Abstract
ジカウイルスNS1タンパク質の260-352番目のアミノ酸配列からなるペプチド断片に結合するモノクローナル抗体及び1-176番目のアミノ酸配列からなるペプチド断片に結合するモノクローナル抗体を用いることにより、ジカウイルスを特異的かつ簡便・迅速に、高感度で検出し得ることを見出した。
Description
本発明は、ジカウイルスNS1タンパク質に特異的な抗体を利用したジカウイルスの免疫学的検出法、及び、当該免疫学的検出を利用してジカウイルス感染症の診断を補助する方法に関する。また、本発明は、これら方法に用いられるキットに関する。
ジカウイルスはフラビウイルス科に属する1本鎖RNAの球形ウイルスであり、2つの遺伝子型(アフリカ型とアジア型)が存在する。ヒトは、ネッタイシマカやヒトスジシマカといった媒介蚊に刺されることにより感染する。臨床的特徴は、2~12日間の潜伏期の後に、全身の不快感、倦怠感といった前駆的症状にはじまり、突然の発熱、頭痛、全身の筋肉痛などが出現する。時にギランバレー症候群を引き起こす可能性や、妊婦への感染では小頭症児の発生が指摘されている。このため、ジカウイルスの流行地域からの入国者や帰国者については、本症の感染の疑いがある場合には、ジカウイルス感染の診断を行う必要がある。
ジカウイルスの診断においては、遺伝子増幅法(RT-PCR)によるウイルス遺伝子の検査やウイルスの分離が行われているが、前者は、特別な設備が必要であり、後者は、多くの時間と手間を要する。
一方、血清中の抗体(IgMやIgG)を検出する血清学的検査も行われているが、フラビウイルス科に属する他のウイルスとの交差反応性(偽陽性)の問題が指摘されている。最近になり、ジカウイルスNS1タンパク質を利用することで、交差反応性の問題を低減させることが可能であることが報告された(非特許文献1)。
しかしながら、血清学的検査は、直接ジカウイルスを検出するものではなく、例えば、ジカウイルスに対する抗体が生じていない感染初期には適用できないという本質的問題を避けることができない。
なお、ジカウイルスと同じフラビウイルス科に属するデング熱ウイルスについては、そのNS1タンパク質に対する抗体を利用した検出法が報告されている(特許文献1)。

本発明は、このような状況に鑑みてなされたものであり、その目的は、ジカウイルスを特異的かつ簡便・迅速に、高感度で検出し得る免疫学的手法を提供することにある。また、本発明は、アジア株かアフリカ株かを問わず、広範にジカウイルスを検出し得る免疫学的手法を提供することをも目的とする。さらなる本発明の目的は、これら免疫学的手法に用いられるキットを提供することにある。
本発明者らは、前記課題を解決すべく鋭意検討を行った。まず、組換えジカウイルスNS1タンパク質を免疫原として多くのハイブリドーマを作製し、その中から、当該ジカウイルスNS1タンパク質に対して高い反応性を示すモノクローナル抗体を産生するハイブリドーマを選抜し、4つのハイブリドーマを取得することに成功した。
次いで、取得したハイブリドーマが産生するモノクローナル抗体のエピトープ解析を行ったところ、3つのモノクローナル抗体が、共通して、ジカウイルスNS1タンパク質の260-352番目のアミノ酸配列からなるペプチド断片に結合することを見出した。そのうち、2つのモノクローナル抗体は、さらに共通して、280-352番目のアミノ酸配列からなるペプチド断片に結合することを見出した。これらモノクローナル抗体は、260-310番目のアミノ酸配列からなるペプチド断片、300-352番目のアミノ酸配列からなるペプチド断片には反応しないことから、いずれもコンフォメーションエピトープを認識する抗体であることが推察された。一方、他の一つのモノクローナル抗体は、ジカウイルスNS1タンパク質の1-176番目のアミノ酸配列からなるペプチド断片に結合した。
これらモノクローナル抗体を用いて、サンドイッチELISA法及びイムノクロマトグラフィーによりジカウイルスNS1タンパク質の検出を行ったところ、いずれの手法によっても、高感度でジカウイルスを検出し得る結果を得た。特に、異なるモノクローナル抗体の組み合わせを用いた場合には、広範なジカウイルスに対して高感度での検出が認められた。さらに、280-352番目のアミノ酸配列からなるペプチド断片に結合する2つの異なるモノクローナル抗体を組合せた場合に、広範なジカウイルスに対する感度がより高くなることが認められた。
以上から、本発明者らは、ジカウイルスNS1タンパク質の上記部位を認識する抗体を利用すれば、ジカウイルスの検出やジカウイルスによる感染症の診断の補助を効率的に行うことが可能であることを見出し、本発明を完成するに至った。
すなわち、本発明は、ジカウイルスに特異的な抗体を利用した、ジカウイルスの免疫学的検出法、当該免疫学的検出を利用してジカウイルス感染症の診断を補助する方法、及び、これら方法に用いられるキットに関し、より詳しくは、以下を提供するものである。

遺伝子増幅法を利用した従来のジカウイルスの検出法では、特別な設備を要するが、本発明の方法によれば、手軽に短時間で試料中のジカウイルスを検出することが可能である。また、試料中の抗ジカウイルス抗体(IgM、IgG)を検出する従来の血清学的手法では、同じフラビウイルス科に属するデングウイルスなどとの交差反応性(偽陽性)が問題となっていたが、本発明の方法に用いられるモノクローナル抗体には、デングウイルスへの反応性はない。さらに、血清学的手法は、直接ジカウイルスの存在を検出するものではなく、例えば、抗体の産生されていない感染初期の患者の診断には不向きであるが、本発明の方法は、ジカウイルスの存在を直接検出し得るため、このような問題もない。さらに、本発明の好ましい態様においては、ジカウイルスのアジア株とアフリカ株との双方を高感度で検出することができる。従って、本発明によれば、特異的かつ簡便・迅速に、高感度で、試料中のジカウイルスを検出することが可能である。
