곰보배추 추출물의 인간 각질세포(human keratinocyte) HaCaT 세포독성을 확인하기 위하여, CCK-8(Cell Counting Kit-8) 어세이를 실시하였다. 96웰 플레이트에서 HaCaT 세포에 시료를 농도별(25, 50, 100, 200㎍/㎖)로 처리하고 24시간 동안 배양하였다. CCK-8을 10% 농도로 첨가한 다음 2시간 동안 37℃에서 반응시킨 후, 마이크로플레이트 분광광도계(Biorad, Hercules, CA, USA)를 이용하여 450nm에서 흡광도를 측정하여 세포 생존율(%)을 계산하였다.
