Abstract
Description
The invention relates to the field of biotechnology, in particular to the stimulation of the destruction of polymer waste by microorganisms - biodestructors.
Currently, packaging consumption is growing rapidly. In Russia, this growth is 5-6% per year. All this makes the problem of packaging disposal urgent. And recycling of polymer packaging comes to the fore. At the same time, the volumes of waste disposal in Russia are at a low level. So, in 2003, polymer waste was disposed of in Russia - only 0.61 million tons (7.7%), which is noticeably less than in 1990 (0.72 million tons and 23.5%, respectively). In general, only 3% of all waste is recycled by industrial methods, the rest is taken to landfills or incinerated. Under landfills and landfills for solid household waste, up to 10 thousand hectares of land are annually alienated, including fertile land withdrawn from agricultural circulation.
The increase in the amount of non-degradable polymer waste in natural conditions leads to environmental pollution and creates a global environmental problem.
т.The accumulation of synthetic polymeric materials, mainly polyolefins (polyethylenes and polypropylenes) is 180 million tons per year. And each year their volume increases by about 25 million tons.
2013. №6. С. 121-123].Active work is underway to develop key areas for cleaning the environment from plastic waste and combating the threatening accumulation of polymer waste. These include different methods of disposal and disposal. Utilization of polymer non-degradable materials includes incineration, pyrolysis, recycling or recycling (RU 2587455 dated 06/20/2016; RU 2617213 dated 04/24/2017). However, none of these methods absolutely improves the environmental situation when burning, highly toxic and supertoxic (furans, dioxins) compounds are formed. And other ways to combat the accumulation of waste with modern technologies for their collection and sorting are quite expensive, because before processing the polymer material, it is necessary to select plastic containers and packaging from household waste, separate the waste by type of plastic, wash, dry, grind and only then recycle into a new polymer product. [Examples O.S., Makeev P.V., Klinkov A.S. Overview of methods for processing waste polymeric materials and analysis of the market for recycled materials // Young scientist. 2013. No.6. S. 121-123].
16-20%.Therefore, the bulk of the polymer waste is stored in landfills, and no more than 16-20% is recycled in developed countries.
One of the possible ways to reduce the giant waste of synthetic plastics is considered composting [Orlova A.M., Popova MN Current problems of municipal solid waste: Monograph. - M .: MGSU, 2010. - 216 p.]. This predominantly aerobic waste recycling process can be considered an analog of nature-like technology. During composting, under the influence of soil and water microorganisms, biopolymers are transformed to humus and then to carbon dioxide and water (end products of the decay of organic matter) in the period from several months to tens of days. The rate of decomposition of materials depends on many factors - the type of polymer, humidity, temperature, light exposure, species structure of the microbial component of the medium, etc.
There are also known methods of microbial decomposition of polymer waste with the participation of accumulative cultures of microorganisms isolated from the soil. So, the dominant forms from the accumulative culture and model systems based on light chestnut soil of the Astrakhan region with the introduction of various polymeric materials are distinguished. The optimal conditions for the development of these microorganisms and the maximum effect of their enzyme systems were studied. When studying the enzymatic activity of the studied microorganisms, special attention was paid to species isolated from the base medium with homogenized polyethylene. After processing the obtained data, it was found that the neutrophilic forms of Bacillus circulans 1, Pseudomonas putida possess the maximum activity spectrum on a base medium with polyethylene; acidophilic forms of Aspergillus, flavus, A. fumigatus,
Great hopes were pinned and are pinned on the use of biodegradable plastics in the national economy [Vlasov SV, Olkhov VV Biodegradable Polymer Materials // Polymer Materials. 2006, No. 7. S. 23-26]. However, the time required for decomposition, for example, of such packaging materials in vivo can be many years and decades. And, according to environmentalists, the simple burial of even biodegradable plastic waste is a “time bomb” and shifting today's problems onto the shoulders of future generations.
In the manufacture of biodegradable polymeric materials, it is taken into account that the process of destruction (destruction) of the base polymer is practically not accelerated. To intensify this process, additives are introduced into the polymer matrix to accelerate its decay under the influence of UV radiation.
There is also a method of stimulating the activity of biodegradable mushrooms by introducing starch into the polymethylacrylate polymer matrix in order to increase the biodegradability of the material. Fungi-resistant samples were considered to have received an assessment of the degree of fouling of not more than 2 points. [Kryazhev D.V. The purpose of this work was to find new ways to accelerate the biodegradation of polymeric materials.
