The present invention provides a method for increasing the viability of microorganisms that are active substances in pesticides, rodenticides, insecticides, and other preparations for pest control (for example, rodents in sanitary and epidemiological practice), as well as agrochemicals, by placing these microorganisms in protective microcontainers, and is intended for use in industry, agriculture, horticulture and forestry.
State of the art
Biological preparations for controlling various pests are known from the prior art, in which various types of microorganisms are used as active substances: fungi, bacteria, viruses, streptomycetes.
In particular, the drug "Rizoplan" is known, with the active substance - the bacteria Pseudomonasfluorescens, which is used as a pesticide and fungicide against helminthosporium rot, powdery mildew, brown rust, spots, late blight of potatoes, gray and fruit rot on fruit and berries, black mucous and vascular bacteriosis of cabbage. Available as a liquid with a concentration of 1 billion CFU / ml. Currently known drug "Biostop" containing Bacillus thuringiensis, Streptomycessp. and Beauveriabassiana, is selective against a wide range of harmful Lepidoptera. Also available in liquid form. Thus, there are various pesticides and fungicides with biological or bacterial preparations.
At the same time, the disadvantages of such biological preparations are known.
So, when using biological products in the field (by spraying a working solution or directly the drug on protected objects) under the influence of ultraviolet radiation (sunlight) and other unfavorable factors, for example, precipitation or wind, a significant part of beneficial microorganisms either die or are washed away during the first days from the treated surfaces, which reduces the effectiveness of the drugs and forces to increase the dosage or requires additional or repeated treatment.
In addition, biological products have a limited shelf life associated with a relatively short period of preservation of beneficial microorganisms in an active state of their viability, and also require special storage conditions (primarily in terms of temperature), which is not always possible to ensure in field conditions. For example, the above preparations Rizoplan and Biostop have a shelf life of 3 months at a temperature of 4 to 10 ° C. All this imposes significant restrictions on use.
There are known methods of increasing the shelf life of drugs by transferring microorganisms to a state of suspended animation by drying. However, the existing methods for drying the culture liquid are either associated with significant losses of beneficial microorganisms. For example, when drying in a spray dryer, production losses can be up to 50% of the original raw material. In the case of lyophilization of biological products that contain a significant amount of moisture, this process is associated with significant energy consumption and the volume of production facilities. Separately, there may be a problem of damage and death of bacterial preparations during lyophilization.
These protectors can differ in their effectiveness depending on the types of microorganisms used, targets and their habitat, i.e. do not differ in universality.
е. увеличить срок хранения препарата), значительная часть способов получения микрокапсул не применимы к биологическим препаратам, так как предполагают проведение реакции в агрессивной для микроорганизмов среде, не способны обеспечить немедленное действие препарата (необходимо время на разрушение оболочки), в силу значительного диаметра (более 50 мкм) не могут быть применены в современных системах опрыскивания (УМО).Known use in pesticides of various microcapsules, consisting of a shell and a core containing both chemical pesticides and other chemical substances (patents No. 2126628, 2518449, 2496483, 2516357, 2488437, 2602196), and living microorganisms (patent No. 2220716). These microcapsules are potentially capable of playing the role of protectors; however, microcapsules, when used in biological pesticides, do not make it possible to dry the drug, transferring microorganisms from an active state to a state of suspended animation (i.e., to increase the shelf life of the drug), a significant part of the methods for obtaining microcapsules are not applicable to biological drugs, since they imply an aggressive reaction for microorganisms in the environment, they are not able to provide an immediate action of the drug (it takes time for the destruction of the shell), due to a significant diameter (more than 50 microns), they cannot be used in modern spray systems (ULV).
The closest analogue is the invention disclosed in patent RU 2581929, which proposes a method for producing a biopesticide, comprising introducing a biologically active suspension in the form of spores of entomopathogenic fungi and a liquid phase into microcontainers, which are hollow containers, in the form of a shell made of a synthetic polymer material, at least , with one hole for the introduction of spores, decantation of the liquid phase and drying at a temperature of 25 ÷ 45 ° C of microcontainers together with spores of entomopathogenic fungi under flow conditions up to constant weight, also a method of protecting plants from pests, including the activation of a biopesticide in an aquatic environment and application of the named preparation on plants. At the same time, the microcontainer fully protects the biopesticide contained in it from harmful UV radiation, maintains the necessary moisture inside itself for the life of microorganisms, protects them from high temperatures, has a prolonged predetermined action time and good "sticking" properties.
At the same time, the invention described in patent RU2581929 assumes the use of only fungal spores as an active substance, which limits the protective effect of a biological pesticide. Accordingly, the present invention is directed to the further development of the invention of patent No. 2581929 based on the principles defined there.
The technical solution according to the present invention makes it possible to expand the range of application of biopesticides with microcontainers against a wider range of plant pests and diseases.

