処理して更に濃縮された生成物を得る、第二の工程と
を有する、
原料液濃縮方法である。
上述したとおり、本明細書では、第一のユニットで行われる工程を「第一の工程」と称し、本実施形態の第二のユニットで行われる工程を「第二の工程」と称している。したがって、当業者は、本実施形態の原料液濃縮システムの第一のユニット及び第二のユニット、並びに誘導溶液再生ユニットについての上記の説明において、「ユニット」を「工程」に読み替えることにより、本実施形態の原料液濃縮方法の詳細を理解することができる。
以下の実施例及び比較例において、各物性は以下の方法により測定した。
溶媒についての正浸透膜の初期透過流速(第一の工程開始時における、溶媒についての正浸透膜の初期透過流速)は、以下の方法により測定した。
運転開始から1分間に、原料液aから誘導溶液へ透過した溶媒bの量(L)を、エー・アンド・デイ社製電子天秤(GX-12K)にて測定した。測定値を数式(2)に代入し、運転によって移動した溶媒の初期透過流束を計算した。計算結果を表1に示す。
誘導溶液中の誘導溶質を原料液中へ移動させる、誘導溶質の逆拡散速度は、以下の方法により測定した。
運転終了後、得られた濃縮原料液に含まれる誘導溶質の量を、Thermo Fishier Scientific社製のICP-MS、形式「iCAP Q」を用いて測定した。測定値を数式(3)に代入し、運転によって移動した誘導溶質の透過流束を計算し、得られた値を誘導溶質の逆拡散速度とした。
各実施例及び比較例で得られた複合中空糸膜モジュールを分解し、モジュールの半径方向の中心、半径の50%の位置、及び最外周部の3箇所から、中空糸をそれぞれ1本ずつサンプリングした。各中空糸を長さ方向に3等分し、9つのサンプルを得た。これらの中空糸サンプルのそれぞれを凍結割断して、中空糸断面サンプルを作成した。
ここで、凍結割断によるサンプル作製は以下のようにして行った。
上記断面サンプルのそれぞれについて、走査型電子顕微鏡観察を行った。該走査型電子顕微観察は、日立製作所製、形式S-4800を使用し、加速電圧1.0kV、WD5mm基準±0.7mm、及びエミッション電流設定10±1μAの条件で行った。顕微鏡像をプリンターで用紙に印刷して分離活性層部分を切り取り、その質量を精密天秤で測定した。この質量を、予め作成したおいた検量線により、分離活性層の厚み(μm)に換算した。そして、9つのサンプルの平均値を分離活性層の平均厚みとし、変動係数を算出した。
≪原料液濃縮システムの作製≫
<正浸透膜oを有する正浸透膜ユニットの作製>
(1)中空糸状支持膜モジュールの作製
ポリエーテルスルホン(PES:BASF社製、商品名「Ultrason」)をN-メチル-2-ピロリドン(和光純薬(株)製)に溶解して、20質量%の中空糸紡糸原液を調製した。二重紡口を装備した湿式中空糸紡糸機に上記の原液を充填し、水を満たした凝固槽中に押し出し、相分離により中空糸を形成した。得られた中空糸は、巻き取り機に巻き取った。得られた中空糸の外径は1.0mm、内径は0.7mm、内表面の微細孔の径は0.05μm、透水性能は1,020kg/(m2×hr)/100kPaであった。
この中空糸を支持層として用いた。
上記中空糸状の支持層130本を束ねた糸束を、2.2cm径、9.2cm長の円筒状プラスチックハウジングに充填し、両端部を接着剤で固定することにより、有効膜内表面積0.023m2の中空糸状支持層のモジュールを作製した。
10L容器に、m-フェニレンジアミン100g及びラウリル硫酸ナトリウム8gを入れ、さらに純水4,892gを加えて溶解し、界面重合に用いる第1溶液を5kg調製した。
別の10L容器に、トリメシン酸クロリド8gを入れ、n-ヘキサン3,992gを加えて溶解し、界面重合に用いる第2溶液4kgを調製した。
