Abstract
Description
The invention relates to medical equipment, namely to surface modification of metal medical implants, in particular vascular stents made of TiNi alloys with shape memory effect, and can be used to increase radiopacity, thromboresistance, biocompatibility and fatigue characteristics of stents and other vascular implants, as well as instruments for endoscopic surgery.
[1-5]. Currently, to increase the radiopacity and biocompatibility of TiNi alloys and stents made from these alloys, thin-film tantalum coatings are used that are deposited by various PVD methods [1-5].
[1,4]. The invention [1], which is closest to the claimed invention in terms of features, describes a method for producing a radiopaque, biocompatible tantalum coating for medical devices, such as vascular stents made of TiNi alloy, capable of withstanding high (up to 8%) deformations inherent to these devices. To deposit the Ta coating on TiNi stents, an unbalanced cylindrical magnetron sputtering system was used, containing two tantalum cathodes and operating on direct or alternating (40 kHz) current. Xenon or krypton was used as the working gas. The total magnetron discharge power for both cathodes was 2 kW or 4 kW, and the bias voltage applied to the stents during deposition of the coating was -50 V or -150 V. Previously, the stents were double-cleaned in an ultrasonic bath, blown with nitrogen and dried with hot air in order to increase the adhesion of the coating. The time the MPC entered the operating mode of coating deposition was 10 min. The time required for the deposition of a Ta coating with a thickness of ~ 10 μm was 2.25 and 4.5 h at a power of 4 and 2 kW, respectively. The equilibrium temperature of the stents during coating deposition was completely determined by the deposition process and ranged from 150 to 450 ° C. The Ta coating had a porous columnar structure oriented perpendicular to the surface of the substrate; peeling of the coating did not exceed 1% of the surface area of the coating with a substrate deformation of ~ 8%. The coating was highly radiopaque [1,4].
There is also a known method of forming functional thin-film Ti-Ta coatings on substrates of Ti and Si [6] and quartz glass [7]. In both cases, Ti-Ta coatings were obtained by simultaneous magnetron sputtering of cathodes made of titanium and tantalum at a working gas pressure (Ar) of 0.67 Pa. The thickness of Ti – Ta coatings was 0.3 μm [6] and ~ 3 μm [7], and the coating described in [7] had, as in [1], a columnar structure oriented perpendicular to the substrate surface.
All known Ta coatings deposited by PVD methods on TiNi alloy substrates (hereinafter referred to as TiNi substrates), including the Ta coating claimed in [1], regardless of their thickness and deposition method, have a polycrystalline structure and have the following disadvantages: limited adhesive coating strength associated with the presence of a relatively sharp coating / substrate interface; thermomechanical incompatibility of the Ta coating and TiNi of the substrate (stent), due to the large difference in the temperature coefficients of linear expansion α and the elastic modulus E of tantalum (α = 6.6 × 10 -6 K -1 , E = 190 GPa) [8] and TiNi alloy (B2 phase ) (α = 11 × 10 -6 K -1 , E = 75 GPa) [9]; the presence of porosity [1,4] and texture [1,3-5] in the coating. Together, these factors pose a danger of delamination of the Ta coating and the occurrence of fatigue cracks in the implant under cyclic bending loads and significant deformations (up to ~ 4-6%), which it undergoes during functioning in the human body, i.e. increase the risk of further reducing the safe life of the implant.
Анизотропная структура покрытия в сочетании с резкой границей с подложкой делает эти покрытия малоперспективными с точки зрения усталостных характеристик системы Ti-Ta (покрытие)/TiNi (подложка).Thin film coatings made of Ti- (30–40) at.% Ta alloys with a high-temperature shape memory effect are largely free from thermomechanical incompatibility. The temperature range of the martensitic transformation of these alloys is 50–100 ° C higher than room temperature [10]; therefore, at the temperature of the human body, they are in highly elastic martensitic states. As a result of this, these alloys have elastic characteristics close to TiNi alloys (for the Ti-35 at.% Ta alloy, the value is E ≈ 70 GPa [11]) and are comparable to muscle tissues. These properties are also preserved in thin-film Ti-Ta coatings of the same composition obtained by simultaneous magnetron sputtering of Ti and Ta targets on various substrates [7, 12]. In addition, Ti-Ta alloys have high corrosion resistance and biocompatibility [13], and the presence of a large weight fraction of Ta (~ 70 wt.%) Provides their increased radiopacity compared to TiNi alloys. Coatings from Ti- (30 ÷ 40) at. % Ta obtained, as well as coatings from pure Ta [1,3-5], by magnetron sputtering, have a columnar nanoscale structure [6]. The anisotropic coating structure in combination with a sharp boundary with the substrate makes these coatings unpromising in terms of the fatigue characteristics of the Ti-Ta (coating) / TiNi (substrate) system.
