カルボン酸8の反応性化合物としては、例えば、酸ハライド(例えば、酸クロリド)、混合酸無水物、イミダゾリド、活性アミド等、アミド縮合形成反応に通常用いられるものを挙げることが出来る。
カルボン酸8を用いる場合は、縮合剤として、例えば、1,1’-カルボニルジイミダゾール(以下、「CDI」という。)、1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド(以下、「EDCI」という)、O-(7-アザベンゾトリアゾール-1-イル)-1,1,3,3-テトラメチルウロニウム ヘキサフルオロホスファート(以下、「HATU」という。)、ジフェニルホスホリルアジドを使用することが出来る。

[工程1]2-(チアゾール-2-イル)アセトニトリルの製造
シアノ酢酸tert-ブチルエステル(28g)のDMF(100mL)溶液に氷冷下、60%水素化ナトリウム(7.9g)を少しずつ加え、10分間撹拌した。ここに2-ブロモチアゾール(25g)を加え、室温で15分撹拌し、ついで120℃で2時間撹拌した。反応混合液に1M塩酸水溶液を加えた後、酢酸エチルで抽出した。有機層を水で洗浄後、無水硫酸マグネシウムで乾燥して減圧濃縮した。得られた残渣をヘキサンで洗浄し、トルエン(200mL)に懸濁し、p-トルエンスルホン酸一水和物(2.0g)を加えて105℃で2時間撹拌した。反応液を酢酸エチルで希釈した後、飽和重曹水で洗浄した。得られた有機層を無水硫酸マグネシウムで乾燥後、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィーで精製し、表題化合物(7.0g)を得た。

[工程2]ピラゾロ[5,1-b][1,3]チアゾール-7-カルボニトリルの製造
工程1で得られた2-(チアゾール-2-イル)アセトニトリル(5g)のジクロロメタン(50mL)溶液に氷冷下でO-(メシチルスルホニル)ヒドロキシアミン(例えば、Organic Process Research & Development,2009,13,263-267.に記載の方法に準じて合成した)のジクロロメタン(20mL)溶液を加え、室温で2時間撹拌した。氷冷下、反応混合物にジエチルエーテルを加えて析出した固形物を濾取した。得られた固形物をオルトギ酸トリエチル(35mL)に懸濁し、120℃で1時間撹拌した。反応溶液を減圧濃縮して得られた残渣をシリカゲルカラムクロマトグラフィーで精製し、表題化合物(2.5g)を得た。

参考例1で得られたピラゾロ[5,1-b][1,3]チアゾール-7-カルボニトリル(6g)のメタノール(150mL)溶液に、28%ナトリウムメトシキドメタノール溶液(24.6mL)を加え、室温で3時間撹拌した。ついで塩化アンモニウム(12.9g)を加え、90℃で1時間撹拌した。反応液を減圧濃縮して得られた残渣にオキサロ酢酸ジエチルナトリウム(33.8g)の5M水酸化ナトリウム水溶液(200mL)を加えて、100℃で終夜撹拌した。反応混合液に濃塩酸を加えて酸性とし、析出した固形物を濾取した。得られた固形物を5M水酸化カリウム水溶液に溶解しクロロホルムで洗浄した。水層に濃塩酸を加えて酸性とし、析出した固形物を濾取して乾燥し、表題化合物(10g)を得た。

参考例2で得られた6-ヒドロキシ-2-(ピラゾロ[5,1-b][1,3]チアゾール-7-イル)ピリミジン-4-カルボン酸(1.4g)をオキシ塩化リン(20mL)に懸濁し、ジエチルアニリン(1.6g)を加えて、130℃で2時間撹拌した。反応溶液を減圧濃縮して氷冷下、反応混合物にメタノール(100mL)を加え、10分間撹拌した。反応液をクロロホルムで希釈した後、水で洗浄した。得られた有機層を無水硫酸マグネシウムで乾燥後、減圧濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィーで精製し、表題化合物(910mg)を得た。