上記で製造した中空糸状支持層のモジュールのコア側(中空糸の内側)に第1溶液を充填し、30分静置した後に液を抜いて、中空糸の内側に第1溶液の薄い液膜を形成した。この状態で、第2溶液をコア側に100mL/分の流量で3分間送液し、界面重合を行った。重合温度は25℃とした。
次いで、モジュールのコア側に50℃の窒素を30分流し、窒素パージによってn-ヘキサンを蒸散除去し、さらに、モジュールのコア側に85℃の熱水を30分間流して、熱水洗浄を行った。
その後、モジュールをオートクレーブ((株)トミー精工製、型名「ES-315」)中に入れ、モジュールのコア側に121℃の高温水蒸気を20分間供給して、高温水蒸気処理を行った。さらに、モジュールのコア側及びシェル側の双方を、20℃の水による30分間の水洗処理を行うことにより、中空糸支持層の内面にポリアミド(PA)から成る分離活性層を有する中空糸状正浸透膜oのモジュールである、正浸透膜ユニットを作製した。
凍結乾燥チャンバーとしては、100mLのナスフラスコを用いた。
(膜蒸留ユニットの作製)
誘導溶液再生工程における希釈誘導溶液濃縮手段としては、以下のように作製した膜蒸留ユニットを使用した。
平均一次粒径0.016μm、比表面積110m2/gの疎水性シリカ(日本アエロジル(株)製、AEROSIL-R972)23質量部とフタル酸時オクチル(DOP)31質量部とフタル酸時ブチル(DBP)6質量部とをヘンシェルミキサーで混合し、これに重量平均分子量が310,000のポリフッ化ビニリデン(SOLVAY社製、Solef(登録商標)6010)40質量部を添加し、再度ヘンシェルミキサーで混合した。この混合物を2軸混練押し出し機で混合し、ペレット化した。
得られたペレットを、2軸混練押し出し機により240℃にて溶融混練し、中空糸状に押出して中空繊維を得た。このとき、押出機先端のヘッド内の押出口に、中空糸成形用紡口を装着し、その押出面にある溶融物押出用円環穴から上記溶融物を押し出して押出物を得ると同時に、溶融物押出用円環穴の内側にある中空部形成流体吐出用の円形穴から窒素ガスを吐出させ、押出物の中空部内に流通させることにより、中空糸状の押出物を得た。得られた中空糸状押出物は、空走距離20cmにて水浴(40℃)中に導入し、20m/分の速度で巻き取った。
実施例1では、L-アラニル-L-グルタミン水溶液の濃縮を行った。
原料液aとしてのL-アラニル-L-グルタミン水溶液は、以下のように調製した。
市販のL-アラニル-L-グルタミン(白色粉末状態、ナカライテスク(株)製)10gを、25℃のイオン交換水/アセトニトリル=85/15(体積比)の混合溶媒中に溶解させて、10g/LのL-アラニル-L-グルタミン水溶液を1L得た。
図5に示した原料液濃縮システムにおける、第一の工程の正浸透膜ユニットに、原料液a(L-アラニル-L-グルタミン水溶液)を流速130mL/分で、誘導溶液dを流速は150mL/分で、それぞれ流した。このとき、原料液aの温度が25℃に保たれ、クロスフロー方式でろ過が行われるように設定した。
誘導溶液dとしては、誘導物質として塩化マグネシウム20質量%を含有する水溶液を使用した。
図5に示した原料液濃縮システムにおける第二の工程では、以下の操作によって、凍結乾燥処理を行った。
4個の凍結乾燥チャンバー(100mLのナスフラスコ)中に、それぞれ、第一の工程で得られた濃縮原料液cを、25gずつ量り取り、液体窒素で完全に凍結させた。
凍結後の濃縮原料液cを含む凍結チャンバー4個を、多岐管を用いて凍結乾燥機に接続して、凍結乾燥を行った。凍結乾燥は、下記の条件にて、24hr実施した。
使用装置:東京理化器械(株)製、EYELA凍結乾燥機FDU-2110型
予備凍結方法:液体窒素で凍結
真空度:約10~20Pa
誘導溶液の誘導濃度を一定に保つための、誘導溶液再生工程には、上記で作製した膜蒸留ユニットを用いた。