The present invention provides a microcontainer, which is a hollow container with a size of up to 40 microns, having at least one opening, containing live microorganisms and / or their spores as an active substance, or eukaryotic cells infected with viruses, while the living microorganisms do not include fungal spores. In this case, living microorganisms and / or their spores are selected from the group: spore-forming and non-spore-forming bacteria; actinomycetes; streptomycetes; conidia, mycelium and blastospores of fungi; rickettsia; protozoa; eukaryotic cells infected with viruses, any part of them, or a mixture of any part of them, or a mixture of these microorganisms. Also, microcontainers may additionally contain mushrooms, or parts thereof, as well as combinations of different types of mushrooms. Also, surfactants (surfactants) in the form of ionic and non-ionic surfactants can be added to the composition of substances placed in microcontainers.
In addition, the present invention provides the use of microcontainers for the production of a pesticide, agrochemical or preparation for the control of pests and diseases of plants and seeds. Also, using the described microcontainers, a method of protecting plants or seeds can be implemented, including application by spraying with a solution that contains microcontainers previously diluted in water, as in a working solution. The described microcontainers can also be used in the method of feeding plants or seeds, including the application of microcontainers by spraying, wherein the microcontainers are previously diluted in water as in a working solution, or by dusting without preliminary dilution in water. A method for regulating soil fertility is also proposed, including the application of microcontainers by spraying, while the microcontainers are previously diluted in water as in a working solution, or by direct application to the soil without preliminary dilution in water. What's more, micro containers can be used to prepare pest control bait and produce rodenticides. Also, the described microcontainers can be used for the preparation of preparations for the preparation of preparations for combating plant diseases.
Например, описанные микроконтейнеры помещают в ферментер в начале стандартного цикла процесса наращивания биомассы, в оптимальном соотношении 1 насыпной литр порошка микроконтейнеров на 3 литра культуральной жидкости (при титре 109 в конце процесса).The present invention provides a method for producing a biopesticide by adding microcontainers obtained according to patent RU2581929, for example, based on polyurea, or obtained in another way, and representing hollow containers, in the form of a shell made of a synthetic polymer material, with at least one hole, into the culture liquid containing microorganisms - active ingredients of biological pesticides. For example, the described microcontainers are placed in a fermenter at the beginning of the standard cycle of the biomass growth process, in the optimal ratio of 1 bulk liter of microcontainer powder to 3 liters of culture liquid (with a titer of 10 9 at the end of the process).
In this case, microorganisms can be bacteria and their spores, for example, bacteria of the genus Pseudomonas, Bacillus, Salmonella Bradyrhizobium japonicum, Azotobacter; streptomycetes; conidia, blastospores and mycelium of fungi; eukaryotic cells and viruses infecting them.
The obtained culture liquid with microcontainers is filtered, and the filtrate is dried in a lyophilic, vacuum or spray dryer until the loss of mass stops, or left in the form of a concentrate with the addition of surfactants (ionic or non-ionic) in the amount of 10 to 90% of the filtered mass of the mixture of culture liquid and microcontainers. In this case, microorganisms go into a state of suspended animation, which increases the shelf life of the biopesticide without losing its properties.
Materials for microcontainers can be natural or artificial materials. For example, suitable polymeric materials can be used as artificial materials, for example, polyureas, polystyrenes and their copolymers, for example, poly (styrene) -b- (poly (methylmetalacrylate) copoly (glycidal methacrylate) copolymer; (polystyrene-b- (PMMA -co-PGMA) and poly (methyl methacrylate), polyacryl cyanoacrylates, polymers and copolymers of polylactides, derivatives of polyethylene glycol and polypropylene glycol, etc. Cellulose and its derivatives, for example, methyl cellulose, methylpropyl cellulose, hydroxymethyl propyl cellulose, and others can be used as natural polymers. and its derivatives, dextran and cyclodextrin of various forms, alginates and their derivatives, glucans and their derivatives, albumin, gelatin and its derivatives, agar and other natural polymers.
In the future, the resulting powder or concentrate of microcontainers with a specific type of microorganisms can be combined with similar microcontainers with other microorganisms, obtaining complex (with several active ingredients) universal biopesticides with an extended spectrum of action on pests and plant diseases.
Before use, biopesticides with microcontainers can be activated by soaking or diluting in water, after which the microorganisms in suspended animation pass into the active phase.

A fermenter with a volume of 1000 l was loaded with a nutrient medium (filling level 65-70% of the volume of the fermenter), for example L-broth, sterilized by heating at 1-1.5 atm for 30-45 minutes, then cooled to a temperature of 25-28 ° FROM. After that, the inoculum, which was a culture of Bacillus thuringiensis cells, was poured into the medium under sterile conditions. The cells were grown for 36-48 hours with continuous stirring and aeration (1-1.5 L of air per 1 L of medium) until the precipitation of bacterial spores was at the level of 70-80%. Then the cultivation process was stopped, the cells were filtered on a microfilter. The resulting concentrate was mixed with microcontainers in a ratio of 1: 1, stirred continuously for 1-2 hours and dried in a spray dryer at an inlet air temperature of 130-150 ° C and an outlet air temperature of 60-70 ° C.

