本発明の摂食障害治療剤は、過食症治療剤として、特に有効である。
本発明によれば、過食症患者特有の過剰な食欲を自然に抑制する一方、正常な食欲(通常の食欲)を抑制することがないので、安全に過食症状の改善が図れる。
本発明の摂食障害治療剤は、ヒト及び動物に対する安全性が高く、安心して長期間継続して日常的に摂取することができる。従って、摂食障害から二次的に生じる種々の疾患に対する治療及び予防に有効である。
本発明の有効成分であるカゼイン加水分解物は、比較的安価な乳等の原料から安定して大量に製造することができるので、安価に提供することができる。さらに、本発明の摂食障害治療剤は、飲食品や飼料の形態として提供することもできる。
本発明の摂食障害治療剤は、満腹感によって食事摂取量を抑制するものではないため、摂取の時期が食事の前後に限定されず、自由である。また、一定期間、効果が持続するので、毎日摂取する必要もない。
本発明の摂食障害治療剤は、摂取するだけで自然に過剰な食欲が抑制するものであるため、一般的な食事療法で問題となる精神的なストレスがほとんどない。
本発明のカゼイン加水分解物の出発物質は、乳蛋白質のカゼインである。カゼインは、ほ乳類(例えば、牛、羊、山羊)の乳由来のカゼインを主成分とするものであれば使用することができ、特に牛乳由来のカゼインを好適に使用することができ、加水分解によってαs2-カゼイン由来の加水分解物を生じるものであれば、如何なるカゼインも使用することができる。具体的には、市販の各種カゼインやカゼイネート、例えば、乳酸カゼイン、硫酸カゼイン、塩酸カゼイン、ナトリウムカゼイネート、カリウムカゼイネート、カルシウムカゼイネート、マグネシウムカゼイネート又はこれらの任意の混合物等が好ましい。また、牛乳、脱脂乳、全脂粉乳、脱脂粉乳から常法により精製して得られるカゼインを原料として使用することができる。好ましい実施の一態様において、カゼインの加水分解では、カゼインを溶解してカゼイン溶液にしたものに酵素等を添加して行われる。したがって、好ましい実施の一態様において、本発明のカゼインはカゼイン水溶液であることが好ましい。
本発明のカゼイン加水分解物は、以下のように製造することができる。
カゼイン加水分解物の製造方法においては、まず、カゼインを水に分散し、溶解してカゼイン溶液とする。カゼイン溶液のカゼイン濃度は特に制限されないが、通常、蛋白質換算で5~15質量%の濃度範囲が効率及び操作性の点から望ましい。
なお、加水分解率の算出方法は、ケルダール法(日本食品工業学会編、「食品分析法」、第102頁、株式会社光琳、昭和59年)により試料の全窒素量を測定し、ホルモール滴定法(満田他編、「食品工学実験書」、上巻、第547ページ、養賢堂、1970年)により試料のホルモール態窒素量を測定し、これらの測定値から分解率を次式により算出する。
加水分解率(%)=(ホルモール態窒素量/全窒素量)×100
本発明の摂食障害治療剤は、過食症に対して特に有効であり、過食症治療剤として使用することができる。本発明の治療剤は、過食症患者の過剰な食欲を抑制することにより、食事摂取量を正常の範囲に戻して過食症を治療することができる。本発明の摂食障害治療剤の投与対象は、摂食障害患者であり、好ましくは過食症患者であり、さらに好ましくは拒食症への移行が懸念される過食症患者である。
結合剤としては、例えば、上記賦形剤の他、ゼラチン;ポリビニルピロリドン;マグロゴール等が挙げられる。
崩壊剤としては、例えば、上記賦形剤の他、クロスカルメロースナトリウム、カルボキシメチルスターチナトリウム、架橋ポリビニルピロリドン等の化学修飾されたデンプン又はセルロース誘導体等が挙げられる。