誘導溶液再生工程の膜蒸留ユニットに、誘導溶液dを流速100cm3/分にて流し、膜蒸留ユニットの気相部の圧力が絶対圧で10kPaになるように真空ポンプで調節して、膜蒸留を行い、濃縮誘導溶液hを得た。
第一の工程で得られた希釈誘導溶液eと、膜蒸留で得られた濃縮誘導溶液hとを、バッファタンク中で混合して誘導溶液dを調製(再生)し、再生誘導溶液gを得て、第一の工程で循環使用した。
得られた生成物fについて、以下の評価を行った。
(4-1)第二の工程(凍結乾燥)における溶媒除去率
得られた生成物fに含まれる水分量、及びアセトニトリル量を、それぞれ、カールフィッシャー水分測定システム(メトローム社製、型名「MATi10」)及びGC/MSシステム(日本電子(株)製、型名「JMS-Q1000GC K9」)により、測定した。
先ず、GC/MSの測定を行った。その結果、生成物f中にアセトニトリルの残留がないことを確認した。
次に、カールフィッシャー測定により、生成物f中の水分量の測定を行った。
さらに、第一の工程前の原料液aの濃度、及び第一の工程前後の原料液の減容率より、第一の工程後の濃縮原料液cに含まれる溶媒量を算出した。
そして、数式{(濃縮原料液cに含まれる溶媒量-生成物fに含まれる水分量)/濃縮原料液cに含まれる溶媒量}×100により、第二の工程(凍結乾燥工程)における溶媒除去率を算出し、以下の基準で評価した。結果を表1に示す。
A:溶媒除去率が100%であった場合
B:溶媒除去率が95%以上100%未満であった場合
C:溶媒除去率が90%以上95%未満であった場合
D:溶媒除去率が90%未満であった場合
第一の工程前の原料液aの濃度、及び第一の工程前後の原料液の減容率より、原料液中の溶質回収率を100%と仮定した場合の、濃縮後原料液cに含まれる理論溶質量を算出した。
次に、第二の工程で得られた凍結乾燥後の生成物fの質量を、天秤で測定した。
そして、数式{(生成物fの質量-生成物f中の水分量)/理論溶質量}×100により、第一の工程及び第二の工程を経た後の溶質回収率を算出し、以下の基準で評価した。結果を表1に示す。
A:溶質回収率が95%以上であった場合
B:溶質回収率が90%以上95%未満であった場合
C:溶質回収率が80%以上90%未満であった場合
D:溶質回収率が80%未満であった場合
原料液aに含まれる溶質の種類、誘導溶液dに含まれる誘導溶質の種類及び濃度、並びに溶媒bの種類を、それぞれ、表1に記載のとおりに変更した以外は、実施例1と同様にして原料液aの濃縮を行い、評価を実施した。結果を表1に示す。
表1中、原料液aの溶媒欄、及び誘導溶液ランの「aq.」は水溶液を表す。
なお、実施例4~7では、正浸透膜ユニットのコア側からシェル側(中空糸状正浸透膜の内側から外側)へ、それぞれ、表1に記載の圧力をかけて、第一の工程を行った。
実施例20における、茶抽出液の凍結乾燥粉末を含む原料液aは、以下のように調製した。
容量3.0LのSUS304製密閉容器に、茶抽出液の凍結乾燥物の粉末10gを入れ、蒸留水1Lを加えて、30分撹拌して得られた溶液を、原料液aとして用いた。
「(2)正浸透膜ユニットの作製」を、以下のように行った以外は、実施例1と同様にして、正浸透膜モジュールを作製し、原料液aの濃縮を行い、評価を実施した。結果を表1に示す。
0.5LL容器に、m-フェニレンジアミン10g及びラウリル硫酸ナトリウム0.8gを入れ、さらに純水489.2gを加えて溶解し、界面重合に用いる第1溶液を0.5kg調製した。
別の0.5L容器に、トリメシン酸クロリド0.8gを入れ、n-ヘキサン399.2gを加えて溶解し、界面重合に用いる第2溶液0.4kgを調製した。
実施例1と同様にして作製した中空糸状支持膜モジュールのコア側(中空糸の内側)に第1溶液を充填し、30分静置した後に液を抜いて、中空糸の内側に第1溶液の薄い液膜を形成した。
