更に、本発明は、式(I)の2H-クロメン化合物若しくはその誘導体又はその塩の有効量を患者に投与することを含む、S1P1の関与する望ましくないリンパ球浸潤によって引き起こされる疾患、殊に、ヒト又は動物における臓器、骨髄又は組織の移植時の拒絶もしくは移植片対宿主病、又は多発性硬化症の予防若しくは治療方法に関する。また、本発明は、S1P1の関与する望ましくないリンパ球浸潤によって引き起こされる疾患、殊にヒト又は動物における臓器、骨髄又は組織の移植時の拒絶もしくは移植片対宿主病、又は多発性硬化症の予防若しくは治療への式(I)の2H-クロメン化合物若しくはその誘導体又はその塩の使用、並びに、S1P1の関与する望ましくないリンパ球浸潤によって引き起こされる疾患、殊にヒト又は動物における臓器、骨髄又は組織の移植時の拒絶もしくは移植片対宿主病、又は多発性硬化症の予防若しくは治療に使用するための、式(I)の2H-クロメン化合物若しくはその誘導体又はその塩をも包含する。
本明細書において、「ハロゲン」は、F、Cl、Br、及びIを意味する。好ましくはF及びClが挙げられる。
「含窒素単環ヘテロアリール」とは、環を構成する原子の一つが必ずNであり、更に環を構成する原子としてN,S及びOから選択されるヘテロ原子を1~2個有していてもよい単環ヘテロアリールを意味する。別の態様としては5~6員環であり、また別の態様としてはピリジル、ピロリル、ピラジニル、ピリミジニル、ピリダジニル、イミダゾリル、トリアゾリル、テトラゾリル、チアゾリル、ピラゾリル、イソチアゾリル、オキサゾリル、イソオキサゾリル、チアジアゾリル、オキサジアゾリル等が挙げられる。別の態様としては、5員環であり、また別の態様としては、ピロリル及びイミダゾリルである。
(1) YがOであり、Qが-C(R2B)=C(R2C)-であり、mが0である2H-クロメン化合物又はその塩。
(2) R4A及びR4Bが、それらが結合するNと一体となって、G群から選択される基で置換され、更に低級アルキル又はハロゲンで置換されていてもよい、アゼチジニル、ピロリジニル、ピペリジニル及びテトラヒドロピリジルから選択される環状アミノである2H-クロメン化合物又はその塩。
(3) G群で示される基が、-C(=O)OH又は-C(=O)NHS(=O)2CH3である、2H-クロメン化合物又はその塩。
(4) Aが、1乃至3個の同一又は異なったR1で置換されていてもよいフェニル、ピリジル又はチエニルである、2H-クロメン化合物又はその塩。
(5) Lが、-(低級アルキレン)-O-、低級アルケニレン又は低級アルキニレンである、2H-クロメン化合物又はその塩。
(6) R2Aが-H又は低級アルキルであり、R2Bが-Hであり、R2Cが-H又はハロゲンであり、R3が-H又はハロゲンであり、R1がハロゲン、低級アルキル、ハロゲノ低級アルキル、フェニル、ピロリル、シクロアルキル、-O-(低級アルキル)、又は-O-(ハロゲノ低級アルキル)であり、かつ、Lが-CH2-O-、-CH=CH-又は3-ブチニレンである、2H-クロメン化合物又はその塩。
(7) R4A及びR4Bが、それらが結合するNと一体となって、-C(=O)OHで置換された、ピペリジニル又はテトラヒドロピリジルであり、Lが-CH2-O-であり、R2Aが-Hであり、R2Bが-Hであり、R2Cが-H又はハロゲンであり、R3が-Hであり、Aが、2個の同一又は異なったR1で置換された、フェニル又はピリジルであり、ここでR1はハロゲン、ハロゲノ低級アルキル、-O-(低級アルキル)又は-O-(ハロゲノ低級アルキル)である、2H-クロメン化合物又はその塩。
(8) R4A及びR4Bが、それらが結合するNと一体となって、-C(=O)OHで置換されたピペリジニルであり、Aが2個の同一又は異なったR1で置換されたフェニルである、2H-クロメン化合物又はその塩。

式(I)の化合物及びその塩は、その基本構造あるいは置換基の種類に基づく特徴を利用し、種々の公知の合成法を適用して製造することができる。その際、官能基の種類によっては、当該官能基を原料から中間体へ至る段階で適当な保護基(容易に当該官能基に転化可能な基)に置き換えておくことが製造技術上効果的な場合がある。このような保護基としては、例えば、ウッツ(P.G.M.Wuts)及びグリーン(T.W.Greene)著、「Greene’s Protective Groups in Organic Synthesis(第4版、2006年)」に記載の保護基等を挙げることができ、これらの反応条件に応じて適宜選択して用いればよい。このような方法では、当該保護基を導入して反応を行なったあと、必要に応じて保護基を除去することにより、所望の化合物を得ることができる。
また、式(I)の化合物のプロドラッグは、上記保護基と同様、原料から中間体へ至る段階で特定の基を導入、あるいは得られた式(I)の化合物を用いてさらに反応を行なうことで製造できる。反応は通常のエステル化、アミド化、脱水等、当業者に公知の方法を適用することにより行うことができる。
以下、式(I)の化合物の代表的な製造法を説明する。各製法は、当該説明に付した参考文献を参照して行うこともできる。なお、本発明の製造法は以下に示した例には限定されない。
Step1の工程は、還元的アミノ化である。化合物(A)と化合物(B)とを等量若しくは一方を過剰量用い、これらの混合物を、還元剤の存在下、反応に不活性な溶媒中、-45℃から還流条件下、特に、0℃~室温において、通常0.1時間~5日間撹拌する。溶媒の例としては、MeOH、EtOH等のアルコール類;Et2O、THF、ジオキサン、DME等のエーテル類;DCM、DCE又はクロロホルム等のハロゲン化炭化水素類;及びこれらの混合溶媒が挙げられる。還元剤としては、NaBH3CN、NaBH(OAc)3、NaBH4等が挙げられる。モレキュラーシーブス等の脱水剤、又は酢酸、塩酸、チタニウム(IV)イソプロポキシド錯体等の酸存在下で反応を行うことが好ましい場合がある。反応中間体であるイミンが、安定な中間体として単離できる場合があり、このイミン中間体を還元すれば化合物(I)を得ることができる。また、前記還元剤の代わりに、MeOH、EtOH、EtOAc等の溶媒中、酢酸、塩酸等の酸の存在下又は非存在下で、還元触媒(例えば、パラジウム炭素、ラネーニッケル等)を用いて反応を行うこともできる。この場合、反応を常圧から50気圧の水素雰囲気下で、冷却から加熱条件下で行う。

化合物(A-1)は、化合物(D)から薗頭反応により製造できる。
Step3-1は、トリフラート化である。化合物(E)は、化合物(D)に無水トリフルオロメタンスルホン酸を作用させることにより製造できる。通常反応を妨げない溶媒、例えば、ハロゲン化炭化水素類中で、ピリジン、TEA、DIPEA等の有機塩基の存在下、-10℃から氷冷条件下に行う。なお、有機塩基は溶媒を兼ねてもよい。
Step3-2は、いわゆる薗頭反応である。化合物(A-1)は、化合物(E)に触媒量のPd(0)触媒及び塩基を加え、末端アセチレンを反応させることにより製造できる。反応系内にヨウ化銅を添加するのが、反応を円滑に進行させる上で有利な場合がある。溶媒の例としては、エーテル類;トルエン、キシレン等の芳香族炭化水素類;DMF、DMSO、EtOAc;及びこれらの混合溶媒が挙げられる。例えば、TEA、ピロリジン等の塩基が溶媒を兼ねていてもよい。反応温度は、室温から還流下で行うことができる。

Step4-1は、ケトンの還元反応である。反応に不活性な溶媒中、冷却下から加熱下、好ましくは-20℃から80℃で、化合物(G)を等量若しくは過剰量の還元剤で、通常0.1時間~3日間処理する。溶媒の例としては、エーテル類;アルコール類;芳香族炭化水素類;DMF、DMSO、EtOAc及びこれらの混合溶媒が挙げられる。還元剤としては、NaBH4若しくはDIBAL等のヒドリド還元剤、ナトリウム、亜鉛、鉄等の金属還元剤、その他、下記文献中の還元剤が好適に用いられる。

Step4-2は、脱水反応である。通常、原料を濃硫酸中で加温条件下で撹拌した後、流出液体が出なくなるまで蒸留を続ける。
Step4-3は、ホルミル化である。化合物(A-2)は、化合物(J)とホルムアミド誘導体との反応により得ることができる。ここで、ホルムアミド誘導体とは、ホルムアミドの窒素原子に同一又は異なった低級アルキル又はアリールが結合しているホルムアミド化合物を意味する。ホルムアミド誘導体とPOCl3の反応により調製されたVielsmeier錯体に対して、芳香環が求核置換して、インモニウム塩が生成する。これを塩基性条件下で加水分解することによりホルミル体が得られる。この反応では、化合物(J)とDMF等価体を等量、或いは一方を過剰に用い、これらの混合物を、反応に不活性な溶媒中又は無溶媒下で、ハロゲン化剤の存在のもとに撹拌する。この反応は、室温~加熱還流下で、通常1時間~5日間行われる。溶媒の例としては、ハロゲン化炭化水素類;エーテル類;又はMeCNが挙げられる。ハロゲン化剤はDMF誘導体をVielsmeier錯体へと導くために用いられるものであり、通常、アルコールのハロゲン化に用いる試薬であれば特に限定はないが、五塩化リン、POCl3等が好適に用いられる。

Step6-2はハロゲン化である。化合物(C)は化合物(M)をハロゲン化して得られる。ハロゲン化剤としては、水酸基をハロゲンに変換するハロゲン化剤が使用される。特に限定はされないが、例えばPBr3、HBr、BBr3、PCl3、PCl5等が用いられる。溶媒は、エーテルが好ましく、例えばTHF、ジエチルエーテル、ジメトキシエタン、メチル-t-ブチルエーテル、ジオキサン、2-メチルテトラヒドロフラン等が用いられる。
化合物(T)は、化合物(N)から、Wittig反応、還元、酸化、クロメン骨格の構築を順次行うことにより製造できる。----が単結合の化合物(T)は、反応を妨げない段階で還元反応を行うことにより得られる。
Step7-1は、リンイリドの形成反応である。化合物(O)は、化合物(N)を通常反応を妨げない溶媒中、例えば、亜リン酸トリエチル等と反応させることにより得られる。溶媒の例としては、芳香族炭化水素類;エーテル類;ハロゲン化炭化水素類;アセトン、エチルメチルケトン等のケトン類;DMF、DMSO、EtOAc、MeCN;及びこれらの混合溶媒が挙げられる。反応温度は通常-20℃から加熱条件下で行われる。
Step7-2は、いわゆるWittig反応である。化合物(O)をアルデヒド化合物(P)と反応させ、化合物(Q)を製造することができる。ホスホリル基置換カルボアニオンのアルデヒド付加により、Wittig類似の機構を経て、オレフィンが得られる。反応温度は0℃から加温条件下である。

