Abstract

본 발명은 CRISPR/Cpf1 시스템을 이용한 유전체 편집용 조성물 및 이의 용도에 관한 것으로서, 보다 상세하게 본 발명은 표적 뉴클레오티드(target nucleotide) 서열과 혼성화 가능한 가이드 서열(guide sequence) 및 상기 가이드 서열의 3'-말단에 연결된 유리딘(U, uridine) 반복 서열을 포함하는 crRNA(CRISPR RNA) 또는 이를 암호화하는 DNA; 및 Cpf1 단백질 또는 이를 암호화하는 DNA을 포함하는 유전체 편집용 조성물, 이를 이용한 유전체 편집 방법, 형질 전환체 제조 방법 및 형질 전환체에 관한 것이다. 본 발명은 CRISPR/Cpf1 시스템을 이용하여 진핵 세포의 유전제 교정을 수행함에 있어서 인델 효율을 높이고 오프-타겟 활성을 줄일 수 있고, 원하는 유전자가 삽입(knock-in) 또는 결실(knock-out)된 형질 전환 세포, 형질 전환 동물 또는 형질 전환 식물을 용이하게 제조할 수 있다.
