Abstract
本發明係有關於包含發酵熟成諾麗、椰子糖或黑砂糖活性成分的組合物用於增進運動能力及疲勞恢復,更具體地,係有關於具有增加組織內的糖原儲存量和肝功能,提高運動執行能力以及抑制血中疲勞物質的蓄積的效果的用於運動執行能力及疲勞恢復增進的組合物。本發明具有在攝取發酵熟成諾麗及椰子糖複合組合物時,增加組織內的糖原儲存量和肝功能,可提高運動持續能力的運動執行能力提高及血中疲勞物質的蓄積的抑制的效果,由此提供食品產業上有用的運動能力提高及抗疲勞用食品組合物。本發明的組合物促進疲勞物質的減少,快速進行疲勞恢復或運動能力增強,由此可製備對疲勞恢復及運動能力增強有效的運動飲料用、功能性食品及藥學組合物製品。
Description
本發明係有關於包含發酵熟成諾麗、椰子糖或黑砂糖活性成分的組合物,更詳細地,係有關於具有增加組織內的糖原儲存量和肝功能,用於增進運動能力以及抑制血液中疲勞物質蓄累的效果,由此在食品產業上有用的用於增進運動能力及疲勞恢復的組合物。
通常,肌肉在不持續運動時,肌肉功能會因老化而降低,減少肌肉量及肌肉神經接點(neuromuscular junction,motor unit),身體容易感到疲勞且無力,導致身體的活力減少,生活品質急劇降低。
若出現感到疲勞的信號,則身體需要可以修養而恢復的時間,但是在忙碌的現代社會上難以正常保持如上疲勞和疲勞恢復的迴圈。因過勞引起的疲勞蓄累,稍微不慎誘發慢性疲勞,還可能誘發消化性潰瘍、高血壓、糖尿病等許多疾病。並且,癌、腦中風、心臟疾病是現代人的主要3大死亡原因,過勞是這些病的主要原因。
疲勞或壓力積累的表現是指身心的功能不順利執行的情況。通常,疲勞可成為肌肉收縮活動中所需的力不能充分發揮的狀態,即運動能力減少的狀態,與身體疲勞相反,壓力可理解為因精神上的超負荷而導致的身體節奏的不均衡。因此,廣義的疲勞是均包括勞累(fatigue)和壓力(stress)的概念,可被視為用於肉體或精神活動的能力的減少,具體地,勞累(fatigue)主要是身體疲勞,其指工作效率降低的狀態,以及壓力是精神疲勞,其指引起體內平衡被擾亂的狀態。
為了防止疲勞或壓力的累積,建議在適當飲食的同時進行定期運動,例如阻力訓練。近年來,隨著健康熱潮,對於享受生活的悠閒,預防和治療各種成人病,都突顯了運動的重要性。如上所述,為了提高生活品質,需要有規律的運動,不僅是運動選手,普通人在日常生活中也需要更多的能量及持久力。因此,用於提高身體運動能力的補充劑、功能性食品、食品組合物等的研究長期持續進行,實際上若服用類固醇、咖啡因等化合物,會增加運動能力。但是,這些藥物有可能伴隨致命的副作用,故而其用途極其受限。
據悉,自然界中存在的植物中含有大量具有防止疾病和抑制老化等生物調節功能的功能性成分,由此正在活躍進行有關天然食品材料的研究。近年來正在活躍進行利用如植物提取物一樣安全性得到保障的天然物來開發功能性補充劑的研究。
對此,本發明人持續進行有關發酵熟成諾麗和椰子糖的研究的結果,確認了通過這些複合物的攝取,可以增加運動能力並抑制血液中疲勞因素的積累,由此完成了本發明。

本發明的目的在於,解決前述問題及與其相關的其他問題。
本發明人在可有效增進運動能力及疲勞恢復的天然成分組合物相關研究中確認包含發酵熟成諾麗、椰子糖或黑砂糖活性成分的組合物具有增強運動能力及疲勞恢復,由此完成了本發明。
本發明的一目的在於,提供包含發酵熟成諾麗、椰子糖及黑砂糖活性成分的組合物用於增進運動能力和/或疲勞恢復。
本發明的另一目的在於,提供包含上述發酵熟成諾麗、椰子糖及黑砂糖的組合物的製備方法。
本發明的另一目的在於,提供運動能力和/或疲勞恢復增進用食品組合物。
本發明的另一目的在於,提供運動能力和/或疲勞恢復增進用藥物組合物。
本發明的另一目的在於,提供運動能力和/或疲勞恢復增進用醫藥外品組合物。
本發明的另一目的在於,提供運動能力和/或疲勞恢復增進用飼料組合物。