Step7-3は、還元反応である。還元剤としては、LiAlH4、LiAlH(OMe)3、DIBALを用い、通常、冷却下から加熱下、THFやエーテルなど反応に不活性な溶媒中で反応できる。
Step7-4は、酸化反応である。酸化剤としては、二酸化マンガンやPDCを用いる。溶媒の例としては、通常ハロゲン化炭化水素類等が挙げられる。反応温度は0℃から加熱条件下で行われ、通常室温で行われる。 その他の例としては、DMSO-POCl3系試薬を用いる方法がある。POCl3を用いる代わりにDCC、酸無水物、塩素、Me2S-NCS系試薬を用いる方法(Corey-Kim酸化)や、Dess-Martin ペルヨージナンを用いることもできる。反応は、通常室温から加温条件下で進行する。溶媒の例としては、特に限定しないが、芳香族炭化水素類;エーテル類;ハロゲン化炭化水素類;MeCN及びこれらの混合溶媒が挙げられる。
Step7-5は、クロメン環構築反応である。化合物(T)は、化合物(S)にアクロレイン誘導体を加え、K2CO3等の無機塩基の存在下に、室温から加熱条件下で攪拌して製造できる。溶媒としては例えば、芳香族炭化水素類;エーテル類;ハロゲン化炭化水素類;MeCN及びこれらの混合溶媒が挙げられる。通常、THF、DME又はジオキサン等のエーテル系の溶媒が用いられる。

例えばアルキル化は、当業者が通常用いるアルキル化反応を採用することができ、エーテル類;芳香族炭化水素類;ハロゲン化炭化水素類;DMF、MeCN;非プロトン性の極性溶媒等の反応に不活性な有機溶媒中、冷却下、冷却~室温下、もしくは、室温~加熱下に、NaH;炭酸アルカリ;炭酸水素アルカリ;アルコキシド;3級アミン;有機塩基などの塩基存在下に行うことができる。
また、例えば、アシル化は当業者が通常用いるアシル化反応を採用することができるが、特に、HOBt存在下、EDCI・HClやCDI、ジフェニルホスホリルシアニド等の縮合剤を反応条件により異なるが、通常、エーテル類;芳香族炭化水素類;ハロゲン化水素類、EtOAc等のエステル類;MeCN;非プロトン性溶媒等の不活性な有機溶媒中、冷却下、冷却~室温下、もしくは室温~加熱下に行われる。
単離、精製は、抽出、分別結晶化、各種分画クロマトグラフィー等、通常の化学操作を適用して行なわれる。
各種の異性体は、適当な原料化合物を選択することにより製造でき、あるいは異性体間の物理化学的性質の差を利用して分離することができる。例えば、光学異性体は、ラセミ体の一般的な光学分割法(例えば、光学活性な塩基又は酸とのジアステレオマー塩に導く分別結晶化や、キラルカラム等を用いたクロマトグラフィー等)により得られ、また、適当な光学活性な原料化合物から製造することもできる。
試験例1: in vitro S1P1受容体アゴニスト作用の評価
(方法1)ヒトS1P1発現細胞の膜を用いたGTP[γ-35S]結合アッセイによる受容体アゴニスト作用の評価方法
本発明化合物のin vitro S1P1アゴニスト作用は、ヒトS1P1発現細胞の膜を用い、GTP[γ-35S]のG蛋白への機能的結合活性の上昇により評価した。ヒトS1P1をコードするcDNAはヒト大腸cDNAライブラリーよりクローニングし、発現ベクターpcDNA3.1に導入し、S1P1-pcDNA3.1を作成した。次にリポフェクトアミン2000(GIBCO)により、S1P1-pcDNA3.1をCHO細胞にトランスフェクションし、10% 牛胎児血清、100 U/mLペニシリン、100 μg/mLストレプトマイシン、1 mg/mL G418二硫酸塩含有Ham’s F-12培地にて培養し、G418に耐性な安定株を得た。培養したヒトS1P1発現細胞は1 mM EDTA・2Na含有PBSにて剥離し、0.1 mM EDTAおよびプロテンインインヒビター含有1 mM Tris HCl(pH7.4)緩衝液中で、ガラス製のホモジナイザーにて氷冷しながら破壊した。1,400×gで10分間遠心分離し、更に上清を4℃、60分間、100,000×gで遠心分離し、1 mM EDTA含有 10 mM Tris HCl(pH7.4)緩衝液に懸濁し膜を精製した。得られた膜(0.13 mg/mL)および50 pM GTP[γ-35S](NEN;非活性1250Ci/mmol)を本発明化合物(10-12~10-5M)と共に100 mM NaCl、10 mM MgCl2、0.1% 脂肪酸非含有BSA、5μM GDPを含む20 mM HEPES(pH7.0)緩衝液中(全量150 μL)で1時間反応させて、セルハーベスター(パッカード、フィルターメート)にてGF-Cフィルタープレート上に膜を回収した。フィルタープレートは50℃で60分間乾燥し、マイクロシンチ0(パッカード)を加えてマイクロプレート用液体シンチレーションカウンター(パッカード、TOP count)にて測定した。本発明化合物および対照化合物のヒトS1P1アゴニスト作用の評価は化合物存在下でのGTP[γ-35S]の結合の飽和状態となる最大反応を100%、化合物非存在下でのGTP[γ-35S]の結合反応を0%とした際の100分率を用い、非線形回帰曲線をプロットし、最大反応の50%を作動するアゴニスト作用を誘導する濃度をEC50値(nM)として定義した。
(方法2)ヒトS1P1発現細胞を用いたCa2+流入アッセイによる受容体アゴニスト作用の評価方法
本発明化合物のin vitro S1P1アゴニスト作用は、ヒトS1P1発現細胞における細胞内Ca2+濃度の上昇により評価した。ヒトS1P1をコードするcDNAはヒト大腸cDNAライブラリーよりクローニングし、発現ベクターpcDNA3.1に導入し、S1P1-pcDNA3.1を作成した。次にリポフェクトアミン2000(GIBCO)により、S1P1-pcDNA3.1をCHO細胞にトランスフェクションし、10% 牛胎児血清、100 U/mLペニシリン、100 μg/mLストレプトマイシン、1 mg/mL G418二硫酸塩含有Ham’s F-12培地にて培養し、G418に耐性な安定株を得た。培養したヒトS1P1発現細胞は1 mM EDTA・2Na含有PBSにて剥離し、10% 牛胎児血清、100 U/mLペニシリン、100 μg/mLストレプトマイシン含有Ham’s F-12培地に懸濁した。この細胞懸濁液を96ウェルプレートに1ウェルあたり50000細胞となるよう分注し、CO2インキュベーター(5%CO2、37℃)内で1晩培養した。培地をカルシウム感受性蛍光試薬(FLIPR(登録商標)カルシウム3アッセイキット、モレキュラーデバイス)を含むloading buffer(ハンクス平衡塩溶液,20mM HEPES,2.5mM probenecid)に置換しCO2インキュベーター(5%CO2、37℃)内で1時間静置した。プレートを創薬スクリーニングシステムFDSS6000(浜松ホトニクス)にセットし、励起波長480nmにおける蛍光強度を1.02秒毎に124回連続測定した。被験化合物(終濃度10-12~10-5M)は12回目の測定と同時に添加し、蛍光強度の変化により細胞内Ca2+濃度変化を評価した。本発明化合物および対照化合物のヒトS1P1アゴニスト作用の評価は、化合物添加後の細胞内Ca2+濃度上昇が飽和状態となる最大反応を100%、溶媒のみを添加したときのCa2+濃度上昇を0%とした際の100分率を用いて非線形回帰曲線をプロットし、最大反応の50%のアゴニスト作用を誘導する濃度をEC50値(nM)として定義した。
ラットを用いて末梢血リンパ球に対する作用を評価した。6~10週齢の雄性Lewisラット(日本チャールスリバー)を無作為に群分けし(n=3)、本発明化合物を0.5%メチルセルロース含有蒸留水に懸濁し、ゾンデにて経口投与した。投与後、4時間後若しくは24時間後にエーテル麻酔下で0.2 mL 眼底採血した。血液サンプルは直ちにEDTA・4Kおよびヘパリンを加え、凝固を防ぎ、全自動血球計算器(シスメックス;XT-2000i)にて血液中のリンパ球数を測定した。同時に行った0.5%メチルセルロース含有蒸留水投与群のリンパ球数を100%として、本発明化合物が末梢血中のリンパ球数を50%減少する投与量をED50値(mg/kg)として算出した。
いくつかの式(I)の化合物についての試験例1ならびに試験例2の結果を、表1並びに表2に示す。表中、A欄は、試験例1の方法1により、但し*の値は方法2により測定した、in vitro S1P1 アゴニスト作用、EC50値(nM)を示す。また、B欄は、試験例2の薬剤投与4時間後若しくは24時間後ののラット末梢血リンパ球を減少作用を、それぞれED504h(mg/kg)若しくは、ED5024h(mg/kg)として示す。
表1並びに表2に示すように、本発明の式(I)の化合物は、良好なS1P1アゴニスト作用を有しており、ラットを用いた薬理試験において投与から4時間後或いは24時間後でも強力なリンパ球数の減少作用を示すことが確認された。
既知のS1P1アゴニスト作用薬に観察された好ましくない作用の1つである、ラットの肺重量増加を評価した。6~10週齢の雄性LewisまたはSDラット(日本チャールスリバー)を無作為に群分けし(n=3~4)、本発明化合物を0.5%メチルセルロース含有蒸留水に懸濁し、ゾンデにて経口投与した。単回投与は投与後24時間後にラットの体重を測定し、ペントバルビタール麻酔下に脱血し、肺を摘出し、重量を測定した。反復投与は1日1回7日間反復投与し、最終投与の24時間後に体重および肺重量を測定した。肺重量増加は0.5%メチルセルロース含有蒸留水投与群の体重に対する肺重量の相対重量の平均に対して、本発明化合物の0.5%メチルセルロース含有蒸留水懸濁液投与群の相対重量の平均が増加した割合を100分率で示し、肺重量増加が10%以上を示した投与量を陽性と判断した。