The technical result consists in expanding the arsenal of technical means to stimulate the activity of microorganisms-biodestructors of polymer waste,
обеспечивающих повышенную агрессивность микроорганизмов в отношении данных отходов.providing increased aggressiveness of microorganisms in relation to these wastes.
The technical result is achieved by the fact that the activity of fungi-biodestructors of polymeric wastes is stimulated by using additives of natural origin, while beer yeast autolysate (APD) is used as an additive with the following ratio of APD to fungi to destructors g: million spores 0.02: 10, respectively .
Brewer's yeast autolysate is a brewing waste, including residual brewer's yeast. The amount of this waste is directly related to the amount of beer produced and is approximately 1.2% of the volume of beer. When converted to absolutely dry matter, the annual volume of spent brewer's yeast in Russia may be 1.0-1.3 million tons. Despite the high nutritional value (TU 9184-001-76373465-2007), these wastes do not find wide practical application, although they are studied comprehensively, in particular, as additives in animal feed [Autolized brewer's yeast in pig feed / M. Kirilov, A Yakhin, M. Prishchep, M. Baburina // Compound feed. 2007. - No. 7. - S. 57]. Until now, however, these wastes have not found rational use in animal husbandry, and are mainly exported to the fields in the form of liquid organic fertilizers.
Another direction of the use of the drug APD relates to pharmacology. APD - a product of induced autolysis of brewer's yeast, which has undergone additional purification from cell membranes, contains a large number of components useful for humans. Most of the proteins are split into small peptide chains and individual amino acids. APD is rich in B1 vitamins, amino acids and minerals. Vitamins, micro and macro elements of yeast are in protein complexes.
So, in the composition of the drug under the commercial name "Nagipol", a food supplement containing brewer's yeast autolysate with a pharmacological effect, it is noted that it contains protein substances, amino acids (including 8 essential), and B vitamins (B1-B6 , Be), E, H, F, etc .; carbohydrates, nucleotides, lipids, polysaccharides, enzymes, higher and lower peptides, and other valuable components; macro- and microelements.
We have used the ADF, that is, the hydrolysed residues of the yeast cells Saccaromyces cerevisiae, tested in this work as additives to cultures
микодеструкторов для стимуляции их активности в процессе их воздействия с полимерными материалами.microdestructors to stimulate their activity during their exposure to polymeric materials.
CO2.Assessment of the physiological activity of mycododestructors when APD was added to them and their subsequent development on polymeric materials was carried out by CO 2 emission.
The following APD concentrations added to mycodestructors and polymer waste were prepared and investigated: 0; 0.01; 0.05; 0.10; 0.15; 0.20 (g / cm 3 ). In this case, the spores of the microdestructor were applied in a single dose (in the form of a suspension at a concentration of 2 million spores / cm 3 ) at the rate of 0.0005 cm 3 per 1 cm 2 of the polymer sample.
The processing of polymer samples was carried out as follows.
A suspension of the APD preparation in sterile distilled water was prepared at operating concentrations of 0.01; 0.05; 0.10; 0.15; 0.20 (g / cm 3 ) immediately before processing samples of polymeric materials. Shake well in a flask and pour (under sterile conditions) into a glass or other container into which it is convenient to immerse samples of polymeric materials / waste.
спор/см3; - в суспензию также добавляли ПАВ Твин-80 (1 капля в 10 мл суспензии).Previously, an aqueous suspension of Aspergillus niger spores (as well as other types of microorganisms separately) was prepared using distilled water and a fungal culture on an agarized Chapek medium aged 14-28 days. The spore concentration was calculated by the direct calculation method in the Goryaev chamber. For the study used a suspension of spores of the fungus with a concentration of 2 million spores / cm 3 ; - Tween-80 surfactant was also added to the suspension (1 drop in 10 ml of suspension).
Then a medical mask and rubber gloves were put on and the polymer material under study was treated (a) with an APD suspension, immersed in a vessel for 4-5 seconds, laid out on the grating bottom of the desiccator and dried for 30-40 minutes. Then, a suspension of spores of microdestructors was carefully sprayed from the atomizer at the rate of 0.0005 ml per 1 cm 2 of a polymer sample, preventing the confluence of drops of the spore suspension of Aspergillus niger or another type of microorganism. At the same time, a control sample was prepared in which a suspension of mycodestructors spores was applied to the polymer sample at the same quantitative concentrations without the use of an additive in the form of APD. A metal cuvette and desiccator were used to work with spores of mycodestructors. The work was carried out under sterile conditions in a laminar box.