参考例3で得られた6-クロロ-2-(ピラゾロ[5,1-b][1,3]チアゾール-7-イル)ピリミジン-4-カルボン酸メチル(1.0g)のDMF(10mL)溶液に、DIPEA(1.8mL)、(1S)-1-シクロプロピルエタンアミン(320mg)を加えて、80℃で3時間撹拌した。反応混合物をシリカゲルカラムクロマトグラフィーで精製し、表題化合物(1.1g)を得た。

[工程2]6-{[(1S)-1-シクロプロピルエチル]アミノ}-2-(ピラゾロ[5,1-b][1,3]チアゾール-7-イル)ピリミジン-4-カルボン酸の製造
工程1で得られた6-{[(1S)-1-シクロプロピルエチル]アミノ}-2-(ピラゾロ[5,1-b][1,3]チアゾール-7-イル)ピリミジン-4-カルボン酸メチル(600mg)のTHF(15mL)、水(5mL)溶液に、水酸化リチウム一水和物(100mg)を加えて、室温で1時間撹拌した。反応混合液に1M塩酸水溶液を加えて酸性とした後、THFを減圧留去し、クロロホルムで抽出した。得られた有機層を無水硫酸ナトリウムで乾燥して減圧濃縮し、表題化合物(470mg)を得た。

水素化ホウ素ナトリウム(19.1g)をTHF(750mL) に加え、N-アミノローダニン(250g)のTHF(500mL)スラリーを5℃以下で分割添加した。5℃以下で30分撹拌後、メタノール(111mL)を滴下し2時間撹拌した。濃塩酸 (44mL)を水 (500mL)で希釈し滴下した後、水(1000mL)を滴下し、10℃以下で1時間撹拌した。析出した結晶をろ過し、水(600mL)で洗浄後、減圧下40℃で乾燥し表題化合物(209.1g)を得た。

ナトリウムメトキシド(14.3g)のシクロペンチルメチルエーテル(300mL)スラリーに、20℃以下でギ酸メチル(17.5g)を滴下した後、クロロアセトニトリル(20g)を滴下し、30℃以下で3時間撹拌した。反応終了後、析出した結晶をろ過してシクロペンチルメチルエーテル(40mL) で洗浄後、減圧下40℃で乾燥し表題化合物(29.2g)を得た。
3-アミノ-4-ヒドロキシ-1,3-チアゾリジン-2-チオン(50g)の2-プロパノール(250mL)スラリーに濃硫酸(48.97g)を添加し、80℃にて1時間加熱撹拌した。冷却し40℃以下でアセトニトリル(500mL)、参考例8で得られた2-クロロ-2-シアノエテン-1-オレートナトリウム(62.65g)を添加し、80℃で4時間撹拌した。冷却し活性炭(10g)添加し室温で30分撹拌した。不溶物をろ過しアセトニトリルで3回(100mL)洗浄した。ろ液を減圧濃縮し、続いてメタノール(100mL)で3回共沸してアセトニトリルを除いた後、濃縮物にメタノール(150mL)とTHF(150mL)を加えた。これを、20℃以下でメタノール (350mL)と塩化アセチル(209g)から調製した溶液に添加した。室温で終夜撹拌した後、THF(350mL)を添加しさらに10℃以下で1時間撹拌した。析出した結晶をろ過してTHF(300mL)で洗浄後、減圧下50℃で乾燥し表題化合物(45.5g)を得た。

6-ヒドロキシ-2-(ピラゾロ[5,1-b][1,3]チアゾール-7-イル)ピリミジン-4-カルボン酸(238g)のDMF(714mL)スラリーにTEA(275.51g)を添加し50℃で30分撹拌した。冷却した後、20℃以下で1,1’-カルボニルジイミダゾール(323.76g)を添加した。30分撹拌後、メチル ピペリジン-4-イルカルバメート トシル酸塩(449.79g)を添加し30分撹拌した。反応終了後、室温下アセトニトリル(3570mL)を滴下し、終夜撹拌した。析出した結晶をろ過してアセトニトリル(480mL)で洗浄後、減圧下60℃で乾燥し表題化合物(377.79g)を得た。

