The invention relates to the field of medicine, primarily to hematology and experimental medicine, and can be used to determine the distribution of red blood cells by resistance.
The purpose of the invention is to simplify and accelerate the method with a more accurate determination of the distribution of red blood cells in resistance.
Figure 1 shows a block diagram of an installation for implementing the method; Fig. 2 is a distribution curve for erig trocytes.
2.The scheme is constructed from a measurement cell 1, which is located between the photodetector 2 and the monochromator 3 of the spectrophotometer A in the path of the transmitted light from the source 5 of the light. The measurement cuvette 1 is equipped with a thermostat 6 and a magnetic stirrer 7. The hemolyzing agent supply unit 8 is connected to the capillary line 9 of the measurement 9. The plotter 10 is connected to the output of the supply unit 8 and through the differentiator 11 to the photoreceiver output 2.
115-1.The unit 8 for the supply of the hemolyzing agent is made on the basis of the industrial dispenser DAJ-115-1.
The following changes have been made to obtain the desired hemolyzing agent feed rate:
используют синхронньй двигатель РД-09 с редуктором, который имеет 1,75 об/мин;use synchronous engine RD-09 with gear, which has 1.75 rev / min;
на корпусе дозатора закреплен кронштейн с .держателем шприца от дозатора да-10; on the case of the dispenser is fixed a bracket with a syringe holder from the dispenser yes-10;
Additional rollers for belt transmission with diameters of 35 and 20 mm are mounted on the drive shaft of the piston lift (to which the reinforcement from the engine is supplied).
с подъемником поршн шприца состыкован реохорд типа ППМЛ-ИЗ (св зан с самописцем).with a syringe piston lifter, a PPML-IZ-type reichord is connected (associated with a recorder).
As a result of modifications, the unit for supplying hemolyzing agents makes it possible to dose hemolyzing substances with low flow rates (0.05, 0.10, 0.22, 0.45, 0.90, 1.35, 1.80 ml / min ), the hemolyzing agent can be easily and quickly replaced by replacing a hemolytic syringe, and record the amount of hemolytic used on the recorder.
To differentiate the signal received from the PMT, a bench-type laboratory instrument is used to differentiate the signals with the following basic parameters of the instrument: input signal 0-2V, output signal 0-100 nV, maximum differentiation time 360 s.
Primer p. Blood is taken in a quantity of 0.1 ml from a patient's finger with a B, j deficiency on an anticoagulant (heparin 25 U / ml). Red blood cells were isolated from whole blood by centrifugation (3000 rpm, 5 min in an OPN-3 centrifuge) and washed three times with saline. The suspension of erythrocytes is diluted to an optical density of D 0.7 at a wavelength of 71,670 nm. The suspension is placed in the measurement cuvette 1 (optical path length 20 mm) in an amount of A ml and the thermostat 6 and the magnetic stirrer 7 j are inserted into the arrow of the measuring writing plotter 10 at the zero mark and at constant temperature t, continuously mixing, in the cuvette 1 measurement the feed unit 8 adds 0.01 M HCt prepared in saline at a constant rate of 0.625 ml / min. As a result of the effect of the hemolyzing agent, the erythrocytes are broken, the light scattering on the erythrocytes decreases, and as a result The transmittance of the suspension is measured at I 670 nm, which is reflected in the increase in the signal removed from the photodetector 2. This signal is differentiated by the differentiator 11 and is fed to the Y coordinate of the plotter 10. The differential (gain) of the signal is proportional to the number of destroyed red blood cells per unit time, i.e. . erythrocyte destruction rate

Завершению процесса гемолиза соответствует возвращение стрелки графопостроител на нулевую линию, units; V is the volume of the hemolyzing agent, ml. The completion of the hemolysis process corresponds to the return of the arrow of the plotter to the zero line,
As can be seen from the curve, in figure 2 the destruction of red blood cells occurs in groups. The graph clearly distinguishes 3 groups of erythrocytes of different resistance. Group I is made up of the most unstable red blood cells, which are destroyed at a constant rate in the region of 0.7-1.8 ml; II - erythrocytes, destroyed in the region of 1.8-3.1 ml; III - red blood cells, destroyed in the region of 3.2-3.6 ml.
The same erythrocyte suspension was i investigated in a known manner. According to the schedule, which has the form of a single peak, it is impossible to isolate groups of red blood cells that are different in resistance, which proves the high resolution of the proposed method.