滑沢剤としては、例えば、タルク;ステアリン酸;ステアリン酸カルシウム、ステアリン酸マグネシウム等のステアリン酸金属塩;コロイドシリカ;ビーガム、ゲイロウ等のワックス類;硼酸;グリコール;フマル酸、アジピン酸等のカルボン酸類;安息香酸ナトリウム等のカルボン酸ナトリウム塩;硫酸ナトリウム等の硫酸塩類;ロイシン;ラウリル硫酸ナトリウム、ラウリル硫酸マグネシウム等のラウリル硫酸塩;無水珪酸、珪酸水和物等の珪酸類;デンプン誘導体等が挙げられる。
安定剤としては、例えば、メチルパラベン、プロピルパラベン等のパラオキシ安息香酸エステル類;クロロブタノール、ベンジルアルコール、フェニルエチルアルコール等のアルコール類;塩化ベンザルコニウム;無水酢酸;ソルビン酸等が挙げられる。
矯味矯臭剤としては、例えば、甘味料、酸味料、香料等が挙げられる。
注射剤用溶剤としては、例えば、水、エタノール、グリセリン等が挙げられる。
かかる飼料は、例えば、カゼイン加水分解物に、トウモロコシ、小麦、大麦、ライ麦、マイロ等の穀類;大豆油粕、ナタネ油粕、ヤシ油粕、アマニ油粕等の植物性油粕類;フスマ、麦糠、米糠、脱脂米糠等の糠類;コーングルテンミール、コーンジャムミール等の製造粕類;魚粉、脱脂粉乳、ホエー、イエローグリース、タロー等の動物性飼料類;トルラ酵母、ビール酵母等の酵母類;第三リン酸カルシウム、炭酸カルシウム等の鉱物質飼料;油脂類;単体アミノ酸;糖類等を配合することにより製造できる。
飼料の形態は、日常的に給餌が可能な形態が好ましい。具体例としては、ペットフード、家畜飼料、養魚飼料等が挙げられる。
実施例の電気泳動(二次元電気泳動)は以下のように行うことができる。
(1)1次元目
始めに、試料を5mg/mlとなるように膨潤用緩衝液(例えばインビトロジェン社製)に溶解し、市販の等電点電気泳動用ストリップゲル(例えばインビトロジェン社製)をこの溶液内で膨潤させて一晩静置した。一晩静置したゲルを175V、20分、175Vから2000Vまでの電圧勾配を45分、更に2000V、30分で泳動する。泳動の終了したストリップゲルは染色液に浸して染色する。
(2)2次元目
1次元目の泳動が終了したストリップゲルを2次元目用のゲル(インビトロジェン社製)にアプライし、200Vの定電圧で35分泳動する。泳動終了後、CBB染色液(インビトロジェン社製)を用いて染色する。
本明細書では、飛行時間型質量分析装置をTOF/MSと記載することがある。
実施例のTOF/MSは、例えば以下のように行うことができる。
上記の電気泳動法によって、2次元電気泳動をした後、各々スポットとして確認されるペプチドについてゲル内消化を行い、個々のペプチドをTOF/MS測定機(ブルカーダルトニクス社製)にて同定する。
1)カゼイン溶液を調製し、殺菌する工程
レンネットカゼイン(フォンテラ社製、蛋白質含量80%)123gを精製水2300gに加え、膨潤させた。これに濃塩酸(和光純薬社製、濃度36.5%)を加えてpHを2.8にした後、85℃、10分保持して殺菌した。
2)カゼイン溶液を消化する工程
殺菌したカゼイン溶液を42℃に冷却してからペプシン(SIGMA社製)20mgを加え、2時間消化した。
3)酵素を失活させる工程
消化後、再度、温度を85℃にして10分保持し、ペプシンを失活させた。
4)遠心分離工程
ペプシンを失活させた溶液を、遠心分離機(日立製作所社製)10,000rpm(20,000G)で20分遠心分離を行い、上清画分(可溶性画分)と沈殿画分(不溶性画分)に分離した。溶液の温度は10℃であった。
5)乾燥工程
遠心分離後の上清画分を凍結乾燥し、カゼイン加水分解物24gを得た。
一方、遠心分離後の沈殿画分を凍結乾燥し、沈殿画分乾燥粉末92gを得た。
1)カゼイン溶液を調製し、殺菌する工程
