目前对紫外线对皮肤损伤的机理认识有:氧化应激、炎症反应、细胞凋亡、胶原和弹性蛋白的降解。皮肤接受过多的紫外线照射,皮肤组织细胞会产生氧化应激,产生过量的活性氧簇ROS,破坏细胞膜的完整性和细胞内的抗氧化***,进一步引起细胞炎症、细胞凋亡及肿瘤的形成。同时过量的紫外线照射使皮肤胶原、透明质酸、弹性蛋白降解,导致皮肤弹性变性、粗糙、形成皱纹。因此,对于紫外线损伤后的修复方法大多是从这些机理出发,使用具有抗氧化、抗炎、抑制基质金属蛋白酶作用的物质。例如白蜡树提取物有效的清除UVB照射后人Fb细胞中生成的大量自由基,降低MMP-1的mRNA和蛋白的表达,从而对抗UVB照射后的皮肤损伤;又比如万寿菊花提取物中含有β胡萝卜素,其通过抑制透明质酸酶、弹性蛋白酶、MMP-1的活性,抑制皱纹产生,从而起到紫外线防护作用。
ATP是细胞能量新陈代谢的关键分子,是皮肤细胞可以直接使用的主要能量源,也是反应细胞健康活力的一项重要指标,ATP的减少,导致能量代谢的减弱,机体会相应的出现功能性的衰退,最明显的莫过于皮肤衰老、暗沉痤疮等。新的研究发现紫外线的强烈照射,能引起ATP生成减少,导致能量代谢的减弱,影响皮肤细胞正常功能。ATP的增加能促进损伤后细胞的增加,促 进细胞能量的新陈代谢,从而有利于紫外线照射造成的皮肤损伤进行修复。
桃花为蔷薇科植物桃Prunus persica(L.)Batsch的花。味苦性平,利水消肿,《神农本草经》里记载桃花"令人好颜色";桃仁为蔷薇科植物桃Prunus persica(L.)Batsch或山桃Prunus davidiana(Carr.)Franch.的干燥成熟种子。味苦甘性平,活血化瘀,兼能润燥。二者相配伍,活血化瘀、润燥利水,能令血瘀、水饮肿满之体恢复气血通调,排除体内气血水瘀滞。
现有技术记载了紫外线的皮肤损伤机制及具有紫外线防护作用的天然产物的研究进展。主要针对紫外线的皮肤损伤机制和天然来源紫外线防护剂的研究进展进行了综述。具体指出:皮肤长期暴露在紫外线下可以引起皮肤损伤,出现皮肤光老化甚至出现皮肤癌变和其他皮肤病变。紫外线引起的皮肤光老化的主要机制有:氧化应激、炎症反应、细胞凋亡、胶原和弹性蛋白的降解等。目前国内外关注的焦点是开发紫外线防护剂,目前主要研究的有黄酮类化合物、萜类化合物、鞣质类和香豆素类等。但是,已有的报道均未提到促进能量代谢和ATP的合成也能够对UVB进行修复。

