Нами було вивчено різні комбінації тіотриазоліну з гліцином - 1:1; 1:2; 1:4; 1:5; 1:7 відповідно, в умовах моделювання гострого порушення мозкового кровообігу (ГПМК). ГАМК моделювали шляхом односторонньої перев'язки загальної сонної артерії у білих безпородних щурів-самців масою 160-170 г. Операцію проводили під тіопентал-натрієвим наркозом. Досліджувані комбінації вводили відразу після виходу тварин з наркозу, один раз на добу по 250 мг/кг, внутрішньошлунково у вигляді водного розчину за допомогою металевого зонда. На четверту добу експерименту у тварин забирали кров з черевної аорти і в сироватці визначали активність цереброспецифічного ізоензиму креатинфосфокінази - ВВ-КФК - маркера пошкодження головного мозку. Для створення ГПМК використовували класичну модель, яка полягає в одночасній перев'язці загальних сонних артерій. Таким чином, можна зробити наступні висновки. Призначення тваринам з ГЛМК фіксованої комбінації гліцину з тіотриазоліном призводило до значної активації окисної продукції енергії на дикарбоновій ділянці циклу Кребса, про що свідчить підвищення рівня малату і підвищення активності СДГ (табл. 3). При цьому спостерігалося підвищення активності цитохром-С- оксидази і рівня ізоцитрату (табл. 3), що забезпечувало підвищення продукції АТФ. Паралельно відмічалося зростання рівня АДФ і зниження рівня АМФ (табл. 2). Гліцин у поєднанні з тіотриазоліном пригнічував активність анаеробного гліколізу (знижував рівень лактату), зменшував ,витрати" гальмівних амінокислот у компенсаторному і енергетично менш вигідному шунті Робертса. Також підвищувався рівень глутамату, ГАМК та гліцину на фоні зниження активності ГДК (глутаматдекарбоксилази) і ГАМК-Т (ГАМК-трансферази) (табл. 4). Збільшення рівня гальмівних амінокислот під дією вказаної комбінації обмежувало дію ЗАК-системи мозку і, тим самим, посилювало сумарну нейропротективну дію препарату. Фіксована комбінація гліцину з тіотриазоліном чинила позитивний вплив на окисну продукцію енергії у головному мозку щурів з ГПМК, та інтенсифікувала транспорт і утилізацію енергії, про що свідчать відповідні підвищення активності м-КФК і АТФф-азної активності у серії тварин, яким вводили гліцин разом з тіотриазоліном (табл. 2).Thus, the following conclusions can be drawn. Glycine in combination with thiotriazoline inhibited the activity of anaerobic glycolysis (reduced the level of lactate), reduced the "expenditure" of inhibitory amino acids in the compensatory and energetically less profitable Roberts shunt. The level of glutamate, GABA and glycine also increased against the background of a decrease in the activity of GDK (glutamate decarboxylase) and GABA-T (GABA-transferases) (Table 4). The increase in the level of inhibitory amino acids under the influence of the indicated combination limited the action of the brain's ZAC system and, thereby, enhanced the total neuroprotective effect of the drug. The fixed combination of glycine with thiotriazoline had a positive effect on oxidative energy production in the main in the brains of rats with HPMC, and intensified the transport and utilization of energy, as evidenced by the corresponding increase in the activity of m-CFC and ATPase activity in a series of animals that were injected with glycine together with thiotriazoline (Table 2).