レンネットカゼイン(フォンテラ社製、蛋白質含量80%)10gを精製水200gに加え、膨潤させた。これに濃塩酸(和光純薬社製、濃度36.5%)を加えてpHを2.8にした後、85℃、10分保持して殺菌した。
2)カゼイン溶液を消化する工程
殺菌したカゼイン溶液を42℃に冷却してから消化酵素としてペプシン(BIOPHEDEX社製)4.88mgを加え、2時間消化した。
3)酵素を失活させる工程
消化後、再度、温度を85℃にして10分保持し、ペプシンを失活させた。
4)分離工程
ペプシンを失活させた溶液を、MF膜(旭化成社製)に通液し、透過液(可溶性画分)と濃縮液(不溶性画分)に分離した。
5)乾燥工程
透過液を凍結乾燥し、カゼイン加水分解物2.2gを得た。
実施例1の方法で得られたカゼイン加水分解物150gに、ラクチュロース粉末(森永乳業社製)100g、マルツデキストリン(松谷化学工業社製)635g、脱脂粉乳(森永乳業社製)85g、ステビア甘味料(三栄源エフ・エフ・アイ社製)1g、ヨーグルトフレーバー(三栄源エフ・エフ・アイ社製)5g、グリセリン脂肪酸エステル製剤(理研ビタミン社製)24gの各粉末を添加して均一に混合し、打錠機(畑鉄鋼所社製)を使用して、1錠当り0.5gとし、12錠/分打錠速度、9.8KPaの圧力で前記混合粉末を連続的に打錠し、カゼイン加水分解物を含有するタブレット(過食症治療剤)1800錠(約900g)を製造した。タブレット1錠当りのカゼイン加水分解物は約15質量%であった。
ホエー蛋白加水分解物(森永乳業社製)10kg、デキストリン(昭和産業社製)36kg、及び少量の水溶性ビタミンとミネラルを水200kgに溶解して、水相をタンク内に調製した。これとは別に、大豆サラダ油(太陽油脂社製)3kg、パーム油(太陽油脂社製)8.5kg、サフラワー油(太陽油脂社製)2.5kg、レシチン(味の素社製)0.2kg、脂肪酸モノグリセリド(花王社製)0.2kg、及び少量の脂溶性ビタミンを混合溶解して油相を調製した。タンク内の水相に油相を添加し、攪拌して混合した後、70℃に加温し、更に、ホモゲナイザーにより14.7MPaの圧力で均質化した。次いで、90℃で10分間殺菌した後、濃縮し、噴霧乾燥して、中間製品粉末約59kgを調製した。この中間製品粉末50kgにショ糖(ホクレン社製)6.8kg、アミノ酸混合粉末(味の素社製)167g、及び実施例1の方法で得られたカゼイン加水分解物1kgを添加し、均一に混合して、カゼイン加水分解物を含有する経腸栄養食粉末約56kgを製造した。
本試験は、本発明品である実施例1のカゼイン加水分解物を同定することを目的とした。
[試験方法]
(1)試料の調製
実施例1で遠心分離後の上清画分を凍結乾燥した本発明のカゼイン加水分解物(試料1、以下、本発明品ということがある)、実施例1で遠心分離後の沈殿画分の凍結乾燥したもの(試料2、以下、沈殿画分ということがある)及びコントロールとして、実施例1でカゼインとして使用したレンネットカゼイン(試料3、フォンテラ社製)を試料とした。
(2)電気泳動
(1次元目)
上記試料をそれぞれ5mg/mlとなるように膨潤用緩衝液(インビトロジェン社製)に溶解し、市販の等電点電気泳動用ストリップゲル(インビトロジェン社製)をこの溶液内で膨潤させて一晩静置した。一晩静置したゲルを175V、20分、175Vから2000Vまでの電圧勾配を45分、更に2000V、30分で泳動した。電気泳動の終了したストリップゲルは染色液に浸して染色した。
(2次元目)
上記(1)で実施した1次元目の電気泳動が終了したストリップゲルを2次元目用のゲル(インビトロジェン社製)にアプライし、200Vの定電圧で35分泳動した。電気泳動終了後、CBB染色液(インビトロジェン社製)を用いて染色した。