ラット腹部異所性心移植モデルの作製はOnoおよびLindseyの方法に準じて行うことができる(Transplantation、1969年、517、pp.225-229)。ドナーとして6~8週齢の雄性ACIラット(日本クレア)を用い、ペントバルビタール麻酔下にて、心臓を露出し、大動脈と肺動脈以外の左右上大静脈、肺静脈および下大静脈を一括して結紮し、大動脈と肺静脈を切り離し、グラフトとして摘出する。6~8週齢の雄性Lewisラット(日本チャールスリバー)をレシピエントとして用い、ペントバルビタール麻酔下にて、グラフトの大動脈端とレシピエントの腹部大動脈を吻合し、さらにグラフトの肺動脈端とレシピエントの下大静脈を吻合してモデルを作製する(1群当たり6~10例になるよう体重により群分けする)。移植心の拒絶判定は、移植後29日まで毎日レシピエントの腹部を触診し、グラフトの拍動の有無により拒絶を判定する。本発明化合物は0.5%メチルセルロース含有蒸留水に懸濁し、移植の当日から14日間、1日1回または2回、経口投与する。対照群として0.5%メチルセルロース含有蒸留水を同期間、同回数、経口投与する。同時にすべての群にタクロリムスを0.02 mg/mL/kg筋肉内投与する。当該試験により、本発明化合物の、タクロリムス併用下における拒絶反応の抑制作用を判定することができる。
雄性Lewisラットをイソフルランで吸入麻酔し、大腿動脈及び静脈にポリエチレンチューブを挿管した。動脈ラインより圧トランスデューサーを介して血圧測定用アンプ・心拍ユニットに接続して動脈血圧及び心拍数を測定した。静脈ラインよりVehicle (10%HCO40/tween 80/PEG, 90%saline)及び本発明化合物を1mL/kg/minの速度で10分間静脈内持続注入した。測定データは投与前値、持続注入開始後1, 2, 5及び10分値、注入終了後1, 2, 5及び10分値をチャートから読み取り(全評価時間20分間)、投与前値の心拍数及び血圧に対して注入中及び注入後で減少率(%)を算出した。
本発明化合物中、例えば実施例230の化合物は、本評価により1mg/kg投与時、心拍数ならびに血圧に影響を与えず、徐脈を発現しないことが確認された。
投与は錠剤、丸剤、カプセル剤、顆粒剤、散剤、液剤等による経口投与、又は、関節内、静脈内、筋肉内等の注射剤、坐剤、点眼剤、眼軟膏、経皮用液剤、軟膏剤、経皮用貼付剤、経粘膜液剤、経粘膜貼付剤、吸入剤等による非経口投与のいずれの形態であってもよい。
経口投与のための液体組成物は、薬剤的に許容される乳濁剤、溶液剤、懸濁剤、シロップ剤又はエリキシル剤等を含み、一般的に用いられる不活性な希釈剤、例えば精製水又はエタノールを含む。当該液体組成物は不活性な希釈剤以外に可溶化剤、湿潤剤、懸濁剤のような補助剤、甘味剤、風味剤、芳香剤、防腐剤を含有していてもよい。

なお、構造式の交差二重結合は、シスとトランスの混合物を意味する。1H-NMRデータにおいて、特に記載のない場合には、テトラメチルシランを内部標準とし、DMSO-d6を測定溶媒とする1H-NMRにおけるシグナルのδ(ppm)(積分値、***パターン)を表す。本明細書中では、NMRは1H-NMR:プロトン核磁気共鳴を示す。なお、MSとESI(EI)の接尾字の+、-は、それぞれポジティブマスデータとネガティブマスデータを示す。
7-[(5-ブロモ-4-フェニル-2-チエニル)メトキシ]-2H-クロメン-3-カルボアルデヒド(120 mg)をDMF(2.4 mL)に溶解した。この反応液に、Zn(CN)2(65 mL)及びPd(PPh3)4(65 mg)を室温で加えた。反応混合物を100℃で5時間攪拌した後、NaHCO3水とEtOAcの1:1混合溶媒中に注ぎ、1時間攪拌した。有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。シリカゲルカラムクロマトグラフィー(ヘキサン:EtOAc=100:0~70:30)で精製し、5-{[(3-ホルミル-2H-クロメン-7-イル)オキシ]メチル}-3-フェニルチオフェン-2-カルボニトリル(83 mg)を淡黄色固体として得た。
メチル 5-ブロモ-4-フェニルチオフェン-2-カルボキシレートのジオキサン溶液に2-イソプロペニル-4,4,5,5-テトラメチル-1,3,2-ジオキサボロランと2M Na2CO3水溶液を加えた。反応混合物中に、酢酸パラジウムとPPh3を加え、100℃で5時間攪拌した。放冷後、飽和NH4Cl水溶液を加え、EtOAcで抽出した。有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮し、シリカゲルカラムクロマトグラフィー(ヘキサン:EtOAc=95:5~80:20)で精製し、メチル 5-イソプロペニル-4-フェニルチオフェン-2-カルボキシレートを無色液体として得た。
DMF(2.5 mL)のDCM(3 mL)溶液にPOCl3(2 mL)を0℃で滴下し、室温で30分攪拌した。続いて、反応液中に、8-(ベンジルオキシ-3,4-ジヒドロー1-ベンズオキセピン-5(2H)-オンのDCM(4 mL)溶液を滴下し、室温で1時間、50℃で3時間攪拌した。反応液に水を加え、EtOAcで2回抽出した。有機層をまとめて、水、brineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置;ヘキサン:EtOAc=97:3~90:10)で精製し、8-(ベンジルオキシ)-5-クロロ-2,3-ジヒドロ-1-ベンズオキセピン-4-カルボアルデヒド(445 mg)を得た。
DMF(2 mL)のDCM(7.5 mL)溶液にPOCl3(1.39 mL)を0℃で滴下し、室温で30分攪拌した。続いて、反応液中に、7-{[tert-ブチル(ジフェニル)シリル]オキシ}-2,3-ジヒドロ-4H-クロメン-4-オン(2.00 g)のDCM(11 mL)溶液を滴下し、室温で1時間、50℃で3時間攪拌した。反応液に水を加え、EtOAcで2回抽出した。有機層をまとめて水、brineで洗浄し、MgSO4で乾燥し、ろ液を濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=100:0~80:20)で精製し、4-クロロ-7-ヒドロキシ-2H-クロメン-3-カルボアルデヒド(720 mg)を得た。
7-(ベンジルオキシ)-2,3-ジヒドロ-4H-クロメン-4-オンをTHFに溶解して、メチルマグネシウムブロマイドのTHF溶液(0.97 M、5 mL)を0℃で滴下した。室温で1時間攪拌し、メチルマグネシウムブロマイドのTHF溶液(0.97M、5 mL))を滴下し、室温で2時間攪拌した。反応液に飽和NH4Cl水溶液を加え、続いて2M塩酸(20 mL)を加え、室温で2時間攪拌後、EtOAcで3回抽出した。有機層をまとめて、水、brineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=95:5~90:10)で精製し、7-(ベンジルオキシ)-4-メチル-2H-クロメン(445 mg)を無色透明液体として得た。
2-ヒドロキシ-4-[(2-メトキシ-4-プロピルフェノキシ)メチル]ベンズアルデヒド(120 mg)のジオキサン溶液(2.4 mL)に、K2CO3(55.2 mg)とアクロレイン(0.267 mL)を25℃で加えた。反応混合物を100℃まで昇温した後、100℃で15時間攪拌した。反応混合物を、25℃まで放冷した後、セライトろ過し、ろ液を濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=100:0~80:20)で精製し、7-[(2-メトキシ-4-プロピルフェノキシ)メチル]-2H-クロメン-3-カルボアルデヒド(104.2 mg)を無色液体として得た。
ジオキサン(40 mL)にK2CO3(835 mg)を懸濁して、2-ヒドロキシ-4-(メトキシメトキシ)ベンズアルデヒド(1 g)、3-メチル-2-ブタナール(0.787 mL)を加え、110℃で終夜攪拌した。EtOAcを加えて、不溶物をセライトろ過で除き、ろ液を減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=95:5~70:30)で精製し、7-(メトキシメトキシ)-2,2-ジメチル-2H-クロメン-3-カルボアルデヒド(320 mg)を黄色オイルとして得た。
0℃で、濃HCl(8 mL)とAcOH(1.6 mL)の混合溶媒にtert-ブチル 3-シアノ-3-(フルオロメチル)アゼチジン-1-カルボキシレート(800 mg)を加えた。25℃まで昇温した後、25℃で1時間攪拌した。その後、100℃で5時間攪拌し、反応液を減圧下濃縮した。次にトルエン(30 mL)で3回共沸した。残渣をアセトン(4.8 mL)と水(8.0 mL)混合溶媒中に溶解し、0℃でNa2CO3(593.7 mg)とDIBOC(1223 mg)を加えた。反応液は、25℃まで昇温した後、25℃で15時間攪拌した。15時間後、反応溶液を濃縮し、アセトンを留去した。残渣にエーテル(50 mL)を加えて3回抽出した(50 mL×3)。水層を集め、0℃に冷却し、0℃で2M HCl(10 mL)を加えて、溶液をpH=2~3に調製した。析出した白色固体をろ取し、ヘキサン(50 mL)で洗浄し、1-(tert-ブトキシカルボニル)-3-(フルオロメチル)アゼチジン-3-カルボン酸(801.2 mg)を白色固体として得た。
tert-ブチル 3-シアノ-3-(ヒドロキシメチル)アゼチジン-1-カルボキシレート(5.0 g)をDCM(100 mL)に溶解した。0℃で、DAST(3.74 mL)を加え、0℃で3時間攪拌した。3時間後、反応液にNaHCO3水溶液(100 mL)を加え、DCM(50 mL)で3回抽出した。有機層をbrineで洗浄し、MgSO4で乾燥後、濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=100:0~50:50)で精製し、tert-ブチル 3-シアノ-3-(フルオロメチル)アゼチジン-1-カルボキシレート(1.24 g)を茶色固体として得た。
2-フルオロ-4,6-ジヒドロキシベンズアルデヒド(12 g)をMeCN(250 mL)に溶解して炭酸セシウム(25.1 g)、クロロメチルメチルエーテル(6.95 mL)を加え、室温で1時間攪拌した。不溶物をセライトろ過で除去し、ろ液を濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=100:0~94:6)で精製し、2-フルオロ-6-ヒドロキシ-4-(メトキシメトキシ)ベンズアルデヒド(11.89 g)を白色粉末として得た。