参考例13で得られた6-{4-[(メトキシカルボニル)アミノ]ピペリジン-1-カルボニル}-2-(ピラゾロ[5,1-b][1,3]チアゾール-7-イル)ピリミジン-4-イル 4-メチルベンゼン-1-スルホナート(1.0g)のアセトニトリル(7.0mL)溶液にDIPEA(0.67g)、(2S)-ブタン-2-アミン(0.4g)を添加した後、封管して100℃で2時間加熱撹拌した。反応終了後、反応液を酢酸エチルで希釈した後、水(30mL)および飽和食塩水(30mL)で洗浄した。有機層を無水硫酸マグネシウムで乾燥して減圧濃縮した後、カラムクロマトグラフィーで精製し表題化合物(650mg)を得た。高速液体クロマトグラフィーにより、得られた表題化合物の光学純度が99%以上であることを確認した(保持時間:35.7分)。

実施例1工程1で得られたメチル [1-({6-[(2S)-ブタン-2-イルアミノ]-2-(ピラゾロ[5,1-b][1,3]チアゾール-7-イル)ピリミジン-4-イル}カルボニル)ピペリジン-4-イル]カルバメート(202mg)にアセトニトリル(5mL)を加え、50℃に加熱した。p-トルエンスルホン酸1水和物(83mg)を加えて、室温で終夜撹拌した。析出した結晶をろ取し、減圧乾燥して表題化合物の結晶(210mg)を得た。粉末X線回折スペクトルを図1に示す。得られた結晶は、元素分析により、一水和物であることが示された。

参考例13で得られた6-{4-[(メトキシカルボニル)アミノ]ピペリジン-1-カルボニル}-2-(ピラゾロ[5,1-b][1,3]チアゾール-7-イル)ピリミジン-4-イル 4-メチルベンゼン-1-スルホナート(1.5g)のアセトニトリル(10mL)溶液にDIPEA(1.0g)、(2R)-ブタン-2-アミン(0.59g)を添加した後、封管して100℃で2時間加熱撹拌した。反応終了後、反応液を酢酸エチルで希釈した後、水(50mL)および飽和食塩水(30mL)で洗浄した。有機層を無水硫酸マグネシウムで乾燥して減圧濃縮した後、カラムクロマトグラフィーで精製し表題化合物(0.93g)を得た。高速液体クロマトグラフィーにより、得られた表題化合物の光学純度が99%以上であることを確認した(保持時間:29.1分)。

[工程2]メチル (1-{[6-{[(1S)-1-シクロプロピルエチル]アミノ}-2-(ピラゾロ[5,1-b][1,3]チアゾール-7-イル)ピリミジン-4-イル]カルボニル}ピペリジン-4-イル)カルバメートトシル酸塩一水和物の製造
実施例3工程1で得られたメチル (1-{[6-{[(1S)-1-シクロプロピルエチル]アミノ}-2-(ピラゾロ[5,1-b][1,3]チアゾール-7-イル)ピリミジン-4-イル]カルボニル}ピペリジン-4-イル)カルバメート(201mg)にアセトニトリル(5mL)を加え、50℃に加熱した。p-トルエンスルホン酸一水和物(81mg)を加えて、室温で終夜撹拌した。析出した結晶をろ取し、減圧乾燥して表題化合物の結晶(237mg)を得た。粉末X線回折スペクトルを図3に示す。得られた結晶は、元素分析により、一水和物であることが示された。

参考例6で得られた6-{[(1S)-1-シクロプロピルエチル]アミノ}-2-(ピラゾロ[5,1-b][1,3]チアゾール-7-イル)ピリミジン-4-カルボン酸(350mg)のDMF(8.0mL)溶液に、N-(ピペリジン-4-イル)シクロプロパンカルボキサミド(268mg)、DIPEA(552μL)、HATU(606mg)を加えて、室温で1時間撹拌した。反応混合物をシリカゲルカラムクロマトグラフィーで精製し、表題化合物(410mg)を得た。得られた表題化合物(350mg)に酢酸エチル(7mL)を加えて加熱溶解し、室温で20時間撹拌した。析出した結晶を濾取し、減圧乾燥して表題化合物の結晶(280mg)を得た。粉末X線回折スペクトルを図5に示す。