(3)上清画分についてのスポットの同定
2次元電気泳動後、各々スポットとして確認されるペプチドについてゲル内消化を行い、個々のペプチドをTOF/MS測定機(ブルカーダルトニクス社製)にて同定した。
試料1(本発明品)には分子量20kDaよりも大きい分子はほとんど消失していた。また、試料2(沈殿画分)では、試料3(レンネットカゼイン)で確認されるカゼインミセルや重合体に由来すると思われる高分子化合物はほぼ消失したが、分子量10~30kDaで検出されるカゼイン画分が、蛋白質のまま残存していた。分子量分布の結果を表1に示す。表中に記載された数値は、それぞれの画分の全体に占める百分率を意味する。
ここで、αs2-カゼインの分子量は、25kDaであることが知られている(山内邦男、他2名「牛乳成分の特性と健康」、株式会社光生館、1993年6月1日初版第1刷発行、p.8参照)。従って、本発明のカゼイン加水分解物の有効成分は、分子量が25kDa以下であることが明らかとなった。
また、本結果から本発明品の分子量分布は、50%以上の成分が1~20kDaであることが明らかとなった。
一方、沈殿画分は、β-カゼインとαs1-カゼインを由来とするカゼイン加水分解物が大部分を占めることが明らかとなった。
なお、20,000Gで20分の条件で遠心分離して得られた画分(試料1)の他に、15,000Gで60分の条件で遠心分離して得られた画分を同様に分析したところ、上記試料1と同様の分子量の分子を含むものとなっていた。
本試験は、本発明のカゼイン加水分解物のアミノ酸分析を行うことを目的とした。
[試験方法]
試験例1で得られた本発明のカゼイン加水分解物(試料1)及び沈殿画分(試料2)を用いた。各試料を塩酸で加水分解した後、ダンシルクロライド(シグマ社製)で標識した。この標品を、C18カラム(Waters社製)で分離し、得られたピークを蛍光検出器(日立製作所社製)にて検出して、解析した。
実施例1のカゼイン加水分解物(試料1)及び沈殿画分(試料2)は、いずれも原料のレンネットカゼインと同じアミノ酸組成を有していた。
本試験は、本発明のカゼイン加水分解物の過食症治療効果を確認することを目的とした。
(1)試料の調製
コントロール:注射用蒸留水をそのまま使用した。
本発明品:実施例1の方法により作製した、本発明品であるカゼイン加水分解物を注射用蒸留水で5mg/mlとなるように希釈した。
沈殿画分:試験例1の試料2を注射用蒸留水で5mg/mlとなるように希釈した。
全カゼイン加水分解物:実施例1で用いたレンネットカゼイン(フォンテラ社製、蛋白質含量80%)を実施例1と同様の方法により、ペプシンで消化後、遠心分離をせずにそのまま取り出して、注射用蒸留水で5mg/mlとなるように希釈した。
GMP(グリコマクロペプチド):市販のGMP(MGニュートリショナルズ社製)を注射用蒸留水で5mg/mlとなるように希釈した。
αs-カゼイン:市販のαs-カゼイン(SIGMA社製)を注射用蒸留水で5mg/mlとなるように希釈した。
κ-カゼイン:市販のκ-カゼイン(SIGMA社製)を注射用蒸留水で5mg/mlとなるように希釈した。
全カゼイン:市販の乳酸カゼイン(フォンテラ社製)を注射用蒸留水で5mg/mlとなるように希釈した。
(2)飼料
市販の乳由来蛋白質を含まない飼料(日本農産社製、商品名:MRストック)を使用した。また、飲料水は水道水とした。
(3)試験動物
過食症のモデル動物としてKK-Ayマウス(オス、5週齢、日本クレア社から購入)を使用した。
上記の飼料による10日間の予備飼育の後、体重と摂餌量がほぼ同等となるように一群を8匹とする8群に分けた。各群には、飼料と水を自由摂取させた。また、一週間のうち5日間を、1日1回のペースでゾンデを用いて試料を0.5ml/匹ずつ投与した。試験期間は5週間とし、週1回の割合で摂餌量を測定した。