Thus, the deposition of pure Ta coatings and Ti-Ta alloys on TiNi substrates and TiNi implants with the aim of increasing their radiopacity and biocompatibility can most likely lead to a deterioration in the mechanical properties of stents, including their fatigue characteristics, which determine the term of safe functioning .
[15,16].The simultaneous increase in X-ray contrast, biocompatibility and fatigue characteristics of TiNi implants while maintaining the characteristics of the shape memory effect and superelasticity at the initial level can be achieved by forming an amorphous or amorphous-nanocrystalline surface layer with a thickness of up to 5-10 μm with a monotonic diffusion transition to the substrate material. As is known, amorphous metal alloys at the micro-scale level have a homogeneous, isotropic, disordered atomic structure. Due to the absence of grain boundaries and other defects characteristic of ordinary polycrystalline alloys, amorphous alloys, including those based on Ti, which are promising metal biomaterials, have high mechanical, including fatigue characteristics [14,15], as well as high corrosion resistance and biocompatibility [15,16].
The technological problem of the invention is the development of a method for the synthesis of a radiopaque surface Ti-Ta-Ni alloy with an amorphous or amorphous-nanocrystalline structure on a TiNi substrate.
The technical result of the invention is the synthesis of a surface Ti-Ta-Ni alloy with an amorphous or amorphous-nanocrystalline structure on a TiNi substrate with an increased level of radiopacity and thromboresistance.
2.The specified technical result is achieved in that the synthesis of a surface Ti-Ta-Ni alloy on a TiNi substrate is carried out by the additive method by repeatedly alternating in a single vacuum cycle the deposition of an amorphous film and its liquid-phase mixing with the components of the substrate; in this case, films of the composition Ti 60-70 Ta 40-30 (at.%) are used as amorphous-forming films, deposited by the magnetron method by simultaneously sputtering cathodes made of titanium and tantalum. The liquid-phase mixing of the film and substrate components with subsequent high-speed quenching of the molten surface layer is carried out using a wide-aperture low-energy high-current electron beam (NSEC) with parameters: pulse duration τ = 2 ÷ 3 μs, energy density E s = 1.5 ÷ 2.5 J / cm 2 .
50.In the proposed method, the thickness of the amorphous Ti-Ta film in each synthesis cycle is selected in the range from 50 to 100 nm; the number of pulses in each synthesis cycle is from 3 to 5; the number of synthesis cycles from 10 to 50.
Before the first cycle of TiNi synthesis, the substrate is irradiated with an NSEP with an energy density of 3 ÷ 5 J / cm 2 ; the number of pulses is from 20 to 50. During the synthesis, the thickness of the molten surface layer in each pulse is approximately an order of magnitude greater than the thickness of the Ti-Ta film, but less than 1–1.5 μm. The total thickness of the surface Ti-Ta-Ni alloy does not exceed 5 μm. The temperature of the TiNi substrate during the synthesis does not exceed 200 ° C.
In the proposed TiNi method, the substrate before the first synthesis cycle is subjected to repeated (20-50 pulses) irradiation of the NSEC at E s = 3 ÷ 5 J / cm 2 for preliminary cleaning of the surface layer. For the synthesis of the surface Ti – Ta – Ni alloy, films of the composition Ti 60–70 Ta 40–30 (at.%) Were used , deposited by the magnetron method, possessing a high temperature shape memory effect and thermomechanical compatibility with the TiNi substrate. The film thickness in each synthesis cycle is d = 50–100 nm. During the synthesis, the thickness of the molten surface layer in each pulse is approximately an order of magnitude greater than the thickness of the Ti-Ta film, but less than 1–1.5 μm. The total thickness of the surface Ti-Ta-Ni alloy, determined mainly by the number of synthesis cycles, does not exceed 5 μm. The temperature of the TiNi substrate during the synthesis of the surface alloy does not exceed 200 ° C. Thin (1 mm) plates made of TiNi alloy and a thin (diameter of 150 μm) wire for vascular stents were used as TiNi substrates.
N. The main processing parameters that determine the elemental composition, microstructure and thickness of the surface Ti-Ta-Ni alloy are: the thickness of the initial Ti-Ta film d deposited on a TiNi substrate, beam energy density Es and the number of pulses n in each cycle of surface alloy synthesis by liquid-phase mixing of the film and substrate components, as well as the number of synthesis cycles N.