アルゴン雰囲気下、参考例5で得られたN-(1-{[6-クロロ-2-(ピラゾロ[5,1-b][1,3]チアゾール-7-イル)ピリミジン-4-イル]カルボニル}ピペリジン-4-イル)シクロプロパンカルボキサミド(550mg)のtert-ブタノール(20mL)溶液に、TEA(534μL)、(2R)-3,3-ジメチルブタン-2-アミン(258mg)を加えて、90℃で終夜撹拌した。反応混合物を減圧濃縮し、得られた残渣をシリカゲルカラムクロマトグラフィーで精製し、表題化合物(570mg)を得た。得られた表題化合物(100mg)に酢酸エチル(2mL)を加えて溶解し、室温で20時間撹拌した。析出した結晶を濾取し、減圧乾燥して表題化合物の結晶(70mg)を得た。粉末X線回折スペクトルを図6に示す。

1.被験物質の調製
被験物質はジメチルスルホキシド(DMSO)で10mMに調製し、さらに1000、100、10、1、0.1、0.01μMの濃度になるようにDMSOでそれぞれ希釈した。JAK1については10mM、1000μM、100μM、10μM、1μM、0.1μMの6濃度、JAK2およびJAK3については1000μM、100μM、10μM、1μM、0.1μM、0.01μMの6濃度のDMSO溶液を用い、アッセイバッファーで20倍に希釈した被験物質溶液を調製した。陰性コントロールにはDMSOをアッセイバッファーで20倍に希釈した溶液を用いた。アッセイバッファーには15mM Tris-HCl(pH7.5)、0.01(v/v)% Tween-20、1mM ジチオスレイトールを用いた。
2.1mM ATP下におけるJAKチロシンキナーゼ阻害活性の測定
活性の測定にはELISA法を用いた。被験物質溶液をStreptavidine coated 96 well plate(DELFIA Strip Plate 8×12well、PerkinElmer社)に10μLずつ添加し(n=2)、基質溶液(ビオチン標識ペプチド基質1250nM(JAK1)、625nM(JAK2、JAK3)、2.5mM ATP(終濃度1mM)、25mM MgCl2、15mM Tris-HCl(pH7.5)、0.01(v/v)% Tween-20、1mM ジチオスレイトール)を20μLずつ添加し撹拌した。最後にJAKチロシンキナーゼ(カルナバイオサイエンス社)(アッセイバッファーにて7.5nM(JAK1)、0.75nM(JAK2、JAK3)に希釈済み)を20μLずつ添加して撹拌し、30℃で1時間反応を行った。プレートを洗浄バッファー(50mM Tris-HCl(pH7.5)、150mM NaCl、0.02(v/v)% Tween-20)で4回洗浄した後、ブロッキングバッファー(0.1% Bovine Serum Albumin、50mM Tris-HCl(pH7.5)、150mM NaCl、0.02(v/v)% Tween-20)を150μLずつ添加して、30℃で1時間ブロッキングを行った。ブロッキングバッファーを取り除き、Horseradish Peroxidase標識抗リン酸化チロシン抗体(BD Biosciences社)(ブロッキングバッファーにて10000倍に希釈済み)を100μLずつ添加し、30℃で30分間インキュベートした。プレートを洗浄バッファーで4回洗浄し、3,3’,5,5’-テトラメチルベンジジン溶液(ナカライテスク社)を100μLずつ添加して10分間発色させた。0.1M硫酸を100μLずつ添加して反応を停止した。マイクロプレートリーダー(BIO-RAD社)にて450nmの吸光度を測定した。
3.測定結果の解析
測定した吸光度について、SASシステム(SAS Institute Inc.)により非線形回帰分析を行い、各チロシンキナーゼ活性を50%阻害する被験物質の濃度(IC50)を算定した。その結果を以下の表1に示す。