各群については、以下の通りである。

表2に投与開始から5週間後の各群の相対摂餌量を示す。
相対摂餌量とは、コントロールの平均摂餌量を1としたときの各群の平均摂餌量である。本発明品では、コントロールに比べ、有意に摂餌量が減少した。一方、沈殿画分では、摂餌量の増加傾向が見られたが、有意な差ではなかった。また、κ-カゼイン由来のGMPでは、摂餌量の減少傾向が見られたが、コントロールと比較して有意な効果ではなかった。また、κ-カゼインでは、摂餌量の減少傾向が見られたが、有意な効果ではなかった。αs-カゼインでは、摂餌量の増加傾向が見られたが、有意な効果ではなかった。全カゼインでは、摂餌量の増加傾向が見られたが、有意な効果ではなかった。全カゼイン加水分解物では、摂餌量の減少傾向が見られたが、有意な効果ではなかった。このことから、全カゼインを加水分解物し、さらに沈殿画分を除去して、上清画分(水溶性画分)を分離すると、分離前には認められなかった有意な過食症治療効果が出現することが明らかとなった。
これらの結果から、本発明品(2群)が過食症治療効果を有していることが明らかとなった。また、本発明のカゼイン加水分解物が、他のカゼイン画分や、全カゼイン加水分解物と対比して、顕著な過食抑制効果を有することが明らかとなった。
本試験は、本発明のカゼイン加水分解物が過食症における血糖値低減効果を有することを確認することを目的とした。
(1)試料の調製
試験例3の試料と同じものを使用した。
(2)飼料
市販の乳由来蛋白質を含まない飼料(日本農産社製、MRストック)を使用した。また、飲料水は水道水とした。
(3)試験動物
過食症のモデル動物としてKK-Ayマウス(オス、5週齢、日本クレア社から購入)を使用した。
上記の飼料による10日間の予備飼育の後、体重と摂餌量がほぼ同じになるように一群を8匹とする8群に分けた。各群に飼料と水を自由摂取させた。また、一週間のうち5日間を、1日1回、ゾンデを用いて試料を0.5ml/匹ずつ投与した。試験期間は5週間とし、週1回の割合で自己検査用グルコースキット(Johnson & Johnson社製)を用いて血糖値を測定した。
各群の試料については、以下の通りである。

表3に投与開始から5週間後の相対血糖値を示す。相対血糖値とは、コントロールの平均血糖値を1としたときの各群の平均血糖値である。本発明品は、コントロールと比較して有意に血糖値が低減した。全カゼイン加水分解物、全カゼイン及びκ-カゼインでは血糖値の低減傾向が見られたが、有意な効果は認められなかった。また、沈殿画分、GMP、αs-カゼインでは血糖値の増加傾向が見られたが、有意な効果は認められなかった。
更に、この血糖値の低減効果と過食症治療効果との相関性について検定した。Pearsonの相関係数は0.593で、この相関係数はp<0.01で有意であった。
本試験は、本発明のカゼイン加水分解物を、過食症状のない正常動物に投与したときの食欲及び血糖値に与える影響を確認することを目的とした。
(1)試料の調製
コントロール:注射用蒸留水をそのまま使用した。
本発明品:実施例1の方法で製造したカゼイン加水分解物を注射用蒸留水で5mg/mlとなるように希釈した。
(2)試料
市販の乳由来蛋白質を含まない飼料(日本農産社製、MRストック)を使用した。また、飲料水は水道水とした。
(3)試験動物
KK-AyマウスのコントロールであるKKマウス(オス、5週齢、日本クレア社より購入)を過食症状のないモデル動物として用いた。