要根據本說明書中公開的發明技術思想來實現的技術問題,並不限定於用於解決以上提及的問題的技術問題,本發明的普通技術人員可通過以下的記載明確理解未被提及的另一技術問題。
[本發明的技術方案]
對其進行具體說明如下。另一方面,本申請中公開的各個說明及實施方式也可適用於各個其他說明及實施方式。即,本申請中公開的多種因素的所有組合屬於本申請的範疇中。並且,本申請的範疇不得被視為限定於以下描述的具體說明。
為了解決如上問題,本發明提供用於增進運動能力及疲勞恢復的組合物,上述組合物作為有效成分包含發酵熟成諾麗、椰子糖和/或黑砂糖。
以下,詳細說明本發明。
在本發明中,“發酵熟成諾麗”是指在諾麗中接種乳酸菌而發酵及熟成的諾麗。其中,諾麗不受其部位的限制,但是作為一個示例可使用諾麗果。
“發酵諾麗”可以是在諾麗果中接種以下7種乳酸菌中的任一個以上的發酵物:
(1)植物乳桿菌(Lactobacillus plantarum);
(2)副乾酪乳桿菌(Lactobacillus paracasei);
(3)鼠李糖乳桿菌(Lactobacillus rhamnosus);
(4)乾酪乳桿菌(Lactobacillus casei);
(5)發酵乳桿菌(Lactobacillus fermentum);
(6)羅伊氏乳桿菌 (Lactobacillus reuteri);以及
(7)乳酸乳球菌乳酸亞種(Lactococcus lactis subsp. lactis)。
上述“發酵諾麗”利用生物轉化能力優秀的乳酸菌來發酵,其可將作為諾麗中所含的配糖體化合物的東莨菪甙(Scopolin)、車葉草甙(Asperuloside)生物轉化為作為非配糖體化合物的東莨菪素(scopoletin)、去乙醯車葉草苷酸(deacetylasperulosidic acid,DAA)及車葉草苷酸(asperulosidic acid)。
在本發明中,上述“發酵諾麗”中生物轉化的東莨菪素的含量為100~2000μg/g(以發酵諾麗提取物固體含量60%為基準),去乙醯車葉草苷酸的含量為0.5~15mg/g(以發酵諾麗提取物固體含量60%為基準),車葉草苷酸的含量為0.2~5mg/g(以發酵諾麗提取物固體含量60%為基準)。
在本發明中,“發酵熟成諾麗”也達到與“發酵諾麗”中所含的功能成分東莨菪素、去乙醯車葉草苷酸及車葉草苷酸的含量相同。
本發明的“發酵熟成諾麗”可以是發酵熟成諾麗本身或從中獲得的提取物。例如,本發明的發酵熟成諾麗可以是在諾麗果接種乳酸菌而發酵及熟成,並將這些發酵熟成的發酵熟成諾麗榨汁而獲得液體的提取物。
在本說明書中,“提取物”是指在本領域中通用為粗提取物(Crude extract)的意思,但是在廣義上海包含將提取物進一步分餾(Fractionation)的分餾物的意思。即,發酵熟成諾麗提取物,不僅包含利用上述提取溶劑來獲得的提取物,還包含在此追加適用精製過程獲得的提取物。例如,使上述提取物經由具有規定的分子量截留(cut off)值的超濾膜而獲得的分餾、通過基於多種層析(大小、電荷、為了基於疏水性或親和性的分離而製備的)的分離等追加實施的多種精製方法獲得的分餾也包括在本發明的發酵熟成諾麗提取物中。
在本說明書中,“活性成分”是指包含實現發酵熟成諾麗或其提取物的功效或活性時充分的量。本發明是從作為天然植物材料的發酵熟成諾麗中提取的組合物,由於過量給藥也對人體無副作用,因此發酵熟成諾麗提取物包含在本發明的組合物的含量上限可由普通技術人員在適當的範圍內選擇而實施。
根據本發明的發酵熟成諾麗提取物是利用有機溶劑來提取的,作為提取溶劑可使用的溶劑為如下。
首先,適合作為極性溶劑的有(i)水,(ii)碳原子數為1至6醇(較佳地,甲醇、乙醇、丙醇、丁醇、正丙醇、異丙醇、正丁醇、1-戊醇、2-丁氧基乙醇或乙二醇),(iii)乙酸,(iv)二甲基甲醯胺(DMFO,Dimethyl-formamide)及(v)二甲基亞碸(DMSO,Dimethyl sulfoxide)等。