Aspergillus fumigatus抽出物(Greer laboratories社)をPBS溶液で400μg/mLとなるように調製した。Day0、Day1、Day7およびDay8に調製したAspergillus fumigatus溶液50μLをマウスに点鼻投与する。点鼻投与は朝の被験物質投与の1時間後に行う。被験物質投与はDay0からDay9まで朝夕1日2回行う。被験物質は0.5%メチルセルロースに10mg/mLの濃度で懸濁させ、10mL/kgの用量で経口投与する。Day10に気管支肺胞洗浄液(bronchoalveolar lavage fluid:BALF)を回収し、BALF中の総白血球数を、Celltac(日本光電社)を用いて計測する。次いで、ADVIA120(Siemens Healthcare Diagnostics社)を用いて、総白血球数中の好酸球数の割合を算出し、総白血球数に乗じることでBALF中の好酸球数を算出する。Aspergillus fumigatus抽出物処置+0.5%メチルセルロース投与群の好酸球数を抑制率0%、Aspergillus fumigatus抽出物未処置+0.5%メチルセルロース投与群の好酸球数を抑制率100%として各被験物質の抑制率を算出する。
1.被験物質の調製
被験物質をジメチルスルホキシド(DMSO)で10mMに調製し、さらに300、100μMの濃度になるようにDMSOで希釈する。さらにRPMI1640培地で100倍に希釈した溶液を被験物質溶液とする。陰性コントロールにはDMSOをRPMI1640で100倍に希釈した溶液を用いる。
2.リン酸化STAT6活性の測定
被験物質溶液または陰性コントロール溶液50μLとDND39細胞液400μL(細胞数:105cells)を混合し37℃で30分間振盪する。刺激物質としてインターロイキン-4(10ng/mL)を50μL添加し15分間振盪する。固定試薬Fixation buffer(BD Biosciences社)を500μL添加し、10分間振盪して反応を停止する。遠心して上清を除去した後、膜透過試薬Perm buffer III(BD Biosciences社)を500μL添加し、4℃で30分間インキュベートする。Stain buffer(BD Biosciences社)にて2度の洗浄操作後、Alexa Fluor 647 Mouse Anti-Stat6(pY641)(BD Biosciences社)を添加し、冷暗所にて30分間インキュベートする。得られた細胞溶液をフローサイトメーターに供する。インターロイキン-4刺激陰性コントロール群蛍光強度のGEOMEAN値を抑制率0%、無刺激陰性コントロール群蛍光強度のGEOMEAN値を抑制率100%として各被験物質の抑制率を算出し、これらの被験物質がIL-4のシグナルを抑制することを確認する。
1.被験物質の調製
被験物質をジメチルスルホキシド(DMSO)で10mMに調製する。さらに、RPMI1640培地で100倍に希釈した溶液を被験物質溶液とする。陰性コントロールにはDMSOをRPMI1640で100倍に希釈した溶液を用いる。
2.リン酸化STAT5活性の測定
ヒト新鮮血100μLに被検物質溶液または陰性コントロール溶液を10μL加え、37℃で30分間振盪する。刺激物質としてインターロイキン-7(100ng/mL)を10μLずつ添加し15分間振盪する。Lyse/fix buffer(BD Biosciences社)を蒸留水で5倍希釈し、1.4mL加える。その後、10分間振盪したのち、遠心し、細胞を分離する。上清を除去後、PBSを1mL加える。遠心し、PBSを除去した後、Perm buffer III(BD Biosciences社)を500μL添加し、4℃で30分間インキュベートする。Stain buffer(BD Biosciences社)にて2度の洗浄操作後、Alexa Fluor 647 Mouse Anti-Stat5抗体(pY694)(BD Biosciences社)を添加し、冷暗所にて30分間インキュベートする。得られた細胞溶液をフローサイトメーターに供する。IL-7刺激陰性コントロール群蛍光強度のGEOMEAN値を抑制率0%、無刺激陰性コントロール群蛍光強度のGEOMEAN値を抑制率100%として各被験物質の抑制率を算出し、これらの被験物質がIL-7のシグナルを抑制することを確認する。