次に、コア側圧力を常圧に設定し、シェル側圧力を絶対圧として10kPaの減圧に設定した。この状態で30分静置した後、この圧力を維持したまま、第2溶液をコア側に1.5L/分の流量で3分間送液し、界面重合を行った。重合温度は25℃とした。
その後、実施例1と同様に、50℃の窒素による窒素パージ、85℃の熱水による熱水洗浄、121℃の高温水蒸気処理による高温水蒸気処理、及び20℃の水による水洗処理を行った。
さらに、モジュールのシェル側(中空糸外側)を100KPaの圧力にて加圧する加圧処理を行っ後、シェル側及びコア側の双方を純水によって洗浄することにより、中空糸支持層の内面にポリアミドから成る分離活性層を有する中空糸状正浸透膜oのモジュールである正浸透膜ユニットを作製した。
「(2)正浸透膜ユニットの作製」において、窒素パージ後の熱水洗浄の熱水温度を90℃とし、高温水蒸気処理の高温水容器の温度を131℃とし、シェル側の加圧処理を行わなかった他は、実施例9と同様にして正浸透膜モジュールを作製した。
得られた正浸透膜モジュールを用いて、実施例2と同様にして、原料液aの濃縮を行い、評価を実施した。結果を表1に示す。
「(2)正浸透膜ユニットの作製」を以下のように行った以外は、実施例1と同様にして、正浸透膜モジュールを作製し、原料液aの濃縮を行い、評価を実施した。結果を表1に示す。
実施例1と同様にして、中空糸状支持層のモジュールのコア側(中空糸の内側)にて、ポリアミドの界面重合を行った。
次いで、モジュールのコア側に50℃の窒素を30分流してn-ヘキサンを蒸散除去した後、シェル側及びコア側の双方を純水にて洗浄した。
さらに、モジュールの内部(コア側及びシェル側双方)に、45℃の酢酸水溶液(pH=2)を流入させて、界面重合で得られたポリアミドを1時間酢酸水溶液に浸漬させる、酢酸処理を行った。1時間経過後、モジュールから酢酸水溶液を取り除き、洗することにより、中空糸支持層の内面に、酢酸水溶液に浸漬させたポリアミドから成る分離活性層を有する中空糸状正浸透膜oのモジュールである、正浸透膜ユニットを作製した。
「(2)正浸透膜ユニットの作製」において、窒素パージ後の熱水洗浄の熱水温度を100℃とし、高温水蒸気処理の高温水容器の温度を141℃とし、シェル側の加圧処理を行わなかった以外は、実施例9と同様にして正浸透膜モジュールを作製した。
得られた正浸透膜モジュールを用いて、実施例2と同様にして、原料液aの濃縮を行い、評価を実施した。結果を表1に示す。
「(2)正浸透膜ユニットの作製」において、酢酸処理に用いた酢酸水溶液(pH=2)の温度を60℃とした以外は、実施例11と同様にして正浸透膜モジュールを作製した。
得られた正浸透膜モジュールを用いて、実施例1と同様にして、原料液aの濃縮を行い、評価を実施した。結果を表1に示す。
「(1)中空糸状支持膜モジュールの作製」において、中空糸状支持膜として、ポリスルホン(PS)から成る中空糸を用いた以外は、実施例1と同様にして、中空糸状支持膜モジュールの作製し、これを用いて正浸透膜ユニットを作製した。
得られた正浸透膜モジュールを用いて、実施例2と様にして、原料液aの濃縮を行い、評価を実施した。結果を表1に示す。
ポリスルホンから成る中空糸を含む、中空糸状支持膜モジュールは、以下のように作製した。
ポリスルホン(ソルベイ社製、P-3500)をN-メチル-2-ピロリドン(和光純薬(株)製)に溶解して、ポリマー濃度19質量%の中空糸紡糸原液を調製した。二重紡口を装備した湿式中空糸紡糸機に上記原液を充填し、水を満たした凝固槽に押し出して、相分離により中空糸を形成した。得られた中空糸は、巻き取り機に巻き取った。得られた中空糸の外径は1.00mm、内径は0.70mmであった。
この中空糸を中空糸状支持膜として用いた。
上記中空糸状支持膜130本を、2cm径、10cm長の円筒状プラスチックハウジングに充填し、両端部を接着剤で固定することにより、有効膜内表面積0.023m2の中空糸支持層モジュールを作製した。