並且,適合作為非極性溶劑的有丙酮、乙腈、乙酸乙酯、乙酸甲酯、氟代烷、戊烷、己烷、2,2,4-三甲基戊烷、癸烷、環己烷、環戊烷、二異丁烯、1-戊烯、1-氯丁烷、1-氯戊烷、o-二甲苯、二異丙基醚、2-氯丙烷、甲苯、1-氯丙烷、氯苯、苯、二***、二乙基硫醚、氯仿、二氯甲烷、1,2-二氯乙烷、苯胺、二乙胺、醚、四氯化碳、二氯甲烷、石油醚及四氫呋喃(THF)等。
作為本發明的提取溶劑的代表性的示例,較佳地使用(a)水,(b)碳原子數為1至4的無水或低級醇(例如,甲醇、乙醇、丙醇、丁醇等),(c)上述低級醇和水的混合溶劑,(d)丙酮,(e)乙酸乙酯,(f)氯仿,(g)乙酸丁酯,(h)1,3-丁二醇,(i)己烷及(j)二***等,為了容易的提取,可利用水、乙醇或水和乙醇的混合物來提取。
並且,在本發明中利用的發酵熟成諾麗提取物,通過減壓蒸餾及冷凍乾燥或噴霧乾燥等之類的追加過程,能夠以粉末狀態製備。
在本發明中,其特徵在於,上述發酵熟成諾麗及椰子糖的重量比為70:30至95: 5,更較佳的重量比為80:20至90:10。
並且,其特徵在於,上述發酵熟成諾麗及黑砂糖的重量比為70:30至95: 5,更較佳的重量比為80:20至90:10。
並且,其特徵在於,上述發酵熟成諾麗、椰子糖及黑砂糖的重量比為90:5:5至70:15:15,更較佳為90:5:5。
根據本發明的一實施方式,本發明係有關於包含發酵熟成諾麗、椰子糖和/或黑砂糖活性成分的組合物用於增進運動能力或疲勞恢復。
上述食品組合物作為一例可包含保健食品組合物,上述保健食品組合物有各種食品類、飲料、口香糖、茶、維生素複合劑、保健食品類等,能夠以粉末、顆粒、片劑、膠囊或飲料的形態使用。各劑型的食品組合物,除了有效成分以外,可根據劑型或使用目的,普通技術人員容易地適當選擇相關領域中通常使用的成分來配合,當與其他原料同時適用時,可產生提升效果。
上述組合物在不損害本發明所目的的主要效果的範圍內,可包含給主要效果可帶來提升效果的其他成分等。例如,為了改善物性,還可包含香料、色素、殺菌劑、抗氧化劑、防腐劑、保濕劑、增粘劑、無機鹽類、乳化劑及合成高分子物質等添加劑。
當本發明的保健食品組合物使用為食品添加物時,原封不動地添加上述發酵熟成諾麗、椰子糖、黑砂糖或者可一起使用其他食品或食品成分,根據常規方法可適當使用。有效成分的混合量可根據其使用目的(預防、健康或治療處置)適當確定。上述食品的種類不受特殊限制。作為可添加上述發酵熟成諾麗、椰子糖、黑砂糖的食品的例,有肉類、香腸、麵包、巧克力、糖果類、速食類、餅乾類、披薩、速食麵、其他面類、口香糖類、包含霜淇淋類的乳製品、各種湯類、飲料水、茶、補液、酒精飲料及維生素複合劑等,均包含常規意義上的保健食品。
當本發明的組合物使用為健康飲料時,如常規的飲料一樣,可包含多種香味劑或天然碳水化合物等作為追加成分。上述的天然碳水化合物為葡萄糖、果糖之類的單糖;麥芽糖、蔗糖之類的二糖;糊精、環糊精之類的多糖;木糖醇、山梨醇、赤蘚糖醇等糖醇。作為甜味劑可使用索马甜、甜菊苷提取物之類的天然甜味劑,或者糖精或阿斯巴甜之類的合成甜味劑等。
本發明還提供包含上述發酵熟成諾麗、椰子糖和/或黑砂糖的組合物的製備方法。
根據本發明的一實施方式,本發明提供包含發酵熟成諾麗及椰子糖和/或黑砂糖的組合物的製備方法,其包括:獲得利用乳酸菌將諾麗進行發酵和/或熟成的發酵熟成諾麗的步驟;以及在上述發酵熟成諾麗追加椰子糖和/或黑砂糖而熟成的步驟。
在本發明中,提供包含發酵熟成諾麗、椰子糖和/或黑砂糖的組合物的製備方法,上述方法包括:步驟(a),在諾麗果接種乳酸菌,發酵及熟成,製備發酵熟成諾麗;步驟(b),將上述發酵熟成諾麗榨汁;以及步驟(c),在上述發酵熟成諾麗追加椰子糖和/或黑砂糖而混合。
在本發明中,製備方法還可包括步驟(d),該步驟(d)在上述步驟(c)中在發酵熟成諾麗混合椰子糖和/或黑砂糖之後,追加熟成。
在本發明中,上述發酵及熟成並不限定於此,在35至40℃下可進行48至168小時。
