At the stage of freeze-drying, 5 ml of a suspension of a liver cell culture containing 2 × 10 6 cells in 1 ml, as well as 2.5 ml of a structurant - a 6% solution of polyvinylpyrrolidone, and a 10% solution of albumin (2 ml) with 10% sucrose solution (0.5 ml), frozen to a temperature (-70°C) in a 50 ml round-bottom flask and placed in the working chamber of the device, drying time 24 hours. Storage is carried out at a temperature of -70°C.
В результате получена лиофилизированная культура клеток печени, обогащенная HGF устойчивая при хранении - фиг. 2., с содержанием концентрации активного вещества - HGF до 240 (198-251) нг/мл. Проводят оценку уровня фактора роста гепатоцитов в лиофилизированных образцах. Содержание HGF в среде после 5-ти суток культивирования составляет 76,8 (68,4-80) нг/мл, в лиофилизате 240 (198-251) нг/мл. Для оценки количества HGF используют метод твердофазного иммуноферментного анализа с использованием иммуноферментного анализатора Stat Fax-2000 (Awareness tehnology INC USA). Влияние лиофилизата культуры клеток печени пролонгированного действия, обогащенной HGF изучали в эксперименте на крысах линии Wistar в течение 6 дней. Моделирование острого токсического повреждения печени выполняют путем подкожного введения четыреххлористого углерода чистого для анализов из расчета 0,5 мг\100г массы животного по А. Фишеру. Через сутки после индукции острого токсического повреждения печени животных приписывали к экспериментальным группам методом случайного распределения, таким образом, в исследование вводили животных с индуцированной острой печеночной недостаточностью. Опытным животным - группа 1, осуществляли подкожную инъекцию 0,5 мл лиофилизата культуры клеток печени через 12 часов после введения ССl4. Контрольным животным - группа 2 осуществляли введение физиологического раствора того же объема. На 3 сутки эксперимента результаты показали, что введение лиофилизата приводит к выживанию 53,3% крыс (рфр.=0,2189) в группе 1, в то время как в группе 2 выжило 40% (рфр=0,0091). К 6-м суткам исследования летальность в группе 2 составила 100%, в то время как в группе 1 количество умерших особей не менялось с Зго дня эксперимента, что составило 46,7%.The effect of a lyophilisate of a long-acting liver cell culture enriched with HGF was studied in an experiment on Wistar rats for 6 days. Введение лиофилизата культуры клеток печени обогащенного HGF, полученного из фармацевтической композиции, при токсическом повреждении печени у животных позволяет улучшить результаты выживания.