「(1)中空糸状支持膜モジュールの作製」において、中空糸状支持膜として、ポリケトン(PK)から成る中空糸を用いた以外は、実施例1と同様にして、中空糸状支持膜モジュールの作製し、これを用いて正浸透膜ユニットを作製した。
得られた正浸透膜モジュールを用いて、実施例2と同様にして、原料液aの濃縮を行い、評価を実施した。結果を表1に示す。
ポリケトンから成る中空糸を含む、中空糸状支持膜モジュールは、以下のように作製した。
エチレンと一酸化炭素とが完全交互共重合した、極限粘度3.4dl/gのポリケトンを、65質量%レゾルシン水溶液に添加し、80℃で2時間攪拌溶解し、脱泡を行って、ポリマー濃度10.7質量%の均一透明な中空糸紡糸原液を得た。
二重紡口を装備した湿式中空糸紡糸機に上記の原液を充填した。水を満たした凝固槽中に、50℃に調温された原液を押し出して、相分離により中空糸を形成した。得られた中空糸は、巻き取り機に巻き取った。得られた中空糸の外径は1.0mm、内径は0.7mm、内表面の微細孔の径は0.15μmであった。
この中空糸を中空糸状支持層として用いた。
上記中空糸状支持膜130本を、2cm径、10cm長の円筒状プラスチックハウジングに充填し、両端部を接着剤で固定することにより、有効膜内表面積0.023m2の中空糸支持層モジュールを作製した。
第一の工程の正浸透膜ユニット、及び第二の工程の凍結乾燥ユニットとしては、それぞれ、実施例1におけるのと同様のものを用いた。
そして、誘導溶液として2-プロパノールを用い、誘導溶液再生工程を行わなかった他は、実施例1と同様にして、原料液aの濃縮を行い、評価を実施した。結果を表1に示す。
誘導溶液dに含まれる誘導溶質の種類及び濃度、並びに溶媒bの種類を、それぞれ、表1に記載のとおりに変更した以外は、実施例23と同様にして原料液aの濃縮を行い、評価を実施した。結果を表1に示す。
そのため、「(4-1)第二の工程(凍結乾燥)における溶媒除去率」では、実施例1と同様に、生成物f中の水分量に基づいて、して、溶媒除去率を算出した。
実施例33では、図2に示した構成の原料液濃縮システムを使用して実施した。
正浸透膜ユニットとしては、有効膜面積が1.65m2である中空糸支持膜モジュールを用いて作製された正浸透膜モジュールを用いた。
希釈誘導溶液濃縮手段としては、有効膜面積が7.5m2の膜蒸留ユニットを用いた。
凍結乾燥チャンバーは、実施例1と同様のものを用いた。
正浸透膜モジュール及び膜蒸留ユニットは、それぞれ、以下のように作製した。
(1)中空糸状支持膜モジュールの作製
実施例1と同様にして得た中空糸を、支持膜として用いた。
この中空糸状支持膜1,750本を束ねた糸束を、5.5cm径、50cm長の円筒状プラスチックハウジングに充填し、両端部を接着剤で固定することにより、有効膜内表面積1.65m2の中空糸状支持膜モジュールを作製した。
上記で得られた中空糸状支持膜モジュールを用い、中空糸の内側に第1溶液の薄い液膜を形成した後に、第2溶液をコア側に送液するときの流量を1.75L/分とした他は、実施例1と同様にして、中空糸支持層の内面にポリアミドから成る分離活性層を有する中空糸状正浸透膜oのモジュールである正浸透膜ユニットを作製した。
(膜蒸留ユニットの作製)
実施例1と同様にして外径1.25mm、内径0.68mm、内表面の微細孔の径0.1μmの中空糸から成る半透膜4,700本を、13cm径、75cm長の円筒状プラスチックハウジングに充填し、両端部を接着剤で固定することによ
第一の工程は、上記で作製した正浸透膜ユニットへの原料液aの供給流速を2L/分とし、誘導溶液dの供給流速を7L/分とした他は、実施例1と同様にして行った。
第二の工程には、実施例1と同様の手法を用いた。
誘導溶液再生工程では、上記で作製した膜蒸留ユニットを用い、希釈誘導溶液eの供給流速を4L/分とした他は、実施例1と同様にして行った。