以將發酵過程中生成的發酵液撒布於發酵物的上部的方式實施上述熟成,該熟成實施1天至3天,更具體地每2天實施。
在本發明中,將椰子(椰子樹)、棕櫚樹、 棗椰樹、尼巴棕櫚葉、桄榔樹等棕櫚科所屬的椰子樹的樹液熬制而得的糖叫做棕櫚糖或香椰糖(palm sugar),椰子糖(coconut sugar)是指由椰子樹(coconut palm)的花蕾莖的樹液生產的香椰糖,其為商業上也能入手的物質。
椰子糖的主要結構成分,包含蔗糖(sucrose)70~79%、葡萄糖(glucose)及果糖(fructose)各自為3~9%(如Beck L., 2014, "Coconut sugar: Is it healthier than white sugar, or just hype?", The Globe & Mail, Retrieved 30 May 2015文獻中記載)。
黑砂糖(Muscovado)是將甘蔗榨汁液(汁)熬制而形成糖蜜、未精製的糖,即乾燥為深褐色的顆粒之後被粉碎而製備的糖,完全不含磷酸、甲酸、二氧化硫、防腐劑或精製的糖中使用的漂白劑或黏度調節劑之類的化學物質的糖。相比於加工的白糖或紅糖,糖蜜含量高,香味濃,顏色偏深。是商業上也能入手的物質。
根據本發明的一實施方式,本發明係有關於包含發酵熟成諾麗、椰子糖和/或黑砂糖活性成分的組合物用於增進運動能力或疲勞恢復。
根據本發明的藥物組合物,單獨包含活性成分或者能夠與藥學上允許的載體以適合的形態一起劑型化,並可追加包含賦形劑或稀釋劑。上述“藥學上允許的”在生理學上允許並給藥到人體時,通常是指不引起胃腸障礙、眩暈症等之類的過敏反應或與其類似的反應的非毒性的組合物。

本發明的組合物,通過任何方法也能給藥到包括人在內的哺乳動物中。例如,能夠口服或非口服給藥。非口服給藥方法不限定於此,但是可以是靜脈內、肌肉內、動脈內、骨髓內、硬膜內、心臟內、經皮、皮下、腹腔內、鼻腔內、腸管、局部或舌下或直腸內給藥。
本發明的藥學組合物根據如上所述的給藥途徑可劑型化為口服用或非口服用製劑。
就口服用製劑而言,本發明的組合物可利用本領域公知的方法劑型化為粉末、顆粒、片劑、丸劑、糖衣片劑、膠囊劑、液劑、凝膠劑、糖漿劑、漿料劑、懸浮液等。例如,口服用製劑,將活性成分與固體賦形劑配合之後將其粉碎,添加適合的補助劑之後,加工為顆粒混合物,由此可獲得片劑或糖衣片劑。作為適合的賦形劑的例,可包含乳糖、葡萄糖、蔗糖、山梨糖醇、甘露醇、木糖醇、赤蘚糖醇及麥芽糖醇等的糖類、包含玉米澱粉、麥澱粉、米澱粉及土豆澱粉等的澱粉類、包含纖維素、甲基纖維素、羧甲基纖維素鈉及羥丙基甲基-纖維素等的纖維素類、明膠、聚乙烯吡咯烷酮等之類的填充劑。並且,根據情況,可添加作為崩解劑的交聯聚乙烯吡咯烷酮、瓊脂、海藻酸或海藻酸鈉等。進而,本發明的藥學組合物還可包含抗凝集劑、潤滑劑、濕潤劑、香料、乳化劑及防腐劑等。

本發明的組合物的總有效量,能夠以單劑量(single dose)給藥到患者,並可通過以多劑量(multiple dose)長期給藥的分次治療方法(fractionated treatment protocol)給藥。本發明的藥物組合物,可根據疾病的程度來改變有效成分的含量。較佳地,對於本發明的藥物組合物的較佳的整體容量而言,按患者體重每1kg,每天可以是約0.01μg至10000mg,較佳地可以是0.1μg至500mg,最較佳地可以是100mg至500mg。但是,上述藥物組合物的容量,不僅考慮製劑化方法、給藥途徑及治療次數,而且也考慮患者的年齡、體重、健康狀態、性別、疾病的重症度、飲食及***率等多種因素來確定有關患者的有效劑量,因此當考慮到這個問題時,只要是本技術領域的普通技術人員就能確定本發明的組合物的適當的有效劑量。根據本發明的藥物組合物,只要呈現本發明的效果,其劑型、給藥途徑及給藥方法就不受特殊限制。
根據本發明的一實施方式,本發明係有關於包含發酵熟成諾麗、椰子糖和/或黑砂糖活性成分的組合物用於增進運動能力或疲勞恢復。