Next, samples of polymeric materials with the studied area, treated with myocodestructors without APD (control sample) and treated with APD and infected from the surface with a spore suspension of mycodoxidator (experimental sample) were placed in separate Petri dishes and / or on the grating bottom of the desiccator at a distance of 1.5-2 cm from each other and incubated under optimal conditions for the development of microorganisms - at a humidity of 80-85% and a temperature of 24-28 ° C, once or twice a week, moving the lid of the desiccator or Petri dishes to the side for 5-10 seconds. Tanks open and close to allow air circulation. Then, after 28-96 days of incubation during free gas exchange, the samples were transferred to penicillin vials with a volume of 15 cm 3 . Then they were tightly closed and incubated for 24-48 hours at a temperature of 24-28 ° C. Then, an aliquot of a gas sample was taken with a syringe from vials with control and experimental samples in an amount of 0.5-1.0 cm 3 . They determined the chromatographic concentration of carbon dioxide in them, calculating the specific emission of carbon dioxide (μmol / cm 2 / h) in the test sample Vo and the control sample Vк by the formula: Vo or Vк = (So or к * 0.002124 * Mr * v) / (t * s) where
S - средняя площадь пика опытной или контрольной пробы на хроматографе при умножении на коэффициент 0,002124 и Mr, показывающий концентрацию CO2 в мкмолях;S is the average peak area of the experimental or control sample on the chromatograph when multiplied by a factor of 0.002124 and Mr, showing the concentration of CO 2 in micromoles;
Mr - молярная масса углекислого газа (равна 44,01 г/моль); v - объем аликвоты газовой пробы из флакона (см3);Mr is the molar mass of carbon dioxide (equal to 44.01 g / mol); v is the volume of an aliquot of the gas sample from the vial (cm 3 );
t - время экспозиции инкубации пробы в закрытом флаконе, час;t is the exposure time of the incubation of the sample in a closed bottle, hour;
2.s is the surface area of the experimental or control sample, cm 2 .
The biostimulating activity of APD is determined by the excess of carbon dioxide emission in the test sample over the control.
To do this, calculate the ratio of specific carbon dioxide emission in the test sample to the control in% (vol), determining the multiplicity of exceeding the concentration of carbon dioxide (k) in the test sample by the formula: k = Vo / Vк.
When k> 2, the stimulating activity of APD is determined.
1.The test results are presented in table 1.
2.APD was applied at the rate of 0.02 ml per cm 2 .
Spores of the mycodestructor (fungus) were applied in the form of a suspension (concentration of 2 million spores / cm 3 ) at the rate of 0.0005 ml per 1 cm 2 of sample. The best ratio of APD to mycostructure, as can be seen from the table, was 0.02 g of APD to 10 million spores of mycostructure.

Makroflex.For the study, a polymer material was used in the form of a dried plate of Makroflex thin polyurethane sealant.
Samples of the material were cleaned of external contaminants by immersion for 1 minute in ethanol and dried. The consumption of alcohol ranged from 0.05 to 0.1 dm 3 / m 2 . Aspergillus niger spore suspension was preliminarily prepared as described above. A suspension of the APD preparation was prepared at a working concentration of 0.01; 0.02 and 0.03 g / cm 2 . Then immersed in the ADF samples of the polymer material for 4-5 seconds.
Under the laminar box, under the conditions of sterility, plates of polymeric material were prepared in the amount of 3 control and 3 experimental. Control samples were immersed in sterile distilled water, and the experimental samples were immersed in a suspension of APD at a concentration of 0.1 g / cm 3 . Then laid on the grating bottom of the desiccator, dried for 30-40 minutes. After that, the experimental and control samples were treated from a bullet machine with a suspension of Aspergillus niger spores, preventing droplets from merging on the surface.
80%.Samples of polymeric materials with a studied surface area (s) of 9 cm 2 infected without APD (control sample) and infected from the surface with a suspension of Aspergillus niger spores with APD (experimental sample) were placed in desiccators, the bottom of which was filled with water, and incubated for 68 days at a temperature of 29 ° C and a humidity of 80%.
