Synthese von 5-Chlor-2-(4-chlorphenyl)-6-(1.3-dioxolan-2-yl)-pyrimidin-4-amin (Tabellenbeispiel-Nr. 2-7)  Synthesis of 5-chloro-2- (4-chlorophenyl) -6- (1,3-dioxolan-2-yl) -pyrimidin-4-amine (Table Example No. 2-7)
1. Synthesis of 2- (4-chlorophenyl) -6- (methoxymethyl) -pyrimidin-4-ol, (pyrimidine 1) 123 g (0.75 mol) of ethyl 4-chloroacetoacetate are added to 535 ml of a 30% sodium methylate solution and stir for another 10 minutes. Thereafter, 143 g (0.75 mol) of 4-chlorobenzene-carboximidamide hydrochloride are added and the reaction mixture is stirred under reflux for 8 hours. Then the reaction mixture is allowed to stand for 48 hours at room temperature (RT) and then concentrated to dryness. The residue is taken up in 500 ml of water, the undissolved solid is filtered off and the aqueous phase is concentrated with conc. HCl to pH3. The solid is filtered off and dried. This gives 100 g (54%) of product as a white solid.
2. After cooling, the reaction mixture is concentrated to dryness, taken up in toluene and concentrated again. The residue is taken up in 400 ml CH 2 Cl 2 and mixed carefully with H 2 O. Thereafter, the organic phase is separated and the aqueous extracted with CH 2 Cl 2. The combined organic phase is dried over Na 2 SO 4 and concentrated by rotary evaporation. The residue thus obtained is chromatographed on silica gel with heptane / ethyl acetate (1: 1) as the eluent. In this way 54 g (49%) of product are obtained as an oil.