7-ヒドロキシ-2,3-ジヒドロ-4H-クロメン-4-オン(900 mg)をDMF(10 mL)に溶解して、tert-ブチル(クロロ)ジフェニルシラン(1.711 mL)、1H-イミダゾール(448 mg)を加え、室温で終夜攪拌した。反応液に水を加え、EtOAcで3回抽出した。有機層をまとめて、水、brineで順次洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=90:10~80:20)で精製し、7-{[tert-ブチル(ジフェニル)シリル]オキシ}-2,3-ジヒドロ-4H-クロメン-4-オン(2.08 g)を無色透明なシロップとして得た。
7-ヒドロキシ-2H-クロメン-3-カルボアルデヒドのDCM溶液にピリジンを0℃で加えた。反応液中にトリフルオロメタンスルホン酸無水物を0℃で滴下した。室温で1時間撹拌した後、0℃で水を加えた。混合物をEtOAcで抽出した。有機層を1M HCl、水、brineで順次洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン:EtOAc=95:5~80:20)で精製し、3-ホルミル-2H-クロメン-7-イル トリフルオロメタンスルホネートを黄色油状として得た。
DMF(1 mL)にPOCl3(0.25 mL)を0℃で滴下し、室温で30分攪拌した。反応混合物に、7-(ベンジルオキシ)-4-メチル-2H-クロメン(280 mg)のDCM(1 mL)溶液を滴下し、室温で3時間攪拌した。反応液を氷水に注ぎ、EtOAcで3回抽出した。有機層をまとめて、水、brineで順次洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=85:15~70:30)で精製し、7-(ベンジルオキシ)-4-メチル-2H-クロメン-3-カルボアルデヒド(234 mg)を淡黄色粉末として得た。
NaH(105.63 mg)のDMF溶液(5.5 mL)を0℃に冷却して、メチル 2-{[tert-ブチル(ジメチル)シリル]オキシ}-4-[(ジエトキシホスホリル)メチル]ベンゾエート(550 mg)を加えた。反応混合物を25℃まで昇温した後に1時間攪拌した。再び0℃に冷却し、2-メトキシ-4-プロピルベンズアルデヒド(235.34 mg)を加えた。反応混合物を25℃に昇温した後に15時間攪拌した。反応液に飽和NH4Cl水溶液(50 mL)を加え、EtOAc(50 mL)で3回抽出した。有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=100:0~70:30)で精製し、メチル2-ヒドロキシ-4-[(E)-2-(2-メトキシ-4-プロピルフェニル)ビニル]ベンゾエート(304.2 mg)を白色固体として得た。
DMF(40 mL)に60%NaH(634 mg)を氷冷下加え、4-フルオロ-3-(トリフルオロメチル)ベンゾニトリル (2 g)のDMF溶液(20 mL)をゆっくり加えた。室温で5時間撹拌した後、反応を飽和NH4Cl溶液でクエンチした。EtOAcで抽出し、有機層をbrineで洗浄、MgSO4で乾燥後ろ過した。ろ液を濃縮することで4-イソプロポキシ-3-(トリフルオロメチル)ベンゾニトリル(2.4 g)を淡黄色固体として得た。
メチル 4-フルオロ-2-(トリフルオロメチル)ベンゾエートのDMF溶液にK2CO3とピペリジンを加え、100℃で3時間撹拌した。反応混合物を0℃に冷却し、水を加え、EtOAcで抽出した。有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン:EtOAc=100:0~90:10)で精製し、メチル 4-ピペリジン-1-イル-2-(トリフルオロメチル)ベンゾエートを無色油状として得た。
メチル 1H-インドール-5-カルボキシレート(1.5 g)のDMF溶液(30 mL)にNaH(410 mg)を0℃で加えた。反応混合物を25℃まで昇温し、0.5時間攪拌した。次に、反応混合物を再び0℃まで冷却した後、ヨードメタン(1.38 mL)を加えた。反応混合物を25℃まで昇温した後、3時間攪拌した。反応液に水(50 mL)を加え、EtOAc(50 mL)で3回抽出した。有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、CHCl3:MeOH=100:0~98:2)で精製し、メチル 1-エチル-1H-インドール-5-カルボキシレート (1465 mg)を白色固体として得た。
4-フルオロ-2-(トリフルオロメチル)安息香酸のMeOH溶液に0℃で濃硫酸を加えた。反応混合物は2日間加熱還流した。反応混合物を減圧下濃縮し、残渣をEtOAcで希釈した。有機層を飽和NaHCO3水溶液で洗浄し、MgSO4で乾燥後、減圧下濃縮し、メチル 4-フルオロ-2-(トリフルオロメチル)ベンゾエートを無色油状として得た。
4-ブロモ-5-エチルチオフェン-2-カルボン酸(800 mg)のMeOH懸濁溶液(4 mL)に0℃でSOCl2(0.50 mL)を滴下した。反応混合物を0℃で1時間攪拌した後、60℃まで昇温した後に15時間攪拌した。反応液を減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=98:2~70:30)で精製し、メチル 4-ブロモ-5-エチルチオフェン-2-カルボキシレート (765.0 mg)を無色液体として得た。
N-イソプロピルプロパン-2-アミン(165.5 mg)のTHF溶液(1 mL)に-78℃でn-ブチルリチウムのヘキサン溶液(1.6 M、0.98 mL)を滴下した。25℃まで昇温した後、30分攪拌した。再び-78℃まで冷却した後、1-tert-ブチル 3-メチル ピロリジン-1,3-ジカルボキシレート(300 mg)のTHF溶液(1 mL)を滴下した。反応混合物を-40℃まで昇温した後に1時間攪拌した。反応混合物を、再び-78℃まで冷却し、N-フルオロ-N-(フェニルスルホニル)ベンゼンスルホンアミド(495.1 mg)のTHF溶液(1 mL)を滴下した。反応混合物は、-78℃で1時間攪拌した後に25℃まで昇温して15時間攪拌した。15時間後、反応液に飽和NH4Cl水溶液(30 mL)を加え、EtOAc(30 mL)で3回抽出した。有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=100:0~80:20)で精製し、1-tert-ブチル 3-メチル 3-フルオロピロリジン-1,3-ジカルボキシレート(154.3 mg)を黄色液体として得た。
メチル 4-フェニルチオフェン-2-カルボキシレート(1.8 g)のDCM溶液(18 mL)に、0℃でピリジニウムトリブロマイド(13.2 g)を少しずつ加えた。反応液を25℃まで昇温した後に45時間攪拌した。反応混合物を0℃に冷却して、飽和Na2S2O3水溶液(100 mL)の中にゆっくりと滴下した。反応混合物をDCM(50 mL)で3回抽出した。有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=100:0~90:10)で精製し、メチル 5-ブロモ-4-フェニルチオフェン-2-カルボキシレート(2.06 g)を無色液体として得た。
4-クロロ-5,5,5-トリフルオロ-3-フェニルペント-3-エン-2-オン(950 mg)とメチル スルファニルアセテート(446 mg)のMeCN溶液(23.8 mL)にDBU(0.63 mL)を25℃で滴下した後に、同温で15時間攪拌した。反応液に飽和NH4Cl水溶液(50 mL)を加え、ジエチルエーテル(50 mL)で3回抽出した。有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=98:2~90:0)で精製し、メチル 3-メチル-4-フェニル-5-(トリフルオロメチル)チオフェン-2-カルボキシレート(1.08 g)を無色液体として得た。
ベンジル 3-シアノピロリジン-1-カルボキシレート(1.0 g)とTEA塩酸塩(2.99 g)のトルエン溶液に、25℃でアジ化ナトリウム(1.41 g)を加えた後に、115℃で5時間攪拌した。反応液を放冷し、DCM(10 mL)を加えた。その後、5%サリチル酸水溶液(100 mL)の中に反応液を滴下した後に25℃で1時間攪拌した。反応液をEtOAc(30 mL)で3回抽出した。有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=100:0~80:20)で精製し、ベンジル 3-(1H-テトラゾール-5-イル)ピロリジン-1-カルボキシレート(10.8 g)を無色液体として得た。
2-{[tert-ブチル(ジメチル)シリル]オキシ}-4-メチル安息香酸メチルエステル(3.4 g)の四塩化炭素(68 mL)溶液に、NBS(2.16 g)及びAIBN(398 mg)を室温で加え、80℃で1時間攪拌した。反応が終了したことをTLCで確認し、反応液に水を加えて、反応を停止させた後、EtOAcで抽出した。有機層をbrineで洗浄し、MgSO4を用いて乾燥後、減圧下濃縮した。シリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=100:0~95:5)で精製し、4-(ブロモメチル)-2-{[tert-ブチル(ジメチル)シリル]オキシ}安息香酸メチルエステル(3.79 g)を無色液体として得た。
(2S)-3-(4-クロロフェニル)-2-メチルプロパン-1-オール (300 mg)のDCM溶液 (20 mL)にN-ブロモスクシンイミド (347 mg)とトリフェニルフォスフィン (511 mg)を氷冷下で加えた。反応液を室温で2時間撹拌した後、反応液を水にあけ、クロロホルムで抽出した。有機層をbrineで洗浄し、MgSO4で乾燥した後、溶媒を減圧下留去した。残渣のシリカゲルカラムクロマトグラフィー (ヘキサン:EtOAc=100:0-90:10)により、1-[(2S)-3-ブロモ-2-メチルプロピル]-4-クロロベンゼン (373 mg)を無色液体として得た。
N-イソプロピルプロパン-2-アミン(11.54 mL)のTHF溶液(50 mL)に-78℃で、n-ブチルリチウムのヘキサン溶液(1.6 M、51.45 mL)を滴下した。反応混合物は、0℃まで昇温した後に30分攪拌した。反応混合物を再び-78℃まで冷却した後、tert-ブチル 3-シアノアゼチジン-1-カルボキシレート(5.0 g)のTHF溶液(30 mL)を滴下した後-78℃で1時間攪拌した。反応混合物に-78℃で1H-ベンゾトリアゾール-1-イルメタノール(8.19 g)のTHF溶液(20 mL)を滴下し、-78℃で3時間攪拌した。反応混合物に飽和NH4Cl水溶液(100 mL)を加え、EtOAc(50 mL)で3回抽出した。有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=100:0~50:50)で精製し、tert-ブチル 3-シアノ-3-(ヒドロキシメチル)アゼチジン-1-カルボキシレート (5.68 g)を白色固体として得た。