由此可见,现有技术均未提及桃花桃仁复方在UVB照射损伤修复和促进能量代谢方面的作用。本发明首次发现桃花桃仁复方具有紫外损伤修复和促进皮肤细胞能量代谢方面的性质。
发明内容
本发明目的在于提供一种具有紫外损伤修复的中草药复方提取物,该中草药复方提取物能够促进ATP合成,对UVB造成的紫外线损伤进行修复。同时可作为功效添加剂加入到化妆品中使用,用于紫外损伤后皮肤护理。
桃花桃仁提取工艺流程如下:
将桃花药材以适宜溶媒浸泡约0.5-1h,以相应方法提取,取滤液,静置过滤,浓缩,干燥成粉。
将桃仁药材以适宜溶媒浸泡约0.5-1h,以相应方法提取,取滤液,静置过滤,浓缩,干燥成粉。
将桃花提取物与桃仁提取物按比例混合,最终制成含1%重量的复合提取物溶液。
配方中中药材可按照比例适当放大或缩小;适宜溶媒包括水(蒸馏水、去离子水、纯净水),不同浓度的乙醇溶液,不同浓度的多元醇溶液以及其他极性相似的溶剂;提取方法可以是煎煮、回流提取、浸渍、渗漉等方法等。
桃花桃仁复方提取物的制取也可采取超临界萃取、亚临界萃取、有机溶剂提取法。有机溶剂提取的溶剂包括正己烷、乙酸乙酯、***、石油醚、 丙酮等中低极性溶剂以及不同浓度的乙醇、甲醇以及多元醇等。提取方法可以是回流提取、浸渍、渗漉等方法。而且,提取过程中还可以使用超声或微波的方法进行辅助。
第一方面,本发明涉及桃花桃仁复方提取物在紫外损伤修复和促进能量代谢中的用途。
第二方面,本发明涉及桃花桃仁复方提取物作为具有紫外损伤修复和促进能量代谢作用的添加剂在化妆品中的用途。
在一个实施方式中,桃花桃仁复方提取物的用量在0.00001-100重量%。在另一个实施方式中,桃花桃仁复方提取物的用量在0.0001-50重量%。在优选的实施方式中,桃花桃仁复方提取物的用量为0.001%-20重量%。
本发明的化妆品选自:洁面乳、化妆水、乳液、膏霜、啫喱、面膜、洗发水、沐浴露、护发素。在一个实施方式中,本发明的化妆品包含桃花桃仁复方提取物以及可接受的辅料。
在一个实施方式中,化妆品包含0.00001-100重量%的桃花桃仁复方提取物。在另一个实施方式中,化妆品包含0.0001-50重量%的桃花桃仁复方提取物。在另一个实施方式中,化妆品包含0.001%-20重量%的桃花桃仁复方提取物。
附图说明
图1显示了正常细胞、UV照射细胞以及各种浓度的本发明桃花桃仁复方提取物的ATP含量百分比。图中,#表示相对于正常组,UVB照射组有显著差异;**表示相对于UVB照射组,具有显著差异(p<0.01)。
具体实施方式
目前针对紫外线损伤后的修复大多从氧化应激、DNA的修复、炎症反应、细胞凋亡、胶原和弹性蛋白的降解等角度出发,提供相应的解决方案。尚未有从ATP能量代谢角度提出紫外线损伤修复的解决方案。本发明首次提出桃花桃仁复方具有紫外损伤修复和促进能量代谢的作用。本发明提供的桃花桃仁复 方提取物能够对UVB照射后的细胞能量损失进行修复,促进皮肤细胞能量代谢和ATP的合成,从而有利于紫外损伤修复。
本发明的发明人在经过了广泛而深入的研究之后发现:桃花桃仁复方提取物具有紫外损伤修复和促进能量代谢作用。
因此,桃花桃仁复方提取物可作为功效添加剂加入到化妆品中。
文献中均未涉及桃花桃仁复方提取物具有紫外损伤修复和促进能量代谢作用,也未提及将其作为功能添加剂加入化妆品中。因此,本发明首次提出桃花桃仁复方提取物具有紫外损伤修复和促进能量代谢作用,可以作为功能添加剂加入化妆品中,用于紫外损伤修复及皮肤细胞能量的补充。
在化妆品应用中,桃花桃仁复方提取物可以是各种形式。例如,桃花桃仁复方提取物应用形式可以是洁面乳、化妆水、乳液、膏霜、啫喱、面膜、洗发水、沐浴露、护发素等任何可以应用于人体皮肤的剂型。
桃花桃仁复方提取物以外的其他成分可以是化妆品配方需要的任何成分,如:水、乙醇、丁二醇、丙二醇、油脂、乳化剂、增稠剂、防腐剂等;也可以添加其他功效成分达到复合功能,如:添加防晒成分使成为促进能量代谢防晒化妆品,添加控油成分使成为促进能量代谢控油化妆品,添加保湿成分成为促进能量代谢保湿化妆品,添加美白成分使成为促进能量代谢美白化妆品,添加抗衰老成分成为促进能量代谢抗衰老化妆品等等。
一般而言,桃花桃仁复方提取物的使用浓度可以在0.00001-100重量%。优选地,桃花桃仁复方提取物的使用浓度可以在0.0001-50重量%。在本发明的一个实施方式中,桃花桃仁复方提取物的使用浓度在0.00001-100重量%。在另一个实施方式中,桃花桃仁复方提取物的浓度在0.001-50重量%。在一个实施方式中,桃花桃仁复方提取物的使用浓度为0.001-20重量%。
在一个实施方式中,桃花桃仁复方提取物的使用浓度为0.001-100ug/ml。在另一个实施方式中,桃花桃仁复方提取物的使用浓度为0.001-50ug/ml。在另一个实施方式中,桃花桃仁复方提取物的使用浓度为0.001-10ug/ml。在另一个实施方式中,桃花桃仁复方提取物的使用浓度为0.01-10ug/ml。在另一个实施方式中,桃花桃仁复方提取物的使用浓度为0.1-10ug/ml。在另一个实施方式中,桃花桃仁复方提取物的使用浓度为1-10ug/ml。
可以将桃花桃仁复方提取物按照0.00001-100重量%的比例加到各种化妆品中。例如,桃花桃仁复方提取物的用量可以是0.00001-25重量%,例如0.0001-50重量%,例如0.001-75重量%,例如0.1-100重量%。在一个优选的实施方式中,化妆品中桃花桃仁复方提取物的用量为0.0001-50重量%。在另一个优选的实施方式中,化妆品中桃花桃仁复方提取物的用量为0.001-20重量%。
桃花桃仁复方提取物中桃花提取物和桃仁提取物的重量比可以是任意比例。例如,在一个实施方式中,桃花桃仁复方提取物中桃花提取物和桃仁提取物的重量比为3:7。
下面结合具体的实施例进一步阐述本发明。但是,应该明白,这些实施例仅用于说明本发明而不构成对本发明范围的限制。下列实施例中未注明具体条件的试验方法,通常按照常规条件,或按照制造厂商所建议的条件。除非另有说明,所有的百分比和份数按重量计。
实施例1:桃花桃仁对UVB照射造成的皮肤细胞能量损伤修复实验
实验样品制备:
称取桃花药材适量,以15倍去离子水提取→过滤,滤液浓缩,干燥成粉得到桃花提取物。
称取桃仁药材适量,以12倍乙醇浸泡过夜→过滤,滤液浓缩,干燥成粉,得到桃仁提取物。
将桃花提取物3份,另取桃仁提取物7份,按比例混合,最终制成含1%重量的复合提取液。
移取浓度为1%重量的桃花桃仁复合提取液,用PBS缓冲液稀释至10ug/ml、1ug/ml、0.1ug/ml,得到样品溶液。
实验方法:
1)细胞复苏,待生长状态良好时,进行细胞同步化处理。
2)同步化后调整浓度接种于细胞板中。第二天,置于UVB照射装置下,再加入正常培养基和含有实施例提取物的培养基,继续培养孵育。
3)经过一段时间孵育后,每孔加入试剂盒中测试液,震荡,室温放置稳定荧光,仪器检测读数。
实验结果:
记录下各孔测得的荧光单位RLU,每组3个复孔取平均值,实验结果进行t检验,P<0.05,表示差异显著;p<0.01表示差异非常显著。同时,以正常细胞100%为基准,计算各组荧光单位ATP含量百分比数值。
结果表明:桃花桃仁复方提取物具有紫外损伤修复和促进细胞能量代谢作用。即使在低浓度下(例如,0.1ug/ml的浓度下),桃花桃仁复方提取物对UVB紫外照射后的细胞能量损失具有较好的修复作用,促进ATP的合成,维持皮肤细胞能量代谢水平。如表1及图1所示,在10ug/ml时修复率可到75%以上,在0.1ug/ml的低剂量浓度下也能达到近50%的修复率。

