Для увеличения квантового выхода флуоресценции и улучшения стабильности синтезированных ядер CdTe ККТ, они были покрыты дополнительными слоями различных полупроводников.In fig. На фиг. 2 представлена схема синтеза сополимера ВП-МА-ЭГДМА. Для дальнейшей модификации ККТ использовался сополимер ВП-МА-ЭГДМА. Данный полимер был выбран ввиду его биосовместимости, гидрофильности, а также по той причине, что после гидролиза данного полимера можно получить большое количество карбоксильных групп на поверхности ККТ, по которым можно проводить как модификацию ККТ, так и конъюгацию с биомолекулами. Структура сополимера подтверждалась ИК-спектрометрией с Фурье преобразованием на приборе Nicolet 6700 (Thermo Scientific, США).In fig. На фиг. 4 представлены спектры поглощения и нормированные спектры флуоресценции CdTe/CdSe/CdS/ZnS-(ТГК-L-cys) - ВП-МА-ЭГДМА, измерения проводились на спектрофлуориметре FluoroLog 3 модель FL3-21 (Horiba Jobin Yvon SAS, Франция) и спектрофотометре UNICO-2100 (UNITED PRODUCTS & INSTRUMENTS, США).In fig. Синтез ККТ CdTe осуществляли в три этапа.The synthesis of CdTe CQDs was carried out in three stages.
Первым этапом синтеза являлось получение NaHTe. В колбу объемом 25 мл помещали навеску теллура Те (0,387 ммоль), навеску боргидрида натрия NaBH4 (1,692 ммоль) и перемешивали. Затем добавляли 5,0 мл дегазированной деионизированной воды и перемешивали реакционную массу при комнатной температуре в течение 1 ч до полного растворения порошка и появления светло-розовой окраски. Синтез ведется в атмосфере аргона.The first stage of the synthesis was the production of NaHTe. Третий этап включал в себя синтез CdTe ККТ. К раствору прекурсора кадмия со стабилизатором нагретому до 98°С быстро добавляли свежеприготовленный раствор NaHTe (прекурсор теллура) при постоянном перемешивании, соотношение Cd : Те составляет 1:0,3. Соблюдение данных условий способствует ускорению роста ККТ. Синтез CdTe ККТ проводят в атмосфере аргона.The third stage included the synthesis of CdTe CQDs. Очищенные CdTe ККТ, стабилизированные ТГК, с соотношением Cd : ТГК равным 1:3, полученные в примере 1, со временем синтеза 180 мин, помещали в трехгорлую колбу, затем к раствору CdTe ККТ медленно добавляли прекурсор Cd и 0,8 мл заранее приготовленного прекурсора Se. В качестве прекурсора селена выступал NaHSe, который предварительно готовили за счет смешивания в 3,0 мл деионизированной воды порошка селена Se (0,01 г) и порошка боргидрида натрия NaBH4 (0,0254 г). Полученные ККТ осаждали с помощью изопропанола в центрифуге при 8000 об/мин в течении 15 мин, декантировали жидкость, осадок промывали изопропанолом. И полученную смесь снова оттитровывали 1 М раствором NaOH до рН=10. Сополимер ВП-МА-ЭГДМА был синтезирован по данной методике: в трехгорлую колбу объемом 50 мл загрузили ВП (28 ммоль) и раствор, содержащий МА (10 ммоль), 2,0 мл уксусной кислоты, 11,0 мл тетрагидрофурана (ТГФ) и ТГК (0,5 ммоль). Смесь дегазировали в течение 20 мин, затем нагревали до 65°С в течение 10 мин при перемешивании в атмосфере аргона. К полученному раствору шприцом добавляли (2,3 ммоль) ЭГДМА и 0,3 г (0,25 ммоль) ТГК, растворенных в 4,0 мл ТГФ. После 10 мин дополнительного нагревания до 65°С реакцию гасили добавлением ТГФ (7,5 ммоль) в 1,0 мл ТГФ и держали при 65°С еще 10 мин. Полученный сополимер очищали за счет осаждения деионизированной водой (3 объема) и последующего центрифугирования при 10000 об/мин. Полученный осадок промывали деионизированной водой и сушили в вакууме при 40°С. The resulting precipitate was washed with deionized water and dried in vacuum at 40°C. The reaction yield was 20%.
Очищенные CdTe/CdSe/CdS/ZnS ККТ, покрытые ТГК и L-cys (полученные в примере 4) помещали в трехгорлую колбу на 100 мл, после чего добавляли 0,06 г сополимера ВП-МА-ЭГДМА и титровали с помощью 0,5 М NaOH до рН=10, смесь дегазировали в течение 30 мин при помощи аргона. Полученные ККТ, покрытые сополимером ВП-МА-ЭГДМА, очищали путем центрифугирования с трехкратным избытком по объему изопропанола при 10000 об/мин в течении 15 мин, декантировали жидкость, осадок промывали изопропанолом. Далее осажденные ККТ сушили при 50°С на воздухе. Данные ККТ имеют гидродинамический радиус 9,7 нм с разбросом менее 10%. Люминесценцию на длинах волн более 650 нм. Растворимость и устойчивость в биологических средах. Квантовый выход более 25%. Исследование характеристик ККТ проводилось на следующем оборудовании: спектрофотометр UNICO-2100 (UNITED PRODUCTS & INSTRUMENTS, США, диапазон длин волн: 190-1100 нм), спектрофлуориметр FluoroLog 3 модель FL3-21 (Horiba Jobin Yvon SAS, Франция, спектральный диапазон возбуждения флуоресценции - 200 - 850 нм), анализатор дзета-потенциала серии Zetasizer Nano Z (Malvern Instruments Ltd, Великобритания, диапазон измерения: 5 нм - 10 микрон), анализатор размера частиц серии Zetasizer Nano S (Malvern Instruments Ltd, Великобритания, диапазон измерения: 0,6 нм - 6 микрон), ИК-спектрометр с Фурье преобразованием Nicolet 6700 (Thermo Scientific, США, спектральный диапазон 25000 - 20 см-1)The resulting CQDs, coated with the VP-MA-EGDMA copolymer, were purified by centrifugation with a threefold excess by volume of isopropanol at 10,000 rpm for 15 minutes, the liquid was decanted, and the precipitate was washed with isopropanol.