本発明は、試料中のジカウイルスを検出する方法、及びジカウイルス感染症の診断を補助する方法を提供する。
本発明の方法は、試料中に含まれるジカウイルスを、ジカウイルスNS1タンパク質に対するモノクローナル抗体又はその組み合わせを用いたイムノアッセイにより検出する工程を含む。
本発明において「ジカウイルス」とは、フラビウイルス科に属する+鎖のRNAウイルスの1種であり、ジカウイルス感染症(ジカ熱)の原因となるウイルスを意味する。
本発明に用いられる「試料」としては、ジカウイルスが存在し得る試料である限り特に制限はない。ジカウイルス感染症の診断の補助を目的とする場合においては、一般的には、診断対象(好ましくはヒト)から採取された血液検体又は尿が用いられる。血液検体は、好ましくは血清又は血漿である。
本発明において「NS1タンパク質」とは、ジカウイルスの非構造タンパク質の1つであり、典型的には、配列番号1に記載のアミノ酸配列を有するものである。
本発明の方法に用いられる「モノクローナル抗体」は、以下の(a)又は(b)の特徴を有するもの(以下、「本発明のモノクローナル抗体」と称する)であり、ジカウイルスと同じフラビウイルス科に属するデングウイルスには反応しないため高い特異性を有する。
(a)配列番号1の260-352番目のアミノ酸配列からなるペプチド断片に結合する(以下、「特徴(a)モノクローナル抗体」と称する)。
(b)配列番号1の1-176番目のアミノ酸配列からなるペプチド断片に結合する(以下、「特徴(b)モノクローナル抗体」と称する)。
本発明の「モノクローナル抗体」は、完全な抗体のみならず、抗体断片であってもよい。抗体断片としては、例えば、F(ab’)2、Fab’、Fab、Fv、単鎖抗体、ダイアボディーなどが挙げられる
本発明の「特徴(a)モノクローナル抗体」の好ましい態様は、さらに、260-310番目のアミノ酸配列からなるペプチド断片と300-352番目のアミノ酸配列からなるペプチド断片には結合しないという特徴を有する(以下、場合により「特徴(a1)モノクローナル抗体」と称する)。このような特徴(a1)モノクローナル抗体としては、本実施例に記載の抗体A、C、及びDが挙げられる(表1)。さらに、本発明の「特徴(a1)モノクローナル抗体」のより好ましい態様は、280-352番目のアミノ酸配列からなるペプチド断片に結合し、かつ、260-310番目のアミノ酸配列からなるペプチド断片と300-352番目のアミノ酸配列からなるペプチド断片には結合しないという特徴を有する(以下、場合により「特徴(a2)モノクローナル抗体」と称する)。このような特徴(a2)モノクローナル抗体としては、本実施例に記載の抗体C及びDが挙げられる(表1)。その結合特性から、抗体A、C及びDに代表されるこれら抗体は、ジカウイルスNS1タンパク質のコンフォメーションエピトープを認識する抗体であると考えられる。
本発明の「特徴(a)モノクローナル抗体」の好ましい態様は、さらに、260-310番目のアミノ酸配列からなるペプチド断片と300-352番目のアミノ酸配列からなるペプチド断片には結合しないという特徴を有する(以下、場合により「特徴(a1)モノクローナル抗体」と称する)。このような特徴(a1)モノクローナル抗体としては、本実施例に記載の抗体A、C、及びDが挙げられる(表1)。さらに、本発明の「特徴(a1)モノクローナル抗体」のより好ましい態様は、280-352番目のアミノ酸配列からなるペプチド断片に結合し、かつ、260-310番目のアミノ酸配列からなるペプチド断片と300-352番目のアミノ酸配列からなるペプチド断片には結合しないという特徴を有する(以下、場合により「特徴(a2)モノクローナル抗体」と称する)。このような特徴(a2)モノクローナル抗体としては、本実施例に記載の抗体C及びDが挙げられる(表1)。その結合特性から、抗体A、C及びDに代表されるこれら抗体は、ジカウイルスNS1タンパク質のコンフォメーションエピトープを認識する抗体であると考えられる。
本発明の「特徴(b)モノクローナル抗体」の好ましい態様は、さらに、配列番号1の83-141番目のアミノ酸配列からなるペプチド断片及び89-264番目のアミノ酸配列からなるペプチド断片に結合するという特徴を有する。このようなモノクローナル抗体としては、本実施例に記載の抗体Bが挙げられる(表1)。
本発明のモノクローナル抗体の作製においては、例えば、本実施例に記載のように、まず、組換えジカウイルスNS1タンパク質を免疫原として多くのハイブリドーマを作製し、その中から、当該ジカウイルスNS1タンパク質に対して高い反応性を示すモノクローナル抗体を産生するハイブリドーマを選抜し、さらに、選抜したハイブリドーマが産生するモノクローナル抗体のエピトープ解析を行って、上記特徴(a)又は(b)を有するモノクローナル抗体を産生するクローンを同定すればよい。