上記の飼料による10日間の予備飼育の後、体重と摂餌量が同じになるように一群8匹とする二群に分けた。そして、一週間のうち5日間を1日1回、ゾンデを用いて試料を0.5ml/匹ずつ投与した。試験期間は5週間とした。試験期間中、週1回の割合で摂餌量及び血糖値を測定した。血糖値の測定は、自己検査用グルコースキット(Johnson & Johnson社製)を用いた。
表4に投与開始から5週間後の相対摂餌量及び相対血糖値の結果を示す。Studentのt検定を行ったが、本発明品にはKKマウスに対する摂餌量抑制及び血糖値の上昇抑制効果を認めなかった。即ち、本願発明品は、過食症状のない正常動物に対しては影響のないことが明らかとなった。このことから、本発明品が、適切に発揮されている通常の食欲(正常な食欲)を抑制することがなく、安全に摂取できることを示すものである。
本試験は、過食症状を有する動物(過食動物)の摂餌量のうち、正常な食欲に起因する摂餌量と過剰な食欲に起因する摂餌量とを特定することを目的とした。
(1)飼料
市販の乳由来蛋白質を含まない飼料(日本農産社製、MRストック)を使用した。また、飲料水は水道水とした。
(2)試験動物
過食症のモデル動物としてKK-Ayマウス(オス、5週齢、日本クレア社から購入)、過食症状のないモデル動物としてKK-AyマウスのコントロールであるKKマウス(オス、5週齢、日本クレア社から購入)、過食症のモデル動物としてZDFラット(オス、ホモ、5週齢、日本チャールズリバー社から購入)、及び過食症状のないモデル動物としてZDFラットのコントロールであるZucker leanラット(オス、5週齢、日本チャールズリバー社から購入)を使用した。
(試験動物)
マウス:
(i) KK-AyマウスのコントロールであるKKマウス(以下、コントロール1と記載することがある)
(ii) KK-Ayマウス
ラット:
(iii) ZDFラットのコントロールであるZucker lean ラット(以下、コントロール2と記載することがある)
(iv) ZDFラット
上記のマウス及びラットについて飼料及び飲料水を自由摂取させた。
マウスについては、入荷後10日間の馴化期間後の摂餌量を比較した。また、ラットについては、入荷後、13週間飼育後の摂餌量を比較した。ラットは13週間飼育後としたが、これはZDFラットが過食症状を示すまでに相当期間を必要とするためである。
表5に5週間後の平均摂餌量の結果を示す。コントロール1はKKマウス、コントロール2はZucker leanラットを意味する。KK-Ayマウスはコントロールの1.22倍の飼料を摂取し、ZDFラットはコントロールの1.23倍の飼料を摂取した。すなわち、過食マウス及び過食ラットのいずれにおいても、過食動物の摂餌量が対照の正常動物の摂餌量と比較して2割程度増加していることが判明した。このことから、過食症における過剰な食欲は、通常の食欲による摂餌量に対して2割程度と見ることができる。
本試験は、本発明の治療剤の用量依存性を確認することを目的とした。
(1)試料の調製
コントロール:注射用蒸留水を使用した。
高用量試料:実施例1のカゼイン加水分解物を注射用蒸留水で5mg/mlとなるように希釈した。
低用量試料:実施例1より作製したカゼイン加水分解物を注射用蒸留水で0.5mg/mlとなるように希釈した。
(2)飼料
市販の乳由来蛋白質を含まない試料(日本農産社製、MRストック)を使用した。また、飲料水は、水道水とした。
(3)試験動物
過食症のモデル動物としてKK-Ayマウス(オス、5週齢日本クレア社から購入)を使用した。
一日1回ずつ週5日、上記試料を0.5ml/匹で投与した。投与期間は5週間とし、飼料及び飲料水は自由摂取とした。
(5)評価方法
KK-Ayマウスの摂餌量と血糖値の測定を毎週1回行った。