本發明中使用的術語“醫藥外品”是指出於診斷、治療、減輕或預防人或動物的疾病的目的來使用的物品中除了不是器具、機械或裝置的物品以及出於給人或動物的結構盒功能帶來藥理學影響的目的使用的物品中除了不是器具、機械或裝置的物品,作為一個具體例,可包含內服用製劑,但並不限定於此,醫藥外品的製劑化方法、容量、利用方法、結構成分等可從技術領域中公知的常規技術中適當選擇。
本發明的醫藥外品組合物,除了上述成分以外,根據需要還可包含藥學上允許的載體、賦形劑或稀釋劑。上述藥學上允許的載體、賦形劑或稀釋劑,只要不損害本發明的效果,就不受限制,例如,可包含填充劑、增量劑、結合劑、濕潤劑、崩解劑、表面活性劑、潤滑劑、甜味劑、芳香劑、保存劑等。
根據本發明的一實施方式,本發明係有關於包含發酵熟成諾麗、椰子糖和/或黑砂糖活性成分的組合物用於增進運動能力或疲勞恢復。
在本發明中,上述飼料組合物,能夠以常規的飼料形態進行劑型化,並可一同包含公知的飼料成分。並且,還可以使用為所用到的飼料中以添加劑形態添加的飼料添加劑。本發明的飼料添加劑相當於飼料管理法上的補助飼料,還可包含重碳酸鈉(蘇打(soda))、膨潤土(bentonite)、氧化鎂、複合礦物質等礦物質製劑,作為鋅、銅、鈷、硒等微量礦物質的礦物製劑,胡蘿蔔素、維生素E、維生素A、D、E、煙酸、維生素B複合體等維生素製劑,蛋氨酸、賴氨酸等氨基酸保護劑,脂肪酸鈣鹽等脂肪酸保護劑,益生菌(乳酸菌製劑)、酵母培養物、黴菌發酵物等益生菌、酵母劑等。
本發明的飼料或飼料添加劑可適用於包括哺乳類、家禽及魚類的多個動物飼料。
[對照先前技術之功效]
根據本發明包含發酵熟成諾麗和椰子糖或黑砂糖活性成分的複合組合物,具有運動能力及疲勞恢復增進效果。並且,上述發酵熟成諾麗,產生在諾麗果的發酵工程中可提高消化吸收的效果,促進消化力相對差的成人、老人的消化力,並促進排便活動,另一方面,由於是天然原料,因此無毒性和副作用,由此具有可放心攝取的效果。
並且,上述發酵熟成諾麗的製備工程,可大幅節減製備成本,由此具有大幅減少消費者的購買費用負擔的效果。
並且,發酵熟成諾麗和椰子糖或黑砂糖複合組合物是相比於以往諾麗,喜好度功能性得到改善,由此消費者無負擔地享受的食品組合物。
以下,通過以下實施例更詳細說明本發明。但是,這些實施例用於例示說明本發明,本發明的範圍並不僅限定於這些實施例。
運動能力及疲勞恢復增進用組合物通過如下過程製備。
實施例1:本發明的運動能力及疲勞恢復增進用組合物的製備
1.1:發酵熟成諾麗的製備
在諾麗果690kg接種複合乳酸菌(7種複合乳酸菌,AON1805(株),Lactomason Co., Ltd.),在37 ℃下發酵45天以上。從發酵起始日起,每7天收集10L發酵液倒在諾麗果上並覆蓋其上,以實施發酵及熟成。此時,使用的複合乳酸菌(7種複合乳酸菌,AON1805(株),Lactomason Co., Ltd.)的組成如以下表1所示。
熟成結束之後,將上述發酵熟成諾麗榨汁,去除固體,獲得發酵熟成諾麗提取物。

1.2:椰子糖的製備
從椰子樹的花莖中採集汁之後,通過傳統方式,在90℃下加熱並搖勻,將上述椰子花莖汁濃縮至蜂蜜茶形態之後使其固化,將完全固化的花汁液細磨,由此製備天然椰子糖。
1.3:黑砂糖(Muscovado)的製備
去除收穫的甘蔗的葉子和雜質,將從甘蔗中榨出的汁放入到大鍋,熬到充分時間之後,去除浮上的雜質和沉澱物,若處於水分蒸發的狀態,則均勻打翻至成為粉末狀態為止,乾燥之後,均勻粉碎,由此製備黑砂糖。
1.4:利用發酵熟成諾麗提取物及椰子糖的複合物的製備
以通過上述1.1至1.3製備的發酵熟成諾麗提取物、椰子糖的不同配合比混合之後,熟成15天,製備複合物。
[複合物的製備]
複合物1:發酵熟成諾麗提取物(重量百分比)90:椰子糖(重量百分比)10。
複合物2:發酵熟成諾麗提取物(重量百分比)80:椰子糖(重量百分比)20。
複合物3:發酵熟成諾麗提取物(重量百分比)70:椰子糖(重量百分比)30。