ハイブリドーマ法としては、代表的には、ケーラー及びミルスタインの方法(Kohler&Milstein,Nature,256:495(1975))が挙げられる。この方法における細胞融合工程に使用される抗体産生細胞は、抗原(標的タンパク質、その部分ペプチド、又はこれらを発現する細胞など)で免疫された動物(例えば、マウス、ラット、ハムスター、ウサギ、サル、ヤギ、ヒツジ、ロバ、ラクダ、アルパカ、ニワトリ)の脾臓細胞、リンパ節細胞、末梢血白血球などである。免疫されていない動物から予め単離された上記の細胞又はリンパ球などに対して、抗原を培地中で作用させることによって得られた抗体産生細胞も使用することが可能である。ミエローマ細胞としては公知の種々の細胞株を使用することが可能である。抗体産生細胞及びミエローマ細胞は、それらが融合可能であれば、異なる動物種起源のものでもよいが、好ましくは、同一の動物種起源のものである。ハイブリドーマは、例えば、抗原で免疫されたマウスから得られた脾臓細胞と、マウスミエローマ細胞との間の細胞融合により産生され、その後のスクリーニングにより、抗原に特異的なモノクローナル抗体を産生するハイブリドーマを得ることができる。抗原に対するモノクローナル抗体は、ハイブリドーマを培養することにより、また、ハイブリドーマを投与した哺乳動物の腹水から、取得することができる。
また、本発明のモノクローナル抗体は、例えば、当該抗体をコードするDNAが取得できれば、組換えDNA法によって作製することができる。この方法は、上記抗体をコードするDNAをハイブリドーマやB細胞などからクローニングし、適当なベクターに組み込んで、これを宿主細胞(例えば哺乳類細胞株、大腸菌、酵母細胞、昆虫細胞、植物細胞など)に導入し、組換え抗体として産生させる手法である(例えば、P.J.Delves,Antibody Production:Essential Techniques,1997 WILEY、P.Shepherd and C.Dean Monoclonal Antibodies,2000 OXFORD UNIVERSITY PRESS、Vandamme A.M. et al.,Eur.J.Biochem.192:767-775(1990))。抗体をコードするDNAの発現においては、重鎖又は軽鎖をコードするDNAを別々に発現ベクターに組み込んで宿主細胞を形質転換してもよく、重鎖及び軽鎖をコードするDNAを単一の発現ベクターに組み込んで宿主細胞を形質転換してもよい(WO94/11523号公報参照)。組換え抗体は、上記宿主細胞を培養し、宿主細胞内又は培養液から分離・精製し、実質的に純粋で均一な形態で取得することができる。抗体の分離・精製は、通常のポリペプチドの精製で使用されている方法を使用することができる。トランスジェニック動物作製技術を用いて、抗体遺伝子が組み込まれたトランスジェニック動物(ウシ、ヤギ、ヒツジ又はブタなど)を作製すれば、そのトランスジェニック動物のミルクから、抗体遺伝子に由来するモノクローナル抗体を大量に取得することも可能である。
本発明の方法においては、本発明のモノクローナル抗体に標識物質を結合させて使用することができる。標識物質としては、抗体に結合させて検出できるものであれば特に制限はないが、例えば、アルカリホスファターゼ(ALP)、ホースラディッシュペルオキシダーゼ(HRP)、βガラクトシダーゼ(β-gal)などの酵素、フルオレセインイソチオシアネート(FITC)やローダミンイソチオシアネート(RITC)などの蛍光色素、アロフィコシアニン(APC)やフィコエリスリン(R-PE)などの蛍光蛋白質、125Iなどの放射性同位元素、金粒子、アビジン、ビオチン、ラテックスなどが挙げられる。
標識物質として酵素を用いた場合には、基質として、発色基質、蛍光基質、あるいは化学発光基質などを添加することにより、基質に応じて種々の検出を行うことができる。
標識物質を結合させた本発明のモノクローナル抗体を用いてジカウイルスNS1タンパク質を直接的に検出する方法以外に、本発明のモノクローナル抗体には標識物質を結合せず、標識物質が結合した二次抗体などを利用して間接的に検出する方法を利用することもできる。ここで「二次抗体」とは、本発明のモノクローナル抗体に対して反応性を示す抗体である。例えば、本発明のモノクローナル抗体をマウス抗体として調製した場合には、二次抗体として抗マウスIgG抗体を使用することができる。ウサギ、ヤギ、マウスなどの様々な生物種に由来する抗体に対して、使用可能な標識二次抗体が市販されており、本発明のモノクローナル抗体の由来する生物種に応じて、適切な二次抗体を選択して使用することができる。二次抗体に代えて、標識物質を結合させたプロテインGやプロテインAなどを用いることも可能である。
本発明のモノクローナル抗体と標識物質との結合には、ビオチン-アビジン系を利用することもできる。この方法においては、例えば、本発明のモノクローナル抗体をビオチン化し、これに、アビジン化した標識物質を作用させ、ビオチンとアビジンの相互作用を利用して、本発明のモノクローナル抗体に標識物質を結合させる。
本発明に用いるイムノアッセイ(免疫学的測定法)の検出原理としては、高感度な検出システムを構築することができる点で、サンドイッチ法が好適である。サンドイッチ法においては、固相化した捕捉用抗体で検出対象物質を捕捉し、それを標識物質が結合した検出用抗体に認識させ、洗浄後、標識物質の種類に応じた検出を行う。固相としては、例えば、プラスチックプレートなどのプレート、ニトロセルロースなどの繊維状物質、磁性粒子やラテックス粒子などの粒子を用いることができる。