4-アミノ-(2-トリフルオロメチル)安息香酸メチル塩酸塩(1.24 g)と2,5-ジメトキシテトロヒドロフラン(773 mg)のAcOH(20 mL)溶液を80℃で12時間攪拌した。反応混合物を減圧下濃縮した後、トルエンで共沸してAcOHを留去して、得られた茶褐色油状物をクロロホルムに溶解させ、飽和NaHCO3水溶液を加えた。有機層を飽和NaHCO3水溶液、水、brineで順次洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(CHCl3:MeOH=97:3)で精製し、 4-(1H-ピロロ-1-イル)-2-(トリフルオロメチル)安息香酸メチルを(11.8 g)得た。
トリメチル(プロ-1-ピン-1-イル)シラン(877 mg)のTHF(60 mL)溶液にn-BuLiのヘキサン溶液(1.58M、4.5 mL)を-78℃で加えた。反応混合物を-78℃で3時間撹拌した後、1-(ブロモメチル)-2,4-ビス(トリフルオロメチル)ベンゼン(2 g)のTHF(10 mL)溶液を滴下した後に1時間撹拌した。反応液にNH4Cl水溶液を加え、エーテルで抽出した。有機層をbrineで洗浄し、MgSO4で乾燥した後に減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン:EtOAc=100:0)で精製し、{4-[2,4-ビス(トリフルオロメチル)フェニル]ブチ-1-ン-1-イル}(トリメチル)シラン (1.8 g)を無色液体として得た。
1-(クロロメチル)-2-メトキシ-4-プルピルベンゼン(1.1 g)をDMF(20 mL)に溶解し、7-ヒドロキシ-2H-クロメン-3-カルボアルデヒド(975 mg)、K2CO3(1.15g)を加えて、80℃で1時間攪拌し、さらに、ヨウ化ナトリウム(416 mg)を加え、80℃で1時間攪拌した。反応が終了したことを確認した後に、反応液に水を加えて反応を停止させ、EtOAcで3回抽出した。有機層をまとめてbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=95:5~80:20)で精製し、7-[(2-メトキシ-4-プロピルベンジル)オキシ]-2H-クロメン-3-カルボアルデヒド(1.21 g)を黄色粉末として得た。
7-ヒドロキシ-2H-クロメン-3-カルボアルデヒド(200 mg)をDMF(5 mL)に溶解して、K2CO3(235 mg)、1-(ブロモメチル)-2,4-ビス(トリフルオロメチル)ベンゼン(0.234 mL)を加え、80℃で30分間攪拌した。反応液を水に注ぎ、生じた粉末をろ取し、減圧乾燥し、7-{[2,4-ビス(トリフルオロメチル)ベンジル]オキシ}-2H-クロメン-3-カルボアルデヒド(455 mg)を淡黄色粉末として得た。
3-ホルミル-2H-クロメン-7-イル トリフルオロメタンスルホネート(520 mg)のDMF(10.4 mL)溶液に1-エチニル-4-(トリフルオロメチル)ベンゼン(330 μL)、ビス(トリフェニルホスフィン)パラジウム(II)ジクロリド(355 mg)、ヨウ化銅(I)(161 mg)及びTEA(470 μL)を室温で加えた。反応混合物を100℃で5時間撹拌した。反応混合物に、氷冷下で水を加え、不溶物をろ別し、ろ液をEtOAcで抽出した。有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン:EtOAc)で精製し、7-{[4-(トリフルオロメチル)フェニル]エチニル}-2H-クロメン-3-カルボアルデヒド(189 mg)得た。
Pd(PPh3)4(542 mg)とTEA(4 mL)のDMF(16 mL)溶液に1-ブロモ-4-イソブチルベンゼン(1 g)とエチニル(トリメチル)シラン(553 mg)を室温で加え、60℃で4時間撹拌した。反応液に1M塩酸を加え、エーテルで抽出した。不溶物をセライトでろ過した。有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン)で精製し、[(4-イソブチルフェニル)エチニル](トリメチル)シラン(459 mg)を黄色液体として得た。
塩化銅(10 mg)とPd(PPh3)4(60 mg)のDMF(2mL)溶液に[(4-イソブチルフェニル)エチニル](トリメチル)シラン(288 mg)と5-フルオロ-3-ホルミル-2H-クロメン-7-イル トリフルオロメタンスルホン酸エステル(340 mg)を室温で加え、80℃で12時間撹拌した。反応液を濃縮し、残渣をシリカゲルカラムクロマトグラフィー(CHCl3)により精製し、5-フルオロ-7-[(4-イソブチルフェニル)エチニル]-2H-クロメン-3-カルボアルデヒド(83 mg)を黄色固体として得た。
1-[4-フェニル-5-(トリフルオロメチル)-2-チエニル]エタノン(1.0 g)のTHF溶液(20 mL)に、DIBAL(0.99M トルエン溶液、9.34 mL)を-78℃で滴下した。反応混合物を25℃に昇温し、3時間攪拌し、飽和ロッシェル塩水溶液(50 mL)を加えた後、EtOAc(50 mL)で3回抽出した。有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=98:2-90:10)で精製し、1-[4-フェニル-5-(トリフルオロメチル)-2-チエニル]エタノール(0.99 g)を無色液体として得た。
3-(トリフルオロメチル)-4-[(1S)-2,2,2-トリフルオロ-1-メチルエトキシ]安息香酸(1.085 g)のTHF溶液(43 mL)に、0℃でBH3・THFのTHF溶液(1M、14 mL)を滴下した。反応混合物を室温まで昇温した後15時間攪拌した。反応液に0℃で1M塩酸を加えて反応を停止させ、30分攪拌した。EtOAcで抽出し、有機層をbrineで洗浄した。MgSO4で乾燥後、減圧下濃縮し、{3-(トリフルオロメチル)-4-[(1S)-2,2,2-トリフルオロ-1-メチルエトキシ]フェニル}メタノール(570 mg)を白色油状物として得た。
メチル 4-ピペリジン-1-イル-2-(トリフルオロメチル)ベンゾエート(955 mg)のTHF(19 mL)溶液にDIBALのヘキサン溶液(1M、10.0 mL)を氷冷下滴下し、同温で2時間撹拌した。反応液にMeOHを滴下した後、飽和ロッシェル塩水溶液を加え、室温で1時間撹拌した。混合物をEtOAcで抽出し、有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(ヘキサン:EtOAc)で精製し、[4-ピペリジン-1-イル-2-(トリフルオロメチル)フェニル]メタノールを(846 mg)得た。
メチル 2-ヒドロキシ-4-[(2-メトキシ-4-プロピルフェノキシ)メチル]ベンゾエート(300 mg)をTHF(15 mL)に溶解した。反応液に0℃でLAH(103.4 mg)加えた。0℃から25℃まで昇温した後、3時間攪拌した。反応液に飽和ロッシェル塩水溶液(30 mL)を加え、EtOAc(30 mL)で3回抽出した。有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=100:0~80:20)で精製し、2-(ヒドロキシメチル)-5-[(2-メトキシ-4-プロピルフェノキシ)メチル]フェノール(245.2 mg)を白色固体として得た。
NaBH4(93.1 mg)のEtOH溶液(15 mL)に、0℃で、7-{[2,4-ビス(トリフルオロメチル)ベンジル]オキシ}-2,3-ジヒドロ-4H-チオクロメン-4-オン(1.0 g)のEtOH溶液(5 mL)を滴下した。反応混合物を25℃に昇温し3時間攪拌した。反応液を減圧下濃縮し、残渣にDCM(20 mL)を加えた後、0℃で飽和NH4Cl水(30 mL)を加え、1時間攪拌し、DCMで3回抽出した(30 mL×3)。有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=100:0~60:40)で精製し、7-{[2,4-ビス(トリフルオロメチル)ベンジル]オキシ}チオクロマン-4-オール (845 mg)を白色固体として得た。
常圧で水素ガス雰囲気下、メチル 4-フェニル-5-ビニルチオフェン-2-カルボキシレート(250 mg)のEtOH溶液(5 mL)にPd/C(50%wet)(50 mg)を25℃で加え、5時間攪拌した。反応混合物をセライトろ過し、ろ液を減圧下濃縮し、メチル 5-エチル-4-フェニルチオフェン-2-カルボキシレート(247.3 mg)を無色液体として得た。
5-フルオロ-7-ヒドロキシ-2H-クロメン-3-カルボアルデヒド(275 mg)と2-(ヒドロキシメチル)-5-メチル-4-フェニル-チアゾール(436 mg)のトルエン溶液(8.2 mL)に氷冷下、ADDP(393 mg)とTBP(315 mg)を加えた。反応液を室温で15時間攪拌した後、IPEを加え、固体をろ過により除去した。ろ液を減圧下濃縮し、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン:EtOAc=90:20~70:30)で精製し、5-フルオロ-7-[(5-5-メチル-4-フェニル-1,3-チアゾール-2-イル)メトキシ]-2H-2H-クロメン-3-カルボアルデヒド(381 mg)を淡黄色固体として得た。
5-({[2’-フルオロ-2-(トリフルオロメチル)ビフェニル-4-イル]オキシ}メチル)-2-(ヒドロキシメチル)フェノール(520 mg)のクロロホルム(10 mL)溶液に二酸化マンガン(1 g)を室温で加えた。反応液を室温で16時間撹拌した後、セライトろ過した。ろ液を減圧下濃縮し、シリカゲルカラムクロマトグラフィー(ヘキサン:EtOAc=95:5~80:20)で精製し、4-({[2’-フルオロ-2-(トリフルオロメチル)ビフェニル-4-イル]オキシ}メチル)-2-ヒドロキシベンズアルデヒド(180 mg)を白色固体として得た.
