Description
明 細 書 打錠時に於ける応力分散方法 [技術分野 ]
本発明は、 打錠時に於けるマイク口カブセルの破壊あるいは薬物 の変性を低減するための応力分散剤を含有することを特徴と した打 錠用組成物および打錠時に於ける応力分散 &法に関する。
[背景技術 ]
従来、 マイ クロカブセル化した医薬品の粉末、 酵素あるいは低融 点物質などは、 打錠時の応力により変質または破壊され、 マイク π 力ブセルの破壊、 酵素活性の消失などの打錠障害等を起こ してい た。 そのため打錠時のマイク口カブセルの破壊を防止するために は、 大量の担体、 例えば、 ラク トース、 微細結晶セルロースおよび 澱粉の混合物が必要であることが知られている。 これにより錠剤は 許容できない程大きなものとなっていた。
また、 打錠時に於けるマイクロカブセルの破壊を低減するための 技術と して、 特開昭 53 - 142520号公報に蠟様物質を配合した技術が 特公昭 57-36893号に打錠圧を調整し成形した多層錠剤の技術が開示 されている。 しカゝしながら、 特開昭 53 - 142520号公報の錠剤は崩壊 性、 薬物の放出性および錠剤の硬度における問題があり、 特公昭 57 -36893号成形が複雑であるという欠点を有している。
[発明の開示]
本発明者らは、 この従来技術の不備な点を解決すべく銳意研究の 結果、 マイク口カブセル化した粉末医薬品にゼラチン微小球及び Z または発泡体を配合することにより打錠時の応力を分散できること を見出し、 この知見に基づいて本発明を完成するに至った。
本発明は、 ゼラチン微小球及び/または発泡体を応力分散剤とし て配合することを特徴とした打錠用組成物および打錠時の応力分散 ガ法でのる。
本発明に用いられるゼラチン微小球の形状に近いものとしては、 特開昭 60-19033号公報に微小中空物、特開昭 60-179132号公報に中空 マイ クロバル一ン、特開昭 60 - 215365号公報に真球ゼラチン粒が知ら れている。 ゼラチン微小球の大きさは、 直径 0. 001〜2. 0咖、 好まし くは 0. 005〜2. 0讓である。 また、 ゼラチン微小球は、 含水量 5〜: L5 %であることが好ましく、 分子量 5, 000〜25, 000、 ゼリー強度 50~350 ブルーム、 更に好ましくは分子量8,000〜20,000、 ゼリー強度 200ブ ルーム以上のゼラチンを用いて製造することが好ましい。 この際、 必要に応じて、 ゼラチン微小球に可塑剤を配合することもできる。 可塑剤と しては、 グリセリン、 プロピレングリコール、 砂糖、 ブド ゥ糖、 水あめ、 ハチミツ等が使用できる。
このゼラチン微小球の製造方法を以下に示す。
ゼラチンの加温した 5〜 50%の水溶液に 必要に応じて可塑剤を 加え、 次いで、 加温したゼラチン水溶液と混ざり合わない液体(流 動パラフ ィ ン、 シリコン油、 バナセート、 ャシ油、 ゴマ油、 小麦胚
芽油、 紅花油など、 ゼラチン水溶液と混ざり合わない液体であれば 何でも使用することができる。 )に入れ、 攪拌機で攪拌してゼラチ ン水溶液を微小液滴と した後、 攪袢しながら冷却し、 ゼラチン水溶 液の微小液滴を凝固させ取り出し、 乾燥後、 へキサン、 アセ トン、 キシレン、 メ タノール、 エタノール、 I P Aなどの有機溶媒でゼラ チン水溶液と混ざり合わない液体を洗い流し、 更に乾燥する。
また本発明に用いられるゼラチン発泡体は、 発泡させたゼラチン を粉砕して粉末と したもので、 大きさは、 l(h200メッシュ、 好まし くは 16〜; 100メ ッシュである。 また、 ゼラチン発泡体は、 含水量は 5~ 15%であるこ とが好まし く、 ゼラチンの分子量5,000〜25,000、 ゼ リ一強度 50~350ブルーム、 更に好ましくは分子量8,000〜20,000、 ゼ リー強度 200ブルーム以上のゼラチンを用いて製造することが好ま しい。
このゼラチン発泡体は、 約 60°Cに加温した 5〜50%のゼラチン水 溶液に発泡剤(例えば、 界面活性剤の様に発泡するものであれば使 用することができる。 )に入れ、 攪拌機で高速攪拌して発泡させた 後、 セノ、 'レター〔ポリエチレンシート、 ポリプロピレンシート、 ポ リエステルシー ト、 紙/ポリエチレンラ ミ ネ一 トフイルムなどのブ ラスチックフ ィルム、 あるいはその表面にシリコン加工を施したも のなど、 乾燥後発泡ゼラチンから剝離することができるものである れば、 何でも使用するこ とができる。 )上に展延し、 lcrc以下の冷 蔵庫で冷凍凝固し、 ゲル(ゼリー)状のシートと し、 乾燥後粉砕機 で粉砕後分級することによ り製造することができる。