血糖値測定は、毎週1回、自己検査用グルコースキット(Johnson & Johnson社製)を用いて測定した。
表6に、投与開始から5週間後の各群の相対摂餌量及び相対血糖値を示す。高用量試料で有意な摂餌量抑制及び血糖値の上昇抑制が認められた。一方、低用量試料では有意な効果が認められなかった。
従って、本発明の過食症治療剤は用量依存性を有することが明らかとなった。
なお、高用量試料群での投与量は、1日当り2.5mg(5mg/ml×0.5ml)であり、投与対象であるKK-Ayマウスの平均体重は43g程度であった。
従って、本発明のカゼイン加水分解物は、毎週、体重当り290mg/kg以上の摂取をすれば、上記効果が得られることが明らかになった。
本試験は、本発明のカゼイン加水分解物を摂取してから効果が発現するまでの期間、及び効果の持続期間を検討することを目的とした。
(1)試料の調製
コントロール:注射用蒸留水を使用した。
本発明品:実施例1で調製した本発明のカゼイン加水分解物を注射用蒸留水で5mg/mlとなるように希釈した。
(2)試験動物
過食症のモデル動物としてZDFラット(オス、5週齢、日本チャールズリバー社から購入)を使用した。ZDFラットは過食症状を発症させるため、12週間予備飼育を行った。
(3)飼料
市販の乳由来蛋白質を含まない飼料(日本農産社製、MRストック)を使用した。また、飲料水は水道水とした。
以下の観察期間の経過後、飼料を投与した。
[観察期間1]
特に何もせず、1週間摂餌量を観察した。
[観察期間2]
開始時に摂餌量を計量後、注射用蒸留水を投与し、1週間摂餌量を観察した。そして、観察期間1の摂餌リズムと比較して、注射用蒸留水の経口ゾンデ投与によっては、摂餌量パターンに変化が見られないことを確認した。観察期間2の経過後、ZDFラットの体重及び血糖値を測定し、摂餌量と併せて均等となるように、コントロール群と本発明品群の2群に分けた。1群当りのZDFラットは8匹とした。
[投与]
観察期間2の後、本発明品群には本発明品を調製した試料を、対照試料群には注射用蒸留水を、1日1回ずつ週5日、毎回2ml投与して、2週間摂餌量を観察した。
試験期間中、週5回摂餌量を測定した。
図1に、観察期間中の摂餌量の測定値を示す。図中、試料の投与及び摂餌量を測定しなかった土曜日、日曜日は省略して記載した。また、注射用水は注射用蒸留水を示す。
観察期間2と観察期間3との間の「sample」は各群に本発明品または注射用蒸留水を投与したことを示す。
観察期間1において、ZDFラットの毎日の摂餌量の変化は±2g程度であることを確認した。観察期間2では注射用蒸留水の投与以降も摂餌パターンに影響はなく、経口ゾンデ投与によるストレスが影響しないことを確認した。
本発明のカゼイン加水分解物の投与後4日目から、本発明品群ではコントロール群と比較して明らかな摂餌量の低下を認めた。そして、驚くべきことに、この効果は投与後9日目まで継続した。
この結果から、本発明のカゼイン加水分解物の摂餌量抑制効果は即時的なものではなく、4日程度経過してから効果を発揮し、その効果は6日間程度持続することが明らかとなった。
この結果は、本発明品は、即時的に膨満感を誘導するものではないことを示す。そのため、本発明品の摂取のタイミングは食事の前後や食間に限定されることがない。さらには、一定期間過食抑制効果が発揮されるので、毎日摂取する必要もない。
本試験は、本発明のカゼイン加水分解物の有効成分の分子量の特定を目的とした。
(1) 試料の調製
下記の試料を調製した。
コントロール:注射用蒸留水を使用した。(コントロール)
本発明品:実施例1で調製したカゼイン加水分解物(本発明品)を注射用蒸留水で5mg/mlの濃度に調製した。