捕捉用抗体は固相に直接固定してもよいが、間接的に固定してもよい。例えば、捕捉用抗体に結合する物質を固相に固定し、当該物質に捕捉用抗体を結合させることにより、補足用抗体を固相に間接的に固定することができる。捕捉用抗体に結合する物質としては、例えば、上記の二次抗体、プロテインG、プロテインAなどが挙げられるが、これらに制限されない。また、捕捉用抗体がビオチン化されている場合には、アビジン化した固相を利用することができる。
サンドイッチ法においては、抗原捕捉用抗体及び検出用抗体の少なくとも一方に、本発明のモノクローナル抗体を用いる。他の一方の抗体は、ジカウイルスNS1タンパク質に結合し得る限り、本発明のモノクローナル抗体以外の抗体を用いることができる。他の一方の抗体は、モノクローナル抗体であっても、ポリクローナル抗体であってもよい。
好ましい態様においては、抗原捕捉用抗体及び検出用抗体の双方が本発明のモノクローナル抗体であり、かつ、ジカウイルスNS1タンパク質に同時に結合可能な(抗原捕捉用抗体と検出用抗体とで互いに異なる)2種の本発明のモノクローナル抗体である。これにより感度及び特異性に特に優れた検出系を構築することができる。このようなジカウイルスNS1タンパク質に同時に結合可能な2種のモノクローナル抗体の組み合わせは、2種の「特徴(a)モノクローナル抗体」であっても、2種の「特徴(b)モノクローナル抗体」であってもよく、「特徴(a)モノクローナル抗体」と「特徴(b)モノクローナル抗体」との組み合わせであってもよい。
2種の「特徴(a)モノクローナル抗体」としては、例えば、本実施例に記載の抗体A、C、及びDから選択される2種のモノクローナル抗体が挙げられる。また、「特徴(a)モノクローナル抗体」と「特徴(b)モノクローナル抗体」との組み合わせとしては、本実施例に記載の抗体A、C、及びDから選択されるモノクローナル抗体と抗体Bとの組み合わせが挙げられる。
また、抗原捕捉用抗体又は検出用抗体の一方に、2種以上の本発明のモノクローナル抗体を混合したものを用いてもよく、抗原捕捉用抗体及び検出用抗体の双方に、それぞれ、2種以上の本発明のモノクローナル抗体を混合したものを用いてもよい。これにより検出感度を向上させ得る。
これらの抗原捕捉用抗体と検出用抗体との組み合わせの中でも、ジカウイルスNS1タンパク質の定量により好適であるという観点からは、抗原捕捉用抗体及び検出用抗体の双方が本発明のモノクローナル抗体であり、かつ、2種の「特徴(a)モノクローナル抗体」であるか、又は、「特徴(a)モノクローナル抗体」と「特徴(b)モノクローナル抗体」との組み合わせ(より好ましくは「特徴(a2)モノクローナル抗体」と「特徴(b)モノクローナル抗体」との組み合わせ)であることが好ましい。また、前記組み合わせが2種の「特徴(a)モノクローナル抗体」の組み合わせである場合には、互いに異なる2種の「特徴(a2)モノクローナル抗体」同士の組み合わせであることがさらに好ましい。
試料中のジカウイルスNS1タンパク質を定量する場合には、ELISA、CLEIA(化学発光酵素免疫測定法)等の、マイクロプレートのウェルやビーズ(自動化の観点から、より好ましくは磁性ビーズ)を固相とするサンドイッチ法が好ましい。得られた測定値からのジカウイルスNS1タンパク質量の特定は、一般的に、標準検体による測定値との比較により行うことができる。この場合、例えば、標準検体による測定値に基づいて作成された標準曲線上のどの位置に、実際の測定値が位置づけられるかを調べることにより、試料中のジカウイルスNS1タンパク質量を求めることができる。
一方、より簡便かつ迅速にジカウイルスを検出するには、イムノクロマトグラフィーが好適である。イムノクロマトグラフィーのデバイスの一態様について説明すると、当該デバイスは、ニトロセルロース膜のような多孔性素材からなるマトリックス上に、捕捉用抗体をライン状に固相化した検出ゾーンと、その上流側に、標識した検出用抗体を担持した標識試薬ゾーンを含む。通常、標識試薬ゾーンは、標識した検出用抗体を担持した多孔性のパッドにより構成される。マトリックスの上流端には、展開液を貯蔵した展開液槽が設けられている。さらに、通常、上記検出ゾーンの下流に、標識抗体の展開を確認するために抗標識抗体をライン状に固相化した展開確認部と、さらにその下流に、展開液を吸収するための多孔性の吸収パッドが設けられた展開液吸収ゾーンが設けられている。さらに、標識が酵素標識である場合には、標識試薬ゾーンよりも上流に、標識酵素の基質を担持した基質ゾーンが設けられる。
使用時には、検体を標識試薬ゾーンに添加し、押し込み部を加圧して突起部を移動させることにより、展開液パッドを展開液槽に挿入し、展開液パッドを通じて展開液をマトリックスに供給する。展開液が基質ゾーンを通過する際に基質が展開液中に溶出され、基質を含む展開液が流動する。展開液が標識試薬ゾーンを通過する際に、標識抗体と検体とが展開液中に溶出され、基質、標識抗体及び検体を含む展開液が流動する。検体中にジカウイルスが含まれる場合には、該ウイルスのNS1タンパク質と標識抗体とが、抗原抗体反応により結合し、これらが検出ゾーンに到達すると、検出ゾーンにおいて、固相化抗体とジカウイルスNS1タンパク質とが抗原抗体反応により結合する。その結果、ジカウイルスNS1タンパク質を介して標識抗体が検出ゾーンに固定される。こうして検出ゾーンにおける標識を測定することにより、ジカウイルスが検出されることになる。検体中にジカウイルスが含まれていない場合には、標識抗体は検出ゾーンに固定されず、さらに下流に移動するため、検出ゾーンにおいて標識は検出されない。なお、検出ゾーンの下流の展開液確認部には、抗標識抗体が固相化されているため、標識抗体は展開液確認部に固定されることになる。よって、展開液確認部に標識が検出された場合、展開液が正しく展開されたことを意味する。展開液は、最終的に、その下流の吸収パッドに吸収される。