1.5:利用發酵熟成諾麗提取物及黑砂糖的複合物的製備
以通過上述1.1至1.3製備的發酵熟成諾麗提取物、黑砂糖的不同配合比混合之後,熟成15天,製備複合物。
[複合物的製備]
複合物4:發酵熟成諾麗提取物(重量百分比)90:黑砂糖(重量百分比)10。
複合物5:發酵熟成諾麗提取物(重量百分比)80:黑砂糖(重量百分比)20。
複合物6:發酵熟成諾麗提取物(重量百分比)70:黑砂糖(重量百分比)30。
1.6:利用發酵熟成諾麗提取物、椰子糖及黑砂糖的複合物的製備
以通過上述1.1至1.3製備的發酵熟成諾麗提取物、椰子糖及黑砂糖的不同配合比混合之後,熟成15天,製備複合物。
[複合物的製備]
複合物7:發酵熟成諾麗提取物(重量百分比)90:椰子糖(重量百分比) 5:黑砂糖(重量百分比)5。
複合物8:發酵熟成諾麗提取物(重量百分比)80:椰子糖(重量百分比)10:黑砂糖(重量百分比)10。
複合物9:發酵熟成諾麗提取物(重量百分比)70:椰子糖(重量百分比)15:黑砂糖(重量百分比)15。
1.7:利用發酵熟成諾麗提取物、食糖的複合物的製備
以通過上述1.1製備的發酵熟成諾麗提取物、非精煉糖的不同配合比混合之後,熟成15天,製備複合物。
[複合物的製備]
複合物10:發酵熟成諾麗提取物(重量百分比)90:非精煉糖(重量百分比)10。
實驗例1:運動能力增進試驗
確認通過上述實施例1.1至1.3製備的單一提取物和通過實施例1.4至1.6製備的複合物的運動能力增進效果。
(1)誘發小白鼠的疲勞
將體重30g的ICR小鼠(奧源生物,韓國(Orient Bio, Korea)),在20℃至24℃的溫度條件及55%的濕度條件下,以明暗週期為12小時(light/dark cycle)適應1周之後,將實驗動物如下分類為各個8只的實驗組。設置為實驗組1(非運動組,由以下Normal表示)、實驗組2(運動對照組,由以下Control表示)、實驗組3(發酵熟成諾麗100mg/kg給藥組,由以下FN表示)、實驗組4(椰子糖100mg/kg給藥組,由以下CS表示)、實驗組5(黑砂糖100mg/kg給藥組,由以下MS表示)、實驗組6(複合物10 100mg/kg給藥組,由以下FNS表示)、實驗組7(複合物1 100mg/kg給藥組,由以下C1表示)、實驗組8(複合物2 100mg/kg給藥組,由以下C2表示)、實驗組9(複合物3 100mg/kg給藥組,由以下C3表示)、實驗組10(複合物4 100mg/kg給藥組,由以下C4表示)、實驗組11(複合物5 100mg/kg給藥組,由以下C5表示)、實驗組12(複合物6 100mg/kg給藥組,由以下C6表示)、實驗組13(複合物7 100mg/kg給藥組,由以下C7表示)、實驗組14(複合物8 100mg/kg給藥組,由以下C8表示)、實驗組15(複合物9 100mg/kg給藥組,由以下C9表示),試驗物質按1次/1天總共給藥4周。
(2)複合物的運動能力測定
實驗動物從實驗開始16小時之前起,以穩定的狀態保持飼養條件之後,在丙烯酸塑膠水槽(70cm×70cm×60cm)內放入約70%左右的水,在水槽中游泳。游泳時間將實驗動物虛脫而不再活動的狀態設定為終點,將開始時間至結束時間未知的時間測定為游泳時間。
其結果,如圖1所示,為了測定運動能力,測定至虛脫時為止的游泳時間。實驗結果,確認到在相同的濃度條件下以對照組為基準,處理本發明的組合物的所有組中,游泳時間相比於單一組(發酵諾麗、椰子糖及黑砂糖給藥組)增加,組合物攝取的運動能力相比於單一組攝取增進,尤其,在複合物1、4及7給藥組中,游泳時間為1081秒至最高1119秒,由此可確認運動能力優秀。
另一方面,發酵熟成諾麗提取物和作為非精煉糖複合物的FNS組的游泳時間為913秒,呈現相比於複合物1、4及7給藥組顯著低的結果。
這些結果呈現如下:比起基於糖的糖分供應的運動能力的增加,基於椰子糖及黑砂糖和發酵熟成諾麗複合物的運動能力的提高結果更優秀。
