Spôsob prípravy tapentadolu vo forme bázy alebo jeho hydrogenchloridu zahŕňa prípravu soli vzorca (III) zo zmesi diastereomérov vzorca (II) pomocou kryštalizáciou indukovanej asymetrickej transformácie (CIAT) cez oxo-enolovú tautomérnu rovnováhu kovalentne viazaných diastereomérov, posunutú smerom k žiadanému diastereoméru bez použitia chirálnej rezolúcie s chirálnymi karboxylovými kyselinami.The process for preparing tapentadol in base form or its hydrogen chloride comprises preparing a salt of formula (III) from a mixture of diastereomers of formula (II) by crystallization-induced asymmetric transformation (CIAT) through an oxo-enol tautomeric equilibrium of covalently bonded diastereomers, displaced toward the desired diastereomer without using a chiral resolution with chiral carboxylic acids.
