Abstract
This invention relates to histology, in particular to laboratory research methods. The aim of the invention is to improve the quality of the sample due to the better preservation of loose tissue. This is achieved by placing the sample after fixation in a 7-8% solution of polyvinyl alcohol for 3-5 minutes. Then it is transferred to a cryostat and frozen sections are prepared at (-5) - (-10) ° C. Sections are mounted on slides pre-coated with the same solution of polyvinyl alcohol. The invention allows to obtain a qualitative sample and good histological. cue slices, especially with loose crumbly fabric.
Description
This invention relates to histology and can be used in the preparation of histological sections from fixed tissues.
The aim of the invention is to obtain a quality sample, especially from friable tissue.
The method is carried out as follows.
образомway
The biopsy specimen is fixed in formalin solutions, but not more than 6 months. Next, the sample is placed in a 7-8% solution of polyvinyl alcohol, which envelops it. Then, cuts are prepared from a sample on a microtome cryostat at a temperature of (-5) - (-10) ° C, which are transferred to a glass that has also been previously coated with poly / vinyl alcohol.
Example 1. Chicken muscle biopsy specimen fixed in 10% formalin for 3 months. Next, the sample is placed in a 7% solution of polyvinyl alcohol for 3 minutes. Then immediately the sample is transferred to a cryostat and histological sections are prepared with a thickness of 5 microns. Sections are transferred to a glass pre-coated with the same solution of polyvinyl alcohol. Stain with hematoxylin-eosin and examine under the microscope. The preparation shows a normal muscle structure.
Example 2C A chicken liver biopsy is fixed in a 10% formalin solution for 30 days.