The thickness of the original Ti-Ta films d, energy density Es and the number of pulses n was chosen based on the following interrelated conditions: the value of Es should be limited from below by the melting threshold of the film / substrate system, and its upper limit should be sufficient for melting the surface layer with a thickness of ≤ 1 μm and substantially less than the evaporation threshold of the film; the film thickness d should be much less than the total diffusion path of the film atoms in the substrate melt in each separate synthesis cycle, which is necessary for uniform liquid-phase mixing of the components in the melt.
The diffusion range of the atoms of the film ℓD ≈ (2D⋅tm)0.5where D is the diffusion coefficient of Ta and Ti atoms in liquid TiNi, tm - melt lifetime. Assuming the diffusion coefficient of less mobile Ta atoms in the TiNi melt, D ≈ 5 ∙ 10-5 cm2/ s, and the melt lifetime tm ~ 10-6with [17], we get that for 1 impulse ℓD ≈100 nm. It follows that, with the thickness of the initial Ti-Ta film d = 50 and 100 nm, the above condition will be reliably satisfied if the number of NSEP pulses in each synthesis cycle is n = 5 and 10, respectively.
At τ = 2.5 μs, the surface melting threshold of the TiNi substrate is reached at E s = 1.3–1.5 J / cm 2 [17]. In turn, experiments on samples [film (Ti 60-70 Ta 30-40 , at.%, 50 nm) / substrate (TiNi)] showed that pulsed melting and mixing of the components occurs at E s ≈ 2 J / cm 2 . The thickness of the molten layer was ~ 1 μm.
N = 20 ÷ 30. The number of synthesis cycles N, which, in addition to the final microstructure, determines the total thickness of the surface alloy, was chosen from the condition that this value, on the one hand, should provide an increase in thromboresistance of the surface (lower limit), and on the other hand, should be sufficient to increase the radiopacity (upper limit). It follows that the thickness of the surface alloy should be from ~ 0.5 to ~ 5 μm. This means that, with the initial Ti-Ta film thickness d = 100 nm, the number N should be in the range from 10 to 50. With these data in mind, most experiments on the formation of surface Ti-Ta-Ni alloys were carried out on film systems [(Ti 60- 70 Ta 40-30 , at.%), 50 and 100 nm)] / substrate (TiNi) at E s = 2 J / cm 2 , n = 5 and N = 20 ÷ 30.
Плотность энергии Es, используемой для предварительной обработки подложки и импульсного плавления систем пленка/подложка, контролировали калориметром с точностью не хуже ±15%.The setup includes an electron gun generating an NSEC (10 - 30 keV, beam current up to 25 kA, τ = 2 ÷ 4 μs, beam diameter up to 80 mm) and a planar magnetron sputtering system (MRS) mounted on a common vacuum chamber. The electron gun includes a housing, an explosion-emission cathode, an anode of a high-current reflective discharge forming a plasma anode, and a solenoid. The energy density E s used for pretreatment of the substrate and pulsed melting of the film / substrate systems was controlled by a calorimeter with an accuracy of no worse than ± 15%.
MPC consists of three unbalanced magnetrons, symmetrically arranged around the circumference (at an angle of 28 ° to the normal to the desktop) and is designed to simultaneously spray three materials onto the substrate. Two magnetrons with corresponding targets with a diameter of 75 mm and a thickness of 4 mm were used to sputter the Ti-Ta films; the purity of both targets was no worse than 99.95%. Argon at a pressure of 0.1 and 0.05 Pa, respectively, was used as the working gas during magnetron sputtering and NSEP treatment. Both magnetrons were powered from direct current sources without applying bias to the substrate. To increase the uniformity of film deposition, the work table with TiNi substrates fixed on it rotated around its axis at a speed of 20 rpm. The deposition rate of a Ti-Ta film during the simultaneous sputtering of Ti and Ta cathodes was 1.8 ± 0.3 nm / s at a magnetron discharge power of 175 and 580 W, respectively.
The substrates were made of a TiNi alloy of the ТН1 grade (MATEK-SPF, Russia) in the form of plates with dimensions of 15 × 10 × 1 mm and wire with a diameter of 150 μm. The chemical composition of the TiNi alloy: Ti-55.08Ni-0.051C-0.03O-0.002N (wt.%), The conversion temperature is A Н = 303 K. The initial ones were TiNi substrates, the surfaces of which were subjected to chemical etching, then electrolytic polishing, and then 3x rinsing with distilled water in an ultrasonic bath.
The substrate moves to position B. During TiNi deposition, the substrate rotates at a speed of 20 rpm.