また、本発明は、上記本発明の方法に用いるためのキットを提供する。本発明のキットは、少なくとも本発明のモノクローナル抗体又はその組み合わせを含む。
サンドイッチ法を検出原理とする方法を利用する場合には、固相化した抗原捕捉用抗体と検出用抗体の少なくとも一方の抗体が、本発明のモノクローナル抗体である。さらに、標準検体試薬(各濃度)、対照試薬、試料の希釈液、希釈用カートリッジ、洗浄液などを組み合わせることができる。酵素標識を利用した場合には、標識の検出に必要な基質や反応停止液などを含めることができる。検出用抗体を標識しない場合には、例えば、当該検出用抗体に結合する物質を標識したものをキットに含めることができる。
サンドイッチ法として、イムノクロマトグラフィーを採用する場合には、抗原捕捉用抗体が検出ゾーンに固定化された膜担体と標識された検出用抗体を担持しているパッドとを備えたデバイスをキットに含めることができる。当該デバイスは、展開液パッドや吸収パッドなど、イムノクロマトグラフィーに適したその他の構成要素を備えることができる。
本発明のキットには、さらに、当該キットの使用説明書を含めることができる。
以下、実施例に基づいて本発明をより具体的に説明するが、本発明は以下の実施例に限定されるものではない。
(実施例1)
[実施例1-1]抗ジカウイルスNS1タンパク質モノクローナル抗体の作製
免疫原としては、組換えジカウイルス(アフリカ株)NS1タンパク質(#ZIKV-NS1、The Native Antigen Company製)を1%SDSの存在下で96℃10分間加熱して変性させた、変性ジカウイルスNS1タンパク質(以下、「変性抗原」とも称する)を使用した。変性抗原をマウスに免疫し(腹腔内投与、投与量10μg/匹、免疫回数3~5回)、常法のハイブリドーマ法により、免疫原に対する抗体を産生するハイブリドーマを作製した。
[実施例1-1]抗ジカウイルスNS1タンパク質モノクローナル抗体の作製
免疫原としては、組換えジカウイルス(アフリカ株)NS1タンパク質(#ZIKV-NS1、The Native Antigen Company製)を1%SDSの存在下で96℃10分間加熱して変性させた、変性ジカウイルスNS1タンパク質(以下、「変性抗原」とも称する)を使用した。変性抗原をマウスに免疫し(腹腔内投与、投与量10μg/匹、免疫回数3~5回)、常法のハイブリドーマ法により、免疫原に対する抗体を産生するハイブリドーマを作製した。
[実施例1-2]抗体スクリーニング
抗体スクリーニングは、ELISAを用いて実施した。前記の変性抗原をPBSで希釈し、0.1~0.2μg/mLの溶液を調製した。これを96穴マイクロウェルプレートに50μLずつ分注し、37℃で1時間、又は4℃で一晩コーティングさせた。1%スキムミルク・PBS、又は2%BSA・PBSでブロッキングした後、1~10倍希釈したハイブリドーマ培養上清を各50μL/ウェル分注し、37℃で1時間反応させた。PBSTで洗浄後、POD標識抗マウスIgG抗体を50μL/ウェル分注し、37℃で30分~1時間反応させた。PBSTで洗浄後、TMB基質系で発色を行い、マイクロプレートリーダーにて450nmの吸光度を測定した。複数のハイブリドーマについて前記のスクリーニングを2回実施し、特に高い吸光度を示す4種のハイブリドーマA~Dを選出した。
抗体スクリーニングは、ELISAを用いて実施した。前記の変性抗原をPBSで希釈し、0.1~0.2μg/mLの溶液を調製した。これを96穴マイクロウェルプレートに50μLずつ分注し、37℃で1時間、又は4℃で一晩コーティングさせた。1%スキムミルク・PBS、又は2%BSA・PBSでブロッキングした後、1~10倍希釈したハイブリドーマ培養上清を各50μL/ウェル分注し、37℃で1時間反応させた。PBSTで洗浄後、POD標識抗マウスIgG抗体を50μL/ウェル分注し、37℃で30分~1時間反応させた。PBSTで洗浄後、TMB基質系で発色を行い、マイクロプレートリーダーにて450nmの吸光度を測定した。複数のハイブリドーマについて前記のスクリーニングを2回実施し、特に高い吸光度を示す4種のハイブリドーマA~Dを選出した。
[実施例1-3]モノクローナル抗体のエピトープ解析(ELISA)
ジカウイルスNS1タンパク質のリコンビナント抗原を調製し、前記4種のハイブリドーマA~Dより産生される4種のモノクローナル抗体(抗体A~抗体D)について、ELISAを用いて各ペプチドとの親和性を調査した。リコンビナント抗原は、次の方法で調製した。352アミノ酸からなるジカウイルスNS1(配列番号1、GenBank Accession No.:KU365780)の全長cDNAを人工合成し、PCR法により所望のDNA断片を増幅した。全長cDNA及び各DNA断片を公知の発現ベクターに組込んで大腸菌に導入し、発現したリコンビナント抗原をカラムにて回収・精製した。この方法により、リコンビナント抗原として、配列番号1の1-176番目、83-141番目、89-264番目、260-352番目、260-310番目、280-352番目及び300-352番目のアミノ酸配列からなるリコンビナントペプチド断片、及び配列番号1の全長からなるリコンビナントNS1タンパク質を調製した。