[2-({[2-(トリフルオロメチル)ビフェニル-4-イル]オキシ}メチル)-4,5-ジヒドロ-1-ベンゾチエン-6-イル]メタノール(1.0 g)のDCM溶液(20 mL)に、PDC(1.36 g)とMS4Å(1.36 g)を25℃で加えた。反応液を3時間攪拌した後、セライトろ過した。ろ液を減圧下濃縮し、残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=100:0~80:20)で精製し、2-({[2-(トリフルオロメチル)ビフェニル-4-イル]オキシ}メチル)-4,5-ジヒドロ-1-ベンゾチオフェン-6-カルボアルデヒド(345 mg)を無色液体として得た。
[1-(tert-ブトキシカルボニル)ピペリジン-4-イル]酢酸(200 mg)のジオキサン(1 mL)溶液に4M 塩化水素ジオキサン溶液(1 mL)を加えた。反応液を、室温で15時間撹拌した後、減圧下濃縮し、ピペリジン-4-イル酢酸塩酸塩 (140 mg)を白色固体として得た。
ベンジル 3-(1H-テトラゾール-5-イル)ピロリジン-1-カルボキシレート(300 mg)を濃塩酸(3 mL)とAcOH(0.6 mL)の混合溶液中に加え、100℃で5時間攪拌した。反応液を濃縮し、次に3回トルエン共沸した(30mL×3)。残渣をアセトン(0.9 mL)と水(1.5 mL)の混合溶液中に溶解した後、0℃まで冷却してNa2CO3(174.5 mg)とDIBOC(359.4 mg)を加えた。反応混合物を25℃まで昇温した後、15時間攪拌した。反応液を濃縮した。残渣にジエチルエーテル(30 mL)を加え、3回抽出した。有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮し、tert-ブチル 3-(1H-テトラゾール-5-イル)ピロリジン-1-カルボキシレート(102.7 mg)を無色液体として得た。
3-フルオロピロリジン-1,3-ジカルボン酸 1-tert-ブチル 3-メチル エステル(100 mg)に0℃で濃塩酸(1 mL)とAcOH(0.2 mL)との混合溶液を加えた。反応混合物を室温まで昇温し、1時間攪拌した後、100℃で5時間攪拌した。原料が消失したことを確認した後、減圧下濃縮し、次に3回トルエン共沸した。残渣をアセトン(0.6 mL)と水(1 mL)の混合液中に溶解し、0℃でNa2CO3(64 mg)とDIBOC(132 mg)を加え、室温まで昇温した後、15時間攪拌した。反応混合物を減圧下濃縮しアセトンを留去した。残渣にジエチルエーテルを加えて分液した。水層を集めて0℃に冷却し、2M塩酸でpH=2~3に調整した。EtOAcを加えて抽出した。有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮し、1-(tert-ブトキシカルボニル)-3-フルオロピロリジン-3-カルボン酸(70 mg)を白色固体として得た。
5-フルオロ-7-(メトキシメトキシ)-2H-クロメン-3-カルボアルデヒド(1.5 g)をアセトン(25 mL)に溶解して1M HCl(20 mL)を加え5時間加熱還流した。反応液を濃縮して残渣をEtOAcに溶解し、水、brineで洗浄し、MgSO4で乾燥し、濾液を濃縮した。残渣をクロロホルムで洗浄し、5-フルオロ-7-ヒドロキシ-2H-クロメン-3-カルバルデヒド(0.95 g)を黄色粉末として得た。
7-(メトキシメトキシ)-2,2-ジメチル-2H-クロメン-3-カルボアルデヒド(300 mg)をEtOH(10 mL)に溶解して、(1S)-(+)-10-カンファースルホン酸(421 mg)を加え、80℃で終夜攪拌した。反応液にシリカゲルを加えて濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=90:10~70:30)で精製し、7-ヒドロキシ-2,2-ジメチル-2H-クロメン-3-カルボアルデヒド(175 mg)を赤色粉末として得た。
KOH (358 mg)のMeOH (30 mL)溶液に、{4-[2,4-ビス(トリフルオロメチル)フェニル]ブチ-1-ン-1-イル}(トリメチル)シラン(1.8 g)を加え、室温で18時間撹拌した。反応液を1M塩酸で中和し、エーテルで抽出した。有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣はシリカゲルカラムクロマトグラフィー(ヘキサン:EtOAc=100:0)で精製し、1-ブチ-3-ン-1-イル-2,4-ビス(トリフルオロメチル)ベンゼン(426 mg)を無色液体として得た。
7-(ベンジルオキシ)-4-メチル-2H-クロメン-3-カルボアルデヒド(230 mg)、1,2,3,4,5-ペンタメチルベンゼン(608 mg)に、TFA(3 mL)を加え、室温で終夜攪拌した。反応液をNaHCO3水溶液中に注ぎ、EtOAcで3回抽出した。有機層をまとめてbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=80:20~20:80)で精製し、7-ヒドロキシ-4-メチル-2H-クロメン-3-カルボアルデヒド(130 mg)を淡黄色粉末として得た。
2-フルオロ-4,6-ジメトキシベンズアルデヒド(22 g)をDCM(110 mL)に溶解し、氷冷下でBBr3のDCM溶液(1M、300 mL)を滴下し、室温で終夜撹拌した。反応の終了を確認した後、反応液を氷水(100 mL)に注ぎ、1時間撹拌した後、EtOAcで3回抽出した。有機層はまとめて、水、brineで順次洗浄し、MgSO4で乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=80:20~60:40)で精製し、2-フルオロ-4,6-ジヒドロキシベンズアルデヒド(12 g)を白色粉末として得た。
7-{[2,4-ビス(トリフルオロメチル)ベンジル]オキシ}チオクロマン-4-オール(800 mg)のトルエン溶液(16 mL)に4-メチルベンゼンスルホン酸(33.7 mg)を加えて、120℃で3時間攪拌した。反応液に飽和NaHCO3水溶液(50 mL)を加え、EtOAc(50 mL)で3回抽出した。有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=100:0~80:20)で精製し、2,4-ビス(トリフルオロメチル)ベンジル 2H-チオクロマン-7-イル エーテル(753.2 mg)を無色液体として得た。
メチル 4-(ブロモメチル)-2-{[tert-ブチル(ジメチル)シリル]オキシ}ベンゾエート(0.30 g)とトリエチルホスファイト(0.17 g)の混合物を、25℃で混合し、その後、130℃で24時間攪拌した。反応液を減圧下濃縮し、トルエンで2回共沸した(30 mL×2)。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=40:60~10:90)で精製し、メチル 2-{[tert-ブチル(ジメチル)シリル]オキシ}-4-[(ジエトキシホスホリル)メチル]ベンゾエート(0.23 g)を無色液体として得た
7-[(4-ブロモ-5-エチル-2-チエニル)メトキシ]-2H-クロメン-3-カルボアルデヒド(150 mg)のジオキサン溶液(4.5 mL)に、25℃で[2-(トリフルオロメチル)フェニル]ボロン酸と2M Na2CO3水溶液を加えた。次に、反応混合物中に酢酸パラジウム(4.44 mg)とPPh3(20.75 mg)を加えた後、100℃に昇温し5時間攪拌した。反応液に飽和NH4Cl水溶液(30 mL)を加え、EtOAc(30 mL)で3回抽出した。有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=95:5~80:20)で精製し、7-({5-エチル-4-[2-(トリフルオロメチル)フェニル]-2-チエニル}メトキシ)-2H-クロメン-3-カルボアルデヒド(134.8 mg)を淡黄色液体として得た。
窒素雰囲気下、[3-クロロ-4-(トリフルオロメチル)フェニル]メタノール(800 ng)とフェニルボロン酸(1.90 g)のトルエン溶液(16 mL)に、リン酸カリウム(1.61 g)、酢酸パラジウム(42.6 mg)及びジシクロヘキシル(2’,6’-ジメトキシビフェニル-2-イル)ホスフィン(195.0 mg)を25℃で加えた。反応混合物は、100℃まで昇温後、15時間攪拌した。反応液に水(30 mL)を加え、EtOAc(30 mL)で3回抽出した。有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置、ヘキサン:EtOAc=70:30~50:50)で精製し、[6-(トリフルオロメチル)ビフェニル-3-イル]メタノール(678.3 mg)を黄色固体として得た。
