Данное изобретение относится к пищевой промышленности и медицины, также касается производства высокоочищенной смеси аминокислот, по их соотношению и количеству полностью соответствующей белку коллагену. Коллаген и его ферментативные гидролизаты широко используются в медицине для лечения опорно-двигательного аппарата и в косметологии. Однако эффективность коллагена многократно возрастает, если использовать его полный гидролизат, т.е. смесь из 15-ти аминокислот коллагена в свободном состоянии. Поступая в желудочно-кишечный тракт, аминокислоты быстрее всасываются в кровь и разносятся по всему организму, обеспечивая воспроизводство коллагена путем биохимического синтеза в кровеносных сосудах, дисках позвоночника, связках костей и в костной ткани. Полный гидролизат коллагена (условно «коллагенит») получают путем высокотемпературного кислотного гидролиза гольевого спилка шкур КРС или другого сырья, содержащего коллаген. Технология не имеет близких аналогов и состоит в следующем: сырье, содержащего коллаген, загружают в эмалированный или стеклянный реактор, затем заливают 16-17% раствор соляной кислоты, герметизируют реактор, нагревают содержимое до +120 градусов и выдерживают при указанной температуре 6 часов. Затем реактор охлаждают до температуры +50-55 градусов, подключают вакуум и отгоняют смесь воды и хлористого водорода, собирая ее в абсорбере, где пары НС1 поглощаются водой, распыляемой сверху форсункой. Оставшийся хлористый водород химически связан с аминокислотами и выделяющимся при гидролизе в виде побочного продукта аммиаком. После отгонки НСl гидролизат фильтруют под вакуумом на нутч-фильтре. Прозрачный фильтрат с рН 1,5-2,0 осветляют, пропуская через ионообменную колонну с ионосорбентом ИА-4-продуктом поликонденсации фенола, формальдегида, уротропина и этиленгликоля микропористой структуры. Осветленный раствор аминокислот пропускают через ионообменную колонну с катеонитом типа ку2 8, где аминокислоты сорбируются вместе с аммиаком. После промывки колонны водой, аминокислоты элюируют с колонны раствором 2Н щелочи, количество которой эквивалентно обменной емкости катионита в колонне. Аммиак отпаривают из элюата под вакуумом, аминокислоты нейтрализуют соляной кислотой рН 4,5-5,0 и осветляют на колонне со смолой МА-4. Entering the gastrointestinal tract, amino acids are more rapidly absorbed into the bloodstream and are distributed throughout the body, ensuring the reproduction of collagen by biochemical synthesis in blood vessels, spinal discs, ligaments of bones and bone tissue. A complete collagen hydrolyzate (conditionally “collagen”) is obtained by high-temperature acid hydrolysis of a glue split of cattle skins or other raw materials containing collagen. The technology has no close analogues and consists in the following: the raw material containing collagen is loaded into an enameled or glass reactor, then a 16-17% hydrochloric acid solution is poured, the reactor is sealed, the contents are heated to +120 degrees and kept at this temperature for 6 hours. Then the reactor is cooled to a temperature of + 50-55 degrees, a vacuum is connected and a mixture of water and hydrogen chloride is distilled off, collecting it in an absorber, where HC1 vapors are absorbed by water sprayed from above by a nozzle. The remaining hydrogen chloride is chemically bound to amino acids and is released by hydrolysis as ammonia by-product. After distillation of the HCl, the hydrolyzate is filtered under vacuum on a suction filter. A transparent filtrate with a pH of 1.5-2.0 is clarified by passing through an ion exchange column with an ion-absorbent IA-4 polycondensation product of phenol, formaldehyde, urotropine and ethylene glycol of a microporous structure. The clarified solution of amino acids is passed through an ion exchange column with ku2 8 type cateonite, where amino acids are sorbed together with ammonia. After washing the column with water, amino acids elute from the column with a 2N alkali solution, the amount of which is equivalent to the exchange capacity of cation exchanger in the column. Ammonia is evaporated from the eluate under vacuum, the amino acids are neutralized with hydrochloric acid, pH 4.5-5.0, and clarified on a column with MA-4 resin. Obtained from a purified solution by freeze drying, the preparation contains at least 90% of free collagen amino acids.

Получают 305 литров 17% соляной кислоты для следующей операции гидролиза. В остаток после отгонки соляной кислоты добавляют 200 литров воды и фильтруют под вакуумом на нутч-фильтре. Получают 17 килограмм черного маслянистого осадка и 300 литров прозрачного фильтрата, содержащего 75 килограмм аминокислот. Фильтрат пропускают через осветляющую ионообменную колонну, загруженную ионосорбентом ИА-4-продуктом поликонденсации фенола, формальдегида, уротропина и этиленгликоля микропористой структуры. Объем колонны составляет 100 литров. Раствор подают снизу со с скоростью 50 л/час. Осветленную фракцию отбирают, когда коэффициент преломления «п» повышается от 1,333 до 1,335 и заканчивают отбирать при снижении «п» до величины 1,334. Сорбированные на катионите аминокислоты элюируют 300 литров 2Н раствора КОН. Элюат представляет собой раствор с запахом аммиака-побочного продукта гидролиза коллагена с рН 11,2. Аммиак удаляют путем вакуум-выпарки при температуре 75 градусов и вакууме 0,8 атм. Упаренный раствор в количестве 250 литров имеет рН 8,7 и содержание сухих веществ 28%. Аминокислоты переводят в Сl- форму добавляя в раствор 46 литров 36% НСl. 296 литров нейтрализованного до рН 5,5 раствора осветляют на колонне, содержащей 100 литров осветляющего сорбента ИА-4. Раствор подают на колонну снизу со скоростью 50 л/час. Контроль процесса ведут по коэффициенту преломления. Фракцию аминокислот отбирают в диапазоне «п» от 1,335 к снижению до 1,334. Получают 376 литров бесцветного раствора без запаха с приятным вкусом и концентрацией аминокислот 23%. Товарная форма продукта-запаянные пакетики из фольги, содержащие 20 миллилитров 20% раствора. The filtrate is passed through a clarifying ion-exchange column loaded with ion-absorbent IA-4 polycondensation product of phenol, formaldehyde, urotropine and ethylene glycol microporous structure. The column volume is 100 liters. The solution is supplied from below with a speed of 50 l / h. The clarified fraction is taken away when the refractive index "p" rises from 1.333 to 1.335 and it is finished taking when the "p" decreases to a value of 1.334. Amino acids are converted to the Cl form by adding 46 liters of 36% Hcl to the solution. 296 liters of a solution neutralized to pH 5.5 are clarified on a column containing 100 liters of clarifying sorbent IA-4. The solution is fed to the column from below at a rate of 50 l / h. The process is controlled by the refractive index. The fraction of amino acids is selected in the range "p" from 1.335 to decrease to 1.334. Get 376 liters of a colorless, odorless solution with a pleasant taste and an amino acid concentration of 23%.