Далее определяли в ней концентрацию диоксида углерода с помощью газового хроматографа (М 3700-4).Then, these samples of polymeric materials, control and experimental, were placed in penicillin vials with a volume of 15 cm 3 . Hermetically closed and incubated for 24 hours at a temperature of 28 ° C. Aliquots of a gas sample (v) in an amount of 1.0 cm 3 were taken from a penicillin vial with a control and experimental samples. Next, the concentration of carbon dioxide was determined in it using a gas chromatograph (M 3700-4).
Vo=(482,81*0,002124*44,01*1)/(24*9)=0,210Three measurements were carried out, while the peak area of the control sample on a chromatograph of three parallel measurements was 352.66; 581.30; 514.46. The calculated average area of the sample peak on the chromatograph (Sк) was equal to 482.81. The molar mass of carbon dioxide (Mr) is 44.01 g / mol. The volume of an aliquot of the gas sample from the bottle (v) was 1 cm 3 . The exposure time of incubation of the sample in a closed vial (t) was 24 hours. The specific emission of carbon dioxide (μmol / cm / h, in the control sample) was calculated by the formula: Vo = (482.81 * 0.002124 * 44.01 * 1) / (24 * 9) = 0.210
Vo=(3253,0*0,002124*44,01*0,5)/(24*4,4)=1,44.Also, three measurements were taken of the peak area of the experimental test in sin parallels on a chromatograph, which amounted to 3549.3; 3015.2; 3195.5. The calculated average area of the sample peak on the chromatograph (Sк) was 3253.00. The molar mass of carbon dioxide (Mr) is 44.01 g / mol. The volume of an aliquot of the gas sample from the bottle (v) was 0.5 cm 3 . The exposure time of incubation of the sample in a closed vial (t) was 24 hours. The specific carbon dioxide emission (μmol / cm2 / hr, in the control sample) was calculated by the formula: Vo = (3253.0 * 0.002124 * 44.01 * 0.5) / (24 * 4.4) = 1.44 .
Затем рассчитывали отношение удельной эмиссии диоксида углерода в опытном образце к контрольному в % (об), определяя кратность превышения концентрации диоксида углерода (k) в опытном образце по формуле: k=1,44/0,21=6,74.Then, the ratio of the specific carbon dioxide emission in the test sample to the control in% (vol) was calculated, determining the multiplicity of exceeding the carbon dioxide concentration (k) in the test sample by the formula: k = 1.44 / 0.21 = 6.74.
Thus, in the prototype, stimulation of the physiological activity of microorganisms was observed, which is reflected in an increase in CO 2 emission. And k is the multiplicity of the excess of the carbon dioxide content (in volume percent) of the experimental over the control amounted to 6.74, which is more than 2. Presented in FIG. 1 illustrations obtained using engineering tomography of a control sample (0.167 mm) and experimental (0.325 mm) show differences in the depth of destruction of the polymer material, which confirms the increased aggressiveness of microorganisms in the variant with the addition of APD.
A study was conducted of the degree of degradation of the polymer material by evaluating the mycelium fouling in points according to GOST 9.048-89. The aggressiveness of biodestructors in the prototype exceeded that in the control, which is presented in FIG. 2, 3 and 4. FIG. 2 shows a control sample of a polymer material treated with A. niger without ADF. The depth of degradation is 0.167 mm. In FIG. 3 shows a prototype polymer material treated with A. niger with ADF. At the same time, APD to microorganisms to destructors g: mln. dispute 0.02: 10, respectively. In FIG. 4 shows samples of biodegradable polyethylene film, where from left to right the first sample is a control not processed by anything; the second sample was treated with a microdestructor - A. niger fungus; the third one was processed only by APD, the fourth sample was processed by A. niger + APD in the stated concentration ratios, and the fifth sample was also processed by A. niger spores + the APD apparatus in the stated concentration ratios and autoclaved. In the fourth and fifth samples, respectively, there is the best effect of the growth of the fungus and its destructive effect on the plastic film.
Thus, the biostimulating drug APD really has a positive effect on the physiological activity of biodestructors and helps to accelerate the degradation of polymer materials.
Given the lack of commercial interest in such brewing waste, the proposed methodology for the processing of polymer waste will not only solve some modern environmental problems, but it is an economically viable waste disposal process.