{3-クロロ-4-[(1S)-2,2,2-トリフルオロ-1-メチルエトキシ]フェニル}メタノール (284 mg)とSOCl2(179 μL)のDCM溶液(7 mL)を室温で2時間撹拌した。反応液を水に注ぎ、クロロホルムで抽出した。有機層をbrineで洗浄し、MgSO4で乾燥後、減圧下濃縮し、2-クロロ-4-(クロロメチル)-1-[(1S)-2,2,2-トリフルオロ-1-メチルエトキシ]ベンゼン(285 mg)を無色液体として得た。
4-ブロモ-3-安息香酸メチル (300 mg)のTHF溶液 (1 mL)にシクロペンチルジンクブロマイド (9.6 mL)とパラジウム - トリ-tert-ブチルホスフィン (1:2) (123 mg)を加えた。反応液を室温で20時間撹拌した。0℃で飽和NH4Cl水溶液を加え、セライト濾過、EtOAcで抽出した。有機層をbrineで洗浄し、MgSO4で乾燥した。減圧下溶媒を留去し、シリカゲルカラムクロマトグラフィー(ヘキサン:EtOAc=100:0-90:10)で精製し、 3-クロロ-4-シクロペンチル安息香酸メチル (280 mg)を黄色固体として得た。
(7-{[2,4-ビス(トリフルオロメチル)ベンジル]オキシ}-5-フルオロ-2H-クロメン-3-イル)メタノール (200 mg)のMeCN溶液 (5 mL)にトリフェニルフォスフィンジブロマイド(240 mg)を加えた。反応液を室温で2時間撹拌し、減圧下濃縮した。残渣にEtOAcとIPEを加え、生じた固体をろ去し、ろ液を濃縮することで7-{[2,4-ビス(トリフルオロメチル)ベンジル]オキシ}-3-(ブロモメチル)-5-フルオロ-2H-クロメン (230 mg)を茶色液体として得た。 60%水素化ナトリウム (24 mg)を0℃でDMF溶液 (5 mL)に加え、続いて1H-ピラゾール-4-カルボン酸エチル (76 mg)を加えた。 混合物を室温で0.5時間撹拌し、 7-{[2,4-ビス(トリフルオロメチル)ベンジル]オキシ}-3-(ブロモメチル)-5-フルオロ-2H-クロメン (220 mg)のDMF溶液 (5 mL)を0℃で加えた。 反応液を室温で13時間撹拌し、飽和NH4Cl溶液でクエンチし、EtOAcで抽出した。有機層をbrineで洗浄し、MgSO4で乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー (ヘキサン:EtOAc=100:0-70:30)で精製して1-[(7-{[2,4-ビス(トリフルオロメチル)ベンジル]オキシ}-5-フルオロ-2H-クロメン-3-イル)メチル]-1H-ピラゾール-4-カルボン酸エチル (111 mg)を淡黄色固体として得た。
4-イソプロポキシ-3-(トリフルオロメチル)ベンゾニトリル (2.4 g)と5M NaOH (50 mL)のEtOH溶液 (50 mL)を18時間加熱還流した。室温に冷却した後、塩酸で酸性とし、クロロホルムで抽出した。有機層をbrineで洗浄し、MgSO4で乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー (クロロホルム:MeOH=100:0-95:5)で精製した。生成物をヘキサンで洗浄して4-イソプロポキシ-3-(トリフルオロメチル)安息香酸 (2.2 g)を白色固体として得た。
5-ブロモ-3-(トリフルオロメチル)-2-{[(2S)-1,1,1-トリフルオロプロパン-2-イル]オキシ}ピリジン(500 mg)をDMSO(5 mL)とMeOH(5 mL)の混合溶媒に溶解する。次にTEA(0.42 mL)を25℃で加えた後、Pd(OAc)2(17 mg)並びにDPPP(60 mg)を25℃で加えた。CO雰囲気下、70℃で15間攪拌した。反応溶液に水 30 mlを加え、EtOAc 20 mlで3回抽出した。有機層をbrineで洗浄し、MgSO4で乾燥後、ろ過し、ろ液を濃縮した。残渣をシリカゲルカラムクロマトグラフィー(自動精製装置。展開液;ヘキサン:EtOAc=100:0-80:20)で精製することにより5-(トリフルオロメチル)-6-{[(2S)-1,1,1-トリフルオロプロパン-2-イル]オキシ}ニコチン酸メチルエステル 403 mgを黄色固体として得た。
(1-メチル-1,2,3,6-テトラヒドロピリジン-4-イル)酢酸エチル (2.05 g)のジクロロエタン溶液 (14 mL)にクロロギ酸-1-クロロエチル (1.5 mL)を0℃で加えた。反応液を2.5時間加熱還流した後、減圧下濃縮した。残渣をMeOH (14 mL)に溶解し、 1時間加熱還流した。減圧下濃縮後、残渣をアミノカラムクロマトグラフィー (クロロホルム:メタノール=100:0-80:20)で精製して1,2,3,6-テトラヒドロピリジン-4-イル酢酸エチル (130 mg)を茶色液体として得た。
5,6-ジクロロニコチン酸メチル (1.5 g)と60%水素化ナトリウム (640 mg)をTHF (45 mL)に溶解した。ここに、1,3-ジフルオロプロパン-2-オール (1.5 g)を0℃で加えた。0℃のままで3時間攪拌し、反応溶液をNH4Cl水でクエンチした。EtOAcで抽出し、有機層をMgSO4で乾燥後、ろ過して乾燥剤を除き、減圧下溶媒を留去した。シリカゲルカラムクロマトグラフィー(ヘキサン:AcOEt=100:0-50:50)により精製し、5-クロロ-6-[(1,3-ジフルオロプロパン-2-イル)オキシ]ニコチン酸メチル (1.56 g)を無色液体として得た。
5-クロロ-6-[(1,3-ジフルオロプロパン-2-イル)オキシ]ニコチン酸メチル (1 g)のTHF溶液 (20 mL)に0.99M DIBALのトルエン溶液 (11.3 mL)を0℃で滴下した。0℃のまま2時間攪拌した後、反応溶液をロッシェル塩水溶液の中に注ぎ、室温で1時間攪拌した。EtOAc水系で抽出し、有機層をbrineで洗浄、MgSO4で乾燥後、ろ過して乾燥剤を除き減圧下溶媒を留去した。シリカゲルカラムクロマトグラフィー(Hex:AcOEt=98:2-70:30)で精製し、{5-クロロ-6-[(1,3-ジフルオロプロパン-2-イル)オキシ]ピリジン-3-イル}メタノール (650 mg)を無色液体として得た。
5,6-ジクロロニコチン酸 (2.2 g)に1,1,1-トリメトキシエタン (4.3 mL)を加え、120℃にてマイクロウエーブを15分間照射した。反応混合物をEtOAcに溶解し、水で洗浄した。有機層をMgSO4で乾燥し、溶媒を減圧下留去した。 残渣のシリカゲルカラムクロマトグラフィー(ヘキサン:EtOAc=85:15-80:20)で精製することにより5,6-ジクロロニコチン酸メチル (2.2 g)を白色固体として得た。
4-ピリジル酢酸エチル (2 g)のMeCN溶液 (20 mL)にヨードメタン (2.3 mL)を加えた。反応液を室温で終夜撹拌し、減圧下濃縮した。残渣にIPEを加え生じた固体をろ取した。固体をMeOHに溶解し水素化ホウ素ナトリウム (916 mg)を15℃以下で加えた。 反応液を室温で6時間撹拌した後、水を加えクロロホルムで抽出した。有機層をbrineで洗浄し、MgSO4で乾燥し、減圧下濃縮した。残渣をカラムクロマトグラフィー (クロロホルム:MeOH=100:0-90:10)で精製して(1-メチル-1,2,3,6-テトラヒドロピリジン-4-イル)酢酸エチル (2.08 g)を淡黄色液体として得た。
{3-(トリフルオロメチル)-4-[(1S)-2,2,2-トリフルオロ-1-メチルエトキシ]フェニル}メタノール(200 mg)のジクロロエタン(5 mL)溶液に塩化チオニル(111 μL)と触媒量のDMFを加え60℃で2時間攪拌した。反応液を減圧下濃縮した後、残渣に(3R)-1-[(7-ヒドロキシ-2H-クロメン-3-イル)メチル]ピペリジン-3-カルボン酸エチル(175 mg)のDMF(8.75 mL)溶液、炭酸カリウム(152 mg)を順次加え、80℃で2時間攪拌した。反応液を室温まで冷却したのち水に注ぎ、EtOAcで抽出した。有機層を水、brineで順次洗浄したのち無水硫酸ナトリウムで乾燥し、溶媒を留去した。残渣をシリカゲルカラムクロマトグラフィーで精製し、(3R)-1-[(7-{[3-(トリフルオロメチル)-4-{[(2S)-1,1,1-トリフルオロプロパン-2-イル]オキシ}ベンジル]オキシ}-2H-クロメン-3-イル)メチル]ピペリジン-3-カルボン酸エチル(271 mg)を黄色油状物質として得た。