200 °C.The last two operations, alternating, are repeated 20 times (N = 20), therefore, the total thickness of the deposited film is 1 μm. During the synthesis of the surface alloy, the temperature of the TiNi substrate, measured using a thermocouple located on the back of the sample, did not exceed 200 ° C.
1. The elemental composition of the initial amorphous Ti-Ta film was determined by energy-dispersive microanalysis (EDMA) using the INCA EDS system (Oxford Instruments, England) installed on an
Example 1. Synthesis of a surface Ti-Ta-Ni alloy with an amorphous nanocrystalline structure.
2, n = 5, N = 20. Synthesis conditions: pretreatment of TiNi of the NSEC substrate at τ = 2 ÷ 2.5 μs, E s = 3.8 ± 0.7 J / cm 2 , n = 32. The elemental composition of the initial Ti-Ta film No. 1: Ti 69.7 Ta 30.3 (at.%) (see table. 1, TiNi substrate 7), d = 50 nm; surface alloy synthesis at τ = 2 ÷ 2.5 μs, E s = 2 ± 0.4 J / cm 2 , n = 5, N = 20.
In FIG. Figure 2 shows an optical micrograph of the surface Ti-Ta-Ni alloy No. 1 (Table 1, TiNi substrate 7). It is seen that the melted surface is free from microcracks. The microrelief and surface roughness are mainly determined by the topographic features of the TiNi surface of the substrate subjected to preliminary NSEC processing. There are also individual small microcraters that have arisen at the final stage of synthesis. According to the SEM / EDMA data (Table 1, substrate 7), the surface Ti-Ta-Ni alloy No. 1 at depths of 0.2–0.8 μm has the following approximate composition: Ti 64 Ta 25 Ni 11 (at.%). The presence of Ni is associated with the capture of this element from the molten TiNi substrate and its displacement to the surface at the solidification front.
In FIG. Figure 3 shows x-ray diffraction patterns of the TiNi substrate in the initial state (a) and after the synthesis of the surface Ti-Ta-Ni alloy No. 1 (b) on it. The presence of two diffuse halo in the diffractogram (b) in the angle ranges 2θ = 30–55 and 65–80 ° indicates the presence of a rather large volume fraction of the amorphous phase in the surface alloy. Against the background of these halo, in addition to intense B2-phase reflections belonging to the TiNi substrate, numerous low-intensity reflections are clearly detected. The broadening of these reflections indicates that nanocrystalline phases are also present in the surface alloy.
4. In FIG. 4 shows a TEM image of the surface Ti-Ta-Ni alloy No. 1 in cross section, composed of a set of successive bright-field TEM images of all sublayers: from the outer surface layer to the last sublayer adjacent to the unmelted TiNi substrate. According to the TEM / MD / EDMA data, the thickness of the surface alloy, in which the Ta concentration is approximately constant (20–25 at.%), Is ~ 1 μm, which is consistent with the total thickness of the Ta films deposited in 20 cycles (50 nm × 20 = 1000 nm); the total thickness of the surface layer doped with Ta is ~ 2 μm. The surface layer has a gradient multilayer structure consisting of at least 7 sublayers that differ in phase composition, atomic structure and grain size. In FIG. 4 separate sublayers are indicated by
4. A continuous diffuse halo on MD2 means that the
1).The outer (near-surface) layer ~ 200 nm thick has a crystalline structure, the grains of which are crystallographically equally oriented relative to the normal to the surface. 2, n = 5, N = 30.Synthesis conditions: pretreatment of TiNi of the NSEC substrate at τ = 2 ÷ 2.5 μs, E s = 3.8 ± 0.7 J / cm 2 , number of pulses n = 32. Elemental composition of the initial Ti-Ta film No. 2: Ti 59.7 Ta 40.3 (at. %) (Table 1, TiNi substrate 8), d = 50 nm; surface alloy synthesis at τ = 2 ÷ 2.5 μs, E s = 2 ± 0.4 J / cm 2 , n = 5, N = 30.
Ta. The melted surface of the surface Ti-Ta-Ni alloy No. 2, as in the previous example 1, has a smoothed microrelief and does not contain microcracks. According to the SEM / EDMA data (Table 1, TiNi substrate 8), the surface Ti-Ta-Ni alloy No. 2 at depths of 0.2–0.8 μm has an approximate composition of Ti 57 Ta 13 Ni 30 (at.%), I.e. a change in the synthesis mode led to an additional (as compared with example 1) enrichment of the surface alloy Ni and a corresponding decrease in the concentration of Ta.