実施例1と同様の方法で行った評価の結果を、表1に示す。
第一の工程の代わりに限外ろ過装置を用いた他は、実施例1と同様にして、原料液aの濃縮を行い、評価を実施した。
限外ろ過膜としては、Hydrosart(登録商標)/Sartocon Slice Casset(排除限界分子量:10K、膜面積:0.1m2、材質:再生セルロース膜、Sartorius AG社製、ポリエーテルスルホン(PES)製)を、膜ホルダー(Sartcon Slice Holder、Sartorius AG社製)に装着して用い、ポンプ(リキポート NE1.300、KNF社製)を用いて、線速100cm/s、運転温度25℃、膜間差圧(TMP)約0.05MPaの条件下で、クロスフロー式ろ過法による処理を行った。
第一の工程の代わりに、逆浸透膜法を用いた以外は、比較例1と同様にして、原料液aの濃縮を行い、評価を実施した。
逆浸透膜には日東電工(株)製の品番「NTR-759HR」、ポリエーテルスルホン(PES)製)を用い、線速10cm/s、運転温度25℃、3.0MPaの操作圧力にて運転した。結果を表2に示す。
第一の工程の代わりに減圧システムを組み込んだ蒸留塔を用い、70℃、10.7~13.3kPa(80~100Torr)にて減圧蒸留を行った以外は、比較例1と同様にして、原料液aの濃縮を行い、評価を実施した。結果を表2に示す。

Claims (45)
- 溶媒及び溶質を含有する原料液と、誘導溶質を含有する誘導溶液とを、正浸透膜を介して接触させ、前記原料液中の前記溶媒を前記誘導溶液中に移動させるとともに前記誘導溶液中の前記誘導溶質を前記原料液中に移動させることにより、濃縮原料液及び希釈誘導溶液を得るための、第一のユニットと、
前記濃縮原料液を凍結乾燥処理して更に濃縮された生成物を得るための、第二のユニットと
を有する、
原料液濃縮システム。 - 前記第一のユニットにおける、前記誘導溶液中の前記誘導溶質を前記原料液中へ移動させる、誘導溶質の逆拡散速度が、200g/(m2×hr)以下である、請求項1に記載の原料液濃縮システム。
- 前記第一のユニットにおける前記誘導溶液中の前記誘導溶質を前記原料液中へ移動させる、誘導溶質の逆拡散速度が、0.001g/(m2×hr)以上、50g/(m2×hr)以下である、請求項1に記載の原料液濃縮システム。
- 前記第一のユニットにおいて、前記溶媒についての前記正浸透膜の初期透過流速が0.15L/(m2×hr)以上35L/(m2×hr)以下である、請求項1~3のいずれか一項に記載の原料液濃縮システム。
- 前記原料液に含まれる前記溶媒が、
水と、
アセトニトリル、メタノール、及び2-プロパノールから成る群から選択される1種又は2種以上と
を含有する、
請求項1~4のいずれか一項に記載の原料液濃縮システム。 - 前記溶媒が、酸性成分をさらに含み、かつ、
前記溶媒のpHが1~4である、

- 前記原料液の溶質が、糖、アミノ酸、オリゴペプチド、酵素、及び核酸から成る群から選択される1種又は2種以上を含む、請求項1~7のいずれか一項に記載の原料液濃縮システム。
- 前記溶質が、数平均分子量が100~6,000の化合物を含む、請求項1~7のいずれか一項に記載の原料液濃縮システム。
- 前記第一のユニットにおける、前記原料液と前記誘導溶液との前記正浸透膜を介する接触が、クロスフロー方式の接触である、請求項1~9のいずれか一項に記載の原料液濃縮システム。
- 前記第一のユニットにおける、前記原料液の温度が5℃以上50℃以下である、請求項1~10のいずれか一項に記載の原料液濃縮システム。
- 前記希釈誘導溶液から前記溶媒を除去して再生誘導溶液を得て、得られた前記再生誘導溶液を前記誘導溶液として使用するための、第一の誘導溶液再生システムをさらに有する、請求項1~11のいずれか一項に記載の原料液濃縮システム。