ジカウイルスNS1タンパク質のリコンビナント抗原を調製し、前記4種のハイブリドーマA~Dより産生される4種のモノクローナル抗体(抗体A~抗体D)について、ELISAを用いて各ペプチドとの親和性を調査した。リコンビナント抗原は、次の方法で調製した。352アミノ酸からなるジカウイルスNS1(配列番号1、GenBank Accession No.:KU365780)の全長cDNAを人工合成し、PCR法により所望のDNA断片を増幅した。全長cDNA及び各DNA断片を公知の発現ベクターに組込んで大腸菌に導入し、発現したリコンビナント抗原をカラムにて回収・精製した。この方法により、リコンビナント抗原として、配列番号1の1-176番目、83-141番目、89-264番目、260-352番目、260-310番目、280-352番目及び300-352番目のアミノ酸配列からなるリコンビナントペプチド断片、及び配列番号1の全長からなるリコンビナントNS1タンパク質を調製した。
各ペプチド断片及びリコンビナント抗原について、PBSで希釈して2μg/mLの溶液を調製し、96穴マイクロウェルプレートに50μLずつ分注した。37℃で1時間コーティングさせた後、2%BSA/PBSを150μL/ウェル加え、4℃で一晩ブロッキングした。PBSTで洗浄し、次いで、抗体A~Dを希釈液A(5%牛血清、1%BSA、0.1%カゼインナトリウム、100μg/mLマウスイムノグロブリンを含むPBS)で2.5μg/mLに調製し、各50μL/ウェル分注し、37℃で1時間反応させた。PBSTで洗浄後、アルカリホスファターゼ標識抗マウスIgG抗体を50μL/ウェル分注し、37℃で1時間反応させた。PBSTで洗浄後、AMPPD溶液(ルミパルス(登録商標)基質液(富士レビオ社製)を50μL/ウェル分注して37℃で5分間発光させ、マイクロプレートリーダーにて発光量を測定した。結果を表1に示す。表1において、リコンビナント抗原の各数字は配列番号1におけるアミノ酸番号(領域)を示す。
表1の結果より、抗体A~Dのエピトープは、配列番号1の表2に示す領域と推察された。抗体Aは、260-352番目のアミノ酸配列からなるペプチド断片には反応するが、260-310、300-352番目のアミノ酸配列からなるペプチド断片には反応しないこと、抗体C及びDは、260-352番目のアミノ酸配列からなるペプチド断片及び280-352番目のアミノ酸配列からなるペプチド断片には反応するが、260-310、300-352番目のアミノ酸配列からなるペプチド断片には反応しないことから、いずれも、コンフォメーションエピトープを認識する抗体であることが推察された。
[実施例1-4]サンドイッチELISAによるジカウイルス検出
抗体A~Dを固相化した96穴マイクロウェルプレートと、抗体A~Dをビオチン標識した標識抗体をそれぞれ組み合わせたサンドイッチELISAにより、ジカウイルス(アフリカ株(AF)・アジア株(BR)各1株)及びデングウイルス(DV)1株のリコンビナントNS1の検出を行った。AFは、実施例1-1に記載のものを使用した。BR(#ZNS118-R-100)はAlpha diagnostics International社より購入し、DV(#PIP047A)はBio-Rad Laboratories社より購入した。各リコンビナント抗原を希釈液B(50mM Tris、150mM 塩化ナトリウム、0.1% Triton X-100、pH8.0)で希釈し、5ng/mLのAF抗原溶液、5ng/mLのBR抗原溶液、及び500ng/mLのDV抗原溶液を調製した。
抗体A~Dを固相化した96穴マイクロウェルプレートと、抗体A~Dをビオチン標識した標識抗体をそれぞれ組み合わせたサンドイッチELISAにより、ジカウイルス(アフリカ株(AF)・アジア株(BR)各1株)及びデングウイルス(DV)1株のリコンビナントNS1の検出を行った。AFは、実施例1-1に記載のものを使用した。BR(#ZNS118-R-100)はAlpha diagnostics International社より購入し、DV(#PIP047A)はBio-Rad Laboratories社より購入した。各リコンビナント抗原を希釈液B(50mM Tris、150mM 塩化ナトリウム、0.1% Triton X-100、pH8.0)で希釈し、5ng/mLのAF抗原溶液、5ng/mLのBR抗原溶液、及び500ng/mLのDV抗原溶液を調製した。
抗体A~DをそれぞれPBSで希釈し、10μg/mLの溶液を調製した。これを96穴マイクロウェルプレートに100μLずつ分注し、37℃で1時間コーティングさせた。次いで、2%BSA・PBS溶液を250μL/ウェル分注し、4℃で一晩ブロッキングした。これをPBSTで洗浄して、各抗体の固相化プレートを調製した。
前記抗体固相化プレートに、前記のAF抗原溶液、BR抗原溶液及びDV抗原溶液をそれぞれ100μL/ウェル分注し、37℃で30分間反応させた。PBSTで洗浄後、ビオチンで標識した抗体A~D(0.05μg/mL希釈液B)を100μL/ウェル分注し、37℃で1時間反応させた。PBSTで洗浄後、希釈液で10000倍希釈したアルカリホスファターゼ標識ストレプトアビジン(#GTX30954、ジェネンテック社製)溶液を100μL/ウェル分注し、37℃で1時間反応させた。PBSTで洗浄後、AMPPD基質液(ルミパルス(登録商標)基質液(富士レビオ社製))を100μL/ウェル分注して10分間発光させ、マイクロプレートリーダーにて発光量を測定した。各ウェルの発光量の値(S)を、同条件でブランク(希釈液Bのみ)を測定した発光量の値(N)で除して、S/N比を求めた。固相化プレートの作製に用いた抗体(固相抗体)とビオチンで標識した抗体(標識抗体)と抗原との各組み合わせ条件(表3の試験例1~16)でのS/N比を表3に示す。
