Abstract

Una formulación de vacuna no acuosa de fase única que contiene un polipéptido inmunogénico disuelto en un solvente polar no acuoso. El polipéptido inmunogénico proviene de un organismo parásito, preferentemente de un organismo seleccionado del grupo compuesto por los nematodos y los insectos. En una realización preferida, el solvente no acuoso comprende dimetilsulfóxido (DMSO),si bien pueden utilizarse también otros solventes polares no acuosos. En su forma más conveniente, el DMSO comprende de hasta un 6% de solvente no acuoso, y el resto comprende un solvente hidroxílico polar. El solvente hidroxílico polar se selecciona del grupo integrado por el glicerol formol y el propilén glicol, ó una mezcla de los mismos. En particular, las formulaciones de la presente invención pueden almacenarse durante períodos prolongados a temperaturas ambientes de hasta 40sC aproximadamente sin perder eficacia, es decir, la capacidad de inducir una respuesta inmune protectora. La presente invención es particularmente útil para la administración conjunta de sustancias farmacéuticas que son inestables en solución acuosa, como ser un compuesto antihelmíntico y de manera más preferida es una lactona marcocíclica del grupo de compuestos integrados por la avermectina y la milbemicina.A single phase nonaqueous vaccine formulation containing an immunogenic polypeptide dissolved in a nonaqueous polar solvent. The immunogenic polypeptide comes from a parasitic organism, preferably from an organism selected from the group consisting of nematodes and insects. In a preferred embodiment, the nonaqueous solvent comprises dimethylsulfoxide (DMSO), although other non-aqueous polar solvents can also be used. In its most convenient form, DMSO comprises up to 6% non-aqueous solvent, and the remainder comprises a polar hydroxylic solvent. The polar hydroxylic solvent is selected from the group consisting of glycerol formaldehyde and propylene glycol, or a mixture thereof. In particular, the formulations of the present invention can be stored for prolonged periods at ambient temperatures of up to about 40sC without losing efficacy, i.e., the ability to induce a protective immune response. The present invention is particularly useful for the co-administration of pharmaceutical substances that are unstable in aqueous solution, such as being an anthelmintic compound and more preferably it is a framework cyclic lactone from the group of compounds composed of avermectin and milbemycin.
