Abstract
This invention relates to medical microbiology and concerns the detection of the virulence of Escherichia. The purpose of the invention is to improve the accuracy of the method. The isolated strains of pathogenic Escherichia are seeded in sectors of 6-8 strains per dish with m seppepton agar, incubated in a thermostat at 37 ± 0.2 ° C for 16-20 hours. pipette 4-5 cm of the azureosin solution according to Romanovsky, additionally diluted with V10 distilled water. Description
This invention relates to medical microbiology and concerns the detection of the virulence of Escherichia.
The purpose of the invention is to improve the accuracy of the method.
The method is carried out as follows.
Выделенные штаммы патогенных эшерихий различных серогрупп засевают секторами по 6-8 штаммов на одну чашку с м сопептонным агаром (МПА) Дл получени более четкого результата МПА осветл ют путем фильтрации в гор чем виде через ватно-марлевый фильтр, последующего отстаивани и декантировани .The isolated pathogenic Escherichia strains of different serogroups are seeded with sectors of 6-8 strains per plate with m of septon agar (MPA). To obtain a more accurate result, MPA is clarified by hot filtration through a cotton gauze filter, followed by settling and decanting.
Crops in the plates are incubated in a thermostat at 37 ± 0.2 ° C for 16–20 h, i.e. until a clear increase in stroke. Carefully, without touching the sowing line and the surface of the nutrient medium, with a pipette, add 4-5 cm3 of an azureosin solution according to Romanovsky, diluted
1:10 distilled water. After 1-2 minutes, the paint is discarded and, in the presence of dark blue (or black) colony staining, the culture is considered virulent.
The colonies and stroke growth of the non-virulent pathogenic Escherichia strains remain colorless or colored only marginally in a light greenish or bluish tint.
To account for the severity of virulence, the following system is used:
4+ - черное окрашивание (резко выраженна реакци );4+ - black staining (pronounced reaction);
3+ - dark blue staining (pronounced reaction) ;.
2+ - синее окрашивание (положительна реакци );2+ —blue staining (positive reaction);
1+ - зеленовато-голубоватое окрашивание всего штриха или колонии (слабо выраженна реакци ;1+ - a greenish-bluish coloration of the entire stroke or colony (mild reaction;
±- слабое окрашивание по краю штриха или колонии (сомнительна реакци );± - faint staining along the edge of the line or colony (questionable reaction);

Virulent strains are considered only those that give a reaction of at least 2 pluses.
In this way, the virulence of 167 pathogenic Escherichia strains isolated from various sources was studied. A positive result was found in 26.9 ± 3.2% of the cultures.
The table shows the comparative data on the detection of the virulence of Escherichia by the proposed method and three others: Hly, CFA and PCF.
Как видно из приведенных данных, предлагаемый способ дал совпадающие ре- з|гльтаты со статистически несущественными различи ми (t 0,8-1,8).As can be seen from the above data, the proposed method gave coinciding results | with the statistically insignificant differences (t 0.8–1.8).
77,78%.The matching results of the proposed method with three others reach 46.7%, and when compared with six other methods (cell cultures, etc.) - up to 77.78%.

In 60 strains, the coincidence of positive results is observed in 3-4 ways, and in 18% - 5-6. The remaining 22% had a coincidence in 2 ways, or none at all.
The advantages of the proposed method consist in its rapidity (3-5 minutes), simplicity, efficiency, clarity of results, high sensitivity, reproducibility on a simple nutrient medium (MPA), available for any bacteriological laboratory.