試験例12においてのみ、100倍濃度のデングウイルス抗原検出時に「+」となったが、他の試験例ではいずれも「-」となった。試験例12についても、ジカウイルス感染判定のカットオフをS/N比15.0以上とすることで、デングウイルスの交差反応の影響を回避できると考えられる。ジカウイルスの検出において、AF、BRの両方について検出可能であったのは、試験例2、5、7、8、10~12及び15であった。ジカウイルス感染判定のカットオフをS/N比3.0以上とした表3では試験例1、6、13の結果がいずれも「-」となったが、下記のイムノクロマトグラフィーによるジカウイルス検出結果でも示されるように、少なくともBRについてはいずれの試験例でも検出可能であった。
[実施例1-5]イムノクロマトグラフィーによるジカウイルス検出
図1に示すように、巾5mm、長さ50mmのニトロセルロース膜のマトリクス2の展開液吸収ゾーン5側の末端から15mmの位置に抗体A~Dのいずれかを含む水溶液0.7μLをニトロセルロース膜に点着し乾燥させ、検出ゾーン6を作製した。さらに2の展開液吸収ゾーン5側の末端から12mmの位置に抗アルカリホスファターゼ(ALP)抗体溶液を点着し乾燥させ、展開確認部10を作製した。次いで、抗体A~DのいずれかをALP標識した標識抗体溶液5μLを点着し乾燥させ、酵素標識試薬パッド4からなる標識試薬ゾーンを作製した。
図1に示すように、巾5mm、長さ50mmのニトロセルロース膜のマトリクス2の展開液吸収ゾーン5側の末端から15mmの位置に抗体A~Dのいずれかを含む水溶液0.7μLをニトロセルロース膜に点着し乾燥させ、検出ゾーン6を作製した。さらに2の展開液吸収ゾーン5側の末端から12mmの位置に抗アルカリホスファターゼ(ALP)抗体溶液を点着し乾燥させ、展開確認部10を作製した。次いで、抗体A~DのいずれかをALP標識した標識抗体溶液5μLを点着し乾燥させ、酵素標識試薬パッド4からなる標識試薬ゾーンを作製した。
展開液パッド3は、巾6mm、長さ20mmのろ紙上に、基質として5-ブロモ-4-クロロ-3-インドリル-リン酸二ナトリウム塩(BCIP)溶液5μLをライン状に点着して乾燥させて基質ゾーン7を作製して調製した。前記マトリクス2、展開液パッド3、酵素標識試薬パッド4及び吸収パッド5(高保水性ろ紙)を、展開液槽11を有するプラスチックケースに固定して、図2及び図3に示すジカウイルス検出器具1を製造した。
製造したジカウイルス検出器具1(イムノクロマト)を使用して、ジカウイルス、デングウイルスへの反応性を試験した。AF抗原、BR抗原、DV抗原は実施例1-4と同じものを使用し、これを希釈液C(100mM Tris、150mM 塩化ナトリウム、0.095% アジ化ナトリウム、3% BSA、1% ニューポール、pH8.0)で希釈して5ng/mLのAF抗原溶液、5ng/mLのBR抗原溶液、及び200ng/mLのDV抗原溶液を調製した。各抗原溶液25μLを検体ゾーン8に滴下した後、変形部材に設けた押し込み部12を下方に加圧して変形させて、変形部材に付設された突起部13によって展開液パッド3を展開液槽11に挿入して展開液を展開液パッド3に供給して測定を開始した。測定開始15分後、展開確認部10の発色によって展開液の展開を確認した後、検出ゾーン6の発色を目視で測定した。検出ゾーン6の作製に用いた抗体(固相抗体)とALPで標識した抗体(標識抗体)と抗原との各組み合わせ条件(表4の試験例1~16)における検出結果を表4に示す。
いずれの条件においても、DV抗原では「-」判定となった。ジカウイルスの検出において、AF、BRの両方について検出可能であったのは、試験例2、5、7、8、10、12、14、及び15であった。
[実施例1-6]CLEIAによるジカウイルス検出
以下の方法で抗体C又はDを単独で、あるいは抗体B、C及びDのうち2種を組み合わせて磁性粒子(平均粒径3μm)に固相化させ、5種の抗ジカウイルス抗体固相化粒子を調製した。すなわち、まず、10mM MES緩衝液(pH5.0)中で磁性粒子0.01g/mLに、抗体C、抗体D、又は抗体B、C及びDのうち2種を等量組み合わせた混合抗体を、それぞれ、計0.2mg/L添加し、25℃で1時間ゆるやかに撹拌しながらインキュベートした。反応後、磁性粒子を磁石で集磁し、洗浄液(50mM Tris、150mM NaCl、2.0% BSA、pH7.0)で洗浄し、抗ジカウイルス抗体固相化粒子を得た。5種の抗ジカウイルス抗体固相化粒子をそれぞれ粒子希釈液(50mM Tris、150mM NaCl、1mM EDTA、0.5% BSA、0.5% Tween 40、pH7.2)で0.04%に懸濁し、各種粒子液を調製した。
以下の方法で抗体C又はDを単独で、あるいは抗体B、C及びDのうち2種を組み合わせて磁性粒子(平均粒径3μm)に固相化させ、5種の抗ジカウイルス抗体固相化粒子を調製した。すなわち、まず、10mM MES緩衝液(pH5.0)中で磁性粒子0.01g/mLに、抗体C、抗体D、又は抗体B、C及びDのうち2種を等量組み合わせた混合抗体を、それぞれ、計0.2mg/L添加し、25℃で1時間ゆるやかに撹拌しながらインキュベートした。反応後、磁性粒子を磁石で集磁し、洗浄液(50mM Tris、150mM NaCl、2.0% BSA、pH7.0)で洗浄し、抗ジカウイルス抗体固相化粒子を得た。5種の抗ジカウイルス抗体固相化粒子をそれぞれ粒子希釈液(50mM Tris、150mM NaCl、1mM EDTA、0.5% BSA、0.5% Tween 40、pH7.2)で0.04%に懸濁し、各種粒子液を調製した。