- 前記第一の誘導溶液再生システムにおける、前記希釈誘導溶液からの前記溶媒の除去が、前記希釈誘導溶液から前記溶媒を蒸発させることである、請求項12に記載の原料液濃縮システム。
- 前記誘導溶液から前記溶媒を除去して濃縮誘導溶液を得て、得られた前記濃縮誘導溶液と前記希釈誘導溶液とを混合して混合物を得て、得られた前記混合物を前記誘導溶液として使用するための、第二の誘導溶液再生システムをさらに有する、請求項1~13のいずれか一項に記載の原料液濃縮システム。
- 前記第二の誘導溶液再生システムにおける、前記誘導溶液からの前記溶媒の除去が、前記誘導溶液から前記溶媒を蒸発させることである、請求項14に記載の原料液濃縮システム。
- 前記誘導溶液が、前記誘導溶質としての無機塩を含む溶液である、請求項1~14のいずれか一項に記載の原料液濃縮システム。
- 前記誘導溶液が、前記誘導溶質としてのアルコールを含む溶液である、請求項1~16のいずれか一項に記載の原料液濃縮システム。
- 前記アルコールが、エタノール、及び2-プロパノールから選択される1種又は2種を含む、請求項17に記載の原料液濃縮システム。
- 前記正浸透膜が、ポリエーテルスルホン、ポリスルホン、ポリケトン、ポリエーテルエーテルケトン、ポリフェニレンエーテル、ポリフッ化ビニリデン、ポリアクリロニトリル、ポリイミン、ポリイミド、ポリベンゾオキサゾール、ポリベンゾイミダゾール、パーフルオロスルホン酸重合体、及びポリアミドから成る群から選ばれる、1種又は2種以上を主成分とする薄膜層を有する膜である、請求項1~18のいずれか一項に記載の原料液濃縮システム。
- 前記正浸透膜が中空糸状の膜である、請求項1~19のいずれか一項に記載の原料液濃縮システム。
- 前記第一のユニットが、複数の中空糸状の前記正浸透膜によって構成される中空糸糸束を有する膜モジュールの形態にあって、
前記中空糸状の正浸透膜が、微細孔性支持膜と、前記微細孔性支持膜の内表面に設けられた、高分子重合体薄膜の分離活性層とを備え、
前記中空糸糸束の膜面積の合計が0.01m2以上であり、
そして前記分離活性層の厚み方向の断面を撮影した走査型電子顕微鏡画像における分離活性層部分の質量を測定する方法により算出された、前記中空糸糸束の半径方向及び長さ方向における前記分離活性層の厚みの変動係数が0~60%である、請求項20記載の原料液濃縮システム。 - 前記第一のユニットで得られた前記濃縮原料液を、前記第一のユニットにおける前記原料液として使用する循環機構を有する、請求項1~21のいずれか一項に記載の原料液濃縮システム。
- 溶媒及び溶質を含有する原料液と、誘導溶質を含有する誘導溶液とを、正浸透膜を介して接触させ、前記原料液中の前記溶媒を前記誘導溶液中に移動させるとともに前記誘導溶液中の前記誘導溶質を前記原料液中に移動させることにより、濃縮原料液及び希釈誘導溶液を得る、第一の工程と、
前記濃縮原料液を凍結乾燥処理して更に濃縮された生成物を得る、第二の工程と
を有する、

- 前記希釈誘導溶液から前記溶媒を除去して再生誘導溶液を得て、得られた前記再生誘導溶液を前記誘導溶液として使用するための、第一の誘導溶液再生工程をさら有する、請求項23~33のいずれか一項に記載の原料液濃縮方法。
- 前記第一の誘導溶液再生工程における、前記希釈誘導溶液からの前記溶媒の除去が、前記希釈誘導溶液から前記溶媒を蒸発させることによって行われる、請求項34に記載の原料液濃縮方法。
- 前記誘導溶液から前記溶媒を除去して濃縮誘導溶液を得て、得られた前記濃縮誘導溶液と前記希釈誘導溶液とを混合して混合物を得て、得られた前記混合物を前記誘導溶液として使用するための、第二の誘導溶液再生工程をさらに有する、請求項23~35のいずれか一項に記載の原料液濃縮方法。

- 前記第一の工程が、複数の中空糸状の前記正浸透膜によって構成される中空糸糸束を有する膜モジュールの形態にある、第一のユニットを用いて行われ、
