Abstract
Bir Cohn fraksiyonu IV çökeltisi gibi bir AAT içeren protein karışımından alfa-1-antiripsini (AAT) saflaştırmaya yönelik geliştirilmiş bir yöntem sağlanır. Buluşun yönteminde, kontamine edici proteinler AAT'yi etkilemeyen, bir ditiyol gibi bir indirgeme reaktifi ile disülfid bağlarının klevajı ile destabilize edilir. Destabilize edilen proteinler daha sonra bir çökelti olarak bir tuz eklenmeksizin, tercihen bir katı protein adsorbe edici materyal üzerinde adsorbe edilir. Solüsyondan katı adsorbanın ayrılması, önceki teknikteki proseslerde olduğu gibi kapsamlı tuz giderme işlemine ihtiyaç olmaksızın kromatografik saflaştırma için doğrudan uygun olan saflaştırılmış bir AAT solüsyonu bırakır. Ticari bir ölçekte yüksek verimde farmasötik saflıkta AAT sağlayan bu yöntemi içeren bir proses ayrıca açıklanır.