一方、抗体B、C又はDを単独で、あるいはこれらのうち2種を等量組み合わせた混合抗体をアルカリホスファターゼ(ALP)でそれぞれ標識し、5種の標識体を調製し、標識体希釈液(1.0mM MgCl2、0.3mM ZnCl2、150mM NaCl、0.5% BSA、0.5% Tween 80、pH6.8)で0.4μg/mLに希釈し、各種標識体液を調製した。
AF抗原、BR抗原は実施例1-4と同じものを使用し、ルミパルス(登録商標)用検体希釈液(富士レビオ社製)で希釈し、10、50、100、1000pg/mLのAF抗原溶液、10、50、100、1000pg/mLのBR抗原溶液を調製した。
各抗原溶液について、上記の各種粒子液、各種標識体液を用いて、自動分析器ルミパルス(登録商標)L2400(富士レビオ社製)によるジカウイルス検出を行った。各抗原溶液50μLを粒子液と混合し、37℃で8分間反応させた。粒子を集磁してルミパルス(登録商標)洗浄液(富士レビオ社製)で洗浄し、標識体液50μLを添加して懸濁させ、37℃で8分間反応させた。再度、粒子を集磁してルミパルス(登録商標)洗浄液で洗浄し、未反応の標識体を除去した後、ルミパルス(登録商標)基質液(富士レビオ社製)を200μL添加し、37℃で4分間発光させ、発光量を測定した。表5に、抗ジカウイルス抗体固相化粒子の作製に用いた抗体(粒子抗体)とALPで標識した抗体(標識抗体)と抗原との各組み合わせ条件(表5の試験例1~9)での実測値(カウント)からブランク値を減じた値を示す。表5中の試験例1~9に示す条件においては、AF抗原、BR抗原ともに10pg/mLの濃度まで十分に検出可能であった。
以上説明したように、本発明によれば、ジカウイルスを、特異的かつ簡便・迅速に、高感度で検出することが可能となる。ジカウイルスは、ジカウイルス感染症(ジカ熱)の原因となるウイルスであることから、本発明は、研究上の利用にとどまらず、ジカウイルスによる疾患の診断においても大きく貢献し得るものである。

Claims (9)
- 試料中のジカウイルスを検出する方法であって、
試料中に含まれるジカウイルスNS1タンパク質を、ジカウイルスNS1タンパク質に対するモノクローナル抗体又はその組み合わせを用いたイムノアッセイにより検出する工程を含み、
前記モノクローナル抗体が、(a)又は(b)の特徴を有するモノクローナル抗体である方法。
(a)配列番号1の260-352番目のアミノ酸配列からなるペプチド断片に結合する、
(b)配列番号1の1-176番目のアミノ酸配列からなるペプチド断片に結合する。 - ジカウイルス感染症の診断を補助する方法であって、
ヒトから採取された検体に含まれるジカウイルスNS1タンパク質を、ジカウイルスNS1タンパク質に対するモノクローナル抗体又はその組み合わせを用いたイムノアッセイにより検出する工程を含み、
前記モノクローナル抗体が、(a)又は(b)の特徴を有するモノクローナル抗体である、方法。
(a)配列番号1の260-352番目のアミノ酸配列からなるペプチド断片に結合する、
(b)配列番号1の1-176番目のアミノ酸配列からなるペプチド断片に結合する。 - 前記イムノアッセイが、抗原捕捉用抗体と検出用抗体とを用いたサンドイッチイムノアッセイであり、前記抗原捕捉用抗体及び前記検出用抗体の少なくとも一方が、(a)又は(b)の特徴を有するモノクローナル抗体である、請求項1又は2に記載の方法。
- 前記抗原捕捉用抗体と前記検出用抗体とが、(a)の特徴を有する互いに異なるモノクローナル抗体同士の組み合わせである、請求項3に記載の方法。
- 前記抗原捕捉用抗体と前記検出用抗体とが、(a)の特徴を有するモノクローナル抗体と(b)の特徴を有するモノクローナル抗体の組み合わせである、請求項3に記載の方法。
- (a)の特徴を有するモノクローナル抗体が、さらに、260-310番目のアミノ酸配列からなるペプチド断片と300-352番目のアミノ酸配列からなるペプチド断片には結合しないという特徴を有する、請求項1から5のうちのいずれか一項に記載の方法。
- (a)の特徴を有するモノクローナル抗体が、280-352番目のアミノ酸配列からなるペプチド断片に結合し、かつ260-310番目のアミノ酸配列からなるペプチド断片と300-352番目のアミノ酸配列からなるペプチド断片には結合しないという特徴を有する、請求項6に記載の方法。
- (b)の特徴を有するモノクローナル抗体が、さらに、配列番号1の83-141番目のアミノ酸配列からなるペプチド断片及び89-264番目のアミノ酸配列からなるペプチド断片に結合するという特徴を有する、請求項1から3、及び5のうちのいずれか一項に記載の方法。
- 請求項1から8のうちのいずれか一項に記載の方法に用いるためのキットであって、
ジカウイルスNS1タンパク質に対するモノクローナル抗体又はその組み合わせを含み、
前記モノクローナル抗体が、(a)又は(b)の特徴を有するモノクローナル抗体であるキット。
(a)配列番号1の260-352番目のアミノ酸配列からなるペプチド断片に結合する、
(b)配列番号1の1-176番目のアミノ酸配列からなるペプチド断片に結合する。
