Abstract

Cette invention concerne des systèmes et des procédés utilisés pour le criblage à fort débit de composés présentant une affinité pour une cible biologique. Le système repose sur l'emploi d'un faisceau de capillaires présentant deux extrémités distinctes, les capillaires étant reliés à une extrémité à des composés stockés dans des réservoirs séparés et étant reliés et traités à l'autre extrémité de manière à former un microréseau bidimensionnel. L'invention porte sur un faisceau de capillaires qui comportent à l'une de leurs extrémités des puits de réaction pour hybridation et réaction des composés. Les capillaires peuvent être photoconducteurs. L'invention concerne également divers procédés permettant d'identifier un composant cible dans un liquide au moyen dudit faisceau de capillaires, ainsi que des procédés de fabrication de ce faisceau. Elle porte également sur une chambre de réaction guidée par effet tensioactif, ainsi que sur des procédés et des opérations chimiques permettant de fabriquer et d'utiliser une telle chambre de réaction pour des essais de liaison et d'hybridation. Sont en outre décrits des systèmes garantissant un dosage précis de liquides dans les capillaires et au niveau des chambres de réaction, y compris du puits de réaction « virtuel » guidé par effet tensioactif. L'invention concerne également des criblages à fort débit réalisés au moyen de capillaires recouverts intérieurement de fibre optique. Les ensembles de capillaires s'utilisent aussi bien avec des sondes liquides qu'avec des sondes immobilisés sur leurs parois.