- - この様にして得られたゼラチン発泡体及び Ζまたは微小球の他に 本発明の打錠剤用組成物には、 添加物と して、 デンプン、 乳糖、 微 結晶セルロース(M C C:)、 白糖、 ブドウ糖、 尿素、 炭酸カルシゥ ム、 メタケイ酸アルミ ン酸マグネシウム、 合成ケィ酸アルミニウム などの賦形剤、 ボリ ビニルアルコール( P V A:)、 ポリ ビニルピロ リ ドン( Ρ V Ρ ) 、 ノヽ イ ド口キシブ口ピルメチルセルロース( Η Ρ M C ) 、 ノヽ ィ ドロキシブ口ビルセルロース( H P C )、 ビニルァセ テー ト、 結晶セルロース、 カルボキシメチルセルロース ( C Μ C ) , 寒天、 アルギン酸ナ トリウム、 デキス トリン、 ゼラチン、 グ ルコース、 ァラ ビヤゴム、 トラガン トゴムなどの結合剤、 C M C— カルシウム、 C M C—ナトリウム、 無水ケィ酸、 ハイ ドロキシブ口 ビルスターチ( H P S )などの崩壤剤、 ステアリン酸、 ステアリン 酸マグネシウム、 ステアリ ン酸カルシウム、 タルク、 リ ン酸水素力 ルシゥム、 カルナゥバロウ、 サラシミツロウ、 軽質無水ケィ酸、 合 成ケィ酸アル ミニウム、 天然ケィ酸アルミニウム、 乾燥水酸化アル ミニゥム · ゲルなどの滑沢剤、 吸収促進剤、 吸着剤、 色素、 香料な どを添加することができる。
本発明の錠剤に適した主薬は、 菌、 酵素あるいは低融点物質、 マ イク口カブセル化された医薬品である。 即ち、 打錠時に変質ある.い はその構造が破壊されるものである。 例えば、 腸溶性あるいは徐放 性のマイ クロ力ブセル化されたァスビリン、 ビタ ミン.、 蛋白分解酵 素、 ビフ ィ プス菌、 イブプロフェンなどである。
本発明の錠剤の製造方法は、 主薬と添加物に乾燥し分級したゼラ
チン発泡体及び または微小球を添加し混合し、 打錠用組成物を得 る。 ゼラチン発泡体及び/または微小球の添加量は 5重量%〜 5 0 重量%、 好ましくは、 1 0重量%〜 4 0重量%である。
この混合物を、 打錠の一般製法によつて各種の錠剤を得ることが でき る。
[発明を実施するための最良の形態 ] 本発明を実施例および試験例を挙げてさらに具体的に説明する。 実施例 1 〜 9
(1)ゼラチン(日局) 150 gに精製水(日局) 150 gを加え水溶中で 60°Cに加温してゼラチンを溶解した。 溶解後、 このゼラチン水溶液 を 60°Cに加温した流動パラフ ィ ン 500 g中に入れ、 攪拌機で攪 拌 (約 1 , 000 r p m ) して、 ゼラチン水溶液を 200〃〜 2. 5mmの球状液 滴と し、 その後流動パラフィ ンを冷却しゼラチン水溶液滴を凝固 し、 更に約 5 °Cの冷蔵庫で 12時間冷却凝固した。 冷却凝固後ゲル化 したゼラチン水溶液滴の微小球を流動バラフィン中から取り出し、 へキサンで流動バラフイ ンを洗い落とし、 温度 23。C;、 湿度 40%以下 の除湿乾燥機に入れ 16時間乾燥しゼラチン微小球よりなる応力分散 剤を得た。 得られたゼラチン微小球は直径が約 100 ^〜1. 5mm、 水分 含量が 11 %であった。
(2)前記のゼラチン微小球を篩で L , M , Sの 3種に分級した。

