Для измерения лейкоцитов в периферической крови методом флуоресцентной проточной цитометрии гематологический анализатор имеет специальную реакционную камеру 1, в которую автоматически подается биоматериал из пробирки и вступает во взаимодействие с двумя реагентами - лизирующим раствором (для лизиса эритроцитов) и флуоресцентной краской (для флуоресцентного окрашивания лейкоцитов). После этого смесь автоматически подается в проточную ячейку 1 и подвергается излучению красного лазера. Под воздействием излучения лейкоциты рассеивают свет, а флуоресцентная краска испускает свет. Рассеянный свет и свет от флуоресцентной краски собирается тремя детекторами - детектор прямого светорассеяния (FSC - forward scattering), детектор бокового светорассеяния (SSC - side scattering) и детектор боковой флуоресценции (SFL - side fluorescense). Собранные сигналы подвергаются обработке и на основе сигналов строятся скатерограммы(точечные графики). График представлен на фиг. 1. На фиг. 1 показано расположение измеряемых субпопуляций лейкоцитов в периферической крови. График WDF построен на основе сигналов от детекторов SSC и SFL (боковое светорассеяние против боковой флуоресценции). Метод флуоресцентной проточной цитометрии позволяет оценивать общее количество лейкоцитов (WBC) и измерять их субпопуляции - абсолютное и относительное количество лимфоцитов (LYMPH%,#), моноцитов (MONO%,#), нейтрофилов (NEUT%,#), эозинофилов (ЕО%,#), базофилов(ВА80%,#), незрелых гранулоцитов (Ю%,#) и показатели NEUT#& и NEUT%&, где NEUT#&=NEUT# - IG# и NEUT%&=NEUT% - IG%, Дополнительно измеряются показатель реактивной интенсивности нейтрофилов (NE-SFL) и коэффициент осцилляции нейтрофилов по оси Y (NE-WY).