实施例2:促进能量代谢抗衰老膏霜的制备
按照表2的成分和用量制备包含本发明所述桃花桃仁复方提取物的促进能量代谢抗衰老膏霜。

制备方法:先用1,3-丁二醇溶解桃花桃仁提取物,然后加入到水中,搅拌均匀,得到水相。将甘油硬脂酸酯柠檬酸酯、甘油硬脂酸酯、鲸蜡硬脂醇、辛酸/癸酸甘油三酯、矿物油加入油相锅中,加热至75℃-80℃,搅拌均匀,得到油相。将油相缓慢加入水相中,并不停搅拌,均匀后加入防腐剂和香精,搅拌均匀,即得。
实施例3:促进能量代谢保湿水的制备
按照表3的成分和用量制备包含本发明所述桃花桃仁复方提取物的促进能量代谢保湿水。

制备方法:先用1,3-丁二醇溶解桃花桃仁提取物,然后加入到水中,搅拌,均匀后加入防腐剂和香精,搅拌均匀,即得。

  1. 桃花桃仁复方提取物在紫外损伤修复和促进能量代谢中的用途。
  2. 桃花桃仁复方提取物作为具有紫外损伤修复和促进能量代谢作用的添加剂在化妆品中的用途。
  3. 如权利要求1或2所述的用途,其特征在于,所述桃花桃仁复方提取物的用量在包含0.00001-100重量%。
  4. 如权利要求1或2所述的用途,其特征在于,所述桃花桃仁复方提取物的用量在包含0.0001-50重量%。
  5. 如权利要求1或2所述的用途,其特征在于,所述桃花桃仁复方提取物的用量包含0.001-20重量%。
  6. 如权利要求2所述的用途,其特征在于,所述化妆品选自:洁面乳、化妆水、乳液、膏霜、啫喱、面膜、洗发水、沐浴露、护发素。
  7. 如权利要求6所述的用途,其特征在于,所述化妆品包含桃花桃仁复方提取物以及可接受的辅料。
  8. 如权利要求6所述的用途,其特征在于,所述化妆品包含0.00001-100重量%的桃花桃仁复方提取物。
  9. 如权利要求6所述的用途,其特征在于,所述化妆品包含0.0001-50重量%的桃花桃仁复方提取物。
  10. 如权利要求6所述的用途,其特征在于,所述化妆品包含0.001-20重量%的桃花桃仁复方提取物。