この分級されたゼラチン微小球を用い、 表 2の処方でマイクロ力 ブセル含有錠剤を製造した。 表 2 ^(の処方( 3錠中)

、 M C Cは微結晶セルロース(旭化成社製, AvicelPli301 )であ る。 これらの組成物を単発式 打錠機(面積 1 c m 2、 平型)を使用 して直接粉 末圧縮法によって 1錠 400m g ,打錠圧 1 t にて 処方 粉体を圧縮成型した。 実施例 1 0〜: I 4
(1)ゼラチン (日局) 150 gに精製水(日局) 250 gを加え水溶中で 60 Cに加温してゼラチンを溶解した。 溶解後、 発泡剤としてラウリ ル硫酸ナ ト リ ウム lgを加えホモミキサ一で高速攪拌(約 7, 000 r p m ) して、 ゼラチン水溶液を発泡させた後、 ボリプロピレンシ一ト 上に約 5mmの厚みに展延し、 5°Cの冷蔵庫に入れゼラチン水溶液を冷 却凝固し、 ゲル化したシートとし、 その後温度 23 C、 湿度 40%以下 の除湿乾燥機に入れ 16時間乾燥し、 粉砕機で粉砕し、 ゼラチン発泡 体を得た。
得られたゼラチン発泡体のサィズは 0. 1-1. 0mm,水分含量は 12%で めった
(2)前記のゼラチン発泡体を篩で 16〜: 100メッシュに分級した。
この分級されたゼラチン発泡体を用い、 表 3の処方のマイクロカ ブセル含有錠剤を表 4の打錠圧力で製造した。

200 )、 M C Cは微結晶セルロース(旭化成社製, AvicelPli301 ) である。 これらの組成物を単発式打錠機(面積 1 c m 2、 平型)を 使用して直接粉末圧縮法によって 1錠 250τ¾の処方粉体を圧縮成型 した。 実施例 1 5〜: L 9

表 5の処方でゼラチンに濃グリセリンと精製水を加え水溶液中で 60。Cに加温してゼラチンを溶解した。 溶解後、 このゼラチン水溶液 を 60。Cに加温した流動パラフィ ン 500g中に入れ、 攪拌機で攪拌( 約 l,000rpm) して、 ゼラチン水溶液を 200 〜 2.5删の球状液滴と し、 その後流動パラフィ ンを冷却しゼラチン水溶液滴を凝固し、 更 に約 5。Cの冷蔵庫で 12時間冷却凝固後ゲル化したゼラチン水溶液滴 の微小球を流動バラフイ ンを洗い落とし、 温度 23。C、 湿度 40%以下 の除湿乾燥機に入れ 16時間乾燥したゼラチン微小球よりなる応力分 散剤を得た。 得られたゼラチン微小球は直径が約 100 ί〜: L.5删、 水 分含量が 11%以下であった。
(2)前記のゼラチン微小球を篩で 16〜: L00メッシュに分級した。 この分級されたゼラチン微小球を用い、.表 6の処¾でマイクロ力 ブセル含有打錠用組成物を製造した。 表 6 錠剤の処^ ( 3錠中) 実施例

ァス ピリ ンの検出は分光光度計(日立製作所製, U-3200型)を用 いて、 ァスピリンの最大吸収波長 275nmにて吸光度を測定して行 なった。

3 . ゼラチン微小球および Zまたは発泡体を 5〜 5 0重量%配合し たことを特徵とする打錠用組成物。
