Animals were selected to provide control and treatment groups of approximately equal average tumour volume.

  • the organic phase was washed with water and with brine, dried over magnesium sulphate and evaporated.
  • the residue was purified by column chromatography on silica using increasingly polar mixtures of methylene chloride and a saturated methanolic ammonia solution as eluent.
  • the material so obtained was dissolved in diethyl ether and a solution of hydrogen chloride in diethyl ether (IM, 2 ml) was added.

  • the 4-aminobenzofuran used as a starting material was prepared as follows :- Allyl bromide (12.5 ml) and sodium iodide (1 g) were added in turn to a stkred mixture of methyl 3-hydroxybenzoate (20 g), potassium carbonate (22 g) and acetone (200 ml) and the resultant mixture was stireed at ambient temperature for 16 hours. The mixture was filtered and the solvent was evaporated. The residue was partitioned between methylene chloride and water. The organic phase was dried over magnesium sulphate and evaporated and the residue was purified by column chromatography using increasingly polar mixtures of petroleum ether (b.p. 60-80°C) and ethyl acetate as eluent.

  • a mixture of benzofuran-4-carboxylic acid (0.5 g), diphenylphosphoryl azide (1.2 ml), triethylamine (0.79 ml) and tert-butanol (1.5 ml) were stirred and heated to reflux for 18 hours. The mixture was cooled to ambient temperature and partitioned between ethyl acetate and water.
  • Trifluoroacetic acid (2.5 ml) was added to a solution of tert-butyl benzofuran- 4-carbamate (0.65 g) and methylene chloride (20 ml) which had been cooled to 0°C and the mixture was stirred for lhour. The mixture was allowed to warm to ambient temperature and stirring was continued for 1.5 hours. The mixture was evaporated and the residue was partitioned between ethyl acetate and water. The organic phase was dried over magnesium sulphate and evaporated. The resultant oil was purified by column chromatography using a 4: 1 mixture of methylene chloride and petroleum ether (b.p. 60-80°C) as eluent.
  • Diethyl azodicarboxylate (0.25 g) was added dropwise to a suspension of 4-chloro- 3-cyano-7-hydroxy-6-methoxyquinoline (0.2 g; , prepared as described in International Patent Application WO 00/68201, disclosed as compound (7) within Preparation 1 thereinj, l-(3-hydroxypropyl)-4-methylpiperazine (0.202 g), triphenylphosphine (0.447 g) and methylene chloride (5 ml) and the mixture was stfrred at ambient temperature for 2 hours.
  • the resultant mixture was evaporated and the residue was purified by column chromatography on silica using initially increasingly polar mixtures of methylene chloride and ethyl acetate followed by increasingly polar mixtures of methylene chloride, ethyl acetate and a saturated methanolic ammonia solution as eluent.
  • the material so obtained was triturated under diethyl ether.

  • the aerosol formulations (h)-(k) may be used in conjunction with standard, metered dose aerosol dispensers, and the suspending agents sorbitan trioleate and soya lecithin may be replaced by an alternative suspending agent such as sorbitan monooleate, sorbitan sesquioleate, polysorbate 80, polyglycerol oleate or oleic acid.