4-クロロ-3-(トリフルオロメチル)ベンゾニトリル ( 1.5 g)、鉄(III)アセチルアセトナート (130 mg)と1-メチルピロリジン-2-オン (4 mL)のTHF溶液 (45 mL)に1MシクロペンチルマグネシウムブロマイドのTHF溶液 (8.8 mL)を5℃で加え室温で0.5時間撹拌した。ジエチルエーテルで希釈し、1M 塩酸をゆっくり加え、EtOAcで抽出した。有機層をbrineで洗浄し、MgSO4で乾燥し、減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー (ヘキサン:EtOAc=100:0-95:5) で精製し、4-シクロペンチル-3-(トリフルオロメチル)ベンゾニトリル (367 mg)を白色固体として得た。
7-(メトキシメトキシ)-2H-クロメン-3-カルバルデヒド(5.00 g)と(3R)-ピペリジン-3-カルボン酸エチル(4.20 mL)のジクロロエタン(150 mL)溶液にナトリウムトリアセトキシボロヒドリド(12.0 g)を加え80℃で4時間攪拌した。反応液を室温まで冷却したのち飽和NaHCO3水を加え、クロロホルムで抽出した。有機層をbrineで洗浄したのち無水硫酸ナトリウムで乾燥し、溶媒を留去した。残渣をシリカゲルカラムクロマトグラフィー(ヘキサン:EtOAc=4:1)で精製し、(3R)-1-{[7-(メトキシメトキシ)-2H-クロメン-3-イル]メチル}ピペリジン-3-カルボン酸エチル(7.30 g)を黄色油状物質として得た。
1-[(7-{[2,4-ビス(トリフルオロメチル)ベンジル]オキシ}-5-フルオロ-2H-クロメン-3-イル)メチル]ピロリジン-3-カルボン酸(98 mg)のDMF(2 mL)溶液にCDI(46 mg)を添加し、70℃で12時間攪拌した。反応液にメタンスルホンアミド(27 mg)及びDBU(43 mg)を順次加え、12時間攪拌した。反応液にAcOHを加えて減圧下濃縮し、残渣を逆相カラムクロマトグラフィー(H2O:MeCN=100:0~90:10)で精製し、黄色アモルファス(70 mg)を得た。この黄色アモルファスをジオキサン(1 mL)に溶解し、4M HCl/ジオキサン溶液(1 mL)を加えて撹拌後、減圧下濃縮した。残渣をヘキサンで洗浄し、1-[(7-{[2,4-ビス(トリフルオロメチル)ベンジル]オキシ}-5-フルオロ-2H-クロメン-3-イル)メチル]-N-(メチルスルホニル)ピロリジン-3-カルボキサミド塩酸塩(60 mg)を淡黄色固体として得た。
ピロリジン-3-カルボン酸塩酸塩のMeOH溶液にTEAを加えて室温で10分撹拌した。反応混合物を減圧下濃縮し、7-{[4-フェニル-5-(トリフルオロメチル)-2-チエニル]メトキシ}-2H-クロメン-3-カルボアルデヒドのMeOH溶液と、AcOHを室温で加えた。反応混合物を70℃まで加熱し2時間攪拌し、放冷して25℃にし、NaBH3CNを室温で加え、70℃で5時間攪拌した。反応液を逆相カラムクロマトグラフィー(MeCN:H2O=20:80~50:50)で精製し、得られた白色固体をジイソプロピルエーテルで洗浄し、1-[(7-{[4-フェニル-5-(トリフルオロメチル)-2-チエニル]メトキシ}-2H-クロメン-3-イ)メチル]ピロリジン-3-カルボン酸を白色固体として得た。
ピロリジン-3-カルボン酸塩酸塩(165 mg)をMeOHに溶解し、TEAを加え、室温で1時間撹拌した。反応混合物を減圧下濃縮した後、7-{[2,4-ビス(トリフルオロメチル)フェニル]エチニル}-5-フルオロ-2H-クロメン-3-カルボアルデヒドのMeOH(8 mL)溶液とAcOH (0.5 mL)を室温で加えた後、70℃で0.5時間攪拌した。室温まで放冷し、反応混合物にNaBH3CN(57 mg)を室温で加え、50℃で2時間攪拌した。LCで反応が終了したことを確認した後、反応液を逆相カラムクロマトグラフィー(MeCN:H2O=20:80-50:50)で精製し、得られたアモルファス(233 mg)をジオキサン(1 mL)に溶解し、4M HCl/ジオキサン溶液(1 mL)を加えた。反応液を減圧下濃縮し、残渣をMeCNで洗浄し、1-[(7-{[2,4-ビス(トリフルオロメチル)フェニル]エチニル}-5-フルオロ-2H-クロメン-3-イル)メチル]ピロリジン-3-カルボン酸塩酸塩(185 mg)を淡黄色固体として得た。
1-[(7-{[2,4-ビス(トリフルオロメチル)ベンジル]オキシ}-5-フルオロ-2H-クロメン-3-イル)メチル]-1H-ピラゾール-4-カルボン酸エチル (100 mg)と1M NaOH水溶液 (0.55 mL)のEtOH/THF (3 mL / 1 mL)溶液を100℃で2時間撹拌した。1M HClで中和し、クロロホルムで抽出した。有機層をbrineで洗浄し、MgSO4で乾燥し減圧下濃縮した。残渣をシリカゲルカラムクロマトグラフィー (クロロホルム:メタノール=100:0-90:10)で精製し、得られた固体をIPEで洗浄して、1-[(7-{[2,4-ビス(トリフルオロメチル)ベンジル]オキシ}-5-フルオロ-2H-クロメン-3-イル)メチル]-1H-ピラゾール-4-カルボン酸 (71 mg)を白色固体として得た。
(3R)-1-[(7-{[3-(トリフルオロメチル)-4-{[(2S)-1,1,1-トリフルオロプロパン-2-イル]オキシ}ベンジル]オキシ}-2H-クロメン-3-イル)メチル]ピペリジン-3-カルボン酸エチル(271 mg)のEtOH(5.4 mL)-THF(2.7 mL)溶液に1M NaOH水溶液(923 μL)を加え、50℃で2時間攪拌した。反応液を室温まで冷却した後、1M 塩酸(923 μL)を加え、溶媒を留去した。残渣を逆相クロマトグラフィー(H2O:MeCN=100:0-30:70)で精製し、黄色油状物質を得た。これをジオキサン(3 mL)に溶解し、4N HCl/dioxane(1 mL)で処理し、IPEで洗浄し、(3R)-1-[(7-{[3-(トリフルオロメチル)-4-{[(2S)-1,1,1-トリフルオロプロパン-2-イル]オキシ}ベンジル]オキシ}-2H-クロメン-3-イル)メチル]ピペリジン-3-カルボン酸塩酸塩(215 mg)を白色粉末として得た。

- 下式(I)で示される2H-クロメン化合物若しくはその誘導体又はその塩。

Aは、低級アルキル、シクロアルキル、アリール、又は、ヘテロアリールであり、ここで、アリール及びヘテロアリールは、それぞれ1から5個の同一又は異なったR1で置換されていてもよく、
R1は、ハロゲン、-CN、-NO2、低級アルキル、低級アルケニル、低級アルキニル、ハロゲノ低級アルキル、アリール、ヘテロアリール、シクロアルキル、-OH、-O-(低級アルキル)、-O-(ハロゲノ低級アルキル)、-O-(アリール)、-O-(シクロアルキル)、-O-(ヘテロアリール)、-NH2、-NH(低級アルキル)、-NH(ハロゲノ低級アルキル)、-N(低級アルキル)2、又は、環状アミノであり、ここで、アリール、ヘテロアリール、シクロアルキル及び環状アミノは、それぞれ、ハロゲン、-CN、低級アルキル、及び、ハロゲノ低級アルキルからなる群から選択される1から5個の同一又は異なった置換基で置換されていてもよく、

1-[(7-{[4-(2,2,2-トリフルオロエトキシ)-3-(トリフルオロメチル)ベンジル]オキシ}-2H-クロメン-3-イル)メチル]ピペリジン-4-カルボン酸。 - 請求項1に記載される、2H-クロメン化合物若しくはその誘導体又はその塩及び製薬学的に許容される賦形剤を含有する医薬組成物。
- S1P1アゴニストである請求項12に記載の医薬組成物。
- S1P1の関与する望ましくないリンパ球浸潤によって引き起こされる疾患の予防若しくは治療用医薬組成物である請求項13に記載の医薬組成物。
- S1P1の関与する望ましくないリンパ球浸潤によって引き起こされる疾患が、ヒト又は動物における臓器、骨髄又は組織の移植時の拒絶もしくは移植片対宿主病、自己免疫疾患又は炎症性疾患である請求項14記載の医薬組成物。
- ヒト又は動物における臓器、骨髄又は組織の移植時の拒絶もしくは移植片対宿主病の予防若しくは治療用医薬組成物である請求項15記載の医薬組成物。
- 多発性硬化症の予防若しくは治療用医薬組成物である請求項15記載の医薬組成物。
- 請求項1に記載される、2H-クロメン化合物若しくはその誘導体又はその塩の有効量を患者に投与することを含む、S1P1の関与する望ましくないリンパ球浸潤によって引き起こされる疾患の予防若しくは治療方法。
- 請求項1に記載される2H-クロメン化合物若しくはその誘導体又はその塩の、S1P1の関与する望ましくないリンパ球浸潤によって引き起こされる疾患の予防もしくは治療への使用。
- S1P1の関与する望ましくないリンパ球浸潤によって引き起こされる疾患の予防もしくは治療に使用するための、請求項1に記載される2H-クロメン化合物若しくはその誘導体又はその塩。