As you can see, the experimental pesticide is more active than the control one. Moreover, as the concentration increases, the effectiveness of the applied pesticide increases.

Activity tests
Planting of rapeseed (Ryazan region) with a concentration of rape beetle 8-9 beetles per 1 sq. m (which exceeds the economic threshold of harmfulness by 3-5 times), was treated with the pesticide Biostop with a consumption
3 liters per 1 ha, containing the same active ingredients as the experimental pesticide Biostop Super, with a consumption of 50 and 100 g per hectare, previously dissolved in water as a working fluid (consumption 3 l per hectare). The outside air temperature was 23 ° С, the weather was cloudy. An OP-2000 sprayer was used to apply the pesticide.
After 8 days, the following results were obtained:

Stability tests
Stability and shelf life tests were carried out.
The pesticides obtained in examples 1a, 1b, as well as the pesticide Biostop without microcontainers, were stored in a bright room at a temperature of 20 ° C, simulating sunny weather conditions.
After storage for every 6 months. Within 2 years, a part of the microcontainers was opened and tests were carried out for the activity of bacteria. As tests have shown, within 2 years the bacteria in microcontainers retained almost 100% activity compared to the initial indicators of activity, while bacteria without microcontainers completely lost their properties in 2 years.
The use of microcontainers leads to a significant decrease in the consumption of the active substance while maintaining efficiency, which is especially important when working in summer in sunny and hot weather.
A fermenter with a volume of 1000 l was loaded with a nutrient medium poor in nitrogen source, while containing 10 g / l of mannose, 2 g / l of sodium phosphate disubstituted, 5 g / l of sodium chloride. The mixture was prepared by the method described in examples 1a and 1b. The apparatus was seeded with nitrogen-fixing organisms Bradyrhizobium (or Azotobacter), cultured analogously to example 1a and 1b. Further, the process of sedimentation, filtration and drying was carried out analogously to examples 1a and 1b.
If the culture released a lot of polysaccharide during its growth and became sufficiently viscous, then after stopping the fermentation, the culture was mixed without sedimentation with microcontainers in a 1: 1 ratio, stirred for 3-4 h and filtered on a suction filter. The resulting filtered mass was sent to freeze drying.
Application of microcontainers
The resulting dry mass of nitrogen-fixing microorganisms (Bradyrhizobium), obtained according to example 5, in an amount of 1 kg was mixed with a dry surfactant (surfactant) in an amount of 100 g or a liquid surfactant in an amount of 500 ml, diluted in 10 l of water. The resulting solution was used to treat legume seeds (soybeans) at the rate of 10 liters of the resulting diluted solution per ton of soybeans. After seed treatment, soybeans were stored for several months and used for sowing without additional inoculation.
This method of using nitrogen fixers in microcontainers made it possible to treat seeds with nitrogen fixers (nitrogen fixing bacteria) in advance, and not immediately before sowing.
Accordingly, the technical solution according to the present invention allows:
1. Снизить воздействие прямого УФ-излучения на микроорганизмы за счет помещения их внутрь микроконтейнера, стенка которого выступает защитным барьером, за счет чего можно значительно повысить их выживаемость;1. To reduce the effect of direct UV radiation on microorganisms by placing them inside a microcontainer, the wall of which acts as a protective barrier, due to which their survival can be significantly increased;
2. Снизить бактериальную нагрузку за защищаемые растения (на Участках 2 и 3 в сравнении с Участками 1 и 1 соответственно в 1000 раз (расчет соотношений титра КОЕ на гектар), при сохранении и достижении лучшей эффективности в отношении уничтожения вредителей;2. Reduce the bacterial load for protected plants (in Plots 2 and 3 in comparison with Plots 1 and 1, respectively, by 1000 times (calculation of the ratio of CFU titer per hectare), while maintaining and achieving better efficiency in terms of pest control;
3. Продлить защитное действие опытного пестицида, что позволяет сократить кратность обработок;3. Extend the protective effect of the experimental pesticide, which allows to reduce the frequency of treatments;
4. Увеличить срок хранения пестицидов (2 года у опытного пестицида по сравнению с 6 месяцами у аналогичного ему биопестицида Биостоп, без микроконтейнеров);4. Increase the shelf life of pesticides (2 years for an experimental pesticide compared to 6 months for a similar biopesticide Biostop, without microcontainers);
5. To increase the shelf life of seeds treated with an agrochemical.
