  • Examples 1 - 2 uses a S. cerevisae yeast strain, which was a xylose and glucose fermenting yeast and commercially available. This yeast strain was propagated in Examples 1 - 2 to produce an amount to inoculate fermentation of Hydrolysate D.

It is understood that the concentrations of glucose, xylose, lactic acid, acetic acid, and ethanol in the hydrolysate-containing propagation medium can be calculated by multiplying the percent of hydrolysate used.

  • the portion of the respective Hydrolysate used for the pre-propagation medium and propagation medium was centrifuged then filtered through glass fiber filters to remove large particulate matter followed by filter sterilization via a 0.45 ⁇ Nalgene filter. Fermentation was performed in 125 mL baffled, one-way vented shake flasks using a cell pitch of 1 g (DW)/L at 32°C and initial pH of 5.8 adjusted using 8 - 10 N KOH.

It is expected that the difference between the yeast propagated according to methods described herein and the control would be greater for hydrolysates that contain ethanol amounts of greater than about 13 - 14 g/L ethanol. For instance, the difference in yields between Examples 1 - 2 at 72-hour fermentation was 4 g/L. It is expected that this difference can increase to greater than 5 g/L or greater than 10 g/L if the hydrolysate contains about 20 g/L endogenous ethanol where propagating the yeast as described herein provides better fermentation performance as the endogenous ethanol inhibitor level goes up.

Abstract

L'invention concerne des procédés, des systèmes et des compositions pour la propagation d'un microorganisme de fermentation qui peut présenter un rendement en produit amélioré pendant la fermentation d'un hydrolysat contenant au moins un inhibiteur de croissance, notamment de l'éthanol. Dans certains modes de réalisation, le procédé consiste : (a) à fournir un produit selon un procédé comprenant l'ensilage d'une biomasse cellulosique pendant au moins 24 heures pour produire de l'éthanol dans la biomasse cellulosique et générer une biomasse cellulosique ensilée, le prétraitement de la biomasse cellulosique ensilée avec une solution comprenant un acide alpha-hydroxysulfonique pour produire une matière prétraitée contenant de l'éthanol, et l'introduction d'une ou de plusieurs cellulases dans la matière prétraitée pour produire l'hydrolysat ; et (b) à fournir un réacteur de propagation contenant : un milieu de propagation comprenant l'hydrolysat en une quantité d'au moins 25 % en volume et du glucose en une quantité supérieure à 14 g/L ; et une première masse cellulaire d'un microorganisme de fermentation.