再消化試料:実施例1で調製したカゼイン加水分解物(本発明品)33gにペプシンを66mg加え、pH2.8及び42℃で2時間消化した後、凍結乾燥した。この凍結乾燥標品を注射用蒸留水で5mg/mlの濃度に調製した。
なお、再消化試料は、電気泳動により分子量が1KDa未満であることを確認した。
(2)試験動物
過食症のモデル動物としてKK-Ayマウス(オス、5週齢、日本クレア社から購入)を使用した。
(3)飼料
市販の乳由来蛋白質を含まない試料(日本農産社製、MRストック)を使用した。また、飲料水は、水道水とした。
体重と摂餌量がほぼ同等となるように一群8匹とする3群に分け、一週間のうち5日間を、1日1回、ゾンデを用いて各試料を0.5ml/匹ずつ投与した。
投与期間は5週間とし、飼料及び飲料水は自由摂取とした。
投与開始から5週間後の各群の相対摂餌量を比較した。本発明品では、コントロールと比較して有意に摂餌量が減少した。
一方、再消化試料はコントロールと差異が見られず、有意な効果が認められなかった。再消化試料が分子量1kDa以下の成分であることから、本発明の過食症治療剤の有効成分は分子量が1kDa以上の成分であることが明らかとなった。
以上の結果から、本発明の過食抑制効果を有する成分は、分子量1kDa~37kDaの範囲に存在することが明らかとなった。本結果と、試験例1の結果とを合わせると、本発明の過食抑制効果を有する成分は、分子量1kDa~25kDaの範囲に存在することが明らかとなった。
また、本発明の治療剤の有効成分である本発明のカゼイン加水分解物は、乳等の原料から大量に製造することができるので、安価に提供することができる。
Claims (14)
- ペプシンによるカゼインの加水分解物の水溶性画分を有効成分として含有してなる、摂食障害治療剤。
- ペプシンによるカゼインの加水分解物の水溶性画分が、分子量1kDa~25kDaのペプチド断片を有効成分として含んでなる画分である、請求項1に記載の摂食障害治療剤。
- ペプシンによるカゼインの加水分解物が、カゼイン1質量部に対して、ペプシン1/10000質量部~1/1000質量部を添加して加水分解された加水分解物である、請求項1~2の何れかに記載の摂食障害治療剤。
- 水溶性画分が、ペプシンによるカゼインの加水分解物を、デカンテーション又は遠心分離によって画分して得られた上清画分である、請求項1~3の何れかに記載の摂食障害治療剤。
- 摂食障害治療剤が、過食症における一日の食事摂取量を、投与開始から5週間後に、投与開始時と比較して、10%以上低下させる摂食障害治療剤である、請求項1~4の何れかに記載の摂食障害治療剤。
- 摂食障害が、過食症である、請求項1~5の何れかに記載の摂食障害治療剤。
- カゼインをペプシンで加水分解して、ペプシンによるカゼインの加水分解物を得る工程、
ペプシンによるカゼインの加水分解物から水溶性画分を得る工程、
を含んでなる、摂食障害治療剤を製造する方法。 - カゼインをペプシンで加水分解して、ペプシンによるカゼインの加水分解物を得る工程が、
カゼイン1質量部に対して、ペプシン1/10000質量部~1/1000質量部を添加して、カゼインをペプシンで加水分解して、ペプシンによるカゼインの加水分解物を得る工程、
である、請求項7に記載の方法。 - ペプシンによるカゼインの加水分解物から水溶性画分を得る工程が、

- 請求項1~6の何れかに記載された摂食障害治療剤を投与することにより、摂食障害を治療する方法。
- 請求項1~6の何れかに記載された摂食障害治療剤を製造するための、ペプシンによるカゼインの加水分解物の水溶性画分の使用。
- 摂食障害の治療に用いられる、ペプシンによるカゼインの加水分解物の水溶性画分。