本發明的組合物,在行動實驗結果顯示,移動距離及運動時間可增加,並可呈現運動能力增強。並且,上述組合物可增加血葡萄糖的濃度,可增加肌肉內的糖原(glycogen)濃度,並可減少乳酸脫氫酶(LDH,lactate dehydrogenase)濃度。
實驗例2:疲勞恢復增進試驗
(1)乳酸含量測定
根據上述實驗例1進行實驗之後,將給藥結束日飼養的小白鼠作為對象,實施強迫游泳試驗,測定作為代表性的疲勞物質的血中乳酸生成。
實驗動物從實驗開始16小時之前起,以穩定的狀態保持飼養條件之後,在丙烯酸塑膠水槽(70cm×70cm×60cm)內放入約70%左右的水,在水槽中游泳。游泳時間為15分鐘,同樣適用在所有實驗動物。游泳結束後,為了測定有氧運動後的肌肉疲勞度,在休息期,按10分鐘單位,從尾部採血,利用乳酸測定儀(Lactate Pro LT-1710, ARKRAY Inc. / JAPAN)測定血清乳酸含量。
其結果顯示,游泳後乳酸的濃度比起穩定期增加6.5倍以上,相比於對照組、發酵熟成諾麗、椰子糖及黑砂糖單獨給藥組,組合物組在相同濃度條件下顯著減少血中乳酸含量。尤其,在相同濃度條件下,本發明的組合物的試驗組,相比於對照組(control),呈現4%至55%的低乳酸濃度,由此可確認到顯著減少作為肌肉疲勞度產生物質的乳酸。
尤其,作為發酵熟成諾麗提取物和非精煉糖複合物的FNS組的乳酸濃度為4.34水準,相比於複合物1(C1)、4(C4)及7(C7)給藥組,調查為顯著高的水準的濃度。
由此,本發明的組合物促進疲勞恢復,由此確認提高運動能力的功效突出。
(2)游泳疲勞後血液化學檢查
根據上述實驗例1進行實驗之後,將給藥結束日飼養的小白鼠作為對象,實施強迫游泳試驗之後,小白鼠犧牲之後,將從心臟中採血之後分離的血清委託給綠十字研究所,利用自動分析儀,測定血中無機磷(Inorganic phosphorus)濃度、肌酸激酶(Creatine kinase,CK)濃度。
運動中,肌肉通過重複的肌肉收縮,提高肌球蛋白(myosin)和肌動蛋白(actin)的親和力的步驟中,ATP水解,因此急劇增加血中無機磷濃度。運動中,若血清無機磷濃度急劇增加,則肌纖維的交聯橋(cross-bridge)弱化,由此能量產生降低。由此,血中的無機磷的濃度用作對於肌肉的疲勞度重要的指標。
確認結果,血清中無機磷的濃度,相比於運動對照組(control),無機磷的濃度在複合物給藥組中均顯著減少。在複合物1、4及7給藥組中,分別以11.01mg/dL、11.18 mg/dL、11.07 mg/dL呈現最低的值,減少作為疲勞度因素的無機磷酸鹽含量,由此可確認對疲勞恢復有效。
但是,發酵熟成諾麗提取物和糖的複合物(FNS)也以12.58mg/dL濃度,在運動對照組(control)中呈現具有顯著性的減少,但是呈現與發酵熟成諾麗單一給藥組類似的水準的無機磷濃度。
另一方面,與複合物1、4及7給藥組相比時,調查為高水準的無機磷濃度。
肌酸激酶(Creatine kinase)是EC 2.7. 3.2. 1943年K. Lohmans從骨骼肌中發現的酶,其為將生成由肌酸和ATP形成磷酸肌酸的反應催化的酶,用於催化磷酸肌酸+ADP肌酸+ATP的的羅曼氏反應(將高能量磷酸基從磷酸肌酸轉移至ADP,合成ATP的反應)。肌肉中多而骨骼肌收縮時,大量消耗ATP的情況下,生成肌肉中大量存在的磷酸肌酸中獲得的ATP而供應。
試驗結果,運動對照組(control)的肌酸激酶(Creatine kinase)調查為562 IU/L數值,單一組及複合物給藥組中相比於運動對照組,均調查為高數值。
尤其,複合物1(C1)、複合物4(C4)及複合物7(C7)給藥組分別為967 IU/L、955 IU/L及966 IU/L,呈現最高數值,在相同濃度條件下促進ATP再合成,由此可確認對肌肉疲勞度減輕有效。
