Abstract
The invention relates to medicine, namely to ophthalmology. The purpose of the invention is to reduce the time. Stimulate corneal repair by instilating a 0.1-1.0% keratan sulfate solution. A stimulating effect of 50-70% of the initial cell population is obtained 48-72 hours after adding the keratan sulfate solution. The method improves the stimulation of corneal cells.
Description
The invention relates to medicine, namely to ophthalmology.
The aim of the invention is to reduce the duration.
The method is carried out as follows.
The animal's cornea is crushed, lyophilized. A solution is prepared from the lyophilized cornea, which is treated with collagenase or trypsin. After filtration, the solution is precipitated with 75% ethanol and again lyophilized. If necessary, the lyophilisate is dissolved with Eagle's medium or medium 199 to 0.1-1.0% concentration and added to keratocyte culture. After 48 h, cells were counted. A stimulating effect of 50-70% of the initial population is obtained 48 hours after the addition of the keratan sulfate solution. A total of 50 samples of keratocytes from IV-VII passages were investigated.
Example 1. A keratan sulfate 0.1% solution in an amount of 0.5 ml to 1.5 ml was added to a keratocyte culture from passage IV (inoculum concentration 10 cells / ml) in an amount of 0.5 ml.
After 72 h of cultivation, the number of cells on a Morphomat device is counted. The number of cells is 48% more than in the control.
For control, keratocytes from passage IV (culture of 104 cells / ml) are added to a culture of 0.1% physiological saline in an amount of 0.5 ml to 1.5 ml of culture medium. After 48 hours of culture, the number of cells does not change from the original seed density Similar results were obtained in all control experiments.
Example 2. In a keratocyte culture from passage IV (104 cell concentration / ml), 0.5 ml of 0.5% keratan sulfate solution was added to the culture medium containing 1% bovine serum. After 72 hours of cultivation, the number of cells increased by 60% compared with the seed concentration.
Example 3. To a culture of keratocytes from passage VII (seeding concentration 104 cells / ml) 0.5 ml of 1% raO was added.

Example 4. A 3.5-kg Chinchilla rabbit under local anesthesia was used to make corneal incisions. Instillation of 1% keratan sulfate solution 3 times a day, 2 drops. 7 days after surgery, keratocytes are activated in the incision area, their number is increased, young collagen appears.

Clinical trials in patients with postoperative corneal edema show a positive effect of the drug on corneal repair after keratotomy.
The use of the invention will improve the stimulation of cells of the affected cornea.