Характер этих гало свидетельствует о том, что доля аморфной фазы в поверхностном Ti-Ta-Ni сплаве №2 существенно выше по сравнению с поверхностным Ti-Ta-Ni сплавом №1.In FIG. Figure 3c shows the diffraction pattern of the surface Ti – Ta – Ni alloy No. 2 on a TiNi substrate. In this diffraction pattern, as in the previous case (Fig. 3b), there are two diffuse halo (at 2θ = 20 ÷ 50 and 65 ÷ 90 °). The nature of these halo indicates that the proportion of the amorphous phase in the surface Ti-Ta-Ni alloy No. 2 is significantly higher compared to the surface Ti-Ta-Ni alloy No. 1.
[21]. In FIG. Figure 5 shows the bright-field TEM image of the cross section of the surface Ti-Ta-Ni alloy No. 2 and three MD patterns obtained from sites located at different depths. The surface layer consists of a surface Ti – Ta – Ni alloy with a thickness of ~ 2 μm and a transition layer with a thickness of ~ 500 nm bordering the TiNi substrate. Continuous diffuse halo on MD1 and MD2 indicate that the entire surface Ti – Ta – Ni alloy No. 2 has a completely amorphous structure. A similar amorphous structure is possessed by surface Ti – Ta – Ni alloys synthesized on

- Ti70Ta30 (ат. %), d = 1 мкм (табл. 1, TiNi подложка 13);- Ti 70 Ta 30 (at.%), D = 1 μm (table 1, TiNi substrate 13);

0.3 Cu. The radiopacity of TiNi substrates was evaluated on an Axiom Artis dFC angiographic unit (Siemens HealthCare GmbH, Germany) at the Research Institute of Cardiology SB RAS (Tomsk) in the imaging mode used for coronary angiography: voltage 56 kV, current 13 mA, series duration 1 s,
Images of TiNi substrates are shown in FIG. 6. The contrast of the obtained images was measured using the
K=[(RGBфон-RGBi)/RGBфон]×100%,K = [(RGB background -RGB i ) / RGB background ] × 100%,
где i – номер подложки.where i is the number of the substrate.
An analysis of the results (Fig. 6) showed that the presence of a Ti 70 Ta 30 (at%) coating (Table 1, TiNi substrate 13) with a thickness of 1 μm resulted in an increase in the contrast of
Estimates showed that in the case of surface Ti-Ta-Ni alloy No. 1 (Example 1), the same effect of increasing the radiopacity of TiNi substrates (7–8%) can be obtained if the thickness of the surface alloy is ~ 5 μm.
Example 4. The increase in thrombotic resistance.
The thromboresistance of the surface was evaluated on flat

- после НСЭП обработки с целью очистки поверхности (табл. 1, TiNi подложка 2); - after NSEP processing to clean the surface (table. 1, TiNi substrate 2);

Thrombotic resistance of the surface was evaluated according to the procedure described in [23]. To measure the density of platelets (platelet count per 1 mm 2 area), 3 substrates of the same type were taken, 5 measurements were made on each of them in different parts of the surface; according to the results of 15 measurements, average platelet density values were calculated.
The results of measurements of the average platelet density, characterizing the adhesive properties and thromboresistance of the metal surface, depending on the type of treatment, are given in table. 2. It presents data on the average platelet density on the surface of TiNi substrates in the initial state (Table 1, TiNi substrate 1) and after preliminary NSEC processing (Table 1, TiNi substrate 2), as well as after the synthesis of surface Ti-Ta– Ni alloy with an amorphous nanocrystalline structure (Example 1; Table 1, TiNi substrate 7). It can be seen that after preliminary NSEC treatment (Table 1, TiNi substrate 2), the platelet density on the surface of the TiNi substrate decreased by ~ 2.4 times, and after the synthesis of surface Ti-Ta-Ni alloy No. 1 (Example 1; Table 1, TiNi substrate 7) - by ~ 6.7 times in comparison with the initial state (Table 1, TiNi substrate 1). This means that platelet adhesion to the surface of a TiNi substrate subjected to multiple pulsed melting (Table 1, TiNi substrate 2) decreased by more than 2 times, and after the synthesis of surface Ti-Ta-Ni alloy No. 1 (Table 1, TiNi substrate 7) - almost 7 times in comparison with the initial state. Thus, the multiple pulsed melting of a TiNi substrate and the subsequent synthesis of a surface Ti – Ta – Ni alloy with an amorphous nanocrystalline structure on it allow one to increase thromboresistance of the surface by ~ 2.4 and ~ 7 times, respectively.