如複合物1、4及7給藥組一樣,對於添加作為糖類成分的糖的發酵熟成諾麗和非精煉糖複合物FNS給藥組而言,調查到肌酸激酶(Creatine kinase)的濃度數值(709 IU.L)顯著低。
由以上的說明應當可理解本發明所屬技術領域的普通技術人員在不變更技術思想或必需特徵的情況下能夠將本發明實施為其他具體方式。與此相關的是,應當要理解以上描述的實施例在整體方面上是示例性的,而不是限定性的。本發明的範圍比起上述詳細的說明,應當要理解所附的權利要求書的意思及範圍及其等同概念中導出的所有變更或變形的方式包括在本發明的範圍內。
圖1為攝取本發明的對照組(control)、單一組及複合組合物之後,強迫游泳之後測定運動執行時間的結果圖表。
圖2為攝取本發明的對照組(control)、單一組及複合組合物之後,強迫游泳之後測定所變化的血中乳酸的濃度的結果圖表。
圖3為攝取本發明的對照組(control)、單一組及複合組合物之後,測定強迫游泳之後測定無機磷酸鹽的結果圖表。
圖4為攝取本發明的對照組(control)、單一組及複合組合物之後,強迫游泳之後測定肌酸激酶(Creatine kinase)濃度的結果圖表。
圖5表示非精煉糖和黑砂糖的結晶及形態的差異。
Claims (18)
- 一種用於增強運動能力或疲勞恢復之食品組合物,其特徵在於,作為一活性成分包含發酵熟成諾麗或其提取物、椰子糖、或黑砂糖。
- 如請求項1所述的食品組合物,其中,作為該活性成分包含選自發酵熟成諾麗或其提取物、椰子糖、及黑砂糖中的兩個以上的組合。
- 如請求項2所述的食品組合物,其中,該發酵熟成諾麗或其提取物及椰子糖的重量比為90:10至70:30。
- 如請求項2所述的食品組合物,其中,該發酵熟成諾麗或其提取物及黑砂糖的重量比為90:10至70:30。
- 如請求項2所述的食品組合物,其中,該發酵熟成諾麗或其提取物、椰子糖、及黑砂糖的重量比為90:5:5至70:15:15。
- 如請求項1所述的食品組合物,其中,該發酵熟成諾麗或其提取物是利用乳酸菌將諾麗進行發酵及熟成而獲得的發酵熟成諾麗。
- 如請求項1所述的食品組合物,其中,該運動能力的增進包括肌力強化、體力增進、肌肉疲勞延遲、線粒體活性增進、平衡感增進、持久力提高、肌肉損傷的預防或肌力萎縮抑制。
- 如請求項1至7中任一項所述的食品組合物,其中,該食品組合物為保健食品。
- 如請求項1所述的食品組合物,其中,該食品組合物為選自粉末、顆粒、丸、片劑、膠囊、糖果、糖漿及飲料中的任一種劑型。
- 一種用於增強運動能力或疲勞恢復之藥物組合物,其特徵在於,作為一活性成分包含發酵熟成諾麗或其提取物、椰子糖、或黑砂糖。
- 一種用於增強運動能力或疲勞恢復之醫藥外品組合物,其特徵在於,作為一活性成分包含發酵熟成諾麗或其提取物、椰子糖、或黑砂糖。
- 一種用於增強運動能力或疲勞恢復之飼料組合物,其特徵在於,作為一活性成分包含發酵熟成諾麗或其提取物、椰子糖、或黑砂糖。
- 一種包含發酵熟成諾麗或其提取物及椰子糖的組合物的製備方法,其特徵在於,包括: 步驟(a),獲得利用乳酸菌將諾麗進行發酵及熟成的發酵熟成諾麗;以及 步驟(b),在該發酵熟成諾麗追加椰子糖而熟成。
- 一種包含發酵熟成諾麗或其提取物及黑砂糖的組合物的製備方法,其特徵在於,包括: 步驟(a),獲得利用乳酸菌將諾麗進行發酵及熟成的發酵熟成諾麗;以及 步驟(b),在該發酵熟成諾麗追加黑砂糖而熟成。
- 一種包含發酵熟成諾麗、椰子糖及黑砂糖的組合物的製備方法,其特徵在於,包括: 步驟(a),獲得利用乳酸菌將諾麗進行發酵及熟成;以及 步驟(b),在該發酵熟成諾麗追加椰子糖及黑砂糖而熟成。
- 如請求項13至15中任一項所述的製備方法,其中,在該步驟(a)之後,還包括將該步驟(a)中獲得的發酵熟成諾麗榨汁而獲得發酵熟成諾麗提取物的步驟。
- 一種組合物,其特徵在於,通過請求項13至15中任一項所述的製備方法獲得。
- 如請求項17所述的組合物,其中,該組合物具有增強運動能力或疲勞恢復能力。
