Не менее острой является проблема инвалидизация выживших после инсульта, большая часть из которых теряет трудоспособность, и зачастую нуждаются в постоянном постороннем уходе. Возраст является одним из наиболее значительных факторов риска инсульта, и уже после 30 лет данный риск начинает непреклонно увеличиваться. Большая часть инсультов (95 %) происходит в возрасте 45 лет и выше. Учитывая процесс демографического старения, который в последнее время охватывает не только развитые, но и развивающиеся страны, очевидна необходимость сконцентрировать усилия в области здравоохранения на улучшение имеющихся и разработку новых профилактических и терапевтических подходов к борьбе с инсультом.Vascular diseases of the brain are an urgent medical and social problem. Синтез гена, кодирующего IGF-1-long человека, проводили методом удлинения частично перекрывающихся олигонуклеотидов, полученных с помощью автоматизированного химического синтеза. Реакции рестрикции и лигирования, селекцию трансформантов, выращивание бактериальных клеток, выделение и электрофорез плазмидных ДНК проводили с использованием стандартных методов генетической инженерии. Синтезированный ген был вставлен в вектор рЕТ28а(+) (Novagen, США) по сайтам NdeI и XhoI, полученной плазмидой рЕТ28-IGF-1 трансформировали клетки E. coli. Для первичного отбора трансформантов, проверки правильности синтеза и стыковки, а также для последующего длительного хранения плазмиды использовали клетки E. coli DH10 B [F- mcrA ∆(mrr-hsdRMS-mcrBC) φ80lacZ∆M15 ∆lacX74 recA1 endA1 araD139 ∆(ara-leu)7697 galU galK λ- rpsL nupG / pMON14272 / pMON7124] (Thermo Fisher Scientific, США). Раствор белка наносили на колонку с SP Sepharose Fast Flow GE Healthcare, уравновешенную стартовым буфером-2 (20 мМ Tris-HCl, рН 8,8; 0,01 % Tween 20). The fractions containing the target protein were combined, the pH of the protein solution was adjusted to a value of 8.8 by the addition of 10 mM NaOH and re-filtered through a 0.2 μm filter. The protein solution was applied to a GE Healthcare SP Sepharose Fast Flow column equilibrated with start buffer-2 (20 mM Tris-HCl, pH 8.8; 0.01% Tween 20). After applying and washing the column with start buffer 2, the protein was eluted with a linear NaCl gradient. Fractions containing the target protein were pooled and dialyzed against 1 × PBS. The bacterial endotoxin content in the preparations was determined by the LAL test (GeneScript, USA). The protein solution was stored at a temperature of -20 ° C with the addition of glycerol up to 10%.