- メチル [1-({6-[(2S)-ブタン-2-イルアミノ]-2-(ピラゾロ[5,1-b][1,3]チアゾール-7-イル)ピリミジン-4-イル}カルボニル)ピペリジン-4-イル]カルバメートトシル酸塩一水和物。
- Cu Kα放射線を用いて得られる粉末X線回折スペクトルにおいて、少なくとも次の回折角2θ:12.6度、13.3度、17.3度、20.0度、20.4度、21.3度及び22.3度に回折ピークを示す、請求項1に記載の化合物の結晶。
- 請求項1または2に記載の化合物を有効成分として含有する医薬組成物。
- メチル [1-({6-[(2R)-ブタン-2-イルアミノ]-2-(ピラゾロ[5,1-b][1,3]チアゾール-7-イル)ピリミジン-4-イル}カルボニル)ピペリジン-4-イル]カルバメートトシル酸塩一水和物。
- Cu Kα放射線を用いて得られる粉末X線回折スペクトルにおいて、少なくとも次の回折角2θ:12.6度、13.3度、17.3度、20.0度、20.4度、21.3度及び22.3度に回折ピークを示す、請求項4に記載の化合物の結晶。
- 請求項4または5に記載の化合物を有効成分として含有する医薬組成物。
- メチル (1-{[6-{[(1S)-1-シクロプロピルエチル]アミノ}-2-(ピラゾロ[5,1-b][1,3]チアゾール-7-イル)ピリミジン-4-イル]カルボニル}ピペリジン-4-イル)カルバメートトシル酸塩一水和物。
- Cu Kα放射線を用いて得られる粉末X線回折スペクトルにおいて、少なくとも次の回折角2θ:12.6度、13.3度、17.2度、20.6度及び21.8度に回折ピークを示す、請求項7に記載の化合物の結晶。
- 請求項7または8に記載の化合物を有効成分として含有する医薬組成物。
- Cu Kα放射線を用いて得られる粉末X線回折スペクトルにおいて、少なくとも次の回折角2θ:12.0度、13.8度、15.0度、16.0度、19.4度、20.9度及び21.9度に回折ピークを示す、エチル (1-{[6-{[(1S)-1-シクロプロピルエチル]アミノ}-2-(ピラゾロ[5,1-b][1,3]チアゾール-7-イル)ピリミジン-4-イル]カルボニル}ピペリジン-4-イル)カルバメートの結晶。
- 請求項10に記載の化合物を有効成分として含有する医薬組成物。
- Cu Kα放射線を用いて得られる粉末X線回折スペクトルにおいて、少なくとも次の回折角2θ:11.1度、12.9度、15.4度、17.8度、21.2度及び22.3度に回折ピークを示す、N-(1-{[6-{[(1S)-1-シクロプロピルエチル]アミノ}-2-(ピラゾロ[5,1-b][1,3]チアゾール-7-イル)ピリミジン-4-イル]カルボニル}ピペリジン-4-イル)シクロプロパンカルボキサミドの結晶。
- 請求項12に記載の化合物を有効成分として含有する医薬組成物。
- Cu Kα放射線を用いて得られる粉末X線回折スペクトルにおいて、少なくとも次の回折角2θ:10.6度、13.0度、14.6度、17.4度、17.7度、21.3度及び21.7度に回折ピークを示す、N-(1-{[6-{[(2R)-3,3-ジメチルブタン-2-イル]アミノ}-2-(ピラゾロ[5,1-b][1,3]チアゾール-7-イル)ピリミジン-4-イル]カルボニル}ピペリジン-4-イル)シクロプロパンカルボキサミドの結晶。
